Reproduction in Fundulus heteroclitus is adapted to the temperateclimate and the tidal environment of the coastal marshes whichare its primary habitat. Egg deposition and spawning are closelylinked to the tidal cycle. Concentration of spawning on springtides gives the fish access to sites in the high intertidalzone. Eggs are deposited near the high water mark, usually insand in New England populations and in Spartina alternifloraor empty Geukensia demissa shells in Middle Atlantic and southernpopulations. Aerial incubation of eggs appears to be essentialfor their survival in the salt marsh. Loss of eggs due to lackof fertilization, mortality during development and predationwas always less than 30 percent in our observations. Hatchingoccurs only when the eggs are inundated, usually on spring tides.An endogenous semilunar gonadal cycle appears to be involvedin control of spawning, but an effective environmental synchronizerhas not been found. Cyclic changes in estradiol-17ßaccompany the gonadal cycle, while spawning behavior is stimulatedby neurohypophyseal hormones. Both temperature and photoperiodhave been implicated in the control of seasonal reproductionin F. heteroclitus. Early workers focused on males and concludedthat increasing temperature was the stimulus that initiatedgonadal recrudescence in spring. These experiments were notrigorous tests of the effects of photoperiod. We have shownthat in females both warm temperatures and long photoperiodsare involved. In female F. heteroclitus, as in most mammalsand birds, it is the timing rather than the duration of thelight exposure which determines its effectiveness in stimulatinggonadal maturation. Maintenance of gonadal maturity in femaleF. heteroclitus depends on long photoperiods, but the ovarymay become refractory and regress even when stimulatory photoperiodsare sustained in the laboratory. Ovarian maturity can be stimulatedwith gonadotropin injection in refractory animals, indicatingthat the brain-pituitary axis is the site of refractoriness.The pathway by which light influences ovarian maturity may involvean encephalic photoreceptor, since neither the eyes nor thepineal gland is necessary.  相似文献

The common mummichog, Fundulus heteroclitus, functions as bothpredator and prey in the trophic structure of east coast tidalmarshes. Although mummichogs are generally considered importantto energy transformations within marshes, few studies have convincinglydemonstrated that predation by F. heteroclitus affects the abundanceof salt marsh benthic invertebrates. Thus far investigationsof this type have dealt only with the direct effects of adultmummichogs. The results of recent experiments have suggestedthat by controlling smaller predators, mummichogs may indirectlyhave a positive effect on the densities of some infaunal marshinvertebrates. Our current knowledge of larval and juvenilemummichogs in their natural habitat is minimal. Unlike the adults,which can utilize the intertidal zone only when it is flooded,the young remain on the marsh even at low tide, inhabiting shallowpuddles of residual tidal water that form between clumps ofvegetation and around fiddler crab (Uca sp.) burrows. The importanceof F. heteroclitus in salt marsh communities will remain incompletelyunderstood unless future studies consider the role of larvaland juvenile mummichogs. Although many species of fishes andwading birds feed on mummichogs, the blue crab (Callinectessapidus) is probably the major predator of adult F. heteroclitusin the intertidal salt marsh. Predation by adult mummichogsand xanthid crabs (e.g., Eurytium limosum) may contribute tothe high mortality of larval and juvenile Fundulus.  相似文献

A brief account of some of the influences that environmentalfactors, such as the lunar cycle, daylength and temperature,may have on thyroid activity in certain salmonids and the killifish,Fundulus heteroclitus, is presented. Four sympatric species of Fundulus (F. heteroclitus, F. majalis,F. diaphanus, and F. luciae) are distributed along tidal heightand salinity gradients such that F. heteroclitus co-occurs witheach species, while the remaining species rarely occur together.Feeding habitats of all the species are similar, and food hasbeen shown to limit population size of F. heteroclitus. Thispaper examines whether competition is an important structuringforce within this guild by addressing two questions: 1) is thespatial separation exhibited by three of the species due tophysiological barriers or due to competitive exlusion? and 2)when F. heteroclitus occurs with other Fundulus species doescompetition for resources take place? Laboratory studies indicate that all four species are tolerantof a wide range of environmental conditions; available evidencesuggests that physiological barriers are an unlikely explanationfor spatial segregation among some members of this guild. Competitiveexclusion seems a likely alternate explanation, but is supportedonly by studies of diet overlap. Similarly, only inference fromdiet overlap is available to answer the second question. A fieldexperiment is presented here in which F. heteroclitus and F.majalis were placed in enclosures separately and together. Competitionbetween these species appears to be important, and similar fieldexperiments are recommended to investigate competitive interactionsamong other species pairs within the genus  相似文献

