Multilocus genotypes using nine DNA microsatellite loci provided an effective and permanent individual identification mark for lemon sharks Negaprion brevirostris , the first example of genetic tagging for any elasmobranch species. The double-tagging provided by micro-satellites afforded a means to determine passive integrated transponder (PIT) tag shed rate in lemon sharks. The effects of passive integrated transponder (PIT) tagging on the growth and survival of kelp grouper Epinephelus bruneus was examined over 1 year. The overall average mass gain for the 1 year period of the abdominal cavity tagged group was higher than the dorsal muscle tagged group. The effects of passive integrated transponder (PIT) tagging on cortisol release, standard metabolic rate (SMR) and daily specific growth rate (GS) were evaluated in the Gulf killifish, Fundulus grandis, a small estuarine fish native to the Gulf of Mexico. Cortisol release by individual fish was measured non-invasively prior to PIT tagging, immediately after tagging and once per week for 1 month following tagging. Within the first 2 h of tagging, cortisol release rates were significantly elevated compared with values measured prior to tagging and significantly higher than that of fish handled identically except not implanted with PIT tags. By 1 week after PIT tagging, cortisol release rates returned to control levels. SMR, determined by intermittent-flow respirometry and GS, defined as per cent change in body mass per day, were measured prior to PIT tagging and weekly for 1 month after tagging. Neither SMR nor GS was significantly different in tagged v. untagged fish for the duration of the study. One month after tagging, haematocrit, plasma cortisol, blood glucose and blood lactate did not differ between tagged and untagged individuals. Therefore, after a transient stress response that subsides within 1 week, PIT tagging had no significant effects on these physiological variables in F. grandis, validating its use as a method of marking this and other small fishes. 相似文献

Gut contents of shovelnose and pallid sturgeon from the lower and middle Mississippi River were obtained by colonic flushing, a safe and easily implemented alternative to gastric lavage. Diets of both species were dominated numerically by immature Trichoptera, Ephemeroptera, and Diptera. Primary prey, based on volume, for shovelnose sturgeon were Trichoptera, and for pallid sturgeon were various fishes. Geographic and seasonal nuances in diet were observed for both species, but the general dichotomy of shovelnose sturgeon as browser on invertebrates and pallid sturgeon as predator on fishes did not change. Data indicate that both species require hard substrates for feeding. Data demonstrate that colonic flushing is an effective technique for describing diet and inferring ecological and behavioral information about sturgeon. 相似文献

Suitability of surgically implanted 8-mm passive integrated transponder tags for small-bodied fishes

Innovative tools that benefit conservation are critical as freshwater fishes are lost at unprecedented rates. Mark-recapture methods can characterize population demographics and life-history traits of diverse fishes, but suitable techniques for tagging for individual recognition of small-bodied fishes are rare. Passive integrated transponder (PIT) tag technology may facilitate the tagging of small fishes and early life stages of larger species. However, relatively little research has evaluated the suitability of these small (8.4 × 1.4 mm) tags for many groups of small fishes. Tag loss (retention and survival) and growth of individuals implanted with PIT tags relative to control and sham treatments were compared for eight fishes of differing morphologies. Additionally, the utility of cyanoacrylate to improve tag retention was tested on a subset of taxa. Fish of each species were equally divided and randomly assigned to one of three treatment groups (handled [control], surgical incision [sham], or surgical incision and PIT tag implantation [PIT]). During the 42-d study period, mortalities and expelled tags were counted daily and growth was measured weekly. Researchers can generally expect little tag loss and uncompromised growth rates for a variety of small-bodied fishes; however, initial fish length was related to the magnitude of physiological effects for some taxa (i.e., blacknose dace Rhinichthys atratulus, johnny darter Etheostoma nigrum, juvenile white sucker Catostomus commersonii). Relatively poor survival (<80%) was observed for two benthic species: johnny darter and blacknose dace, often when incision wounds became inflamed before healing. Prevalent tag loss for johnny darter, tadpole madtom Noturus gyrinus, and a dorsal-laterally compressed cyprinid can be reduced by closing the wounds with cyanoacrylate, but with substantially increased mortality rates. This research demonstrates the broad applicability of PIT tagging technology for ecological studies of small-bodied fishes and alleviates many concerns when surgically implanting tags into several fishes. 相似文献

