Single unit responses elicited through noxious mechanical stimulation of orofacial receptive fields were recorded, with glass micro-electrodes, within the rat ventrobasal complex of the thalamus. The evoked activities were compared before and after de-afferentation of the caudal sub-nucleus by trigeminal tractotomy at the level of the obex. Only units responding to noxious stimulation of oral receptive fields were unaffected by tractotomy. These results provide evidence that the rostral part of the spinal nucleus is involved in trigeminal painful sensation.  相似文献

Dentin of human teeth is a vital hydrated tissue. It is strongly sensitive to dehydration and drying that are commonly used in preparation of samples for scanning electron microscopy. Experience in examination of dentine surfaces of extracted human third molars using contact mode atomic force microscopy under moist conditions is described. The examined dentine surfaces are modified by laser radiation produced by a pulsed Nd:YAG laser that leads to sealing of open dentinal tubules under suitable conditions that are reached after covering dentine surfaces with dye agents. Out of four investigated dye agents erythrosin solution in water has been found the most suitable and the lower and upper limits of pulse energies for sealing of dentinal tubules have been set.  相似文献

It is known that removal of the tooth pulp from mandibular molar teeth in adult rats alters the mechanoreceptive field properties of many low-threshold mechanoreceptive neurons in the trigeminal brainstem nuclear complex. The present study investigates one possible way that such deafferentation-induced receptive field changes could occur: altered central projections of uninjured trigeminal low-threshold mechanoreceptive primary afferent fibers. Intra-axonal injection of horseradish peroxidase (n = 22) or neurobiotin (n = 44) into characterized fibers was performed ipsilateral to, and 10–32 days after, removal of the coronal pulp from the left mandibular molars in adult rats. Collaterals were reconstructed, quantified, and compared by means of multivariate analyses of variance to equivalent fibers stained in normal adult rats.

Stained mechanosensitive fibers from experimental animals were rapidly conducting and responded to light mechanical stimulation of one vibrissa, one tooth, oral mucosa, facial hairy skin, or guard hairs. Their central projections were indistinguishable from those of control axons in all four trigeminal subnuclei. The numbers of collaterals, areas subtended by collateral arbors, numbers of boutons per collateral, and arbor circularity did not differ from those of control afferents. Collateral somatotopy was also unaffected.

These data suggest that following pulpotomy, the central collaterals of uninjured trigeminal afferents display normal morphologies and maintain normal somatotopy. Changes in the morphology of low-threshold primary afferents cannot account for the changes that occur in the receptive field properties of trigeminal brainstem neurons after pulp deafferentation.  相似文献

We have studied the premaxillary teeth in three armoured catfish, Corydoras aeneus, C. arcuatus and Hoplosternum littorale , by means of light and electron microscopy, in order to compare their development, fine structure and mode of attachment with that of odontodes and other teleost teeth. A premaxillary dentition consisting of small (50–100 μm long) slender pointed teeth showing no true replacement is only present in larval and juvenile stages and is subsequently lost, possibly in relation to a change in feeding mode from predatory to bottom feeder. Like odontodes, teeth are composed of dentine surrounding a pulp cavity and are covered by a hypermineralized cap. Particular features, also found in odontodes, are the absence of dentinal tubules and of nerves and capillaries in the pulp cavity, both possibly related to the small size of the teeth. The irregular pattern of implantation and the variability in attachment mode (primary and/or secondary attachment bone, fusion, mere apposition or ligamentous connection) distinguish the teeth from most other teleost teeth and from odontodes and are interpreted as reflecting considerable differences in dynamics of remodeling of the supporting element (premaxillary bone vs scute). This comparison of teeth and odontodes strongly supports current views according to which teeth and odontodes are two very closely related phenotypic expressions of a single, modifiable, morphogenetic system probably rooted in the earliest stages of vertebrate evolution.  相似文献

Affinity-purified antibodies have been used in combination with the peroxidase-antiperoxidase technique to study the distribution of osteonectin and collagen types I and III in porcine dental tissues. Tissue sections (2 mm thick), including unerupted (fetal) or erupted (adult) teeth, were fixed in periodate-lysine-paraformaldehyde, demineralized in 12% w/v ethylenediaminetetraacetic acid, and after embedding, 6 micron sections were prepared for immunolocalization. Strong staining for osteonectin was observed in dentine of unerupted teeth and in the associated alveolar bone. Light to moderate staining was observed in the dental pulp, stratum intermedium, stellate reticulum, and the reticular elements in the endosteal spaces. In erupted teeth, osteonectin staining in dentine was concentrated around dentinal tubules and the associated alveolar bone stained with variable intensity. Cementum was poorly stained. However, the periodontal ligament and reticular material in the endosteal spaces showed moderate to strong staining. Weaker staining was apparent in the pulp and lamina propria of the gingiva. In comparison, type I collagen showed a similar distribution to osteonectin in both fetal and adult tissues, whereas type III collagen was generally restricted to the periodontal ligament, reticular elements of the endosteal spaces, and Sharpey's fibers in bone and cementum. Both odontoblast and ameloblast layers in fetal tissues stained for osteonectin and type III collagen.  相似文献