Salt marshes and similar tide-dominated habitats present anunusual challenge for reproduction of resident aquatic organisms.Strong currents, siltation and hypoxia can potentially contributeto reproductive failure through mortality of the eggs or flushingof the eggs and larvae from the habitat. Fundulus heteroclitus,a small brackish water killifish, is a common resident of tidalmarshes along the east coast of North America from Newfoundlandto Florida. The reproductive strategy of this and related speciesis based on aerial incubation of eggs in the high intertidalzone. The eggs are resistant to desiccation and, when fullydeveloped, hatch on immersion. Copulatory behavior and anatomyof accessory reproductive structures are adapted to placementof eggs in protected incubation sites. The gonads of both malesand females mature rhythmically with an endogenous circasemilunarperiod, which is synchronized with the "spring" tides of newand full moons. Spawning occurs on high tides. Embryos developin 9–15 days, and usually hatch on the succeeding springtide series. Reproductive cyclicity in F. heteroclitus and relatedfishes ensures that spawning fish will have access to the highintertidal zone, thereby permitting aerial incubation of eggs.  相似文献

Teleosts, the bony fishes, inhabit both freshwater and seawaterenvironments. Some euryhaline fish, such as Fundulus heteroclitus,alternate between the two milieux several times daily. Regardlessof adaptation, the gills of these animals possess a highly specializedcell type called the chloride cell. This cell contains numerousmitochondria and exhibits a greatly amplified basolateral cellsurface richly endowed with Na,K-ATPase. Recent studies on isolatedopercular epithelia containing chloride cells have demonstratedactive chloride secretion and passive transepithelial sodiummovements, and have established the chloride secretory roleof this cell type in seawater-adapted teleosts. Current modelssuggest that chloride transport occurs via a transcellular route.Seawater chloride cells exist in multicellular units and sharesimple, shallow tight junctions which are thought to be theroute for passive sodium movement. Freshwater chloride cells,whose function remains to be elucidated, are generally describedas existing in a unicellular configuration. However, recentobservations in Fundulus heteroclitus adapted to salinitiesas low as 1% sea water reveal that chloride cells persist inmulticellular complexes with apical crypts. The killifish,Fundulus heteroclitus, is a euryhalineteleost fish capable of adapting rapidly to transfer from freshwater (FW) to four times seawater (SW). To investigate osmoregulation at amolecular level, a 5.7-kilobase cDNA homologous to human cysticfibrosis transmembrane conductance regulator (hCFTR) was isolated froma gill cDNA library from SW-adapted killifish. This cDNA encodes aprotein product (kfCFTR) that is 59% identical to hCFTR,the most divergent form of CFTR characterized to date. Expression ofkfCFTR in Xenopus oocytes generatedadenosine 3',5'-cyclic monophosphate-activated,Cl-selective currentssimilar to those generated by hCFTR. In SW-adapted killifish,kfCFTR was expressed at high levels in the gill, opercular epithelium, and intestine. After abrupt exposure of FW-adapted killifish to SW, kfCFTR expression in the gill increasedseveralfold, suggesting a role for kfCFTR in salinity adaptation. Undersimilar conditions, plasma Na+levels rose significantly after 8 h and then fell, although it is notknown whether these changes are directly responsible for the changes inkfCFTR expression. The killifish provides a unique opportunity to understand teleost osmoregulation and the role of CFTR.

Recent studies have shown distinct patterns of variation inthe egg morphology (oil droplet number, diameter and densityof chorionic filaments) of Fundulus heteroclitus along the eastcoast (including Chesapeake and Delaware bays). We tested thehypothesis that variation in adult morphology and in electromorphsis concordant with that of egg morphology over the same rangeby subjecting data for these character sets to the Mantel test.This analysis was based on data for three egg characters, nineteenadult morphological (meristic and morphometric) characters andfive protein characters from locations along the Atlantic coast.If the geographical differences observed among locations reflectgenetic differences between two forms that have establishedsecondary contact after isolation, we would expect general concordancebetween the different character sets. We would expect littleconcordance between the sets if they are each affected by differentselectional and/or developmental responses to environmentalregimes. Our analyses indicate that patterns of variation amonglocalities for meristic, egg, and genetic character sets areall concordant, and thus the observed differences among locationsare likely the result of secondary intergradation between twopreviously isolated forms. These conclusions are supported bysimilar patterns of geographical variation in spawning sitepreference and body coloration. Thus, these results supportour earlier suggestion that these morphs should be treated asseparate taxa, with F. h. heteroclitus occurring along the eastcoast from New Jersey south to Florida including lower Chesapeakeand Delaware bays and F. h. macrolepidotus distributed fromConnecticut north to Newfoundland with disjunct populationsin upper Chesapeake and Delaware bays. Studies of 16 polymorphic loci in the fish Fundulus heteroclitushave uncovered significant directional changes in gene frequencieswith latitude (i.e., clines). These spatial patterns could havearisen by primary and/or secondary intergradation. While wecannot presently distinguish between these two models, mitochondrialDNA analyses indicate that if previous isolation occurred asrequired for secondary intergradation, it must have been relativelyrecent and of short duration. Herein we discuss the roles ofgenetic drift, random migration, nonrandom migration, selectionand others as potential driving forces for both modes of clineformation. In addition, we address the potential role of thelast glacial period in (1) creating isolating barriers, (2)the opening of unoccupied habitats for range extension, and(3) as a mechanism for thermal selection. While some evidenceexists that at least one of the driving forces is selection,the extent to which it and other deterministic forces participateas opposed to various stochastic processes must await furtheranalysis  相似文献

