The interaction of K+ with mammalian ribosomes was studied by equilibrium dialysis and compared with that of other univalent cations. The heavy K+ analogue, Tl+, binds more firmly than K+ to ribosomes and, unlike K+, has a practically useful isotope. With 204Tl+ as a marker of K+-selective binding the ribosome-cation interaction could be followed down to levels below 0.1 average Tl+-occupied site per ribosome. The Tl+/ribosome ratio varied with the free Tl+ concentration in a multiple way. At high Tl+ saturation Tl+ was easily displaced by Mg2+. With decreasing Tl+ saturation the competitive activity of Mg++ was strikingly reduced, indicating that Tl+ and Mg++ compete with different efficiency for different classes of sites.The experiments on univalent cations were performed at 1.5 mM Mg2+ under two complementary conditions: (1) Ribosomes were pretreated with 5 × 10?2, 5 × 10?3, and 5 × 10?4 M LiNO3, NaNO3, KNO3, and CsNO3, and then equilibrated with different concentrations of 204TlNO3 in the same buffers. (2) Ribosomes were pretreated with 10?2, 10?4, and 10?6 M 204TlNO3, and then equilibrated with different concentrations of LiNO3, NaNO3, KNO3, and CsNO3 (displacement experiments). At high Tl+ saturation Na+ and Li+ were about as active as K+ and Cs+ in competing with 204Tl+. With decreasing Tl+ saturation a differentiation occurred in favor of K+ and Cs+, with some preference for K+. It is concluded that ribosomes contain a limited number of sites with pronounced ion specificity. Of physiological cations K+ is most firmly bound to these sites.  相似文献

Purpose

The goal of this study was to compare the tumor uptake kinetics and diagnostic value of three 68Ga-DOTA-labeled somatostatin analogues (68Ga-DOTATOC, 68Ga-DOTANOC, and 68Ga-DOTATATE) using PET/CT in a murine model with subcutaneous meningioma xenografts.

Methods

The experiment was performed with 16 male NUDE NU/NU mice bearing xenografts of a human meningioma cell line (CH-157MN). 68Ga-DOTATOC, 68Ga-DOTANOC, and 68Ga-DOTATATE were produced in a FASTLab automated platform. Imaging was performed on an Argus small-animal PET/CT scanner. The SUVmax of the liver and muscle, and the tumor-to-liver (T/L) and tumor-to-muscle (T/M) SUV ratios were computed. Kinetic analysis was performed using Logan graphical analysis for a two-tissue reversible compartmental model, and the volume of distribution (Vt) was determined.

Results

Hepatic SUVmax and Vt were significantly higher with 68Ga-DOTANOC than with 68Ga-DOTATOC and 68Ga-DOTATATE. No significant differences between tracers were found for SUVmax in tumor or muscle. No differences were found in the T/L SUV ratio between 68Ga-DOTATATE and 68Ga-DOTATOC, both of which had a higher fraction than 68Ga-DOTANOC. The T/M SUV ratio was significantly higher with 68Ga-DOTATATE than with 68Ga-DOTATOC and 68Ga-DOTANOC. The Vt for tumor was higher with 68Ga-DOTATATE than with 68Ga-DOTANOC and relatively similar to that of 68Ga-DOTATOC.

Conclusions

This study demonstrates, for the first time, the ability of the three radiolabeled somatostatin analogues tested to image a human meningioma cell line. Although Vt was relatively similar with 68Ga-DOTATATE and 68Ga-DOTATOC, uptake was higher with 68Ga-DOTATATE in the tumor than with 68Ga-DOTANOC and 68Ga-DOTATOC, suggesting a higher diagnostic value of 68Ga-DOTATATE for detecting meningiomas.  相似文献

