Chronic myeloid leukemia (CML) is characterized by a Ph1 chromosome that derives through a translocation between chromosomes 9 and 22, i.e., t (9;22). Identifying the Ph1 chromosome through cytogenetic analysis is an important aspect of CML diagnosis. The aim of this study was to determine the significance of cytogenetic analysis in the diagnosis of CML as well as to find out a relationship between chromosomal abnormalities and CML patients in different stages of treatment. Six CML patients were investigated for this study. The presence of Ph1 chromosome was detected at different times of treatment using GTG banding on peripheral blood or bone marrow aspirations, and the results were analyzed using cytovision workstation. Hematological features were compared between newly diagnosed patients and patients under treatment. The Ph1 chromosome was strongly associated with all cases of CML. The regression of Ph1 chromosomes differed for each patient depending on the treatments and individual response to specific treatments.  相似文献

ObjectivesDespite advances in the development of novel targeted therapies, the need for B-ALL alternative treatments has not been met. Anlotinib could blunt the proangiogenic activity of VEGFR, PDGFR, and FGFR, and has shown strong antitumor activities across multiple tumors. However, anlotinib cytotoxicity against B-ALL has not ever been evaluated, thus prompting us to initiate this study.MethodsExpression2Kinases program was used to identify potential treatment targets. Cell viability and apoptosis were determined by CCK-8 and Annexin V/PI staining kit, respectively. qRT-PCR and Western blotting were utilized to investigate the molecular mechanisms. In vivo antileukemia activity of Anlotinib was evaluated in a Ph+ B-ALL patient-Derived Xenograft (PDX) model.ResultsCompared with treatment-naive B-ALL cases, RR B-ALL patients had higher activities in the VEGF/VEGFR signaling and the PI3K/AKT/mTOR pathway. Exposure of Ph and Ph+ B-ALL cells to anlotinib resulted in significant cell viability reduction, apoptosis enhancement, and cell cycle arrest at G2/M phase. Importantly, anlotinib treatment led to remarkably decreased leukemia burdens and extended the survival period in a Ph+ B-ALL PDX model. Blockade of the role of the proangiogenic mediators, comprising VEGFR2, PDGFR-beta, and FGFR3, played a critical role in the cytotoxicity of anlotinib against Ph and Ph+ B-ALL. Moreover, anlotinib dampened the activity of PI3K/AKT/mTOR pathway that resides in the convergence of the three mentioned proangiogenic signals.ConclusionThis work provides impressive preclinical evidence of anlotinib against Ph and Ph+ B-ALL and raises a rationale for future clinical evaluation of this drug in the management of Ph and Ph+ B-ALL.  相似文献

