The neuronal organization of the lateral basilar region (LBR) of gray matter in the cervical portion of the cat spinal cord was studied by light and electron microscopy. It was found that LBR neurons form a homogeneous group with regard to the size of their soma. The ordinary pale ultrastructure of the cytoplasm is found in 96.8% of neurons examined. The ultrastructure of the cytoplasm of the small cells (3.2%) is dark and their matrix has high electron density. Most endings on LBR neurons have spherical vesicles (of the S-type). Endings with flattened vesicles (F-type) are next in order of numerical frequency. In some endings, besides the ordinary synaptic vesicles, there are other vesicles with an osmiophilic center, and endings with a dense matrix and numerous spherical vesicles. Endings of the F-type are relatively more numerous on dendrites of LBR neurons than on their soma. Axodendritic synapses form 87.8% of the synaptic connections of the LBR, and axo-somatic synapses 9.2%. The few axo-axonal synapses are formed by small endings with small synaptic vesicles and large plaques with spherical vesicles. The latter frequently make contact with several dendrites simultaneously. The functional role of the various neuronal structures of LBR in the transmission of descending and afferent influences is discussed.A. A. Summary After in vivo njections of 3H-glycine into the cat spinal cord autoradiography at the light and electron microscopic level revealed high concentrations of radioactivity over certain nerve endings, often containing flat vesicles, over glial cells and over neuronal cell bodies probably representing spinal interneurons. Microelectrode discharges of potentials have been realized from segmentary interneurons of the dorsal horn and intermediate nucleus of the spinal cord in cat at the L6–L7 level by electrical stimulation of the sensorimotor region of the brain cortex. It has been established that corticifugal influences on segmentary interneurons of the system of the flexor reflex and on neurons activated by high threshold muscle afferents (groups Ib, II, and III), or high threshold cutaneous afferents are predominantly excitatory. Interneurons activated by muscle afferents of group Ia or by the lowest threshold cutaneous fibers are weakly subjected to pyramidal influences. The mean latencies of excitatory postsynaptic potentials (EPSP's) and discharges evoked under the influence of pyramidal volley, for the neurons under study in the system of afferents of the flexor reflex are equal to 11.8±2.6 and 20.1±1.8 msec, respectively; for interneurons, excited only by high threshold muscle afferents, they are equal to 15.5±3.6 and 16.3±2.2 msec, respectively; and for interneurons, excited by high threshold cutaneous fibers they are equal to 11.8±2.6 and 18.3±1.4 msec, respectively. Templates of the membrane potential profiles from lateral (LI) interneurons and motoneurons during glutamate- and N-methyl-D-aspartate (NMDA)-induced fictive locomotion showed pronounced plateau phases. In contrast, crossed caudal (CC) interneurons had a less obvious and steeper plateau region that was followed by a clear notch coinciding with the end of the lateral interneuron plateau phase. In experiments on spinal cats, when branches of the pelvic nerve innervating the rectum were electrically stimulated at the same time as the peroneal nerve it was found that 12 out of 20 neurons, to which the effects of both these supplies converged, were activated by both A- and C-fibers. Responses to stimulation of the pelvic nerve were apparently mediated via fibers with a conduction velocity not less than 2.2 m·s–1. In studies in spinal rats it was shown that distension of the more distal regions of the large intestine could excite neurons in laminae IV–V, and inhibit neurons in deeper laminae. In seven out of 18 cases the inhibition evoked by visceral stimulation was due to a direct effect on the postsynaptic membrane of these cells, and in 11 cases it was localized to presynaptic structures. Naloxone, strychnine, and atropine did not block this inhibition, thus providing evidence against the possible participation of opioids, glycine, and acetylcholine in its generation. Phaclofen, a GABAB-receptor antagonist, was also without effect, but bicuculline suppressed this inhibition in three out of 12 cases, indicating that GABAA-receptors are involved.I. M. Calcium binding proteins (CBPs) regulate intracellular levels of calcium (Ca2+) ions. CBPs are particularly interesting from a morphological standpoint, because they are differentially expressed in certain sub-populations of cells in the nervous system of various species of vertebrate animals. However, knowledge on the cellular regulation governing such cell-specific CBP expression is still incomplete. In this work on the L7 segment of the cat spinal cord, we analyzed the localization and morphology of neurons expressing the CBPs calbindin-28 KD (CB), parvalbumin (PV), and calretinin (CR), and co-expressing CB and PV, CB and CR, and PV and CR. Single CBP-positive (+) neurons showed specific distributions: (1) CB was present in small neurons localized in laminae I, II, III and X, in small to medium size neurons in laminae III–VI, and in medium to large neurons in laminae VI–VIII; (2) PV was present in small size neurons in laminae III and IV and in medial portions of laminae V and VI, medium neurons and in lamina X at the border with lamina VII, in medium to large neurons in laminae VII and VIII; (3) CR labeling was detected in small size neurons in laminae I, II, III and VIII, in medium to large size neurons in laminae I and III–VII, and in small to medium size neurons in lamina X. Double labeled neurons were a small minority of the CBP+ cells. Co-expression of CB and PV was seen in 1 to 2% of the CBP+ cells, and they were detected in the ventral and intermediate portions of lamina VII and in lamina X. Co-localization of CB and CR was present in 0.3% of the cells and these cells were localized in lamina II. Double labeling for PV and CR occurred in 6% of the cells, and the cells were localized in ventral part of lamina VII and in lamina VIII. Overall, these results revealed distinct and reproducible patterns of localization of the neurons expressing single CBPs and co-expressing two of them. Distinct differences of CBP expression between cat and other species are discussed. Possible relations between the cat L7 neurons expressing different CBPs with the neurons previously analyzed in cat and other animals are suggested.  相似文献

