The mechanism of nitrate transport across the tonoplast of barley root cells   总被引:14,自引:0,他引:14  
Nitrate-selective microelectrodes were used to measure not only nitrate activity in the cytoplasm and vacuole of barley (Hordeum vulgare L.) root cells, but also the tonoplast electrical membrane potential. For epidermal cells, the mean cytoplasmic and vacuolar pNO3 (-log10 [NO3]) values were 2.3±0.04 (n=19) and 1.41±0.03 (n=35), respectively, while for cortical cells, the mean cytoplasmic and vacuolar nitrate values were 2.58±0.18 (n=4) and 1.17±0.06 (n=13), respectively. These results indicate that the accumulation of nitrate in the vacuole must be an active process. Proton-selective microelectrodes were used to measure the proton gradient across the tonoplast to assess the possibility that nitrate transport into the vacuole is mediated by an H+/NO 3 antiport mechanism. For epidermal cells, the mean cytoplasmic and vacuolar pH values were 7.12±0.06 (n=10) and 4.93±0.11 (n=22), respectively, while for cortical cells, the mean cytoplasmic and vacuolar pH values were 7.24±0.07 (n=3) and 5.09±0.17 (n=7), respectively. This paper deals with the effect of calcium binding in the cell wall on the measured 45Ca influx in Chara corallina Klein ex Will. esk. R.D. Wood. Calcium in the cell wall was in the range 687–1197 (mol · m–2 compared to the sap which contained only 144–256 mol · m–2. In dilute culture solutions the calcium content of the cell wall was relatively independent of external calcium at concentrations above about 0.1 mol · m–3. The half-times for exchange of calcium from 45Ca-labelled cell walls varied from 45 min at 0.05 mol · m–3 to less than 2 min at 2 mol · m–3. The effectiveness of other cations in displacing calcium from cell walls was in the order La > Zn > Co > Ni > Mg. Rinsing of 45Ca-labelled cell walls in 2 mol · m–3 LaCl3 for 20 min removed more than 99% of the bound 45Ca. However, the residual 45Ca activity in isolated cell walls following La3+ rinsing was similar to that in whole cells. It is concluded that in whole cells 45Ca influx cannot normally be distinguished from extracellular binding of calcium. Methods are described for the measurement of 45Ca fluxes in charophyte cells by isolation of intracellular 45Ca after the uptake period using techniques which avoid contamination from the large amount of tracer bound in the cell wall. At an external calcium concentration of 1 mol · m–3, the plasmalemma influx was approx. 0.2 nmol · m–2 · s–1 of which about half entered the vacuole and half was effluxed back into the external solution. The cytoplasm filled with calcium with a half-time of 40–50 min with an apparent pool size of 50 mmol · m–3. After 2 h the net flux to the cell was almost the same as the vacuolar flux. Uptake of 3H-labelled (±)-abscisic acid (ABA) into isolated barley (Hordeum vulgare L.) epidermal cell protoplasts (ECP) was followed over a range of pH values and ABA concentrations. The present results show that ABA uptake is not always linearly correlated with the external concentration of undissociated ABA (ABAH). At pH 7.25, ABA uptake exhibited saturation kinetics with an apparent K m value of 75 mmol·m–3 to tal ABA. This saturable transport component was inhibited by pretreating the protoplasts with 1 mol·m–3 p-chloromercuribenzenesulfonic acid at pH 8.0, conditions that minimized the uptake of this acid sulfhydryl reagent. Moreover, the rate of (±)-[3]HABA uptake was reduced by addition of 0.1 mol·m–3 (±)-ABA to 41%, whereas the same concentration of (±)-ABA was approximately half as effective (46% of the inhibitory effect). Thus, it was concluded that only (±)-ABA competes for an ABA carrier that is located in the epidermal cell plasma membrane. The permeability of the epidermal cell plasma membrane was studied by performing a Collander analysis. Thylakoids isolated from cells of the red alga Porphyridium cruentum exhibit an increased PS I activity on a chlorophyll basis with increasing growth irradiance, even though the stoichiometry of