Fundulus heteroclitus, the mummichog or Atlantic killifish, is the dominant small-bodied fish species of the east coast estuaries and salt marshes of Canada and the USA, where it is present as two subspecies, the northern F. h. macrolepidotus and the southern F. h. heteroclitus. Recently identified as the premier teleost model in environmental biology, the species has long been of value in understanding evolved tolerance to toxicants and more lately in adding to our knowledge about reproductive effects of environmental endocrine disruptors. The body of literature on F. heteroclitus ovarian physiology and reproduction, from both field and laboratory studies, provides the foundation for present work focused on understanding the reproductive effects and modes of action of environmental toxicants. In this paper, we review the environmental and endocrine factors controlling ovarian and reproductive cycling in F. heteroclitus, noting specifics related to field and laboratory studies on the two subspecies as well as key research gaps compared to other fish species. We also summarize recent development of methodologies to study the effects of environmental contaminants on endocrine signalling and egg production in F. heteroclitus. Continued efforts to progress both our fundamental understanding of reproductive physiology in mummichog, coupled with studies focused on the modes of action of environmental contaminants, have high potential to further develop this teleost model. While the model may presently lag behind those based on other species of fish, the unique biochemical and physiological adaptations which allow F. heteroclitus to adapt to changing environmental and toxic conditions provide a valuable experimental system for comparative physiologists, ecotoxicologists and evolutionary biologists.  相似文献

A morphological basis for osmoregulation in the teleosts was studied by comparing the fine structure of chloride cells found in epithelia of the gills of three species of fish: Fundulus heteroclitus which can survive in a wide range of salinities, and F. similis and F. chrysotus which are usually restricted to salt water and fresh water environments, respectively. Gills were removed from F. heteroclitus which had been laboratory adapted to either sea water or pond water. For a comparison, gills were also removed from the marine F. similis and the fresh water F. chrysotus which had been adapted to their natural environments. Gill-filaments were fixed in Millonig's phosphate buffered (pH 7.4), 1 per cent osmium tetroxide and were embedded in Epon. Thin sections of filaments were stained with lead hydroxide. The cytoplasm of chloride cells of all three species of Fundulus is heavily populated with mitochondria and is filled with tubules of the agranular endoplasmic reticulum (ER). An orderly secretory cycle was indicated for chloride cells of salt water adapted F. heteroclitus and the marine F. similis. An amorphous material is observed in the agranular ER. Its density increases towards the apical end of the cell. In the apical cytoplasm, tubules of the agranular ER appear to converge and to discharge the amorphous material into an apical cavity. Except for the actual opening of the apical cavity, the distal end of salt water adapted chloride cells is characteristically shielded from the hypertonic environment by thin cytoplasmic flanges projecting from the neighboring epithelial cells. Chloride cells of the fresh water F. chrysotus resemble chloride cells of pond water adapted F. heteroclitus, in that these cells do not have apical cavities with the functional appearance of those in the sea water adapted forms. The distal end of fresh water adapted chloride cells is typically exposed to the free surface of the gill-filament. The possible function of the cell type is discussed.  相似文献

