In the present work, in order to investigate the electronic excited-state intermolecular hydrogen bonding between the chromophore coumarin 153 (C153) and the room-temperature ionic liquid N,N-dimethylethanolammonium formate (DAF), both the geometric structures and the infrared spectra of the hydrogen-bonded complex C153–DAF+ in the excited state were studied by a time-dependent density functional theory (TDDFT) method. We theoretically demonstrated that the intermolecular hydrogen bond C1?=?O1···H1–O3 in the hydrogen-bonded C153–DAF+ complex is significantly strengthened in the S1 state by monitoring the spectral shifts of the C=O group and O–H group involved in the hydrogen bond C1?=?O1···H1–O3. Moreover, the length of the hydrogen bond C1?=?O1···H1–O3 between the oxygen atom and hydrogen atom decreased from 1.693 Å to 1.633 Å upon photoexcitation. This was also confirmed by the increase in the hydrogen-bond binding energy from 69.92 kJ mol?1 in the ground state to 90.17 kJ mol?1 in the excited state. Thus, the excited-state hydrogen-bond strengthening of the coumarin chromophore in an ionic liquid has been demonstrated theoretically for the first time.  相似文献

To understand the chemical behavior of uranyl complexes in water, a bis-uranyl [(phen)(UO2)(μ2–F)(F)]2 (A; phen?=?phenanthroline, μ2?=?doubly bridged) and its hydrated form A?·?(H2O)n (n?=?2, 4 and 6) were examined using scalar relativistic density functional theory. The addition of water caused the phen ligands to deviate slightly from the U22–F)2 plane, and red-shifts the U–F-terminal and U?=?O stretching vibrations. Four types of hydrogen bonds are present in the optimized hydrated A?·?(H2O)n complexes; their energies were calculated to fall within the range 4.37–6.77 kcal mol-1, comparable to the typical values of 5.0 kcal mol-1 reported for hydrogen bonds. An aqueous environment simulated by explicit and/or implicit models lowers and re-arranges the orbitals of the bis-uranyl complex.

A bis(uranyl) complex [(phen)(UO2)(μ2–F)(F)]2 (A) and its solvated form A?·?(H2O)n were examined using scalar relativistic density functional theory. Hydrogen bonds cause the phen ligand to slightly deviate from the equatorial plane of the uranyl ion, resulting in a pronounced red-shift of the U–F-terminal and U?=?O asymmetric stretching vibrations. The calculated energies fall within 4.4?–6.8 kcal/mol. Explicit and/or implicit aqueous solvation re-arranges the molecular orbitals of the complex  相似文献

The character of the bridged hydrogen atom (Hb) of B2H6 has become a hot issue in recent years. In this work, the complexes B2H6?·?·?·?NH3, B2H2X4?·?·?·?nNH3 (n?=?1, 2) and 2HF?·?·?·?B2H2X4?·?·?·?2NH3 (X?=?Cl, Br, I) were constructed and studied based on the M06-2X calculations to investigate how to enhance the Hb?·?·?·?N hydrogen-bonded interaction. When the terminal hydrogen atoms (Ht) of B2H6 were replaced by X (X?=?Cl, Br, I) atoms, the Hb?·?·?·?N hydrogen bond were strengthened. According to the electrostatic potentials in B2H2X4, two HF molecules were added to the interspace of the B-H-B-H four-membered ring of the B2H2X4?·?·?·?2NH3 complexes, and H?·?·?·?X hydrogen bond formed, resulting in further enhancing effect of Hb?·?·?·?N hydrogen bond. As a result, the positive cooperative effect of Hb?·?·?·?N hydrogen bond and H?·?·?·?X hydrogen bond do enhance the interactions of each other. The two measures not only enhance the strength of Hb?·?·?·?N hydrogen bond, but also achieve the goal to make the Hb?·?·?·?N hydrogen bond perpendicular to B?·?·?·?B direction.

