Summary Production of ethanol from cellodextrins, as large as cellohexose, byCandida lusitaniae andC. wickerhamii was studied.C. lusitaniae fermented only glucose and cellobiose, whereasC. wickerhamii efficiently fermented cellodextrins. Maximum ethanol yields of 29.2 g/liter from 54 g/liter cellodextrins were achieved byC. wickerhamii in 3–4 days.  相似文献

Plants were regenerated from leaf explants of Centrosema brasilianum cultured in vitro. Callus and buds were produced on Murashige and Skoog medium (MS), 0.8% agar, 0.1 mg/l NAA and 1 mg/l BAP. Regeneration of multiple shoots was achieved by transferring callus onto fresh medium containing 0.01 and 1 mg/l of NAA and BAP, respectively. Shoots formed roots upon transfer to MS with 0.01 mg/l NAA. Plantlets were succesfully transferred to soil. Leaf-derived calli of Centrosema arenarium, C. macrocarpum, C. pascuorum, C. pubescens, and C. virginianum did not produce shoots when cultured in vitro.  相似文献

Summary Thermophilic degradation of sugar beet pulp was studied in batch cultures at 55°C by different associations of bacteria, includingClostridium thermocellum,Methanobacterium sp. andMethanosarcina MP.C. thermocellum produced acetate, succinate, methanol, ethanol, H2 and CO2. The coculture ofC. thermocellum andMethanobacterium sp. produced trace amounts of ethanol and succinate; acetate concentration was about three times higher than in theC. thermocellum monoculture. The association of this coculture withMethanosarcina MP produced 5.5 mmol CH4/g dry weight sugar beet pulp.  相似文献

Summary Four intracellular enzymes from two species of breadmaking yeasts- S. cerevisiae and C. boidinii- have been measured as a function of time during its disruption using a bead mill in batch operation. The amount and rate of enzyme released was dependent on its location inside the cell as well as on the kind of yeast. The maximum amount of invertase, a-D-glucosidase, alcohol dehydrogenase and fumarase was obtained at 2,5,10,15 min. respectively for S. cerevisiae. C. boidinii did not show either invertase nor a-D glucosidase activity and the maximum amount of alcohol dehydrogenase and fumarase were reached at 5 and 20 min. respectively.  相似文献

Summary A total of 55 yeast strains selected from 7 genera known to ferment carbohydrates to ethanol were screened for their ability to ferment glucose to ethanol in shaken flask culture at 37°, 40° and 45°C. Yields of more than 50% of the theoretical maximum were obtained with 28 strains at 37°C, but only 12 at 40°C. Only 6 could grow at 45°C, but they produced poor yields. In general Kluyveromyces strains were more thermotolerant than Saccharomyces and Candida strains, but Saccharomyces strains produced higher ethanol yields. The 8 strains with the highest yields at 40°C were evaluated in batch fermentations. Three of these, two Saccharomyces and one Candida, were able to meet minimum commercial targets set at 8% (v/v) ethanol from 14% (w/v) glucose at 40°C.  相似文献

Summary Temperatures as high as 36°C and 40°C did not negatively affect the ethanol productivity of Jerusalem artichoke (J.a.) juice batch fermentation and the final concentrations of ethanol were close to those produced at lower temperatures. At higher process temperatures (36–40°C), ethanol toxicity inKluyveromyces marxianus was less important during the fermentation of J.a. juice as compared with a simple medium. In simple medium, the heat-sticking of fermentation was observed and the percentage of unfermented sugars steeply increased from 28°C up to 40°C.  相似文献

Summary A basic procedure was developed to produce a tempe-like product using the mouldRhizopus oligosporus and black common beans (Phaseolus vulgaris) as substrate. The initial pH of the substrate was 5.8, and fermentation was conducted at 37°C with a relative humidity of 70% for 72 hrs. Levels of soluble solids and soluble protein increased dramatically as a result of fermentation. Some changes were as well observed in fatty acid contents of fermented samples. It was concluded that the common bean used was an acceptable substrate for preparing this product.  相似文献

