Using X-ray photography and flow cytometry, the internal morphologyand DNA replication activity of wild type (wt), GA- (gib-1 )and ABA-deficient (sitw ) tomato (Lycopersicon esculentum Mill.cv. Moneymaker) mutant seeds were studied. During seed formation,from 30 to 45 d after pollination (DAP) the endosperm becomessolid and the seed starts to gain desiccation tolerance. Atthis time significant changes occur in the amounts of DNA inradicle tip cells. At 30 DAP, radicle tip cells of the threegenotypes manifest about 60% of 2C, 30% of 4C and 10% of 8Camounts of DNA. Upon maturation (45 DAP onwards), most cellsin the seeds of the three genotypes arrest in the G1phase ofthe cell-cycle with 2C amounts of DNA. However, a relativelyhigh proportion of cells with 4C amounts of DNA was detectedin the radicle tip cells ofsitw compared with wild type andgib-1. At the well-matured stage (60 DAP), there were about 2% ofseeds with free space in wild type andgib-1 , and about 13%insitw . At the over-matured stage (75 DAP), even more seedswith free space were found insitw , whereas no increase in theproportion of the seeds with free space was detected in theother two genotypes. In -1.0 MPa PEG-6000 with or without 10µM GA4+7, no germination occurred in well-matured wildtype andgib-1 seeds, whether or not they were dried after harvest.However,sitw seeds were able to germinate both in over-maturefruit and in -1.0 MPa PEG-6000. Priming of dried seeds in -1.0MPa PEG induced a large amount of free space in almost all seedsof the three genotypes, and nuclear DNA synthesis in the radicletip cells of wild type andsitw seeds. However, PEG priming offresh (non-dried) seeds had no effect on the amount of freespace and 2C/4C DNA ratios in wild type orgib-1 seeds, but didinduce free space in about 20–25% ofsitw seeds and provoked4C signals insitw seeds. Removal of the endosperm and testaopposite the radicle tip of seeds resulted in root protrusion,the induction of free space and an increase of 4C DNA signalsin the three genotypes. It is concluded that ABA is crucialfor the efficient arrest of tomato embryo radicle tip cellsin G1phase upon maturation, whereas GAs play an important rolein re-initiating 4C DNA levels upon germination. Damage and degradation of cellular proteins is observed duringage-induced seed deterioration. L-Isoaspartyl protein methyltransferase(EC 2.1.1.77 [EC] ) is an enzyme hypothesized to play a role in limitingand repairing age-induced damage to proteins. Tomato (Lycopersiconesculentum Mill. ‘New Yorker’) seeds were assayedfor changes in L-isoaspartyl methyl-transferase activity duringaccelerated ageing and after osmotic priming. Accelerated ageingof seeds for 1–4 d at 45C and 100% relative humidityreduced germination from 94% to 71%, increased the mean timeof germination (MTG) from 2.4 to 5.8 d, and was accompaniedby a correlative decrease in L-isoaspartyl methyltransferaseactivity (r2=0.90). Aged and untreated seeds were primed for7 d at 20C in darkness using aerated solutions of 3% KNO3 orpolyethylene glycol 8000 (PEG) with equivalent osmotic potential(–1.25 MPa). Priming with KNO3 decreased the MTG, butdid not improve germination percentage for untreated seeds.Priming did not affect L-isoaspartyl methyltransferase activityin untreated seeds, but restored activity in aged seeds primedin KNO3 to levels near that of untreated seeds. Priming withPEG did not effectively improve the MTG or increase L-isoaspartylmethyltransferase activity. During germination, L-isoaspartylmethyltransferase activity remained constant for 48 h post-imbibitionand then declined, suggesting that the enzyme was developmentallyregulated and inactivated or degraded as radicle emergence occurred. The Effects of Priming and Ageing on Seed Vigour in Tomato   总被引:1,自引:0,他引:1  
A comparison was made of the effects of seed priming or ageingtreatments on the performance of tomato (Lycopersicon esculentumMill. cv. UC204C) seeds according to a number of indices ofseed vigour. A single lot of tomato seeds was primed in 120mol m–3 K2HPO4 + 150 mol m–3 KNO3 for 5 d at 20?C, or aged at 13% moisture content (dry weight basis) and 50?C for 6 d. Germination percentage (>98%) was unaffectedby priming and reduced to 85% by ageing. X-ray photographs andlongitudinal sections revealed the formation of free space surroundingthe embryo in dry primed seeds, which was not evident in controlor aged seeds. Priming increased the rate of germination atall temperatures above the base temperature (Tb), while ageingdecreased it. Tb was unaffected by priming and only slightlyincreased by ageing. The variation in individual times to germinationwas approximately doubled in both primed and aged seed comparedto the control, based upon the slopes of probit germinationpercentage versus log thermal time