Changes in net photosynthesis, respiration, transpiration andcontents of total C, NO3-N and reduced N were followed throughoutthe life of leaf 6 of nitrate-dependent plants of castor beanexposed to moderate salinity stress (71 mol m–3 NaCl).Salt treatment was applied for measuring mineral flows in aparallel study (Jeschke and Pate, 1991b). Concurrent measurementswere made of solute composition and C: N molar ratios and concentrationsof reduced N and collected NO3-N in phloem sap bleeding fromshallow incisions in the top and at the base of petioles andin xylem exudates from flaps of proximal leaf midribs followingpressurization of the root system. The resulting data were usedto construct empirical models of the respective economies ofC, total N, NO3 and reduced N for a sequence of defined phasesof leaf life. Water use efficiency increased 3-fold from emergenceto a maximum of 1·5 mmol CO2 mol–1 H2O before decliningto 0·5 mmol CO2 mol–1 H2O at senescence. Xylemmolar ratios of C:N varied from 1·2–2·8,with nitrate always a smaller component than reduced N. Phloemsap C:N increased from 10–40 with leaf expansion and wasthen maintained in the range of 40–50 until falling steeplyto 20 at leaf senescence. Nitrate comprised less than 1% oftotal N in all phloem sap samples. The models of C uptake, flow,and utilization showed a major role of phloem import and thenincreasingly of laminar photosynthesis in providing C for leafgrowth. The carbon budget was thereafter characterized by ratesof phloem export closely matched to net rates of CO2 fixationby the lamina. Corresponding data for total N depicted an earlymajor role of both xylem and phloem import, but the eventualdominance of xylem import as the N source for leaf growth. Cyclingof N by xylem to phloem exchange commenced before the leaf hadachieved maximum N content, and was the major contributor tophloem export until leaf senescence when mobilized N providedmost exported N. The nitrate economy of the leaf was characterizedby early establishment of tissue pools of the ion in the petioleand to a lesser extent in the lamina, continued high rates ofnitrate reduction in the lamina but negligible assimilationin the petiole, and a release through xylem of previously accumulatedNO3 from petiole to lamina. Related data for reduced N illustratedthe much greater importance of this form of N than nitrate intransport, storage and cycling of N at all stages of leaf andpetiole life. Xylem to phloem interchanges of reduced N in petiolewere minimal in comparison with cycling through the lamina.The ratio of CO2 reduction to NO3 reduction in the lamina wasat first low (57 mol mol–1) increasing to a peak valueof 294 during mature leaf functioning before declining to 190during the presenescence phase of leaf development. This patternreflected age-related effects on water use efficiency, changesin NO3 levels in the xylem stream entering the lamina, and therelatively low photosynthetic performances of very young andsenescent laminae. Fully expanded leaves of Erythroxylum coca var. coca Lam. (Erythroxylaceae)35-70-d-old were harvested from 21-month-old plants grown undergreenhouse conditions. Each harvested leaf was placed on a plexiglasssilhouette according to its dimensions and divided into fourprimary sections (petiole, base, mid and anterior) and threesubsections (lamina periphery, false mid-rib, and true mid-rib)to determine the distribution and content of hygrine, cuscohygrine,trans-cinnamoylcocaine, cis -cinnamoylcocaine, tropacocaine,tropinone, methyl ecgonine and cocaine. The leaf sub-sectionsand petiole were extracted and analysed for alkaloid content(%) by HPLC (cocaine alkaloid only) gas chromatography and GC/MS.Cocaine, methyl ecgonine and hygrine were highest in the laminaperiphery with a content of 0·48, 0·46 and 0·32%,respectively. Trans-cinnamoylcocaine was pre-eminent of thecinnamoylcocaines and was most abundant in the petiole at acontent of 0·24%. Studies of in vivo nitrate reductase activity (NRA) in Eichhorniacrassipes showed that adding 5% isopropanol gave highest NRAin all parts of the plant, but it was also necessary to flushthe assay media containing leaves with N1 to obtain the maximalactivity. There were differences in the pH optima for NRA, theroot optimum being 6·5 leaf 7·5–8·0,and the petiole showing a major peak at 6·5 and a minorpeak at 7·.5–8·0. Removal of the blade from the leaf subtending the first flowerbud on Cyclamen persicum ‘Swan Lake’ plants causedthe petiole