Rat hepatocytes were incubated in the presence of a high concentrationof the hepatopathogenic agent D-galactosamine (GalN), and theeffect on the cellular concentrations of pyrimidine nucleotidesand nucleotide sugars was determined. The UTP pool became depleted.The pools of UMP and CMP in RNA decreased to 72%, indicativefor an inhibition of RNA synthesis. UDP-HexNAc (where HexNAcis GlcNAc + GalNAc) and UDP-HexN (where HexN is GlcN + GalN)levels increased, and those of UDP-hexose and UDP-GlcA (whereGlcA is glucuronic acid) decreased. The cellular concentrationof CTP did not change, whereas that of CMP-NeuAc (where NeuAcis N-acetylneuraminic add) showed a 2-fold increase. Labellingwith [14C]orotic acid and [3H]cytidine showed that the metabolicflow via the de novo pathway was not changed. The depletionof the so-called overflow pool of UTP [Pels Rijcken et al, Biochem.J., 293, 207–213, 1993] caused a release of the feedbackinhibition by UTP and thus an increased flow through the salvagepathway. Finally, it appeared that GalN, when added to hepatocytes,gives rise to a pool of UDP-GlcNAc (where GlcNAc is N-acetylglueosamine)that is separate from the pool of UDP-GlcNAc that is derivedfrom GlcN. Using an in situ approach, we have evaluated the phosphorusinputs from zooplankton in a high-mountain oligotrophic lake.Values of the specific gross release rate (SGRR) fluctuatedbetween 0.2 and 2.9 µg P mg–1 dry weight h–1,and were higher when the nauplii of Mixodiaptomus laciniatusdominated the zooplankton community. The rate of P recyclingby the zooplankton was high, reaching 1.6 µg P l–1day–1, and showed a highly consistent seasonal patternfrom one year to the next, with maxima in midsummer. Zooplanktonsize accounted for as much as 85% of the variance obtained inthe measurements of the specific rate of P release, while otherfactors, such as the quality or quantity of food, did not significantlyinfluence the SGRR changes. Among the models tested, only theone proposed by Peters (Int, Ver. Theor. Angew. Limnol. Verh.,19, 273–279, 1975) was useful for predictions in thissystem. The stoichiometric model of Hessen and Andersen (Arch.Hydrobiol., 35, 111–120, 1992), applied in this oligotrophicsystem, adequately predicted the phyto- and zooplankton dynamics,whereas the values of P release estimated using this model werefar higher than the excretion rates obtained experimentally.These differences were related to the type of egestion (formationof faecal pellets) of metazooplankton and to the relative importanceof the food resistance to digestion. We believe that in communitieswhere copepods constitute a substantial percentage of the zooplankton,an evaluation of the P release which is readily available [solublereactive phosphate (SRP), total dissolved phosphate (TDP)] toalgae and bacteria would not fit the predictions of generalmodels of mass balance; under these circumstances, assimilationefficiency proves to be the key parameter for predicting thereadily available P (excreted P).  相似文献

In vitrorates of gross and net oxygen production were measuredas a function of light intensity in some plankton communitiescollected from Bedford Basin, Nova Scotia, and in a monoclonalculture of Synechococcus. The rate of gross oxygen productionwas measured by a technique in which the stable oxygen isotope,18O, serves as a photosynthetic tracer Net oxygen productionwas measured by automated Winkler technique. The rate of communityrespiration in the light was then determined by the differencebetween gross and net rates of oxygen production. In the naturalpopulations examined, neither gross nor net oxygen productionrates were significantly inhibited at the highest light intensitymeasured (500–800 µE m–2 s–1) In a samplein which the dark respiration rate was small relative to themaximal rate of production [Pmax;sensu Platt et al (1980) JMar. Res., 38, 687–701] the rates of ‘light’respiration were 3 times greater. In two other communities,with high rates of dark respiration relative to Pmaxthe ratesof ‘light’ respiration were closer to rates of darkrespiration. In the Synechococcus clone, both gross and netoxygen production rates were inhibited at high light intensities.Rates