Photosystems I and II in such cells shows little change (Cunningham et al. (1989) Plant Physiol 91: 1179–1187). PS I activity was 26% greater in thylakoids of cells acclimated at 280 mol photons · m–2 · s–1 (VHL) than in cells acclimated at 10 mol photons · m–2 · s–1 (LL), indicating a change in the light absorbance capacity of PS I. Upon isolating PS I holocomplexes from VHL cells it was found that they contained 132±9 Chl/P700 while those obtained from LL cells had 165±4 Chl/P700. Examination of the polypeptide composition of PS I holocomplexes on SDS-PAGE showed a notable decrease of three polypeptides (19.5, 21.0 and 22 kDa) in VHL-complexes relative to LL-complexes. These polypeptides belong to a novel LHC I complex, recently discovered in red algae (Wolfe et al. (1994a) Nature 367: 566–568), that lacks Chl b and includes at least six different polypeptides. We suggest that the decrease in PS I Chl antenna size observed with increasing irradiance is attributable to changes occurring in the LHC I-antenna complex. Evidence for a Chl-binding antenna complex associated with PS II core complexes is lacking at this point. LHC II-type polypeptides were not observed in functionally active PS II preparations (Wolfe et al. Hydrogenase was solubilized from the membrane of acetate-grown Methanosarcina barkeri MS and purification was carried out under aerobic conditions. The enzyme was reactivated under reducing conditions in the presence of H2. The enzyme showed a maximal activity of 120±40 mol H2 oxidized · min–1 · min–1 with methyl viologen as an electron acceptor, a maximal hydrogen production rate of 45±4 mol H2 · min–1 · mg–1 with methyl viologen as electron donor, and an apparent K m for hydrogen oxidation of 5.6±1.7 M. The molecular weight estimated by gel filtration was 98,000. SDS-PAGE showed the enzyme to consist of two polypeptides of 57,000 and 35,000 present in a 1:1 ratio. The native protein contained 8±2 mol Fe, 8±2 mol S2–, and 0.5 mol Ni/mol enzyme. Cytochrome b was reduced by hydrogen in a solubilized membrane preparation. The hydrogenase did not couple with autologous F420 or ferredoxin, nor with FAD, FMN, or NAD(P)+. Summary Absorption of nitrate and ammonium was studied in water culture experiments with 4 to 6 weeks old plants of barley (Hordeum vulgare L.), buckwheat (Fagopyrum esculentum L. Moench) and rape (Brassica napus L.). The plants were grown in a complete nutrient solution with nitrate (5.7±0.2 mM) or nitrate (5.6±0.2 mM) + ammonium (0.04±0.02 mM). The pH of the nutrient solution was kept at 5.0 using a pH-stat. It was found that phosphorus deficiency reduced the rate of nitrate uptake by 58±3% when nitrate was the sole N source and by 83±1% when both nitrate and ammonium were present. The reduction occurred even before growth was significantly impeded by P deficiency. The inhibition of the uptake of ammonium was less,i.e. ammonium constituted 10±1% of the total N uptake in the P sufficient plants and 30±5% in the P deficient plants. The reduction of nitrate absorption greatly decreased the difference between the uptake of anions and cations. It is suggested that P deficiency reduced the assimilation of NO 3 into the proteins, which might cause a negative feedback on NO 3 influx and/or stimulate NO 3 efflux.  相似文献

The concentrations of vacuolar solutes in different cells of the upper epidermis of the third leaf of barley (Hordeum vulgare L.) were studied in leaves of different ages grown under different irradiances (120 or 400 mol photons·m–2·s–1). Vacuolar saps were extracted from individual cells located at various positions between adjacent veins and were analysed for their osmolality and the concentrations of K+, Ca2+, Cl, NO 3 and malate. Each ion showed a cell-specific distribution within the epidermis that was both quantitatively and qualitatively dependent on the leaf developmental stage and on the light level. During leaf ageing, Ca2+ accumulated preferentially in interstomatal cells (i.e. those located between longitudinally adjacent stomata) at concentrations