of that leaf to senesce, but had no effect on thegrowth of the flower peduncle in the debladed petiole's axil.A 10 mg NAA l–1 application generally had no effect onpetiole senescence, peduncle elongation or flowering date whenapplied to the cut end of the petiole after blade removal. A25 mg GA3 l–1 application or a combination of 25 mg GA3l–1 application or a combination of 25 mg GA3 l–1plus 10 mg NAA l–1 delayed petiole senescence and enhancedpeduncle elongation and subsequent flowering. The physical pathway of a systemic signal linking local woundingand systemic synthesis of proteinase inhibitors was investigatedin tomato (Lycopersicon esculentum Mill. ‘Moneymaker’)plants. Lucifer Yellow was used to visualize wound induced flowin the xylem. Cuts under water or severe wounds (heat or largecrushing wounds) induced flows in the xylem to other parts ofthe plant in a pattern determined by the vascular architecture.The detailed distribution of systemic proteinase inhibitor activityfollowing these wounds was similar to the pattern of wound inducedflow in the xylem. Steaming the petiole of the wounded organdid not prevent the systemic induction of proteinase inhibitorby a severe wound. It was concluded that elicitors releasedby a severe wound were distributed systemically in the xylem.Small crushing wounds did not induce systemic flow in the xylembut did induce proteinase inhibitor activity in organs importingvia the phloem. Steaming the petiole of the wounded leaf preventedsystemic induction of proteinase inhibitor by small crushingwounds, a result which is consistent with the translocationof elicitors in the phloem. These results indicate the participationof more than one signalling pathway in the systemic inductionof proteinase inhibitor synthesis by wounding. An experimentally-based modelling technique was developed todescribe quantitatively the uptake, flow, storage and utilizationof NO3-N over a 9 d period in mid-vegetative growth of sandcultured castor bean (Ricinus communis L.) fed 12 mol m–3nitrate and exposed to a mean salinity stress of 128 mol m–3NaCl. Model construction used information on increments or lossesof NO3-N or total reduced N in plant parts over the study periodand concentration data for NO3-N and reduced (amino acid) Nin phloem sap and pressure-induced xylem exudates obtained fromstem, petiole and leaf lamina tissue at various levels up ashoot. The resulting models indicated that the bulk (87%) of incomingnitrate was reduced, 51% of this in the root, the remainderprincipally in the laminae of leaves. The shoot was 60% autotrophicfor N through its own nitrate assimilation, but was oversuppliedwith surplus reduced N generated by the root and fed to theshoot through the xylem. The equivalent of over half (53%) ofthis N returned to the root as phloem translocate and, mostly,then cycled back to the shoot via xylem. Nitrate comprised almosthalf of the N of most xylem samples, but less than 1% of phloemsap N. Laminae of leaves of different age varied greatly inN balance. The fully grown lower three leaves generated a surplusof reduced N by nitrate assimilation and this, accompanied byreduced N cycling by xylem to phloem exchange, was exportedfrom the leaf. Leaf 4 was gauged to be just self-sufficientin terms of nitrate reduction, while also cycling reduced N.The three upper leaves (5–7) met their N balance to varyingextents by xylem import, phloem import (leaves 6 and 7 only)and assimilation of nitrate. Petioles and stem tissue generallyshowed low reductase activities, but obtained most of theirN by abstraction from xylem and phloem streams. The models predictedthat nodal tissue of lower parts of the stem abstracted reducedN from the departing leaf traces and transferred this, but notnitrate, to xylem streams passing further up the shoot. As aresult, xylem sap was predicted to become more concentratedin N as it passed up the shoot, and to decrease the ratio ofNO3-N to reduced N from 0·45 to 0·21 from thebase to the top of the shoot. These changes were reflected inthe measured N values for pressure-induced xylem exudates fromdifferent sites on the shoot. Transfer cells, observed in thexylem of leaf traces exiting from nodal tissue, were suggestedto be involved in the abstraction process. Air-embolism formation in xylem vessels of Populus tremuloidesMichx. was quantified by its reduction of hydraulic conductivityin branch segments. Embolism was induced by increasing xylemtension in drying stems, or by