Synaptic responses evoked in propriospinal neurons of the upper lumbar segments (L3–L4) by reticulo-, vestibulo-, and corticospinal impulses were studied in experiments on cats and monkeys. Propriospinal cells, identified by antidromic stimulation, were stained with Procion red, so that they could be localized in the different zones of the ventral horn. Monosynaptic reticular and vestibular excitatory influences were discovered in cats; convergence of these influences on the same neurons was demonstrated. In monkeys bulbospinal monosynaptic effects were supplemented by monosynaptic influences arriving from the motor cortex; convergence of monosynaptic excitatory influences from all supraspinal sources studied was found on some propriospinal neurons. It is shown by the use of Golgi's method in Antonova's modification that the neuronal structure of the periaqueductal gray matter (PGM) in the frontal plane is characterized by the presence of small and medium-sized cells of "reticular type," which can be subdivided into three types: fusiform, triangular, and multipolar. On the basis of the visual distribution of these types of neurons and also of statistical analysis of 800 identified neurons, two regions can be distinguished: medial, directly surrounding the aqueduct of Sylvius, containing small neurons, among which the fusiform kind predominate significantly (P<0.001), and a lateral region with larger neurons, with significantly (P<0.001) more triangular cells. Neurons in the medial region show a characteristic and strong (P<0.001) tendency for their dendrites to be oriented toward the lumen of the aqueduct, and through them the physiologically active substances of the CSF may influence the functional activity of neurons of PGM.Central Research Institute of Reflex Therapy, Moscow City Council Main Health Board, Moscow. Cord dorsum potentials were recorded in the acute experiments on cats. The relationship between the N-component of the cord dorsum potentials and the strength of stimulation of the big femoral nerve was studied. After the transsection of the half of midbrain tegmentum the increased curve slope indicating the rise in cord interneuron excitability was mainly observed on the contralateral side. The same results were observed after the removal of the contralateral somatosensory cortex or contralateral section of midbrain basis. The data indicate the tonic inhibitory influence of cortical structures on cord interneurons, which participate in the transmission of somatosensory signals.  相似文献

The latest research reveals that nitric oxide as a gas messenger may diffuse into the surrounding extracellular fluid and act locally upon neighboring target cells. However, several observations raise the possibility that nitric oxide may also be released at a greater distance from the neuronal cell body. The catalytic nitric oxide synthase (cNOS) activity was therefore studied in the cervicothoracic and lumbosacral segments of the spinal cord of rabbits, including the white matter of dorsal columns (DC), lateral columns (LC) and ventral columns (VC), as well as the gray matter of dorsal horns (DH), intermediate zone (IZ) and ventral horns (VH). Lower cNOS activity was found in the white matter of both cervicothoracic (47%) and lumbosacral (30%) regions, whereas that detected in the gray matter of the lumbosacral part of the spinal cord was considerably higher (70%). Enzyme activity varied from 43.4 to 77.2 dpm/microg protein in the cervicothoracic segments of the gray matter in the descending order: DH>VH>IZ. Similar cNOS activity was found in the white matter of the cervicothoracic segments (42.1-62.8 dpm/microg protein). When the activity of cNOS was compared in the lumbosacral segments, the highest enzyme activity was found in DH of the gray matter (198.7 dpm/microg protein) and the lowest cNOS in DC (45.8 dpm/microg protein) of the white matter. It was concluded that the white matter of the spinal cord contains similar cNOS activity in comparison to the gray matter.  相似文献

