One hundred and thirty-five microsatellite markers were developed for hop Humulus lupulus L. from di- and trinucleotide-enriched libraries. Seventy-eight primers showed amplification in two tested genotypes. Twenty-four loci were further characterized on a population of 34 hop samples and the number of alleles per locus, observed heterozygosity and expected heterozygosity ranged from two to 20 (9.7 on average), from 0.0294 to 0.9412 (0.6234 on average) and from 0.0294 to 0.9170 (0.6720 on average), respectively. These microsatellite markers will be further used for studying population structures and relationships and for identifying important qualitative and quantitative loci of hop.  相似文献

The occurrences and distributions of hop stunt (HSVd) and hop latent (HLVd) viroids were assessed by a nucleic acid hybridisation assay, using samples from 476 commercial hop plantings in the UK. These samples represented about half of the UK production.
HLVd was detected in c. 17% of the samples, with infection in different cultivars ranging from 0% to 89%. This viroid was found in all cultivars sensitive to Verticillium wilt except cv. Sunshine, an old cultivar grown on only one farm in the UK. Two minor wilt-tolerant cultivars were also found to be infected at low frequencies, but the main commercially-important wilt-tolerant cultivars were all uninfected. A high proportion of the nuclear stock mother plants in the "A +" house at the Institute of Horticultural Research Dept of Hops Research, Wye College were infected. Circumstantial evidence, based on the planting dates of infected gardens, suggests that infection became established in the hop propagation system during the late 1970s and that there was a major increase in the prevalence of HLVd as a result. Whether this contamination of propagating material arose because of spread from long-standing infections or because the viroid was newly introduced into the UK, is not known.
All samples were also tested for HSVd but this viroid was not detected in any UK hop material.  相似文献

Hop (Humulus lupulus L.) cultivars are vegetatively propagated and it is difficult to differentiate them during the process of propagation. Fingerprinting with molecular markers based on DNA could be a useful means of identifying different cultivars. Simple sequence repeats, or microsatellite markers, are the most suitable marker for genetic fingerprinting because they are multi‐allelic and co‐dominant. For this purpose, we have developed primers for 10 new polymorphic microsatellite loci that are suitable for genetic fingerprinting in hop.  相似文献

 A cryopreservation procedure using encapsulation/dehydration was established for shoot-tips obtained from in vitro-grown shoots of hop. After dissection, shoot-tips were encapsulated in medium with alginate and 0.5 M sucrose. Optimal conditions consisted of preculture for 2 days in solid medium with 0.75 M sucrose, or in increasing sucrose concentrations, desiccation for 4 h with silicagel in a flow cabinet (16% water content) followed by rapid freezing and slow thawing. Shoot recovery after freezing 60 min in liquid nitrogen was around 80%. No phenotypical changes were observed in the recovered plants from cryopreserved shoot-tips growing in the field. Chalcones xanthohumol (X) and desmethylxanthohumol (DMX), present in hops (Humulus lupulus L.), and the corresponding flavanones isoxanthohumol (IX, from X), 8-prenylnaringenin (8-PN, from DMX), and 6-prenylnaringenin (6-PN, from DMX), have been examined in vitro for their anti-proliferative activity on human prostate cancer cells PC-3 and DU145. X proved to be the most active compound in inhibiting the growth of the cell lines with IC50 values of 12.3±1.1 μM for DU145 and 13.2±1.1 μM for PC-3. 6-PN was the second most active growth inhibitor, particularly in PC-3 cells (IC50 of 18.4±1.2 μM). 8-PN, a highly potent phytoestrogen, exhibited pronounced anti-proliferative effects on PC-3 and DU145 (IC50 of 33.5±1.0 and 43.1±1.2 μM, respectively), and IX gave comparable activities (IC50 of 45.2±1.1 μM for PC-3 and 47.4±1.1 μM for DU145). DMX was the least active compound. It was evidenced for the first time that this family of prenylated flavonoids from hops effectively inhibits proliferation of prostate cancer cells in vitro.  相似文献

Incubation of nuclei from rat liver or human leukemic cells in the presence of 3H-UTP2 and other factors results in th incorporation of label into a material precipitable by acid, alcohol or ether. This materials is isolated by phenolsds extraction, is sensititve to ribonuclease digestion and presumed to be RNA.