Nigericin is a monocarboxylic polyether molecule described as a mobile K+ ionophore unable to transport Li+ and Cs+ across natural or artificial membranes. This paper shows that the ion carrier molecule forms complexes of equivalent energy demands with Li+, Cs+, Na+, Rb+, and K+. This is in accordance with the similar values of the complex stability constants obtained from nigericin with the five alkali metal cations assayed. On the other hand, nigericinalkali metal cation binding isotherms show faster rates for Li+ and Cs+ than for Na+, K+, and Rb+, in conditions where the carboxylic proton does not dissociate. Furthermore, proton NMR spectra of nigericin-Li+ and nigericin-Cs+ complexes show wide broadenings, suggesting strong cation interaction with the ionophore; in contrast, the complexes with Na+, K+, and Rb+ show only clear-cut chemical shifts. These latter results support the view that nigericin forms highly stable complexes with Li+ and Cs+ and contribute to the explanation for the inability of this ionophore to transport the former cations in conditions where it catalyzes a fast transport of K+>Rb+>Na+.Part of the results of this paper were presented at the 14th International Congress of Biochemistry in Prague, Czechoslovakia.  相似文献

Summary When Chlorella pyrenoidosa photoassimilates 3H–14C-acetate glycollic acid rapidly becomes labelled with both tritium and 14C. The 3H/14C ratio was 10 in glycollate, (compared with 4 in the acetate added) and the only other intermediates showing similar 3H/14C ratios to glycollate were glycerate and serine. This suggests a glycollate pathway for the formation of serine was operating in Chlorella pyrenoidosa during the photoassimilation of acetate. When Chlorella pyrenoidosa assimilated 3H–14C-acetate in the dark glycollate was not labelled with either 14C or tritium. Although glycerate and serine both became labelled with 14C and tritium in the dark they did not show the high 3H/14C ratios recorded in the light. When cells were aerated with unlabelled 5% CO2 during the photoassimilation of 3H–14C-acetate, the 3H/14C ratios of glycollate, glycerate and serine were slightly decreased. Similarly, under anaerobic conditions in the light the 3H/14C ratio was decreased compared with aerobic conditions.  相似文献

1. (1) VO3 combines with high affinity to the Ca2+-ATPase and fully inhibits Ca2+-ATPase and Ca2+-phosphatase activities. Inhibition is associated with a parallel decrease in the steady-state level of the Ca2+-dependent phosphoenzyme.
2. (2) VO3 blocks hydrolysis of ATP at the catalytic site. The sites for VO3 also exhibit negative interactions in affinity with the regulatory sites for ATP of the Ca2+-ATPase.
3. (3) The sites for VO3 show positive interactions in affinity with sites for Mg2+ and K+. This accounts for the dependence on Mg2+ and K+ of the inhibition by VO3. Although, with less effectiveness, Na+ substitutes for K+ whereas Li+ does not. The apparent affinities for Mg2+ and K+ for inhibition by VO3 seem to be less than those for activation of the Ca2+-ATPase.
4. (4) Inhibition by VO3 is independent of Ca2+ at concentrations up to 50 μM. Higher concentrations of Ca2+ lead to a progressive release of the inhibitory effect of VO3.

Side-by-side with inhibition of the Na+,K+-ATPase ouabain and other cardiotonic steroids (CTS) can affect cell functions by mechanisms other than regulation of the intracellular Na+ and K+ ratio ([Na+]i/[K+]i). Thus, we compared the doseand time-dependences of the effect of ouabain on intracellular [Na+]i/[K+]i ratio, Na+,K+-ATPase activity, and proliferation of human umbilical vein endothelial cells (HUVEC). Treatment of the cells with 1-3 nM ouabain for 24-72 h decreased the [Na+]i/[K+]i ratio and increased cell proliferation by 20-50%. We discovered that the same ouabain concentrations increased Na+,K+-ATPase activity by 25-30%, as measured by the rate of 86Rb+ influx. Higher ouabain concentrations inhibited Na+,K+-ATPase, increased [Na+]i/[K+]i ratio, suppressed cell growth, and caused cell death. When cells were treated with low ouabain concentrations for 48 or 72 h, a negative correlation between [Na+]i/[K+]i ratio and cell growth activation was observed. In cells treated with high ouabain concentrations for 24 h, the [Na+]i/[K+]i ratio correlated positively with proliferation inhibition. These data demonstrate that inhibition of HUVEC proliferation at high CTS concentrations correlates with dissipation of the Na+ and K+ concentration gradients, whereas cell growth stimulation by low CTS doses results from activation of Na+,K+-ATPase and decrease in the [Na+]i/[K+]i ratio.  相似文献