inserting one end of a hydratedstem in a pressure bomb and increasing air pressure in the bomb.Both treatments produced the same response suggesting that embolismby water stress was caused by air entering water-filled vessels,presumably through inter-vessel pits. In rapidly-growing P.tremuloides branches, the vessels of the outer growth ring werefunctional whereas vessels in older xylem were mostly embolized.This selective embolizing of older vessels was associated witha marked increase in permeability of their inter-vessel pitsto air, relative to pits of younger vessels. Air-injection pressuresless than 1·0 MPa completely embolized older vesselsthat had been re-filled in the laboratory, whereas pressuresover 4·0 MPa were required to embolize young vessels.Greater permeability of old vessels was due to degradation oftheir pit membranes as seen in the scanning electron microscope;large openings were present that were not seen in pit membranesof young vessels. These holes would allow air to penetrate vesselends at low pressure differences causing embolism. Degradationof pit membranes causing the selective dysfunction of oldersapwood may be a general phenomenon initiating heartwood formationin many species. An enlargement of the peduncle and rachis of the terminal racemeand the petiole of the uppermost mainstem leaf was observedin soybean plants [Glycine max (L.) Merr.] treated with thecytokinin, 6-benzylaminopurine (BAP). Histological studies wereperformed to determine the timing and extent of anatomical changesaccompanying BAP treatment. Swelling of treated ‘Tracy-M’peduncles, rachises, and petioles was observed within 4–6d after treatment initiation. A significant increase in totalcross-sectional tissue area was observed at lower and intermediateinternodes of treated rachises after 11 d. Rachis enlargementwas due to increases in both cell size and cell number, particularlyof the vascular tissue. In treated petioles of IX93-100, procambialcells of vascular bundles were the first to respond to the BAPtreatment. Proton Fluxes and the Activity of a Stelar Proton Pump in Onion Roots   总被引:6,自引:2,他引:4  
The xylem vessels of excised adventitious roots of onion, Alliumcepa, were perfused with unbuffered nutrient solution adjustedinitially to either pH 9·3 or 3·9; the pH of thesolution after passage through the xylem, at rates not lessthan 2 xylem volume changes min–1, was close to pH 6·5in both instances. The flux of H+ across the xylem/symplastboundary into mildly alkaline, phosphate-buffered solutionsperfusing the vessels could be increased greatly with increasingbuffer strength, up to a maximum value between 0·5–1·0pmol H+ mm–2 s–1. The apparent neutralization ofacidic malic acid buffers had a slightly lower maximum capacity,equivalent to –0·3 to –0·5 pmol H+mm–2 s–1. The addition of 5·0 pmol m–3fusicoccin (FC) to the xylem perfusion solution stimulated theentry of H+ into the xylem; in unbuffered perfusion solutionsthe pH fell to pH 3·6 after a lag of 25–35 min.FC additions to phosphate-buffered solutions also stimulatedthe H+ flux to an extent similar to that in unbuffered solution,viz. 0·2–0·4 pmol mm–2 s–1. The release of K+ (36Rb-labelled) into xylem sap transientlyincreased as the [K+] in weakly buffered perfusion solutionswas raised stepwise; a very marked increase being seen whenthe concentration was raised to 100 mol m–3 from 40 molm–3. The addition of 5·0 mmol m–3 FC to theperfusing solution containing 100 mol m–3 K+ rapidly decreasedthe K+ flux to the xylem as the H+ flux increased. Fusicoccinalso inhibited the flux of K+ into unbuffered perfusion solutionsbut the effect appeared reversible. Addition of 10 mmol m–3abscisic acid (ABA) to the perfusion solution quickly producedtransient increases in both K+ and H+ fluxes into the xylem.In this and other experiments using weakly phosphate-bufferedperfusing solutions, H+ fluxes were comparable in size to thoseof K+ The results are consistent with the idea that the stele of onionroots contains a proton trarislocating ATPase whose activityresponds to the pH of the xylem sap. It is evident that theactivity of the proton secreting and proton neutralizing mechanismsin the xylem parenchyma control the movement of other ions acrossthe xylem/symplast boundary. Cortical Bundles in the Persistent, Photosynthetic Stems of Cacti   总被引:2,自引:2,他引:0  
