Efficiency of linked cross-sectional scheme has been compared with pure longitudinal schemes for generalised estimation of norm relaxing the condition of constancy of variances. The relevant theory of estimation has been developed and the expression for the optimum estimators alongwith their variances have been derived. The percentage gain in efficiency has been calculated. It has been observed that for higher values of Q, percentage gain in efficiency of proposed scheme over cross-sectional is very high.  相似文献

Frequently in threshold logic it is necessary to realize a switching function f of n variables in a two or more stages realization, both because it is not possible to realize f in a single gate and for reliability considerations. This means that the given function must be decomposed into a certain number of threshold functions, each of them having a number of variables less than n. When a geometric representation of switching functions is considered, the problem is equivalent to that of looking for functions whose patterns (on-sets) cover, in an adequate way, the on-set of the function f to be realized. In this paper a heuristic covering approach has been introduced which provides a manageable method for searching for a two stage realization of f. Furthermore a class of switching functions whose patterns possess a particular geometric shape (shell structure) has been considered. It has been proved that such functions (shell functions) are linearly separable and that one of their separating systems strictly depends on the centre and on the diameter of the shell structure. Gates implementing shell functions have been considered and their use has been revealed more convenient than that of majority gates in the solution of the problem of network synthesis. A farm population of the sable Martes zibellina L. has been selected for darker coat color during 40 years. Correlations between fitness characters and correlations of these characters with the selected character have been monitored. Correlation analysis has shown that the female fertility in the first year of reproduction is a promising predictor of how valuable the female will be for further breeding. Artificial directional selection has been shown to change the correlations that have been formed in natural populations of the sable. The relationship of this phenomenon with a decrease in the overall fitness during selection that has been observed in the sable population is discussed.  相似文献

Abstract

Twenty-eight moss species have been investigated cytologically. Mitotic karyograms have been presented for most of these species and these show a remarkable similarity in the n = 11 karyotype. Nine of the eleven chromosomes have terminal or sub-terminal centromeres. Lightly staining heterochromatic bands frequently occur along the axis of the chromosomes and very often these are the sites of chromosome bending; it has been suggested that these may be areas of neocentric activity. The karyotypes investigated show a reduction series of chromosome number which is paralleled by an increase in the number of long metacentrics; since the number of major chromosome arms is maintained in most of the species it has been suggested that Robertsonian fusion has been an important mechanism in the evolution of the Bracytheciaceae, and probably in all Diplolepidous mosses. Polyploidy has also played an important role in speciation. Finally it has been proposed that n = 11 is the primary basic number in the Diplolepideae.  相似文献

Several adducts of sulfur trioxide with electron-donor molecules have been prepared. Of these the air-stable triphenylphosphine-SO3 complex has been shown to be the most useful in the synthesis of peptides. It has been demonstrated that a variety of solvents may be used for couplings, but that in order for reaction to occur the solvent must itself be able to form an adduct. This behaviour is due to the fact that SO3 is transferred from the triphenylphosphine-SO3 complex to the solvent, thus forming an intermediate complex which is responsible for activation of the carboxylate unit. The carboxyl component is generally present as its tetramethylguanidinium salt. The method has been used to synthesize several model peptides and to prepare a fully biologically active sample of methionine-enkephalin.  相似文献

Synopsis Formaldehyde reacts with unsaturated fatty acids in tissues during histological fixation. The reaction of formaldehyde with oleic acid has been found to given rise to compounds (adducts) with the following structures: Two other compounds were isolated but their nature is still open to doubt.The equilibrium constant for the initial part of the reaction has been approximately estimated as 0·042 at a room temperature of 22°C. The endothermic heat of reaction has been estimated as approximately 12·6 kcal.The occurrence of these adducts in tissues explains why it is that less lipid can be demonstrated histologically in material that has been stored in form-aldehyde for a considerable length of time.  相似文献