Thirty-nine patients with trigeminal neuralgia, not controlled by medical treatment, were treated by radio-frequency thermocoagulation of the Gasserian ganglion and its posterior rootlets. Thirty-six received satisfactory pain relief. In 30 patients touch sensation in the treated territory was preserved. The corneal reflex was affected in only six patients, two of whom subsequently developed keratitis. There were no other complications apart from a minor unpleasant sensation in eight patients. By selectively destroying pain fibres this technique offers the scope of preserving touch sensation in the treated area. Moreover, the zone of analgesia can be restricted to the affected region by sensory mapping through electrode stimulation before thermocoagulation. Its simplicity, low morbidity, associated short hospital stay, and the increased ability to preserve touch sensation, especially of the cornea, seem to make it preferable to other forms of surgical management for trigeminal neuralgia.  相似文献

Teeth are commonly used model systems for the study of epithelial-mesenchymal interactions during organogenesis. We describe here the ultrastructural characteristics of developing pharyngeal replacement teeth in juvenile zebrafish, an increasingly important model organism for vertebrate development. Replacement teeth develop in close association with the dental organ of a functional tooth. Morphogenesis is well advanced prior to the start of cytodifferentiation. Fibrillar enameloid matrix is formed first, followed by the deposition of predentine. Initial mineralization of the enameloid proceeds quickly; maturation involves the presence of ruffled-bordered ameloblasts. Dentine mineralization is inotropic and is mediated by matrix vesicles. Woven-fibred attachment bone matrix is deposited before completion of dentine mineralization. Eruption of fully ankylosed teeth is a fast process and may involve degenerative changes in the pharyngeal epithelium. Mononucleated osteoclasts and clastic cells located in the pulp cavity intervene in tooth resorption prior to shedding. Structural differences with larval, first-generation zebrafish teeth include the presence of dentinal tubules and the absence of an electron-dense covering membrane. Part of these differences may relate to size differences of the teeth. Others, like the site of the replacement tooth bud, suggest that initiation may take place in already committed epithelium from the first initiation event in the larval stage.  相似文献

Dentinogenesis imperfecta (DI) is the result of a dominant genetic defect and affects both the deciduous and permanent dentitions. It is characterized by opalescent teeth composed of irregularly formed and undemineralized dentin which obliterates pulp chamber and root canal. DI can appear as a separate disorder or with osteogenesis imperfecta (OI). The teeth with DI show a grayish-blue to brown hue with dislodged enamel, dysplastic dentine with irregular dentinal tubules and interglobular dentine, short roots and pulpal obliteration, which all may lead to rapid and extensive attrition which require adequate crown reconstruction. The aim of this study was to show a reconstruction of frontal teeth in upper jaw with direct composite veneers in young adult patient with DI.  相似文献

The structure of the dentin, consists of the following elements: the odontoblastic processes, dentinal tubules and their periodontoblastic spaces. The odontoblasts are aligned in a single layer in the periphery of the dental pulp and secrete the organic components of dentin. The vitality of dentin is mediated too by the nerve fibres. The ultrastructure of the trigeminal sensory nerves in dentin, especially in relation to odontoblasts remains to be clarified. We studied the third molars and young premolars. The specimens were fixed in glutaraldehyde immediately after extraction. Our investigations give evidence to prove that the distribution of the dentinary tubules is homogeneous, containing a principal odontoblastic prolongation in the regions of the inner dentine, and only in special cases more than one. The area of the dentinary tubules and the odontoblastic prolongations' area were studied. The nervous fibres appeared accompanying 30-70% of the odontoblastic prolongations and their synapsis-like relation with the odontotoblastic processes was demonstrated. The existence of very few periodontoblastic spaces, and intradentinal sensory axons, as well as the intercellular connections will allow us to discover more about the mechanisms of the dentinary permeability, and its significance in maintenance and repair of the human pulpodentinal complex.  相似文献