up to 180 mM. Under low light conditions, this was accompanied by a more or less equal decrease in K+ concentration. Epidermal malate was found only in plants grown continuously or transiently under the high irradiance and reached highest concentrations in trough and interstomatal cells (60 to 150mM). Chloride concentration was highest in cells overlying the veins (designated as ridge cells) and lowest in cells located between the veins (trough cells), while NO 3 exhibited the reverse distribution, although the precise patterns were age-dependent. Epidermal osmolality increased with age, but the intercellular differences in the osmolalities were small compared to differences in vacuolar solute composition. The pericarp of the dormant sugarbeet fruit acts as a storage reservoir for nitrate, ammonium and -amino-N. These N-reserves enable an autonomous development of the seedling for 8–10 d after imbibition. The nitrate content of the seed (1% of the whole fruit) probably induces nitrate-reductase activity in the embryo enclosed in the pericarp. Nitrate that leaks out of the pericarp is reabsorbed by the emerging radicle. Seedlings germinated from seeds (pericarp was removed) without external N-supply are able to take up nitrate immediately upon exposure via a low-capacity uptake system (vmax = 0.8 mol NO 3 - ·(g root FW)–1·h–1; Ks = 0.12 mM). We assume that this uptake system is induced by the seed nitrate (10 nmol/seed) during germination. Induction of a high-capacity nitrate-uptake system (vmax = 3.4 mol NO 3 - ·(g root FW)–1·h–1; Ks = 0.08 mM) by externally supplied nitrate occurs after a 20-min lag and requires protein synthesis. Seedlings germinated from whole fruits absorb nitrate via a highcapacity uptake mechanism induced by the pericarp nitrate (748 nmol/pericarp) during germination. The uptake rates of the high-capacity system depend only on the actual nitrate concentration of the uptake medium and not on prior nitrate pretreatments. Nitrate deprivation results in a decline of the nitrate-uptake capacity (t1/2 of vmax = 5 d) probably caused by the decay of carrier molecules. Summary The transepithelial water permeability in frog urinary bladder is believed to be essentially dependent on the ADH-regulated apical water permeability. To get a better understanding of the transmural water movement, the diffusional water permeability (P d) of the basolateral membrane of urinary bladder was studied. Access to this post-luminal barrier was made possible by perforating the apical membrane with amphotericin B. The addition of this antibiotic increasedP d from 1.12±0.10×10–4 cm/sec (n=7) to 4.08±0.33×10–4 cm/sec (n=7). The effect of mercuric sulfhydryl reagents, which are commonly used to characterize water channels, was tested on amphotericin B-treated bladders. HgCl2 (10–3 m) decreasedP d by 52% andpara-chloromercuribenzoic acid (pCMB) (1.4×10–4 m) by 34%. The activation energy for the diffusional water transport was found to increase from 4.52±0.23 kcal/mol (n=3), in the control situation, to 9.99±0.91 kcal/mol (n=4) in the presence of 1.4×10–4 m pCMB. Our second approach was to measure the kinetics of water efflux, by stop-flow light scattering, on isolated epithelial cells from urinary bladders.pCMB (0.5 or 1.4×10–4 m) was found to inhibit water exit by 91±2%. These data strongly support the existence of proteins responsible for water transport across the basolateral membrane, which are permanently present.  相似文献

