The aim of this study was to determine the isotopic‐turnover rate (RIT) and trophic‐discrimination factor (FTD) in muscle tissues of Lebranche mullet Mugil liza fed an experimental diet (δ13C = ?27·1‰; δ15N = 1·0‰). Juvenile M. liza exhibited a relatively fast RIT, with a half‐life (t50) of only 16 and 14 days for δ13C and δ15N respectively and a nearly complete isotopic turnover (t95) of 68 and 60 days for δ13C and δ15N.  相似文献

There is a basal level of enzyme activity for trypsin, aminopeptidase, amylase, and lipase in the gut of unfed larval (L6) Spodoptera frugiperda. Trypsin activity does not decrease with non‐feeding, possibly because of the low protein levels in plants along with high amino acid requirements for growth and storage (for later reproduction in adults). Therefore, trypsin must always be present so that only a minimal protein loss via egestion occurs. Larvae, however, adjust amylase activity to carbohydrate ingestion, and indeed amylase activity is five‐fold higher in fed larvae compared to unfed larvae. Gut lipase activity is low, typical of insects with a high carbohydrate diet. A flat‐sheet preparation of the ventriculus was used to measure the release of enzymes in response to specific nutrients and known brain/gut hormones in S. frugiperda. Sugars greatly increase (>300%) amylase release, but starch has no effect. Proteins and amino acids have little or no effect on trypsin or aminopeptidase release. The control of enzyme release in response to food is likely mediated through neurohormones. Indeed, an allatostatin (Spofr‐AS A5) inhibits amylase and trypsin, and allatotropin (Manse‐ AT) stimulates amylase and trypsin release. Spofr‐AS A5 also inhibits ileum myoactivity and Manse‐AT stimulates myoactivity. The epithelial secretion rate of amylase and trypsin was about 20% of the amount of enzyme present in the ventricular lumen, which, considering the efficient counter‐current recycling of enzymes, suggests that the secretion rate is adequate to replace egested enzymes. © 2009 Wiley Periodicals, Inc.  相似文献

The emergence of strains of multidrug‐resistant Gram‐negative bacteria mandates a search for new types of antimicrobial agents. Alyteserin‐2a (ILGKLLSTAAGLLSNL.NH2) is a cationic, α‐helical peptide, first isolated from skin secretions of the midwife toad, Alytes obstetricans, which displays relatively weak antimicrobial and haemolytic activities. Increasing the cationicity of alyteserin‐2a while maintaining amphipathicity by the substitution Gly11→ Lys enhanced the potency against both Gram‐negative and Gram‐positive bacteria by between fourfold and 16‐fold but concomitantly increased cytotoxic activity against human erythrocytes by sixfold (mean concentration of peptide producing 50% cell death; LC50 = 24 µm ). Antimicrobial potency was increased further by the additional substitution Ser7→Lys, but the resulting analogue remained cytotoxic to erythrocytes (LC50 = 38 µm ). However, the peptide containing d ‐lysine at positions 7 and 11 showed high potency against a range of Gram‐negative bacteria, including multidrug‐resistant strains of Acinetobacter baumannii and Stenotrophomonas maltophilia (minimum inhibitory concentration = 8 µm ) but appreciably lower haemolytic activity (LC50 = 185 µm ) and cytotoxicity against A549 human alveolar basal epithelial cells (LC50 = 65 µm ). The analogue shows potential for treatment of nosocomial pulmonary infections caused by bacteria that have developed resistance to commonly used antibiotics. A series of laboratory bioassays with each consisting of low, medium and high concentration treatments of the fungal biocontrol agent Beauveria bassiana alone or supplemented with an increasing sublethal rate of imidacloprid were conducted to quantify the fungal and chemical interactions on chrysanthemum aphid Macrosiphoniella sanborni (0.01‐0.05 a.i. μg mL?1) and green peach aphid Myzus persicae (0.05‐0.5 a.i. μg mL?1). During one week after exposure to a 1 mL spray onto a 95 cm2 area in a Potter Spray Tower, M. sanborni was either more susceptible to B. bassiana or more sensitive to imidacloprid than M. persicae. The time–concentration–mortality (TCM) responses of each aphid species in each of five bioassays fit well to a TCM model, indicating a strong dependence of the fungal and chemical interactions on both concentration and post‐spray time. Adding imidacloprid to B. bassiana sprays at the rates of 0.025–0.05 μg mL?1 against M. sanborni or 0.1–0.5 μg mL?1 against M. persicae significantly enhanced or accelerated the fungal action. Based on the LC50 or LC90 estimates and their variances determined by the fitted TCM relationships for each aphid species, the relative potencies of an imidacloprid‐inclusive bioassay over those with B. bassiana alone or together with a lower sublethal rate ranged from a few to hundreds of times and varied over days after spray. These results suggest an alternative tactic for practical control of the aphid pests by a combined formulation or application of B. bassiana and imidacloprid and manage aphid resistance to the chemical insecticide.  相似文献