of ‘light’ respiration were found to varyas a function of light intensity. The greatest rates of respirationwere measured in samples incubated at light intensities thatwere just saturating (100 µE m–2 s–1). Recent evidence from algal physiology and molecular biologyconfirms that photoinhibition is directly related to D1 proteindamage and recovery, and D1 protein damage leads to a decreasein electron transfer or an increase in turnover time of theelectron transfer chain. In this study, the turnover time ofthe electron transfer chain is defined as a function of therelative concentration of D1 protein in reaction centre II andthe photoinhibition processes due to D1 protein degradationare incorporated into a model of photosynthesis, initiated byDubinsky et al. (Plant Cell Physiol., 27, 1335–1349, 1986)and developed by Sakshaug et al. (Limnol. Oceanogr., 34, 198–205,1989). Skin fibroblasts treated with brefeldin A produce a recyclingvariant of glypican (a glycosyiphosphatidylinositol-anchoredheparan-sulfate proteoglycan) that is resistant to inositol-specificphospholipase C and incorporates sulfate and glucosamine intoheparan sulfate chains (Fransson, L.-Å. et al., Glycobiology,5, 407–415, 1995). We have now investigated structuralmodifications of recycling glypican, such as fatty acylationfrom [3H]palmitate, and degradation and assembly of heparansulfate side chains. Most of the 3H-radioactivity was recoveredas lipid-like material after de-esterification. To distinguishbetween formation of heparan sulfate at vacant sites, elongationof existing chains or degradation followed by re-elongationof chain remnants, cells were pulse-labeled with [3H]glucosamineand then chase-labeled with [14C]glucosamine. Material isolatedfrom the cells during the chase consisted of proteoglycan andmostly [3H]-labeled heparan-sulfate degradation products (molecularmass, 20–80 kDa) showing that the side chains were degradedduring recycling. The degradation products were initially glucuronate-rich,but became more iduronate-rich with time. The glypican proteoglycanformed during the chase was degraded either with alkali to releaseintact side chains or with heparinase to generate distally locatedchain fragments that were separated from the core protein, containingthe proximally located, covalently attached chain remnants.All of the [14C]-radioactivity incorporated during the pulsewas found in peripheral chain fragments, and the chains formedwere not significantly longer than the original ones. We thereforeconclude that newly made heparan-sulfate chains were neithermade on vacant sites, nor by extension of existing chains butrather by re-elongation of degraded chain remnants. Protoplasts were successfully isolated from internodal callustissues of both Oxalis glaucifolia and O. rhombeo-ovata whenthey were digested in a solution containing 0.1% (w/v) MacerozymeR-10, 0.5% (w/v) cellulase Onozuka R-10 and 0.3 mmol m–3sucrose. Protoplasts proliferated to give cell colonies on Gamborget al.'s B5 medium supplemented with 0.3 mmol m–3 mannitol,0.5 mg dm–32, 4-D, and 2.0 mg dm–3 kinetin. Calluswas produced upon transfer of cell colonies to Murashige andSkoog medium containing 2.0 mg dm–3 l-naphthaleneaceticacid (NAA) and 0.1 mg dm–3 kinetin for O. glaucifolia,or with 5.0 mg dm–3 NAA and 0.5 mg dm–3 6-benzylaminopurine,for O. rhombeo-ovata. Plants were regenerated from O. glaucifoliaprotoplasts on a medium containing 0.1 mg dm–3 NAA, 1.0mg dm–3 kinetin and 1.0 mg dm–3 gibberellic acid,but only vascular nodules were differentiated by O. rhombeo-ovataprotoplast-derived calli. Feeding and metabolism of the siphonophore Sphaeronectes gracilis   总被引:1,自引:0,他引:1  
