A sizable component of small peptide is probably an important store of amino acid in Calliphora and Drosophila. Quantitative amino acid analysis of the peptides in adult male Calliphora erythrocephala and Drosophila subobscura show that their composition is very similar. It is shown in Calliphora that the amount of peptide present in adults varies with age and feeding regime. Further, the peptide composition of the haemolymph indicates that the peptides are stored intracellularly. However, there is evidence which suggests that their hydrolysis takes place in the haemolymph, thus providing the whole fly with free amino acid and the haemolymph with osmotic stability.  相似文献

The amounts of nearly every amino acid in Calliphora remain unchanged inspite of stress. Thus the free amino acid pool is regulated. The amounts of free amino acid in the haemolymph account for only a small part of the total free amino acid in a fly and therefore most free amino acid is located and regulated intracellularly. A comparison of the rates of conversion and turnover of glycine-C14 in flies fed protein and sugar and in flies fed sugar alone indicates that in the absence of a dietary source of amino acid, not only is the rate of total turnover appreciably lower but the rates of conversion to other amino acids also change. With a dietary source of amino acid, the rates of incorporation into protein account for only a part of the total turnover of each amino acid. With a sufficiency of amino acid, substantial amounts are probably used as an energy source.  相似文献

Changes in the ‘free’ amino acids, betaine, and trimethylamine oxide during the development of the eggs of Balanus balanoides (L.) and B. balanus (L.) have been determined; the results are given in terms of μM/g dry wt, μM/g water, and μM/106 eggs. The amino acids are derived from the yolk proteins the net composition of which is known. Free amino acids are present in considerable quantity, as is commonly the case with crustacean tissue. Changes in the individual amino acids are discussed. B. balanus eggs contain large, and relatively constant, amounts of sarcosine; its function is unknown but large quantities are present in the more highly evolved cirripedes so far examined. A possible relation between betaine glycine, and sarcosine relative to choline metabolism is considered. Large amounts of taurine are present. There is a striking increase in β-alanine in the late stages of development; in B. balanoides it comes to be the most, and in B. balanus the third most common amino acid; its possible involvement in purine metabolism is considered. The relation between the amounts of the various entities in the eggs and in the bodies of the adult are examined.  相似文献

In hydrolysates of the eastern subterranean termite, Reticulitermes flavipes, the most abundant protein amino acids (μmoles) were glycine, alanine, and glutamic acid; the least abundant were methionine and histidine. Sawdust from both sound and Lenzites trabea-decayed sapwood blocks of sugar maple, loblolly pine, and slash pine was force-fed to termites. A diet of decayed rather than sound wood had little effect on protein amino acid composition of the termites; glycine content varied the most. In contrast, diet affected the free amino acid composition. Except for glutamic acid, the major protein amino acids of the termites were not the predominant free amino acids. Tyrosine and histidine were relatively more abundant as free than as protein amino acids. Greatest differences in protein amino acid compositions of sound and decayed wood were in contents of glycine, leucine, lysine, and arginine.  相似文献

Amino acid composition and fatty acid composition were determined on seed samples of a range of white lupin (Lupinus albus) cultivars and accessions grown in either of two environments.Variability between genotypes was found for lysine, arginine and glutamic acid content, but not for the concentrations of other amino acids. The deficiency in sulphurcontaining amino acids, typical of legume proteins, was evident, with methionine and cyst(e)ine totalling only 2.2% of the protein. Variability was limited, indicating that improvement by breeding would be impracticable. Lupinus albus differed slightly from other lupin species in amino acid composition, having higher levels of threonine, tyrosine and isoleucine, but a lower level of glutamic acid than both L. angustifolius and L. luteus. Four low-alkaloid lines of L albus each had higher lysine content than the high-alkaloid line, but ‘Kiev Mutant’, despite earlier claims, had a lysine level no higher than the other three low-alkaloid lines.Fatty acid composition of the seed oil varied considerably between genotypes. Oleic acid ranged from 43.6 to 54.4% and linolenic acid from 6.7 to 15.2%, these two fatty acids being negatively correlated at one site. Linoleic acid content varied between 17.2 and 26.9% and was not correlated with other fatty acids. Total oil content averaged 9.6% with little variability between lines.It is concluded that, relative to other lupin species, L. albus has a more favourable amino acid profile for its utilisation in cereal-based diets for animals, particularly if the energy source is wheat, which is deficient in threonine. The higher oil content would be an important energy benefit to such diets and may allow their protein/energy balance to be maintained at higher levels of incorporation of L. albus seed meal than is possible with other lupin species.  相似文献

