Glycosyltransferases are members of the multigene superfamily in plants that can transfer single or multiple activated sugars to a range of plant molecules,resulting in the glycosylation of plant compounds.Although the activities of many glycosyltransferases and their products have been recognized for a long time,only in recent years were some glycosyltransferase genes identified and a few functionally characterized in detail.Glycosylation is thought to be one of the most important modification reactions towards plant secondary metabolites,and plays a key role in maintaining cell homeostasis,thus likely participating in the regulation of plant growth,development and in defense responses to stress environments.With advances in plant genome projects and the development of novel technologies in analyzing gene function,significant progress could be made in gaining new insights into the properties and precise biological roles of plant secondary product glycosyltransferases,and the new knowledge will have extensive application prospects in the catalytic synthesis of glycoconjugates and metabolic engineering of crops.In this review,we summarize the current research,highlighting the possible biological roles,of plant secondary metabolite glycosyltransferases and discuss their potential applications as well as aspects to be further studied in the near future.  相似文献

植物激素对植物的生长发育有重要的调节作用。由于激素的作用依赖于其浓度, 所以植物内源活性激素的水平必须受到严格控制, 而糖基化修饰被认为是调控激素活性水平的重要方式之一。随着植物激素糖基化修饰相关糖基转移酶基因不断被克隆与鉴定, 多种植物激素的糖基化修饰机制和功能作用逐渐被揭示。该文重点介绍了近年来植物生长素、细胞分裂素、脱落酸、油菜素内酯、水杨酸、茉莉酸等植物激素的糖基转移酶活性鉴定与功能研究进展。同时, 对植物激素糖基化修饰领域存在的问题和发展前景进行了讨论。  相似文献

Glycosylation is a widespread modification of plant secondary metabolites. It is involved in various functions, including the regulation of hormone homeostasis, the detoxification of xenobiotics and the biosynthesis and storage of secondary compounds. In plants, these reactions are controlled by a specific subclass of the ubiquitous glycosyltransferase family. Although these enzymes have been studied intensively for many years, to date only a handful have been characterized in planta. Plant genome projects have uncovered unsuspected complexity within this family that is hindering the characterization of single genes. However, genome information also paves the way for the development of functional genomic approaches. Here, we highlight recent progress and the outcomes of novel strategies developed to uncover the physiological roles of these glycosyltransferases.  相似文献

Family-1 UDP glycosyltransferases (UGTs) from plants transfer sugar moieties from activated sugar donors to a wide range of small molecules, and control many metabolic processes during plant growth and development. Here, we report a genome-wide analysis of maize that identified 147 Family-1 glycosyltransferases based on their conserved PSPG motifs. Phylogenetic analysis of these genes with 18 Arabidopsis UGTs and two rice UGTs clustered them into 17 groups (A–Q). The patterns of intron gain/loss events, as well as their positions within UGTs from the same group, further aided elucidation of their divergence and evolutionary relationships between UGTs. Expression analysis of the maize UGT genes using both online microarray data and quantitative real-time PCR verification indicates that UGT genes are widely expressed in various tissues and likely play important roles in plant growth and development. Our study provides useful information on the Family-1 UGTs in maize, and will facilitate their further characterization to better understand their functions.  相似文献

On plants, microbial populations interact with each other and their host through the actions of secreted metabolites. However, the combined action of diverse organisms and their different metabolites on plant health has yet to be fully appreciated. Here, the multifactorial nature of these interactions, at the organismal and molecular level, leading to the biological control of plant diseases is reviewed. To do so, we describe in detail the ecological significance of three different classes of secondary metabolites and discuss how they might contribute to biological control. Specifically, the roles of auxin, acetoin, and phenazines are considered, because they represent very different but important types of secondary metabolites. We also describe how studies of the global regulation of bacterial secondary metabolism have led to the discovery of new genes and phenotypes related to plant health promotion. In conclusion, we describe three avenues for future research that will help to integrate these complex and diverse observations into a more coherent synthesis of bacterially mediated biocontrol of plant diseases.  相似文献

Glycosyltransferases of plant secondary metabolism transfer nucleotide-diphosphate-activated sugars to low molecular weight substrates. Until recently, glycosyltransferases were thought to have only limited influence on the basic physiology of the plant. This view has changed. Glycosyltransferases might in fact have an important role in plant defense and stress tolerance. Recent results obtained with several recombinant enzymes indicate that many glycosyltransferases are regioselective or regiospecific rather than highly substrate specific. This might indicate how plants evolve novel secondary products, placing enzymes with broad substrate specificities downstream of the conserved, early, pivotal enzymes of plant secondary metabolism.  相似文献

