用PCR方法从4种山茶属(Camellia)(山茶科)(Theaceae)植物的总DNA中分别扩增到CHS基因外显子2的部分序列,经克隆、测序得到16个该基因的序列,这些序列与来自GenBank的该属另一种植物的3个序列及作为外类群的大豆(Glycine max (L.) Merr.)的2个序列一起进行分析.研究表明,山茶属CHS基因家族在进化过程中已分化为A、B、C三个亚家族,包括A1、A2、A3、B1、B2、C 等6类不同的基因成员;其中只有A2类成员为全部被研究的5种植物所共有,而其他5类成员只在部分被研究的植物中发现;所有这些CHS成员具有很高的同源性:在核苷酸水平上同一亚家族内基本上高于90%,不同亚家族间也在78%以上.从推测的氨基酸组成看,山茶属内CHS基因的功能已发生了分化,各类成员的碱基替代率有较大差异; 从分子系统发育树和可能的氨基酸组成分析,山茶属具有新功能的基因成员是在经过基因重复后,或是由少数几个位点的突变而成,或是由逐渐积累的突变而形成的.进一步分析认为,该属CHS基因的分化直到近期还在活跃地进行,并且不同种的进化式样有一定的差别,这种不同的进化式样可能是物种形成后受不同环境因素影响而形成的.  相似文献

In the present study, the nucleotide sequences of the CHS1 gene from dermatophytes and related fungi in the genera Chrysosporium, Epidermophyton, Microsporum and Trichophyton were investigated using molecular methods. About 440-bp genomic DNA fragments of the CHS1 gene from 21 species were amplified by polymerase chain reaction (PCR) and sequenced. The CHS1 nucleotide sequences of these fungi showed more than 83% similarity. The molecular taxonomy of the CHS1 gene sequences revealed that Microsporum was genetically distinct from Chrysosporium and Trichophyton, as classified by morphological characteristics. The sequences of the large subunit of mitochondrial ribosomal RNA (LsmtrRNA) gene of Malassezia species were analysed. The sequences of the seven species of Malassezia are well separated in each species. Therefore the LsmtrRNA gene is thought to be one of the gene targets for species identification in the genus Malassezia. The dendrogram obtained from this gene supports the previous study of Malassezia species based upon the chromosomal genes. This is the first report of taxonomic analysis of Malassezia species based upon the mitochondrial gene.  相似文献

Nucleotide sequences of chitin synthase 1 (CHS1) gene of dermatophytes, Arthroderma benhamiae, A. simii, A. vanbreuseghemii, Trichophyton mentagrophytes var. interdigitale (T. interdigitale), and T. rubrum were analyzed for their phylogenetic relationship. About 620-bp genomic DNA fragments of the CHS1 gene were amplified from these dermatophytes by polymerase chain reaction (PCR) and sequenced. The CHS1 nucleotide sequences of these five dermatophytes showed more than 90% similarity between the species. The phylogenetic analysis of their sequences revealed that A. benhamiae, A. simii, A. vanbreuseghemii, and T. rubrum were genetically distinct from one another, but T. interdigitale was genetically very close to A. vanbreuseghemii. On the other hand, a specific restriction endonuclease site of HinfI was present in the CHS1 gene fragment of T. rubrum but not in those of A. benhamiae, A. simii, A. vanbreuseghemii and T. interdigitale. The molecular analysis of CHS1 genes will provide useful information for the identification of these Trichophyton species and the understanding of their evolution. 利用PCR与TAlL-PCR方法,从半月苔(Lunularia cructata(L.)Dum.ex Lindb)中获得了一段长约l 000 bp的基因片段,它与已知的CHS基因在核苷酸水平上的相似性大于56%,在氨基酸水平上的相似性大于60%,所推断的氨基酸序列中酶反应的4个催化位点与已知晶体结构的紫花苜蓿MCHS2A上的催化位点相同,首次证明了苔类植物中可能存在类CHS基因,将CHS基因的起源时间推到苔藓类植物出现之前.以该序列和两种蕨类植物(Psilotumnudum(L.)Griseb.和Equisetum arvense L.)的CHS序列作为外类群,应用邻接法、最大简约法和最大似然法分别构建了被子植物的CHS的分子系统树.结果表明,大部分科中的CHS分布在不同的分支上,而十字花科、可科和禾本科各自聚成一个单系类群.以邻接树为依据,对茄科、旋花科和菊科的CHS基因进行了相对碱基替换速率的检测,发现这三个科内或科间序列的替换速率不一致.被子植物的CHS基因在基因拷贝数目、碱基替换速率以及重复/丢失事件的发生上都存在较大的差异,这种差异可能与被子植物的生活史、生活环境、花的特性以及对外界的防御系统等的多样性相关.  相似文献

