Nitrogen (N) isotope systematics were investigated at two high-elevation ombrotrophic peat bogs polluted by farming and heavy industry. Our objective was to identify N sources and sinks for isotope mass balance considerations. For the first time, we present a time-series of δ15Ν values of atmospheric input at the same locations as δ15Ν values of living Sphagnum and peat. The mean δ15Ν values systematically increased in the order: input NH4 + (?10.0‰) < input NO3 ? (?7.9‰) < peat porewater (?5.6‰) < Sphagnum (?5.0‰) < shallow peat (?4.2‰) < deep peat (?2.2‰) < runoff (?1.4‰) < porewater N2O (1.4‰). Surprisingly, N of Sphagnum was isotopically heavier than N of the atmospheric input (P < 0.001). If partial incorporation of reactive N from the atmosphere into Sphagnum was isotopically selective, the residual N would have to be isotopically extremely light. Such N, however, was not identified anywhere in the ecosystem. Alternatively, Sphagnum may have contained an admixture of isotopically heavier N. Ambient air contains such N in the form of N215ΝN2 = 0‰). Because high energy is required to break the triple bond, microbial N fixation is likely to proceed only under limited availability of pollutant N. Also for the first time, a δ15Ν comparison is presented between anoxic deeper peat and porewater N2O. Isotopically light N is removed from anoxic substrate by denitrification, whose final product, N2, escapes into the atmosphere. Porewater N2O is an isotopically heavy residuum following partial N2O reduction to N2.  相似文献

The stable isotope composition of spines produced serially from the apex of columnar cacti has the potential to be used as a record of changes in climate and physiology. To investigate this potential, we measured the δ18O, δ13C and F14C values of spines from a long-lived columnar cactus, saguaro (Carnegiea gigantea). To determine plant age, we collected spines at 11 different heights along one rib from the stem apex (3.77 m height) to the base of a naturally occurring saguaro. Fractions of modern carbon (F14C) ranged from 0.9679 to 1.5537, which is consistent with ages between 1950 and 2004. We observed a very strong positive correlation (r = 0.997) between the F14C age of spines and the age of spines determined from direct and repeated height measurements taken on this individual over the past 37 years. A series of 96 spines collected from this individual had δ18O values ranging from 38‰ to 50‰ [Vienna standard mean ocean water (VSMOW)] and δ13C values from ?11.5‰ to ?8.5‰ [Vienna Peedee belemnite (VPDB)]. The δ18O and δ13C values of spines were positively correlated (r = 0.45, P < 0.0001) and showed near-annual oscillations over the ~15-year record. This pattern suggests that seasonal periods of reduced evaporative demand or greater precipitation input may correspond to increased daytime CO2 uptake. The lowest δ18O and δ13C values of spines observed occurred during the 1983 and 1993 El Niño years, suggesting that the stable isotope composition recorded in spine tissue may serve as a proxy for these climate events. We compared empirical models and data from potted experimental cacti to validate these observations and test our hypotheses. The isotopic records presented here are the first ever reported from a chronosequence of cactus spines and demonstrate that tissues of columnar cacti, and potentially other long-lived succulents, may contain a record of past physiological and climatic variation.  相似文献

We conducted a comparative analysis of the fatty acid (FA) composition and the ratios of stable isotopes of carbon (δ13C) and nitrogen (δ15N) in soft tissues of ten species of bivalve mollusks collected simultaneously on adjacent biotopes in shallow Vostok Bay (the Sea of Japan). Comparison of the FA composition of the lipids of digestive gland and all soft tissues showed that the percentages of C16 and C18 marker FAs were greater in the digestive gland and the levels of marker C20 and C22 FAs were, in most cases, higher in soft tissues. According to the results of cluster analysis and principal component analysis, four groups of samples were identified with a similarity of the FA composition of more than 80% within groups. The carbon stableisotope ratios varied within very wide limits in the studied species of bivalves; the range of δ13C variations was 8.1‰. The range of δ15N variations was much smaller, 2.5‰. Two pairs of species of mollusks (Saxidomus purpurataProtothaca euglypta and P. jedoensisDiplodonta semiasperoides) did not differ in the values of both δ15N and δ13C, the remaining species differed in at least one of these parameters. The greatest similarity of the FA composition and stable-isotope ratios was found in species that inhabit similar substrates, except Macoma irus and D. semiasperoides. Particularly marked differences in the FA composition and stable-isotope ratios were found between a filter-/surface deposit-feeder M. irus and filter-feeders Arca boucardi and Mytilus coruscus that live next to this species.  相似文献