This paper describes pathological alterations in the permanent canines (tusks) of four male wild boars. The mandibular tusks of all individuals, and also some of the maxillary tusks, exhibited an extended enamel hypoplasia in their apical portion, denoting an impairment of secretory ameloblast function. Moreover, the pulp cavities of the mandibular tusks were exposed through cleft-like openings in the wear (whetting) surfaces of the teeth. Presence of a plug of reparative dentine within the pulp cavity was observed in a split mandibular tusk of one individual. In a second boar, the presence of a plug of reparative dentine within the pulp cavity of the mandibular tusks was indicated radiographically. These findings suggested a reparation process attempting to demarcate a vital, apical pulp portion from a necrotic, incisal portion. The enamel hypoplasias observed in the teeth are regarded to be sequelae of the pulp inflammation caused by bacterial invasion in the mandibular tusks. Most likely, bacterial invasion of the dental pulp occurred through the cleft-like openings in the tusks whetting surfaces, the openings resulting from insufficient formation of secondary dentine. It is, however, also conceivable that pulp inflammation and partial necrosis occurred as a consequence of bacterial invasion of patent dentinal tubules, and that the openings in the whetting surface developed secondarily as a consequence of the pulp changes. One mandibular tusk showed marked signs of resorption apically, suggesting a spread of the inflammation from the pulp into the periodontium.  相似文献

A useful method is described in this paper for studying the mediators released from tooth pulp of the dog during the electrical stimulation of dentine. This method is based upon the perfusion of the pulp and superfusing the return continuously over the isolated, in cascade, cat jejunum and rat stomach fundus strip. The presented evidences in this study indicate the possibility of the release of bradykinin and prostaglandin - like materials from the tooth pulp during the electrical stimulation of dentine. Possible relationship of these mediators and tooth pain due to the electrical stimulation of dentine is discussed.  相似文献

Initial responses of odontoblasts and immunocompetent cells to cavity preparation by laser ablation were investigated in rat molars. In untreated control teeth, intense heat shock protein (Hsp) 25 immunoreactivity was found in the cell bodies of odontoblasts, whereas cells immunopositive for the class II major histocompatibility complex (MHC) antigen were predominantly located beneath the odontoblast layer in the dental pulp. Cavity preparation caused the destruction of the odontoblast layer and the shift of most class-II-MHC-positive cells from the pulp-dentin border toward the pulp core at the affected site. Twelve hours after cavity preparation, numerous class-II-MHC-positive cells appeared along the pulp-dentin border and extended their processes deep into the exposed dentinal tubules, but subsequently disappeared from the pulp-dentin border together with Hsp-25-immunopositive cells by 24 h after the operation. By 3–5 days postoperation, distinct abscess formation consisting of polymorphonuclear leukocytes was found in the dental pulp. The penetration of masses of oral bacteria was recognizable in the dentinal tubules beneath the prepared cavity. 本文是利用扫描电子显微镜对陆栖恐龙tvrannosaurid与海栖渐增mosasaurid”牙齿结构进行的比较解剖学研究。化石采自加拿大RedDeerhiverValley上白里统Horse-shoeCanyon组。通过研究地层中出现的生物化石,特别是动物牙齿的组织结构,可以了解动物为了适应生活环境而发生的进化过程,也可以推测它们的系统发育关系。tyrannosaund与mosas。id都拥有锥状的同形齿,牙齿侧向扁平,且略向后弯曲。研究结果确认了tyrannosaurid的牙齿由于薄层的无柱釉质bPrismaticenamel)向齿质的侵人而造成许多的凹凸构造,此锯齿状构造沿着牙齿的前后缘,由牙齿的顶端分布至基部。因此tyrannosaurid的牙齿呈现着锐利的切缘;在这些凹凸状切缘的沟与小窝的深部可观察到有机物的沉积。但是类似的锯齿状构造只能在齿冠呈钝圆状的mosasaurid牙齿的基部附近观察到。我们以扫描电子显微镜(SEM)检索,确认两爬行类的齿质皆是属于中间型的真性齿质(intermediatetypeorthodentine);所谓orthodentine即是细管齿质(tubulardentine)。tyrannosaurid的真性齿质的齿质小管只在齿质一釉质相接处Uentino—enamaljunction)附近放散出规则性的分歧与末枝。但mosasaurid的真性齿质的齿质J。管,在齿质的中间层与表层中,呈现着由复杂  相似文献

Traditionally, physical anthropologists have focused their dental studies on features of teeth that can be observed with the unaided eye, measured with calipers, or observed with light microscopes. With the advent of the scanning electron microscope, there has been renewed interest in the use of the microstructure of enamel in phylogenetic reconstruction and taxonomic classification. The microstructure of dentine has received far less research attention than has enamel although several investigators have proposed that dentine has taxon specificity. We report the first results of a study in which we investigated the taxon specificity of dentine. In our study, we exposed the dentinal tubules on the mesial root surfaces of 12 Canis, 11 Papio, and 12 Homo mandibular first molars and compared tubule density and pattern among the three taxa. Based on these parameters, 91.43% of the teeth were correctly identified as being dog, baboon, or human, respectively. We conclude that dentine has taxon-specific structural characteristics, which can be used in anthropological investigations. To our knowledge, this is the first comparative study of the micro-structure of teeth utilizing statistical analysis.  相似文献