The in situ predation rate of the siphonophore Sphaeronectesgracilis was estimated from gut content analysis of hand-collectedsiphonophores and from laboratory data on digestion rates ofprey organisms. At daytime prey densities of 0.25 copepods 1–1,S. gracilis was estimated to consume 8.1 – 15.4 prey day–1siphonophore–1. From data on abundances of siphonophoresand copepods, S. gracilis was estimated to consume 2–4%of the copepods daily. In laboratory experiments, ingestionrates averaged 13.8 prey day–1 siphonophore–1 atprey densities of 5 copepods 1–1 and 36.9 at 20 copeods1–1. This was equivalent to a specific ingestion rate(for both carbon and nitrogen) of –17% day–1 and45% day–1, respectively, while specific ingestion in situwas only 2% day–1. Ammonium excretion averaged 0.095 µg-atsiphonophore–1 day–1 at 5 prey 1–1, and 0.162at 20 prey 1–1. The seasonal variability of phytoplankton in the EquatorialAtlantic was analysed using Sea-viewing Wide Field-of-view Sensor(SeaWiFS)-derived chlorophyll a (Chl a) concentration data from1998 to 2001, together with in situ Chl a and primary productiondata obtained during seven cruises carried out between 1995and 2000. Monthly averaged SeaWiFS Chl a distributions werein agreement with previous observations in the Equatorial Atlantic,showing marked differences between 10° W in the EasternTropical Atlantic (ETRA) and 25° W in the Western TropicalAtlantic (WTRA) provinces (Longhurst et al. 1995. J. PlanktonRes., 17, 1245–1271). The seasonal cycle of SeaWiFS-derivedChl a concentration calculated for 0–10° S, 0–20°W (ETRA) is consistent with in situ Chl a measurements, withvalues ranging from 0.16 mg m–3, from February to April,to 0.52 mg m–3 in August. Lower variability was observedin 10° N–10° S, 20–30° W (WTRA) whereminimum and maximum concentrations occurred in April (0.15 mgm–3) and in August (0.24 mg m–3), respectively.A significant empirical relationship between depth-integratedprimary production and in situ measured sea surface Chl a wasfound for ETRA, allowing us to estimate the seasonal cycle ofdepth-integrated primary production from SeaWiFS-derived Chla. As for Chl a, this model was verified in a small area ofthe Eastern Equatorial Atlantic (0–10° S, 0–20°W), although in this instance it was not completely able todescribe the magnitude and temporal variability of in situ primaryproduction measurements. The annual euphotic depth-integratedprimary production rate estimated for ETRA by our empiricalmodel was 1.4 Gt C year–1, which represents 16% of theopen ocean primary production estimated for the whole AtlanticOcean.  相似文献

Feeding on natural plankton populations and respiration of thesmall cyclopoid copepod Oithona similis were measured duringthe warm season in Buzzards Bay, Massachusetts, USA. AlthoughO.similis did not significantly ingest small autotrophic andheterotrophic flagellates (2–8 µn), this copepodactively fed on >10 µm particles, including autotrophic/heterotrophic(dino)flagel-lates and ciliates, with clearance rates of 0.03–0.38ml animal–1 h–1. The clearance rates increased withthe prey size. O.similis also fed on copepod nauplii (mainlycomposed of the N1 stage of Acartia tonsa with a clearance rateof 0.16 ml animal–1 h–1. Daily carbon ration fromthe combination of these food items averaged 148 ng C animal–1day–1 (41% of body C day–1), with ciliates and heterotrophicdino-flagellates being the main food source ({small tilde}69%of total carbon ration). Respiration rates were 020–0.23µl O2 animal–1 day–1. Assuming a respiratoryquotient of 0.8 and digestion efficiency of 0.7, the carbonrequirement for respiration was calculated to be 125–143ng C animal–1 day–1, close to the daily carbon rationestimated above. We conclude that predation on ciliates andheterotrophic dinoflagellates was important for O.similis tosustain its population in our study area during the warm season.  相似文献

We previously showed that plasma membrane Ca2+-ATPase (PMCA) activity accounted for 25–30% of relaxation in bladder smooth muscle (8). Among the four PMCA isoforms only PMCA1 and PMCA4 are expressed in smooth muscle. To address the role of these isoforms, we measured cytosolic Ca2+ ([Ca2+]i) using fura-PE3 and simultaneously measured contractility in bladder smooth muscle from wild-type (WT), Pmca1+/–, Pmca4+/–, Pmca4–/–, and Pmca1+/–Pmca4–/– mice. There were no differences in basal [Ca2+]i values between bladder preparations. KCl (80 mM) elicited both larger forces (150–190%) and increases in [Ca2+]i (130–180%) in smooth muscle from Pmca1+/– and Pmca1+/–Pmca4–/– bladders than those in WT or Pmca4–/–. The responses to carbachol (CCh: 10 µM) were also greater in Pmca1+/– (120–150%) than in WT bladders. In