curves. Root growth aftergermination tests and seedling growth in both greenhouse andfield tests were not influenced by either priming or ageing.The conductivity test was found to be unreliable as a vigourtest for tomato seeds. The results identify several indiceswhich can be used to quantify seed vigour in tomato. They alsoillustrate that seed priming can enhance seed performance accordingto some criteria, while having no effect or decreasing qualityaccording to other criteria. Seed vigour can apparently be separatedinto various components which can be independently influencedby seed enhancement treatments. Flow cytometric determination of DNA levels in embryos of fullymatured dry tomato (Lycopersicon esculenium) seeds revealedlarge amounts of 2C DNA signals, indicating that most cellshad arrested in the cell cycle at the presynthetic G1 phaseof nuclear division. After imbibition in water, an augmentationof the 4C signal in the embryonic root tip region was found.This increase could be ascribed to cells entering the syntheticphase of nuclear division leading towards the doubling of chromosomalmaterial. In the root tip cells, 4C:2C ratios increased I dafter imbibition in water though radicle emergence started 2d later. Apparently, DNA synthesis preceded germination. Onlya small increase in the number of cells with 4C DNA levels wasfound in the rest of the embryonic tissues. In whole dry seeds,DNA histograms revealed both a 2C signal and a considerable6C peak, the latter originating from the endoreduplicated endosperm. A priming period of 14 d in PEG-6000 considerably enhanced therate and uniformity of germination. In the ungerminated seeds,the 4C DNA signal of root tip cells started to increase after3 d incubation in PEG. The ratio of 4C:2C steadily increasedduring the 14 d priming period, though did not reach the levelobtained after hydration in water. Upon priming, the 4C:2C ratiowas constant after redrying the seeds towards the original moisturecontent, indicating that the chromosomal material in the rootcells had stably ceased cell cycle activity at the G2 phase.The present results indicate that the beneficial effects ofpriming on seedling performance are associated with the actionof replicative DNA synthetic processes prior to germination. The bases of differences in germination rates (GRg = inverseof time to germination [tg] of percentage g) among three cold/salt-toleranttomato (Lycopersicon esculentum Mill.) accessions (PI 341988,PI 120256, and PI 174263) and one cold/salt-sensitive tomatocultivar (T5) were investigated. The effects of seed priming(6 d imbibition in aerated –1.2 MPa polyethylene glycolsolution at 20 ?C followed by redrying) and of removing theendosperm/testa cap covering the radicle on the temperaturesensitivity of GRg, and the interaction of these treatmentswith genotypes, were also examined. GRg decreased linearly withdecreasing temperature for all genotypes and seed treatments.The minimum or base temperatures for germination (Tb) variedby 1 ?C among the tomato lines, so genotypic differences inGRg were due to differing thermal time requirements for germination.The mean thermal time requirement for germination of T5 seeds was 22% and 19% greater than that of PI 341988 andPI 120256 seeds, respectively, but only 9% greater than thatof PI 174263 seeds. Seed priming did not lower Tb of any genotype,but significantly reduced by 24, 49, 41, and 49% in T5, PI 341988, PI 120256, and PI 174263, respectively,indicating that priming increased the rate at which the seedsprogressed towards germination when T>Tb, but did not lowerthe minimum temperature at which germination could occur. Primingincreased the GRg of T5 seeds to equal or exceed those of control(non-primed) seeds of the cold/salt-tolerant genotypes at anyT>Tb, but the PI lines exhibited an even greater responseto priming. Times to germination within each seed lot were normallydistributed on a logarithmic scale. Priming increased the variancein tg within a seed lot when compared to control seeds. However,the variation in thermal time for germination between the 10thand 90th percentiles of the seed population (T(10–90))was relatively unaffected by priming due to the reduction in in primed seeds. Removing the endosperm cap and testa opposite the radicle tip decreased almost 6-fold and and reduced Tb by 5 ?C in T5 and PI 341988,implicating processes in the endosperm/testa as the limitingfactors in germination at suboptimal temperatures. Germination of akinetes of Anabeana vaginicola v. fertilissimaPrasad in response to environmental stress was studied. Additionof nitrate to the medium induced early and maximum germination(96 per cent), whereas less than half of the akinetes germinatedwhen either nitrate or phosphate was omitted from the medium.The pH range