The results of three long-term breeding and genetic experiments with Drosophila melanogaster performed at different times are summarized. Selections for different fitness components led to similar results. The data on the concentration of viability mutations in the inbred strains LA, LA+, and LA- after 400 generations of selection and the strain ULA during its breeding are presented for the first time. The results of studying the genetic heterogeneity and spontaneous mutational process in Drosophila inbred strains comply with the idea of M.E. Lobashev that "a change in the direction of selection or acceleration of its rate is always accompanied by a concurrent increase in mutational variation".  相似文献

The effect of intraplantarly (i.pl.)-injected methionine-enkephalin (ME) on Concanavalin A (Con A)-induced paw edema in Dark Agouti (DA) and Albino Oxford (AO) rats was investigated. ME suppressed edema in DA rats, which was antagonized with naloxone (non-selective opioid receptor antagonist) and naltrindole (delta opioid receptors antagonist). Potentiating effect of ME in AO rats was blocked by naloxone, nor-binaltorphimine (kappa opioid receptors antagonist) and beta-funaltrexamine (mu opioid receptors antagonist). Dexamethasone suppressed edema in both rat strains. These findings suggest that strain-dependent differences in the effects of ME on inflammation in DA and AO rats could be related to diversity in opioid receptors expression in these strains.  相似文献

The airway and systemic arterial smooth muscle responsiveness to cholinergic agents of two strains of rats, Rat Albino (RA) and Brown Norway (BN), was compared in vivo and in vitro. In vivo, we measured the doses of carbachol that induced a 100% increase in lung resistance (PD100 RL), a 50% decrease in dynamic lung compliance (PD50 Cdyn), and the value of systolic blood pressure at the carbachol dose of 10 micrograms (Pa 10 micrograms). In vitro airway smooth muscle and systemic arterial smooth muscle responsiveness was assessed by measuring the maximal response to acetylcholine, the slope of the linear portion of the dose-response curve, and the negative logarithm of the molar concentration of acetylcholine producing 50% of the maximal response (pD2). PD100 and PD50 were about four times greater in BN rats than in RA rats. In contrast, Pa 10 micrograms was 1.5 lower in the BN rats. These differences persisted after bivagotomy. Tracheal pD2 was 25% greater in the RA than in the BN strain. The mean dose-response curve of parenchymal strips of RA rats was situated upward and to the left of the BN curve, but the reverse was observed for aortic smooth muscle dose-response curves. Thus 1) airway smooth muscle responsiveness to cholinergic agents is greater in RA strain than in BN, but the reverse is true for systemic arterial smooth muscle responsiveness; and 2) these differences are not due to factors extrinsic to the smooth muscle, since they occurred in vitro and may depend on different densities of muscarinic receptors.  相似文献

Coitus is a unique physiological event which may result in internal fertilization, whereby active allogeneic spermatozoa are presented to the female host which can invade her tissues. Sterile coitus on the other hand, stimulates the uterine cervix in the rat and induces a physiological state of pseudopregnancy which interrupts the normal pattern of the oestrous cycle and establishes a persistent dioestrous state. This state is hormonally controlled and in its early stages is not dissimilar from that of true pregnancy. Pregnancy hormones are known to influence the immunological status of the female. Thymic observations during early pseudopregnancy showed interstrain differences in weight, cell content and thymocyte proliferation. Pooled data indicated increased cell density on days 3, 4 and 5 of pseudopregnancy associated with a peak of mitotic activity on day 3 in both strains. The timing of this proliferative activity seems to reflect the needs of the immune system in its putative response to mating and implantation.  相似文献