Summary The optimal growth rate ofLipomyces starkeyi, with dextran as sole carbon source, was found within the pH range 2.5–4.0, and temperature between 25–30°C. This yeast was unable to grow above 33°C. Dextranase production optima paralleled growth optima, except at pH 2.5. Decrease in enzyme yield at this pH could not be attributed to poor yeast growth or enzyme stability.  相似文献

The plasmid pDHDP5812 carryingdapA gene ofC. glutamicum led to increase in lysine production in theseE. coli strains. Threonine and lysine co-producerE. coli TF1 with pDHDP5812 produced lysine with small amount of threonine. Summary Four diverse strains of Agrobacterium tumefaciens (C58, Ach5, GV3111, and A281) were capable of inducing tumors at a high frequency on inoculated stems of lentil (Lens culinaris Medik. cultivar Laird) in vivo, and on excised shoot apices in vitro. GV3111 and Ach5 produced the largest and heaviest tumors in vivo, while A281 produced the heaviest tumors in vitro. Tumor formation and opine production are indicative of plant cell transformation and tumors produced appropriate opines: nopaline (C58), octopine (Ach5 and GV3111), and agropine and mannopine (A281). Southern analysis of DNA from a tumor line produced by strain C58 showed that a T-DNA fragment had been transferred into the lentil genome.  相似文献

Summary Enzymatically hydrolysed cassava starch was used for C. utilis cultivation. Highly efficient starch hydrolysis was achieved with a 92% DE syrup obtained after 15–20h. Cyanide content fell during cassava processing to very low levels in the hydrolysate. Comparison of biomass yields and protein of C. utilis using molasses and cassava hydrolysate as substrates demonstrates the potential of the latter for yeast production.  相似文献

Summary Cell suspension cultures of a Ceratocystis ulmi-resistant (Ulmus pumila) and a -susceptible elm (U.campestris) were established from leaf callus tissue. Treatment of cultures with spores of C.ulmi induced a large increase in the activity of phenylalanine ammonialyase, only in the cells of the resistant species U.pumila with a maximum after 24 h. Inoculated U.pumila cells also excreted a red unidentified chemical into the culture medium. Neither responses were induced in inoculated U.campestris cultures. The results are discussed in relation to the development of the elm cell culture system as a model for studying the differential biochemical mechanisms of disease resistance in elms.  相似文献

Summary Unusually low culture temperature, such as 20°C, was shown to be preferable for the synthesis of active human interferon- (IFN-) inE. coli harboring a recombinant plasmid. TheE. coli cells cultured at 20°C gave 8.6-fold higher IFN- activity than those cultured at 37°C. However, almost the equal amounts of IFN- protein were accumulated in both cells cultured at 20°C and at temperature higher than 20°C, suggesting that IFN- might exist as an active form in the cells cultured at 20°C, while as a rather denatured form in the cells cultured at higher temperature.  相似文献

Electrochemical energy as a source of reducing equivalent was applied to the cultures ofClostridum acetobutylicum to understand the effects of the pressure by reducing equivalent on anaerobic bacteria. The fermentation byC. acetobutylicum with methyl viologen and electrochemical energy produced more butanol (up to 26%) than the control culture, whilst less acetone (up to 25%) was produced. But no effect was observed on the growth of the culture. These results were indirectly supported byin vitro electrochemical reduction of NAD+ and artificial electron carriers.  相似文献

Summary Alcohol producing top and bottom yeasts were employed individually and together to assess their role in enhancing the rate of ethanol production, in cane molasses fermentation, at 30°C. The combination of top yeastS.cerevisiae NCIM 3281, and bottom yeastS.uvarum NCIM 3509, improved the enthanol production rate by 32.6% in batch fermentation and 25.2% in recycling yeasts cell fermentation as compared to their mean value of individual ethanol production activity.  相似文献
