Abstract— [U-14C]Ribose was given by subcutaneous injection to young rats aged 2–56 days. During the first week after birth 14C in the brain was found mainly combined in glucose, fructose and sedoheptulose which contained 46–57 per cent of the 14C in the acid soluble metabolites in the rat brain. In contrast, during the critical period (10–15 days after birth) the 14C in the free sugars decreased from 24 to 3 per cent, while the 14C content of amino acids in the brain increased from 11 to 44 per cent of the total perchloric acid-soluble 14C. The increase in labelling of amino acids during the critical period was attributed to increased glycolysis and increased oxidation of pyruvate. The relative specific radioactivity of y -aminobutyrate and aspartate in the rat brain at 28 days after birth was equal to or greater than the relative specific radioactivity of glutamate. Assuming that the increase in amino acid content following the cessation of cell proliferation in the brain is located mainly in cell processes (cytoplasm of axons, dendrites, glial processes and nerve terminals), tentative values were estimated for the pool sizes of glutamate, glutamine, aspartate and y -amino butyrate.  相似文献

MEASUREMENT OF THE RATE OF GLUCOSE UTILIZATION BY RAT BRAIN IN VIVO   总被引:17,自引:15,他引:2  
Abstract— A method is described by which the rate of glucose utilization by whole brain of conscious rats may be measured. The basis is the uptake of 14C derived front [2-14C] glucose into the acid-soluble metabolite pool of brain. Catheters are placed in the femoral artery and vein under light ether anesthesia. After full recovery of consciousness a single intravenous injection of [2-14C] glucose is given and arterial blood samples taken at intervals. Simultaneous with the last sample the brain is removed and frozen within 1 s. The accumulation of 14C into the acid-soluble metabilite pool is measured and the rate of glucose utilization is calculated according to the equation:

The integral is calculated from the plasma glucose specific activity curve and evidence is presented to justify this procedure. The rate of glucose utilization measured by this method was 0·62 μmol/min per g in conscious rats and 0·28 μmol/min per g in sodium pentobarbital anesthetized rats.  相似文献

Abstract— Guinea pig neocortical tissues were incubated with [14C]adenine, dispersed in cold isotonic sucrose and subcellular fractions prepared by centrifugation. Some 98 per cent of the assimilated 14C was found as acid-soluble nucleotides in the incubated tissues. In primary fractions obtained by differential centrifugation, about 60 per cent of the [14C]-nucleotides were in supernatant fractions, in distinction to ATP of which the greatest molar quantity (61 per cent of that in the dispersion) was in the crude mitochondrial fraction. When the crude mitochondrial fraction was separated by density gradient centrifugation, most 14C was found in synaptosomal fractions and about 85 per cent of this 14C was adenine nucleotides.
Electrical stimulation of incubating tissues immediately prior to their dispersion and centrifugation greatly diminished the proportion of 14C subsequently found in nucleotides (collectively) in the supernatant fraction, and increased their inosine and hypoxanthine. Stimulation increased the tissue's cyclic AMP but a preferential localization for this was not established. Results are tentatively interpreted in terms of liberation of an adenine derivative on excitation, and its action or reuptake at a tissue component different from that from which it was liberated. Fractionation of tissues which had been incubated with both [14C]-adenine and [3H]adenosine suggested that of the two compounds, more adenosine was taken up by synaptic regions in preference to other cellular regions of the tissue.  相似文献

Abstract— Myelin, synaptosomal and mitochondrial fractions obtained from homogenates of whole mouse brain contain K+ which can exchange with 42K+ at 2º in 0·32 m -sucrose. The content and rates of exchange of K+ were greater at pH 8·2 than at 6·1. In the synaptosomal preparations, the rates of exchange and content of 42K+ and K+ declined progressively with decreasing pH.
Of the total synaptosomal K+, 95 per cent could exchange with external 42K+. At pH 7·5, 20 per cent of the K+ and 78 per cent of the Na+ appeared to reside in osmotically insensitive pools. Synaptosomal K+ at 2º was slowly displaced by NaCl (0·18 m ) and the rate of exchange between 42K+ and K+ was retarded. KCI (0·18 m ) did not readily displace endogenous Na+. Synaptosomal K+ exchanged with exogenous K+ more rapidly than with exogenous Na+.
These observations have been discussed in terms of possible roles for ion exchange as the principal means by which K+ traverses the plasma membrane at 2º.  相似文献