over which germination occurred was 7.0–9.0.The desiccated akinetes after rehydration germinated after acertain lag period, depending upon the dehydration state. Thetemperature optimum for germination and vegetative growth wasthe same (25 °C) and germination did not occur at 5 °Cor above 35 °C. The limit of heat shock tolerated was 55°C for 4 min. In addition to white light, only the red partof the visible spectrum induced germination. Ultraviolet radiationreduced germination rate presumably by inducing thymine dimersin DNA. The photoreactivating system (s) in akinetes is certainlynon-photosynthetic. LD50 photon flux densities were 300 Jm–2for akinetes and 240 Jm–2 for vegetative cells. Several models have been proposed to describe germination rates,but most are limited in statistical analysis and biologicalmeaning of indices. Therefore, a mathematical model is proposedto utilize the logistic function. The function was defined asan overall response including time, temperature, and the interactionbetween time and temperature. Cumulative germination percentagesover time were used to develop the model. Germination tests were conducted on indiangrass (Sorghastrumnutans (L.) Nash) strain ‘IG-2C-F1’, at constanttemperatures of 9, 12, 15, 20, 25, and 30 °C. The functionfitted the observed data over six temperatures at r2 = 0.99.Time to reach 10% of final germination (Gt10) increased from2.5 d at 30 °C to 44.0 d at 9 °C, and Gt50 (time toreach 50% of final germination) increased from 3.6 d at 30 °Cto 53.8 d at 9 °C. True germination rate (% d–1) foreach temperature was maximum at Gt50. A linear model of 1/Gt50versus temperature was used to estimate the base temperatureof 8.3 °C for germination. An Arrhenius plot indicated achange occurred between 20 °C and 25 °C for temperatureresponse of germination. Published data on hypocotyl growthof Cucumis melo L. were recalculated using the model. Absolutegrowth rates showed a temperature response similar to the publishedweighted-mean elongation rates. Base temperature for hypocotylgrowth of C. melo was estimated as 8.8 °C. The rate of net photosynthesis (P) of whole plant stands oftomato (Lycopersicon esculentum Mill.), cucumber (Cucumis sativusL.) and sweet pepper (Capsicum annuum L.) was measured in sixlong-term experiments in large greenhouses under normal operatingconditions and CO2-concentrations between 200 and 1200 µmolmol-1. The objective was to quantify the responses to lightand carbon dioxide and to obtain data sets for testing simulationmodels. The method of measuring canopy photosynthesis involvedan accurate estimation of the greenhouse CO2 balance, usingnitrous oxide (N2O) as tracer gas to determine, on-line, theexchange rate between greenhouse and outside air. The estimatedrelative error in the observed P was about ± 10%, exceptthat higher relative errors could occur under particular conditions. A regression equation relating P to the photosynthetically activeradiation, the CO2 concentration and the leaf area index explained83-91% of the variance. The main canopy photosynthesis characteristicscalculated with the fitted regression equations were: canopyPmax 5-9 g m-2 h-1 CO2 uptake; ratio Pmax/LAI 1·5-3 gm-2 h-1; light compensation point 32-86 µmol s-1 m-2;light use efficiency (quantum yield) at low light 0·06-0·10µmol µmol-1 and CO2 compensation point 18-54 µmolmol-1. Controlled environment experiments were performed to determinethe effects of temperature and water potential on germination,radicle elongation and emergence of mungbean (Vigna radiata(L.) Wilczek cv. IPB-M79-17-79). The effects of a range of constant temperatures (15–45°C) and water potentials (0 to –2.2 MPa) on germinationand radicle elongation rates were studied using an osmoticumtechnique, in which seeds were held against a semi-permeablemembrane sac containing a polyethylene glycol solution. Linearrelationships were established between median germination time(Gt50) and water potential at different temperatures, and betweenreciprocal Gt50 (germination rate) and temperature at differentwater potentials. Germination occurred at potentials as lowas –2.2 MPa at favourable temperatures (30–40 °C),but was fastest at 40 °C when water was not limiting, withan estimated base temperature (Tb) of about 10 °C. Subsequentradicle elongation, however, was restricted to a slightly narrowertemperature range and was fastest at 35 °C. The conceptof thermal time was used to develop an equation to model thecombined effects of water potential and temperature on germination.Predictions made using this model were compared with the actualgermination obtained in a related series of experiments in columnsof soil. Some differences observed suggested the additionalimportance of the seed/soil/water contact zone in influencingseed germination