Increase of the atmospheric concentration of halogenated organic compounds is partially responsible for a change of the global climate. In this work we have investigated the interaction between halogenated ether and water, which is one of the most important constituent of the atmosphere. The structures of the complexes formed by the two most stable conformers of enflurane (a volatile anaesthetic) with one and two water molecules were calculated by means of the counterpoise CP-corrected gradient optimization at the MP2/6–311++G(d,p) level. In these complexes the CH…Ow hydrogen bonds are formed, with the H…Ow distances varying between 2.23 and 2.32 Å. A small contraction of the CH bonds and the blue shifts of the ν(CH) stretching vibrations are predicted. There is also a weak interaction between one of the F atoms and the H atom of water, with the Hw…F distances between 2.41 and 2.87 Å. The CCSD(T)/CBS calculated stabilization energies in these complexes are between ?5.89 and ?4.66 kcal?mol?1, while the enthalpies of formation are between ?4.35 and ?3.22 kcal?mol?1. The Cl halogen bonding between enflurane and water has been found in two complexes. The intermolecular (Cl···O) distance is smaller than the sum of the corresponding van der Waals radii. The CCSD(T)/CBS stabilization energies for these complexes are about ?2 kcal?mol?1.

The capacity of SX2 (X = F, Cl, and Br) to engage in different kinds of noncovalent bonds was investigated by ab initio calculations. SCl2 (SBr2) has two σ-holes upon extension of Cl (Br)?S bonds, and two σ-holes upon extension of S?Cl (Br) bonds. SF2 contains only two σ-holes upon extension of the F?S bond. Consequently, SCl2 and SBr2 form chalcogen and halogen bonds with the electron donor H2CO while SF2 forms only a chalcogen bond, i.e., no F···O halogen bond was found in the SF2:H2CO complex. The S···O chalcogen bond between SF2 and H2CO is the strongest, while the strongest halogen bond is Br···O between SBr2 and H2CO. The nature of these two types of noncovalent interaction was probed by a variety of methods, including molecular electrostatic potentials, QTAIM, energy decomposition, and electron density shift maps. Termolecular complexes X2S···H2CO···SX′2 (X = F, Cl, Br, and X′ = Cl, Br) were constructed to study the interplay between chalcogen bonds and halogen bonds. All these complexes contained S···O and Cl (Br)···O bonds, with longer intermolecular distances, smaller values of electron density, and more positive three-body interaction energies, indicating negative cooperativity between the chalcogen bond and the halogen bond. In addition, for all complexes studied, interactions involving chalcogen bonds were more favorable than those involving halogen bonds.

MP2(full)/aug-cc-pVDZ(-PP) computations predict that new triangular bonding complexes (where X? is a halide and H–C refers to a protic solvent molecule) consist of one halogen bond and two hydrogen bonds in the gas phase. Carbon tetrabromide acts as the donor in the halogen bond, while it acts as an acceptor in the hydrogen bond. The halide (which commonly acts as an acceptor) can interact with both carbon tetrabromide and solvent molecule (CH3CN, CH2Cl2, CHCl3) to form a halogen bond and a hydrogen bond, respectively. The strength of the halogen bond obeys the order CBr4???Cl? > CBr4???Br? > CBr4???I?. For the hydrogen bonds formed between various halides and the same solvent molecule, the strength of the hydrogen bond obeys the order C-H???Cl? > C-H???Br? > C-H???I?. For the hydrogen bonds formed between the same halide and various solvent molecules, the interaction strength is proportional to the acidity of the hydrogen in the solvent molecule. The diminutive effect is present between the hydrogen bonds and the halogen bond in chlorine and bromine triangular bonding complexes. Complexes containing iodide ion show weak cooperative effects.