Abstract— [14C]Leucine was injected intracranially into the brainstem reticular formation at the level of the upper medulla by the stereotaxic method. Subcellular fractions prepared 3 hr after injection showed that the specific activities of leucine-incorporated proteins decreased in the order soluble, microsomal, nuclear and mitochondrial fractions. Specific activities of proteins in the sera were 2·2 per cent of whole homogenate proteins.
The results from 27 experiments showed that 66·6 per cent of the mean specific activities of proteins extracted from whole homogenates fell within ·1 s.d . and 100 per cent within ± 2 s.d . (close to a normal distribution). The coefficients of variation were between 40 and 50 per cent for whole homogenates, sera and all subcellular fractions. Reproducibility of results and factors concerned with possible errors in the technique are discussed.  相似文献

1. Growth, density and δ13C of wood and leaf area were measured in two adjacent stands of 6 year-old Eucalyptus globulus growing in the 600–700 mm year–1 rainfall region of south-western Australia. Study sites were identical except for differences in the availability of water owing to physical properties of soil profiles and location of sites within the landscape.
2. Abundance of 13C (expressed as δ13C) in wood of trees growing on the drought-prone site (– 24·8‰±1·4) was greater than in other trees (– 25·8‰±1·2, P <0·001) throughout the 6 years and, with further development, the δ13C signatures of wood may become useful indices of drought-susceptibility in plantations within a few years of establishment. The seasonal pattern of δ13C of wood appeared to reflect seasonal variation in water availability and duration of cambial activity.
3. Basic density of wood of trees growing on the more drought-prone site (496±14·0 kg m–3) was reduced compared to other trees (554±5·3 kg m–3, P <0·001). δ13C of wood across boundaries of growth-rings suggested that drought stopped cambial activity resulting in less production of late wood and less dense wood.
4. The stand growing on the drought-prone site had reduced growth, wood yield and leaf area but identical specific leaf area. Annual growth was correlated with the previous season's rainfall. Together, these results suggested that within the same evaporative climate, drought reduces growth primarily by reducing leaf area and that there is a lag between onset of drought and reduced productivity.  相似文献

In the present study, profiles of stable isotope composition were characterized for two species with partially migratory populations in rivers along the latitudinal gradient of Patagonia, brown trout Salmo trutta and rainbow trout Oncorhynchus mykiss . The effects of factors ( e.g. ontogeny of fishes, location, species and fasting) that may influence the stable isotope analysis (SIA) were evaluated, as was SIA evaluated as a tool to assign individual fish to their corresponding ecotype. Anadromous fishes exhibited enriched δ15N (15·2 ± 1·0‰; mean ± s . d .) and δ13C (−19·2 ± 1·3‰) relative to resident fishes'δ15N (8·8 ± 1·1‰) and δ13C (−23·2 ± 2·5‰). For both species, the difference in δ15N was larger between resident (range 6·8–10·7‰) and anadromous (range 14·3–17·8‰) fishes than that in δ13C. Values of δ13C, while not as dramatically contrasting in rainbow trout, provided a powerful anadromy marker for brown trout in the region. Increases were found in both δ15N and δ13C during the spawning migration of anadromous rainbow trout, most likely due to fasting. Differences in stable isotopes between location, size and species were found, suggesting different stable isotopes base levels in freshwater environments and different trophic levels and feeding location of anadromous populations. The SIA was demonstrated as a powerful tool for ecotype discrimination in Patagonian Rivers, overriding any effect of sampling location, size or species.  相似文献