The concentration of free amino acids, total soluble protein, and haemoglobin in the haemolymph of fourth instar Chironomus tentans was investigated.The concentration of the free amino acid pool increases between the early (15.7 mM/l) and mid-(33.9 mM/l) fourth larval stages followed by a decline during the late (16.9 mM/l) fourth larval period. Alanine, serine, and the amides of aspartic acid and glutamic acid are the predominant free amino acids at all stages. Physiological fluid analysis of late fourth instar haemolymph detected 32 ninhydrin positive components including 18 common amino acids plus homoarginine, ornithine, citrulline, β-alanine, α-aminoadipic acid, α-aminoisobutyric acid, and sarcosine.The concentration of total soluble protein steadily increases during fourth instar larval development to a maximum of 9.3 g100 ml followed by a decline during the pharate pupal period. A similar pattern of variation occurs in haemoglobin content which comprises from 51 to 66% of Chironomus tentans haemolymph protein.The mM percentage of individual amino acids of total haemolymph protein varies little during the fourth instar. At all stages alanine and aspartic acid are the predominant amino acids.  相似文献

In Phycomyces blakesleeanus the total level of free amino acids and their relative distributions change strikingly during the development of sporangiophores. Very high levels of serine, glutamate and histidine are found. The variety of amino acids becomes more restricted in sporangiophores grown on nitrogen-rich media; levels of arginine increase dramatically. The distribution differs in mycelia. The utilization of the blood meal by mosquitoes was investigated by first feeding females quantities of blood ranging from 1 to 5 mg, and then analyzing the faeces for the various by-products of protein catabolism that were subsequently eliminated. The nitrogeneous waste products in order of importance were uric acid, histidine, ammonia and arginine. Only traces of the other amino acids were excreted.The total amount of each faecal substance varied linearly with the quantity of blood ingested, however their relative proportions did not change. Regardless of blood meal size the quantily of uric acid and ammonia produced indicates that about 80% of the non-histidine and arginine amino acids are deaminated and utilized for metabolic purposes other than egg protein synthesis.Most of the histidine and about one half of the arginine content of the blood were excreted as free amino acids, but the other amino acids were lost in trace amounts.Nineteen per cent of the total ingested amino acids was incorporated into soluble yolk proteins and this proportion was constant even for small blood meals that result in a reduction in the numbers of eggs produced.The comparative aspects of nitrogen partitioning and blood meal utilization by haematophagous insects, as well as the factors that affect blood meal utilization and fecundity in A. aegypti are discussed.  相似文献

l-Cystathionine and l-selenocystathionine have been isolated from the foliage of Astragalus pectinatus. In addition to these two amino acids, some S-methylcysteine and trace amounts of Se-methyl-selenocysteine were also detected in the foliage extracts. The seeds of A pectinatus were found to contain significant amounts of all four of these amino acids plus the γ-glutamyl peptides of S-methylcysteine and Se-methylselenocysteine.  相似文献

