The male (CBA X C57BL) FI mice received 125 mg of hydrocortisone per kg body weight intraperitoneally. The functional activity of neutrophils has been evaluated by means of nitroblue terazolium test (NBT-test) values taken before or after heat-killed S. marcescens cell stimulation in vitro by 2, 12, 24 h 3, 7 or 14 days post hormonal treatment. Throughout the 1st day after hydrocortisone administration the NBT-test values taken prior to as well as post microbial neutrophil stimulation were steadily increased. This effect could be seen as early as 2 h post hormone administration and it was linked with growing leukopenia and total decrease of blood granulocyte content. By the 3rd day the same very values turned up to become normal. The NBT-test values after microbial stimulation of neutrophils were 1.7 or 2.4 lower after hydrocortisone had been added to blood in vitro in a dose 3.5 X 10(-6) M or 7 X 10(-5) M.  相似文献

The consequences of fasting or overfeeding during 2 days on energy expenditure were investigated by continuously monitoring O2 consumption in unrestrained, unanesthetized rats. O2 consumption decreased by 15% on the 1st day of fasting and then by an additional 15% on the 2nd day. On the 3rd day, when rats were fed again, energy intake increased by 30% above control (prefasting) values, whereas energy expenditure rapidly increased but no more than control values. On the other hand, when ad libitum fed animals were offered a sucrose solution (32%) for 2 days, energy intake increased by 30% and energy expenditure by 9-12%. On the 3rd day, when the rats were fed with their normal diet, energy intake significantly decreased under control (preoverfeeding) values during one day, but energy expenditure rapidly returned to normal values. The results show that fasting decreases, whereas hyperphagia increases 24-h energy expenditure during the treatments. When the treatments are terminated, energy expenditure rapidly returns to normal values, but fasting induces a postfasting increase of energy intake (during 2 days), whereas hyperphagia, on the contrary, results in a transient decrease of appetite. This indicates that alterations of food intake induce compensatory changes of energy expenditure during the treatments, but that after the treatments, energy balance is normalized via regulatory adjustments in the ratio of energy expenditure over energy intake.  相似文献

We have previously found two stages of amnesia evoked by disruption of memory reconsolidation with MK-801 (NMDA glutamate receptors antagonists) application in food aversion conditioned snails. Repeated conditioning restored the food aversion at early stage of amnesia development (<10 days), whereas repeated conditioning 10 days after MK-801 application did not restore the food aversion. In present work, amnesia was induced with MK-801/reminding 24 hours after food aversion conditioning, and antiamnestic effects of NMDA receptor glycine site agonist d-cycloserine were studied at early (3rd day) or late (12th day) stages of amnesia development. D-cycloserine injection and reminding restored memory only 3 days after amnesia induction whereas d-cycloserine injection without reminding was ineffective. D-cycloserine injection and reminding as well as repeated learning 12 days after amnesia induction were also ineffective in memory restoration. Thus, for the first time, it is revealed that NMDA receptor agonist d-cycloserine influences the memory restoration processes only at early but not the later stages of amnesia development.  相似文献

Neurokinin A (NKA), which selectively inhibits only cellular dehydration (CD)-induced drinking in adult rats, exerts a more general antidipsogenic effect in pups in which it also inhibits drinking induced by angiotensin II (AII) or suckling deprivation (SD). The inhibition of drinking is precocious (1st-3rd day) and never involves the intake of milk. The inhibition of CD-induced drinking increases with age, while that of AII- or SD-induced drinking progressively decreases and disappears on day 12-15. In the rat, NKA is therefore a precocious and selective inhibitor of drinking behavior and its selectivity is achieved ontogenetically.  相似文献

Rats were raised in litters of 3 or 14 from the 3rd postnatal day. Already, 2 days later on day 5, those raised in groups of three had elevated blood cholesterol and insulin levels and a very much lower hepatic 3-hydroxy-3-methyl-glutaryl-CoA (HMG) CoA) reductase activity, than the other group. When aged 40 days, hepatic HMG CoA reductase activity was higher in the group raised three rats per litter than in the large litter group.  相似文献

The concentration of calcium ions in soleus muscle fibers in Mongolian gerbils and rats has been shown to increase for the first day of gravitational load by 4.5 and 2.8 times, respectively, compared to the control. This increase is preserved for 3 days, while, after 12 days, it decreases, but remains above the control level. The resting calcium level in the control in gerbils is lower than in rats and, as a consequence, its change for the first day of unloading is greater. It is possible that this leads to earlier changes in the muscle fiber isoform composition in gerbils; a statistically significant shift in the ratio of fibers that contain fast and slow calcium pumps towards pumping faster is noted as early as the first day, while that of fibers that express the corresponding isoforms of myosin heavy chains (MHCs) is observed after 3 days of functional unloading. In rats, similar changes are revealed as late as on the 7th day. After 12 days, in both species, a tendency to return to the control level is observed that is more pronounced in gerbil.  相似文献