Passive integrated transponder (PIT) tags allow a range of individual‐level data to be collected passively and have become a commonly used technology in many avian studies. Although the potential adverse effects of PIT tags have been evaluated in several species, explicit investigations of their impacts on small (<12 g) birds are limited. This is important, because it is reasonable to expect that smaller birds could be impacted more strongly by application of PIT tags. In this study, we individually marked Black‐capped Chickadees (Poecile atricapillus), a small (circa 10 g) passerine, at the University of Alberta Botanic Garden to evaluate potential lethal and sublethal effects of two PIT tagging methods: attachment to leg bands or subcutaneous implantation. We used a Cox proportional hazards model to compare the apparent survival of chickadees with leg band (N = 79) and implanted PIT tags (N = 77) compared with control birds that received no PIT tags (N = 76) over the subsequent 2 years based on mist net recaptures. We used radio‐frequency identification (RFID) redetections of leg band PIT tags to evaluate sex‐specific survival and increase the accuracy of our survival estimates. We also used a generalized linear regression model to compare the body condition of birds recaptured after overwintering with leg band PIT tags, implanted PIT tags, or neither. Our analysis found no evidence for adverse effects of either PIT tagging method on survival or body condition. While we recommend carefully monitoring study animals and evaluating the efficacy of different PIT tagging methods, we have shown that both leg band and subcutaneously implanted PIT tags ethical means of obtaining individualized information in a small passerine. 相似文献

Individual identification of organisms is a crucial part of assessing the processes that influence small-scale distribution patterns and the maintenance of social organizations. This study evaluates the use of passive integrated transponder (PIT) tags to quantify small-scale space use in a marine damselfish, Pomacentrus amboinensis (Pomacentridae). Implanting small PIT tags in the body cavity of fish down to 5.2 g with a hypodermic needle had no adverse effect on mortality or growth over a 47-day tank study. A 63-day field study also showed that tags had no influence on body condition as measured by Fultons condition factor (K), gonadosomatic indices, or plasma cortisol levels (a physiological indicator of stress). The utility of PIT-tag technology is illustrated by preliminary information from a study on the diel periodicity of visitations to male nest sites by female P. amboinensis. Information suggests that there is a strong periodicity in visitations. Females first entered the nest site at 03:20 h, with 88% of the total of 4 h 45 min that females spent within a single nest occurring prior to dawn. Despite this, 81% of the total visitations to the nest occurred after dawn, with most being very brief, averaging 17 s. No one female monopolized access to the nest, with nine females accounting for 90% of the time that females spent within the nest site. This methodology will be particularly useful in the study of activity rhythms, patch dynamics, and social interactions in a wide range of marine organisms.Communicated by: Ecological Editor P.F. ABSTRACT. Double-observer methodology requires independent collection of data to accurately estimate population parameters. Use of visual nest markers to facilitate matching, relocating, and monitoring nests as part of a double-observer study violates this assumption, but few reliable alternatives exist, especially when working with cryptic nests and high nest densities in homogeneous habitat. We used passive integrated transponder (PIT) tags to nonvisually mark the nests of ground-nesting birds at the Yukon Delta National Wildlife Refuge in western Alaska in a double-observer study of nest density. We marked 70 nests with PIT tags and naïve observers subsequently detected tags at 44 of 50 re-scanned nests (88% correct identification). Failed detections were likely due to either suboptimal tag orientation or tags falling through nest material, and such failures may be an inherent, but uncommon, feature of this nest-marking technique. PIT tags facilitated nest monitoring among independent observers, uniquely and reliably marked nests, provided a minimum of cues to potential nest predators, and allowed us to estimate densities in a double-observer framework while not violating assumptions. These tags should be useful in other studies of nesting birds where nonvisual, reliable nest markers are needed, and they provide a new tool for double-observer studies. 相似文献