Methane-derived carbon (MDC) can subsidize lake food webs. However, the trophic transfer of MDC to consumers within macrophyte vegetation is largely unknown. We investigated the seasonality of δ13C in larval chironomids within Nelumbo nucifera (Gaertn.) and Trapa natans var. Japonica (Nakai) vegetation in the shallow, eutrophic Lake Izunuma in Japan. Over the past several years, N. nucifera has rapidly expanded across more than 80% of the lake surface. Prior to the expansion of N. nucifera (2007–2008), a previous study reported extremely low larval δ13C levels with peak sediment methane concentrations in August or September. After the expansion of N. nucifera (2014–2015), we observed extreme hypoxia as low as or lower than 1 mg l?1 among the macrophyte coverage during June and August. During August and September, no larvae could be found among N. nucifera, and larvae in T. natans showed relatively high δ13C levels (>???40‰). In contrast, larvae were markedly 13C–depleted (down to ??60‰) during October and November. The renewed supply of oxygen to the lake bottom may stimulate MOB activity, leading to an increase in larval assimilation of MDC. Our results suggest that macrophyte vegetation can affect the seasonality of MDC transfer to benthic consumers under hypoxic conditions in summer.  相似文献

Effects of oxygen transfer on recombinant protein production by Pichia pastoris under glyceraldehyde-3-phosphate dehydrogenase promoter were investigated. Recombinant glucose isomerase was chosen as the model protein. Two groups of oxygen transfer strategies were applied, one of which was based on constant oxygen transfer rate where aeration rate was Q O/V = 3 and 10 vvm, and agitation rate was N = 900 min?1; while the other one was based on constant dissolved oxygen concentrations, C DO = 5, 10, 15, 20 and 40 % in the fermentation broth, by using predetermined exponential glucose feeding with μ o = 0.15 h?1. The highest cell concentration was obtained as 44 g L?1 at t = 9 h of the glucose fed-batch phase at C DO = 20 % operation while the highest volumetric and specific enzyme activities were obtained as 4440 U L?1 and 126 U g?1 cell, respectively at C DO = 15 % operation. Investigation of specific enzyme activities revealed that keeping C DO at 15 % was more advantageous with an expense of relatively higher by-product formation and lower specific cell growth rate. For this strategy, the highest oxygen transfer coefficient and oxygen uptake rate were K L a = 0.045 s?1 and OUR = 8.91 mmol m?3 s?1, respectively.  相似文献

The variability of modern Cyprideis salebrosa and Cyprideis americana (Ostracoda) from the northern Neotropics were investigated in order to understand site specific influences on the isotopic composition of their valves (δ18O, δ13C) in comparison to their host water and to connect this to morphological features of their valves (valve size, nodosity). C. salebrosa was found in a stream (Shell Creek, Florida) and a slightly brackish lake (Laguna del Rincon, Dominican Republic; salinity <0.7 psu) while C. americana occurred in a coastal lake with polyhaline waters (Parrotee Pond, Jamaica; salinity: >20 psu). Valve size and position of nodes differed between the two species. A reverse temperature dependency have been considered to influence Shell length (seasonally in Shell Creek, summer: 1076 µm; winter: 1092 µm, supposedly permanently in Laguna del Rincon, 1035 µm). But, regarding the small dataset other factors could not be excluded to influence ostracod valve size. A decline of node frequency of C. salebrosa is mainly related to an increase in salinity. Isotopic values of ostracod valves reflect the trend in stable isotopes of their host water. Variations in Cyprideis salebrosa δ18O and δ13C values signify differences in their host water. Offsets of ostracod valves to a theoretical calcite precipitated in their host water with an uncertain time lag (+0.015 to +2.63 ‰) needs to be clarified. This study presents a contribution to the understanding of environmental influences on modern ostracod shell characters as basis for paleontological applications.  相似文献