Extraction of Ca2+ from the oxygen-evolving complex of photosystem II (PSII) in the absence of a chelator inhibits O2 evolution without significant inhibition of the light-dependent reduction of the exogenous electron acceptor, 2,6-dichlorophenolindophenol (DCPIP) on the reducing side of PSII. The phenomenon is known as “the decoupling effect” (Semin et al. Photosynth Res 98:235–249, 2008). Extraction of Cl? from Ca2+-depleted membranes (PSII[–Ca]) suppresses the reduction of DCPIP. In the current study we investigated the nature of the oxidized substrate and the nature of the product(s) of the substrate oxidation. After elimination of all other possible donors, water was identified as the substrate. Generation of reactive oxygen species HO, H2O2, and O 2 ·? , as possible products of water oxidation in PSII(–Ca) membranes was examined. During the investigation of O 2 ·? production in PSII(–Ca) samples, we found that (i) O 2 ·? is formed on the acceptor side of PSII due to the reduction of O2; (ii) depletion of Cl? does not inhibit water oxidation, but (iii) Cl? depletion does decrease the efficiency of the reduction of exogenous electron acceptors. In the absence of Cl? under aerobic conditions, electron transport is diverted from reducing exogenous acceptors to reducing O2, thereby increasing the rate of O 2 ·? generation. From these observations we conclude that the product of water oxidation is H2O2 and that Cl? anions are not involved in the oxidation of water to H2O2 in decoupled PSII(–Ca) membranes. These results also indicate that Cl? anions are not directly involved in water oxidation by the Mn cluster in the native PSII membranes, but possibly provide access for H2O molecules to the Mn4CaO5 cluster and/or facilitate the release of H+ ions into the lumenal space.  相似文献

In this study are reported the syntheses of three bis(diarylhydrazonecarbonyl)methylene derivatives [{ArPhCNNH C(O)}2CH2] [Ar = 2 C5H4N (5), C6H5 (6), and 2‐C4H3S (7)], obtained by condensation of corresponding hydrazones with carbon suboxide, C3O2. The solid‐state self‐assembly of these carbonyl derivatives, giving rise to polymeric and dimeric networks, is described. In the formation of these structural features, in addition to N—H· · ·OC intermolecular hydrogen bonds, stabilizing intramolecular NH· · · π (systems) and intermolecular CO· · ·π (systems) interactions also seem to play an important role. Solution 1H‐nmr data of compounds 5–7 indicate that the polymeric and dimeric structures are not maintained in solution and show the occurrence of keto‐enolic equilibria. © 1999 John Wiley & Sons, Inc. The reaction of [VCl3(PMe2Ph)3] with HSSSSH (where the HS are thiophenolate and the S′ thioether functions, respectively), H21, yields [VCl(μ-SSSS)]2 (3) with one of the thiolate groups of each of the two ligands in the bridging mode. Reaction of Na21 with [VOCl2(thf)2] leads to a polymeric product of composition [VO(SSSS)]x (4). The products obtained from the reaction between [VOCl2(thf)2] and NaSNNSNa, Na22, (S is thiophenolate, N the amine function) depend on subtle changes in the diamine backbone of this ligand: If the amine functions are linked by -CH2CH2– (2a), the tetranuclear VIV complex [V(SNNS)μ-O]4 (5) is formed alongside the VIII complex [VCl(SNNS)]. If the backbone is -CH(Me)CH(Me)- (2b), [VO(SNNS)] (7) and the dinuclear, asymmetrically oxo-bridged VIV complex [{(SNN S)(thf)V}μ-O{V(SNN S)}] (8) are obtained. In 8, one amine of each of the two ligands is deprotonated to the amide group. In either case, the complexation is accompanied by oxidation of the thiolates to disulfides, leading to the generation of teraazatetrathio-cycloeicosanes (6a/b). Compounds 5 and 8·2THF have been structurally characterized by X-ray analyses. The connectivities have further been established for 3·2CH2Cl2 and for 6b, which exhibits the same conformation as formally characterized 6a. The cluster compound 5 is stabilized by an extended intramolecular N-H...O and N-H...S) hydrogen-bonding network. In 7·2THF, one of the THFs of crystallization is hydrogen-bonded to the NH of the penta-coordinated {VO(SNN S)} moiety; further, there is an intramolecular hydrogen bond between one of the thiolates of this tetragonal-pyramidal half of the molecule and the NH of the octahedral {VO(SNN S)thf} half. The generation of the ligand 2b from its precursor compound, the zinc complex [Zn(SNNS)] (9) leads to the structural characterization of 9·CH3OH with a large SZnS bite angle and a strong hydrogen bond between the methanolic OH and one of the thiolate sulfurs. The relevance of these compounds in biological systems is discussed.  相似文献

