This paper explores the effects of preservation and taphonomy on the ultrastructure of recent and fossil (Quaternary and Neogene) Globigerinoides using scanning electron microscopy and thin section petrography. We show preservation states from: pristine (plankton tow) specimens that are “glassy”, have a microcrystalline test structure, and bear sharply defined interpore ridges with delicate spines; through core top and fossil specimens that have gametogenic calcite veneers of euhedral or lumpy deposits covering the interpore ridges and spine bases; to fossil material with extensive dissolution of the test wall and diagenetic calcite formed during shallow burial (less than 300 m below the sea floor). The latter produce a “grainy” texture to the test. Although we cannot unravel the precise effects of ecology and taphonomy on calcification temperature at every site, we show that for well-preserved fossil material with gametogenic calcite, temperature estimates are typically 2-3 °C cooler than modern sea surface, and are similar to those recorded using the δ18O of core top material from tropical latitudes. In contrast, at tropical sites with poor fossil preservation, estimates of calcification temperature are significantly cooler, sometimes by more than 10 °C than expected from present observations.  相似文献

The oxygen isotopic composition of plant cellulose is commonly used for the interpretations of climate, ecophysiology and dendrochronology in both modern and palaeoenvironments. Further applications of this analytical tool depends on our in-depth knowledge of the isotopic fractionations associated with the biochemical pathways leading to cellulose. Here, we test two important assumptions regarding isotopic effects resulting from the location of oxygen in the carbohydrate moiety and the biosynthetic pathway towards cellulose synthesis. We show that the oxygen isotopic fractionation of the oxygen attached to carbon 2 of the glucose moieties differs from the average fractionation of the oxygens attached to carbons 3-6 from cellulose by at least 9%, for cellulose synthesized within seedlings of two different species (Triticum aestivum L. and Ricinus communis L.). The fractionation for a given oxygen in cellulose synthesized by the Triticum seedlings, which have starch as their primary carbon source, is different than the corresponding fractionation in Ricinus seedlings, within which lipids are the primary carbon source. This observation shows that the biosynthetic pathway towards cellulose affects oxygen isotope partitioning, a fact heretofore undemonstrated. Our findings may explain the species-dependent variability in the overall oxygen isotope fractionation during cellulose synthesis, and may provide much-needed insight for palaeoclimate reconstruction using fossil cellulose.  相似文献

Common terminal sterilization methods are known to alter the natural structure and properties of soft tissues. One approach to providing safe grafts with preserved biological properties is the combination of a validated chemical sterilization process followed by an aseptic packaging process. This combination of processes is an accepted method for production of sterile healthcare products as described in ANSI/AAMI ST67:2011. This article describes the validation of the peracetic acid and ethanol-based (PAAE) chemical sterilization process for allograft dermal tissues at the Musculoskeletal Transplant Foundation (MTF, Edison, NJ). The sterilization capability of the PAAE solution used during routine production of aseptically processed dermal tissue forms was determined based on requirements of relevant ISO standards, ISO 14161:2009 and ISO 14937:2009. The resistance of spores of Bacillus subtilis, Clostridium sporogenes, Mycobacterium terrae, Pseudomonas aeruginosa, Enterococcus faecium, and Staphylococcus aureus to the chemical sterilization process employed by MTF was determined. Using a worst-case scenario testing strategy, the D value was calculated for the most resistant microorganism, Bacillus. The 12D time parameter determined the minimum time required to achieve a SAL of 10?6. Microbiological performance qualification demonstrated a complete kill of 106 spores at just a quarter of the full cycle time. The validation demonstrated that the PAAE sterilization process is robust, achieves sterilization of allograft dermal tissue to a SAL 10?6, and that in combination with aseptic processing secures the microbiological safety of allograft dermal tissue while avoiding structural and biochemical tissue damage previously observed with other sterilization methods such as ionizing irradiation.  相似文献

Objectives

Holocene hunter‐gatherers adapted to climatic and environmental changes over time. Carbon and nitrogen stable isotope analysis of human skeletal remains from the Inariyama shell mound of the Final Jomon period have revealed large dietary variations in the population. This study analyzed radiocarbon dates of these individuals to test temporal changes in diet and its relationship with tooth ablation.

Materials and Methods

Twenty‐nine human skeletal remains from Inariyama were included in this study. Extracted bone collagen samples were purified to graphite. Then, radiocarbon dating of these samples was performed using the accelerator mass spectrometer.

Results

The radiocarbon ages of Inariyama ranged about, 3,230–2,140 cal BP and showed three peaks of occupation. In the early and late phases, terrestrial resource consumption and incisor extraction were observed, while marine resource consumption and canine extraction were observed in the middle phase.

Discussion

These temporal changes of diet and tooth ablation types occurred in parallel with climatic cooling and environmental change and help reveal how Holocene hunter‐gatherers adapted to the changing environments.