in soil. Seedling emergence appeared to reflectfurther the radicle elongation results by occurring within anarrower range of temperatures and water potentials than germination.Emergence had an estimated Tb of 12.6 °C and was fastestat 35 °C. The effects of primingand ‘natural’ differences in quality amongst onionseed lots on the response of the rate of germination to temperatureand the identification of the characteristics under genotypiccontrol —J. exp. Bot. 39: 935–950. A screening procedure was applied to define the response ofthe rate of seed germination to sub-and supra-optimal temperaturesfor different lots or sub-lots of two onion (Allium cepa L.)cultivars.Three sub-lots of the cultivar White Lisbon were derived froma control lot by osmotic priming (–1.4 MPa, 20 °C.7 d) alone, by priming and drying and by priming, drying andsubsequently storing the seeds for 7 weeks at 2–5 °C.The major effect of priming was to reduce the thermal time forgermination at both sub- and supra-optimal temperatures. Primingalone also altered the distribution of thermal times at sub-optimaltemperatures. A new equation is presented to describe this variation.In contrast, priming had no consistent effect on base temperature(Tb and little effect on the distribution of ceiling temperatures[Te(G)]. For the control lot of White Lisbon Tb was 4°C,whilst the best common estimate of Tb for all four sub-lotswas 3.5°C. The mean estimate of Tc(50) for the control,primed and primed and dried sub-lots was 35.5°C.Comparisonof three lots of the cultivar Senshyu Semi Globe Yellow of widely-differingviability showed substantial differences in the thermal timefor germination at sub-optimal temperatures, but no significantdifferences in Tb (P>0.10), the common estimate being 4°C.There was a significant negative correlation between probitpercentage viability and the logarithm of the thermal time for50% germination at sub-optimal temperatures amongst the threelots (P<0.05). The work suggests that base temperature forgermination is a genotypic characteristic which is unaffectedby differences in seed quality. It also shows that the effectof priming, quantified as a reduction in thermal time requirementsfor germination, varies amongst the seeds within a lot. A range of post-storage priming treatments were evaluated todevelop a protocol for priming pea seeds. Post-storage primingtreatments at 16 °C with PEG-8000 (-0·5, -1·0and -1·2 MPa), ABA (10-1 M) and distilled water for 3,5 and 7 d ameliorated some of the damage which resulted fromageing. Most of the benefits occurred during the first 3 d withPEG or ABA and during the first 5 d distilled water. Primingtreatments increased the final germination and decreased themean germination time (MGT) and the frequency of chromosomalaberrations, possibly due to the repair of some age-induceddamage. The results of the priming experiment suggest that thecritical moisture content that facilitates repair of chromosomaldamage in pea seeds is likely to be between 32 and 46%. Chloroplast preparations from the young primary leaves of Phaseolusvulgaris L. cv. Canadian Wonder carry out the DNA-dependentincorporation of UTP into RNA at rates between 8 and 14 pmolUTP µg–1 chlorophyll h–1. It is estimatedthat 90% of the activity was localized in the chloroplasts.The incorporation proceeded for between 20 and 30 min at 35°C. The maximum rates of RNA synthesis were attained atpH 8.3, in the presence of 15 mM MgCl2. Chloroplasts were alsoactive, to a lesser extent, with 1.5 mM MnCl2. The simultaneouspresence of MnCl2 and MgCl2 resulted in inhibition of activity.Nuclear material prepared from young P. vulgaris leaves incorporatedUTP at a rate of about 12 pmol UTP µg–1 DNA h–1.On a chloroplast (Tritonsoluble) DNA basis chloroplast activitywas over 40-fold that of nuclei. Methods of solubilizing chloroplastRNA polymerase were explored. Yields of over 75% were achieved,but methods suitable for one species were not always successfulwhen applied to another. The highest yields of the P. vulgarisenzyme were obtained using EDTA and KCl. All methods resultedin solubilization of DNA. RNA synthesis by the soluble P. vulgarisenzyme proceeded for more than 40 min at 35 °C.  相似文献

