Aims
The present study was planned to investigate the diversity of 1-aminocyclopropane-1-carboxylate (ACC) deaminase producing bacteria from the rhizosphere of wheat plants and subsequent evaluation of selected PGPR on growth enhancement of wheat seedlings under drought and saline conditions.Methods
ACC deaminase producing plant growth promoting rhizobacteria (PGPR) were isolated from the rhizosphere of wheat and identified using 16S rRNA gene sequence analysis. Isolates were evaluated for various direct and indirect plant growth promoting (PGP) traits. Plant inoculation experiment was conducted using isolates IG 19 and IG 22 in wheat to assess their plant growth promotion potential under salinity and drought stress.Results
Thirty-eight ACC deaminase producing PGPR were isolated which belonged to 12 distinct genera and falling into four phyla γ-proteobacteria, β-proteobacteria, Flavobacteria and Firmicutes. Klebsiella sp. was the most abundant genera and followed by Enterobacter sp. The isolates exhibited ACC deaminase activities ranging from 0.106–0.980 μM α- ketobutyrate μg protein?1 h?1. The isolates showed multiple PGP traits such as IAA production, phosphate, zinc, potassium solubilization and siderophore production. Enterobacter cloacae (IG 19) and Citrobacter sp. (IG 22) inoculated wheat seedlings showed notable increases in fresh and dry biomass under non-stress as well as under stressed condition.Conclusion
To the best of our knowledge this is the first report of presence of ACC deaminase activity and other PGP traits from the genus Citrobacter and Empedobacter. Our finding revealed that the γ-proteobacteria group dominated the wheat rhizosphere. Plant inoculation with PGPR could be a sustainable approach to alleviate abiotic stresses in wheat plants. These native PGPR isolates could be used as potential biofertilizers for sustainable agriculture.2.

Introduction
Mediterranean winter crops are commonly and increasingly exposed to irregular rainfall and high temperatures, which lead to transient drought events of different degrees, adversely affecting growth and yield. Hence, exploring the diverse degrees of tolerance to drought existing in the crop and the molecular strategies behind it is pivotal for the development of ad hoc breeding programs.Objective
We investigated the physiological and metabolic response of six commercial wheat cultivars to transient water stress at the tillering and grain-filling stages.Methods
Drought experiments in lysimeters were set up at two developmental stages including six wheat cultivars. Newly expanded youngest leaves and flag leaves were sampled during the drought and following recovery. Metabolite profiles were generated using a GC–MS based protocol. Data on transpiration were continually acquired by measuring the weight variation of pots using electronic temperature compensated load cells.Results
The tillering stage in wheat is more sensitive to droughts than the grain filling stage. The former stage was characterized by pronounced metabolic alterations also during recovery from the drought, and plants exhibited reduced transpiration. Notably, cultivars varied considerably in their susceptibility to drought. Exceptionally only in cv Zahir was transpiration not reduced at tillering. During recovery, the transpiration rate of Yuval and Zahir was not significantly affected, while except Ruta the other varieties maintained lower values. At grain-filling, a moderate decrease in transpiration in response to drought was evident in Bar-Nir, Yuval and Zahir varieties as compared with the stronger response of Gedera, Galil and Ruta. The transpiration trend during recovery remained lower than the control plants, particularly in Gedera and Zahir, while it reached higher values than control plants in Yuval and Ruta varieties. Metabolite profiling of leaves across cultivars showed varietal specific trends of response. Particularly during tillering, amino acid metabolism was differentially regulated across cultivars. For instance, Ruta and Zahir exhibited major changes in central carbon nitrogen metabolism during stress response, accumulating large amounts of proline and threonine during tillering, while in Bar-Nir a general decrease in relative amino acid content was noted. Changes in stress related GABA were common to Galil, Ruta, Yuval and Zahir. Desiccation related raffinose family oligosaccharides were mostly associated with a later stage of grain-filling and recovery stages of response.Conclusion
The results indicate the occurrence of stage-dependent metabolic diversification along with a physiological response during transient droughts among wheat cultivars. It can be concluded that the most tolerant cultivar was Zahir, where a combination of stomatal closure deregulation and a significant accumulation rate of stress-related metabolites were evident.3.
Rewatering after drought is beneficial to plants subjected to moderate drought stress, and selenium (Se) could increase the tolerance of plants to stressful environment. The role of Se in rewatering of drought-treated wheat seedlings (Triticum aestivum L., cv Hengmai5229) was studied. The objective was to elucidate whether Se could improve recovery of wheat seedlings at rewatering after drought stress. Drought stress induced a significant reduction in growth parameters, total chlorophyll and soluble protein contents, and increased the rate of superoxide radical (O 2 ·? ) production, MDA content, and the activities of peroxidase, catalase (CAT), and superoxide dismutase in wheat seedlings. Rewatering after drought did not significantly affect biomass accumulation of seedlings over drought treatment, although it decreased the rate of O 2 ·? production and MDA content. However, the combined treatment of rewatering and Se evidently promoted biomass accumulation of seedlings over drought treatment and rewatering alone; and the rate of O 2 ·? production, MDA content, soluble protein content and CAT activity were recovered to the control values. This indicates that Se improved recovery of wheat seedlings at rewatering after drought stress. 相似文献