The addition of Cu++ to the incubation system is necessary to inhibit RNA breakdown and allows the isolation of undegraded RNA without interefering with th incorporation of radiosactivity. The time patterns of labl incorporation by the two nuclei preparations are different. Whereas label incorporation by th two nuclei preparations are different. Whereas labelincorporation by liver nuclei continues to increase up to 60 minutes, incorporation by th leukemic nuclei is high during the first 10 minutes and continues at a slower rate up to 45 minutes of incubation. further, th two nuclei preparations also synthesize diferent RNA species. While liver nuclei synthesize RNA sedimenting at 4.5S and 7S to 13S, leukemic nuclei synthesize a heterogeneous, polydisperse type of RNA.  相似文献

Hop flavonoids are being regarded as attractive molecules to prevent or treat certain forms of cancer. Studies have focused mainly on xanthohumol, the most abundant prenylated chalcone existing in hops extract. However, during the production of beer, or after its ingestion, xanthohumol originates different metabolites, among which isoxanthohumol and 8-prenylnaringenin. The aim of this work was to study the effect of the prenylflavonoids xanthohumol, isoxanthohumol and 8-prenylnaringenin on the breast cancer Sk-Br-3 cell line proliferation, apoptosis and activity of the enzyme aromatase (estrogen synthase). Aromatase activity was determined by a tritiated water assay, cell proliferation was assessed by [3H]thymidine incorporation, sulforhodamine B protein measurement and Ki-67 immunostaining and apoptosis was determined by TUNEL. Our results show that all tested prenylflavonoids were able to inhibit aromatase activity and thus, estrogen formation. Additionally, breast cancer cell line proliferation was decreased and apoptosis induced by all three compounds. The presence of 17β-estradiol in treatment medium was able to revert the effect of the prenylflavonoids on cellular proliferation. These observations strengthen the idea that hop flavonoids may have anti-breast cancer effects and shed new light on a possible mechanism of action by which these effects occur, namely through their ability to decrease estrogen synthesis.  相似文献

以啤酒花的幼嫩茎段作为外植体,研究出理想的一次性成苗培养基:MS+6-BA0.01mg/L+IAA0.1mg/L+琼脂6.5g/L+葡萄糖2%。试管苗月增殖率4.2,生根率100%,成苗率90%。采用试管苗微扦插的方式进行快速繁殖,经锻炼后直接移入经改造后的土壤中,移栽成活率达93.8%。  相似文献

AIMS: To examine the resistance of beer isolates of lactic acid bacteria (LAB) towards a mixture of tetrahydroiso-alpha-acids (Tetra) by growth experiments as well as by measurement of intracellular pH. METHODS AND RESULTS: Beer LAB isolates were identified to species level by SDS-PAGE of whole-cell proteins. Beer isolates of Lactobacillus brevis showed better ability for growth in the presence of Tetra than nonbeer isolates of the L. brevis or other species of LAB including beer and nonbeer isolates. The antimicrobial effect of Tetra was also examined by noninvasive measurement of intracellular pH by fluorescence ratio imaging microscopy for selected beer isolates of L. brevis and Pediococcus inopinatus. Strains of L. brevis showing limited decrease of intracellular pH during exposure to Tetra also showed better ability for growth in the presence of these compounds as well as in commercial beer products. CONCLUSIONS: It was possible to apply a method for noninvasive measurement of intracellular pH to predict the resistance of beer spoilage LAB towards the Tetra hop analogue compounds. SIGNIFICANCE AND IMPACT OF THE STUDY: This study demonstrated the usability of a new rapid method for detecting hop-resistant variants of known beer spoilage LAB species.  相似文献