The amino acids of four species of Microsporidia parasitizing crustaceans were investigated: three species — Thelohania maenadis, Ormieresia carcini and Ameson pulvis — are parasites of Carcinus mediterraneus; Inodosporus sp. is a parasite of Palaemon serratus. Seventeen protein amino acids were identified of which aspartic acid, glutamic acid and lysine were quantitatively the most important, being quantitatively similar in all four parasites. The relative amounts of the next most abundant amino acids were found to vary and might serve as a systematic criterion at the genus level. For example, phenylalanine is predominant in Ormieresia and leucine in Inodosporus: serine in Thelohania and glycine in Ameson are also predominant, but to a lesser extent. The free amino acids composition shows little qualitative variation among the four genera, but quantitative differences are found in the composition of Microsporidia parasitizing the same host species; this may reflect variations in the amino acid metabolism of the parasite. The urea cycle in Ormeresia is most remarkable in this connection. The free amino acid level in the parasites was generally found to be in inverse proportion to the level in the host; the amino acids which are found to exist at high levels in the parasites correspond to essential amino acids of the Crustacea. Monoterpenoid composition of samples from 10 populations of Satureja douglasii in a 250-ha area were analyzed in Spring 1974. Two distinct compositional types were found: Type A contained large amounts of limonene and carvone but very small quantites of piperitenone, piperitone, pulegone and isomenthone (3-oxy compounds), whereas Type B was essentially the reverse. This variation is attributed to a genotypic difference. The amounts of 3-oxy compounds varied greatly among the Type B populations. Increasing shade conditions were correlated with increased amounts of the more reduced compounds isomenthone and piperitone. Clonal lines were established from two Type B populations whose habitats represented extremes of sun and shade conditions. The two clonal lines, which had very different compositions in the field, produced similar oils under controlled conditions. Growth under different temperature regimes had little effect on composition. Developmental changes in composition implied the precursor-product relationships piperitenone→piperitone and pulegone→isomenthone. The data suggest that S douglasii monoterpenoid compositional patterns are significantly influenced by genetic and environmental factors (probably light intensity) as well as developmental processes.  相似文献

Under accelerated ageing at high relative humidity and high temperature for 4 days germination and membrane permeability remained unaffected both in sunflower and chick pea seeds. However, the steryl glycoside concentration in the pooled leachate increased progressively with ageing. Total sterols, as well as steryl glycosides and free sterols of the seeds, increased with a concomitant decline in steryl esters under accelerated ageing. Pretreatment with the sterol biosynthesis inhibitor SK & F 7997A3 prevented the increase of total sterols under accelerated ageing conditions but there were increases in the amounts of steryl glycosides and free sterols and a decrease in steryl ester after such treatment, therefore, indicating interconversions of the various sterol types. Accelerated ageing also caused increases in free amino acids and soluble carbohydrate. Low relative humidity-high temperature and high relative humidity-low temperature failed to produce such effects.  相似文献

Staphylococcin 1580 increased the relative amount of diphosphatidylglycerol and decreased the amount of phosphatidylglycerol in cells of Staphlococcus aureus, while the amounts of lysylphosphatidylglycerol, phosphatidic acid and total phospholipid remained constant.Treatment of cells of Escherichia coli and S. aureus with colicin A and staphylococcin 1580, respectively, did not affect proton impermeability but subsequent addition of carbonylcyanide-m-chlorophenylhydrazone resulted in a rapid influx of protons into the cells.Bacteriocin-resistant and -tolerant mutants of E. coli and S. aureus were isolated. The bacteriocins caused leakage of amino acids preaccumulated into membrane vesicles of resistant mutants and had no significant effect on membrane vesicles of tolerant mutants.The uptake of amino acids into membrane vesicles was inhibited by both bacteriocins, irrespective of the electron donors applied. The bacteriocin inhibition was noncompetitive. The bacteriocins did not affect oxygen consumption and dehydrogenases in membrane vesicles.Both bacteriocins suppressed the decrease in the fluorescence of 1-anilino-8-naphthalene sulfonate caused by d-lactate or α-glycerol phosphate when added to membrane vesicles.It is concluded that the bacteriocins uncouple the transport function from the electron transport system.  相似文献