Wolterbeek, H. Th. and De Bruin, M. 1986. The import and redistributionof several cations and anions in tomato leaves.—J. exp.Bot. 37: 331–340. The upward movements in the xylem and redistribution from theleaf of Na+ , K+ , Rb+, Cs+ and four anions were examined insub-systems of tomato plants (Lycopersicon esculentum, Mill.cv. Tiny Tim). There was a delay with respect to the redistributionof newly imported elements from the source leaf of about 16–20h for all four alkali ions. This is considerably less than theapparent delay for the anions Sb(SO4) WO42– Mo7O246–and AsO43– The prolonged delay for the anions is suggestedto be a consequence of metabolic transformation in the leaf.Reduction of the source-sink activity ratio did not decreasethe delay period from the source leaf, but apparently causedincreased Na+ transfer from the xylem. It is concluded thatthe application of a detailed mathematical descnption of upwardelement movement has considerable potential possibilities forunderstanding circulation of nutrients in the plant. The response of the germination of seeds of Barbarea vema (Mill.)Aschers, Brassica chinensis L., Brassica juncea (L.) Czern.& Coss., Brassica oleracea L. var. gongylodes L., Camelinasaliva (L.) Crantz, Eruca saliva Mill., Lepidium sativum L.,Nasturtium officinale R. Br., and Rorippa palustris (L.) Besserto white fluorescent light of different photon flux densitiesapplied for different daily durations in a diurnal alternatingtemperature regime of 20 °C/30 °C (16 h/8 h) was quantifiedby linear relations between probit percentage germination andthe logarithm of photon dose, the product of photon flux densityand duration. The low energy reaction, in which increasing dosepromotes germination, was detected in all the seed populationsbut in Barbarea vema and Brassica Juncea the lowest photon doseapplied (10–5–2 and 10–5 7 mol m–2 d–1,respectively) was sufficient to saturate the response. Comparisons,where possible, between photoperiods demonstrated reciprocity,i.e. germination was proportional to photon dose irrespectiveof photoperiod, for the low energy reaction in Brassica oleracea(1 min d–1 to 1 h d–1), Camelina saliva (1 min d–1to 8 h d–1), Eruca saliva (1 min d–1 to 24 h d–1),Lepidium sativum (I min d–1 to 8 h d–1) and Rorippapalustris (1 min d–1 to 8 h d–1), but not in Brassicachinensis and Nasturtium officinale. The high irradiance reaction,in which increasing dose inhibits germination, was detectedin Barbarea vema, Brassica chinensis, Brassica juncea, Brassicaoleracea, and Camelina saliva. The minimum dose at which inhibitionwas detected was lO–0–3 mol m–2 d–1.These results are discussed in the context of devising optimallight regimes for laboratory tests intended to maximize germination The response of germination to photon dose was also quantifiedwith 3 x 10–4 M GA2, co-applied (Brassica chinensis, Camelinasaliva, and Lepidium sativum) and with 2 x 10–2 M potassiumnitrate co-applied (Brassica chinensis). In the latter casepotassium nitrate had no effect in the dark and inhibited germinationin the light, but GA2, promoted germination substantially inall three species. Variation amongst seeds in the minimum photondose required to stimulate germination was not affected by co-applicationof GA2, in Brassica chinensis and Camelina saliva, whereas seedsof Lepidium salivum showed a narrower distribution of sensitivitiesto the low energy reaction in the presence of GA2 Barbarea vema (Mill.) The effect of alternating temperatures on the times taken byseeds of lentil (Lens culinaris Medikus) to germinate was investigatedusing a two-way temperature-gradient plate. Between 5 and 25°C,warmer temperatures increased the rate of germination. Variationamong the individual seeds in the times required for germinationat different constant temperatures within this range were describedwell by a log-normal distribution of thermal times, accumulatedabove a base temperature of 1·5°C. Even with amplitudesas great as 20°C, no effect of alternation per se on thethermal time required for germination was detected—whetherthe cool temperature was applied for 8 or 16 h d-1. Similarly,in alternating temperature regimes where the minimum temperatureof the diurnal cycle was between 0°C and the base temperature,the thermal times required for germination (where no thermaltime accrued during the periods when temperature was below Tb)were in close agreement with those values provided by the modeldetermined at warmer constant temperatures. However, where theminimum temperature applied was < 0°C the germinationof all but the earliest germinators was delayed beyond modelpredictions, and more so where the sub-zero minimum temperaturewas applied for 16 rather than 8 h d-1. The results, therefore,contradict the view that alternation in temperature per se reducesthe thermal time required for seed germination. Effects of chilling (5 °C) period, light and applied nitrogen(N) on germination (%), rate of germination (d to 50% of totalgermination; T50%) and seed imbibition were examined inClematisvitalba L. In the absence of chilling, light and N, germinationwas minimal (3%). When applied alone, both chilling and N increasedgermination. Chilling for 12 weeks increased germination to64%, and 2.5 mM NO-3or NH+4increased germination to 10–12%.Light did not increase germination when applied alone, but didwhen applied in combination with