The aim of our current study was to investigate the effect of acute exposure to electric tail shock stress (ES) and to a stress witnessing procedure (SW), as models for physical and psychological stress paradigms, respectively, on phagocytosis and H(2)O(2) production in peritoneal macrophages isolated from Albino Oxford (AO) and Dark Agouti (DA) rats. In addition, we studied the in vitro effects of methionine-enkephalin (ME) on phagocytosis and H(2)O(2) production in peritoneal macrophages isolated from both AO and DA rats that had been exposed to ES and SW procedures. The results showed that peritoneal macrophages isolated from DA rats were less sensitive to the suppressive effects of ES and SW than macrophages isolated from AO rats. In vitro treatment of macrophages isolated from AO rats with ME mimicked to some extent the suppressive effects of ES and SW on phagocytosis and H(2)O(2) production and additionally diminished H(2)O(2) release in macrophages isolated from AO rats previously exposed to ES or SW. ME did not have any effect on phagocytosis in macrophages isolated from DA rats, but changed H(2)O(2) production in a concentration-dependent manner. In macrophages isolated from DA rats previously exposed to stress the effect of ME was dependent on the macrophage function tested and the particular stress paradigm employed. Our results emphasise the fact that both beneficial and detrimental effects of stress on immune system functions could be attributed to the individual variations in the macrophage's response to stress mediators.  相似文献

Genetic differences in the testicular hormonal responsiveness to in vivo administration of chorionic gonadotropin (CG) between adult male mice of eight inbred strains (A/Sn, CBA/Lac, CC57Br, C57Bl/6J, DBA/2J, GR, PT, and YT) were determined. In addition, the genetic variation of the body and testis weights was estimated as related to the responsiveness to stimulation of steroidogenesis with CG. Adult males were subcutaneously injected with 10 IU of CG or physiological saline 120 min before decapitation. It was found that the baseline testosterone level in the blood serum and its content in the testes only slightly varied in males of the strains studied. Administration of CG increased these parameters by a factor of 3–45, depending on the strain. The results of the study indicate genetic differences in the testicular reactivity to CG. In addition, it has been found that the response to administration of CG, as compared to the baseline levels, provides the most reliable information on the genetic characteristics of the hormonal potential of the testes. The given set of inbred mouse strains may be a promising genetic model for studying the physiological and hereditary variations of testicular steroidogenesis.  相似文献

In this study, we present a pharmacoproteomic investigation of response to antidepressants two inbred strains. Our aim was to uncover molecular mechanisms underlying antidepressant action and identify new biomarkers to determine therapeutic response to two antidepressants with proven efficacy in the treatment of depression but divergent mechanisms of action. Mice were treated with the pro-noradrenergic drug nortriptyline, the pro-serotonergic drug escitalopram or saline. Quantitative proteomic analyses were undertaken on hippocampal tissue from a study design that used two inbred mouse strains, two depressogenic protocols and a control condition, (maternal separation, chronic mild stress, control), two antidepressant drugs and two dosing protocols. The proteomic analysis was aimed at the identification of specific drug-response markers. Complementary approaches, 2DE and isobaric tandem mass tagging (TMT), were applied to the selected experimental groups. To investigate the relationship between proteomic profiles, depressogenic protocols and drug response, 2DE and TMT data sets were analysed using multivariate methods. The results highlighted significant strain- and stress-related differences across both 2DE and TMT data sets and identified the three gene products involved in serotonergic (PXBD5, YHWAB, SLC25A4) and one in noradrenergic antidepressant action (PXBD6).  相似文献

Nineteen inbred strains of the laboratory rat (Rattus norvegicus) were investigated for genetic differences by starch and agarose gel electrophoretic techniques. For six out of 21 loci, interstrain differences were observed: Tf, 6PGD, Es-1, Es-4, Es-5, and Svp. The variants are briefly described and the allelic distributions are tabulated. The 6GPD locus was found to be linked neither to the major histocompatibility gene region nor to the coat color genes c, a, h.This investigation was supported by the Deutsche Forschungsgemeinschaft (Be 352/10 and Gu 105).  相似文献

