An isoperoxidase (EC 1.11.1.7) purified from avocado ( Persea americana Mill. cv. Topa Topa) leaves, lost 35% of its glycosidic moiety after incubation with glycopeptidase F (EC 3.2.2.18). The partial removal of carbohydrate chains caused a decrease in the Km for reductor substrates tguaiacol, syringaldazine and indole-3-acetic acid) and favoured the enzyme activation by Ca2+. A decrease in stability against temperature was also observed. Our results are consistent with the hypothesis that the carbohydrate moiety located on the surface of the peroxidase molecule acts as a molecular shield.  相似文献

A soluble anionic isoperoxidase (EC 1,11,1,7) was purified from peach ( Prunus persica L. Batsch cv. Merry) seeds. Purification was achieved by DEAE-Sephacel, Sephacryl S-300 and CM-cellulose chromatography. The purified isoperoxidase de-carboxylated indole-3-acetic acid (S0.5 0.13 m M , Hill coefficient 1.7). Molecular mass, determined by gel filtration and sodium dodecyl sulfate polyacrylamide gel electrophoresis, was ca 60 kDa. Polyclonal antibodies were raised in rabbit against this isoperoxidase. Using immunoprecipitation this isoenzyme was found to be immunologically different from other soluble anionic isoperoxidases isolated from peach seeds.  相似文献

Amylase activity extracted from tulip ( Tulipa gesneriana L. cv. Apeldoorn) bulbs that had been stored for 6 weeks at 4°C was resolved to 3 peaks by anion-exchange chromatography on diethylaminoethyl-Sephacel. These 3 amylases exhibited different relative mobilities during non-denaturing polyacrylamide gel electrophoresis (PAGE). The most abundant amylase form (amylase I) was purified to apparent homogeneity using hydrophobic interaction chromatography, gel filtration and chromatofocusing. The apparent molecular mass of the purified amylase was estimated to be 51 kDa by sodium dodecyl sulfate-PAGE and 45 kDa by gel filtration chromatography. The purified amylase was determined to be an endoamylase (EC 3.2.1.1) based on substrate specificity and end-product analysis. The enzyme had a pH optimum of 6.0 and a temperature optimum of 55°C. The apparent Km value with soluble starch (potato) was 1.28 mg ml−1. The presence of Ca2+ increased the activity and thermal stability of the enzyme. The presence of dithiothreitol enhanced the activity, while β -mercaptoethanol and reduced glutathione had no significant effect. When pre-incubated in the absence of the substrate, N-ethylmaleimide and 5,5'-dithiobis-(2-nitrobenzoic acid) partially inhibited the enzyme. α -cyclodextrins or β -cyclodextrins had no effect on enzyme activity up to 10 m M . In addition to CaCl2, CoCl2 slightly enhanced activity, while MgCl2 and MnCl2 had no significant effect at a concentration of 2 m M . ZnCl2, CuSO4, AgNO3 and EDTA partially inhibited enzyme activity, while AgNO3 and HgCl2 completely inhibited it at 2.0 m M .  相似文献

The effects of chemicals such as metal ions and typical organic enzyme inhibitors on the activity of deglycosylated β-fructofuranosidases ( P -1 and P -2) from Aureobasidium were observed and compared with those of native enzymes. P -1 and P -2 enzymes became sensitive to metal ions, such as Fe2+, Co2+, Ni2+ and Al3+, after deglycosylation. The enzymes were also inhibited by a sulphydryl reagent (monoiodoacetic acid), a peptide-hydrolysing reagent (hydroxylamine) and chelating reagents (sodium citrate, EDTA and sodium azide) after deglycosylation. The importance of deglycosylation for the determination of the true characteristics of the enzymes against chemicals is discussed.  相似文献