High performance liquid chromatography (HPLC) coupled with specific radioimmunoassays for methionine-enkephalin-Arg6-Gly7-Leu8 (Met-E-Arg6-Gly7-Leu8), methionine-enkephalin (Met-E), leucine-enkephalin (Leu-E) and methionine-enkephalin-Arg6-Phe7 (Met-E-Arg6-Phe7) has demonstrated that Met-E-Arg6-Gly7-Leu8 exists together with Met-E, Leu-E and Met-E-Arg6-Phe7 in the brain of guinea pig, rat and golden hamster. The content of Met-E-Arg6-Gly7-Leu8 was comparable to those of Leu-E and Met-E-Arg6-Phe7, whereas that of Met-E was the highest among the four opioid peptides. These results are compatible with the recent studies on the nucleotide sequence of cloned cDNA for preproenkephalin from bovine adrenal medulla, which reveal that this precursor molecule contains four copies of Met-E and one copy each of Leu-E, Met-E-Arg6-Phe7 and Met-E-Arg6-Gly7-Leu8. The co-existence of Met-E-Arg6-Gly7-Leu8 with Met-E, Leu-E and Met-E-Arg6-Phe7 suggests that their biosynthetic pathway in the brain is similar to that in the adrenal medulla.  相似文献

The major histocompatibility (B) complex of a distinct commercial pure White Leghorn chicken line was characterized using serological, biochemical and restriction fragment length polymorphism (RFLP) typing. Line B chickens displayed a high recombination frequency within the B complex. Three recombinant haplo-types were identified. The influence of these haplotypes was determined in relation to the haplotypes Bl9 and B21 on their resistance to Marek's disease (MD) in an experimental infection with the virus. Offspring of sires with a recombinant haplotype in combination with B19 or B21, and dams, which were homozygous B19/B19 or B21/B21 were infected. The B type of the offspring had a significant effect upon survival. Animals with B complex types B21/B21, B134/B21 and B234/B21 were relatively resistant to MD (24–32% mortality), whereas B19/B19 birds were highly susceptible (68% mortality). Animals with a recombinant halpotype B19r21 (B-G21, B-F19) were equally susceptible to MD as birds with the complete B19 haplotype. In contrast to earlier publications, resistance was not inherited as a dominant trait. Apparently, B19 was associated with a dominant susceptibility. The gene(s) associated with the B complex and involved in resistance to MD were localized within the B-F/B-L region. However, the association with a presumably non-coding subregion of B-G could not be excluded.  相似文献

Myeloid-derived suppressor cells are increased in the peripheral blood of advanced-stage cancer patients; however, no studies have shown a correlation of these immunosuppressive cells with clinical outcomes in melanoma patients. We characterized the frequency and suppressive function of multiple subsets of myeloid-derived suppressor cells in the peripheral blood of 34 patients with Stage IV melanoma, 20 patients with Stage I melanoma, and 15 healthy donors. The frequency of CD14+ MDSCs (Lin? CD11b+ HLA-DR? CD14+ CD33+) and CD14? MDSCs (Lin? CD11b+ HLA-DR? CD14? CD33+) was increased in the peripheral blood of Stage IV melanoma patients relative to healthy donors. The frequency of CD14+ and CD14? MDSCs correlated with each other and with the increased frequency of regulatory T cells, but not with classically defined monocytes. CD14? MDSCs isolated from the peripheral blood of Stage IV melanoma patients suppressed T cell activation more than those isolated from healthy donors, and the frequency of these cells correlated with disease progression and decreased overall survival. Our study provides the first evidence that the frequency of CD14? MDSCs negatively correlates with clinical outcomes in advanced-stage melanoma patients. These data indicate that suppressive MDSCs should be considered as targets for future immunotherapies.  相似文献

