The esteroproteolytic enzyme, a serine protease from porcine pancreas, has been crystallized and characterized by X-ray diffraction. Two closely related crystal forms were observed. The unit cell dimensions of the first form are a = 59.2 A?, b = 96.4 A? and c = 47.4 A?, space group P21212 with one molecule (mol. wt 30,000) per asymmetric unit. The unit cell dimensions of the second form are a = 59.2 A?, b = 96.4 A? and c = 94.8 A?, space group P212121. Mercury derivatives of enzyme inhibitors were used to obtain multiple isomorphous heavy-atom substituents suitable for phase determination.  相似文献

Tonin is an enzyme isolated from the submaxillary glands of old rats. This enzyme is involved in the direct conversion of angiotensinogen or the tetradecapeptide renin substrate to the octapeptide angiotensin II, the agent implicated in hypertension. Large well-formed crystals of tonin were grown from 2.5 m-(NH4)2SO4, 0.01 m-phosphate buffer (pH 6.2) and 1% methylpentanediol. The crystals are trigonal, space group P3121 (P3221) with unit cell dimensions: a = b = 94 A?, c = 66 A?, γ = 120°. The value of Vm = 2.6 A?3/dalton corresponds to one molecule of molecular weight 32,000 per asymmetric unit. Definitive classification of tonin into one of the four proteolytic enzyme classes (carboxyl protease, serine protease, thiol protease or metalloprotease) has not been firmly established.  相似文献

Crystals of N- and C-terminal fragments of elongation factor G (EF-G) from Escherichia coli have been grown from the preparations obtained by limited tryptic hydrolysis. Molecular masses of these fragments are equal to about 49,000 and 25,000, respectively. In the form of an additive complex they appear to be a major part of the native spatial structure of EF-G (Alakhov et al., 1979). Crystals of N-terminal fragment belong to space group P41212 or P43212 with unit cell dimensions a = b = 76.6 A? and c = 191.6 A?. Crystals of C-terminal fragment belong to space group P4122 or P4322 with unit cell dimensions a = b = 77.1 A? and c = 75.0 A?. In both cases the assumed number of protein molecules per asymmetric part of the unit cell is one.  相似文献

Crystals of plastocyanins from pea and corn leaves have been obtained. Both are suitable for X-ray structure analysis with a resolution up to 1.8 Å. The crystal form of plastocyanin from pea leaves belongs to the space group P212121 with unit cell dimensions: a = 49.0 A?, b = 53.3 A?, c = 82.6 A?. The assumed number of protein molecules per asymmetric unit of the unit cell is two. Crystals of the oxidized (Cu2+) and reduced (Cu+) forms are isomorphic. No essential differences in spot intensities for the main zone with a resolution of 3 Å were revealed. The crystal form of plastocyanin from corn leaves belongs to the space group P1 with unit cell parameters: a = 24.8 A?, b = 30.0 A?, c = 58.5 A? and α = 96° 10′, β = 87°08′, γ = 78°40′. A toxic protein from the venom of the scorpion Centruroides sculpturatus has been crystallized. The crystals are orthorhombic, with unit cell dimensions a = 52.2 A?, b = 42.0 A? and c = 28.5 A?, space group P212121. Three new crystal forms of cow α-lactalbumin are described. A trigonal form in space group P3121 or P3221 has unit cell dimensions: a = b = 57.4 A?, c = 75.0 A?. A hexagonal form in space group P622 has unit cell dimensions: a = b = 94.0 A?, c = 67.1 A?. A second trigonal form grown in the presence of calcium ions belongs to space group P321 with unit cell dimensions: a = b = 93.7 A?, c = 66.9 A?. The structural protein, δ-crystallin, has been purified and crystallized from adult turkey lens. The crystals are orthorhombic and normally belong to space group P21212 with unit cell dimensions a = 99.9(2) A?, b = 133.4(3) A? and c = 69.1(2) A?. This corresponds to two molecules of molecular weight approximately 200,000 per unit cell. A second crystal form has also been found in which b and c increase to 135.4(3) Å and 140.0(3) Å. respectively, indicating four molecules per unit cell.  相似文献

A lectin from the fava bean (Vicia faba) has been purified and crystallized in a form suitable for high-resolution crystallographic structure analysis. This protein binds glucose- and mannose-like saccharides, and it is mitogenic for lymphocytes. The fava lectin crystallizes in the orthorhombic space group. P212121 with unit cell dimensions a = 90.0, b = 89.3, and c = 67.4 A?. The mass of protein in the asymmetric unit is 53,000 daltons, corresponding to the molecular weight of the protein in solution.  相似文献

A complex of ribosomal proteins B-L8 and B-L13 from Bacillus stearothermophilus has been isolated and crystallized. Levansucrase, an exocellular enzyme, has been isolated from a high producer mutant, the BS5C4 constitutive strain, of Bacillus subtilis. Three crystalline forms have been obtained, all three belonging to the orthorhombic space group P212121. The most suitable form for a three-dimensional structure investigation has cell dimensions, a = 68 A?, b = 125 A?, c = 54 A?. There is one molecule in the asymmetric unit.  相似文献

Large single crystals of ω-amino acid: pyruvate aminotransferase, were prepared by dialysis of the enzyme solution against 2.2 m-ammonium sulphate solution at pH 7.8. X-ray diffraction patterns show that the crystals belong to the orthorhombic space group I222 or I212121 with unit cell dimensions a = 124.1 A?, b = 137.9 A?, and c = 61.2 A?. The dd-carboxypeptidase-endopeptidase of low penicillin sensitivity that is excreted by Streptomyces albus G has been crystallized from a polyethylene glycol (Mr 6000 to 7500) solution at pH 8.0. X-ray examination of the prismatic crystals shows that the space group is P21 with unit cell dimensions a = 51.1 A?, b = 49.7 A?, c = 38.7 A?, β = 100.6 ° and one molecule in the asymmetric unit. A crystal suspension made in 50 mm-Tris · HCl buffer (pH 8.0) supplemented with 5 mm-MgCl2 and 16% (wv) polyethylene glycol exhibits enzyme activity on the substrate Ac2-l-Lys-d-Ala-d-Ala.  相似文献