Interneurons of the lumbar division of the cat spinal cord responding after a short latent period with intensive excitation to stimulation of the medullary pyramids and red nucleus but not responding (or excited after a long latent period) to stimulation of peripheral nerves were investigated by microelectrode recording. Most of these neurons, located in the lateral zones of Rexed's laminae IV–VII of the gray matter, were identified as propriospinal cells sending axons into the dorsolateral funiculus of the white matter (mean velocity of antidromic conduction in the group 34.6 m/sec). Marked convergence of corticofugal and rubrofugal excitatory influences was found on the overwhelming majority of neurons. Some neurons were activated monosynaptically by fast-conducting fibers of both descending systems. The minimal and mean values of the latent periods of the pyramidal EPSPs for the neurons tested were 4.5 and 6.28 msec, and for the rubral EPSPs 3.3 and 4.94 msec respectively. A distinguishing feature of the activation of these neurons is the intensive potentiation of their synaptic action on the arrival of a series of corticofugal and rubrofugal waves.A. A. Stereotaxic microinjections of horseradish peroxidase (HP) were made into different parts of the rostral and caudal periaqueductal gray (PAG) in cats to study corticofugal projections to the PAG. The method of retrograde axonal transport of HP demonstrated labeled neurons in the I and II somatosensory areas, frontal, cingular and insular cortex of the brain. It was shown that the II somatosensory cortex projects to all the areas of the rostral and caudal PAG. The frontal cortex projects to the dorsolateral quadrant of the PAG. The findings obtained enabled the detection of the morphological substrate of the corticofugal effects on one of the antinociceptive brain structures--the PAG.  相似文献

Using an indirect immunoperoxidase technique, the location of cell bodies and fibers containing substance P, neurokinin A and neurokinin B was studied in the cat spinal cord. The former two neuropeptides showed a widespread distribution throughout the whole spinal cord, whereas the distribution of neurokinin B was more restricted. Neurokinin A-immunoreactive structures showed a more widespread distribution and a higher density than the immunoreactive structures observed to contain substance P. In the cat spinal cord, we observed cell bodies containing neurokinin A, but no cell bodies containing neurokinin B or substance P were found. These cell bodies were located in laminae V (sacral 1 and 2 levels), VI (sacral 1 and 3), VII (lumbar 7, sacral 1 and 3, caudal 1) and X (sacral 1). Laminae I and II showed the highest density of immunoreactive fibers for each of the three tachykinins studied, being in general lamina IV who showed the lowest number of immunoreactive fibers containing substance P, neurokinin A or B. The anatomical distribution of the three tachykinins studied in the cat spinal cord indicates that the neuropeptides could be involved in the neurotransmission and/or in the neuromodulation of nociceptive information, as well as in autonomic and affective responses to pain. Moreover, the involvement of substance P, neurokinin A or B in other functions unrelated to the transmission of pain is also possible (autonomic and motor functions). The distribution of the neuropeptides studied in the cat is compared with the location of the same neuropeptides in the spinal cord of other species. The possible origin of the tachykinergic fibers in the cat spinal cord is also discussed.  相似文献

The activity of neurons in Deiters' lateral vestibular nucleus was recorded in decerebrate cats before, during and after spinal cord stimulation. An almost equal number of units were inhibited and excited early during stimulation. Later during stimulation the majority of units was inhibited. Early after cessation of stimulation an ever larger number of units were inhibited to an even larger extent (for about 2 imp/s on the average). Later after stimulus cessation the predominant inhibitory effect could still be noted, as well as excitation in some units. The results could support the hypothesis that the inhibition of Deiters' neurons during and for some time after epidural cord stimulation may play a part in the decrease of limb spasticity. The mechanism of inhibitory and excitatory unitary responses, side effects during stimulation and differences between the experimental model and human state are discussed.  相似文献

The intermediate laminae of the lumbosacral spinal cord are suggested to contain a small number of specialized neuronal circuits that form the basic elements of movement construction ("movement primitives"). Our aim was to study the properties and state dependence of these hypothesized circuits in comparison with movements elicited by direct nerve or muscle stimulation. Microwires for intraspinal microstimulation (ISMS) were implanted in intermediate laminae throughout the lumbosacral enlargement. Movement vectors evoked by ISMS were compared with those evoked by stimulation through muscle and nerve electrodes in cats that were anesthetized, then decerebrated, and finally spinalized. Similar movements could be evoked under anesthesia by ISMS and nerve and muscle stimulation, and these covered the full work space of the limb. ISMS-evoked movements were associated with the actions of nearby motoneuron pools. However, after decerebration and spinalization, ISMS-evoked movements were dominated by flexion, with few extensor movements. This indicates that the outputs of neuronal networks in the intermediate laminae depend significantly on descending input and on the state of the spinal cord. Frequently, the outputs also depended on stimulus intensity. These experiments suggest that interneuronal circuits in the intermediate and ventral regions of the spinal cord overlap and their function may be to process reflex and descending activity in a flexible manner for the activation of nearby motoneuron pools.  相似文献