Recently, the fruit fly Drosophila melanogaster has been introducedas a model system to study the molecular bases of a varietyof ethanol-induced behaviors. It became immediately apparentthat the behavioral changes elicited by acute ethanol exposureare remarkably similar in flies and mammals. Flies show signsof acute intoxication, which range from locomotor stimulationat low doses to complete sedation at higher doses and they developtolerance upon intermittent ethanol exposure. Genetic screensfor mutants with altered responsiveness to ethanol have beencarried out and a few of the disrupted genes have been identified.This analysis, while still in its early stages, has alreadyrevealed some surprising molecular parallels with mammals. Theavailability of powerful tools for genetic manipulation in Drosophila,together with the high degree of conservation at the genomiclevel, make Drosophila a promising model organism to study themechanism by which ethanol regulates behavior and the mechanismsunderlying the organism's adaptation to long-term ethanol exposure.  相似文献

Social monogamy without biparental care has evolved in manytaxa, and a number of hypotheses have been developed to explainthis phenomenon. Several authors have suggested the importanceof male mate-guarding behavior in the evolution of social monogamy,although empirical support for this hypothesis is lacking. Inthe caridean shrimp genus Alpheus, social monogamy may resultfrom selection on males for long-term guarding of females becausemating is temporally restricted to a short time after the female'smolt. I used Alpheus angulatus to test two predictions of theextended mate-guarding hypothesis: Males should (1) be physiologicallycapable of predicting the timing of female sexual receptivity,and (2) prefer to associate with (guard) females that are closerto sexual receptivity. Data from a Y-maze experiment testingfor distance chemical communication showed that males of A.angulatus were attracted to water treated by exposure to premoltfemales, repulsed by water treated by exposure to intermoltmales and females, and did not appear to respond in either directionto water treated by exposure to premolt males. In mate choiceexperiments, significantly more males paired with premolt femalesthan with postmolt females. These data suggest that males ofA. angulatus engage in precopulatory mate-guarding behavior.Other factors (population density, sex ratio) may have playeda role in the temporal extension of mate guarding to socialmonogamy.  相似文献

The coccolith-bearing phase of Pleurochrysis scherffelii hasbeen examined with the electron microscope for the first time.Coccolith and scale structure, haptonema morphology, and themicroanatomy of the protoplast have been recorded and shownto agree closely with those of Hymenomonas carterae and theHymenomonas stage of Apistonema previously investigated by thepresent author. Comparative observations have been made on thevegetative phase and non-coccolith-bearing swarmers of Pleurochrysisscherffelii and these differ significantly in scale morphologyfrom the coccolith-bearing phase although the haptonema of theswarmers is similar. Certain developmental observations havebeen included in the investigation and the taxonomic significanceof the findings is discussed in a preliminary way.  相似文献

The results of pilot experiments lead to the conclusion thatD. glomerata exhibits a number of developmental stages: firstly,a juvenile stage during which the plant is insensitive to environmentalconditions which later stimulate flowering; secondly, an inductivestage, when the plant responds to periodic exposure to darknessat the conclusion of which it is fully induced or ripe to flower,and finally, a post-inductive stage during which inflorescencesare initiated and undergo further development; these are long-dayprocesses. In four populations studied the juvenile stage lasts about fiveweeks. In north European material daily exposure to seven hoursof darkness is near the minimum for induction although thereis considerable within-population variation. Further, it appearsthat the daily dark requirement becomes less as the plant ages. Comparisons are made of the flowering behaviour of D. glomerataand Lolium perenne. The differences between these species resultfrom the presence of a juvenile stage in Dactylis and the possibilityof satisfying its inductive requirement by long days. Inductionin Lolium requires short days or low temperature. The significance of these results is discussed in the lightof previous work on the environmental control of flowering inherbage grasses. The existence of three developmental stagescan explain the wide differences in interpretation of the floweringrequirements of Dactylis previously held. 1. The euryhaline fish Fundulus heteroclitus has an incipient lethal pH between 3.75 and 4.0 in fresh water.2. Fish exposed to pH 3.5 in sea water or fresh water died in about 3 hr, and had greatly elevated or depressed body sodium concentrations, respectively. The direction and degree of change in body sodium level depended on the sodium diffusion gradient between the environment and the fish. This is the first time that the death of fish in sea water at low pH has been shown to be associated with hypernatremia.3. Yet, sodium fluxes during the first hour of exposure to pH 3.5 in water of 3.5 or 35 ppt salinity were not different from controls, and body and plasma sodium concentration did not change during 2hr exposure to pH 3.5. This initial insensitivity of gill sodium regulation to blockage by low pH is quite different from the response of previously studied freshwater fish.4. The degree of acid tolerance displayed by F. heteroclitus is surprising considering its estuarine habits. This paradoxical tolerance appears to be a secondary consequence of its ability to adjust sodium balance in relation to rapid changes in salinity.  相似文献