目的:观察慢性吗啡处理及戒断后大鼠杏仁核中Parvalbumin(PV)的表达变化,为其功能的研究提供形态学依据。方法:将30只健康雄性SD大鼠随机分为吗啡依赖组和生理盐水对照组。吗啡依赖组大鼠腹膜腔注射吗啡,2次/d,起始剂量为5 mg/kg,逐日递增5mg,至第10d为50mg/kg;对照组注射同体积的生理盐水。于末次注射后动物分别存活3h、3 d和14d。用免疫组化方法和相对平均灰度值检测杏仁核内PV的表达。结果:在生理盐水处理组各存活时间点,杏仁核内PV的表达相同。和生理盐水对照组相比,3h时杏仁核内PV的表达明显增加(P<0.05)。第3d时,杏仁核内PV的表达减少,明显低于第3 h组(P<0.05)。至第14d时,PV的表达又开始增加,明显高于第3 d组(P<0.05)。结论:本结果提示慢性吗啡处理及戒断后杏仁核PV的表达具有时相特异性;这种变化在戒断早期可能主要与躯体依赖相关,而戒断晚期主要与精神依赖相关。  相似文献

Several indices of free radical generation were determined in limbic structures after kainate (KA)-induced seizure activity in adult and postnatal day (PND) 12 and 17 rats. Superoxide dismutase, catalase, and glutathione peroxidase activities were measured in piriform cortex and hippocampal subfields at 8, 16, 48 h, and 5 days after KA injection in adults and pups, and also at 3 weeks postinjection in adults. KA-induced seizure activity had no significant effect on enzyme activities in PND 12 and 17 rats. In adults, superoxide dismutase and catalase activities were significantly increased at 5 days after KA administration, and returned to preinjection levels by 3 weeks. Glutathione peroxidase activity was also increased significantly at 5 days postinjection, but remained elevated at 3 weeks. Lipid peroxidation, as indicated by malondialdehyde (MDA) concentration, exhibited an early significant increase at 8 and 16 h, followed at 48 h and 5 days by a significant decrease. At 3 weeks postinjection, MDA levels were still significantly decreased in CA3 and dentate gyrus. KA administration in PND 12 and 17 rats had no significant effect on MDA content. KA-induced seizure activity in adults also resulted in a large and sustained increase in protein oxidation in piriform cortex and hippocampus. The early increase in MDA and protein oxidation in adult rats strongly suggests the involvement of oxygen free radicals in the initial phases of KA-induced pathology, whereas the changes in scavenging enzyme activities and MDA content at 5 days and 3 weeks post KA injection possibly reflect glial proliferation subsequent to neuronal death.  相似文献

Arginine-vasopressin reduced the tonic-clonic seizures' latency as well as the duration of the seizures brain-stem generalisation on the 3rd and 5th postpartum days in rats. The reduced latency was also observed after the PACAP38 low doses administration, whereas higher doses diminished and then enhanced the threshold of generalised hyperthermia-induced seizures on the 3rd and 5th days and the 7th and 9th days, resp. The arginine-vasopressin-treated animals had a dramatically enhanced duration of the tonic-clonic seizures up to the epileptic status on the 9th postpartum day. The findings suggest the PACAP involvement in mechanisms of experimental febrile seizures through its effect upon arginine-vasopressin neurosecretion.  相似文献

In the present study we investigated the effect of chronic hypermethioninemia on rat performance in the Morris water maze task, as well as on acetylcholinesterase (AChE) activity in rat cerebral cortex. For chronic treatment, rats received subcutaneous injections of methionine (1.34–2.68 μmol/g of body weight), twice a day, from the 6th to the 28th day of age; control rats received the same volume of saline solution. Groups of rats were killed 3 h, 12 h or 30 days after the last injection of methionine to AChE assay and another group was left to recover until the 60th day of life to assess the effect of early methionine administration on reference and working spatial memory of rats. AChE activity was also determined after behavioral task. Results showed that chronic treatment with methionine did not alter reference memory when compared to saline-treated animals. In the working memory task, we observed a significant days effect with significant differences between control and methionine-treated animals. Chronic hypermethioninemia significantly increased AChE activity at 3 h, 12 h or 30 days after the last injection of methionine, as well as before or after behavioral test. The effect of acute hypermethioninemia on AChE was also evaluated. For acute treatment, 29-day-old rats received one single injection of methionine (2.68 μmol/g of body weight) or saline and were killed 1, 3 or 12 h later. Results showed that acute administration of methionine did not alter cerebral cortex AChE activity. Our findings suggest that chronic experimental hypermethioninemia caused cognitive dysfunction and an increase of AChE activity that might be related, at least in part, to the neurological problems presented by hypermethioninemic patients.  相似文献