The isotope composition of fruits and seeds of Hordeum vulgare L. (barley), Triticum aestivum/durum (wheat) and Vicia faba var. minor (faba bean) from three chronological phases between 2200 and 1321 cal bc of the settlement Cerro del Castillo de Alange (SW Iberian Peninsula) have been studied. The δ13C values for cereals were between ?24.40 and ?20.39‰ (V-PDB), with a mean of ?22.01‰, the discrimination (Δ) being 15.96‰. The legumes registered similar values, between ?26.25 and ?20.49‰ (mean = ?22.59‰), with a differential for the period of 16.51‰. In both cases, a change was noted from the first phase, where water availability is clearly a limiting factor for plant development. In subsequent phases the growing conditions appear be wetter. By comparison, we measured samples of Quercus ilex-coccifera (oak) charcoal, which shows similar values throughout the series. This suggests that there was no significant climate shift to moister conditions that could explain the above results, but rather they were the consequence of a change in crop management. In addition, we measured samples of a current rainfed Triticum sp. (year 2014), which averaged Δ13C 15.56‰. The changes between the three phases could indicate the development of mixed models of exploitation that combine strategies based on the use of rainfed and potentially irrigated areas during the 2nd millennium bc. The implementation of such a cropping technique, taking advantage of the river banks, could be a response to the processes of climatic degradation that begin in the Middle Holocene. The situation of the Cerro del Castillo reservoir between the rivers of the Guadiana and Matachel rivers is consistent with the development of this type of practice indicated by the isotopic and archaeobotanical data.  相似文献

Objective

To produce δ-decalactone from linoleic acid by one-pot reaction using linoleate 13-hydratase with supplementation with whole Yarrowia lipolytica cells.

Results

Whole Y. lipolytica cells at 25 g l?1 produced1.9 g l?1 δ-decalactone from 7.5 g 13-hydroxy-9(Z)-octadecenoic acid l?1 at pH 7.5 and 30 °C for 21 h. Linoleate 13-hydratase from Lactobacillus acidophilus at 3.5 g l?1 with supplementation with 25 g Y. lipolytica cells l?1 in one pot at 3 h produced 1.9 g l?1 δ-decalactone from 10 g linoleic acid l?1 via 13-hydroxy-9(Z)-octadecenoic acid intermediate at pH 7.5 and 30°C after 18 h, with a molar conversion yield of 31 % and productivity of 106 mg l?1 h?1.

Conclusion

To the best of our knowledge, this is the first production of δ-decalactone using unsaturated fatty acid.