Aims: Producing granular cultures of obligate aphid pathogen Pandora nouryi for improved sporulation and storage. Methods and Results: Small millet–gel granules were made of the mixtures of 80–95% millet powder with 5–20% polymer gel (polyacrylamide, polyacrylate or acrylate‐acrylamide copolymer) and inoculated with mycelia at 30 mg biomass g?1 dry granules plus 87·5% water, followed by static incubation at 20°C for 4–12 days. The fungus grew well on 12 preparations but best on that including 10% copolymer. An 8‐day culture of this preparation discharged maximally 58·5 × 104 conidia mg?1 granule at 100% RH and was capable of ejecting conidia at the nonsaturated regimes of 86–97% RH. During storage at 6°C, granular cultures with >85% water content had twofold longevity (120 days) and half‐decline period (34–36 days) of those stored at room temperature. The steadily high water content preserved the cultures better than that decreasing at 6°C. However, conidia from 70‐day‐stored granules were less infective to Myzus persicae nymphs than those from fresh ones based on their LC50s. Conclusions: The millet–gel granules had higher sporulation capacity than reported Pandora cultures and a capability of spore discharge at nonsaturated humidity. Significance and Impact of the Study: The granular cultures are more useful for aphid control.  相似文献

In order to investigate virulence enhancement of entomopathogenic fungi, a Beauveria bassiana-sourced Pr1 protease (CDEP-1) was expressed by a methylotrophic yeast Pichia pastoris and then used as an additive to three gradient sprays of B. bassiana strain (Bb0062) onto apterous green peach aphid Myzus persicae adults in six bioassays. The resultant data fit well to a time–concentration–mortality model. Generally, the LC50 estimates of the fungal pathogen against the aphid species decreased with increasing CDEP-1 concentrations from 0 to 100 µg mL?1. The LC50s on days 5–7 after spray were reduced by 1.5–2.5-fold at the concentrations of 20–100 µg mL?1. However, sprays of 20–100 µg CDEP-1 mL?1 aqueous solution alone had no significant effect on aphid mortality compared to water spray only. Neither did inclusion of inactivated CDEP-1 at a concentration of 50 µg mL?1 affect significantly the fungal virulence to aphids. Our results confirm for the first time that the cuticle-degrading protease CDEP-1 enhanced fungal virulence due to acceleration of conidial germination and cuticle penetration. This suggests a new approach to utilising the protease in microbial control.  相似文献

1. Despite the ubiquity and abundance of water striders (Hemiptera: Gerridae) in temperate streams and rivers and their potential usefulness as sentinels in contaminant studies, little is known about their feeding ecology and lipid dynamics. 2. In this study we used stable isotopes of carbon (δ13C) and nitrogen (δ15N) and elemental carbon to nitrogen ratios (C/N) to assess dietary habits and lipid content, respectively, for water striders. 3. To determine diet‐tissue fractionation factors, nymphs of the most common species in New Brunswick, Canada, Aquarius remigis were reared in the laboratory for 73 days and exhibited rapid isotopic turnover in response to a switch in diet (C half‐life = 1.5 days, N half‐life = 7.8 days). Their lipid content increased towards the end of the growing season and resulted in lower δ13C values. Diet‐tissue fractionation factors were established after correction of δ13C data for the confounding effect of de novo lipid synthesis (strider δ13Cadj– diet δ13Cadj = 0.1‰, strider δ15N – diet δ15N = 2.7‰). 4. Water striders from the majority of 45 stream sites (83%) in New Brunswick had less than 50% contribution of aquatic carbon to their diets but showed a gradual increase in the contribution of this carbon source to their diet with increasing stream size. 5. These data indicate that striders exhibit a strong connection to terrestrial carbon sources, making them important users of energy subsidies to streams from the surrounding catchment. However, this dependence on terrestrial organic matter may limit their utility as indicators of contamination of aquatic systems by heavy metals and other pollutants.  相似文献