Carnation ( Dianthus caryophyllus L. cv. White Sim) petals contained two distinct invertases (EC 3.2.1.26) based on chromatographic behavior on DEAE-cellulose. Both are soluble in 20 m M sodium phosphate buffer (pH 6.5) and exhibit acid pH optimum of 5.5. Extraction of a cell wall preparation from petals with 1 M NaCl released little additional activity. Furthermore, only traces of activity remained associated with the NaCl-extracted cell wall preparation. One of the soluble invertases, representing over 75% of the total activity, was partially purified by (NH4)2SO4 fractionation and sequential chromatography over diethylaminoethyl-cellulose, concanavalin-A sepharose and polyacrylamide P-200. The enzyme was purified 38-fold with a recovery of 12%. It had an apparent native molecular weight of 215 kDa. The partially purified invertase is a β-fructofuranosidase (EC 3.2.1.26) based on its specificity for sucrose. The Km for sucrose was 3.3 m M . Accumulation of reducing sugars and increased invertase activity during expansive petal growth indicates that sucrose is the major source of carbon for petal growth.  相似文献

Acid phosphatase, purified from the yeast Saccharomyces cerevisiae, was completely deglycosylated by endo-beta-N-acetylglucosaminidase H or by HF treatment. Three protein bands were obtained on sodium dodecyl sulfate (SDS)-electrophoresis, with molecular weights of 73,000, 71,000 and 61,500. The released carbohydrate chains varied in size from 12 to 142 mannose units. To study the role of carbohydrate chains in the structure and function of acid phosphatase, a comparison of the properties of the partially deglycosylated enzyme with the native one was performed. The 60% deglycosylated enzyme retained the original activity, and CD and fluorescence spectra showed that the native conformation of the enzyme was preserved. The 90% deglycosylated enzyme showed a pronounced loss of enzyme activity, accompanied by the disruption of the three-dimensional structure. The partially deglycosylated enzyme was less soluble and more susceptible to denaturing effects of heat, pH, urea, and guanidine hydrochloride. Under conditions of electrophoresis, the partially deglycosylated enzyme dissociated, indicating a possible role of carbohydrate chains in maintaining the dimeric structure of the enzyme. Susceptibility of acid phosphatase toward proteolysis was drastically increased by deglycosylation.  相似文献

Sucrose-phosphate (SPS) from source leaves of soybean ( Glycine max (L.) Merr. cv. Ransom II) was purified 74-fold to a final specific activity of 1.8 U (mg protein)1. The partially purified preparation was free from phosphoglucoseisomerase (EC 5.3.1.9), pyrophosphatase (EC 3.6.1.1), phosphoenolpyruvate-phosphatase (EC 3.1.3.-), phosphofructokinase (EC 2.7.1.11), and uridine diphosphatase (EC 3.6.1.6), and was used for characterization of the kinetic and regulatory properties of the enzyme. The enzyme showed hyperbolic saturation kinetics for both fructose-6-phosphate (Km=0.57 m M ) and UDPGlucose (UDPG) (Km=4.8 m M ). The activity of SPS was inhibited by the product UDP. In vitro this inhibition could be partially overcome by the presence of Mg2+. Inorganic orthophosphate was only slightly inhibitory (35% inhibition at 25 m M phosphate). Glucose-6-phosphate (up to 20 m M ) had no effect on activity, and did not show any significant interaction with phosphate inhibition. A range of potential effectors was tested and had no effect on SPS activity: Glucose-1-phosphate, fructose-1, 6-bisphosphate, α-glycero-phosphate, dihydroxyacetone-phosphate, 3-phosphoglyceric acid, (all at 5 m M ), sucrose at 100 m M and pyrophosphate at 0.1 m M . The apparent lack of allosteric regulation of soybean SPS makes this enzyme markedly different from SPS previously characterized from spinach and maize.  相似文献