The purpose of this study was to investigate the short-term effects of maize (Zea mays)-fallow rotation, residue management, and soil water on carbon mineralization in a tropical cropping system in Ghana. After 15 months of the trial, maize–legume rotation treatments had significantly (P?C 0 (μg CO2–C g?1) than maize–elephant grass (Pennisetum purpureum) rotations. The C 0 for maize–grass rotation treatments was significantly related to the biomass input (r?=?0.95; P?=?0.05), but that for the maize–legume rotation was not. The soil carbon mineralization rate constant, k (per day), was also significantly related to the rotation treatments (P?k values for maize–grass and maize–legume rotation treatments were 0.025 and 0.036 day?1 respectively. The initial carbon mineralization rate, m 0 (μg CO2–C g?1 day ?1), was significantly (P?θ. The m 0 ranged from 3.88 to 18.67 and from 2.30 to 15.35 μg CO2–C g?1 day?1 for maize–legume and maize–grass rotation treatments, respectively, when the soil water varied from 28% to 95% field capacity (FC). A simple soil water content (θ)-based factor, f w, formulated as: \(f_{\text{w}} = \left[ {\frac{{\theta - \theta _{\text{d}} }}{{\theta _{{\text{FC}}} - \theta _{\text{d}} }}} \right]\), where θ d and θ FC were the air-dry and field capacity soil water content, respectively, adequately described the variation of the m 0 with respect to soil water (R 2?=?0.91; RMSE?=?1.6). Calcite-rich columnar stromatolites grew in perennially ice-covered Lake Joyce in the McMurdo Dry Valleys, Antarctica, during a period of environmental change associated with rising lake level. Stromatolite calcite contains carbon and oxygen isotope records of changes to microbial activity in response to variable light environments and water chemistry through time. The stromatolites grew synchronously with correlative calcite zones. The innermost (oldest) calcite zone has a wide range of δ13Ccalcite values consistent with variable photosynthetic effects on local DIC 13C/12C. Subsequent calcite zones preserve a progressive enrichment in δ13Ccalcite values of approximately + 2.6‰ through time, with δ13Ccalcite values becoming less variable. This enrichment likely records the removal of 12C by photosynthesis from the DIC reservoir over decades, with photosynthetic effects decreasing as light levels became lower and more consistent through time. Mean δ18Ocalcite values of the innermost calcified zone were at least 1‰ lower than those of the other calcified zones (t test p-level < 0.001). The significant difference in δ18Ocalcite values between the innermost and other calcified zones could be a product of mixing source waters with different isotopic values associated with the initiation of lake stratification associated with rising lake level. Overall, Lake Joyce stromatolites record significant lateral variability in relative photosynthetic rate and long-lived lake water stratification with microbial modification of the DIC pool. Such processes provide criteria for interpreting microbial activity within polar paleolake deposits and may shed light on variability in lake environments associated with changing climate in the McMurdo Dry Valleys.  相似文献

The biotransformation of hexachlorocyclohexane isomers (HCH) by two Dehalococcoides mccartyi strains (195 and BTF08) and an enrichment culture was investigated and compared to conversion by the obligate anaerobic strain Clostridium pasteurianum strain DSMZ 525. The D. mccartyi strains preferentially transformed γ-HCH over α-HCH and δ-HCH isomers while β-HCH biotransformation was not significant. In case of the enrichment culture, γ-HCH was preferentially transformed over the δ-HCH, β-HCH and α-HCH isomers. Major observed metabolites in both cases were tetrachlorocyclohexene and as end products monochlorobenzene (MCB) and benzene. Dechlorination of the γ-HCH isomer was linked to an increase in cell numbers for strain 195. γ-HCH transformation was linked to considerable carbon stable isotope fractionation with the enrichment factor εc?=???5.5?±?0.8‰ for D. mccartyi strain 195, εc?=???3.1?±?0.4‰ for the enrichment culture and εc?=???4.1?±?0.6‰ for co-metabolic transformation by C. pasteurianum.  相似文献

Carbon balancing within the plant species is an important feature for climatic adaptability. Photosynthesis and respiration traits are directly linked with carbon balance. These features were studied in 20 wild rice accessions Oryza spp., and cultivars. Wide variation was observed within the wild rice accessions for photosynthetic oxygen evolution or photosynthetic rate (A), dark (R d), and light induced respiration (LIR) rates, as well as stomatal density and number. The mean rate of A varied from 10.49 μmol O2 m?2 s?1 in cultivated species and 13.09 μmol O2 m?2 s?1 in wild spp., The mean R d is 2.09 μmol O2 m?2 s?1 and 2.31 μmol O2 m?2 s?1 in cultivated and wild spp., respectively. Light induced Respiration (LIR) was found to be almost twice in wild rice spp., (16.75 μmol O2 m?2 s?1) compared to cultivated Oryza spp., Among the various parameters, this study reveals LIR and A as the key factors for positive carbon balance. Stomatal contribution towards carbon balance appears to be more dependent on abaxial surface where several number of stomata are situated. Correlation analysis indicates that R d and LIR increase with the increase in A. In this study, O. nivara (CR 100100, CR 100097), O. rufipogon (IR 103404) and O. glumaepatula (IR104387) were identified as potential donors which could be used in rice breeding program. Co-ordination between gas exchange and patchiness in stomatal behaviour appears to be important for carbon balance and environmental adaptation of wild rice accessions, therefore, survival under harsh environment.  相似文献