Sesquiterpene lactones (STLs) constitute a large group of secondary metabolites that are widely distributed in several angiosperm plant families and a few bryophytes, including liverworts. These metabolites are particularly diversified in the family Asteraceae, in which more than 5,000 compounds have been reported so far. In addition to their pharmacological importance and potential therapeutic applications, most STLs display a wide range of protective activities in plants, including acting as anti-herbivory and antimicrobial substances or inhibiting growth of competing plants. These activities are mainly related to their characteristic α,β-unsaturated structural elements, which can participate in Michael-type additions with biological nucleophiles that contain sulfhydryl groups. Supporting the protective roles of STLs, they are mainly located in glandular trichomes of aerial parts because the highly nonspecific toxicity of such compounds necessitates compartmentalization to prevent autotoxicity. However, STLs have also been reported in other aerial and underground organs, where they are assumed to exhibit other biological activities. Recent studies have suggested that these metabolites not only display protective activities due to toxicity but also play key physiological roles in mediating rhizosphere communication among plants, soil microorganisms and plant parasites. STLs have been directly implicated in plant phototropism, resulting in differential growth of a plant organ due to auxin inhibition when accumulated in response to a light stimulus. This review therefore not only highlights the protective roles of STLs in producing plants but also explores the physiological roles of these metabolites, thus providing insights for new research approaches for understanding the roles of STLs in plants and their potential future applications.  相似文献

Flavonoid glycosides are required for a number of crucial roles in planta and have the potential for development in a variety of agricultural, medicinal, and biotechnological applications. A number of recent advancements have been made in characterizing glycosyltransferases, the enzymes that are responsible for the synthesis of these important molecules. In this review, glycosyltransferases are considered with regard to biochemical properties, expression patterns, levels of enzyme activity during development, and structure/function relationships. This is presented with historical context to highlight critical findings, particularly with regard to the innovative work that has come from research on citrus species. The plant glycosyltransferase crystal structures that have been solved over the past decade, either alone or in complex with sugar donor and/or acceptor molecules, are discussed. The application of results from these structures to inform current structure/function work as well as implications and goals for future crystallography and tertiary modeling studies are considered. A thorough understanding of the properties of glycosyltransferases will be a critical step in any future biotechnological application of these enzymes in areas such as crop improvement and custom design of enzymes to produce desired compounds for nutritional and/or medicinal usage.  相似文献

A unique characteristic of carbohydrates is their structural diversity which is greater than that of many other classes of biological compounds. Carbohydrate-containing natural products show many different biological activities and some of them have been developed as drugs for medical use. The biosynthesis of carbohydrate-containing natural products is catalysed by glycosyltransferases. In this review we will present information on the function of glycosyltransferases involved in the biosynthesis of oligosaccharide antibiotics focusing especially on urdamycins and landomycins, two angucycline antibiotics with interesting antitumor activities. We will also discuss the use of glycosyltransferases in combinatorial biosynthesis to generate new "hybrid" antibiotics.  相似文献

The functional elucidation of plant cell wall biosynthesis (CWB) related genes is important for understanding various stress tolerance responses as well as enhancing biomass in plants. Despite their significant role in physiology and growth of the plant, the function of a limited number of CWB related genes have been identified. Major obstacles such as functional redundancy and limited functional information pose challenges in the characterization of CWB genes. Here, a genome-wide analysis of CWB genes using meta-expression data revealed their roles in stress tolerance and developmental processes. The identification of coexpressed CWB genes suggests functional modules for plant cell wall biosynthesis associated with specific tissue types, biotic stress, abiotic stress, and hormone responses. More interestingly, we identified that glycosyl hydrolases are specialized for root and pollen development, glycosyltransferases for ubiquitous function and leaf development, and carbohydrate esterases for pollen development. A T-DNA insertional mutant of OsCESA9 showing internode preferred expression revealed severe dwarfism and a co-expression network analysis of OsCESA9 in oscesa9 mutant suggest downstream pathways for secondary cell wall biosynthesis and DNA repair processes. Data from our studies will facilitate functional genomic studies of CWB genes in rice and contribute to the enhancement of biomass and yield in crop plants.  相似文献