Trotlines were used to capture pallid sturgeon in the free‐flowing Mississippi River, which extends from the Gulf of Mexico to the mouth of the Missouri River. Trotlines were baited with worms, and set overnight usually along the channel border. The pectoral fin rays of 165 pallid sturgeon caught in the Mississippi River were aged; 118 were from the lower Mississippi River (LMR) between the Gulf and mouth of the Ohio River, and 47 were from the middle Mississippi River (MMR) between the mouths of the Ohio and Missouri rivers. Initial agreement within ±1 year between two readers ranged from 53% for the LMR specimens, which were read first, to 84% for the MMR. Final age was agreed upon by both readers. For LMR pallid sturgeon, final age estimates ranged from 3 to 21 years with a mean (±SD) of 11.0 ± 4.7. For MMR pallid sturgeon, final age estimates ranged from 5 to 14 years with a mean of 9.5 ± 2.1. Seven pallid sturgeon marked with coded wire tags (CWT), indicating hatchery origin, were collected in the MMR. Age estimates for CWT fish were 7–8 years representing 1997 stocked fish, and 11–12 years representing 1992 progeny stocked in 1994. Von Bertalanffy growth equations for length indicated that pallid sturgeon in the MMR had higher growth rates for a given age than pallid sturgeon in the LMR. However, there were no significant differences (anova , P > 0.5) in the length–weight relationships between reaches. In the LMR, pallid sturgeon fully recruited to trotlines at age 11 and instantaneous total mortality (Z; slope of catch curve) was estimated at −0.12 (n = 10 year classes, r2 = 0.55, P = 0.01). Of the 118 sectioned rays from the LMR, 28 could not be reliably aged (only one section from the MMR could not be aged). Therefore, age was predicted from length using the von Bertalanffy equation. The catch curve was re‐calculated using the predicted ages of the 28 pallid sturgeon in the LMR resulting in Z = −0.07. In the MMR, pallid sturgeon fully recruited to trotlines at age 9 and Z was estimated at −0.36 (n = 6 year classes, r2 = 0.67, P = 0.04), which was significantly higher (anova , P = 0.04) than the LMR estimate. Higher mortality in the MMR may be due to habitat limitations compared to a larger, more diverse channel in the LMR, and incidental take of larger, older individuals during commercial harvesting of shovelnose sturgeon. Commercial take of shovelnose does not occur in the LMR except in the northern portion of the reach. Considering the presence of pallid sturgeon with CWT, recruitment of older individuals in the MMR may have been influenced by stocking a decade earlier. Management strategies for this endangered species should consider the differences in mortality rates among reaches, the impacts of commercial fishing on recovery of pallid sturgeon in the MMR, and the long‐term effects of hatchery fish now recruiting into the free‐flowing Mississippi River. 相似文献

Ultrasonic telemetry is a preferred method for fish-movement studies. Despite surgical tag implantation being the most common method for affixing tags, many studies lack tests addressing the assumption that tagging has no effect on fish performance or survival. The threatened, anadromous green sturgeon, Acipenser medirostris, has little documentation concerning its movements. We evaluated the effects of surgery and tag implantation in juveniles. We compared three groups: tagged fish with dummy transmitters implanted in the peritoneal cavity, sham fish that underwent surgery without tag implantation, and control fish that were handled and anesthetized but did not undergo surgery. We found no differences in growth or critical swimming velocity among groups. Photos of incisions were taken towards the beginning and at the end of the study to assess inflammation and to score each incision for closure and suture retention. Inflammation declined similarly for tagged and sham fish during the study. Ucrit was not related to the extent of inflammation or to post-surgery time. All fish showed healing during the study (ca. 140 day duration) and 10 % of tagged and sham fish showed signs of inflammation by the study end. These results suggest that current ultrasonic surgical tagging methods do not significantly affect the short-term growth or swimming performance of juvenile green sturgeon. Additionally, effects of surgery can be mitigated by minimizing the number of suture entry points and by using rapid-absorbing sutures. 相似文献

We tested the feeding behaviour of small European perch (Perca fluviatilis) in a laboratory study during the first 24 h after handling and 23 mm passive integrated transponder (PIT) tag implantation. Feeding commenced almost immediately following tagging and overall feeding patterns were unaffected by tagging. However, untagged perch had more feeding events than PIT-tagged individuals. This discrepancy could be attributed to post-tagging effects or/and reduced room for food due to the presence of the tag in the body cavity. 相似文献