Chemical components of callus and other tissues (leaf, root, and seed) of rice were investigated to find the variation in callus metabolism. The isolated compounds were squalene, three sterols, three triterpenes and a fatty alcohol; the last being present only in the callus. Cylindrin, isoarborinol and chlorophyll were not found in the callus. Fatty acids were detected in all tissues in varying amounts, the callus containing lauric acid in greater quantity than the leaves, roots, and seeds.  相似文献

We studied Holocene speleothems and tufa samples collected in numerous caves and rivers in the Dinaric Karst of Croatia, Slovenia, Bosnia and Herzegovina, as well as Serbia and Montenegro. Differences in the formation process of tufa and speleothems are discussed in the context of their isotopic composition (14C, 13C and 18O), as well as the chemistry of surface water (rivers, lakes) and drip water (in caves). The physical and chemical parameters monitored in the surface water (tufa precipitation) and drip water (speleothem precipitation) show that more stable conditions accompany speleothem rather than tufa formation. This is particularly obvious in the water temperature variations (2-22°C in surface water and 7-12°C in drip water) and in saturation index variation (3-11 in surface water and 1-6 in drip water). The range of 14C ages recorded by Holocene speleothems (∼12?000 yr) is wider by several thousands years than that of Holocene tufa samples (∼6000 yr). δ13C values for tufa samples range from −12‰ to −6‰ and for speleothem samples from −12‰ to +3‰ reflecting higher soil carbon and/or vegetation impact on the process of tufa than on speleothem formation. The differences in δ18O values of tufa and speleothem samples from different areas reflect different temperature conditions and differing isotopic composition in the water. The study shows that speleothems from the Dinaric Karst can be used as global palaeoclimatic records, whereas tufa records changes in the local palaeoenvironment.  相似文献

The brown coal mines of the Geiseltal in Germany were among the most important middle Eocene fossil localities and constitute the reference sites for the Geiseltalian stage of the European Land Mammal Mega Zones. Here, an updated review of the Geiseltal avifauna is given. Thirteen species are represented by diagnostic bones and can be referred to avian higher-level taxa, but various indeterminable albeit distinct fossils indicate a higher diversity of the Geiseltal avifauna. The majority of the Geiseltal fossils belong to taxa, which are well known from early to mid-Ypresian fossil sites of Europe and North America. Some of the species from the Geiseltal are larger than their early Eocene relatives, but it remains elusive whether this indicates an evolutionary size increase in some avian lineages or reflects different palaeohabitats of the involved fossil sites. The Geiseltal avifauna exhibits some taphonomic peculiarities that have not yet been addressed. Most bird fossils consist of isolated bones or partial skeletons and here it is hypothesized that at least some of the fragmentary specimens represent feeding remains of crocodilians. For the first time, medullary bone, which is indicative of breeding females, is reported for birds from the Geiseltal. The comparative abundance of the coliiform species Eoglaucidium pallas is remarkable, and either this species was extremely abundant in the Geiseltal ecosystem or it had ecological preferences, which favored its preservation.  相似文献

The concentrations of cucumber mosaic (CMV) and alfalfa mosaic viruses, present in various types of cultured plant tissues, were significantly less when ribavirin (syn. Virazole) at 50 or 100 mg/litre was present in the culture medium. CMV concentrations decreased within 24 days in infected cultures treated with ribabirin, but incubation periods of up to 127 days were required before the virus was undetectable. However, virus-free cultures were obtained from CMV-infected meristem-tips irrespective of the presence or absence of ribavirin. Kinetin at concentrations up to 25.6 mg/litre had no persistent antiviral effect.  相似文献

Planktonic foraminifera collected from a sediment trap deployed off Hainan in the northwestern South China Sea (SCS-NW) between July 2012 and April 2013 were studied to evaluate their seasonal variability and ecology as well as to infer the factors controlling their shell fluxes. The total planktonic foraminifera flux, as well as the fluxes of the dominant species (Globigerinoides ruber, Globigerinoides sacculifer and Neogloboquadrina dutertrei), showed three distinct maxima during SW-monsoon in August 2012, the SW-NE intermonsoon in October 2012 and the NE-monsoon in December 2012–February 2013. These periods were characterized by upwelling, aerosol fallout, and intense wind mixing, respectively, from which the foraminiferal assemblages benefitted, as indicated by the close correlation between wind speed, sea surface temperature (SST), chlorophyll a concentration (Chl-a), δ18O of G. ruber and the shell fluxes. The correlation also suggests that temperature and food availability might have been the primary drivers of the observed changes in foraminiferal abundance. The offset between the SST deduced from flux-weighted of G. ruber δ18O and annual mean SST is only ∼0.3 °C, much lower than ∼5.2 °C between the summer and winter temperature, indicating a balanced seasonality bias in the shell flux. The linear regression between the satellite-derived sea surface temperature and G. ruber δ18O reveals the strong potential of this species, at least in the studied region, as an ecological indicator for past oceanic environments.  相似文献