The light-dependent modulation of ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) activity was studied in two species: Phaseolus vulgaris L., which has high levels of the inhibitor of Rubisco activity, carboxyarabinitol 1-phosphate (CA1P), in the dark, and Chenopodium album L., which has little CA1P. In both species, the ratio of initial to fully-activated Rubisco activity declined by 40–50% within 60 min of a reduction in light from high a photosynthetic photon flux density (PPFD; >700 mol · m–2 · s–1) to a low PPFD (65 ± 15 mol · m–2 · s–1) or to darkness, indicating that decarbamylation of Rubisco is substantially involved in the initial regulatory response of Rubisco to a reduction in PPFD, even in species with potentially extensive CA1P inhibition. Total Rubisco activity was unaffected by PPFD in C. album, and prolonged exposure (2–6 h) to low light or darkness was accompanied by a slow decline in the activity ratio of this species. This indicates that the carbamylation state of Rubisco from C. album gradually declines for hours after the large initial drop in the first 60 min following light reduction. In P. vulgaris, the total activity of Rubisco declined by 10–30% within 1 h after a reduction in PPFD to below 100 mol · m–2 · s–1, indicating CA1P-binding contributes significantly to the reduction of Rubisco capacity during this period, but to a lesser extent than decarbamylation. With continued exposure of P. vulgaris leaves to very low PPFDs (< 30 mol · m–2 · s–1), the total activity of Rubisco declined steadily so that after 6–6.5 h of exposure to very low light or darkness, it was only 10–20% of the high-light value. Transformed Nicotiana plumbaginifolia plants with constitutive expression of nitrate reductase (NR) activity were grown at different levels of nitrogen nutrition. The gradients in foliar NO 3 content and maximum extractable NR activity observed with leaf order on the shoot, from base to apex, were much decreased as a result of N-deficiency in both the transformed plants and wild type controls grown under identical conditions. Constitutive expression of NR did not influence the foliar protein and chlorophyll contents under any circumstances. A reciprocal relationship between the observed maximal extractable NR activity of the leaves and their NO 3 content was observed in plants grown in nitrogen replete conditions at low irradiance (170 mol photons·m–2 ·s–1). This relationship disappeared at higher irradiance (450 mol photons·m–2·S–1) because the maximal extractable NR activity in the leaves of the wild type plants in these conditions increased to a level that was similar to, or greater than that found in constitutive NR-expressors. Much more NO 3 accumulated in the leaves of plants grown at 450 mol photons·m–2·s–1 than in those grown at 170 mol photons·m–2·s–1 in N-replete conditions. The foliar NO 3 level and maximal NR activity decreased with the imposition of N-deficiency in all plant types such that after prolonged exposure to nitrogen depletion very little NO 3 was found in the leaves and NR activity had decreased to almost zero. The activity of NR decreased under conditions of nitrogen deficiency. This regulation is multifactoral since there is no regulation of NR gene expression by NO 3 in the constitutive NR-expressors. We conclude that the NR protein is specifically targetted for destruction under nitrogen deficiency. The ion content of compartments within cortical cells of mature roots of the halophyte Suaeda maritima grown at 200 mol·m-3 NaCl has been studied by X-ray microanalysis of freeze-substituted thin sections. Sodium and Cl were found in the vacuoles at about four-times the concentration in the cytoplasm or cell walls, whereas K was more concentrated in the cell walls and cytoplasm than in vacuoles. The vacuolar Na concentration was 12- to 13-times higher than that of K. The Na concentration of cell walls of cortical cells was about 95 mol·m-3 of analysed volume. The cytoplasmic K concentration within the mature cortical cells was estimated to be 55 mol·m-3 of analysed volume.  相似文献