Little is known about interstrain variations in baseline lung functions or smooth muscle contractility in murine lungs. We therefore examined basal lung mechanics and airway, as well as vascular reactivity to methacholine, thromboxane (using U-46619), and endothelin-1 (ET-1), A/J, AKR, BALB/c, C3H/HeN, C57BL/6, and SCID mice. All experiments were performed with isolated perfused mouse lungs. Except AKR mice (which were excluded from further analysis), all other strains showed stable pulmonary compliance, pulmonary resistance, and pulmonary arterial pressure within a control period of 45 min. Among these strains, C3H/HeN mice exhibited higher dynamic pulmonary compliance and lower pulmonary resistance, whereas SCID mice had higher baseline pulmonary resistance than the other strains. Concentration-response experiments with methacholine showed a lower airway reactivity for C57BL/6 mice compared with the other strains. Perfusion with 1 microM U-46619 or 100 nM ET-1 revealed a similar pattern: the agonist-inducible broncho- and vasoconstriction was lower in C57BL/6 mice than in all other strains, whereas it tended to be higher in SCID mice. The present study demonstrates a correlation between airway and vascular responsiveness in all tested strains. SCID mice are hyperreactive, whereas C57BL/6 mice are hyporeactive, to smooth muscle constrictors. Lung mechanics, as well as airway and vascular responsiveness, appear to be genetically controlled.  相似文献

In vitro fertilization with cryopreserved inbred mouse sperm   总被引:8,自引:0,他引:8  
Sperm from C57BL/6J, DBA/2J, BALB/cJ, 129S3/SvImJ, and FVB/NJ inbred mice were cryopreserved in 3% skim milk/18% raffinose cryoprotectant solution. The post-thaw sperm from all strains were evaluated for their viability and fertility by comparing them against B6D2F1 sperm used as a control. The protocol used for freezing mouse sperm was effective in different strains, because the motility was decreased by 50% after cryopreservation similar to other mammalian sperm. However, the progressive motility and the fertility of each inbred strain were affected differently. The C57BL/6J, BALB/cJ, and 129S3/SvImJ strains were the most affected; their fertility (two-cell cleavage) decreased from 70%, 34%, and 84% when using freshly collected sperm to 6%, 12%, and 6% when using frozen/thawed sperm, respectively. Live newborns derived from frozen/thawed sperm were obtained from all strains in the study. These results corroborate the genetic variation among strains with regard to fertility and susceptibility to cryopreservation.  相似文献

In this study, differences between two strains of inbred mice in aspects of neutrophil function, namely Rac1 expression, chemotaxis, nicotinamide adenine dinucleotide phosphate oxidase activity and formation of neutrophil extracellular traps (NETs), were determined. Neutrophils from CBA/CaH mice exhibited weaker Rac1 expression and a slower chemotactic gradient than BALB/c mice. Furthermore, PMA‐ or fMLP‐stimulated neutrophils from CBA/CaH mice generated much less superoxide and NETs than similarly stimulated neutrophils from BALB/c mice. These findings suggest that neutrophils from BALB/c mice are functionally more efficient than those from CBA/CaH mice.  相似文献

Hypoxic ventilatory responses differ between rodent strains, suggesting a genetic contribution to interindividual variability. However, hypoxic ventilatory responses consist of multiple time-dependent mechanisms that can be observed in different respiratory motor outputs. We hypothesized that strain differences would exist in discrete time-dependent mechanisms of the hypoxic response and, furthermore, that there may be differences between hypoglossal and phrenic nerve responses to hypoxia. Hypoglossal and phrenic nerve responses were assessed during and after a 5-min hypoxic episode in anesthetized, vagotomized, and ventilated rats from four inbred strains: Brown Norway (BN), Fischer 344 (FS), Lewis (LW), and Piebald-viral-Glaxo (PVG). During baseline, burst frequency was higher in PVG than LW rats (P < 0.05), phrenic burst amplitude was higher in PVG vs. other strains (P < 0.05), and hypoglossal burst amplitude was higher in PVG and BN vs. FS and LW (P < 0.05). During hypoxia, burst frequency did not change in BN or LW rats, but it increased in PVG and FS rats. The phrenic amplitude response was smallest in PVG vs. other strains (P < 0.05), and the hypoglossal response was similar among strains. Short-term potentiation posthypoxia was slowest in FS and fastest in LW rats (P < 0.05). Posthypoxia frequency decline was absent in PVG, but it was observed in all other strains. Augmented breaths were observed during hypoxia in FS rats only. Thus genetic differences exist in the time domains of the hypoxic response, and these are differentially expressed in hypoglossal and phrenic nerves. Furthermore, genetic diversity observed in hypoxic ventilatory responses in unanesthetized rats may arise from multiple neural mechanisms.  相似文献