L-GLUTAMIC ACID DECARBOXYLASE IN NON-NEURAL TISSUES OF THE MOUSE   总被引:7,自引:5,他引:2  
Abstract— Low levels of γ-aminobutyric acid (GABA) and of glutamic acid decarboxylase (GAD) activity have been detected in mouse kidney, liver, spleen and pancreas. Quantitation of both 14CO2 and [14C]GABA produced in radiometric assays from [U-14CJglutamic acid has shown that measurement of 14CO2 evolution alone is not, in all cases, a valid estimate of true GAD activity. As evidenced by increased ,14CO2 production upon addition of NAD and CoA to assay mixtures, radiometric assay of GAD activity in crude homogenates may yield 14CO2 via the coupled reactions of glutamic acid dehydrogenase and a-ketoglutarate dehydrogenase. The addition of 1 mM aminooxyacetic acid (AOAA) to assays of kidney homogenates inhibited [,14C]GABA production 92 per cent while 14CO2 production was inhibited only 53 per cent. No evidence was found to confirm the reported existence of a second form of the enzyme, GAD II. previously described by Haber el al. (H aber B., K uriyama K. & R oberts E. (1970) Biochem. Pharmac. 19, 1119-1136). Based on sensitivity-to AOAA and chloride inhibition, the GAD activity in mouse kidney is. apparently, indistinguishable from that of neural origin.  相似文献

Abstract— When [2-3H]glycerol was injected intracranially into young rats, it was presented as a pulse label, leaving the brain rapidly and giving up much of its labelled hydrogen to water. [2-3H]glycerol was efficiently incorporated into brain lipids, especially into choline and ethanolamine phospholipids. Following injection of a mixture of [3H]- and [14C]-labelled glycerol, the ratio of 3H to 14C in the phospholipids of both whole brain and the microsomal fraction decreased as a function of time after injection. This finding indicated less recycling of the tritium label. This lack of recycling was further indicated by the finding that 94 per cent of the tritium label of phosphatidyl choline was in the glycerol portion of the molecule rather than in the fatty acids. At 2 weeks following injection with [3H]glycerol, 93 per cent of the total radioactivity in brain appeared in the lipid fraction. In contrast, following injection with [14C]glycerol, only 57 per cent of the radioactivity appeared in lipid, with about 20 per cent in protein.  相似文献

(1) Treatment with cortisol acetate (0.2 mg daily during the first 4 days after birth) reduced the rate of growth in the rat: at 35 days of age the body weight was reduced by 50 per cent and the brain weight, depending on the region, by up to 30 per cent. (2) In the brain the normal increase in cell number was severely inhibited during the period of cortisol treatment; this resulted in a final deficit in cell number of about 20 per cent in the cerebrum and 30 per cent in the cerebellum. (3) To determine whether cortisol affected primarily cell formation or cell destruction the labelling of brain DNA was studied 1 h after a subcutaneous injection of 20 Ci/100 g [2-14C]thymidine. In the controls the amount of labelled DNA increased by a factor of two in the cerebrum and seven in the cerebellum during the period 2-13 days, and it decreased to 40 and 27 per cent of the peak values in the cerebrum and cerebellum respectively in the following 7 days. The results indicated that mitotic activity is higher in the cerebellum than in the cerebrum in the 2nd week of life. It would appear that in the cerebrum appreciable cell death accompanies new cell formation, especially during the period 13-35 days of age. (4) Cortisol treatment affected cell division rather than cell destruction in the brain since it strongly inhibited the incorporation of [2-14C]thymidine into DNA. The inhibition was severe during the period of treatment but it did not result in a lasting fall in mitotic activity. At the age of 13 days the amount of labelled DNA formed approached the normal level and it was twice that in controls at 20 days, indicating a tendency for compensating cell deficit by an accelerated mitotic activity. Nevertheless, massive cell proliferation ceased at about the same age as in normals; the labelling of DNA decreased markedly between 13 and 20 days after birth, and the DNA content did not increase after the age of 20 days. (5) In contrast to the marked effect on cell number, cortisol treatment did not influence significantly the maturational changes related to average cell size (DNA concentration) or the chemical composition of cells (RNA/DNA and protein/DNA).  相似文献