Two cases of chronic myelogenous leukaemia were studied during the final blastic crisis. The results show in case I a stemline of 50 (2 Ph1) intact and 2 fragmentary chromosomes, with a considerable variation from this mode, and in case II a stemline of 48 chromosomes (2 Ph1), with only a slight variation. Different aspects of the presentation of aneuploidy in CML are discussed in relation to the findings in both cases, and special attention is given to the formation of stemlines.Director: Prof. Dr. H. The tetra-chelating ligands 1,2-bis[(5H-dibenzo[a,d]cyclohepten-5-yl)phenylphosphanyl]-ethane, bis(troppPh)ethane, and 1,3-bis[(5H-dibenzo[a,d]cyclohepten-5-yl)phenylphosphanyl]-propane, bis(troppPh)propane, were synthesised. For the binding of transition metals, these ligands offer two olefin moieties and two phosphorus centres and form mixtures of diastereomers with a R,S-configuration at the phosphorus centres (meso), or a R,R(S,S)-configuration (rac), respectively. meso/rac-bis(troppPh)ethane was separated by fractional crystallisation and reacted with [Ir(cod)2]OTf (cod=cylcooctadiene, OTf=CF3SO3 −) to give the penta-coordinated complex-cations meso/rac-[Ir(bis(troppPh)ethane)(cod)]+, where the bis(troppPh)ethane serves as tridentate ligand merely. One olefin unit remains non-bonded, however, a slow intra-molecular exchange between this olefin and the coordinated olefin unit was established (meso-[Ir(bis(troppPh)ethane)(cod)]+: k<0.5 s−1; rac-[Ir(bis(troppPh)ethane)(cod)]+: k≈35 s−1). The ligand meso/rac-bis(troppPh)propane reacts with [Ir(cod)2]OTf to give the corresponding complexes containing the tetra-coordinated 16-electron complex-cations meso/rac-[Ir(bis(troppPh)propane)]+. The diastereomers were separated by fractional crystallisation. The complex rac-[Ir(bis(troppPh)propane)]+ is reduced at relatively low potentials (E11/2=−0.95 V, E21/2=−1.33 V versus Ag/AgCl) to give the neutral 17-electron complex [Ir(bis(troppPh)propane)]0 and the 18-electron anionic iridate [Ir(bis(troppPh)propane)], respectively. With acetonitrile, [Ir(bis(troppPh)propane)]+ reacts to give the penta-coordinated complex rac-[Ir(MeCN)(bis(troppPh)propane)]+ (K=45 M−1, kf=6×103 M−1 s−1, kd=1×102 s−1) and with chloride to yield the relatively stable complex rac-[Ir(Cl)(bis(troppPh)propane)] (kd<0.5 s−1). Compared to the rac-isomer, the meso-[Ir(bis(troppPh)propane)]+ shows significantly cathodically shifted reduction potentials (E11/2=−1.25 V, E21/2=−1.64 V versus Ag/AgCl), an acetonitrile complex could not be detected, and the chloro-complex, meso-[Ir(Cl)(bis(troppPh)propane)], is much more labile (kd≈20′000 s−1). meso-[Ir(bis(troppPh)propane)]+ reacts with one equivalent H2 to give the trans-dihydride complex-cation, meso-[Ir(H)2(bis(troppPh)propane)]+, while the rac-isomer, rac-[Ir(bis(troppPh)propane)]+, reacts with two equivalents H2 to give rac-{Ir(H)2(OTf)[(troppPh)(H2troppPh)propane]}, a cis-dihydride complex containing a hydrogenated 10,11-dihydro-5H-dibenzo[a,d]cycloheptene unit, H2troppPh. The triflate anion in this complex is rather firmly bound and dissociates only slowly (k=29 s−1). All differences between the different stereoisomers are attributed to the fact that the ligand backbone in the meso-isomer, meso-[Ir(bis(troppPh)propane)]+, enforces a planar coordination sphere at the metal. On the contrary, already in the tetra-coordinated rac-[Ir(bis(troppPh)propane)]+, the metal has a tetrahedrally distorted coordination sphere which does not impede the reduction to the d9-Ir(0) and d10-Ir(−1) complexes and allows more easily a distortion towards a trigonal bipyramidal (tbp) or octahedral structure for penta- or hexa-coordinated complexes, respectively. A comparison of the NMR data for iridium bonded olefins in equatorial or axial positions in tbp structures shows that the latter experience only modest metal-to-ligand back-donation, while the olefins in the equatorial positions have a high degree of metallacyclopropane character.  相似文献