The enantioselectivities of individual enantiomers of furalaxyl in acute toxicity and bioaccumulation in the earthworm (Eisenia foetida) were studied. The acute toxicity was tested by filter paper contact test. After 48 h of exposure, the calculated LC50 values of the R‐form, rac‐form, and S‐form were 2.27, 2.08, and 1.22 µg cm‐2, respectively. After 72 h of exposure, the calculated LC50 values were 1.90, 1.54, and 1.00 µg cm‐2, respectively. Therefore, the acute toxicity of furalaxyl enantiomers was enantioselective. During the bioaccumulation experiment, the enantiomer fraction of furalaxyl in earthworm tissue was observed to deviate from 0.50 and maintained a range of 0.55–0.60; in other words, the bioaccumulation of furalaxyl was enantioselective in earthworm tissue with a preferential accumulation of S‐furalaxyl. The uptake kinetic of furalaxyl enantiomers fitted the first‐order kinetics well and the calculated kinetic parameters were consistent with the low accumulation efficiency. The comparative toxicity of five essential oil vapours was tested against four aphid species, the black bean aphid Aphis fabae, the pea aphid Acyrthosiphon pisum, the chrysanthemum aphid Macrosiphoniella sanborni, the green peach aphid Myzus persicae and on two of the most common coccinellid predators, the seven-spotted ladybird Coccinella septempunctata and the two-spotted ladybird Adalia bipunctata. All essential oils were highly toxic to the aphid species tested with LC50 and LC99 values ranging between 0.17 and 1.92 and 0.44 and 4.83 µL/L air, respectively, depending on the aphid species and on the essential oil. Coccinellid predators were also highly susceptible to the essential oil vapours and the selective toxicity ratio varied depending on aphid species, coccinellid predator and essential oil. The possibilities for the utililization of essential oils as aphicides, especially in IPM programmes in glasshouses are discussed with regard to the present findings.  相似文献

Abstract: Three questions regarding possible benefits of mixed diets for the specialist aphid predator, Coccinella septempunctata larvae were investigated. (1) Do aphids species from different host plants complement each other nutritionally? (2) Is a mixed diet of high‐quality aphids beneficial? (3) How does the quality of mixed diets depend on the quality of constituent species? All mix‐combinations of aphid species of high (Metopolophium dirhodum), intermediate (Myzus persicae), and poor food quality (Aphis sambuci), and the three single‐species diets were compared. A mixed diet of two high‐quality species (Sitobion avenae and M. dirhodum) was also compared with single‐species diets. Larvae that were given pure A. sambuci and a mixed diet of A. sambuci + M. persicae died within 18 days and none of the larvae developed to fourth instar. Metopolophium dirhodum was generally of higher quality as food than M. persicae, whereas the mixed diet of M. dirhodum + M. persicae was intermediate. Sitobion avenae and M. dirhodum were found to have approximately the same food value. Coccinella septempunctatam larvae that were offered a mixed diet of these two high‐quality aphids gained no extra advantage. Overall, no benefit from mixing of aphid species was found. The quality of mixed diets depended on the quality of the constituent species.  相似文献

To contribute the development of new products to control plant‐parasitic nematodes, 12 chalcone analogues were synthesized and screened for activity against Meloidogyne incognita. Three caused mortality greater than negative controls in second‐stage juvenile M. incognita, with values varying from 19.9% to 100%. The most active chalcone analogue was (1E,4E)‐1,5‐di(4‐nitrophenyl)‐2‐butylpenta‐1,4‐dien‐3‐one (compound 6 ), which had an LC50 value of 41 µg/ml. Under the same conditions, the commercial nematicide Carbofuran® (2,2‐dimethyl‐2,3‐dihydro‐1‐benzofuran‐7‐yl methylcarbamate) presented an LC50 equal to 101 µg/ml. When this chalcone analogue was applied to tomato plants infested with M. incognita, reductions in the numbers of galls and eggs of 51% and 68% were observed, respectively. According to in silico studies, the enzyme target of compound 6 in M. incongita is cytochrome P450, which is important for the oxidation of several substances in the nematode. Therefore, compound 6 is potentially useful for the development of new products to control M. incongita.  相似文献