Rubbing applied to a young tomato internode inhibited the elongation of this internode and increased soluble peroxidase activity. These morphological and biochemical changes were observed both at the site of rubbing (local response) and in the neighbouring internode (systemic response). The cellular, biochemical, and molecular mechanisms leading to inhibition of internode elongation are not fully understood. It was proposed that mechanical stimuli increased the oxidation of IAA, via the induction of specific peroxidases and stimulated the lignification processes. In order to gain more information about the role of these enzymes, analysis of changes in peroxidase activities were performed. Qualitative analysis of isoperoxidases, by means of native cathodic PAGE, showed four induced isoforms termed C1, C2, C3, and C4. The major isoform (C2) was purified to homogeneity and partially characterized. This isoform is probably unglycosylated, with a molecular mass of 36 kDa and a neutral pI of 7.1. The effects of pH and temperature on the activity were determined with guaiacol as electron donor. Optima were obtained at pH 5 and at a temperature of 55°C. The activity of the purified enzyme was not affected by Ca2+, Mg2+ and Mn2+ as was reported for some basic peroxidases. Analysis of substrate specificity revealed that this isoperoxidase acted on ABTS, o -dianisidine, pyrogallol, guaiacol, coniferyl alcohol (monolignol) and IAA but not on syringaldazine. Activitiy of C2 isoperoxidase on coniferyl alcohol and IAA suggests a possible role of peroxidase C2 in inhibition of internode elongation, observed in rubbed plants, probably via an increase in lignification processes and regulation of IAA levels in internode tissues.  相似文献

Cyclic nucleotide phosphodiesterase (3',5'-cyclic nucleotide nucleotidohydrolase, EC 3.1.4.17) activity isolated from Phaseolus vulgaris L. cv. Limberg seedlings was partially purified and characterized by fractional (NH4)2SO4 precipitation, DEAE-cellulose chromatography, chromatography on 3',5'-cAMP-agarose, gel permeation chromatography and chromatofocusing. A crude enzyme preparation, a 30–65% (NH4)2SO4 pellet, showed an acidic pH optimum. The enzyme activity was stimulated by imidazole and divalent cations such as Ca2+, Mg2+ and Mn2+, whereas NaF, PPi and Fe3+ were inhibitory. Isobutylmethylxanthine had no significant effect on the plant enzyme. An MI of 42 000 was estimated by gel permeation high performance liquid chromatography. By chromatography on 3',5'-cAMP-agarose a phosphodiesterase was resolved that produced 5'-AMP as sole reaction product.  相似文献

The effects of pretreatment with HCN on the level of fructose 2.6-bisphosphate (F-2,6-P2). on activity of fructose 6-phosphate 2-kinase (EC 2.7.1.150; F-6-P. 2K. enzyme synthesizing F-2,6-P2), as well as on activities of PP1-dependent and ATP-dependent phosphofructokinases (EC 2.7.1.90. PP1-PFK and EC 2.7.1.11, ATP-PFK) were studied in cultured, dormant embryos of apple ( Malus domestica Borb. cv. Antonówka). HCN increased the F-2.6-P2 level and F-6-P, 2K activity in embryonal axes (3-fold), but had no effect in cotyledons. HCN pretreatment of embryos or the addition of F-2.6-P2 to enzyme extract stimulated PP1, -PFK activity, whereas the activity of ATP-PFK was slightly inhibited by HCN in axes and in cotyledons. Glycolysis is one of the first processes in the germination of apple embryos, and the stimulation of glycolysis by HCN may be the result of F-6-P, 2K activation in the axes. This will lead to accumulation of F-2,6-P2, which, in turn, enhances glycolysis by activation of PP1PKF.  相似文献

The metabolism of hydrogen peroxide by the scavenging system was studied in Chlamydomonas grown in a selenium-lacking and a selenium-containing medium. In cells of the former, 40% of external hydrogen peroxide (H2O2) was scavenged by ascorbate peroxidase (AsAP; EC 1.11.1.11) and the residual H2O2 by catalase (EC 1.11.1.6). The enzymes involved in the ascorbate-glutathione cycle including AsAP. were localized in the chloroplast. In cells of the latter, glutathione peroxidase (GSHP; EC 1.11.1.9) functioned primarily in the removal of external H2O2. GSHP was located solely in the cytosol. The Chlamydomonas AsAP was relatively stable in ascorbate-depleted medium as compared with chloroplast AsAP of higher plants. No inactivation of the enzyme was found upon its incubation with hydroxyurea, an inhibitor of the chloroplast enzyme of higher plants. The enzyme showed higher specificity with pyrogallol than with ascorbate. The amino acid sequences in the N-terminal region of Chlamvdomonas AsAP showed no significant similarity to any other AsAP from higher plants and Euglena . The enzyme had a molecular mass of 34 kDa. The Km values of the enzyme for ascorbate and H2O2 were 5.2±0.3 and 25±3.4 μ M , respectively. Hydrogen peroxide was generated at a rate of 6.1±0.8 μmol mg-1 chlorophyll h-1 in intact chloroplasts isolated from Chlamydomonas cells grown in the presence of Na-selenite, and it diffused from the organelles into the medium.  相似文献