Abstract— Synaptosomes took up [14C]choline, about half or more of which was converted to [I4C]acetylcholine when incubated in an appropriate medium containing 1 to 5 μ M-[14C] choline and neostigmine. The amount of [14C]acetylcholine synthesized in synaptosomes increased in parallel with the increase of Na+ concentration in the incubation medium. The effect of Na+ on the uptake of [I4C]choline into synaptosomes was dependent on the concentration of choline in the incubation medium.
About 25 per cent of [14C]acetylcholine synthesized in synaptosomes was released rapidly into the medium by increasing the K+ concentration in the medium from 5 m m to 35 m m . The change of Na+ concentration hardly affected the release of [14C]acetylcholine. The effect of K+ on the release of [14C]choline was rather small compared to that on [14C] acetylcholine. Ouabain promoted the release of [14C]acetylcholine.  相似文献

CHOLINESTERASE ACTIVITY OF THE MOTOR ENDPLATE IN ISOLATED MUSCLE MEMBRANE   总被引:2,自引:0,他引:2  
Abstract— The cholinesterase activity of motor endplates in tibialis anterior muscle of rats accounted for about 20 per cent of the total cholinesterase activity of the muscle. In the isolated muscle membrane preparation of rat intercostal muscle, the cholinesterase activity was localized solely in the motor endplate, as shown by cholinesterase staining. The cholinesterase activity of the membrane per unit of nitrogen was 26·9 times that of the muscle homogenate. The membrane (endplate) cholinesterase had an optimal pH of 8, Km value of 3·1 m m , and was stable at 4° for at least 13 days. Cholinesterase of a motor endplate hydrolysed 2·69 × 108 acetylcholine molecules in 1 msec. Since it is estimated that 108 cholinesterase active sites are present in a motor endplate, the turnover time (time necessary for one enzyme site to hydrolyse one acetylcholine molecule) is calculated to be 372 μ sec, and the turnover number (molecules of acetylcholine hydrolysed by one enzyme site/min) to be 1·61 × 105. From studies with cholinesterase inhibitors, cholinesterase activity was estimated to be due mostly to acetylcholinesterase, and only a minor part to pseudocholinesterase. The muscle membrane preparation seems to be useful for the study of other properties of the motor endplate.  相似文献

IN VIVO METHYLATION AND TURNOVER OF RAT BRAIN HISTONES   总被引:3,自引:1,他引:2  
Abstract— The turnover of the different histone components from brain nuclei was studied after the administration of l -[3H]lysine and l -[14C-methyl]methionine to newborn rats. The radioactivities of the different histone subfractions as well as other proteins were determined over a 280-day period. Biphasic type decay curves (3H and 14C) were obtained for total brain histones and all the subfractions. From 6 to 40 days of age the half life of total brain histones was 19 days. After reaching brain maturity the half life was 132 days. The lysine rich histone (F1) was found to turnover the fastest of all the histones, having half lives of 13 and 112 days, respectively. The decay curve for the slightly lysine rich histones (F2a2, F2b) gave half lives of 25 days up to 40 days of age and 189 days after reaching brain maturity. The arginine rich histones (F2a1, F3) gave a half life of 32 days up to 40 days of age, while no turnover was observed after maturity. The turnover rates of the methyl groups and/or methionyl residues did not vary significantly from the turnover rates of the lysyl residues in the F2 and F3 histones. The lysine-rich histones did not contain significant amounts of methionyl residues or methyl groups.
Amino acid analysis of the brain histones revealed that about 3·6 per cent of the lysyl residues in the slightly lysine rich histones were methylated, mainly as ε-N-dimethyllysine. About 13 per cent of the lysyl residues in the arginine rich histones were methylated, mainly as ε-N-monomethyllysine and ε-N-dimethyllysine.  相似文献

