Activation of cholinergic neurons in the brain is produced by administration of the acetylcholinesterase inhibitors physostigmine and diisopropylfluorophosphate (DFP). This activation has a biphasic effect on tyrosine hydroxylase (EC 4.14.3-) activity. The acute effect of DFP, 1 mg/kg, intraperitoneally, or physostigmine, 0.2 mg/kg, intravenously, or 10 mug, intraventricularly, was a rapid reduction in tyrosine hydroxylase activity in the hypothalamus. The activities of DOPA decarboxylase (EC 4.1.1.28) and dopamine-beta-hydroxylase (EC 1.14.17.1) were not changed. In contrast to the acute effect, chronic administration of physostigmine, 0.2 mg/kg, intravenously, twice daily for 7 days produced an increase in tyrosine hydroxylase activity in the hypothalamus. The rapid acute effects may be due to an allosteric inactivation of tyrosine hydroxylase, while the chronic effects may reflect enzyme induction.  相似文献

Exposure to cold caused an increase in AChE activity of the different brain regions of both Arvicanthis and Columbia and a decrease in the enzyme activity of Scincus midbrain. Heat exposure provoked variable changes in AChE activity of the various brain regions of the three experimental species. The changes in AChE activities may be one of the mechanisms by which birds and mammals tend to acclimatize themselves to various forms of stress. In reptiles, AChE activity varied with changes in ambient temperature and this is probably due to the adaptive significance of thermally directed changes in enzyme substrate affinity.  相似文献

Effect of acute, chronic-predictable and chronic-unpredictable stress on memory and acetylcholinesterase (AChE) was investigated in rats. The animals were subjected to 3 type of stressors--(1) acute immobilization stress, (2) chronic-predictable stress i.e., immobilization daily for 5 consecutive days and (3) chronic-unpredictable stress that included reversal of light/dark cycle, over-night fasting, forced-swimming, immobilization and forced exercise in random unpredictable manner daily for 5 consecutive days. Learning and memory function was studied by single trial Passive avoidance test. AChE activity was assayed spectrophotometrically in the detergent (DS) and salt (SS) soluble fractions in different brain regions. Learning was obtained in acute and chronic-predictable stress groups but not in chronic-unpredictable group. Acute, chronic-predictable and chronic-unpredictable stress caused significant decrease in AChE activity in the DS fraction of cortex, hippocampus and hypothalamus as compared to control. Results indicate that AChE in DS fraction is predominantly affected in stressed and stressed-trained group but cognition is affected only by chronic-unpredictable stress. In acute and chronic-predictable groups the decreased AChE activity in the hippocampal DS fraction during learning may be responsible to maintain cognitive function by enhancing the cholinergic activity.  相似文献

The effect of reserpine and meprobamate on the total acetylcholine content and acetylcholinesterase activity in the brain tissue of the kusu rat, Arvicanthis niloticus, was studied. The total acetylcholine content and acetylcholinesterase activity were determined 1 hr after i.p. injection of different doses of reserpine (0.25, 0.5 and 1 mg/ml/100 g body wt) and meprobamate (6.25, 12.5 and 25 mg/ml/100 g body wt). The effect of different time intervals (1, 10, 30 min, 1, 2.5, 5, 8, 12, 24 and 48 hr) on the total acetylcholine content and acetylcholinesterase activity was investigated after i.p. injection of 0.5 mg of reserpine and 12.5 mg of meprobamate/ml/100 g body wt. Both reserpine and meprobamate caused an increase in the total ACh content in the brain tissue of Arvicanthis niloticus which was suggested to be due to a decrease in the release of ACh, since both reserpine and meprobamate inhibited AChE activity after some tested periods. The effect of meprobamate was observed to be stronger than that of reserpine.  相似文献

A two-fold increase in acetylcholine, that can randomly be released by brain synaptosomes, is registered 60 min following whole-body X-irradiation of rats with a dose of 0.21 C/kg; depolarization of the synaptosome membranes by potassium chloride increases the release of acetylcholine the augmentation of the release in this case being lower than that in the control. The initial rate of spontaneous neuromediator release from synaptosomes grows by 80 per cent whereas after depolarization of synaptosome membranes by potassium chloride, by 15 per cent. There is a 2.5-fold increase in the maximum rate of a highly specific uptake of choline with Km value being constant. Acetylcholine content of gray substance of irradiated rat brain is invariable.  相似文献

Eight hours after a single tube-feeding of tryptophan the activity of acetylcholinesterase in the cerebral hemisphere of well-fed (non-fasted) normal and adrenalectomized rats was 28 and 53% higher, respectively, compared to the corresponding water-fed control. On the other hand, the enzyme activity in the cerebellum of both normal and adrenalectomized rats remained essentially unchanged following tryptophan administration. Pretreatment of adrenalectomized rats with actinomycin-D totally abolished the tryptophan-mediated stimulation of cerebral acetylcholinesterase activity. The pattern of response of cerebral acetylcholinesterase in well-fed adrenalectomized rats over a period of 24-hr following a dose of tryptophan was found to be biphasic.  相似文献