Following a single acute exposure to chlorine gas, persistenteffects on epicuticular waxes, cuticular transpiration, treegrowth and mortality were studied in foliage of Pinus ponderosaand Pseudotsuga menziesii for three growing seasons. Chlorinegas exposure caused foliar injury to both exposed foliage andfoliage that flushed after exposure (P < 0.05). The tendencyto form films of water rather than droplets was greater in directlyexposed foliage (P < 0.001). Rates of cuticular transpirationwere higher for directly and indirectly exposed foliage of Pinusponderosa up to 1 year after exposure and up to 6 months afterexposure for directly exposed Pseudotsuga menziesii(P < 0.001),after which P. menziesii needles defoliated. Total water content(TWC) and relative water content were significantly correlatedwith foliar injury (P < 0.05). TWC was lower for directlyexposed foliage up to 1 year after exposure (P < 0.001).There was no persistent negative effect on Fv/Fm ratios after1 year. Exposure to chlorine gas did not affect needle lengthor annual shoot increment growth, but exposure was correlatedwith increased bud production. Needle longevity of foliage thatflushed 2 months after exposure was reduced significantly (P< 0.001). Annual stem increment growth for both species decreasedover at least three growing seasons following chlorine gas exposure(P < 0.001), and depended on distance from the spill site.Cone production was lower for exposed Pinus ponderosa treescompared to controls (P < 0.05), and tree mortality was higherwithin approx. 50 m of the release site forPseudotsuga menziesii. Growth responses for both conifers agreed well with predictedpatterns of carbon allocation after defoliation caused by chlorinegas exposure. Four cDNAs for spermidine synthase (SPDS), which converts thediamine putrescine to the higher polyamine spermidine usingdecarboxylated S-adenosylmethionine as the co-factor, were isolatedfrom Nicotiana sylvestris, Hyoscyamus niger, and Arabidopsisthaliana. When the N. sylvestris SPDS cDNA was expressed ina SPDS-deficient E. coli mutant, the recombinant protein showedhigh SPDS activity, but did not have any spermine synthase activity.The plant SPDSs have molecular masses of about 34 kDa, possessthe co-factor binding motifs which have been proposed for S-adenosylmethionine,and are more homologous in amino acid sequence to tobacco putrescineN-methyltransferase (PMT) than to SPDSs from mammals and E.coli. The SPDS gene is expressed in root, stem, and leaf inN. sylvestris, whereas the PMT gene is expressed only in root.The potential evolution of plant SPDS and PMT, and their evolutionaryrelationships with animal SPDS are discussed. The effect of ultraviolet A (UVA) on growth and photosyntheticrate was studied in diatoms (Melosira spp.) of the phytoplanktonof a eutrophic lake and a cultured green alga Chloretla ellipsoidea.The cells were incubated under photosynthetically active radiation(PAR) (–UVA) or PAR + UVA conditions (+UVA). Growth ofC.ellipsoidea was retarded under +UVA, as shown by an increasein the lag period, but the rate of exponential growth was almostthe same in + and –UVA conditions. The photosyntheticrate was depressed markedly by UVA in Chlorella cells grownunder –UVA. In contrast, cells grown in +UVA showed onlyslight inhibition by UVA and after exposure to UVA for 6 daysthere was no inhibition. During the growth experiment, the cellularchlorophyll a content was higher in +UVA than +UVA grown cells.A similar effect was observed in diatoms from the eutrophicLake Suwa. In vivo fluorescence with (Fa) and without 3-(3,4-dichloropheny)-l,l-dimethylurea (DCMU) (Fb) and the photosynthetic rate were measured forC.ellipsoidea and the diatoms for 5 h under + and –UVAconditions. Soon after C.ellipsoidea had been subjected to +UVA,Fb and Fa / Fb decreased quickly and reached minima after 40min and 1 h, respectively. The suppressed in vivo fluorescenceresumed and full recovery was achieved after 4 h. This suggeststhat reactivation of the photosystem is acquired under prolongedexposure to UVA. A similar shift of Fa + Fb, but no change inFb, was found in diatoms by exposure to UVA. Changes in photosyntheticoxygen evolution by C.ellipsoidea under +UVA were similar tochanges in Fa + Fb. Degradation of chlorophyll a extracted inmethanol was enhanced by UVA. The rate of degradation by UVAwas independent of temperature from 15 to 34°C, suggestinga photochemical reaction. The results indicate that C.ellipsoideaand Melosira spp. acclimatize to prolonged UVA exposure by reactivationof the photosystem and enhanced cellular chlorophyll a synthesis.The ecological importance of these results to phytoplanktonproductivity in natural aquatic environments is discussed.  相似文献