Hypoxia-inducible factor 1 (HIF-1) mediates many of the systemic and cellular responses to intermittent hypoxia (IH), which is an experimental model that simulates O2 saturation profiles occurring with recurrent apnea. IH-evoked HIF-1α synthesis and stability are due to increased reactive oxygen species (ROS) generated by NADPH oxidases, especially Nox2. However, the mechanisms by which IH activates Nox2 are not known. We recently reported that IH activates xanthine oxidase (XO) and the resulting increase in ROS elevates intracellular calcium levels. Since Nox2 activation requires increased intracellular calcium levels, we hypothesized XO-mediated calcium signaling contributes to Nox activation by IH. We tested this possibility in rat pheochromocytoma PC12 cells subjected to IH consisting alternating cycles of hypoxia (1.5% O2 for 30 sec) and normoxia (21% O2 for 5 min). Kinetic analysis revealed that IH-induced XO preceded Nox activation. Inhibition of XO activity either by allopurinol or by siRNA prevented IH-induced Nox activation, translocation of the cytosolic subunits p47phox and p67phox to the plasma membrane and their interaction with gp91phox. ROS generated by XO also contribute to IH-evoked Nox activation via calcium-dependent protein kinase C stimulation. More importantly, silencing XO blocked IH-induced upregulation of HIF-1α demonstrating that HIF-1α activation by IH requires Nox2 activation by XO.  相似文献

To characterise the NADH oxidase activity of both xanthine dehydrogenase (XD) and xanthine oxidase (XO) forms of rat liver xanthine oxidoreductase (XOR) and to evaluate the potential role of this mammalian enzyme as an O2 •− source, kinetics and electron paramagnetic resonance (EPR) spectroscopic studies were performed. A steady-state kinetics study of XD showed that it catalyses NADH oxidation, leading to the formation of one O2 •− molecule and half a H2O2 molecule per NADH molecule, at rates 3 times those observed for XO (29.2 ± 1.6 and 9.38 ± 0.31 min−1, respectively). EPR spectra of NADH-reduced XD and XO were qualitatively similar, but they were quantitatively quite different. While NADH efficiently reduced XD, only a great excess of NADH reduced XO. In agreement with reductive titration data, the XD specificity constant for NADH (8.73 ± 1.36 μM−1 min−1) was found to be higher than that of the XO specificity constant (1.07 ± 0.09 μM−1 min−1). It was confirmed that, for the reducing substrate xanthine, rat liver XD is also a better O2 •− source than XO. These data show that the dehydrogenase form of liver XOR is, thus, intrinsically more efficient at generating O2 •− than the oxidase form, independently of the reducing substrate. Most importantly, for comparative purposes, human liver XO activity towards NADH oxidation was also studied, and the kinetics parameters obtained were found to be very similar to those of the XO form of rat liver XOR, foreseeing potential applications of rat liver XOR as a model of the human liver enzyme.  相似文献

The reactions of R2SnX2 (R = Ph, Me; X = Cl, Br) with excess halide, tributylphosphine, tricyclohexylphosphine and tributylphosphine oxide have been investigated in dichloromethane solution by tin-119 and phosphorus-31 NMR techniques. R2SnX2 form five coordinate 1:1 adducts with halide and phosphine (phos) ligands whilst both 1:1 and 1:2 adducts are formed with tributylphosphine oxide (L). Tin-119 spectra imply that Ph2SnX2(phos) has the phosphine in the equatorial position of a trigonal bipyramid. At low temperature there is evidence for a slow intramolecular twist mechanism between octahedral isomers of Ph2SnCl2L2. The stereochemistry of the complexes Ph2SnX2L2 differ between chloro and bromo compounds and no mixed halide complex is observed. In the case of the bromo system only, the 1:3 adduct [Ph2SnBrL3]+Br is formed. Ph3SnCl does not react with phosphines but it does give 1:1 adducts with Cl, L and pyridine. All the adducts have similar tin-119 chemical shifts which is consistent with the phenyl groups being equatorial in the five coordinate trigonal bipyramidal adducts. Ph4Sn does not form adducts with X, L or phosphine.  相似文献