Objective : Development of a model for the prediction of δ13Cprotein from δ13Ccollagen and Δ13Cap‐co. Model‐generated values could, in turn, serve as “consumer” inputs for multisource mixture modeling of paleodiet. Methods : Linear regression analysis of previously published controlled diet data facilitated the development of a mathematical model for predicting δ13Cprotein (and an experimentally generated error term) from isotopic data routinely generated during the analysis of osseous remains (δ13Cco and Δ13Cap‐co). Results : Regression analysis resulted in a two‐term linear model (δ13Cprotein (%) = (0.78 × δ13Cco) ? (0.58× Δ13Cap‐co) ? 4.7), possessing a high R‐value of 0.93 (r2 = 0.86, P < 0.01), and experimentally generated error terms of ±1.9% for any predicted individual value of δ13Cprotein. This model was tested using isotopic data from Formative Period individuals from northern Chile's Atacama Desert. Conclusions : The model presented here appears to hold significant potential for the prediction of the carbon isotope signature of dietary protein using only such data as is routinely generated in the course of stable isotope analysis of human osseous remains. Mannose‐binding lectins were shown to be useful in creating transgenic plants resistant to insects, including many phloem‐feeding Hemiptera. Before these plants can be used extensively, it is important to understand how these lectins exert their toxic effects on the target organisms. We investigated the feeding alterations induced by presenting the pea aphid, Acyrthosiphon pisum (Harris) (Hemiptera: Aphididae), with a diet containing the lectin from Canavalia ensiformis (ConA). A series of behavioural experiments were carried out to detect potential sensory mediation of lectin activity. Choice tests performed with a 400 µg ml?1 ConA diet (3.7 µm of native tetramer) showed that A. pisum quickly rejected the ConA diet, but that this reaction was not typical of a sensory‐mediated phagodeterrent effect. In addition, the aphids did not develop a conditioned taste aversion to the lectin. Diet uptake was evaluated using a radioactive tracer (14C‐methylated inulin), and showed depression of ingestion only after 16 h at 200 µg ml?1 or after 8 h at 400 µg ml?1 ConA. This effect was reversible under our test conditions. No evidence was obtained for early detection of the lectin, even by intoxicated aphids. An electrical penetration graph technique was adapted to artificial diets and provided short‐term continuous analysis on feeding/probing events. At the 400 µg ml?1 level, adults were affected and had reduced ingestion durations as early as in the first 4 h of contact, but experienced an adaptation to the behavioural alterations induced by lectin feeding. Overall, feeding deterrency following exposure to mannose lectins appeared to be a consequence of intoxication, and not due to a sensory mediated process.  相似文献

Understanding the ecological patterns of invasive species and their habitats require an understanding of the species’ foraging ecology. Stable carbon (δ13C) and nitrogen (δ15N) isotope values provide useful information into the study of animal ecology and evolution, since the isotope ratios of consumers reflect consumer's dietary patterns. Nevertheless, the lack of species‐ and element‐specific laboratory‐derived turnover rates could limit their application. Using a laboratory‐based dual stable isotope tracer approach (Na15NO3 and NaH13CO3), we evaluated the δ15N and δ13C isotope turnover rates in full‐grown adult invasive Limnomysis benedeni from Lake Constance. We provide δ15N and δ13C turnover rates based on nonlinear least‐squares regression and posterior linear regression models. Model precisions and fit were evaluated using Akaike's information criterion. Within a couple of days, the δ15N and δ13C of mysids began to change. Nevertheless, after about 14 days, L. benedeni did not reach equilibrium with their new isotope values. Since the experiment was conducted on adult subjects, it is evident that turnover was mainly influenced by metabolism (in contrast to growth). Unlike traditional dietary shifts, our laboratory‐based dual stable isotope tracer approach does not shift the experimental organisms into a new diet and avoids dietary effects on isotope values. Results confirm the application of isotopic tracers to label mysid subpopulations and could be used to reflect assimilation and turnover from the labeled dietary sources. Field‐based stable isotope studies often use isotopic mixing models commonly assuming diet‐tissue steady state. Unfortunately, in cases where the isotopic composition of the animal is not in equilibrium with its diet, this can lead to highly misleading conclusions. Thus, our laboratory‐based isotopic incorporation rates assist interpretation of the isotopic values from the field and provide a foundation for future research into using isotopic tracers to investigate invasion ecology.  相似文献