contrast, the responses in Pmca4–/– and Pmca1+/–Pmca4–/– bladders to CCh were significantly smaller (40–50%) than WT. The rise in half-times of force and [Ca2+]i increases in response to KCl and CCh, and the concomitant half-times of their decrease upon washout of agonist were prolonged in Pmca4–/– (130–190%) and Pmca1+/–Pmca4–/– (120–250%) bladders, but not in Pmca1+/– bladders with respect to WT. Our evidence indicates distinct isoform functions with the PMCA1 isoform involved in overall Ca2+ clearance, while PMCA4 is essential for the [Ca2+]i increase and contractile response to the CCh receptor-mediated signal transduction pathway. Loliun perenne L. (cv.S. 23) was grown on vermiculite in winterin a heated greenhouse for 8 weeks under factorial combinationsof two potassium regimes (nominally 6 parts/106 and 156 parts/106in Hewitt's solution) and three densities of artificially supplementedvisible radiation flux (36.1, 7.3, and 2.2 W m–2). Growthand potassium uptake were studied through the calculation ofvarious growth functions from fitted curves. There was little effect of potassium treatment but the experimentalmaterial responded markedly to light. Leaf-area ratio in thethree treatments showed extreme plasticity in increasing from2–3 x 10–2 through 6 x 10–2 to 8–9 x10–2 m2 g–1 as light intensity decreased. Correspondingdecreases in unit leaf rate, however, caused over-all reductionsin relative growth rate. Specific absorption rates for potassium (AK, dry-weight basis)were strongly reduced at the lower light intensities but alsodisplayed complex ontogenetic drifts. Values of the allometricconstant, k (the ratio of root and shoot relative growth rates),decreased from c. 0.7 at 36.1 W m–2 through c. 0.3 at7.3 W m–2 to a value not significantly different fromzero (P < 0.05) at 2.2 W m–2. In material grown under the two higher light intensities a constantinverse relationship was found between the mass ratio of rootand shoot and the corresponding activity ratio. The resultsconform to this model: Mass ratio = –0.001+45.0 (1/activityratio) where activity ratio is expressed as specific absorptionrate for potassium (in µg g root–1 h–1)/unitshoot rate (rate of increase of whole-plant dry weight per unitshoot dry weight, in mg g shoot–1 h–1). The relationships between photosynthesis and photosyntheticphoton flux densities (PPFD, P-l) were studied during a red-tideof Dinophysis norvegica (July-August 1990) in Bedford Basin.Dinophysis norvegica, together with other dinoflagellates suchas Gonyaulax digitate, Ceratium tripos, contributed {small tilde}50%of the phytoplankton biomass that attained a maximum of 16.7µg Chla 1 and 11.93 106 total cells I–1.The atomic ratios of carbon to nitrogen for D.norvegica rangedfrom 8.7 to 10.0. The photosynthetic characteristics of fractionatedphytoplankton (>30 µm) dominated by D.norvegica weresimilar to natural bloom assemblages: o (the initial slope ofthe P-l curves) ranged between 0.013 and 0.047 µg C [µgChla]–1 h–1 [µmol m s–1]–1the maximum photosynthetic rate, pBm, between 0.66 and 1.85µg C [µghla]–1 h–1; lk (the photoadaptationindex) from 14 to 69 µ,mol m–2 s–1. Carbonuptake rates of the isolated cells of D.norvegica (at 780 µmolm–2 s–1) ranged from 16 to 25 pg C cell–1h and were lower than those for C.tripos, G.digitaleand some other dinoflagellates. The variation in carbon uptakerates of isolated cells of D.norvegica corresponded with PBmof the red-tide phytoplankton assemblages in the P-l experiments.Our study showed that D.norvegica, a toxigenic dinoflagellate,was the main contributor to the primary production in the bloom.  相似文献