Magnetic isotope and magnetic field effects on the rate of DNA synthesis catalysed by polymerases β with isotopic ions 24Mg2+, 25Mg2+ and 26Mg2+ in the catalytic sites were detected. No difference in enzymatic activity was found between polymerases β carrying 24Mg2+ and 26Mg2+ ions with spinless, non-magnetic nuclei 24Mg and 26Mg. However, 25Mg2+ ions with magnetic nucleus 25Mg were shown to suppress enzymatic activity by two to three times with respect to the enzymatic activity of polymerases β with 24Mg2+ and 26Mg2+ ions. Such an isotopic dependence directly indicates that in the DNA synthesis magnetic mass-independent isotope effect functions. Similar effect is exhibited by polymerases β with Zn2+ ions carrying magnetic 67Zn and non-magnetic 64Zn nuclei, respectively. A new, ion–radical mechanism of the DNA synthesis is suggested to explain these effects. Magnetic field dependence of the magnesium-catalysed DNA synthesis is in a perfect agreement with the proposed ion–radical mechanism. It is pointed out that the magnetic isotope and magnetic field effects may be used for medicinal purposes (trans-cranial magnetic treatment of cognitive deceases, cell proliferation, control of the cancer cells, etc).  相似文献

The activation of desoxyribonuclease on desoxyribonucleate, known to occur with Mg++ and Mn++, has been shown to occur equally well with Co++, to nearly the same extent with Fe++, and to a lesser extent with Ca++, Ba++, Sr++, Ni++, Cd++, and Zn++. The conditions under which the optimal activation is revealed vary among these ions. Thus, Mg++, Mn++, and Co++ may show marked activation under conditions in which Fe++ is nearly ineffective. Since too high a concentration of an ion may be as ineffective as too little, concentration-activation curves were determined for each ion. Per micromole of nucleic acid phosphorus, the optimal effective amount of each ion in micromoles is as follows: Mg++ 3, Mn++ 3, Co++ 3, Fe++ 0.3, Ni++ 0.3, Ba++ 1.7, Ca++ 3, Sr++ 3, Zn++ 0.3, and Cd++ 0.3.The optimum pH for the activation with Mg++, Co++, and Ca++ is about 6.5, that with Fe++ is at 5.7, while Mn++ shows two optima at pH 6.8 and 8.0.Experiments conducted in Pyrex and in quartz vessels showed the same results, and indicated that there was no activation of desoxy-ribonuclease in the absence of added salts.  相似文献

A set of three experiments is described which correlate aromatic resonances of histidine and tryptophan residues with amide resonances in 13C/15N-labelled proteins. Provided that backbone 1H and 15N positions of the sequentially following residues are known, this results in sequence-specific assignment of histidine 1Hδ2/13Cδ2 and 1Hε1/13Cε1 as well as tryptophan 1Hδ1/13Cδ1, 1Hζ2/13Cζ 2, 1Hη2/13Cη2, 1Hε3/13Cε3, 1Hζ3/13Cζ3 and 1Hε1/15Nε1 chemical shifts. In the reverse situation, these residues can be located in the 1H–15N correlation map to faciliate backbone assignments. It may be chosen between selective versions for either of the two amino acid types or simultaneous detection of both with complete discrimination against phenylalanine or tyrosine residues in each case. The linkages between δ-proton/carbon and the remaining aromatic as well as backbone resonances do not rely on through-space interactions, which may be ambiguous, but exlusively employ one-bond scalar couplings for magnetization transfer instead. Knowledge of these aromatic chemical shifts is the prerequisite for the analysis of NOESY spectra, the study of protein–ligand interactions involving histidine and tryptophan residues and the monitoring of imidazole protonation states during pH titrations. The new methods are demonstrated with five different proteins with molecular weights ranging from 11 to 28 kDa.  相似文献

Bumetanide is a potent diuretic drug which has some structural features in common with furosemide. The steady-state exchange of K+ and Cl? was investigated in Ehrlich ascites tumor cells treated with bumetanide. This agent did not alter the cellular content of K+ or Cl? but the self-exchange of both ions was depressed. K+ self-exchange was inhibited by 55% at bumetanide concentrations as low as 10?6 M. Cl? self-exchange was less sensitive to this drug but at low concentrations (between 10?6 and 10?3 M) bumetanide was a more effective inhibitor of Cl? transfer than furosemide. The steady-state K+ flux of cells equilibrated in NO3? media was compared with the K+ flux in cells treated with 10?4 or 10?3 M bumetanide; the Cl? -sensitive K+ exchange was equivalent to the bumetanide-sensitive K+ exchange. Since the results suggested that a bumetanide-sensitive (Cl?, K+) cotransport could be operative in steady-state cells, the stoichiometry of the bumetanide-sensitive fluxes was determined by measuring Cl? and K+ fluxes simultaneously in the same cell suspension. At 5 · 10?4 and 10?3 M bumetanide concentrations, the ratio of these fluxes was 0.98 ? 0.07 (S.E.) and 1.04 ? 0.06, respectively, consistent with the postulated cotransport mechanism. At 10?4 and 10?5 M, however, the ratio of the bumetanide-sensitive Cl?/K+ flux was significantly less than 1.0. Since the magnitude of the bumetanide-sensitive K+ flux at 10?4 M was close to that of the Cl?-sensitive flux, a ratio of less than 1.0 at this drug level indicates that Cl? sensitivity and drug sensitivity may not reflect inhibition of the same process under all circumstances.  相似文献