萜类化合物由于其结构类型丰富多样而被称为"terpenome".除了参与植物生长发育、环境应答等生理过程,萜类化合物还应用于医药、有机化工等领域.萜类的生物合成大致可分为前体形成、骨架构建以及后修饰三部分,基本骨架通常由萜类合酶催化形成,进一步在后修饰酶的作用下产生数以万计的萜类化合物.结合我们对香茶菜二萜生物合成的初步研究结果,本文主要针对近年来植物萜类生物合成中的一些有代表性的后修饰酶包括P450单氧酶、双键还原酶、酰基转移酶和糖基转移酶,进行研究现状分析与展望.  相似文献

人类对植物次生代谢产物(天然产物)的早期研究源于它们的应用价值,近些年来人们越来越认识到植物次生代谢产物广泛的生物学效应,开始重新评价这些化合物在植物生命活动以及生态系统中可能扮演的角色。植物的次生代谢是植物在长期进化中与环境(生物的和非生物的)相互作用的结果,次生代谢产物在植物提高自身保护和生存竞争能力、协调与环境关系上充当着重要的角色。介绍了植物次生代谢及其产物的特点,概述了植物次生代谢与温度、水分、光照、养分、CO2浓度、UV-B辐射、环境污染等非生物环境以及与化学防御、化感作用、菌根共生、微生物病害的关系。研究植物次生代谢与环境的关系,可以从更深的层次发掘植物与环境的内在联系,为全面、深入认识植物与环境的相互关系提供新的研究途径,同时也有利于人类更有效、合理地利用植物的次生代谢产物。  相似文献

Jasmonates and related compounds have been highlighted recently in the field of plant physiology and plant molecular biology due to their significant regulatory roles in the signaling pathway for the diverse aspects of plant development and survival. Though a considerable amount of studies concerning their biological effects in different plants have been widely reported, the molecular details of the signaling mechanism are still poorly understood. This review sheds new light on the structural requirements for the bioactivity/property of jasmonic acid derivatives in current computational perspective, which differs from previous research that mainly focus on their biological evaluation, gene and metabolic regulation and the enzymes in their biosynthesis. The bioactivity of many natural products produced by microorganisms can be attributed to their sugar substituents. These substituents are transferred as nucleotide-activated sugars to an aglycon by glycosyltransferases. Engineering these enzymes can broaden their substrate specificity and can therefore have an impact on the bioactivity of the secondary metabolites.In this review we present the generation of a glycosyltransferase gene toolbox which contains more than 70 bacterial glycosyltransferases to date. Investigations of the function, specificity and structure of these glycosyltransferases help to understand the great potential of these enzymes for natural product biosynthesis.  相似文献

Flavonoids are one of the largest classes of small molecular secondary metabolites produced in different parts of the plant. They display a wide range of pharmacological and beneficial health effects for humans, which include, among others, antioxidative activity, free radical scavenging capacity, coronary heart disease prevention and antiatherosclerotic, hepatoprotective, anti-inflammatory, and anticancer activities. Hence, flavonoids are gaining high attention from the pharmaceutical and healthcare industries. Notably, plants synthesize flavonoids in response to microbial infection, and these compounds have been found to be a potent antimicrobial agent against a wide range of pathogenic microorganisms in vitro. Antimicrobial action of flavonoids results from their various biological activities, which may not seem very specific at first. There are, however, promising antibacterial flavonoids that are able not only to selectively target bacterial cells, but also to inhibit virulence factors, as well as other forms of microbial threats, e.g. biofilm formation. Moreover, some plant flavonoids manifest ability to reverse the antibiotic resistance and enhance action of the current antibiotic drugs. Hence, the development and application of flavonoid-based drugs could be a promising approach for antibiotic-resistant infections. This review aims to improve our understanding of the biological and molecular roles of plant flavonoids, focusing mostly on their antimicrobial activities.

Studies of the glycosyltransferases (GTs) of small molecules have greatly increased in recent years as new approaches have been used to identify their genes and characterize their catalytic activities. These enzymes recognize diverse acceptors, including plant metabolites, phytotoxins and xenobiotics. Glycosylation alters the hydrophilicity of the acceptors, their stability and chemical properties, their subcellular localisation and often their bioactivity. Considerable progress has been made in understanding the role of GTs in the plant and the utility of GTs as biocatalysts, the latter arising from their regio- and enantioselectivity and their ability to recognize substrates that are not limited to plant metabolites.  相似文献