We examined 62 species in 45 genera of the cactus subfamilyCactoideae; all had collateral cortical bundles that permeatedthe broad, water-storing inner cortex and extended to the baseof the outer, photosynthetic palisade cortex. Mean distancebetween cortical bundles was 0.75 mm, similar to the mean spacing(0.74 mm) of veins in leaves of Pereskia, a genus of relictleaf-bearing cacti. In 16 species, both young and extremelyold stem cortex was available for study: in all of these, olderbundles had larger amounts of phloem than did younger bundles,indicating that phloem had been produced for many years. Inten species, older bundles also had more xylem than youngerbundles. In two genera (Rhipsalis and Selenicereus) there werecaps of primary phloem fibres, and in a single species (Pilosocereusmortensenii) cortical bundle xylem contained libriform fibres.All cortical bundle tracheary elements were narrow (radius range,0.91–8.2 µm; mode, 1.8–2.7 µm), similarto Pereskia leaf vein elements (radius range, 1.8–2.7µm); this was much narrower than stem wood vessels (radiusrange, 10–42 um; mode, 23–28 µm). Longitudinalconduction of water and nutrients probably occurs predominantlyin stem wood, with cortical bundles maintaining the broad, voluminouscortex, the outer part of which is the plant's photosynthetictissue and the inner part of which stores water and starch.The cortex of the Cactordeae contains numerous leaflike characters;homeotic genes may be involved in its morphogenesis. A root excision technique was used to estimate the proportionof total resistance to water flux residing in the soil, theroot, and the xylem of lodgepole pine (Pinus contorta Douglex. Loud.) trees in the field. Root excision at mid-day alwaysresulted in rapid recovery of leaf water potential when waterwas supplied to the cut stem, suggesting a high soil-root resistance.Transpiration was unaffected if leaf water potential beforecutting was not limiting leaf conductance. By mid-June wateruptake by the excised stem always exceeded calculated crowntranspiration indicating recharge of internal sapwood storage.Predawn leaf water potential before root excision was highlycorrelated with total soil-plant resistance (r2 = 0·89)and calculated root water uptake (r2 = 0·92).  相似文献

The quantitative relationship between turgor and the pressureexerted by the inner tissues (cortex, vascular tissue, and pith)on the peripheral cell walls (longitudinal tissue pressure)was investigated in hypocotyls of sunflower seedlings (Helianthusannuus L.) In etiolated hypocotyls cell turgor pressures, asmeasured with the pressure probe, were in the range 0·38to 0·55 MPa with an average of 0·48 MPa. In irradiatedhypocotyls turgor pressures varied from 0·40 to 0·57MPa with a, mean at 0·49 MPa. The pressure exerted bythe inner tissues on the outer walls was estimated by incubatingpeeled sections in a series of osmotic test solutions (polyethyleneglycol 8000). The length change was measured with a transducer.In both etiolated and irradiated hypocotyls an external osmoticpressure of 0·5 MPa was required to inhibit elongationof the inner tissues, i.e. the average cell turgor and the longitudinaltissue pressure are very similar quantities. The results indicatethat the turgor of the inner tissues is displaced to and borneby the thick, growth-limiting peripheral cell walls of the hypocotyl. An investigation has been made of the relation between the sizesof the lamina and of the petiolar xylem of both mature and immatureleaves of the runner bean (Phaseolus multiflorus Willd.). The ratios xylem area/lamina area and the number of vessels/laminaarea are lowest for mature leaves. Immature leaves gave higherbut somewhat more variable values for these two ratios. There is a constant growth ratio between the lpminn area andthe xylem area, such that k is approximately o·61 inthe allometry formula. The significance of the results is briefly considered from thepoint of view that the xylem area is related to the water requirementsof the leaf.  相似文献