The antagonistic effect of calcium (Ca2+), zinc (Zn2+) and selenium (Se4+) at different concentrations (10−2–10−6 M) against cadmium (Cd2+) induced genotoxic effects in root cells of Hordeum vulgare were studied. The results showed that 10−3–10−5 M could induce chromosomal aberrations and micronuclei formation. But in the treatment with 10−2–10−6 M of Ca2+, Zn2+ and Se4+ together with Cd2+ (10−3–10−5 M), respectively, the frequencies of chromosomal aberrations and micronuclei effectively decreased after 48 h of treatment. The treatment with 10−4–10−6 M of Ca2+ together with 10−4–10−5 M Cd2+, 10−6 M of Zn2+ together with 10−5 M Cd2+ and 10−6 M of Se4+ together with 10−5 M Cd2+ suggested rather obvious antagonistic effects. The order of the antagonisms of Ca2+, Se4+ and Zn2+ against Cd2+ toxicity was Ca2+>Se4+>Zn2+. The degree of antagonisms of Ca2+, Se4+ and Zn2+ against Cd2+ related to their concentration ratio.  相似文献

Rabbit lymph node cells (Ig+Ig?) were rosetted with anti-Ig antibody-coated erythrocytes and the rosetted Ig+ cells (B cells) were separated from unrosetted Ig? cells (T cells) by centrifugation through Ficoll-Hypaque medium. The Ig? cells were recovered from the top and the Ig+ cells from the bottom of the Ficoll-Hypaque layer. Some of the purified Ig+ cells lost their ability to form rosettes when cultured with the mitogen associated with streptolysin O. This suggested that the Ig+ population might contain two distinct subpopulations. The response of Ig+Ig?, Ig+, and Ig? cells to various mitogens was studied. The Ig? cells incorporated more 3H-TdR when they were incubated by themselves than when they were cultured with Ig+ cells in an Ig+Ig? culture. On the other hand, the Ig+ cells incorporated less 3H-TdR when they were incubated by themselves than when they were incubated with Ig? cells in an Ig+Ig? culture. Thus, Ig+ cells suppressed the response of Ig? cells whereas Ig? cells enhanced the response of Ig+ cells. We conclude that rabbit Ig+ cells (B cells) and Ig? cells (T cells) interact with a feedback pattern of regulation.  相似文献

Relationships among several of the ion movements associated with the acrosome reaction of S. purpuratus were investigated. Egg jelly initiates 45Ca2+ and 22Na+ uptake, and K+ and H+ efflux. H+ efflux and 22Na+ uptake occur with approximately equivalent stoichiometries as rapidly as the appearance of acrosomal rods, perhaps reflecting a linked process. Most K+ loss, as measured either by 42K+ efflux or K+-ion-selective electrodes, occurs after the acrosome reaction is complete. Since an elevation of seawater K+ (from 10 to 15 mM) or the addition of 0.5 mM tetraethylammonium (TEA), an inhibitor of K+ channels, inhibits the acrosome reaction half-maximally, K+ movements or alterations of K+-dependent membrane potentials may regulate the triggering by jelly. Most, but not all, of the 45Ca2+ influx is inhibited with a mixture of 10 μM FCCP, 1 mM CN?, and 2 μg/ml oligomycin, suggesting that the mitochondria store most of the Ca2+. The extracellular Na+ concentration affects Ca2+ fluxes: sperm placed into 5 mM Na+ seawater have enhanced 45Ca2+ uptake, but do not undergo the acrosome reaction, unless 30 mM Na+ is also added. Low Na+ concentrations lead to spontaneous triggering, by allowing for both Ca2+ influx and Na+-dependent H+ efflux. At least one early Ca2+ requirement precedes the Na+ and H+ movements, as inferred from attempts at reversing the inhibitors of jelly induction of the acrosome reaction. When sperm are incubated with jelly in the absence of Ca2+, then washed and incubated with jelly in the presence of Ca2+, the acrosome reaction is triggered only upon the second incubation. However, when sperm are mixed with jelly in the presence of the other inhibitors (verapamil, TEA, 5 mM Na+ seawater, low pH, or elevated K+), they are altered so that even upon subsequent washing, jelly-mediated triggering is no longer possible. This suggests the existence of an intermediate state in the reaction pathway, that follows an event for which Ca2+ is required, but that precedes the Na+ and H+ movements, which are inhibited by all inhibitors of the acrosome reaction. These data are used to develop a partial sequence of ionic changes associated with the triggering mechanism.  相似文献