The interaction studies of CuII nalidixic acid–DACH chemotherapeutic drug entity, [C36H50N8O6Cu] with serum albumin proteins, viz., human serum albumin (HSA) and bovine serum albumin (BSA) employing UV–vis, fluorescence, CD, FTIR and molecular docking techniques have been carried out. Complex [C36H50N8O6Cu] demonstrated strong binding affinity towards serum albumin proteins via hydrophobic contacts with binding constants, K?=?3.18?×?105 and 7.44?×?104 M–1 for HSA and BSA, respectively implicating a higher binding affinity for HSA. The thermodynamic parameters ΔG, ΔH and ΔS at different temperatures were also calculated and the interaction of complex [C36H50N8O6Cu] with HSA and BSA was found to be enthalpy and entropy favoured, nevertheless, complex [C36H50N8O6Cu] demonstrated higher binding affinity towards HSA than BSA evidenced from its higher binding constant values. Time resolved fluorescence spectroscopy (TRFS) was carried out to validate the static quenching mechanism of HSA/BSA fluorescence. The collaborative results of spectroscopic studies indicated that the microenvironment and the conformation of HSA and BSA (α–helix) were significantly perturbed upon interaction with complex [C36H50N8O6Cu]. Hirshfeld surfaces analysis and fingerprint plots revealed various intermolecular interactions viz., N–H····O, O–H····O and C–H····O linkages in a 2–dimensional framework that provide crucial information about the supramolecular architectures in the complex. Molecular docking studies were carried out to ascertain the preferential binding mode and affinity of complex [C36H50N8O6Cu] at the target site of HSA and BSA. Furthermore, only for Transmission electroscopy microscopy micrographs of HSA and BSA in presence of complex [C36H50N8O6Cu] revealed major protein morphological transitions and aggregation which validates efficient delivery of complex by serum proteins to the target site.

A new pyrazole-based ligand, namely 1,3-bis(3,5-dimethylpyrazol-1-yl)-2-butanoic acid (Hbdmpb) was synthesised together with its copper complex Na[Cu(bdmpb)2(OOCCH3)H2O] · 4H2O. Both the free ligand and its Cu compound were fully characterised and their crystal structures were determined by X-ray analysis. The free-ligand molecular structure is uneventful. The Cu compound is highly unusual, as the pyrazole nitrogen atoms do not bind to the Cu ion. The copper(II) ion is coordinated by four nearly coplanar oxygen atoms from two dehydronated ligands bdmpb (CuO(1a) 1.942(4), CuO(1b) 1.933(4) Å), a monodentate acetate group (CuO(1) 1.927(3) Å) and a water molecule (CuO(1w) 1.937(4) Å). The nitrogen atoms of the pyrazole rings do not coordinate to the metal center, but instead are involved in strong intramolecular hydrogen bonds. The coordinated water molecule is strongly H-bonded to two pyrazole N atoms from two bdmpb ligands (N(12a) ? HO(1w) 2.762(7), N(12b) ? HO(1w) 2.774(7) Å). The other two pyrazole N atoms with a lone pair are hydrogen-bonded to water molecules in the lattice (N(22a) ? HO(2w) 2.763(7), N(22b) ? HO(6w) 2.892(7) Å). The sodium ion is six-coordinated by the oxygen atom O(2) of the acetato ligand and by five water molecules. The EPR spectrum recorded in the solid state shows a characteristic signal for an axial anisotropic S = 1/2 species. The spectrum recorded in methanol glass confirms the absence of the coordination of pyrazole nitrogen atoms to the copper centers.  相似文献

