In light, malformin completely abolished the ability of Ag+to inhibit Ethrel-induced leaf abscission from cuttings of Vignaradiata, even though Ag+ was applied 24 hr before malformin.Malformin itself did not induce abscission in the light. However,Ag+ was active on cuttings which had been pre-treated with malforminfor 2 days in the light. No evidence was obtained to suggestreaction between malformin and Ag+. In vitro cultivated apple pedicels without a primary abscission layer can form a secondary (adventitious) abscission layer, especially under the influence of auxins. In the apple cv. Cox's Orange Pippin abscission can only be induced by auxins, while the site of the abscission layer, a few millimetres from the basal ends of the pedicels, is fixed and independent of the auxin concentration. The auxin treatment has to last at least 5–6 days to induce abscission, which is not affected by the presence of a flower. A secondary layer does not occur when pedicels are placed inverted on the media. Although abscission occurs both in light and in darkness, it is strongly promoted by light. Abscission is also accelerated by raising the temperature from 9°C to 21–25°C. High concentrations of 2,3,5-triiodobenzoic acid reduce the percentage of auxin-induced abscission. Sugar is required, but the presence of macro-elements is not essential. 相似文献

在众多遗传转化法中,农杆菌(Agrobacterium tumefaciens)介导法以易操作、低费用、插入片段明确、拷贝数低等独特优点成为植物遗传转化的首选。然而,至今仍有许多物种不能被农杆菌转化。研究表明,农杆菌的转化能力是由位于染色体基因组之外Ti质粒上的vir基因决定的。在所有vir基因中,除virA和virG组成型表达外,其它vir基因的表达均需酚类化合物的诱导;糖类物质可增强酚类化合物对vir基因的诱导;低磷酸和酸性pH环境也可促进vir基因的诱导表达。文章论述了酚类化合物、糖类物质、低磷酸、酸性pH和培养温度等因素对农杆菌vir基因诱导表达的影响,以期为更好地利用这一天然载体及为提高转化效率提供依据。 相似文献

SYNOPSIS. The effect of osmotic pressure, different electrolytes and organic compounds on cyst formation in Acanthamoeba palestinensis has been tested. The optimal osmolarity for encystment was similar to that of the growth medium. Iso-osmotic solutions of NaCl, KCl, MgCl2 , CaCl2 , glycine and sucrose led to maximum cyst formation. 以黄瓜品种‘长春密刺’幼苗为材料,研究了亚精氨(Spd)诱导黄瓜幼苗对白粉病的抗性,并测定Spd处理和白粉菌接种对黄瓜叶片4种防御酶活性及3种防卫基因表达的影响。结果显示:(1)0.2~1.0mmol.L-1 Spd对黄瓜幼苗白粉病抗性均有不同程度的诱抗效果,并以0.8mmol.L-1 Spd处理效果最明显,诱导效率可达55.3%。(2)喷施Spd或接种白粉菌均可提高黄瓜叶片过氧化物酶(POD)、苯丙氨酸解氨酶(PAL)、几丁质酶和β-1,3-葡聚糖酶的活性,且诱导并接种处理的植株叶片上述酶活性均比只诱导不接种处理的上升速度更快;同时,Spd处理和接种白粉菌可以提高植株叶片中POX、PAL、PR-1a基因的表达量。研究表明,Spd处理可以诱导防卫基因表达的增强,提高防御酶活性,显著降低病情指数,增强黄瓜幼苗对白粉病的抗性。 相似文献

Photosynthesis, Dark Respiration and Bud Sugar Concentrations in Pepper Cultivars Differing in Susceptibility to Stress-induced Bud Abscission 总被引:2,自引:0,他引:2
Pepper (Capsicum annuum L.) cultivars differ in susceptibilityto stress-induced abscission. Previous research indicates thatthe stress susceptible cultivar 'Shamrock' undergoes a largerreduction in net assimilation rate (NAR) under low light stress,and partitions less dry matter (DM) to reproductive structuresand more to leaves than the more tolerant cultivar 'Ace'. Todetermine if photosynthetic rates under low light stress couldexplain NAR differences, photosynthesis was measured on 'Ace'and 'Shamrock'. Assimilate partitioning was compared throughmeasurement of leaf and bud respiration rates and analysis ofbud sugar concentrations. Photosynthetic rates per unit leafarea of leaves fully exposed to incident light revealed no cultivardifferences under low light conditions. Bud respiration ratesfell to a lower level in 'Shamrock' than 'Ace' in low light-stressedplants, while expanded leaves respired at higher rates in 'Shamrock'than 'Ace' under both full and low light. Bud sugar concentrationswere significantly lower in 'Shamrock' than 'Ace' after 3 dof low light stress. Seventy-seven patients with severe brachial plexus injuries were interviewed two or more years later to determine their success in returning to work and the factors that had led to good or bad resettlement. For most of them these were crucial issues potentially influencing the rest of their lives. When interviewed virtually all had regular jobs in open industry, but many had endured long delays and most were working entirely one-handed. Failure of communication was regrettably common. Too often advice by doctors had been lacking, and there was evidence that the services for vocational resettlement could be improved. 相似文献