Turner syndrome is caused by haploinsufficiency of the short arm of X-chromosome, and is usually diagnosed by karyotyping. This procedure is time-consuming, expensive and unfeasible for population screening. We propose molecular detection of 45XO Turner patients based on the ability of HpaII, a methylation sensitive endonuclease, to induce the cleavage of non-methylated DNA in the active X-allele. Genomic DNA was obtained from 22 patients with Turner syndrome confirmed by karyotype (45XO, N = 18; 45XO/46XX, N = 4). After digestion, DNA was amplified with primers directed to exon 1 of the androgen receptor (AR) gene and to the GAPDH control gene. Normal control females or mosaic patients, with a second methylated X-chromosome, escaped from HpaII digestion and produced a band corresponding to AR gene amplification. 45XO patients have just one active non-methylated X-chromosome, completely digested by HpaII, thus preventing the amplification of the AR gene. Three of the 45XO cases gave amplified bands, suggesting low-frequency mosaicisms that are not detected by karyotyping. Compared to classical karyotype studies for the detection of 45XO Turner patients, this new molecular method is simpler, faster and less expensive.  相似文献

The complexes [Me2(Meclo)SnOSn(Meclo)Me2]2 (2) and [Ph3Sn(Meclo)] (3) where HMeclo is meclofenamic acid, N-(2,6-dichloro-m-tolylanthranilic acid)], have been prepared and structurally characterized by means of vibrational, 1H and 13C NMR spectroscopies. The crystal structure of complexes (2) and (3) have been determined by X-ray crystallography. Three distannoxane rings are present to the dimeric tetraorganodistannoxane of planar ladder arrangement of (2). The structure is centro symmetric and features a central rhombus Sn2O2 unit two additional tin atoms linked at the oxygen atoms. Five- and six-coordinated tin centers are present in the dimer distannoxane. X-ray analysis of (3) revealed a penta-coordinated structure containing Ph3Sn coordinated to the chelated carboxylato group. The polar imino hydrogen atom participates in intra-molecular hydrogen bonds. Complexes (2) and (3) are self-assembled via π → π, C-H-π, stacking interactions and intra-molecular hydrogen bonds. Meclofenamic acid and [Ph3Sn(Meclo)] have been evaluated for antiproliferative activity in vitro against three human cancer cell lines: MCF-7 (human breast cancer cell line), T24 (bladder cancer cell line), A-549 (non-small cell lung carcinoma) and a mouse L-929 (a fibroblast-like cell line cloned from strain L). The [Ph3Sn(Meclo)] complex exhibited high cytotoxic activity against all the cancer cell lines. Meclofenamic and [Ph3Sn(Meclo)] were tested for anti-mycobacterial activity against Mycobacterium tuberculosis H37Rv. The [Ph3Sn(Meclo)] complex was found to be a promising anti-mycobacterial lead compound, displaying high activity against M. tuberculosis H37Rv.  相似文献

Diploid-like chromosome pairing in polyploid wheat is controlled by several Ph (pairing homoeologous) genes with major and minor effects. Homoeologous pairing occurs in either the absence of these genes or their inhibition by genes from other species (Ph I genes). We transferred Ph I genes from Triticum speltoides (syn Aegilops speltoides) to T. aestivum, and on the basis of further analysis it appears that two duplicate and independent Ph I genes were transferred. Since Ph I genes are epistatic to the Ph genes of wheat, homoeologous pairing between the wheat and alien chromosomes occurs in the F1 hybrids. Using the Ph I gene stock, we could demonstrate homoeologous pairing between the wheat and Haynaldia villosa chromosomes. Since homoeologous pairing occurs in F1 hybrids and no cytogenetic manipulation is needed, the Ph I gene stock may be a versatile tool for effecting rapid and efficient alien genetic transfers to wheat.Contribution no. 93-435-J from the Kansas Agricultural Experiment Station, Kansas State University, Manhattan, KS 66506-5502, USA  相似文献