The effect of ovariectomy and estrogen treatment on the brain acetylcholinesterase activity and cognition in rats was investigated in this study. Ovariectomized and nonovariectomized rats were treated subcutaneously with estradiol dipropionate for 8 d. In the single-trial, passive-avoidance test all the groups showed significant learning and retention of memory as evident by the increase in transfer latency time in trial 2 as compared with trial 1. No-transfer response was significantly increased in the estradiol-dipropionate-treated ovariectomized (80%) and nonovariectomized (60%) group as compared with the ovariectomized (30%) group. Specific activity of acetylcholinesterase was assayed spectrophotometrically in salt-soluble and detergent-soluble fractions of various brain areas: frontal cortex, cerebral cortex, striatum, hippocampus and hypothalamus, thalamus, pons, medulla, and cerebellum. The effect of ovariectomy and estradiol dipropionate was varied in both fractions of these brain areas. Estradiol dipropionate treatment could restore the acetylcholinesterase activity to the control level only in the detergent-soluble fraction of hypothalamus and salt-soluble fraction of hypothalamus, thalamus, and medulla in ovariectomized rats. The results indicate that ovariectomy alters acetylcholinesterase activity in the brain areas but not in a uniform manner and affects only qualitative aspects of cognitive function, which could be improved by estrogen supplementation.  相似文献

The effect of muscle training and hypokinesia on the content of catecholamines in the adrenals of rats in the postnatal ontogenesis has been studied. A systematic swimming training promotes the process of catecholamine accumulation in the adrenals, while hypokinesia slows down that process. An unitary carrying-out of maximum swimming exertion causes a sharp decrease of total content and concentration of catecholamines, at this their minimum concentration in the adrenals of developing animals does not depend on the adaptation to the locomotive regime.  相似文献

Changes in the noradrenaline (NA) content in the hypothalamus, dopamine (DA) and homovanillic acid (HVA) in the striatum were determined in rats after chronic alcohol administration. A single injection of alcohol (2.5 g/kg i.p.) provoked a 30% decrease in NA only in rats predisposed to ethanol intake. Voluntary intake of 15% ethanol for 10 days made the NA content return to normal, the 4-month use of ethanol did not change whereas the 8-month use reduced the NA content by 17%, DA by 31% and raised the content of HVA by 25%. Twenty-four hours after alcohol abstinence the HVA content dropped by 13%. It is concluded that the noradrenergic system is involved in the formation and development of alcohol motivation and that the dopaminergic system participates in the development of the physical dependence and abstinence.  相似文献

The homocarnosine content and homocarnosine synthetase activity were studied in the brain of rats in normal state and under hyperoxia. The homocarnosine content is higher in phylogenetically old brain areas as compared with that in the cerebral hemispheres. Its nonuniform distribution in the brain is associated with different activity of homocarnosine-carnosine synthetase in the corresponding brain areas. At the preconvulsive stage of oxygen poisoning the homocarnosine content in all the brain areas does not change, the homocarnosine-carnosine synthetase activity is 32% lower. At the convulsive stage of hyperoxia the homocarnosine amount in the cerebral hemisphere decreases by 33%, in the midbrain and diencephalon -- by 70, in the medulla oblongata -- by 60, in the cerebellum -- by 58%. The decrease in the homocarnosine content correlates with that in the activity of homocarnosine-carnosine synthetase in the corresponding brain areas; in the cerebral hemispheres -- by 33%, in the midbrain and diencephalon -- by 50, in the medulla oblongata -- by 49, in the cerebellum -- by 40%.  相似文献

Summary Areas containing AChE-positive capillaries were mapped in the brain of the cat and the guinea pig. Regions with AChE-positive capillaries mostly also contain neuronal elements with AChE activity. Electron-microscopical cytochemistry revealed localization of AChE in basement membranes of endothelial cells and pericytes very often in continuity with activity of the extracellular space. Intraendothelial AChE activity was seen only in pinocytic vesicles. The vascular AChE is thought to be of neuronal origin since no cytochemical evidence has been obtained for a synthesis of this enzyme in endothelial or other non-neuronal cells in the CNS.Drs. H. The level of acetylcholinesterase (AChE) in brain regions of O. mossambicus at different intervals showed the extent of phosalone toxicity. Significant inhibition of AChE at the end of 96 hr in the brain regions was observed. In contrast to AChE inhibition, the monoamine oxidase (MAO) activity showed significant increase in the regions of cerebral hemispheres, dien/mesencephalon, cerebellum and medulla oblongata. The increase of MAO activity in the brain regions under phosalone toxicity is considered to be one of the mechanisms to maintain the amines level in O. mossambicus.  相似文献

In order to test the effects of chronic lithium (Li) administration on learning and memory, 21 day old rats were subjected to different degrees of environmental stimulation (enriched condition, EC and impoverished condition, IC) with and without Li for 144 days. Li was administered with food (2.18 mEq/Kg weight/day). Average plasma Li concentration at the end of the experiment was 0.41 +/- 0.04 mu Eq/ml. Both Li treatment and the environmental condition showed an overall significant effect on the cortex/subcortex weight ratio and learning ability index, but not on AChE activity in occipital cortex. A similar pattern of brain Li distribution was observed in both EC-Li and IC-Li, with occipital cortex having the highest levels. Li tissue/protein/plasma ratio was higher in EC than in IC, in all the brain areas studied. Other organs (liver and kidney) did not show EC-IC differences in the tissue/protein/plasma Li ratio.  相似文献