Introduction – The rising trend to consume herbal products for the treatment and/or prevention of minor ailments together with their chemical and pharmacological complexity means there is an urgent need to develop new approaches to their quality and stability. Objectives – This work looks at the application of one‐dimensional diffusion‐edited 1H‐NMR spectroscopy (1D DOSY) and 1H‐NMR with suppression of the ethanol and water signals to the characterisation of quality and stability markers in multi‐component herbal medicines/food supplements. Material and Methods – The experiments were performed with commercial tinctures of Valeriana officinalis L. (valerian), expired and non‐expired, as well as its combination with Hummulus lupulus L. (hops), which is one of the most popular blends of relaxant herbs. These techniques did not require purification or evaporation of components for the qualitative analysis of the mixture, but only the addition of D2O and TSP. Results – The best diagnostic signals were found at δ 7 ppm (H‐11, valerenic acid), δ 4.2 ppm (H‐1, hydroxyvalerenic acid) and δ 1.5‐1.8 ppm (methyl groups in prenylated moieties, α‐acids/prenylated flavones). Conclusion – This work concludes on the potential value of 1D DOSY 1H‐NMR to provide additional assurance of quality in complex natural mixtures. Copyright © 2016 John Wiley & Sons, Ltd.  相似文献

The aim of this study was to assess the effects of hop cones (Humulus lupulus L.) from two varieties Aurora and Dana, differing in their α- and β-acid contents, on rumen microbial activity measured with in vitro gas production kinetics and short-chain fatty acids (SCFA) production. Hop cones were added to the total mixed dairy cow ration (CONT) in concentrations simulating a cow’s daily intake of 50, 100 and 200 g of hop cones – the concentrations of hop cones expressed on a substrate basis were 43, 82 and 153 mg/g of substrate. Substrates were anaerobically incubated in glass syringes, and gas production kinetic parameters were determined by fitting data with the Gompertz model. Gas produced after 24 h (Gas24), maximum fermentation rate (MFR) and time of maximum fermentation rate (TMFR) were calculated from the estimated gas production kinetic parameters. After 24 h of incubation, the fermentation liquids of each substrate were taken for the determination of SCFA. Increasing the hop cone concentration decreased the total potential gas production, Gas24, MFR and shortened TMFR. The highest hop cone concentration significantly decreased acetic and butyric acid productions and total SCFA production after 24 h of incubation, but not propionic acid production, resulting in a decreased ratio between acetic acid and propionic acid.  相似文献

The genetic diversity of 159 representative genotypes of native hop (Humulus lupulus var. lupuloides E. Small, Cannabaceae) from 34 selected populations was assessed by relative magnitudes and ranges of alpha acids (AA), beta acids (BA), and the cohumulone (CoH) component of alpha acids, with reference to temporal changes between 1989-1990 and 2001, and to the same attributes in American and European hop cultivars, principally H. lupulus var. lupulus L. Chemical profiles of these genotypes were generated by high pressure liquid chromatography (HPLC) of methanol extracts from their processed samples (cones). The alpha ratio (AR, alpha acids / alpha+beta acids) measured the degree to which alpha acids predominated in cone extracts. Synchronous ranges of AR and CoH were also selected for graphic portrayals of native hop genotypic diversity. Cones sampled and analyzed from eight populations that were accessible in both 1989 and 2001 were distinct in chemical attributes, indicating a succession of genotypes, and suggesting temporal cycling of H. lupulus var.lupuloides germplasm. The principal distinctions between the two sub-species were a markedly higher proportion of CoH (38-88% vs. 19-41%) in alpha acids of H. l. var. lupuloides, and generally higher concentrations of AA in cultivars of both American and European commercial hop cultivars, predominantly H. lupulus var. lupulus. All of the 159 native hop genotypes also contained detectable levels of xanthohumol and xanthogalenol, prenylflavonoids recently reported to have mammalian anti-cancer activity. Some native genotypes had previously exhibited natural repellence of insect and mite pests; thus H. lupulus var. lupuloides germplasm offers a diverse resource of underutilized and yet undefined biochemicals.  相似文献