Background and aims
Drought events, agricultural practices and plant communities influence microbial and soil abiotic parameters which can feedback to fodder production. This study aimed to determine which soil legacies influence plant biomass production and nutritional quality, and its resistance and recovery to extreme weather events.Methods
In a greenhouse experiment, soil legacy effects on Lolium perenne were examined, first under optimal conditions, and subsequently during and after drought. We used subalpine grassland soils previously cultivated for two years with grass communities of distinct functional composition, and subjected to combinations of climatic stress and simulated management.Results
The soil legacy of climatic stress increased biomass production of Lolium perenne and its resistance and recovery to a new drought. This beneficial effect resulted from higher nutrient availability in soils previously exposed to climatic stresses due to lower competitive abilities and resistance of microbial communities to a new drought. This negative effect on microbial communities was strongest in soils from previously cut and fertilized grasslands or dominated by conservative grasses.Conclusion
In subalpine grasslands more frequent climatic stresses could benefit fodder production in the short term, but threaten ecosystem functioning and the maintenance of traditional agricultural practices in the long term.5.

Alternative pathway (AP) is involved in the tolerance of highland barley seedlings to the low-nitrogen stress by dissipating excessive reducing equivalents generated by photosynthesis and maintaining the cellular redox homeostasis.Abstract
Low nitrogen (N) is a major limiting factor for plant growth and crop productivity. In this study, we investigated the roles of the alternative pathway (AP) in the tolerance of two barley seedlings, highland barley (Kunlun12) and barley (Ganpi6), to low-N stress. The results showed that the chlorophyll content and the fresh weight decreased more in Ganpi6 than those in Kunlun12 under low-N stress, suggesting that Kunlun12 has higher tolerance to low-N stress than Ganpi6. AP capacity was markedly induced by low-N stress; and it was higher in Kunlun12 than in Ganpi6. Comparatively, the cytochrome pathway capacity was not affected under all conditions. Western-blot analysis showed that the protein level of the alternative oxidase (AOX) increased under low-N stress in Kunlun12 but not in Ganpi6. Under low-N stress, the NAD(P)H content and the NAD(P)H to NAD(P)++NAD(P)H ratio in Ganpi6 increased more than those in Kunlun12. Furthermore, photosynthetic parameters (Fv/Fm, qP, ETR and Yield) decreased markedly and qN increased, indicating photoinhibition occurred in both barley seedlings, especially in Ganpi6. When AP was inhibited by salicylhydroxamic acid (SHAM), the NAD(P)H content and the NAD(P)H to NAD(P)++NAD(P)H ratio dramatically increased under all conditions, resulting in the marked accumulation of H2O2 and malondialdehyde in leaves of both barley seedlings. Meanwhile, the malate–oxaloacetate shuttle activity and the photosynthetic efficiency were further inhibited. Taken together, AP is involved in the tolerance of highland barley seedlings to low-N stress by dissipating excess reducing equivalents and maintaining the cellular redox homeostasis.6.