We report differences in the length of a specific region of the circadian clock gene period (per) that correlate with different capacities for pupal diapause in the flesh fly, Sarcophaga bullata. The conspicuous difference is located in a region we refer to as the putative C-terminal photoperiodic (CP) region. The length of the CP region correlates inversely with the incidence of diapause. A deletion of 33 amino acids in this region correlates with a significant increase in the incidence of diapause, from 78.1% to 93.0%, and an insertion of 9 amino acids in the same area correlates with a drop in the diapause incidence to 4.0%. This correlation suggests a possible functional role for this region of per in photoperiodism.  相似文献

A qualitative and quantitative analysis of free amino acids and related compounds in the haemolymph of Rhynchosciara americana was carried out for different periods of the fourth larval instar. Threonine, serine, proline, and glutamic acid make up 50 per cent of the total free amino acids in R. americana haemolymph just before the larvae start spinning the communal cocoon; after this the titre of most of the amino acids declines continuously. There are few peptides but these are present in high titres; they consist of two to three amino acid residues, of which the most important are histidine and aspartic acid. The fall in the haemolymph amino acid and peptide titres is insufficient to account for the silk protein which accumulates on the communal cocoon during the same period. The results are consistent with a silk protein origin from haemolymph proteins and haemolymph free amino acids. The origin and metabolic rôle of some haemolymph ninhydrin-positive compounds are discussed.  相似文献

The two diastereoisomers, (2S,4R)-4-hydroxyglutamic acid and (2S,4S)-4-hydroxyglutamic acid, occur in characteristic concentration ratios in Phlox species. The second of these compounds is the principal free amino acid in the green parts of the plants. The presence of (2S,4R)-4-hydroxyglutamic acid in plants is reported for the first tiine. No other 4-substituted acidic amino acids were detected in the Phlox species analysed, although special attention was paid to the possible presence of 4-hydroxy-4-methylglutamic acids which have previously been reported in plants. It was found, however, that both diastereoisomers of (2S)-4-hydroxy-4-methylglutamic acid co-exist in Ledenbergia roseoaenea and also in Pandanus veitchii. Although the presence of 3,4-dihydroxyglutamic acids in green parts of Lepidium sativum and Rheum rhaponticum has been previously reported, we were not able to detect or isolate any of the possible diastereoisomers from the green parts or seeds of these plants. We did isolate glutathione which was found to have some properties in common with those reported for the dihydroxy compounds.  相似文献

The amounts of sugar solution consumed by Drosophila melanogaster flies were determined. Starved and desiccated flies of a wild type strain (QA) consume 7?9 × 10?2 λ of a 0.3 M sucrose solution per fly during the first hour and less later. They consume more of the 0.3 M sucrose solution than of the more diluted and the more concentrated solutions. In preference-aversion tests the flies discriminated between water and various sugar solutions, and between different sugar concentrations. Contrary to other fly species these flies did not prefer 0.05 M fructose over 0.05 M glucose. 0.3–0.5 M NaCl added to 0.1 M sucrose turned a preference over 0.01 sucrose into an aversion. A mutant, Lot-94, selected for its increased consumption of a 1 M NaCl solution was found to consume more of all test solutions. The amount of NaCl that had to be added to 0.1 M sucrose to turn the preference over 0.01 M sucrose by the mutant flies into aversion was not different from that found for the wild type flies.  相似文献

The media, in which a butterfly cell line (Px 58), derived from pharate adult ovaries of Papilio xuthus cultured for 8 days, were analysed to examine the changes in free amino acids in the medium during cultivation. Beta-alanine, arginine, glycine, histidine, lysine, phenylalanine, proline, serine, and tryptophan did not change markedly. Asparagine, aspartic acid, cystine, glutamine, isoleucine, leucine, methionine, threonine, tyrosine, and valine decreased to some extent with culturing. Alpha-alanine increased markedly, and glutamic acid did so to a lesser extent. Requirements of amino acids by the cell line were examined by deleting amino acids one at a time. Deletion of alpha-alanine, beta-alanine, asparagine, glutamic acid, glycine, and phenylalanine did not cause deterioration of the cell. These amino acids were thought to be non-essential or required only a little. Deletion of other amino acids impaired the cell growth severely. These amino acids would appear to be essential for growth of the Px 58 cell line.  相似文献