Ecological applications of stable isotope data require knowledge on the isotopic turnover rate of tissues, usually described as the isotopic half-life in days (T 0.5) or the change in mass (G 0.5). Ecological studies increasingly analyse tissues collected non-destructively, such as fish fin and scales, but there is limited knowledge on their turnover rates. Determining turnover rates in situ is challenging, with ex situ approaches preferred. Correspondingly, T 0.5 and G 0.5 of the nitrogen stable isotope (δ15N) were determined for juvenile barbel Barbus barbus (5.5 ± 0.6 g starting weight) using a diet-switch experiment. δ15N data from muscle, fin and scales were taken during a 125 day post diet-switch period. Whilst isotopic equilibrium was not reached in the 125 days, the δ15N values did approach those of the new diet. The fastest turnover rates were in more metabolically active tissues, from muscle (highest) to scales (lowest). Turnover rates were relatively slow; T 0.5 was 84 (muscle) to 145 (scale) days; G 0.5 was 1.39 × body mass (muscle) to 2.0 × body mass (scales), with this potentially relating to the slow growth of the experimental fish. These turnover estimates across the different tissues emphasise the importance of estimating half-lives for focal taxa at species and tissue levels for ecological studies.  相似文献

Abstract

Annual tree-ring cellulose oxygen isotope values (δ18O) of Fokienia hodginsii provide a promising proxy of monsoon-season precipitation in Southeast China. Measuring intra-annual cellulose δ18O values may reveal the seasonal variability of precipitation and the associated climate influences. Here, we examine intra-annual variation of cellulose δ18O values in Fokienia hodginsii and Cryptomeria fortune from Fujian Province, Southeast China. Both species exhibited considerable intra-annual variations in cellulose δ18O (range ~6 ‰) with a consistent pattern of enriched values near the annual ring boundary and depleted values in the central portion of the ring. Seasonal patterns in the tree-ring δ18O values generally followed changes in precipitation δ18O values. Compared with annual tree-ring cellulose δ18O, intra-annual cellulose δ18O has potential to reconstruct precipitation with higher resolution and stronger signal intensity. July tree-ring cellulose δ18O is significantly correlated (r = ?0.58, p < 0.05) with July precipitation, and June–August tree-ring cellulose δ18O and annual tree-ring cellulose δ18O, respectively, explain 52 and 41 % of the actual variance of April–August precipitation. In addition, May–October cellulose δ18O values during El Niño years are higher than in La Niña years, and April to October rainfall is lower in El Niño years than in La Niña years. Combining the significant correlations between inter-annual cellulose δ18O values and sea surface temperatures in the central tropical Pacific, our results support the hypothesis that El Niño–Southern Oscillation affects tree-ring cellulose δ18O in Southeast China by modulating seasonal precipitation.

Water deficit is one of the key factors that limits the carbon (C) assimilation and productivity of plants. The effect of variable water deficit on recently root-derived bicarbonate assimilation in Camptotheca acuminate seedlings was investigated. Three-month-old seedlings were subjected to three water regimes, well-watered (WW), moderate stress (MS), and severe stress (SS) induced by polyethyleneglycol, in conjunction with relatively high (H) and low (L) natural 13C-abundance of NaHCO3-labeled treatments in hydroponics for 14 days. The δ13C of the newly expanded leaves in H were generally more enriched in heavy isotopes than were those in L, indicative of the involvement of bicarbonate in aboveground tissues. The C isotope fractionation of newly expanded leaves relative to air (?13Cair-leaves) ranged from 17.78 to 21.78‰ among the treatments. The ?13Cair-leaves under the MS and SS treatments in H were both more negative than was that in L. A linear regression between Ci/Ca and ?13Cair-leaves in both L and H were different from the theoretical regression. On the basis of the two end-member mixing model, the proportion of fixed CO2 supplied from bicarbonate contributing to the total photosynthetically inorganic C assimilation were 10.34, 20.05 and 16.60% under the WW, MS, and SS treatments, respectively. These results indicated that the increase in water deficit decreased the atmospheric CO2 gain but triggered a compensatory use of bicarbonate in C. acuminate seedlings.  相似文献