Exploring novel chemotherapeutic agents is a great challenge in cancer medicine. To that end, 2-substituted benzimidazole copper(II) complex, [Cu(BMA)Cl2]·(CH3OH) (1) [BMA = N,N′-bis(benzimidazol-2-yl-methyl)amine], was synthesized and its cytotoxicity was characterized. The interaction between complex 1 and calf thymus DNA was detected by spectroscopy methods. The binding constant (K b = 1.24 × 10M?1) and the apparent binding constant (K app = 6.67 × 10M?1) of 1 indicated its moderate DNA affinity. Complex 1 induced single strand breaks of pUC19 plasmid DNA in the presence of H2O2 through an oxidative pathway. Cytotoxicity studies proved that complex 1 could inhibit the proliferation of human cervical carcinoma cell line HeLa in both time- and dose-dependent manners. The results of nuclei staining by Hoechst 33342 and alkaline single-cell gel electrophoresis proved that complex 1 caused cellular DNA damage in HeLa cells. Furthermore, treatment of HeLa cells with 1 resulted in S-phase arrest, loss of mitochondrial potential, and up-regulation of caspase-3 and -9 in HeLa cells, suggesting that complex 1 was capable of inducing apoptosis in cancer cells through the intrinsic mitochondrial pathway.  相似文献

A comparative theoretical investigation into the change in strength of the trigger-bond upon formation of the Na+, Mg2+ and HF complexes involving the nitro group of RNO2 (R?=? –CH3, –NH2, –OCH3) or the C?=?C bond of (E)-O2N–CH?=?CH–NO2 was carried out using the B3LYP and MP2(full) methods with the 6-311++G**, 6-311++G(2df,2p) and aug-cc-pVTZ basis sets. Except for the Mg2+?π system with (E)-O2N–CH?=?CH–NO2 (i.e., C2H2N2O4?Mg2+), the strength of the trigger-bond X–NO2 (X?=?C, N or O) was enhanced upon complex formation. Furthermore, the increment of bond dissociation energy of the X–NO2 bond in the Na+ complex was far greater than that in the corresponding HF system. Thus, the explosive sensitivity in the former might be lower than that in the latter. For C2H2N2O4?Mg2+, the explosive sensitivity might also be reduced. Therefore, it is possible that introducing cations into the structure of explosives might be more efficacious at reducing explosive sensitivity than the formation of an intermolecular hydrogen-bonded complex. AIM, NBO and electron density shifts analyses showed that the electron density shifted toward the X–NO2 bond upon complex formation, leading to a strengthened X–NO2 bond and possibly reduced explosive sensitivity.

Introducing cations into explosives is more efficacious at reducing sensitivity than H-bond formation  相似文献

For the Fe–O2(S = 0) linkages of oxyhemes, valence bond (VB) structures are re-presented for the McClure [FeII(S = 1) + O2(S = 1)], Pauling–Coryell [FeII(S = 0) + O2*(S = 0)], and Weiss [FeIII(S = ½) + O2 ?(S = ½)] models of bonding. The VB structures for the McClure and Weiss models are of the increased-valence type, with more electrons participating in bonding than occur in their component Lewis structures. The Fe–O bond number and O–O bond order for the McClure structure are correlated with measured Fe–O and O–O bond lengths for oxymyoglobin. Back-bonding from O 2 ? to FeIII of the Weiss structure gives a restricted form of the McClure structure. The McClure and Weiss increased-valence structures are used to provide VB formulations of mechanisms for the oxyhemoglobin + NO reaction. The products of these two formulations are Hb+ and NO3 ? (where Hb is hemoglobin) and Hb+ and OONO?, respectively. Because Hb+ and NO3 ? are the observed products, they provide an experimental procedure for distinguishing the McClure and Weiss models. It is also shown that the same type of agreement between McClure-type theory and experiment occurs for oxycoboglobin + NO, cytochrome P450 monooxygenases, and related hydrogen atom transfer reactions. In the appendices, the results of density functional theory and multireference molecular orbital calculations for oxyhemes are related to one formulation of the increased-valence wavefunction for the McClure model, and theory is presented for the calculation of approximate weights for the Lewis structures that are components of the McClure increased-valence structure.  相似文献