The purpose of this study was to evaluate the inhibitory effect of renierol, extracted from marine sponge Halicdona.SP., on xanthine oxidase (XO) and its hypouricemic effect in vivo. Renierol and a positive control, allopurinol, were tested for their effects on XO activity by measuring the formation of uric acid and superoxide radical from xanthine. Renierol inhibited XO in a concentration-dependent and competitive manner. IC50 value was 1.85 μg·ml? 1 through the measuring of uric acid and was 1.36 μg.ml? 1 through the measuring of superoxide radical. Renierol was found to have an in vivo hypouricemic activity against potassium oxonate-induced hyperuricaemia in mice. After oral administration of renierol at doses of 10, 20 and 30 mg.kg? 1, there was a significant decrease in the serum urate level (4.08 ± 0.09 mg.dl? 1, P < 0.01), (3.47 ± 0.11 mg.dl? 1, P < 0.01) and (3.12 ± 0.08 mg.dl? 1, P < 0.01), when compared to the hyperuricaemic control (6.74 ± 0.23 mg.dl? 1). Renierol was a potent XO inhibitor with hypouricemic activity in mice.  相似文献

Recent reports show that the natural β-diketone curcumin displays important biological properties regarding the intercellular adhesion molecule-1 (ICAM-1), which plays a critical role in the immune responses and inflammation. In this study the ICAM-1 inhibitory activity of β-diketone compounds, which are curcumin models lacking aromatic peripheral hydroxyl and methoxy groups, along with some metal derivatives is investigated. β-Diketones are systematically more active than metal complexes and the best obtained inhibition is 75% for both groups. The best inhibitors are 4-benzoyl-3-methyl-1-phenyl-pyrazol-5-one (HQPh) among the ligands, and sodium benzoylacetonato among metal derivatives. These results appear in line with the reported antitumor activity of related species. Since 4-acyl-5-pyrazolones posses four tautomeric forms, those corresponding to HQPh were investigated using density functional theory. Docking of all HQPh tautomers on ICAM-1 protein was performed suggesting one keto-enol form favored to act in biological environment.  相似文献

Hypoxanthine–xanthine oxidase (HX–XO) system is a classical system that can generate superoxide anions. The inductive effect of the HX–XO system for lambda prophage was investigated in this study. The results showed that the system can induce lambda prophage from the lysogenic state to lytic growth. The inductive effect was directly proportional to the concentration of HX and XO and inversely related to the time of preliminary incubation of HX with XO.The cell density of the lysogenic bacteria also greatly affected the inductive effect. The maximal PFU number of 2.9 × 104 PFU/ml was recorded at 0.86 mM HX, 1.6 × 10–2 U/ml XO, and a cell density of 108 cells/ml. The inductive effect of the HX–XO system was inhibited in the suspensions by glutathione, superoxide dismutase, and catalase. The results provide evidence that free radicals are the primary factors in the induction of lambda prophage in lysogenic bacteria.  相似文献

Four new triphenyltin(IV) complexes of composition Ph3SnLH (where LH = 2-/4-[(E)-2-(aryl)-1-diazenyl]benzoate) (1-4) were synthesized and characterized by spectroscopic (1H, 13C and 119Sn NMR, IR, 119Sn Mössbauer) techniques in combination with elemental analysis. The 119Sn NMR spectroscopic data indicate a tetrahedral coordination geometry in non-coordinating solvents. The crystal structures of three complexes, Ph3SnL1H (1), Ph3SnL3H (3), Ph3SnL4H (4), were determined. All display an essentially tetrahedral geometry with angles ranging from 93.50(8) to 124.5(2)°; 119Sn Mössbauer spectral data support this assignment. The cytotoxicity studies were performed with complexes 1-4, along with a previously reported complex (5) in vitro across a panel of human tumor cell lines viz., A498, EVSA-T, H226, IGROV, M19 MEL, MCF-7 and WIDR. The screening results were compared with the results from other related triphenyltin(IV) complexes (6-7) and tributyltin(IV) complexes (8-11) having 2-/4-[(E)-2-(aryl)-1-diazenyl]benzoates framework. In general, the complexes exhibit stronger cytotoxic activity. The results obtained for 1-3 are also comparable to those of its o-analogs i.e. 4-7, except 5, but the advantage is the former set of complexes demonstrated two folds more cytotoxic activity for the cell line MCF-7 with ID50 values in the range 41-53 ng/ml. Undoubtedly, the cytotoxic results of complexes 1-3 are far superior to CDDP, 5-FU and ETO, and related tributyltin(IV) complexes 8-11. The quantitative structure-activity relationship (QSAR) studies for the cytotoxicity of triphenyltin(IV) complexes 1-7 and tributyltin(IV) complexes 8-11 is also discussed against a panel of human tumor cell lines.  相似文献