Biomineralization is the process by which metazoa form hard minerals for support, defense, and feeding. The minerals so formed, e.g., teeth, bones, shells, carapaces, and spicules, are of considerable interest to chemists and materials scientists. The cell biology underlying biomineralization is not well understood. The study of the formation of mineralized structures in developing organisms offers opportunities for understanding some intriguing aspects of cell and developmental biology. Five examples of biomineralization are presented: (1) the formation of siliceous spicules and frustules in sponges and diatoms, respectively; (2) the structure of skeletal spicules composed of amorphous calcium carbonate in some tunicates; (3) the secretion of the prism and nacre of some molluscan shells; (4) the development of skeletal spicules of sea urchin embryos; and (5) the formation of enamel of vertebrate teeth. Some speculations on the cellular and molecular mechanisms that support biomineralization, and their evolutionary origins, are discussed.  相似文献

We have measured the (14)C content of human femoral mid-shaft collagen to determine the dynamics of adult collagen turnover, using the sudden doubling and subsequent slow relaxation of global atmospheric (14)C content due to nuclear bomb testing in the 1960s and 1970s as a tracer. (14)C measurements were made on bone collagen from 67 individuals of both sexes who died in Australia in 1990-1993, spanning a range of ages at death from 40 to 97, and these measurements were compared with values predicted by an age-dependent turnover model. We found that the dataset could constrain models of collagen turnover, with the following outcomes: 1) Collagen turnover rate of females decreases, on average, from 4%/yr to 3%/yr from 20 to 80 years. Male collagen turnover rates average 1.5-3%/yr over the same period. 2) For both sexes the collagen turnover rate during adolescent growth is much higher (5-15%/yr at age 10-15 years), with males having a significantly higher turnover rate than have females, by up to a factor of 2. 3) Much of the variation in residual bomb (14)C in a person's bone can be attributed to individual variation in turnover rate, but of no more than about 30% of the average values for adults. 4) Human femoral bone collagen isotopically reflects an individual's diet over a much longer period of time than 10 years, including a substantial portion of collagen synthesised during adolescence.  相似文献

In 1982, Chisholm et al. used δ13C data for human burials from shell midden sites widely distributed on the coast of British Columbia (BC) to show the extreme dependence of these individuals on high trophic level marine consumers, principally salmon and marine mammals. Here, we present previously unpublished analyses of δ15N for some of the same individuals as well as δ13C data for additional individuals. Nitrogen isotope data show that the diet was dominated by high trophic level marine fauna including carnivorous fish and marine mammals. Although most burials were found in shell middens, marine mollusks made up of only a minor component of diet. The data for δ13C demonstrate that terrestrial faunal foods are undetectable in the diet of the majority of individuals, and seldom constitute more than 10% of the dietary protein of individuals living on the coast although terrestrial fauna were widely available as a potential source of protein. This dietary pattern of exclusion of land‐based animals from their diet persisted for almost 6,000 years along a wide expanse of coastline. In contrast, people from the BC interior (100 km or more from the coast) consumed a mixed diet of terrestrial and marine foods including spawning salmon. Although abnormal hard tissue mineralization is a recognized complication of cystic fibrosis (CF), the pathogenesis leading from the defective cystic fibrosis transmembrane conductance regulator (CFTR) protein is poorly understood. We hypothesized that CFTR plays a direct role in the mineralization of bone and teeth and tested the hypothesis using CF mouse models [CFTR(−) mice]. In vivo measurements by dual-emission X-ray absorpitometry (DEXA) indicated that bone mineral density (BMD) was reduced in CF mice as compared to gender-matched littermates. However, no change was evident after correction of BMD for the covariant of body weight. The latter finding was confirmed in isolated femurs and nasal bones by standard dry-ashing and instrumental neutron activation analysis (INAA). INAA of the continuously growing hypsodont incisor teeth from CFTR(−) mice revealed reduced Ca and normal P in the enamel layer—a finding consistent with changes in the deciduous teeth of CF children. Interestingly, enamel fluoride was increased in the CFTR(−) incisors and may associate with abnormal enamel crystallite formation. The iron content of the incisor enamel was reduced, explaining the loss of yellow pigmentation in CFTR(−) incisors. In contrast to the incisors, the mineral content of the slow-growing brachydont molar teeth was not different between CFTR(−) and CFTR(+) mice. It was concluded that CFTR does not play a direct role in the mineralization of bones or brachydont teeth in mice. Functional CFTR is apparently required for normal mineralization of the hypsodont incisors. However, multiple changes in the mineral composition of the CF incisors suggest an indirect role for CFTR, perhaps by maintaining a normal salivary environment for continuous tooth eruption. Vitamin B12 deficiency is a risk factor for bone disorders via mechanisms not fully understood. In this study, an increase in serum inorganic phosphorus (Pi) concentrations was associated with a vitamin B12 deficiency. Napi2a, a renal cotransporter for Pi reabsorption, accumulated on plasma membranes in a vitamin B12 deficiency suggests that vitamin B12 plays an important role in Pi homeostasis.  相似文献