In the summer and fall of 2014, Nebraska Game and Parks Commission crews monitored juvenile pallid sturgeon, Scaphirhynchus albus (Forbes & Richardson, 1905) (age‐1 and age‐4) implanted with telemetry tags that were stocked in a side channel of the Missouri River in Nebraska, USA to gain knowledge into post‐stocking survival and dispersal. For this study, specific questions were asked: (i) what is the short‐term survival of stocked pallid sturgeon, (ii) do pallid sturgeon stocked in summer exhibit decreased survival rates due to increased water temperatures or high river discharge, and (iii) how quickly do pallid sturgeon disperse from a stocking site? Detection histories for two rounds were used to estimate apparent survival (Ø) and detection (p) rates using maximum likelihood estimators based on the standard mark‐recapture Cormack‐Jolly‐Seber model structure within program MARK. Overall apparent survival was 98.6% and dispersal was rapid; therefore, moratoriums on stocking pallid sturgeon during summer months or during high flow events do not appear to be warranted. 相似文献

We validated the effects of a passive integrated transponder (PIT) tagging process on the oriental weather loach Misgurnus anguillicaudatus. Laboratory experiments were conducted to assess the effects of PIT tagging on fish survival, growth, wound healing, and tag omission. Two tagging protocols, standard syringe injection versus insertion through a small hole pierced by a fine needle-shaped awl, were compared using a 12.5 × 2.07 mm2 tag. A control group was also included. In comparison with the awl technique, syringe injection heightened the mortality of the loach and delayed healing of the wound caused by tag insertion. No effects of either PIT tagging method were detected on the growth of surviving loach. We also field-tested similarly tagged populations within a river-based irrigation system of Sado Island, Japan. Two different sized tags (long, 12.5 × 2.07 mm2; short, 8.5 × 2.12 mm2) were compared by using antenna loggers which detected fish movement through gates and automatically logged tagged fish’s tag IDs and timestamps. By comparing logged data and actual fish collection surveys both below and above the gates, 77% and 30% of actual loach movements were confirmed to have been successfully logged for the long and short tags, respectively. The awl insertion technique with the longer tag is therefore recommended for use in similar studies of smaller fish species. 相似文献

Retention and mortality associated with visible implant elastomer (VIE) and passive integrated transponder (PIT) tagged juvenile chub [ Leuciscus cephalus (L.)], dace [ Leuciscus leuciscus (L.)] and roach [ Rutilus rutilus (L.)] were evaluated. PIT tag retention (96.6–100%) was higher than VIE over the 6-month duration of the experiment. VIE retention was significantly better in the head (96.3–98.8%) than in the fins (78.8–90.9%) the first month after tagging, but the opposite was found after 6 months (head = 21.5–57.5%; fins = 77.2–88.8%). Survival was not significantly different from controls for any treatment, except dace tagged with 23-mm PIT (significantly influenced by mass of fish at tagging) and sham PIT tagged dace, because of initial losses. PIT tags are recommended as the most suitable method for tagging individual juvenile chub, dace and roach based on high retention and survival. VIE implantation in the head (studies < 30 days) and fins (studies > 30 days) could provide a cheap, batch-marking alternative, provided retention rates are monitored. 相似文献

Passive Integrated Transponder tags (PIT tags) are recommended as the most suitable method for tagging fish on the basis of their high retention and fish survival rates. The objective of this study was to determine the long‐term effects (between 1 and 11 years) of the PIT tag on the growth of the yellow European eel (Anguilla anguilla). A difference of more than 50% was observed in the growth of marked and unmarked yellow eels. If this is a general long‐term effect in all eels, it would seriously restrict the use of PIT tags for studying the dynamics of European eel populations, and so for their management. 相似文献

The effects of tagging on small cetaceans are difficult to assess due to logistical difficulties in recapturing the whales. In this study two narwhals, Monodon monoceros, and five harbor porpoises, Phocoena phocoena, were recaptured between 297 and 767 days after instrumentation with satellite transmitters. The transmitters were mounted by pins that were pushed through the fins of the porpoises or the backs of the narwhals. Overall body condition seemed unaffected by the instrumentations. Macroscopical examination revealed that umbilicalization of the tissue surrounding the pins was almost complete. On one of the narwhals the reepithelialization created a closed tunnel where the pins were isolated from the subdermal tissue, however the reepithelialization was incomplete around the middle of the pin and a low‐grade inflammation increased with decreasing thickness of epidermis. The inflammation consisted of mononuclear cells, mainly lymphocytes. With increasing inflammation the number of neutrophils and macrophages increased. In the lymphoid follicular hyperplasia macrophages and a few neutrophils were found, in one case accompanied by Splendore‐Hoeppli material with radiating eosinophilic clubs and Gram‐positive cocci. Immunohistochemical staining of the cocci for Staphylococcus aureus was positive. The observations from the recaptured cetaceans suggest that the instrumentations caused only temporary and low‐grade inflammatory responses. 相似文献