The interactions of L-aminoglucosidic stereoisomers such as rhodostreptomycins A (Rho A) and B (Rho B) with cations (Mg2+, Ca2+, and H+) were studied by a quantum mechanical method that utilized DFT with B3LYP/6-311G**. Docking studies were also carried out in order to explore the surface recognition properties of L-aminoglucoside with respect to Mg2+ and Ca2+ ions under solvated and nonsolvated conditions. Although both of the stereoisomers possess similar physicochemical/antibiotic properties against Helicobacter pylori, the thermochemical values for these complexes showed that its high affinity for Mg2+ cations caused the hydration of Rho B. According to the results of the calculations, for Rho A–Ca2+(H2O)6, ΔH = ?72.21 kcal?mol?1; for Rho B–Ca2+(H2O)6, ΔH = ?72.53 kcal?mol?1; for Rho A–Mg2+(H2O)6, ΔH = ?72.99  kcal?mol?1 and for Rho B–Mg2+(H2O)6, ΔH = ?95.00  kcal?mol?1, confirming that Rho B binds most strongly with hydrated Mg2+, considering the energy associated with this binding process. This result suggests that Rho B forms a more stable complex than its isomer does with magnesium ion. Docking results show that both of these rhodostreptomycin molecules bind to solvated Ca2+ or Mg2+ through hydrogen bonding. Finally, Rho B is more stable than Rho A when protonation occurs.

Rho B–H showed higher stability since it is considered a proton pump inhibitor, and is therefore a stronger inhibitor of Helicobacter pylori  相似文献

Superoxide (O 2 ·? ) overproduction, by decreasing the nitric oxide (·NO) bioavailability, contributes to vascular complications in type 1 diabetes. In this disease, the vascular O 2 ·? can be produced by the NADPH oxidase (NOX), nitric oxide synthase (NOS), and xanthine oxidase (XO). This study aimed to determine the contribution of each enzymatic pathway in hyperglycemia-induced O 2 ·? overproduction, and the effects of an endurance training program and insulin therapy, associated or not, on the O 2 ·? production (amount and related enzymes) in diabetic rats. Forty male Wistar rats were divided into diabetic (D), diabetic treated with insulin (D-Ins), diabetic trained (D-Tr), or diabetic insulin-treated and trained (D-Ins + Tr) groups. An additional healthy group was used as control. Insulin therapy (Glargine Lantus, Sanofi) and endurance training (treadmill run: 60 min/day, 25 m/min, 5 days/week) started 1 week after diabetes induction by streptozotocin (45 mg/kg), and lasted for 8 weeks. At the end of the protocol, the O 2 ·? production in aorta rings was evaluated by histochemical analyses (DHE staining). Each production pathway was studied by inhibiting NOX (apocynin), NOS (L-Name), or XO (allopurinol) before DHE staining. Diabetic rats exhibited hyperglycemia-induced O 2 ·? overproduction, resulting from NOX, NOS, and XO activation. Insulin therapy and endurance training, associated or not, decreased efficiently and similarly the O 2 ·? overproduction. Insulin therapy reduced the hyperglycemia and decreased the three enzymatic pathways implicated in the O 2 ·? production. Endurance training decreased directly the NOS and XO activity. While both therapeutic strategies activated different pathways, their association did not reduce the O 2 ·? overproduction more significantly.  相似文献