The characterization of 66 kDa protein molecule, a major protein component which is produced from femoral-diaphyseal tissues with fracture healing (Igarashi and Yamaguchi [2002] Int. J. Mol. Med. 9:503-508), was investigated. Weaning rats were killed at 7 and 14 days after femoral fracture. When the femoral-diaphyseal tissues with fracture healing were cultured for 48 h in a serum-free medium, many proteins in the bone tissues were released into the medium. Analysis with sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) showed that a protein molecule of approximately 66 kDa was markedly increased in culture medium from bone tissues with fracture healing. N-terminal sequencing of 66 kDa protein indicated that its N-terminus was identical to that of rat albumin. Western blot analysis of medium 66 kDa protein showed expression of albumin. This expression was significantly enhanced by fracture healing. The expression of albumin was seen in the diaphyseal (cortical bone) and metaphyseal (trabecular bone) tissues of rat femur. When the femoral-diaphyseal tissues obtained at 7 days after femoral fracture were cultured in a serum-free medium containing either vehicle, parathyroid hormone (1-34) (10(-7) M), insulin-like growth factor-I (10(-8) M) or zinc acexamate (10(-4) M), medium albumin was significantly increased in the presence of those bone-stimulating factors. The addition of albumin (0.5 or 1.0 mg/ml of medium) caused a significant increase in calcium and deoxyribonucleic acid contents in the femoral-diaphyseal and -metaphyseal tissues obtained from normal rats in vitro. The present study demonstrates that fracture healing induces a remarkable production of albumin which is a major protein component produced from femoral-diaphyseal tissues of rats, and that albumin has an anabolic effect on bone components.  相似文献

Cryopreservation of human cells and tissue has generated great interest in the scientific community since 1949, when the cryoprotective activity of glycerol was discovered. Nowadays, it is possible to reach the optimal conditions for the cryopreservation of a homogeneous cell population or a one cell-layer tissue with the preservation of a high pourcentage of the initial cells. Success is attained when there is a high recovery rate of cell structures and tissue components after thawing. It is more delicate to obtain cryopreservation of composite tissues and much more a whole organ. The present work deals with fundamental principles of the cryobiology of biological structures, with special attention to the transfer of liquids between intra and extracellular compartments and the initiation of the formation and aggregation of ice during freezing. The consequences of various physical and chemical reactions on biological tissue are described for different cryoprotective agents. Finally, we report a review of results on cyropreservation of various tissues, on the one hand, and various organs, on the other. We also report immunomodulation of antigenic responses to cryopreserved cells and organs.  相似文献

A new technique is described for taphonomic investigation of fossil vertebrates with a high degree of skeletal articulation and completeness, and applied to analysis of the taphonomy of Archaeopteryx. The known skeletal remains of Archaeopteryx can be assigned to two preservational types: (A) well-articulated and almost complete skeletons, and (B) less complete and more disarticulated skeletons, but with some well-articulated sub-units. Differences between these categories are most likely a function of time elapsed between death and burial, and these groups are interpreted as samples of a larger possible range of taphonomic variation. The specimens represent parts of a decay spectrum rather than a decay sequence, and there is no evidence for a regional drift pattern. Digital crossover in the hands of Archaeopteryx , previously considered an anatomical condition, is interpreted as a post-mortem artefact.  相似文献

This study investigated stable‐isotope ratio evidence of weaning for the late Anglo‐Saxon population of Raunds Furnells, Northamptonshire, UK. δ15N and δ13C values in rib collagen were obtained for individuals of different ages to assess the weaning age of infants within the population. A peak in δ15N values at about 2‐year‐old, followed by a decline in δ15N values until age three, indicates a change in diet at that age. This change in nitrogen isotope ratios corresponds with the mortality profile from the site, as well as with archaeological and documentary evidence on attitudes towards juveniles in the Anglo‐Saxon period. The pattern of δ13C values was less clear. Comparison of the predicted age of weaning to published data from sites dating from the Iron Age to the 19th century in Britain reveals a pattern of changing weaning practices over time, with increasingly earlier commencement and shorter periods of complementary feeding in more recent periods. Such a change has implications for the interpretation of socioeconomic changes during this period of British history, since earlier weaning is associated with decreased birth spacing, and could thus have contributed to population growth.