Woody tissue photosynthesis might play a key role in maintaining plant carbon economy and hydraulic function under unfavourable conditions such as drought stress.Abstract
Within trees, a portion of respired CO2 is assimilated by bark and woody tissue photosynthesis, but its physiological role remains unclear, in particular under unfavour able conditions like drought stress. We hypothesised that woody tissue photosynthesis will contribute to overall tree carbon gain both under sufficient water supply and during drought, and plays a role in maintaining the hydraulic function. We subjected half of the trees to a stem and branch light-exclusion treatment to prevent bark and woody tissue photosynthesis. Then, we measured leaf gas exchange and stem growth in Populus deltoides x nigra ‘Monviso’ trees both under well-watered and dry conditions. We additionally measured cavitation using acoustic emission in detached control and light-excluded branches to illustrate the role of woody tissue photosynthesis in xylem embolism repair. Under well-watered conditions, light exclusion resulted in reduced stem growth relative to control trees by 30 %. In response to drought, stem shrinkage of light-excluded trees was more pronounced as compared to control trees. During drought stress also maximum photosynthesis and transpiration rate tended to decrease more rapidly in light-excluded trees compared to control trees. Leaf fall in light-excluded branches together with the larger number of acoustic emissions in control branches indicates that in the latter more xylem vessels were still hydraulically functional under drought. Therefore, our study highlights that photosynthesis at branch and stem level might be a key factor in the resilience of trees to drought stress by maintaining both the plant carbon economy and hydraulic function.7.
Background
Maize seedlings are constantly exposed to inorganic phosphate (Pi)-limited environments. To understand how maize cope with low Pi (LP) and high Pi (HP) conditions, physiological and global proteomic analysis of QXN233 genotype were performed under the long-term Pi starvation and supplementation.Methods
We investigated the physiological response of QXN233 genotype to LP and HP conditions and detected the changes in ion fluxes by non-invasive micro-test technology and gene expression by quantitative real-time polymerase chain reaction. QXN233 was further assessed using vermiculite assay, and then proteins were isolated and identified by nano-liquid chromatography-mass spectrometry.Results
A negative relationship was observed between Na+ and Pi, and Na+ efflux was enhanced under HP condition. Furthermore, a total of 681 and 1374 were identified in the leaves and roots, respectively, which were mostly involved in metabolism, ion transport, and stress response. Importantly, several key Pi transporters were identified for breeding potential. Several ion transporters demonstrated an elaborate interplay between Pi and other ions, together contributing to the growth of QXN233 seedlings.Conclusion
The results from this study provide insights into the response of maize seedlings to long-term Pi exposure.8.

A higher mortality of dominant trees under drought stress is explained by impacts of tree size, canopy- and root structure and the hydraulic transport system.Abstract
Drought stress can trigger tree mortality but the impact depends on stress intensity (water demand and availability) and on the vulnerability of the individual. Therefore, most research focusses on the species-specific properties such as water use efficiency or hydraulic conductivity that determine vulnerability. At the ecosystem scale, however, tree properties that have been found important for drought sensitivity or resistance vary with individual size and resource availability within a forest—also within the same species. This is caused by different environmental conditions for each tree and hence different growth histories of individuals generating specific anatomical and physiological features. Individual drought stress sensitivity might thus be considerably different from stand scale sensitivity. Indeed, empirical evidence shows that drought stress impact depends on tree social position which can be defined in degrees of suppression but correlated to resource availability, stress sensitivity and stress exposure. In this review, we collect such evidence and discuss the role of microclimate and soil water distribution as well as anatomical and physiological adjustments, which might serve as foundation for better-adapted management strategies to mitigate drought stress impacts. Finally, we define model requirements aiming to capture stand-scale drought responses or management impacts related to drought stress mitigation.11.

The effects of soil water regime and wheat cultivar, differing in drought tolerance with respect to root respiration and grain
yield, were investigated in a greenhouse experiment. Two spring wheat (Triticum aestivum) cultivars, a drought sensitive (Longchun 8139-2) and drought tolerant (Dingxi 24) were grown in PVC tubes (120 cm in length and 10 cm in diameter) under an automatic rain-shelter. Plants were subjected
to three soil moisture regimes: (1) well-watered control (85% field water capacity, FWC); (2) moderate drought stress (50%
FWC) and (3) severe drought stress (30% FWC). The aim was to study the influence of root respiration on grain yield under
soil drying conditions. In the experiment, severe drought stress significantly (p < 0.05) reduced shoot and root biomass, photosynthesis and root respiration rate for both cultivars, but the extent of the
decreases was greater for Dingxi 24 compared to that for Longchun 8139-2. Compared with Dingxi 24, 0.04 and 0.07 mg glucose m−2 s−1 of additional energy, equivalent to 0.78 and 1.43 J m−2 s−1, was used for water absorption by Longchun 8139-2 under moderate and severe drought stress, respectively. Although the grain yield of both cultivars decreased with declining
soil moisture, loss was greater in Longchun 8139-2 than in Dingxi 24, especially under severe drought stress. The drought tolerance cultivar (Dingxi 24), had a higher biomass and metabolic activity under severe drought stress compared to the sensitive cultivar (Longchun 8139-2), which resulted in further limitation of grain yield. Results show that root respiration, carbohydrates allocation (root:shoot
ratio) and grain yield were closely related to soil water status and wheat cultivar. Reductions in root respiration and root
biomass under severe soil drying can improve drought tolerant wheat growth and physiological activity during soil drying and