We compared the DNA sequences of the genus Metagonimus: M. yokogawai, M. takahashii, and M. miyatai. We obtained 28S D1 ribosomal DNA (rDNA) and mitochondrial cytochrome c oxidase subunit I (mtCOI) fragments from the adult worms by PCR, that were cloned and sequenced. Phylogenetic relationships inferred from the nucleotide sequences of the 28S D1 rDNA and mtCOI gene. M. takahashii and M. yokogawai are placed in the same clade supported by DNA sequence and phylogenic tree analysis in 28S D1 rDNA and mtCOI gene region. The above findings tell us that M. takahashii is closer to M. yokogawai than to M. miyatai genetically. This phylogenetic data also support the nomination of M. miyatai as a separate species.  相似文献

During a study on the occurrence of lipid-dependent Malassezia spp. in domestic animals, some atypical strains, phylogenetically related to Malassezia sympodialis Simmons et Guého, were shown to represent novel species. In this study, we describe two new taxa, Malassezia caprae sp. nov. (type strain MA383=CBS 10434), isolated mainly from goats, and Malassezia equina sp. nov. (type strain MA146=CBS 9969), isolated mainly from horses, including their morphological and physiological characteristics. The validation of these new taxa is further supported by analysis of the D1/D2 regions of the 26S rRNA gene, the ITS1-5.8S-ITS2 rRNA, the RNA polymerase subunit 1 and chitin synthase nucleotide sequences, and the amplified fragment length polymorphism patterns, which were all consistent in separating these new species from the other species of the genus, and those of the M. sympodialis species cluster, specifically.  相似文献

Nucleotide sequence fragments of the mitochondrial DNA gene encoding cytochrome b were examined in 26 individuals belonging to the seven species of Barbus endemic to the Iberian Peninsula: Barbus haasi, B. bocagei, B. graellsii, B. sclateri, B. comiza, B. guiraonis, and B. microcephalus. Six of the seven currently recognized species can be distinguished on the basis of their cytochrome b nucleotide sequences. Sequence divergence estimates for Spanish species of Barbus (0-6.5%) are, in general, low in comparison to those reported for other fish species, and hybrid individuals were found. All of these observations suggest a recent radiation. The inferred phylogenetic tree has two main clades, one including B. graellsii, B. guiraonis, and B. microcephalus, and the other the remaining species groups.  相似文献

用PCR方法从石笔木(Tutcheria spectabilis Dunn)的总DNA中扩增CHS基因外显子2的部分序列以代表该基因进行研究,经克隆后测序,得到长约740-780bp的序列共12个。以EMBL数据库中得到的紫花苜蓿和欧洲赤松各一个序列作为参照,进行排序和系统树的构建分析。结果显示,所有被测定的序列同源性均高于70%,为CHS基因家庭的成员,且这些序列由3大类共5种不同的基因拷贝组成:第一类家族成员因碱基的插入和缺失改变了读码框,推测已失去了CHS基因的功能,成为假基因,其中个别拷贝存在较大缺失,这在以前的研究中未见报道;第二类家族成员因活性位点的氨基酸发生突变,可能具有新的基因功能;第三类家族成员则具有原CHS基因的功能。综上结果,可预测,山茶科CHS基因家族较大,且有着复杂的进化式样。  相似文献

An inheritable bleeding disorder with light coat color caused by an autosomal recessive gene has been reported in a population of Japanese black cattle. The disease has been diagnosed as Chediak-Higashi Syndrome (CHS) of cattle which correspond to a human inheritable disorder caused by mutation in LYST gene. To characterize the molecular lesion causing CHS in cattle, cDNAs encoding bovine LYST were isolated from a bovine brain cDNA library. The nucleotide and deduced amino acid sequences of bovine LYST had 89.6 and 90.2% identity with those of the human LYST gene, respectively. In order to identify the mutation within the LYST gene causing CHS in cattle, cDNA fragments of the LYST gene were amplified from an affected animal by RT-PCR and their nucleotide sequences were completely determined. Notably, a nucleotide substitution of A to G transition, resulting in an amino acid substitution of histidine to arginine (H2015R) was identified in the affected animal. The presence of the substitution was completely corresponding with the occurrence of the CHS phenotype among 105 members of pedigrees of the Japanese black cattle and no cattle of other populations had this substitution. These findings strongly suggested that H2015R is the causative mutation in CHS of Japanese black cattle. Received: 25 May 1999 / Accepted: 26 July 1999  相似文献