The stable isotope composition (δ13C and δ15N values) of harbor seals (Phoca vitulina) is influenced by their diet. Young‐of‐the‐year during lactation and postweaning fast are expected be enriched in 15N compared to foraging seals. We studied the temporal variation of stable isotope composition of young‐of‐the‐year and adults to determine from which point in time the young‐of‐the‐year tissues (i.e., muscles and vibrissae) are influenced by independent foraging only. These results were compared with the development of trophically transmitted parasitic infections. The δ15N values in young‐of‐the‐year muscles decreased from June (20.3‰ ± 0.5‰) to October (18.5‰ ± 0.4‰), while those of foraging seals were all year long below 19.2‰. This decrease coincides with the increase of parasitic infections in young‐of‐the‐year, reflecting a shift to fish diet. Together these results suggest that the muscles of the young‐of‐the‐year older than 5–6 mo reflect independent foraging and that they can therefore be used in community diet studies. The nursing signal in vibrissae was unclear, as the δ15N values of young‐of‐the‐year were stable over time, whereas those of adults varied seasonally. However, δ15N values of nursing pups were significantly higher than those of adults in May and June, maybe due to their reliance on milk.  相似文献

We report isotopic data (δ2H, δ18O n = 196; δ13C, δ15N n = 142; δ34S n = 85) from human hair and drinking water (δ2H, δ18O n = 67) collected across China, India, Mongolia, and Pakistan. Hair isotope ratios reflected the large environmental isotopic gradients and dietary differences. Geographic information was recorded in H and O and to a lesser extent, S isotopes. H and O data were entered into a recently developed model describing the relationship between the H and O isotope composition of human hair and drinking water in modern USA and pre‐globalized populations. This has anthropological and forensic applications including reconstructing environment and diet in modern and ancient human hair. However, it has not been applied to a modern population outside of the USA, where we expect different diet. Relationships between H and O isotope ratios in drinking water and hair of modern human populations in Asia were different to both modern USA and pre‐globalized populations. However, the Asian dataset was closer to the modern USA than to pre‐globalized populations. Model parameters suggested slightly higher consumption of locally producedfoods in our sampled population than modern USA residents, but lower than pre‐globalized populations. The degree of in vivo amino acid synthesis was comparable to both the modern USA and pre‐globalized populations. C isotope ratios reflected the predominantly C3‐based regional agriculture and C4 consumption in northernChina. C, N, and S isotope ratios supported marine food consumption in some coastal locales.   • 1 The mean supercooling points of first instar and adult Myzus persicae (Sulzer) maintained at 20°C and cooled at 1°C min?1 were ?26.6 and ?25.0°C respectively.
  • 2 The LT50 (temperature) of the same age groups drawn from the same population and cooled at the same rate were ?8.1 and ?6.9°C, indicating extensive pre-freeze mortality in M.persicae under laboratory conditions.
  • 3 Acclimation at 10 and 5°C did not affect supercooling but depressed the LT50 of both first instars and adult aphids.
  • 4 Freezing of leaves during feeding did not increase mortality above that expected from the direct effects of low temperature.
  • 5 The level of cold in different winters can be expressed in terms of the total number of frost days, and the frequency of abnormally cold days. Winter temperatures differ markedly in a vertical profile from the soil to the soil or grass surface, and then to the air (and foliage) above.
  • 6 The time of the first record of M.persicae in suction trap samples is correlated with January and February temperatures except in the west of England and Wales. Further north December and January temperatures are relatively more important.
  • 7 Winter temperatures and the resultant aphid mortality is a primary determinant of the timing of the spring migration.