Summary Na+–H+ exchange activity in renal brush border membrane vesicles isolated from hyperthyroid rats was increased. When examined as a function of [Na+], treatment altered the initial rate of Na+ uptake by increasingV m (hyperthyroid, 18.9±1.1 nmol Na+ · mg–1 · 2 sec–1; normal, 8.9±0.3 nmol Na+ · mg–1 · 2 sec–1), and not the apparent affinityK Na + (hyperthyroid, 7.3±1.7mm; normal, 6.5±0.9mm). When examined as a function of [H+] and at a subsaturating [Na+] (1mm), hyperthyroidism resulted in the proportional increase in Na+ uptake at every intravesicular pH measured. A positive cooperative effect on Na+ uptake was found with increased intravesicular acidity in vesicles from both normal and hyperthyroid rats. When the data were analyzed by the Hill equation, it was found that hyperthyroidism did not change then (hyperthyroid, 1.2±0.06; normal, 1.2±0.07) or the [H+]0.5 (hyperthyroid, 0.39±0.08 m; normal, 0.44±0.07 m) but increased the apparentV m (hyperthyroid, 1.68±0.14 nmol Na+ · mg–1 · 2 sec–1; normal 0.96±0.10 nmol Na+ · mg–1 · 2 sec–1). The uptake of Na+ in exchange for H+ in membrane vesicles from normal and hyperthyroid animals was not influenced by membrane potential. H+ translocation or debinding was rate limiting for Na+–H+ exchange since Na+–Na+ exchange activity was greater than Na+–H+ exchange activity. Hyperthyroidism caused a proportional increase and hypothyroidism caused a proportional decrease in Na+–Na+ and Na+–H+ exchange. We conclude that hyperthyroidism leads to either an increase in the number of functional exchangers in the membrane or exactly proportional increases in the rate-limiting steps for Na+–Na+ and Na+–H+ exchange activity.  相似文献

Nitrogen fixation was measured in four subarctic streams substantially modified by beaver (Castor canadensis) in Quebec. Acetylene-ethylene (C2H2 C2H4) reduction techniques were used during the 1982 ice-free period (May–October) to estimate nitrogen fixation by microorganisms colonizing wood and sediment. Mean seasonal fixation rates were low and patchy, ranging from zero to 2.3 × 10–3 µmol C2H4 · cm–2 · h–1 for wood, and from zero to 7.0 × 10–3 µmol C2H4 · g AFDM–1 · h–1 for sediment; 77% of all wood and 63% of all sediment measurements showed no C2H2 reduction. Nonparametric statistical tests were unable to show a significant difference (p > 0.05) in C2H2 reduction rates between or within sites for wood species or by sediment depth.Nitrogen contributed by microorganisms colonizing wood in riffles of beaver influenced watersheds was small (e.g., 0.207 g N · m–2 · y–1) but greater than that for wood in beaver ponds (e.g., 0.008 g N · m–2 · y–1) or for streams without beaver (e.g., 0.003 g N · m–2 · y–1). Although mass specific nitrogen fixation rates did not change significantly as beaver transform riffles into ponds, the nitrogen fixed by organisms colonizing sediment in pond areas (e.g., 5.1 g N · m–2 · y–1) was greater than that in riffles (e.g., 0.42 g N · m–2 · y–1). The annual nitrogen contribution is proportional to the amount of sediment available for microbial colonization. We estimate that total nitrogen accumulation in sediment, per unit area, is enhanced 9 to 44 fold by beaver damming a section of stream.  相似文献

Summary We have investigated the effect of a purified preparation of Charybdotoxin (CTX) on the Ca-activated K+ (Ca–K) channel of human red cells (RBC). Cytosolic Ca2+ was increased either by ATP depletion or by the Ca ionophore A23187 and incubation in Na+ media containing CaCl2. The Ca–K efflux activated by metabolic depletion was partially (77%) inhibited from 15.8±2.4 mmol/liter cell · hr, to 3.7±1.0 mmol/liter cell · hr by 6nm CTX (n=3). The kinetic of Ca–K efflux was studied by increasing cell ionized Ca2+ using A23187 (60 mol/liter cell), and buffering with EGTA or citrate; initial rates of net K+ efflux (90 mmol/liter cell K+) into Na+ medium containing glucose, ouabain, bumetanide at pH 7.4 were measured. Ca–K efflux increased in a sigmoidal fashion (n of Hill 1.8) when Ca2+ was raised, with aK m of 0.37 m and saturating between 2 and 10 m Ca2+. Ca–K efflux was partially blocked (71±7.8%, mean ±sd,n=17) by CTX with high affinity (IC500.8nm), a finding suggesting that is a high affinity ligand of Ca–K channels. CTX also blocked 72% of the Ca-activated K+ efflux into 75mm K+ medium, which counteracted membrane hyperpolarization, cell acidification and cell shrinkage produced by opening of the K+ channel in Na+ media. CTX did not block Valinomycin-activated K+ efflux into Na+ or K+ medium and therefore it