Ligand-biased receptor signaling has been proposed for several G-protein coupled receptors including the niacin receptor GPR109A. Coupling to the Gi/o pathway has been shown to be responsible for the well described triglyceride lowering effect of nicotinic acid in mice, while activation of the β-arrestin pathway has been suggested to be responsible for its peripheral vasodilatory effect that causes cutaneous flushing. Several ligands have been described to selectively induce triglyceride lowering without inducing flushing.Cellular impedance has been demonstrated to determine G-protein coupled receptors activation in a G-protein specific manner. Agonists, which induce triglyceride lowering, but not flushing show a profile in cellular impedance that is distinct from the one induced by niacin and those compounds that induce triglyceride lowering as well as flushing. The strength of the signal correlates with the activation of β-arrestin.  相似文献

Background  

Until today, analysis of 16S ribosomal RNA (rRNA) sequences has been the de-facto gold standard for the assessment of phylogenetic relationships among prokaryotes. However, the branching order of the individual phlya is not well-resolved in 16S rRNA-based trees. In search of an improvement, new phylogenetic methods have been developed alongside with the growing availability of complete genome sequences. Unfortunately, only a few genes in prokaryotic genomes qualify as universal phylogenetic markers and almost all of them have a lower information content than the 16S rRNA gene. Therefore, emphasis has been placed on methods that are based on multiple genes or even entire genomes. The concatenation of ribosomal protein sequences is one method which has been ascribed an improved resolution. Since there is neither a comprehensive database for ribosomal protein sequences nor a tool that assists in sequence retrieval and generation of respective input files for phylogenetic reconstruction programs, RibAlign has been developed to fill this gap.  相似文献

From cell lysates of a recombinant Escherichia coli strain, Bacillus ginsengihumi phytase has been isolated and studied for the first time. The enzyme has been purified to homogeneity, and its primary structure has been determined. It has been concluded that phytase belongs to the class of β-propeller phosphatases. The molecular weight of the protein is 41 kDa, and pI is 4.8. Some physicochemical properties of the enzyme have been investigated.  相似文献

Malaria is a widespread and infectious disease that is a leading cause of death in many parts of the world. Eradication of malaria has been a major world health goal for decades, but one that still remains elusive. Other diseases have been eradicated using vaccination, but traditional vaccination methods have thus far been unsuccessful for malaria. Infection by Plasmodium species, the causative agent of malaria, is currently treated with drug-based therapies, but an increase in drug resistance has led to the need for new methods of treatment. A promising strategy for malaria treatment is to combine transmission blocking vaccines (TBVs) that prevent spread of disease with drug-based therapies to treat infected individuals. TBVs can be developed against surface protein antigens that are expressed during parasite reproduction in the mosquito. When the mosquito ingests blood from a vaccinated individual harboring the Plasmodium parasite, the antibodies generated by vaccination prevent completion of the parasites life-cycle. Animal studies have shown that immunization with Pfs48/45 results in the production of malaria transmission blocking antibodies; however, the development of this vaccine candidate has been hindered by poor expression in both prokaryotic and eukaryotic hosts. Recently, the chloroplast of Chlamydomonas reinhardtii has been used to express complex recombinant proteins. In this study, we show that the C-terminal antigenic region of the Pfs48/45 antigen can be expressed in the chloroplast of the green algae C. reinhardtii and that this recombinant protein has a conformation recognized by known transmission blocking antibodies. Production of this protein in algae has the potential to scale to the very large volumes required to meet the needs of millions at risk for contracting malaria.  相似文献

1. It has been shown that the glass electrode method as previously described by the author for measurements of transient rates in photosynthesis is free of systematic errors. 2. Justification has been given for the use of the steady state-gas flow method for measuring rates of assimilation under conditions in which the solution is known not to be seriously depleted of CO2. 3. The decline of photosynthetic rates under light saturation at CO2 pressures less than several tenths per cent of an atmosphere has been shown to be a real phenomenon. It has been suggested that there may be a real discrepancy between this finding and those of some other investigators due to a difference in the previous history of the algal suspensions.  相似文献