Stable carbon (δ13C) and nitrogen (δ15N) isotopes were employed to elucidate energy flows and trophic interactions in Lake Apopka, a hypereutrophic lake in central Florida, U.S.A. Isotope compositions of lake biota ranged from −27·1 to −3·0‰ for δ13C, and from 3·7 to 13·9‰ for δ15N. The food web was based primarily on plankton production with diatoms, Microcystis and zooplankton dominating the diet of fish. Carbon isotope evidence showed that pico- and nano-phytoplankton were not a direct carbon source for fish, but were important to zooplankton. δ15N mass balance estimates indicated that planktivorous fish obtained 48–85% of their diets from zooplankton. The ∼3‰ range of δ15N in gizzard shad reflected increasing dependence on zooplankton as fish grew whereas the positive relationship between total length and δ15N of largemouth bass reflected increasing predation on larger planktivorous fish with growth. The broad ranges of δ13C (−25·9 to −9·5‰) and δ15N (5·8 to 14·4‰) of blue tilapia were indicators of diet diversity. Two presumed omnivores (brown bullhead and white catfish) and piscivores (black crappie, largemouth bass and Florida gar) were found to depend on planktivorous fish. However, stable isotope data revealed no trophic links between blue tilapia, an abundant fish in the near-shore area, and piscivores.  相似文献

Abstract— –The rates of incorporation of 14C from [U-l4C]glucose into intermediary metabolites have been measured in rat brain in vivo. The time course of labelling of glycogen was similar to that of glutamate and of glucose, which were all maximally labelled between 20 and 40min, but different from lactate, which lost radioactivity rapidly after 20min. The extent of labelling of glycogen (d.p.m./ μ mol of glucose) was of the same order as that of glutamate at 20 and 40 min after injection of [14C]glucose. However, calculations of turnover rates showed that glutamate turns over some 8-10 times faster than glycogen. Insulin, intracisternally applied, produced after 4-5 h a 60 per cent increase in glucose-6-P and a 50 per cent increase in glycogen. There was no change in the levels of glucose, glutamate or lactate, nor in the activity or properties of the particulate and soluble hexokinase of the brain. The injection of insulin affected neither the glycogen nor glucose contents of skeletal muscle from the same animals. The effects of insulin on the incorporation of l4C into the metabolites contrasted with its effects on their levels. The specific activities of glycogen and glucose were unchanged and there was a slight but non-significant increase in the specific activity of glutamate. The time course of incorporation into lactate was unaffected up to 20 min, but a significant delay in the loss of 14C after 20 min occurred as a result of the insulin injection. At 40 min, the specific activity of cerebral lactate was 60 per cent higher in insulin-treated animals than in control animals. The results are interpreted in terms of an effect of insulin on glucose uptake to the brain, with possibly an additional effect on a subsequent stage in metabolism, which involves lactate.  相似文献

Abstract— Isolated brains from three adult monkeys were perfused for 1 hr with [2-14C]glucose. Glycogen was isolated from the brain stem, cerebral hemispheres, cerebellum and the hypothalamic area at completion of the perfusions. The distribution of 14C in carbons of the glucose unit of glycogen was determined and from this the contribution of the pentose cycle to metabolism of glucose was calculated. The data indicate a maximum contribution by the pentose cycle of 5–8 per cent in brain. No significant difference was observed in the various portions of brain. Oxygen consumption was noted to be low in relation to the amount of glucose utilized, as measured in these experiments.  相似文献

Abstract— Desheathed rat dorsal root ganglia were incubated in a medium containing amino-oxyacetic acid and [3H]GABA. Under these conditions, [3H]GABA is taken up exclusively by the satellite glial cells in the ganglia. Efflux of [3H]GABA from the tissue was measured after passing the ganglia through a series of wash solutions. The spontaneous efflux of radioactivity, mostly [3H]GABA, was more rapid in the absence of amino-oxyacetic acid in the incubation and wash media.
Raising the potassium concentration in the wash media caused an increase in the efflux of [3H]GABA. This increase was sigmoidally related to the potassium concentration in the wash media, reaching a maximum at 64 m m -K+. The releasing effect of K+ was inhibited by removing calcium from the media. Reducing the calcium and raising the magnesium concentration in the wash solutions inhibited the increased efflux of [3H]GABA due to 64 m m -K+ by 48 per cent, while 5 mM-La3+ and diphenylhydantoin (0·005 and 0·5 m m ) had no effect on this increase.
Only a small increase in the efflux of [14C]glutamate was produced by 64 m m -K+ and it had no effect upon the effluxes of [3H]glycine, [3H]alanine or [3H]leucine. The efflux of lactate dehydrogenase was similarly unaffected by 64 mM-K+. The results suggest that glial cells in spinal ganglia can respond to depolarizing concentrations of potassium by releasing GABA in a calcium-dependent process.  相似文献