Association mapping of drought-related traits in barley was used to increase the density of existing QTL maps without recreating mapping populations.Abstract
We used 109 spring barley genotypes exhibiting high or low drought tolerance to elucidate the associations between diversity array technology sequencing (DArTseq) and single nucleotide polymorphism (SNP) markers and various physiological parameters related to plant responses to drought conditions. The study was performed in controlled conditions (growth chambers), drought tolerance was phenotyped in the four-leaf seedlings. We identified 58 associations including 34 new markers (i.e., 16 DArTseq and 18 SNP markers). The results for three markers were consistent with the data obtained in an earlier traditional biparental QTL mapping study. The regions neighboring markers on linkage group 2H contained the highest number of significant marker–trait associations. Five markers related to the photosynthetic activity of photosystem II were detected on chromosome 4H. The lowest number of associations were observed for the sequences neighboring DArT markers on linkage group 6H. A chromosome 3H region related to water use efficiency and net photosynthesis rate in both biparental QTL, and association study, may be particularly valuable, as these parameters correspond to the ability of plants to remain highly productive under water deficit stress. Our findings confirm that association mapping can increase the density of existing QTL maps without recreating mapping populations.14.

Background
R-wave synchronised atrial pacing is an effective temporary pacing therapy in infants with postoperative junctional ectopic tachycardia. In the technique currently used, adverse short or long intervals between atrial pacing and ventricular sensing (AP–VS) may be observed during routine clinical practice.Objectives
The aim of the study was to analyse outcomes of R-wave synchronised atrial pacing and the relationship between maximum tracking rates and AP–VS intervals.Methods
Calculated AP–VS intervals were compared with those predicted by experienced pediatric cardiologist.Results
A maximum tracking rate (MTR) set 10 bpm higher than the heart rate (HR) may result in undesirable short AP–VS intervals (minimum 83 ms). A MTR set 20 bpm above the HR is the hemodynamically better choice (minimum 96 ms). Effects of either setting on the AP–VS interval could not be predicted by experienced observers. In our newly proposed technique the AP–VS interval approaches 95 ms for HR > 210 bpm and 130 ms for HR < 130 bpm. The progression is linear and decreases strictly (? 0.4 ms/bpm) between the two extreme levels.Conclusions
Adjusting the AP–VS interval in the currently used technique is complex and may imply unfavorable pacemaker settings. A new pacemaker design is advisable to allow direct control of the AP–VS interval.16.

Introducing genes from wild relatives is the best option to increase genetic diversity and discover new alleles necessary for wheat improvement. A population harboring genomic fragments from the diploid wheat progenitor Aegilops tauschii Coss. in the background of bread wheat (Triticum aestivum L.) was developed by crossing and backcrossing 43 synthetic wheat lines with the common wheat cultivar Norin 61. We named this population multiple synthetic derivatives (MSD). To validate the suitability of this population for wheat breeding and genetic studies, we randomly selected 400 MSD lines and genotyped them by using Diversity Array Technology sequencing markers. We scored black glume as a qualitative trait and heading time in two environments in Sudan as a quantitative trait. Our results showed high genetic diversity and less recombination which is expected from the nature of the population. Genome-wide association (GWA) analysis showed one QTL at the short arm of chromosome 1D different from those alleles reported previously indicating that black glume in the MSD population is controlled by new allele at the same locus. For heading time, from the two environments, GWA analysis revealed three QTLs on the short arms of chromosomes 2A, 2B and 2D and two on the long arms of chromosomes 5A and 5D. Using the MSD population, which represents the diversity of 43 Ae. tauschii accessions representing most of its natural habitat, QTLs or genes and desired phenotypes (such as drought, heat and salinity tolerance) could be identified and selected for utilization in wheat breeding.20.