目的:观察caspase-12抑制剂Z-ATAD-FMK对百草枯染毒大鼠肝脏caspase-12蛋白表达和肝细胞凋亡的影响,以及通过血清SOD和MDA的检测,探讨其对百草枯中毒大鼠肝脏保护作用和对机体氧化应激反应的作用。方法:50只健康Wistar大鼠随机分三组:A组为正常对照组;B组为模型组(PQ染毒);C组抑制剂组(PQ/Z-ATAD-FMK),于3d、7d处死B组和C组各10只大鼠。取血检测ALT、TBIL、SOD和MDA;HE染色观察肝组织病理学变化;免疫组化法检测caspase-12蛋白;二苯胺法检测细胞凋亡率。SPSS18.0统计分析。结果:Z-ATAD-FMK抑制剂组ALT、TBIL、MDA值低于模型组(P0.05),SOD高于模型组(P0.05);caspase-12蛋白表达和细胞凋亡率亦低于模型组(P0.05);HE染色显示凋亡和坏死程度也较模型组轻。结论:Caspase-12抑制剂Z-ATAD-FMK能减少百草枯中毒大鼠肝脏caspase-12蛋白表达和肝细胞凋亡率,减轻肝细胞坏死,对肝脏具有保护作用,同时可以抑制百草枯中毒大鼠的氧化应激反应。  相似文献

Progesterone and oestradiol serum level was investigated in WAG/Rij rats with genetically determined absences. Blood samples were drawn before and after the pregnancy following the parturition. The serum concentration of progesterone increased after the 3rd day of pregnancy. There is no increasing of oestradiol during pregnancy as large as this. The progesterone is kept high to the 18th day of pregnancy and drastically decreased before the parturition. Common duration of absences--spontaneous spikewave discharges (SWD), frequency and the duration of every SWD decreased from 3rd to 19th days of pregnancy before the parturition. On the basis of these data and modern investigations, regulation of GABAA receptor expression during pregnancy by progesterone (Brusaartd A. B. et al., 1999) it can be assumed that the changes in the parameters of SWD are possibly correlated with the progesterone changes in serum during pregnancy in WAG/Rij rats.  相似文献

Hypermetabolism and anorexia are significant problems associated with major burn trauma. Recent studies have shown that hypothalamic corticotropin releasing factor (CRF) elevates metabolic rate, while neuropeptide Y (NPY) reduces it. CRF also elicits anorexia, while NPY stimulates feeding. We hypothesized that elevation of CRF and decrease of NPY may be mediators of these negative effects of burn trauma. Therefore, we assessed concentrations of CRF and NPY in hypothalamus of burned rats one, three, and twenty-one days after a 30% body surface area, full-thickness, open flame burn. In addition we determined whether a biochemical lesion of CRF receptors using 3rd ventricle injections of a saporin-CRF conjugated peptide would decrease resting energy expenditure (REE). We found a three-day period of anorexia, with REE significantly increasing three days after the burn trauma. Concentrations of NPY were increased in the PVN-containing dorsomedial region of the hypothalamus 1 and 3 days after burn trauma, but were increased further in the day 1 pair-fed rats suggesting this change was a consequence of the anorexia. Levels of CRF were decreased in the ventromedial region of the hypothalamus in day 1 and day 3 burned and PF rats. Treatment with the saporin-CRF conjugate normalized REE and reduced CRF receptor-2 density in the hypothalamus of burned rats, and blocked CRF-induced hypermetabolism in sham-burned rats. Although these results suggest a role of CRF receptors in mediating burn-induced hypermetabolism, CRF itself may not be the principle ligand, as suggested by the significant elevation of hypothalamic urocortin 15 days after burn injury.  相似文献

The present study investigates the effect of the mammalian tachykinin neurokinin A on salt intake in the rat. Intracerebroventricular injection of neurokinin A inhibited salt intake elicited by sodium depletion, by subchronic deoxycorticosterone treatment and by adrenalectomy. It also inhibited the need-free salt intake of female rats that had been previously depleted of sodium. Since different brain mechanisms elicit salt intake in these experimental models, it is concluded that neurokinin A exerts a general antinatriorexic effect. Apparently, its inhibitory effect on salt intake is not due to malaise or competing behaviors, as shown by the fact that the doses of neurokinin A which suppress salt intake do not suppress milk intake. In comparison to the amphibian tachykinin kassinin, neurokinin A possesses a similar spectrum of antinatriorexic activities, but is markedly less potent and less effective in all the experimental models investigated. These findings suggest that activation of neurokinin A receptors cannot solely account for the potent antinatriorexic effect of kassinin and of other nonmammalian tachykinins.  相似文献