In order to characterize the phylogenetic relationship and deep-sea adaptation process of the deep-sea fish genus Coryphaenoides, the nucleotide sequences of the mitochondrial (mt) 12 S rRNA and COI gene sequences for seven Coryphaenoides species were analyzed. Our molecular phylogenetic tree shows a new arrangement of seven Coryphaenoides species, which form two distinct groups, abyssal and nonabyssal species, and differs from the results of previous taxonomic studies. Using the mutation rate of mitochondrial genes, the divergence time between abyssal and nonabyssal Coryphaenoides was found to be 3.2-7.6 million years ago. Our study suggests that hydraulic pressure plays an important role in the speciation process in the marine environment.  相似文献

Members of the yeast genus Malassezia, including atypical strains, are lipophilic except for Malassezia pachydermatis. New physiological features that characterize atypical Malassezia strains are mainly associated with alteration in Tween assimilation pattern — such isolates still require lipids for growth. We isolated three non-lipid-dependent strains of Malassezia from patients with diagnosed atopic dermatitis (AD). These isolates could not be identified to the species level via their physiological properties. Phylogenetic trees, based on the D1/D2 regions of the 26S rDNA gene sequences and nucleotide sequences of the ITS1-5.8S-ITS2 rRNA region, showed the isolates to belong to Malassezia furfur. Three non-lipid dependent isolates from AD skin were conspecific, and sequences analysis proved them to be M. furfur.  相似文献

The Nucleotide sequence of the chitin synthase1 (CHS1) gene of Epidermophyton floccosum, an anthrophophilic dermatophyte which is the type species of the genus Epidermophyton was analyzed to determine its phylogenetic relation to eight other dermatophyte species belonging to the genera Microsporum and Trichophyton, which were sequenced in our previous studies. A genomic DNA fragment about 620 bp in length of the CHS1 genewas amplified from E. floccosum by polymerasechain reaction (PCR) and was sequenced. The CHS1 nucleotide sequence showed more than 85% similarity with sequences derived from the other dermatophytes. Phylogenetic analyses of the sequences from E. floccosum revealed that the genus Epidermophyton may be genetically distinct from Microsporum and Trichophyton. The phylogenetic relationships and taxonomic position of sea stars of the family Porcellanasteridae have been a subject of debate for over a hundred years. In this paper, the methods of molecular phylogenetic analysis were used to solve these issues. The nucleotide sequences of fragments of the mitochondrial gene of subunit 1 of cytochrome oxidase (COI) and those of the 16S rRNA gene were obtained for 9 and 6 species of porcellanasterids for the first time, respectively. The phylogenetic trees including these sequences indicate the nearest relationships of the family Porcellanasteridae with the families Ctenodiscidae and Goniopectinidae, thereby supporting the validity of the allocation of these three families in the suborder Cribellina.  相似文献

将自测的我国直翅目蝗总科7科7种和从GenBank中下载的17种直翅目昆虫的18S rDNA序列片段进行了同源性比较,用似然比检验的方法对序列比对结果进行了碱基替代模型的选择,以蚱总科的Paratettix cucullatus和蜢总科的Stiphra robusta作外群,用NJ、MP、ML和贝叶斯法构建了分子系统树。在获得的1 849 bp的序列中,有205个变异位点,74个简约信息位点; A、T、C和G的碱基平均含量分别为23.9%、24.3%、23.8%和28.0%,碱基组成基本上无偏异。分子系统树表明:所研究的内群聚为4支,锥头蝗科、瘤锥蝗科、斑腿蝗科、网翅蝗科、槌角蝗科和剑角蝗科都不是单系。建议将蝗总科分为4科,即锥头蝗科、大腹蝗科、癞蝗科和蝗科。  相似文献

We determined nucleotide sequences of homologous 0.9-kb fragments of mitochondrial DNAs (mtDNAs) derived from four species of old-world monkeys, one species of new-world monkeys, and two species of prosimians. With these nucleotide sequences and homologous sequences for five species of hominoids, we constructed a phylogenetic tree for the four groups of primates. The phylogeny obtained is generally consistent with evolutionary trees constructed in previous studies. Our results also suggest that the rate of nucleotide substitution for mtDNAs in hominines (human, chimpanzee, and gorilla) may have slowed down compared with that for old-world monkeys. This evolutionary feature of mitochondrial genes is similar to one found in nuclear genes.   相似文献