The kinetics of the reactions between anhydrous HCl and trans-[MoL(CNPh)(Ph2PCH2CH2PPh2)2] (L=CO, N2 or H2) have been studied in thf at 25.0 °C. When L=CO, the product is [MoH(CO)(CNPh)(Ph2PCH2CH2PPh2)2]+, and when L=H2 or N2 the product is trans-[MoCl(CNHPh)(Ph2PCH2CH2PPh2)2]. Using stopped-flow spectrophotometry reveals that the protonation chemistry of trans-[MoL(CNPh)(Ph2PCH2CH2PPh2)2] is complicated. It is proposed that in all cases protonation occurs initially at the nitrogen atom of the isonitrile ligand to form trans-[MoL(CNHPh)(Ph2PCH2CH2PPh2)2]+. Only when L=N2 is this single protonation sufficient to labilise L to dissociation, and subsequent binding of Cl gives trans-[MoCl(CNHPh)(Ph2PCH2CH2PPh2)2]. At high concentrations of HCl a second protonation occurs which inhibits the substitution. It is proposed that this second proton binds to the dinitrogen ligand. When L=CO or H2, a second protonation is also observed but in these cases the second protonation is proposed to occur at the carbon atom of the aminocarbyne ligand, generating trans-[MoL(CHNHPh)(Ph2PCH2CH2PPh2)2]2+. Addition of the second proton labilises the trans-H2 to dissociation, and subsequent rapid binding of Cl and dissociation of a proton yields the product trans-[MoCl(CNHPh)(Ph2PCH2CH2PPh2)2]. Dissociation of L=CO does not occur from trans-[Mo(CO)(CHNHPh)(Ph2PCH2CH2PPh2)2]2+, but rather migration of the proton from carbon to molybdenum, and dissociation of the other proton produces [MoH(CO)(CNPh)(Ph2PCH2CH2PPh2)2]+.  相似文献

Univalent sex chromosomes in spermatocytes of Sxr-carrying mice   总被引:1,自引:0,他引:1  
Pachytene configurations of the sex chromosomes were studied in whole-mount, silver-stained preparations of spermatocytes in mice with XY,Sxr, XX,Sxr, XO,Sxr, XO,Sxr+512 and T(X;4)37H,YSxr chromosomes, and non-Sxr-carrying controls. XY,Sxr males showed an increased number of X and Y univalents and of self-synapsed Y chromosomes. In T(X;4)37H,YSxr males an increased proportion of trivalent+Y configurations was also accompanied by higher numbers of self-paired Y univalents; the proportion of trivalent+X4 was not increased, but that of self-synapsed X4 univalents was. There was more selfsynapsis in cells containing one univalent than in cells containing two univalents. Spermatocytes of XX,Sxr mice contained single univalent X, which was never seen to be self-synapsed, but self-synapsis of the X occurred in a proportion of cells in XO,Sxr males. There were no self-paired X chromosomes in the XO,Sxr+512 mouse although lowlevel pairing of the 512 chromosome occurred. All four XX,Sxr and XO,Sxr males contained testicular sperm, and testicular sperm were also present in one T(X;4)37H male, while another such male had sperm in the caput. It is concluded that (1) self-synapsis of univalents is affected by variable conditions in the cell as well as by the DNA sequences of the chromosome, and (2) that the level of achievable spermatogenesis is not always rigidly predetermined by a chromosome anomaly but can be modulated by the genetic background.  相似文献