chilling and/or N. Half theseed germinated when light was combined with 2.5 mM NO-3or NH+4.The influence of chilling, light and/or N on germination wasgreater when combined, than when either factor was applied alone.Both oxidized (NO-3) and reduced (NH+4) forms of N increasedgermination, but non-N-containing compounds did not, suggestingthe response was due to N and not ionic or osmotic effects. Without additional N, T50%decreased from 16–20 d at zerochilling, to around 5 d at 8 and 12 weeks chilling. AlthoughT50%was not influenced by an increase in NO-3or NH+4from 0.5to 5.0 mM , it did increase with additional applied N thereafter.However, the magnitude of the N effect was small compared tothat of chilling. Like germination, seed imbibition increasedwith a longer chilling period, but in contrast imbibition decreasedslightly with increased applied NO-3or NH+4. It is argued thatincreased imbibition is not directly related to an increasein total germination, but that it may be related to the rateof germination. Possible mechanisms involved in the reductionin dormancy ofC. vitalba seed are discussed. Plants of Lolium perenne L. cv. S23 were grown in sand culturesupplied with either ammonium (NH4+) or nitrate (NO3)in an otherwise complete nutrient solution at 12°C or 20°C.Three weeks after germination, plants were clipped weekly tosimulate grazing. After 10 weeks growth all nitrogen (N) wassupplied enriched with 15N to quantify the effects of form ofN supply and temperature on the relative ability of currentroot uptake and remobilization to supply N for laminae regrowth. The form of N supply had no effect on the dry matter partitioning,while at 20°C more dry weight was allocated to laminae regrowthand less to the remaining plant material. The current root uptakeof N, which subsequently appeared in the laminae regrowth, wassimilar for plants supplied with NH4+ or NO3, and bothwere equally reduced at the lower temperature of growth. Remobilizationof N to laminae regrowth was greater for plants receiving NH4+than NO3; remobilization with either form of N supplywas reduced at the lower temperature of growth. Remobilizationwas reduced to a lesser extent at 12°C than current rootuptake. It was concluded that remobilization became relativelymore important in supplying N for regrowth of laminae at lowertemperatures. The intact dormant seeds of Dioscorea tokoro germinate slowlyif at all between 11-23°C; for full and rapid germinationthey require prior chilling treatment [Okagami and Kawai (1982)Bot. Mag. Tokyo 95: 155]. The germination abilities of zygoticembryos detached from dormant seeds of this species were studiedunder various nutritional and temperature regimes. For germinationof embryos, the minimum nutritional components in Murashigeand Skoog's (1962) medium that were required were sucrose andNO3 or SO2–4. As the source of carbohydrate forgermination of detached embryos, sucrose, mannose and maltosewere effective; glucose and fructose were less effective; andrhamnose was entirely unable to support germination. Embryos detached from dormant seeds, incubated with the sucroseplus KNO3, germinated more rapidly with increasing temperatureup to 35°C. However, application of sucrose and KNO3 didnot induce germination of intact seeds above 26°C. Therefore,it is very possible that the endosperm exerts an inhibitoryfunction on germination at such high temperatures. When seeds were incubated after a cut was made over a smallpart of the edge of the endosperm in which the radicle of theembryo is encased, germination occurred rapidly but the increasein germination percentage was slight. This result suggests thatthe endosperm suppots part of the germination inhibition bymeans of a mechanical barrier or its impermeability to wateror gases. Physiological features of the endosperm alone or interactionsbetween the embryo and endosperm may contribute significantlyto the characteristics of dormancy of intact seeds of this species. The vapour pressures of aqueous solutions of polyethylene glycol6000 have been measured (by equilibration with sucrose solutions)up to the saturation point at 25 °C (1.45 g g–1 water).The reduced-osmotic-pressure (/c), when plotted versus concentration(c), rapidly and linearly increased up to a concentration ofabout 0.8 g g–1 (crossing the similar plot for sucrose).Above this concentration, the reduced-osmotic-pressure rosemore slowly, but still more rapidly than sucrose. The maximumosmotic pressure achieved at saturation was nearly 18 MPa. Usingthe virial equation: /c= RT/M + RTA2c, the calculated secondvirial coefficient (A2) for the linear part is 4.5 x 10–3mol g–1, a value slightly greater than most literaturevalues at 25 °C. Data are cited showing that A2 varies linearlyfrom 5–6 x 10x3 at 0 °C, to zero at 80–90 °C  相似文献