Developmental patterns of lateral roots and their vascular differentiationwere investigated for Vitis vinifera L. cv. Shiraz to assessthe likely contribution of lateral roots to total water uptakeof plants subjected to different irrigation regimes. Correlationanalyses showed a significant positive correlation between mainroot diameter and the diameter of first order lateral rootsof well-watered plants, but in water-stressed plants the twowere not significantly correlated. The correlations betweendiameters of first order lateral roots and the diameters ofmain roots were greater than correlations between the lengthsof first order laterals and the diameters of main roots. Thesuberised surface area of well-watered main roots increasedfrom 4% of total surface area at 0·25 cm to 100% at 10cm from the tip, whereas that of stressed plants increased from15% at 0·25 cm to 100% at 5 cm from the tip. In all treatmentsthe highest linear density of first order laterals was about7 laterals cm-1 of main root. More than 50% of first order lateralshad diameters less than 0·05 cm, and more than 90% ofthem had lengths less than 5 cm. Calculations of axial resistancesbased on xylem diameter measurements suggest that the axialresistances of root segments may not be uniform along rootsas is often assumed in models of water uptake. Water flow intothe main roots via the lateral root pathway is likely to bemuch smaller than that via the direct radial flow pathway asonly about 1% of surface area of main roots is directly occupiedby lateral roots, leaving the other 99% of main root surfacearea available for the direct radial flow pathway.Copyright1994, 1999 Academic Press Axial resistance, grapevine (Vitis vinifera L. cv. Petiole development and formation of xylem vessels have been investigated in Xanthium leaves from early ontogeny to maturity. Kinetics of growth was presented in terms of absolute and relative elemental rates of elongation. The process of vascularization was assessed by the number of differentiated xylem vessels. The leaf plastochron index (LPI) developed by Erickson and Michelini (1957) was used for designating the various stages of development. An exponential increase in petiole length was observed between the LPIs –3 and +4 indicating a constant relative rate of 0.20 or 20% increase per day. After cessation of lamina elongation at LPI 8, petiole elongation continued for an additional 5 day period, to LPI 9.5. Relative elemental rate analysis revealed that the basipetal pattern of elongation was maintained throughout the leaf development. At a specific plastochron age, the only growth was due to the petiole elongation. Leaves which ceased elongating had not completed their internal development, since the process of xylem formation continued for several plastochrons, or about 8 days. The highest rate of xylem formation was ten vessels per day at LPI 5. On the average, about five xylem vessels differentiated per day in the middle portion of a Xanthium petiole. Mature petioles contained an average of 218 xylem vessels. About 12 canals of schizogenous origin preceeded the development of the vascular tissue.  相似文献

In vigorously growing shoots of apple and plum 14C-assimilateswere translocated from a ‘fed’ leaf to particularsectors of other leaves in a distribution pattern associatedwith the phyllotaxis; the same sectorial and distribution patternswere produced by 32P phosphate solution taken into the shootthrough a cut petiole. The frequency with which a given sectorialpattern occurred at a particular position on the phyllotacticspiral was ascertained. Such patterns were not observed abovethe third rolled leaf in the apple shoot apex. Killing the phloem in the petiole prevented egress of labelledassimilate but not of 32P solution. Barkringing above the sourceleaf reduced, but did not completely prevent, assimilate movementup the stem, suggesting some translocation in the xylem. Distribution of label from 45CaCl2, 86RbCl and [3H]asparagine,incorporated through cut petioles, did not follow the same patternas label from 32P solutions. A sequential study describing the content (%) of alkaloids inleaves of Erythroxylum coca var. coca Lam. from leaf bud developmentto leaf drop has not previously been conducted. The length oftime the leaf resides on the plant and its concurrent metabolicactivity also has not been defined. In the present study, cocaine,methyl ecgonine, hygrine, tropinone, trans -cinnamoylcocaine,cis-cinnamoylcocaine, tropacocaine and cuscohygrine were monitoredto determine: (1) their content and patterns of accumulationfrom leaf bud to leaf drop; (2) the time leaves resided on theplant; and (3) whether leaves were metabolically active untilleaf drop. E. coca plants were grown in a controlled environmentfor 37 months. Leaves similar in chronological age were extractedand analysed for alkaloid content by gas chromatography (GC)and gas chromatography/mass spectrometry (GC/MS). Cocaine washighest in 7 d old rolled leaves (0·75%) and declinedto 0·39% at leaf drop. Cocaine, methyl ecgonine, hygrine,tropinone, trans -cinnamoylcocaine, cis-cinnamoylcocaine, cuscohygrineand tropacocaine content in 35 d old (mature) leaves was 0·61,0·59, 0·68, 0·08, 0·31, 0·55,0·52, and 0·05%. respectively. Cocaine, methylecgonine, hygrine, cis -cinnamoylcocaine, and cuscohygrine displayeda gradual decline from week 2 to week 36 of leaf duration. Tropinoneand tropacocaine were the least abundant of the alkaloids monitored.Cis-cinnamoylcocaine content exceeded cocaine at week 12, 16,and weeks 19 to 23 of leaf duration. Trans -cinnamoylcocainewas highest in rolled leaves (week 1) and in expanded leavesafter week 30. The monitored alkaloids appeared to be part ofthe metabolically active pool of the leaf. Lolium temulentum seedlings were grown on a nutrient mediumcontaining NH4NO2 at 0, 0·1, 0·5, 1·0 and4·3 mmoll–1 as the sole N source. Relative andabsolute extension rates, maximal leaf size, duration of extensiongrowth, rate of leaf appearance and plastochron index were determinedfrom the parameters of Richards functions fitted to lengthsof laminae measured at intervals after sowing. The final lengthof leaf I was relatively insensitive to N whereas mean relativeextension rate was increased and duration of growth decreasedwith increasing NH4NO2 concentration. Leaves 2 and 3 enlargedprogressively with N at concentrations up to 1·0 mmoll–1but were unresponsive thereafter. There was no significant correlationbetween final length and mean relative extension rate for leaves1 to 3. Leaves 4 to 6 continued to show increasing length beyond1·0 mmoll–1 N and final length was significantlycorrelated with mean relative extension rate. Increasing N increasedthe rate of leaf appearance by decreasing the duration of leafextension and plastochron. These results are discussed in relationto the control of leaf and N turnover. The development of the first pair of leaves of the Runner Bean(Phaseolus multiflorus Willd.) under various conditions is described. 1. Under damp conditions and also when one leaf of a pair wascovered with a black paper envelope, the rate of developmentwas altered but no significant changes in the relative developmentof the parts of the leaf occurred. 2. Removal of a portion of the lamina at an early stage of developmentresulted, at maturity, in a leaf having relatively less petiolarxylem, although the ratio xylem area/lamina area was slightlyhigher. This treatment also delayed a ‘sudden’ increasein area of the vessels which was seen in normal plants. 3. An attempt was made to estimate the transporting power ofthe petiolar xylem and to compare it with the lamina area. Thedata suggest that the maximum possible area of the lamina atany stage in development may be determined by the transportingpower of the xylem, but that the attainment of this maximumpossible area may be prevented by external factors as is seenin some of the experimental results presented.  相似文献

A controlled environment experiment investigated whether thered:far-red (R:FR) ratio of light at the apical bud of the mainstolon could alter plant morphogenesis in clonal cuttings ofwhite clover (Trifolium repens L.) The apical bud included theapical meristem, five to six developing leaf primordia withassociated axillary bud primordia and stipules and the firstemerged folded leaf until development was greater than 0·3on the Carlson scale. Three light regimes were imposed on theapical bud by collimating light from R or FR light-emittingdiodes so that the R:FR ratio of light incident at the apicalbud was set at 0·25, 1·6 or 2·1, withoutsignificantly altering photosynthetically active radiation.The effect of these light regimes on white clover seedling growthwas also tested. At a low R:FR ratio seedling hypocotyl and cotyledon lengthswere significantly longer. However, with the cuttings, the lighttreatments did not alter node appearance rate or internode lengthof the main stolon, petiole length, area of leaves or totalshoot dry matter. There was one significant photomorphogeneticresponse in the cuttings, a delay of 0·5 of a phyllochronin the appearance of branches from axillary buds in the lowR:FR ratio treatment relative to the other treatments. Wherebranch appearance was delayed plants had fewer branches. Thisdifference could be ascribed solely to a delay in branch appearanceas there were no significant treatment effects on either theinitiation of axillary bud primordia within the apical bud,the probability of branching or on the rate of growth of branchesafter appearance.