Leaf respiration in the dark and its C isotopic composition (δ13CR) contain information about internal metabolic processes and respiratory substrates. δ13CR is known to be less negative compared to potential respiratory substrates, in particular shortly after darkening during light enhanced dark respiration (LEDR). This phenomenon might be driven by respiration of accumulated 13C‐enriched organic acids, however, studies simultaneously measuring δ13CR during LEDR and potential respiratory substrates are rare. We determined δ13CR and respiration rates (R) during LEDR, as well as δ13C and concentrations of potential respiratory substrates using compound‐specific isotope analyses. The measurements were conducted throughout the diel cycle in several plant species under different environmental conditions. δ13CR and R patterns during LEDR were strongly species‐specific and showed an initial peak, which was followed by a progressive decrease in both values. The species‐specific differences in δ13CR and R during LEDR may be partially explained by the isotopic composition of organic acids (e.g., oxalate, isocitrate, quinate, shikimate, malate), which were 13C‐enriched compared to other respiratory substrates (e.g., sugars and amino acids). However, the diel variations in both δ13C and concentrations of the organic acids were generally low. Thus, additional factors such as the heterogeneous isotope distribution in organic acids and the relative contribution of the organic acids to respiration are required to explain the strong 13C enrichment in leaf dark‐respired CO2.  相似文献

The terrestrial carbon cycle is influenced by environmental variability at scales ranging from diurnal to interannual. Here, we present 5‐years of growing season (day 131–275) observations of the carbon isotope ratio of ecosystem respiration (δ13CR) from a semiarid woodland. This ecosystem has a large necromass component resulting from 97%Pinus edulis mortality in 2002, is dominated by drought‐tolerant Juniperus monosperma trees, and experiences large variability in the timing and intensity of seasonal and synoptic water availability. Mean growing season δ13CR was remarkably invariant (?23.57±0.4‰), with the exception of particularly enriched δ13CR in 2006 following a winter with anomalously low snowfall. δ13CR was strongly coupled to climate during premonsoon periods (~May to June), including fast (≤2 days) responses to changes in crown‐level stomatal conductance (Gc) and vapor pressure deficit (vpd) following rain pulses. In contrast, δ13CR was relatively decoupled from Gc and environmental drivers during monsoon and postmonsoon periods (July–August and September, respectively), exhibiting only infrequent couplings of δ13CR to vpd and soil water content (SWC) with longer lags (~8 days) and variable response slopes (both positive and negative). Notably, δ13CR exhibited consistent dynamics after rainfall events, with depleted δ13CR occurring within 1 h, progressive hourly δ13CR enrichment over the remainder of the night, and net δ13CR depletions over the multiple nights postevent in monsoon and postmonsoon periods. Overall this ecosystem demonstrated strong dependence of δ13CR on precipitation, with an apparent dominance by the autotrophic δ13C signal in premonsoon periods when deep soil moisture is abundant and surface soil moisture is low, and weaker coupling during monsoonal periods consistent with increasing heterotrophic dominance when deep soil moisture has declined and surface moisture is variable.  相似文献

Stable isotope ratios (δ13C and δ15N) were analyzed from the bone collagen of individuals (n = 8) from a Lapita burial ground (ca. 2800–2350 BP) on Watom Island, located off northeast New Britain in the Bismarck Archipelago. The aim of this study was to assess the diet and subsistence strategies of humans that lived during the later Lapita period in Near Oceania. To aid in the interpretation of the human diet we analyzed the stable isotope ratios of faunal material from the site (n = 27). We also aim to assess methods of animal husbandry at the site over time from an analysis of the stable isotope ratios (δ13C and δ15N) of pig bones (n = 22) from different temporal periods (Lapita, post‐Lapita, and late prehistoric). The protein diet of the humans consisted of marine organisms from the inshore environment and some deep‐water species, most likely marine turtle, in addition to higher trophic level terrestrial foods, likely pig and native animals (e.g., fruit bat, Cuscus and bandicoot). Although the sample sizes were small, females (n = 4) displayed more variable δ13C and δ15N values compared with males (n = 4), which may be associated with the movement of adult females to the island. The stable isotope analysis of the pig bones indicated that there were few differences between the diets of the pigs from the Lapita and post‐Lapita layers, suggesting that the method of pig husbandry was similar between these two periods and was likely relatively free‐range.