The developmental profile of the activities of some enzymes involved in malate metabolism, namely phosphoenolpyruvate carboxylase (PEPC; EC 4. 1. 1. 31), NAD+-linked (EC 1. 1. 1. 37) and NADP+-linked (EC 1. 1. 1. 82) malate dehydrosenase (MDH), NAD+linked (EC 1. 1. 1. 39) and NADP+-linked (EC 1. 1. 1. 40) malic enzyme (ME), has been determined in leaves of peach [ Prunus persica (L.) Batsch cv. Maycrest], a woody C3 species. In order to study the role of these enzymes, their activities were related to developmental changes of photosynthesis, respiration, and capacity for N assimilation. Activities of PEPC, NAD(P)+-MDH and NADP+-ME were high in young expanding leaves and decreased 2- to 3-fold in mature ones, suggesting that such enzymes play some role during the early stages of leaf expansion. In leaves of peach, such a role did not seem to be linked to C3 photosynthesis or nitrate assimilation, in that photosynthetic O2 evolution and activities of nitrate reductase (EC 1. 6. 6. 1) and glutamine synthetase (EC 6. 3. 1. 2) increased during leaf development. In contrast, leaf respiration strongly decreased with increasing leaf age. We suggest that in expanding leaves of this woody species the enzymes associated with malate metabolism have anaplerotic functions, and that PEPC may also contribute to the recapture of respiratory CO2.  相似文献

A survey for the enzyme L-myo-inositol-1-phosphate synthase (EC 5.5.1.4) has been conducted among various members of the lower plant groups, mainly algac, bryophytes and fungi; some properties of the partially purified enzyme from Euglena gracilis Z . are presented. The enzyme was detected in Chloropycean algae, Marchantiales and the Basidiomycetous fungi. The enzyme from Euglena had a pH optimum at 7.5. The Km for glucose-6-P was 2.1 m M and for NAD+ 80 μ M . When assayed in the absence of added NAD+, the enzyme showed a basal activity suggesting the presence of bund NAD+ in the system. NH4Cl increased the enzyme activity two-fold, altough the enzyme was inactivated by (NH4)2SO4.  相似文献

Pectins from persimmon ( Diospyros kaki L.) fruit pericarp were sequentially extracted with 0. 05 M trans -1,2-diaminocyclohexane-N,N, N', N'-tetraacetic acid (CDTA), 0. 05 M Na2CO3 (1°C) and Na2CO3 (20°C) and the carbohydrate composition and metabolism during development determined. Young persimmon fruits contained a large proportion of pectins, 46% by dry weight, that decreased to 20% with ripening. This decrease occurred in the CDTA and Na2CO3 (1°C) fractions, mainly composed of uronic acids, and represents a net loss of uronic acids, arabinose and galactose. The amount of non-cellulosic neutral sugars was especially high in the Na2CO3 (20°C) fraction. The loss of pectins was also accompanied by a depolymerisation of the polysaccharides extracted in the three pectic fractions. However, none of these changes can be attributed to the action of polygalacturonase activity. Proteins were extracted from the pericarp tissue, but endopolygalacturonase (EC 3. 2. 1. 15) activity, determined as a decrease in viscosity of polygalacturonic acid, was not observed in the extract. Determination of exopolygalacturonase (EC 3. 2. 1. 67) activity by measuring the release of reducing groups from polygalacturonic acid was also negative. The results presented indicate that polygalacturonase is not responsible for the metabolism of pectins during persimmon fruit ripening.  相似文献