does not inhibit K+ movement coupled to anion conductive permeability.TheV max, but not theK m–Ca of Ca–K efflux showed large individual differences varying between 4.8 and 15.8 mmol/liter cell · min (FU). In red cells with Hb A,V max was 9.36±3.0 FU (mean ±sd,n=17). TheV max of the CTX-sensitive, Ca–K efflux was 6.27±2.5 FU (range 3.4 to 16.4 FU) in Hb A red cells and it was not significantly different in Hb S (6.75±3.2 FU,n=8). Since there is larger fraction of reticulocytes in Hb S red cells, this finding indicates that cell age might not be an important determinant of theV max of Ca–K+ efflux.Estimation of the number of CTX-sensitive Ca-activated K+ channels per cell indicate that there are 1 to 3 channels/per cell either in Hb A or Hb S red cells. The CTX-insensitive K+ efflux (2.7±0.9 FU) may reflect the activity of a different channel, nonspecific changes in permeability or coupling to an anion conductive pathway.  相似文献

Summary Paired toad urinary bladders were prepared without or with an osmotic gradient (175 mosm) across them, stimulated for 2.5 (n=6), 5 (n=6), 30 (n=6) or 60 (n=6) min with ADH (20 mU/ml), and studied by freeze-fracture electron microscopy. Water permeability at these times was assessed in additional bladders (n=6 for each case) after tissue fixation according to the technique of Eggena. After both 60 and 30 min of ADH stimulation, the presence of a gradient compared with the absence of one was associated with fewer aggregates (242±35vs. 382±14 ×235 m–2 at 60 min,P<0.01; 279±36vs. 470±51 ×235 m–2 at 30 min,P<0.01) and lower water permeability (8.4±1.1vs. 18.8±1.8g×min–1×cm–1 ×mosm –1 at60min,P<0.005; 9.2±1.0vs. 22.0±2.1 g ×min–1×cm–2×mosm –1 at 30 min,P<0.001). In addition, with a gradient both maximum water permeability and maximum aggregate frequency were reached nearly together; a similar correspondence occurred without a gradient. We conclude that in the presence of an osmotic gradient both the ADH-associated aggregates and the water permeability response to ADH are prevented from reaching the higher levels observed in bladders not exposed to a gradient.  相似文献

Summary Methods have been used for monitoring either volume flows or pressure changes, simultaneously with membrane potentials, in giant algal cells ofChara australis during an action potential. The volume flows were measured from the movement of a mercury bead in a capillary tube recorded by a photo-transducer. The pressure changes were measured by monitoring the deflection of a thin wedge, resting transversely across a cell, and using the same photo-transducer, the deflection of the wedge being directly related to the cell's turgor pressure. The average maximum rate of volume flow per unit area during an action potential was 0.88±0.11 nliter·sec–1·cm–2 in the direction of an outflow from the cell (total volume outflow being about 3 nliter·cm–2 per action potential). Similarly, the maximum rate of change of pressure was 19.6±3.8×10–3 atm·sec–1 (peak change being 19.3±2.9×10–3 atm equivalent to 14.7±2.2 mm Hg). The volume flow and pressure changes followed the vacuolar potential quite closely, the peak rate of volume flow lagging behind the peak of the action potential by 0.17±0.08 sec and the peak rate of pressure change leading it by 0.09±0.07 sec.  相似文献

Summary The water and potassium content and the relative vacuolar volume ( = Vvacuole/Vcell) of mesophyll cells of the needles of healthy 21-yearold spruce trees [Picea abies (L.) Karst.] were determined. In 5-year-old needles was 0.626 ± 0.178 (ovx ± SD). Potassium concentrations in the bulk tissue water ranged from about 65 to 105 mM. Simulations were made using this information and a simple two-compartmental model of the cell with the bulk cytoplasm and the vacuole and assuming that the minimum cytoplasmic and vacuolar K+ concentrations are 100–150 mM and 10–15 mM respectively. It is shown that a K+ content of needles below 50 mmol/1 tissue water would be precarious for maintenance of normal physiological and metabolic performance.  相似文献