In this study, it was shown that calcium levels in the fibers of m. soleus of Mongolian gerbils after one day of unloading increased four and a half times in comparison with the control. In rats, the situation differed: 2.8 times increase after the 1st day of hypogravity persisted until the 3rd day and then calcium levels slightly decreased to the 12th day. However, the base concentrations of calcium in the control group of gerbils were significantly lower than in control rats, thus there was larger shift of this parameter during the first day. Probably, this was the reason for an earlier start of the protein phenotype shift in gerbils: statistically significant increase in the number of fibers with "fast" SERCA isoforms was detected after one day of unloading and therefore slow-to-fast shift in myosin phenotype was observed on the 3rd day of unloading. The same changes in rat muscle could be detected only after the 7th and the 12th day, respectively. In both species, there was a tendency to restore their initial parameters to the 12th day but in gerbils it was much more intensive.  相似文献

Following the lens removal from the left eye of the newborn rats, animals were obtained with one normal (control) and another microphtalmic eye. The animals were sacrificed on the 2nd, 3rd, 5th, 7th and 9th days of postnatal development after four injections of 3H-thymidine during 19 hrs. The number of labelled nuclei and mono- and binuclear cells in the central zone of the eye fundus was counted on the autographs. After the initial increase of the index of labelled nuclei in the operated eyes (on the 2nd, 3rd and 5th days) it fell below the control level (on the 7th and 9th days). The number of binuclear cells in the operated eyes, as well as in the control, attains on the 5th day 50% of the total number of cells and remains at this level up to the end of the experiment, whereas in the control eyes the number of binuclear cells increases up to 60% on the 7th and 80% on the 9th day. The results obtained have shown that in rats the factors of total eye growth participate in the control of proliferative activity and polyploidization of the pigment epithelium cells in the retina.  相似文献

Changes in the ultrastructural aspect of the glyoxysome fraction obtained from maize scutella by density gradient centrifugation were followed during the first 6 days of germination. During the first 2 days the fraction consists of very electron-dense bodies about 0.3 to 0.5 micron in size while at the 4th day it is formed by larger and less dense membrane-bound particles. Some intermediate form between the two types of organelles can be seen at the 3rd day. Between the 4th and the 6th days of germination the glyoxysomes are destroyed, and their enzymes are released into the cytosol. At the peak of their development (4th day) the glyoxysomes contain 75 to 80% of the total isocitratase and 65% of the total malate synthetase of the scutellum. These values drop to very low levels during the next 2 days. Catalase bound to glyoxysomes amounts to 30 to 35% of the total activity present in the scutellum at the 1st day of germination: this value decreases steadily during the following days.  相似文献

To determine whether pretraining with the water-filled straight channel affects learning acquisition, we studied the water filled multiple T-maze learning ability in 8-weeks-old SPF Wistar-Imamichi rats. The performance time for the straight channel was markedly shortened at the 2nd and 3rd trial compared to the time at the 1st trial on the 1st day. But at subsequent trials on days 2 and 3 it was longer than at the 3rd trial on day 1. At the 1st trial on day 1, the performance time of the group unexperienced in the straight channel was more than three times that of the experienced group. In subsequent trials, however, both groups showed similar performance times. More errors were observed in the unexperienced group than in the experienced group at the 1st trial on day 1. No difference was found between the two groups in subsequent trials. These results indicate that the learning acquisition was largely influenced by pretraining in the straight channel at the 1st trial on day 1.  相似文献

药物成瘾被认为是药物长期作用于脑而产生的一种慢性复吸性脑疾病,长期反复的药物(如吗啡)滥用会导致一系列严重后果,如药物依赖、药物耐受、强迫性药物寻求等。本实验利用条件化位置偏好(conditioned place preference,CPP)模型来检测大鼠对吗啡依赖和心理渴求等过程;采用双声刺激听觉诱发电位来研究大鼠在慢性吗啡给予、戒断以及再给药过程中海马感觉门控(N40)的动态变化。吗啡组大鼠注射吗啡(10mg/kg,i.p.)12d,经历第一次戒断12d,再次注射吗啡(2.5mg/kg,i.P.)1d,之后经历第二次戒断2d;对照组大鼠注射同体积生理盐水,其余实验条件与吗啡组相同。CPP实验表明,这种药物给予方法促使大鼠对吗啡产生药物依赖和心理渴求。双声刺激诱发电位实验表明,吗啡组大鼠在吗啡给予期间海马感觉门控受到损伤;第一次戒断期的第1~2天海马感觉门控能力减弱,第3天增强,第4~12天逐渐恢复到正常水平;再次给予吗啡后海马感觉门控能力与对照组相比显著降低,并且随后2d的戒断期内海马感觉门控能力也一直保持较低水平,表明再次给药使大鼠海马感觉门控对吗啡更加敏感化。结果提示,长期反复的吗啡给予及再给药干扰了海马的感觉门控能力,吗啡成瘾对大脑可能产生长期影响。  相似文献