Dental thermal pain is a significant health problem in daily life and dentistry. There is a long-standing question regarding the phenomenon that cold stimulation evokes sharper and more shooting pain sensations than hot stimulation. This phenomenon, however, outlives the well-known hydrodynamic theory used to explain dental thermal pain mechanism. Here, we present a mathematical model based on the hypothesis that hot or cold stimulation-induced different directions of dentinal fluid flow and the corresponding odontoblast movements in dentinal microtubules contribute to different dental pain responses. We coupled a computational fluid dynamics model, describing the fluid mechanics in dentinal microtubules, with a modified Hodgkin-Huxley model, describing the discharge behavior of intradental neuron. The simulated results agreed well with existing experimental measurements. We thence demonstrated theoretically that intradental mechano-sensitive nociceptors are not "equally sensitive" to inward (into the pulp) and outward (away from the pulp) fluid flows, providing mechanistic insights into the difference between hot and cold dental pain. The model developed here could enable better diagnosis in endodontics which requires an understanding of pulpal histology, neurology and physiology, as well as their dynamic response to the thermal stimulation used in dental practices.  相似文献

Peritubular dentine is a mineralised deposit formed centripetally in the dentine tubules with advancing age, so that the tubular diameter is smaller in teeth from older persons. The aim of the present study was to investigate the relationship between age in humans and the amount of peritubular dentine and the extent of the consequent obliteration of the tubules, and to find out whether this relationship was strong enough to be used as a parameter for age estimation. Fifty mandibular central and lateral human incisors were ground on the lingual aspect of the root and examined in a scanning electron microscope (SEM). The number of open tubules was counted and the diameter of the tubules measured both before and after etching with 35% orthophosphoric acid. The difference in the number of tubules in unetched and etched specimens was taken to be the number of occluded tubules, and the difference in radii before and after etching to be the thickness of peritubular dentine. The results did not demonstrate a significant relationship between age and the reduction in the number of tubules. One explanation might be that a certain age has to be reached before obliterated dentine tubules can be observed. The correlation between age and the thickness of peritubular dentine was not significant in teeth extracted because of periodontal disease, so these teeth were excluded from the regression analysis with age as the dependent variable. Only the thickness of peritubular dentine was included in the regression (r=0.69); this factor was a better indicator of age than the tubular diameter, but not so closely related to age that it can be recommended for general use in forensic and archaeological age estimations.  相似文献

This study was performed on the effects of stimulating the midbrain central gray matter (CGM) on neuronal response in the association medial thalamic nuclei evoked by stimulation of A-alpha and A-delta fibers of the infraorbital nerve and the caudal nucleus of the spinal trigeminal tract (CN STT) and tooth pulp stimulation using cats anesthetized by thiopental-chloralose as experimental animals. Stimulating the CGB with trains of stimuli was found to evoke an excitatory response in a percentage of the neurons tested, in which latency fluctuated between 15 and 40 msec. Applying conditioned stimuli to the CGM caused suppression of response to afferent impulses in neurons belonging to the "convergent" and "low" and "high" threshold groups. Responses induced by stimulating tooth pulp and A-delta fibers showed 100% inhibition as compared with 86% during A-alpha fiber and infraorbital nerve stimulation. The fact that stimulating the CGM produces an inhibitory effect on the response of thalamic neurons evoked by stimulation of both peripheral afferents and the CN STT would indicate that the CGM can exert a direct action on thalamic neuronal activity.A. Our small experiences with electrical stimulation in the VPL and VPM for dysesthetic pain show that it provoked only paresthesia and induced some relief of pain. It does not increase the beta-endorphin level in CSF. To clarify the anatomical substrata in VPL stimulation, neuroanatomical studies were done about the inputs to VPL in man, monkey and cat by the Fink-Heimer method. The spinothalamic tract terminates in VPL in a patchy fashion in the monkey. The corticothalamic fibers from SI and SII cortex project to VPL and VPM in somatotopical organization in the cat. SI and SII cortices have reciprocal connections, in addition to projections to area 5 or SIII cortex. The corticofugal fibers to the magnocellular and gigantocellular tegmental fields are suggested in addition to the dorsal column nuclei, spinal trigeminal nuclei and spinal posterior horn in cat. The medial lemniscus input to VPL and the above neural circuits are thought to be associated with VPL stimulation.  相似文献