Uptake of Co2+ by three nickel-resistant strains (NiR1, NiR2, and NiR3) ofNeurospora crassa that differed in resistance to Co2+ has been studied. Uptake was linear with Co2+ concentration (up to 1 mM), with time (up to 6 h), and with pH between 3 and 6. Uptake rates were in the order NiR2>NiR1>NiR3. In all strains, there was gradual increase in Co2+ uptake between 10° and 28°C, with a much sharper increase between 28° and 40°C. Metabolic inhibitors decreased Co2+ uptake partially in all strains, except for KF in NiR3. About 50–80 g Co2+/100 mg dry weight was surface bound. Ni2+, Zn2+, and Mn2+ competed with Co2+, the effects being strain specific. Mg2+ inhibited Co2+ uptake in all strains with preformed mycelia. In NiR1 and NiR2 only with young mycelia (40 h old) was Mg2+ inhibitory to Co2+ uptake,during growth in the presence of Co2+. The results suggested the presence of two transport systems for Co2+ in NiR1 and NiR2, only one of which was sensitive to Mg2+; in contrast, NiR3 had a single system, which was sensitive to Mg2+.  相似文献

The specificity of thermitase (EC 3.4.21.14), a microbial thermostable serine proteinase fromThermoactinomyces vulgaris, with several oligo- and polypeptide substrates was investigated. Preferred hydrolysis of peptide bonds with a hydrophobic amino acid at the carboxylic site was observed. The proved carboxypeptidolytic splitting of Leu5-enkephalin and bradykinin, as well as the noncleavability of casomorphins by thermitase, can be explained by the position of the glycine and proline residues in these substrates. Major cleavage sites in the oxidized insulin B chain in a 15-min incubation with thermitase at Gln4-His5, Ser9-His10, Leu11-Val12, Leu15-Tyr16 and in the oxidized insulin A chain at Cys SO3H11-Ser12, Leu13-Tyr14, and Leu16-Glu17 were observed. Additional cleavages of the bonds His5-Leu6, Arg22-Gly23, Phe24-Phe25, Phe25-Tyr26, and Tyr26-Thr27 in the oxidized B chain and Cys SO3H6-Cys SO3H7 and Tyr19-Cys SO3H20 in the oxidized A chain in 2-h incubations with thermitase were also noted. Hydrolysis of salmine A I component in a 10-min incubation was observed mainly at four peptide bonds: Arg5-Ser6, Ser6-Ser7, Arg18-Val19, and Gly27-Gly28. The cleavage sites of thermitase in both insulin chains were similar to those reported in the studies of subtilisins.  相似文献

Cytokinin-dependent tobacco (Nicotiana tabacum) tissue was grown in the presence of N6-benzyladenine (6-benzylaminopurine) labeled in the phenyl ring with 3H and in the 8-position of the purine ring with 14C. The 3H/14C ratio of N6-benzyladenosine recovered from labeled tobacco callus transfer RNA preparations was compared with the corresponding ratio for N6-benzyladenine supplied in the medium. By this means, with suitable controls, the incorporation of the synthetic cytokinin, N6-benzyladenine, into the transfer RNA of tobacco callus tissue was shown to involve the intact moiety.