Multiple activity peaks of neutral invertase (EC 3.2.1.26) were found in chicory roots ( Cichorium intybus L. var. foliosum cv. Flash). The main activity peak was purified by a combination of anion-exchange chromatography, hydrophobic interaction chromatography, chromatofocusing and gel filtration. This protocol produced a 77-fold purification and a specific activity of 1.6 μmol (mg protein)−1 min−1. The mass of the enzyme was 260 kDa as estimated by gel filtration and 65 kDa on SDS-PAGE. Optimal activity was found between pH 7 and 7.5. The purified enzyme exhibited hyperbolic saturation kinetics with a Km between 10 and 20 mM for sucrose. No other products than glucose and fructose could be detected. Raffinose was hydrolyzed at a rate of 2.4% relative to sucrose whereas the enzyme did not hydrolyze maltose, cellobiose, trehalose, 1-kestose, 1.1-nystose or inulin. Neutral invertase activity was completely inhibited by HgCl2 and AgNO3 and partially inhibited by CoCl2, and ZnSO4 (1 mM). Pyridoxal phosphate (Ki≅ 500 μ M ), Tris (Ki≅ 1.2 m M ), glucose and fructose (Ki≅ 16 m M ) were strong inhibitors of the enzyme. Fructose and Tris behaved as competitive inhibitors. A possible role for the enzyme's activity in vivo is discussed.  相似文献

Ornithine decarboxylase (ODC, EC 4.1.1.17) was studied in crude extracts of parenchyma slices of dormant tubers activated for 12 h, tuber shoots and shoot apices. It was highest in shoot apices. The enzyme activity was measured by the production of 14CO2 from labelled ornithine; Vmax was 450 nmol (mg protein)-1h-1, Km for ornithine and pyridoxal phosphate were, respectively, 30 m M and 5μ M . Only when partially purified, the 14CO2 production was inhibited by α-difluoromethylornithine, while in crude extracts dithiothreitol was inhibitory. Ornithine and arginine decarboxylase (ADC, EC 4.1.1.19) activities from parenchyma tubers were not greatly altered by exogenously supplemented ornithine, even though its endogenous pool increased. Exogenously supplemented arginine enhanced ornithine decarboxylase activity, whereas putrescine decreased it slightly. The possibility of artifactual activities in the crude extract is also discussed.  相似文献

Plasma membrane vesicles were purified from 8-day-old oat ( Avena sativa L. cv. Brighton) roots in an aqueous polymer two-phase system. The plasma membranes possessed high specific ATPase activity [ca 4 μmol P1 (mg protein)−1 min−1 at 37°C]. Addition of lysophosphatidylcholine (lyso-PC) produced a 2–3 fold activation of the plasma membrane ATPase, an effect due both to exposure of latent ATP binding sites and to a true activation of the enzyme. Lipid activation increased the affinity for ATP and caused a shift of the pH optimum of the H+ -ATPase activity to 6.75 as compared to pH 6.45 for the negative H+-ATPase. Activation was dependent on the chain length of the acyl group of the lyso-PC, with maximal activition obtained by palmitoyl lyso-PC. Free fatty acids also activated the membrane-bound H+-ATPase. This activation was also dependent on chain length and to the degree of unsaturation, with linolenic and arachidonic acid as the most efficient fatty acids. Exogenously added PC was hydrolyzed to lyso-PC and free fatty acids by an enzyme in the plasma membrane preparation, presumably of the phospholipase A type. Both lyso-PC and free fatty acids are products of phospholipase A2 (EC 3.1.1.4) action, and addition of phospholipase A2 from animal sources increased the H+-ATPase activity within seconds. Interaction with lipids and fatty acids could thus be part of the regulatory system for H+-ATPase activity in vivo, and the endogenous phospholipase may be involved in the regulation of the H+-ATPase activity in the plasma membranne.  相似文献