The genesis of the present research paper is to develop a revised exact analytical solution of thermal profile of 1-D Pennes’ bioheat equation (PBHE) for living tissues influenced in thermal therapeutic treatments. In order to illustrate the temperature distribution in living tissue both Fourier and non-Fourier model of 1-D PBHE has been solved by ‘Separation of variables’ technique. Till date most of the research works have been carried out with the constant initial steady temperature of tissue which is not at all relevant for the biological body due to its nonhomogeneous living cells. There should be a temperature variation in the body before the therapeutic treatment. Therefore, a coupled heat transfer in skin surface before therapeutic heating must be taken account for establishment of exact temperature propagation. This approach has not yet been considered in any research work. In this work, an initial condition for solving governing differential equation of heat conduction in biological tissues has been represented as a function of spatial coordinate. In a few research work, initial temperature distribution with PBHE has been coupled in such a way that it eliminates metabolic heat generation. The study has been devoted to establish the comparison of thermal profile between present approach and published theoretical approach for particular initial and boundary conditions inflicted in this investigation. It has been studied that maximum temperature difference of existing approach for Fourier temperature distribution is 19.6% while in case of non-Fourier, it is 52.8%. We have validated our present analysis with experimental results and it has been observed that the temperature response based on the spatial dependent variable initial condition matches more accurately than other approaches.  相似文献

For decades, the bio-duck sound has been recorded in the Southern Ocean, but the animal producing it has remained a mystery. Heard mainly during austral winter in the Southern Ocean, this ubiquitous sound has been recorded in Antarctic waters and contemporaneously off the Australian west coast. Here, we present conclusive evidence that the bio-duck sound is produced by Antarctic minke whales (Balaenoptera bonaerensis). We analysed data from multi-sensor acoustic recording tags that included intense bio-duck sounds as well as singular downsweeps that have previously been attributed to this species. This finding allows the interpretation of a wealth of long-term acoustic recordings for this previously acoustically concealed species, which will improve our understanding of the distribution, abundance and behaviour of Antarctic minke whales. This is critical information for a species that inhabits a difficult to access sea-ice environment that is changing rapidly in some regions and has been the subject of contentious lethal sampling efforts and ongoing international legal action.  相似文献

Amiodarone is an effective antiarrhythmic that has been used in Europe for over a decade and has been available for investigational use in North America for a shorter time. It has several well recognized side effects. Recent reports have related pulmonary disorders to the use of this drug; fibrosing alveolitis has been found by lung biopsy. Amiodarone''s toxicity to the lung does not appear to be dose-related. Besides cessation of amiodarone administration, management of this complication includes steroid therapy. A case is described of nonspecific diffuse alveolar damage syndrome in a patient who had received amiodarone.  相似文献

Although the nuclear processes responsible for genomic DNA replication and repair are well characterized, the pathways involved in mitochondrial DNA (mtDNA) replication and repair remain unclear. DNA repair has been identified as being particularly important within the mitochondrial compartment due to the organelle's high propensity to accumulate oxidative DNA damage. It has been postulated that continual accumulation of mtDNA damage and subsequent mutagenesis may function in cellular aging. Mitochondrial base excision repair (mtBER) plays a major role in combating mtDNA oxidative damage; however, the proteins involved in mtBER have yet to be fully characterized. It has been established that during nuclear long-patch (LP) BER, FEN1 is responsible for cleavage of 5′ flap structures generated during DNA synthesis. Furthermore, removal of 5′ flaps has been observed in mitochondrial extracts of mammalian cell lines; yet, the mitochondrial localization of FEN1 has not been clearly demonstrated. In this study, we analyzed the effects of deleting the yeast FEN1 homolog, RAD27, on mtDNA stability in Saccharomyces cerevisiae. Our findings demonstrate that Rad27p/FEN1 is localized in the mitochondrial compartment of both yeast and mice and that Rad27p has a significant role in maintaining mtDNA integrity.  相似文献