In some environments, species may exhibit trophic plasticity, which allows them to extend beyond their assigned functional group. For Gammarus minus, a freshwater amphipod classified as a shredder or detritivore, cave populations have been observed consuming heterotrophs as well as shredding leaves, and therefore may be exhibiting trophic plasticity. To test this possibility, we examined the C and N stable isotope and C/N ratios for cave and spring populations of G. minus. A 15-day feeding experiment using leaves and G. minus from a spring population established that the diet-tissue discrimination factor was 3.2 ‰ for δ15N. Cave G. minus were 8 ‰ higher in δ15N relative to cave leaves, indicating they did not derive nitrogen from leaves, whereas field collected spring populations were 2–3 ‰ higher than spring leaves, indicating that they did. Cave G. minus were 2.6 ‰ higher in δ15N than the cave isopod, Caecidotea holsingeri. Relative to spring populations, Organ Cave G. minus were 15N enriched by 6 ‰, suggesting they occupied a different trophic level, or incorporated an isotopically distinct N source. While stable isotopes cannot tell what the cave G. minus are eating, the isotopes certainly show that G. minus are not eating leaves and are trophically distinct form the surface populations. Differences in C/N ratios were observed, but reflect the size of the G. minus examined and not feeding group or habitat. The isotope data strongly support the hypothesis that cave populations of G. minus have become generalist or omnivorous by including animal protein in their diet.  相似文献

Atmospheric nitrogen deposition poses a major threat to global biodiversity. Tropical epiphytic plants are especially at risk given their reliance on atmospheric sources of nutrients. The leaf, pseudobulb, and root carbon and nitrogen content, C:N ratio, as well as the nitrogen isotopic composition were studied for individuals of Laelia speciosa from a city and from an oak forest in Mexico. The nitrogen content of leaves was similar between the city and the oak forest, reaching 1.3 ± 0.2 % (dry mass). The δ15N of leaves, pseudobulbs, and roots reached 5.6 ± 0.2 ‰ in the city, values found in sites exposed to industrial and vehicular activities. The δ15N for plant from the oak forest amounted to –3.1 ± 0.3 ‰, which is similar to values measured from sites with low industrial activities. Some orchids such as Laelia speciosa produce a single pseudobulb per year, i.e., a water and nutrient storage organ, so the interannual nitrogen deposition was studied by considering the ten most recent pseudobulbs for plants from either site formed between 2003 and 2012. The C:N ratio of the ten most recent pseudobulbs from the oak forest, as well as that of the pseudobulbs formed before 2010 for plants in the city were indistinguishable from each other, averaging 132.4 ± 6.5, while it was lower for the two most recent pseudobulbs in the city. The δ15N values of pseudobulbs from the oak forest averaged ?4.4 ± 0.1 ‰ for the entire series. The δ15N ranged from 0.1 ± 1.6 ‰ for the oldest pseudobulb to 4.7 ± 0.2 ‰ for the pseudobulb formed in the city from 2008 onwards. Isotopic analysis and the C:N ratio for L. speciosa revealed that rates of nitrogen deposition were higher in the city than in the forest. The δ15N values of series of pseudobulbs showed that it is possible to track nitrogen deposition over multiple years.  相似文献