Three new supramolecular complexes based on a 2-(pyrazin-2-yl)-1H-benzimidazole (Hpbi) and a series of Cd(II) salts have been solvothermally synthesized and structurally characterized by single-crystal X-ray diffraction analysis. Reaction of CdCl2·2.5H2O with Hpbi afforded a one-dimensional chain [Cd(Hpbi)Cl2] (1), which exhibits a three-dimensional (3-D) supramolecular architecture through intermolecular X-H···Cl (X = N and C) hydrogen bonds and π-π stacking interactions. When using CdBr2·4H2O instead of CdCl2·2.5H2O under similar reaction conditions, a bisnuclear complex [Cd(Hpbi)2Br2] (2) is obtained, which obviously exhibits intermolecular X-H···Br (X = N and C) hydrogen bonds and π-π stacking interactions. When CdI2 take place of CdCl2·2.5H2O, a mononuclear complex, [Cd(Hpbi)2I2] (3), is isolated, which shows a 3D supramolecule framework formed by intermolecule hydrogen bonds and π-π packing interactions. Interestingly, the Hpbi ligand exhibits the same coordination modes in complexes 1-3. It is noteworthy that the radius of anions plays an important role in affecting the structures and luminescent intensity of the final products. The TGA for 1-3 have been investigated and discussed in detail.  相似文献

Interaction between D-glucuronic acid and alkaline earth metal ions leads to the formation of the complexes such as M(D-glucuronate)X· nH2O and M(D-glucuronate)2 · nH2O, where M = Mg(II), Sr(II), and Ba(II), X = Cl? or Br?, and n = 2–4. Owing to the distinct spectral similarities with the structurally known Ca(D-gluguronate)Br · 3H2O compound, the metal cations bind to three sugar moieties (through O6, O5 of the first, O6', O4 of the second, and O1, O2 of the third residue) and to two H2O molecules, forming an eight-coordination geometry around each metal ion, in M(D-glucuronate)X · nH2O (except for Mg(II) ion, which is six-coordination). The metal ions in M(D-glucuronate)2-nH2O show six-coordination in different structural environments. The strong hydrogen bonding network of the free acid is weakened upon metalation and the sugar moiety crystallizes as α-anomer, in these series of metal-sugar complexes.  相似文献

The responses of the antioxidant defense system in plant species to drought stress are still relatively unknown. In order to further understand how the system responds to drought stress, the leaves of Fargesia denudata seedlings were investigated. Antioxidant enzyme activities, antioxidant contents, hydrogen peroxide (H2O2), superoxide anion (O 2 ·? ) and MDA contents in the seedling leaves were measured under well-watered (WW), moderate drought-stressed (MD), and severe drought-stressed (SD) treatments. Although drought stress significantly increased H2O2 and O 2 ·? levels in F. denudata leaves, only weak lipid peroxidation was observed. This is attributed to the higher superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX), glutathione reductase (GR), monodehydroascorbate reductase (MDHAR), and dehydroascorbate reductase (DHAR) activities in F. denudata leaves during the entire drought period. Reduced and oxidized ascorbate (AsA and DHA) contents were almost not affected by drought except that DHA under SD showed an obvious increase on day 30. Furthermore, reduced glutathione (GSH) content under drought stress significantly decreased, while oxidized glutathione (GSSG) markedly increased under SD on days 30 and 45 as well as under MD on day 30; as a result, the ratio GSH/GSSG declined considerably. These results indicated that GSH was involved in scavenging H2O2 and O 2 ·? under drought stress and it was more sensitive to drought stress in scavenging H2O2 and O 2 ·? than AsA. As a result, a highly efficient antioxidant defense system in drought-stressed F. denudate leaves operated mainly through the synergistic functioning of SOD, CAT, APX, MDHAR, DHAR, GR, and GSH against oxidative damage.  相似文献