Species-specific differences in the assimilation of atmosphericCO2 depends upon differences in the capacities for the biochemicalreactions that regulate the gas-exchange process. Quantifyingthese differences for more than a few species, however, hasproven difficult. Therefore, to understand better how speciesdiffer in their capacity for CO2 assimilation, a widely usedmodel, capable of partitioning limitations to the activity ofribulose-1,5-bisphosphate carboxylase-oxygenase, to the rateof ribulose 1,5-bisphosphate regeneration via electron transport,and to the rate of triose phosphate utilization was used toanalyse 164 previously published A/Ci, curves for 109 C3 plantspecies. Based on this analysis, the maximum rate of carboxylation,Vcmax, ranged from 6µmol m–2 s–1 for the coniferousspecies Picea abies to 194µmol m–2 s–1 forthe agricultural species Beta vulgaris, and averaged 64µmolm–2 s–1 across all species. The maximum rate ofelectron transport, Jmax, ranged from 17µmol m–2s–1 again for Picea abies to 372µmol m–2 s–1for the desert annual Malvastrum rotundifolium, and averaged134µmol m–2 s–1 across all species. A strongpositive correlation between Vcmax and Jmax indicated that theassimilation of CO2 was regulated in a co-ordinated manner bythese two component processes. Of the A/Ci curves analysed,23 showed either an insensitivity or reversed-sensitivity toincreasing CO2 concentration, indicating that CO2 assimilationwas limited by the utilization of triose phosphates. The rateof triose phosphate utilization ranged from 4·9 µmolm–2 s–1 for the tropical perennial Tabebuia roseato 20·1 µmol m–2 s–1 for the weedyannual Xanthium strumarium, and averaged 10·1 µmolm–2 s–1 across all species. Despite what at first glance would appear to be a wide rangeof estimates for the biochemical capacities that regulate CO2assimilation, separating these species-specific results intothose of broad plant categories revealed that Vcmax and Jmaxwere in general higher for herbaceous annuals than they werefor woody perennials. For annuals, Vcmax and Jmax averaged 75and 154 µmol m–2 s–1, while for perennialsthese same two parameters averaged only 44 and 97 µmolm2 s–1, respectively. Although these differencesbetween groups may be coincidental, such an observation pointsto differences between annuals and perennials in either theavailability or allocation of resources to the gas-exchangeprocess. The relationship between chlorophyll a (Chl a) and primary productivity(PP) in the uppermost water layer and the water column-based(0–15 m) integral values of those variables were examinedusing measurements taken in Lake Kinneret (Israel) from 1990to 2003. In 81% of all Chl a profiles examined, the distributionwas fairly uniform within the entire 0–15 m water column,and 12.3% of instances showed a prominent subsurface maximum,when the lake phytoplankton was dominated by the dinoflagellatePeridinium gatunense. Chl a can be reliably estimated by remotesensing techniques in the productive and turbid water of LakeKinneret, since Chl a concentration at surface layers can beextrapolated to the entire water column. Light vertical attenuationcoefficient average for wavelengths from 400 to 700 nm, Kd,ranged from 0.203 to 1.954 m–1 and showed high degreeof temporal variation. The maximal rate of photosynthetic efficiency,PBopt [average 3.16 (±1.50)], ranged from 0.25 to 8.85mg C m–3 h–1 mg Chl a–1. Using measured dataof Chl a, PBopt, and light as an input, a simple depth-integratedPP model allowed plausible simulation of PP. However, a lackof correlation between photosynthetic activity and temperature(or other variable with remotely sensed potential) renders theuse of models that require input of photosynthetic efficiencyto calculate integrated PP of little value in the case of productiveand turbid Lake Kinneret.  相似文献

We have examined the long-term effects of NO3 concentrationson NO3 (15NO3) fluxes and cellular pool sizesin roots of intact 30-d-old wheat (Triticum aestivum cv. Courtot)grown hydroponically. Compartmental analysis was performed understeady-state conditions at five different levels of NO3concentration (from 0.1 up to 5 mol m–3 taking into accountmetabolism and secretion into the xylem (Devienne et al., 1994).Nitrate and reduced nitrogen levels in the tissues were largelyindependent of external NO3 concentration although below1.5 mol m–3 NO3; concentration limited plant growth.In the chamber, marked diurnal variations in net uptake occurredand, in the light, higher NO3 concentrations yieldedhigher NO3 uptake rates. After transfer of the plantsto the laboratory, the increase in net uptake linked to elevationof NO3; concentrations was even larger (from 0.1 to 8.8µmolh–1 g–1 FW) as a result of a marked increase (x10–11) in the unidirectional influx at the plasmalemmawhile NO3 efflux was less enhanced (x 4–5). Underthese conditions, influx into the vacuole was also higher (x2–4) while efflux from the vacuole was little affected(x 1–3). NO3 concentrations within the cell compartmentswere estimated under the clas sical assumptions. The vacuolarconcentration was a little modified by NO3 availabilitywhereas that in the cytosol increased from about 10 mol m–3to about 20 mol m–3 indicating that (1) the absolute valuefor the cytosol was high and (2) it displayed only a small increasedespite very large changes in NO3 fluxes. Three marine phytoplankton species (Skeletonema costatum, Olisthodiscusluteus andGonyaulax tamarensis) were grown in batch culturesat 15°C and a 14:10 L:D cycle at irradiance levels rangingfrom 5 to 450 µEinst m–2 s–1. At each irradiance,during exponential growth, concurrent measurements were madeof cell division, carbon-specific growth rate, photosyntheticperformance (both O2 and POC production), dark respiration,and cellular composition in terms of C, N and chlorophyll a.The results indicate that the three species were similar withrespect to chemical composition, C:N (atomic) = 6.9 ±0.4, photo-synthetic quotient, 1.43 ± 0.09, and photosyntheticefficiency, 2.3 ±0.1 x 10–3 µmol O2 (µgChl a)–1 h–1 (µEinst m–2 s–1)–1.Differences in maximum growth rate varied as the –0.24power of cell carbon. Differences in growth efficiency, werebest explained by a power function of Chl a:C at µ = 0.Compensation intensities, ranged from 1.1 µEinst m–2s–1 for S. costatum to 35 forG. tamarensis and were foundto be a linear function of the maintenance respiration rate.The results indicate that interspecific differences in the µ–Irelationship can be adequately explained in terms of just threeparameters: cell carbon at maximum growth rate, the C:Chl aratio (at the limit as growth approaches zero) and the respirationrate at zero growth rate. Acclimation of single plants of Lolium temulentum to changing[CO2] was studied on plants grown in controlled environmentsat 20°C with an 8 h photoperiod. In the first experimentplants were grown at 135 µ;mol m–2 s–1 photosyntheticphoton flux density (PPFD) at 415µl l–1 or 550µll–1 [CO2] with some plants transferred from the lowerto the higher [CO2] at emergence of leaf 4. In the second experimentplants were grown at 135 and 500 µmol m–2 s–1PPFD at 345 and 575 µl l–1 [CO2]. High [CO2] during growth had little effect on stomatal density,total soluble proteins, chlorophyll a content, amount of Rubiscoor cytochrome f. However, increasing [CO2] during measurementincreased photosynthetic rates, particularly in high light.Plants grown in the higher [CO2] had greater leaf extension,leaf and plant growth rates in low but not in high light. Theresults are discussed in relation to the limitation of growthby sink capacity and the modifications in the plant which allowthe storage of extra assimilates at high [CO2]. On sheltered rocky beaches in Hong Kong, Linatella caudata preysupon the lower-littoral and sub-littoral fringe fauna. Ascidiantests are cut with the radula, while the proboscis is insertedbetween the parted shell valves of bivalves. The salivary glandssecrete sulphuric acid that is not used in prey penetration,as in the Cassidae, but is more likely used in digestion ordefence. In preference experiments, prey were chosen in the sequence:Barbatia virescens (Bivalvia: Arcidae) > Styela plicata (Ascidiacea)/Saccostreacucullata (Bivalvia: Ostreidae) > Lunella coronata (Gastropoda:Turbinidae) > Balanus amphitrite (Crustacea: Cirripedia).52% of all attacks were upon Barbatia Lunella and Balanus wereattacked rarely. Prey preferences by individual gastropods werealso demonstrated. Calibration plots of total weight against wet and dry tissueweight of prey (Barbatia virescens) and, finally, the predatorwere obtained and used in estimations of consumption. On average,an adult Linatella caudata (10-15g) consumed three B. virescens.week–1 The mean weight of B. virescens flesh consumed.week–1 ranged between 0. 208–0.412g dry weight prey.gdry weight predator–1, i.e., a mean of 28.2% of the predator'body weight.week–1 or 4%.day–1.