Excitatory amino acid transporters (EAATs) are crucial in maintaining extracellular levels of glutamate, the most abundant excitatory neurotransmitter, below toxic levels. The recent three-dimensional crystal structure of GltPh, an archaeal homolog of the EAATs, provides elegant structural details of this family of proteins, yet we know little about the mechanism of the bacterial transporter. Conflicting reports in the literature have described GltPh as an aspartate transporter driven by Na+ or a glutamate transporter driven by either Na+ or H+. Here we use purified protein reconstituted into liposomes to thoroughly characterize the ion and substrate dependence of the GltPh transport. We confirm that GltPh is a Na+-dependent transporter that is highly selective for aspartate over other amino acids, and we show that transport is coupled to at least two Na+ ions. In contrast to the EAATs, transport via GltPh is independent of H+ and K+. We propose a kinetic model of transport in which at least two Na+ ions are coupled to the cotransport of each aspartate molecule by GltPh, and where an ion- and substrate-free transporter reorients to complete the transport cycle.  相似文献

Inhibitory activity of Fe-ion-catalyzed radical oxidation products from 22 types of phenolic compounds toward xanthine oxidase (XO) was investigated. Phenols are readily oxidizable compounds in nature and, thus, showed potent antioxidant activities. Among the phenols screened in this study, noticeable activity was observed in the oxidation product of caffeic acid, whereas almost no XO-inhibitory activity of caffeic acid was observed. Assay-guided purification of the oxidation product of caffeic acid afforded a highly potent XO inhibitor, with an IC50 value that was calculated to be 60 nmol L−1, which indicated XO-inhibitory activity much stronger than that of allopurinol (IC50 = 1 μmol L−1), a potent XO inhibitor and excellent medicine for the treatment of gout. Background

Xanthine oxidoreductase (XOR) existing in two interconvertible forms, xanthine dehydrogenase (XDH) and xanthine oxidase (XO), catabolises xanthine to uric acid that is further broken down to antioxidative agent allantoin. XOR also produces free radicals serving as second messenger and microbicidal agent. Large variation in the XO activity has been observed among various species. Both hypo and hyper activity of XOR leads to pathophysiological conditions. Given the important nutritional role of buffalo milk in human health especially in south Asia, it is crucial to understand the functional properties of buffalo XOR and the underlying structural basis of variations in comparison to other species.

Methods and Findings

Buffalo XO activity of 0.75 U/mg was almost half of cattle XO activity. Enzymatic efficiency (k cat/K m) of 0.11 sec−1 µM−1 of buffalo XO was 8–10 times smaller than that of cattle XO. Buffalo XOR also showed lower antibacterial activity than cattle XOR. A CD value (Δε430 nm) of 46,000 M−1 cm−1 suggested occupancy of 77.4% at Fe/S I centre. Buffalo XOR contained 0.31 molybdenum atom/subunit of which 48% existed in active sulfo form. The active form of XO in buffalo was only 16% in comparison to ∼30% in cattle. Sequencing revealed 97.4% similarity between buffalo and cattle XOR. FAD domain was least conserved, while metal binding domains (Fe/S and Molybdenum) were highly conserved. Homology modelling of buffalo XOR showed several variations occurring in clusters, especially close to FAD binding pocket which could affect NAD+ entry in the FAD centre. The difference in XO activity seems to be originating from cofactor deficiency, especially molybdenum.

Conclusion

A major fraction of buffalo milk XOR exists in a catalytically inactive form due to high content of demolybdo and desulfo forms. Lower Fe/S content and structural factors might be contributing to lower enzymatic efficiency of buffalo XOR in a minor way.  相似文献