Levels of activity of the sucrose catabolizing enzymes, acid invertase (EC 3.2.1.26) and sucrose synthase (EC 2.4.1.13), were measured during development of new leaves of Citrus sinensis (L.) Osbeck cv. Shamouti. Soluble acid invertase showed a peak activity of 32 nkat (g fresh weight)−1 at ca 60% of full leaf expansion and rapidly declined toward and after full expansion. There was no concomitant increase in an insoluble form of the enzyme. Sucrose synthase activity, measured in the synthesis direction, declined from 33% of full leaf expansion [10 nkat (g fresh weight)−1] 10, and following, full expansion. Highest sucrose synthase activity, measured in the cleavage direction, was 6 nkat (g fresh weight)−1 and showed little change during development. Acid invertase has a Km of 5 m M for sucrose, while sucrose synthase had a Km of 118 m M for sucrose. Changes in acid invertase activity correlated with changes in the reducing sugar:sucrose ratio. These results suggest that soluble acid invertase activity is the primary enzyme responsible for sucrose catabolism in the expanding Citrus leaf. Changes in leaf expansion rate and invertase activity did not correlate positively with changes in endogenous free IAA level, as determined by enzyme linked immunoassay.  相似文献

NADP+-dependent malic enzyme (L-malate : NADP+ oxidoreductase, decarboxylating, EC 1.1.1.40) was extracted from the leaves of yellow lupine. The purification procedure included fractionation with (NH4)2SO4 and Sephadex G-25 chromatography, followed by purification on DEAE-cellulose and Sephadex G-200 columns. The enzyme was purified 122-fold. The enzyme affinity towards L-malate was found to be significantly higher with Mn2+ than with Mg2+. The Hill coefficient for Mg2+ depended on concentration and was 1.6 for the lower and 3.9 for the higher concentrations. The dependence of the enzyme activity on NADP+ followed a hyperbolic curve. Km values and Hill coefficients for NADP+ were similar with both Mn2+ and Mg2+. The enzyme activity was strictly dependent on divalent cations and followed a sigmoidal curve at least for Mg2+. The enzyme had 4-fold higher affinity towards Mn2+ than towards Mg2+, the Km values being 0.3 and 1.15 m M respectively. Of several tested organic acids, oxalate was the most effective inhibitor followed by oxaloacetate while succinate was the strongest activator.  相似文献

Stomatal phosphoenolpyruvate carboxylase (PEPCase EC 4.1.1.31), extracted from abaxial epidermal peels of Vicia faba L. cv. Frühe Weiβkeimige, was partially purified by ammoniumsulfate precipitation, and molecular sieve (Sepharosc S-400) and ion exchange (DEAE-Sepharose) chromatography. The partially purified enzyme, essentially free of a PEPCase isoform existing in mesophyll and epidermal cells, had a specific activity of 300 nkat mg-1 protein at 25°C. Western immunoblot analysis revealed that the stomatal enzyme had two bands (M: of 110000 and 112000), crossreacting with PEPCase antibodies raised against PEPCase from Ka-lanchoe daigremontiana . The native molecular mass of the enzyme (467000) points to a tetrameric subunit structure. The temperature optimum was found to be 35°C; cold treatments of PEPCase before assaying were accompanied by inactivation. The energy of activation was calculated to 51 kJ mol-1. The kinetic behaviour of the enzyme at fixed MgCl2 concentrations is characterized by a pH optimum between pH 8.0–8.2 with or without 1 m M malate or 5 m M glucose-6-phosphate (Glc-6-P), but a combination of both effectors resulted in a shift of the optimum to pH 7.6. The enzyme showed a pH sensitive inhibition by 1 m M malate and an activation by Glc-6-P. At low pH (6–7), Glc-6-P was able to compensate for the malate induced inhibition of the enzyme. Malate and Glc-6-P both affected Km(PEP), drastically and influenced Vmax at pH 7, but not at pH 8.3. The inhibition constant of malate was determined to be 1.2 m M at pH 7. From the Dixon plot, a competitive inhibition of malate was assumed under defined assay conditions.  相似文献