We compared the effectiveness of passive gill nets, hoop nets, set lines, and drifted trammel nets, towed beam trawls and otter trawls to develop criteria to best determine the mean catch per unit effort (CPUE) for juvenile pallid sturgeon (Scaphirhynchus albus) based on selectivity and seasonal efficiency in various habitats of the Missouri River downstream of Fort Randall Dam, South Dakota. Sampling occurred from April to November in 2003 and 2004 and from March to November in 2005. We captured 29 juvenile pallid sturgeon in a total of 498 overnight gill net sets, 55 in 870 drifted trammel nets, 19 on 1683 set lines, and six in 166 otter trawl tows. No pallid sturgeon were captured in 515 beam trawl tows or 520 overnight hoop net sets. Seasonal trends in mean CPUE were found and the relative precision was the greatest in October and November for gill nets, in August for trammel nets, in April for set lines, and in October for otter trawls. A higher proportion of pallid sturgeon captures for gill nets were in the inside bend macrohabitat generally associated with lower water velocities, trammel nets over sand substrate and in the outside bend macrohabitat typically associated with higher water velocities and greater depths, set lines in lower water velocities, and otter trawls in depths >2.5 m and over sand substrate. Although we found trends among seasons and habitats for gill nets, trammel nets, set lines, and otter trawls, the catch rates were low and annual point estimates of relative abundance are not adequate to detect changes in relative abundance of juvenile pallid sturgeon in this reach of the Missouri River. Independently, gill nets, trammel nets, and otter trawls likely captured the size structure of the population of pallid sturgeon in the Missouri River downstream of Fort Randall Dam. Based on our results, a standardized protocol can now be established to systematically monitor juvenile pallid sturgeon, an essential element for determining responses to recovery efforts in the Missouri River. 相似文献

A multiyear study of pallid sturgeon distribution and relative abundance was conducted in the lower and middle Mississippi river (LMR and MMR, respectively). The LMR and MMR comprise the free‐flowing Mississippi River extending 1857 river kilometers (rkm) from its mouth at the Gulf of Mexico upstream to the mouth of the Missouri River. A total of 219 pallid sturgeon and 6018 shovelnose sturgeon was collected during the periods 1996–1997 and 2000–2006. Trotlines baited with worms were the primary collecting gear. The smallest pallid sturgeon captured on trotlines was 405 mm FL and the largest was 995 mm FL. Mean size of pallid sturgeon was statistically smaller in the Mississippi River below the Atchafalaya River near Baton Rouge, LA (621 mm FL). Mean abundance (catch per trotline night) of pallid sturgeon was highest at water temperatures around 10°C. There was a latitudinal trend in mean abundance of pallid and shovelnose sturgeon, but the pattern differed between species. Pallid sturgeon abundance was statistically (P < 0.05) higher (0.3 fish per trotline night) in the lower reach between the Atchafalaya River and New Orleans (rkm 154–507), and at the Chain of Rocks (COR), a low water dam near the mouth of the Missouri River. Pallid sturgeon abundance between these two locations was statistically the same (0.12–0.23). Shovelnose sturgeon abundance increased going upstream, but was disproportionally higher at the COR (22 fish per line compared with <6 fish per line in other reaches). Overall, the ratio between pallid and shovelnose sturgeon varied from a high of 1 : 6 at the lower reach, and gradually decreased upstream to a low of 1 : 77 at the COR. Based on differences in sturgeon abundance, size and habitat characteristics, the free‐flowing Mississippi River can be divided into two reaches in the MMR (i.e. COR is a separate location), and four reaches (i.e., including the Atchafalaya River) in the LMR where management goals may differ. 相似文献