A Gram-stain negative, aerobic, motile by flagella, rod-shaped strain (THG-T16T) was isolated from rhizosphere of Hibiscus syriacus. Growth occurred at 10–40 °C (optimum 28–30 °C), at pH 6.0–8.0 (optimum 7.0) and at 0–1.0% NaCl (optimum 0%). Based on 16S rRNA gene sequence analysis, the near phylogenetic neighbours of strain THG-T16T were identified as Nibribacter koreensis KACC 16450T (98.6%), Rufibacter roseus KCTC 42217T (94.7%), Rufibacter immobilis CCTCC AB 2013351T (94.5%) and Rufibacter tibetensis CCTCC AB 208084T (94.4%). The DNA G+C content of strain THG-T16T was determined to be 46.7 mol%. DNA–DNA hybridization values between strain THG-T16T and N. koreensis KACC 16450T, R. roseus KCTC 42217T, R. immobilis CCTCC AB 2013351T, R.tibetensis CCTCC AB 208084T were 33.5?±?0.5% (31.7?±?0.7% reciprocal analysis), 28.1?±?0.2% (25.2?±?0.2%), 17.1?±?0.9% (10.2?±?0.6%) and 8.1?±?0.3% (5.2?±?0.1%). The polar lipids were identified as phosphatidylethanolamine, two unidentified aminophospholipids, an unidentified aminolipid and three unidentified lipids. The quinone was identified as MK-7 and the polyamine as sym-homospermidine. The major fatty acids were identified as C16:1 ω5c, C17:1 ω6c, iso-C15:0, summed feature 3 (C16:1 ω7c and/or C16:1 ω6c) and summed feature 4 (iso-C17:1 I and/or anteiso-C17:1 B). On the basis of the phylogenetic analysis, chemotaxonomic data, physiological characteristics, and DNA–DNA hybridization data, strain THG-T16T represents a novel species of the genus Nibribacter, for which the name Nibribacter flagellatus sp. nov. is proposed. Horse heart carboxymethylated cytc (CM-cytc) displays myoglobin-like properties. Here, the effect of cardiolipin (CL) liposomes on the nitrite reductase activity of ferrous CM-cytc [CM-cytc-Fe(II)], in the presence of sodium dithionite, is reported between pH 5.5 and 7.6, at 20.0 °C. Cytc-Fe(II) displays a very low value of the apparent second-order rate constant for the NO2 ?-mediated conversion of cytc-Fe(II) to cytc-Fe(II)-NO [k on = (7.3 ± 0.7) × 10?2 M?1 s?1; at pH 7.4], whereas the value of k on for NO2 ? reduction by CM-cytc-Fe(II) is 1.1 ± 0.2 M?1 s?1 (at pH 7.4). CL facilitates the NO2 ?-mediated nitrosylation of CM-cytc-Fe(II) in a dose-dependent manner, the value of k on for the NO2 ?-mediated conversion of CL–CM-cytc-Fe(II) to CL–CM-cytc-Fe(II)-NO (5.6 ± 0.6 M?1 s?1; at pH 7.4) being slightly higher than that for the NO2 ?-mediated conversion of CL–cytc-Fe(II) to CL–cytc-Fe(II)-NO (2.6 ± 0.3 M?1 s?1; at pH 7.4). The apparent affinity of CL for CM-cytc-Fe(II) is essentially pH independent, the average value of B being (1.3 ± 0.3) × 10?6 M. In the absence and presence of CL liposomes, the nitrite reductase activity of CM-cytc-Fe(II) increases linearly on lowering pH and the values of the slope of the linear fittings of Log k on versus pH are ?1.05 ± 0.07 and ?1.03 ± 0.03, respectively, reflecting the involvement of one proton for the formation of the transient ferric form, NO, and OH?. These results indicate that Met80 carboxymethylation and CL binding cooperate in the stabilization of the highly reactive heme-Fe atom of CL–CM-cytc.  相似文献

The thermostable bifunctional CMCase and xylanase encoding gene (rBhcell-xyl) from Bacillus halodurans TSLV1 has been expressed in Escherichia coli. The recombinant E. coli produced rBhcell-xyl (CMCase 2272 and 910 U L?1 xylanase). The rBhcell-xyl is a ~62-kDa monomeric protein with temperature and pH optima of 60 °C and 6.0 with T1/2 of 7.0 and 3.5 h at 80 °C for CMCase and xylanase, respectively. The apparent K m values (CMC and Birchwood xylan) are 3.8 and 3.2 mg mL?1. The catalytic efficiency (k cat/K m ) values of xylanase and CMCase are 657 and 171 mL mg?1 min?1, respectively. End-product analysis confirmed that rBhcell-xyl is a unique endo-acting enzyme with exoglucanase activity. The rBhcell-xyl is a GH5 family enzyme possessing single catalytic module and carbohydrate binding module. The action of rBhcell-xyl on corn cobs and wheat bran liberated reducing sugars, which can be fermented to bioethanol and fine biochemicals.  相似文献