Induction of resistance to crown rust, caused by Puccinia coronata Cda. var, avenae Frazer et Led., occurred in seedlings of three genetic lines of oats (Avena sativa L.) inoculated sequentially with one of three inducer-challenger combinations of the pathogen. There was significant reduction in both the number of pustules per leaf and the weight of urediospores harvested in all three host genotypes tested. There were highly significant differences between individual spore harvests correlated with age of the pustules. Development of telia by the challenger was found to occur considerably later, and was less intense, when the challenger was inoculated alone in contrast to treatments in which the challenger followed pre-inoculation with the inducer. There was no correlation between the numbers of resistant and susceptible lesions within the range of inoculum density used in these experiments. 相似文献

目的:探讨人腭部涎腺腺样囊性癌细胞(NACC)经顺铂(DDP)短期诱导后耐药性产生情况及耐药机制。方法:采用恒定浓度DDP反复间歇诱导法诱导NACC细胞,获得耐药细胞NACC/DDP3,MTT法检测细胞耐药指数;实时荧光定量PCR检测存活蛋白(Survivin)及核苷酸切除修复交叉互补基因1(ERCC1)的mRNA表达;裸鼠右侧腋部皮下接种NACC及NACC/DDP3细胞,成瘤后将荷瘤裸鼠随机分为生理盐水对照组和DDP 2 mg·kg~(-1)组,比较2 mg·kg~(-1) DDP对两组荷瘤裸鼠的抑瘤率,HE染色观察肿瘤组织形态。结果:诱导后的NACC/DDP3对DDP耐药性增强,耐药指数为1.44;在NACC/DDP3细胞中,Survivin及ERCC1的mRNA表达明显上调,分别为亲本细胞的2.02(P0.01)和1.59(P0.05)倍;2 mg·kg~(-1) DDP对NACC组抑瘤率为(31.64±1.15)%,对NACC/DDP3组抑瘤率为(16.66±0.76)%,两组间差异具有统计学意义(P0.01),HE染色观察两组瘤体标本均符合腺样囊性癌实体型组织学表现。结论:NACC细胞经DDP短期诱导即产生一定耐药性,NACC/DDP3细胞中Survivin及ERCC1的mRNA表达上调,提示Survivin及ERCC1可能参与了腺样囊性癌细胞对DDP的耐药。相同剂量DDP对NACC/DDP3所建立的皮下移植瘤模型抑瘤率显著低于NACC组,提示NACC/DDP3接种到裸鼠体内仍具有耐药性,这将为体内研究耐药腺样囊性癌的治疗提供良好的平台。 相似文献

以链格孢菌Alternaria alternata和烟草Nicotiana tabacum品种云烟87为试材,研究不同浓度茉莉酸甲酯(MeJA)和水杨酸(SA)处理下,链格孢菌菌丝生长情况以及成熟期烟叶防御酶活性与多酚含量变化,探讨植物生长调节剂对烟草抗链格孢菌的影响。结果表明,1 mmol·L–1 MeJA对链格孢菌的抑制效果最好,抑菌率高达59%以上,其次是3.5 mmol·L–1SA;MeJA和SA对链格孢菌的抑制效果随药剂浓度升高而增强;0.1 mmol·L–1 MeJA和2.5 mmol·L–1 SA能诱导烟草叶片SOD、POD、CAT等防御酶活性,并能降低H2O2活性氧含量,尤其以MeJA诱导效果较好。2种植物生长调节剂处理并接种链格孢菌能诱导提高烟叶多酚代谢相关酶PPO及PAL活性,但对烟草多酚类物质影响较小;成熟烟叶中含量较高的前3种多酚物质是绿原酸、芸香苷、隐绿原酸。 相似文献

To induce somatic embryogenesis in Quercus robur L. immature zygotic embryos at different developmental stages were collected in weekly intervals from June until September
in three consecutive years from four open pollinated trees at two Vienna sites. Acorns were surface sterilised and cultured
firstly on P24 medium with 5μM 2,4-dichlorophenoxyacetic acid (2,4-D) and 0.5 μM 6-benzylaminopurine (BAP) or on hormone-free
P24 medium and secondly on P24 medium with 0.9 μM BAP. The formation of white-yellow globular structures of somatic embryos
started during the fourth week after the induction treatment. High induction frequencies of 30 - 80 % were achieved on 2,4-D/BAP
medium, whereas rates on hormone-free medium were below 20 %. The initiation of somatic embryogenesis was favoured in the
heartshaped and early cotyledonary stage of the zygotic embryo in all three years and lasted until the acorns reached maximum
size in August.

观察了肽类生长因子对2BS细胞中抗癌基因表达的影响。结果表明,EGF或FGF均可明显诱导2BS细胞中Rb基因的表达,其幅度分别为305%、243%;EGF也可诱导2BS细胞中TGFβ1基因表达增加30-50%,但FGF对TGFβ1的表达无明显影响;EGF或FGF对p53基因的表达均无明显诱导作用。上述结果为生长因子作用机制研究及生长因子与抗癌基因的关系提供了新的线索。 相似文献