Abstract— The total lipid extract of shrimp muscle ( Artemisia longinaris ) was precipitated with ether. The supernatant containing 95 per cent of the phospholipids and 50 per cent of the protein showed binding for L-[14C]glutamatc in the first peak of protein. The sediment, redissolved in chloroform-methanol was chromatographed on a Sephadex LH-20 column. A single peak was eluted in the chloroform (20-40 ml) having no lipid phosphorous and high affinity binding for [14C]GABA. The saturation was achieved at 1 mole per 80.000 g protein and the curve revealed a single type of binding site. The purification achieved was of about 4000-fold. There was no binding of L-[14C]glulamate to the ether precipitate. The specificity of the binding of [14C]GABA was further supported by competition experiments with bicuculline. picro-toxin and muscimol. It is suggested that the hydrophobic protein isolated by us represents the GABA receptor. The findings presented in the two papers of the series suggest that the excitatory and inhibitory receptor from crustacean muscle can be separated as two different proteins.  相似文献

The diet of the dusky grouper Epinephelus marginatus , from the Balearic Islands, western Mediterranean, consisted primarily of crustaceans, molluscs and fishes, but diet composition varied with body size. The smaller dusky groupers (<300 mm L T) fed primarily on crustaceans, and particularly on brachyurans, which accounted for 46% of the prey identified. As dusky groupers grew, cephalopods became increasingly important and constituted 10 to 40% of the prey identified in subadult and adult specimens. The largest dusky groupers fed primarily on fishes that represented 40·9% of prey identified. These shifts in diet were accompanied by a positive selection of increasingly large prey and by an expansion of trophic niche. δ 15N values of dusky grouper white muscle ranged from 8·8 to 13·1% and 71% of the variation in δ 15N was explained by differences in dusky grouper size. δ 13C values ranged from – 17·9 to – 15·9%, and no significant body size effect on δ 13C was detected. Stomach content and 13C values indicated that from 1 year old, the diet of E. marginatus was based on the benthic food web. The enrichment in δ 15N registered from juvenile to large males was c . 3·8%. Overall, there was good agreement between gut content and stable isotope data indicating that the latter may be useful as a tool in trophic studies in marine protected areas where sampling for fishes is not allowed.  相似文献

We investigated variation in intrinsic water-use efficiency during the past century by analysing δ 13C in tree rings of beech growing in north-eastern France. Two different silvicultural systems were studied: high forest and coppice-with-standards. We studied separately effects related to the age of the tree at the time the ring was formed and effects attributable to environmental changes. At young ages, δ 13C shows an increase of more than 1‰. However, age-related trends differ in high forest and coppice-with-standards. Changes in microenvironmental variables during stand maturation, and physiological changes related to structural development of the trees with ageing, could explain these results. During the past century, δ 13C in tree rings shows a pattern of decline that is not paralleled by air δ 13C changes. Isotopic discrimination has significantly decreased from 18·1 to 16·4‰ in high forest and varied insignificantly between 17·4 and 16·9‰ in coppice-with-standards. As a consequence, intrinsic water-use efficiency has increased by 44% in high forest and 23% in coppice-with-standards during the past century. These results accord with the increased water-use efficiency observed in controlled experiments under a CO2-enriched atmosphere. However, other environmental changes, such as nitrogen deposition, may be responsible for such trends.  相似文献