N-Methyl-d-aspartate (NMDA), which is a selective agonist for the NMDA receptor, has recently been shown to be present in various biological tissues. In mammals, the activity of d-aspartate N-methyltransferase (DDNMT), which produces NMDA from d-aspartate, has been detected only in homogenates prepared from rat tissues. Moreover, the enzymatic properties of DDNMT have been poorly studied and its molecular entity has not yet been identified. In this report, we show for the first time that the activity of DDNMT is present in mouse tissues and succeed in obtaining a partially purified enzyme preparation from a mouse tissue homogenate with a purification fold of 1900 or more, and have characterized the enzymatic activity of this preparation. The results indicate that DDNMT, which is highly specific for d-aspartate and is S-adenosyl-l-methionine-dependent, is a novel enzyme that clearly differs from the known methylamine-glutamate N-methyltransferase (EC 2.1.1.21) and glycine N-methyltransferase (EC 2.1.1.20).  相似文献

A recombinant plasmid, pI26, has been constructed by cloning into pBR322 a transforming gene of murine sarcoma virus (a Moloney strain, clone 124, MSV) synthesized by detergent-treated virions. From this plasmid a XbaI-HindIII fragment has been isolated which contains only mos-specific sequences. This mos-specific probe has been used for screening a human gene library cloned in bacteriophage λ Charon 4A. Of these, 19 clones have been isolated containing mos-related sequences. By physical mapping and molecular hybridization it has been shown that these sequences are neighboured by DNA regions related to Moloney murine leukemia virus. Recombinant phages have also been found containing human inserts related to MLV, not to the mos gene. The possible existence of murine-like endogenous retroviruses in the normal human genome, including that of a sarcoma type, is discussed. By Northern blotting, expression of the cellular c-mos gene has been detected in mouse liver treated with a hepatocarcinogen. The general significance of the suggested model for evaluating the relationship between chemical carcinogenesis and oncogene expression is discussed.  相似文献

The mei-41 gene of Drosophila melanogaster plays an essential role in meiosis, in the maintenance of somatic chromosome stability, in postreplication repair and in DNA double-strand break repair. This gene has been cytogenetically localized to polytene chromosome bands 14C4-6 using available chromosomal aberrations. About 60 kb of DNA sequence has been isolated following a bidirectional chromosomal walk that extends over the cytogenetic interval 14C1-6. The breakpoints of chromosomal aberrations identified within that walk establish that the entire mei-41 gene has been cloned. Two independently derived mei-41 mutants have been shown to carry P insertions within a single 2.2 kb fragment of the walk. Since revertants of those mutants have lost the P element sequences, an essential region of the mei-41 gene is present in that fragment. A 10.5 kb genomic fragment that spans the P insertion sites has been found to restore methyl methanesulfonate resistance and female fertility of the mei-41 D3 mutants. The results demonstrate that all the sequences required for the proper expression of the mei-41 gene are present on this genomic fragment. This study provides the foundation for molecular analysis of a function that is essential for chromosome stability in both the germline and somatic cells.This Paper is dedicated to the memory of Professor James B. In the early stages of a primary infection with N. brasiliensis in rats, adults are characteristically located about one-third of the distance anad of the pyloric sphincter. As one of a series of experimental investigations of this phenomenon, surgical manipulation of the site has been attempted. The normal site has been surgically removed, it has been reversed while keeping the blood system intact and the mesenteric arterial and venous supply has been locally restricted. In no case did surgical intervention significantly lower the number of adult parasites. In all cases there was a shift from the normal dispersion to one more orad. It has been conclusively demonstrated that there is no specific structural or physiological factor in the normal site of N. brasiliensis, which is critical for its survival. All results indicate as proposed in an earlier paper, that N. brasiliensis is a lumenal dweller and is most frequently recovered from the intestinal contents.  相似文献