Female flowers of hop (Humulus lupulus) are an essential source of terpenoid‐related compounds, which are mainly used as flavoring in the beer brewing process. The compounds involved are bitter acids, terpenophenolics, as well as mono‐ and sesquiterpenoids. In this work, we analyzed the proteome of purified glandular trichomes (lupulin glands) from female flowers, which produce and accumulate these compounds. An extensive 2D‐LC‐MS/MS analysis identified 1015 proteins. Of these, most correspond to housekeeping and primary metabolism‐related proteins, albeit predominantly including amino acid and lipid metabolism, which feeds the specialized (secondary) metabolism. Indeed, 75 proteins belong to the specialized metabolism. No less than 40 enzymes are involved in the synthesis of terpenoid‐derived compounds and 21 are predicted transporters, some of which might be involved in the transport of specialized metabolites. We discuss the possible routes involved in the intra‐ and intercellular translocation of terpenoids and their precursors. This comprehensive proteomic map of the glandular trichomes of hop female flowers represents a valuable resource to improve our knowledge on the function of glandular trichomes.  相似文献

Investigations of cell wall teichoic acid structures of various staphylococci were carried out by a rapid method based on the gas-liquid chromatographic separation of products obtained after treatment of phenol-extracted cells with 70% hydrofluoric acid. In most of the strains teichoic acids of the poly(glycerolphosphate) and/or poly(ribitolphosphate) type were found. Teichoic acids of the poly(glycerolphosphate-N-acetylglucosaminephosphate) type and polymers consisting of N-acetylglucosaminephosphate were present in few strains.The results obtained by the rapid chemical screening method were compared with data obtained by serological analysis of teichoic acid structures using specific antisera and the lectin wheat germ agglutinin. Teichoic acid components occurring in low concentrations could only be detected with the chemical and not with the serological method. A number of strains of species of the genus Staphylococcus have been studied using these rapid methods. Summary An experiment in which perennial ryegrass (Lolium perenne L.) was grown in solution cultures containing various forms of radiosilver (110mAg) is ptake of labelled Ag from solutions containing either AgNO3, Ag2SO4, AgCH3COO, Ag-humic acid (HA) or Ag-fulvic acid (FA) complexes is compared. In each case concentrations of Ag in the plant roots was several fold higher than in the plant shoots. However the Ag concentration in roots from the Ag-HA preparations was only about 5% of that from the AgNO3 solutions; from Ag-FA it was 18%. Transport index values were highest for plants grown in the Ag-HA solution, indicating greater mobility of Ag within these plants. Preliminary data for a selective extraction procedure performed on Ag-HA preparations suggest sorption onto various active sites. The relevance of these findings to the cycling of Ag within soils is discussed.  相似文献

利用GCMS计算机联用方法从我国阿魏属(FerulaL.)15种1变种根部挥发油中分离鉴定出214种成分,其中广布成分有22种。挥发油成分中80%是萜类,其次是多硫化合物。萜类中主要有单萜和倍半萜两大类,均以环化结构成分为主。通过全成分和萜类成分的多种聚类分析,对样品进行归类,结合形态特征、核型、黄酮成分等已有资料以及地理分布,讨论了挥发油成分在阿魏属分类中的意义,同时对所分析种类进行了类群划分和调整  相似文献

To cryopreserve sugar beet shoot tips using an encapsulation-dehydration technique, cold hardening of in vitro plants was needed to obtain high survival rates after freezing. Cold acclimation not only enhanced dehydration and freezing tolerance, but also induced several changes in sugar beet shoots. Plants contained greater amounts of sucrose, D-glucose and D-fructose and the fatty acid composition of lipids changed. Furthermore, the unsaturation level of membrane lipids, estimated by the (C18:2 + C18:1)/C16:0 ratio, increased after cold hardening. These changes were correlated with better survival rates after cryopreservation.  相似文献

Vitex rotundifolia L. is widely distributed along the sea coast of China. The aim of this study was to investigate the anti-nociceptive and anti-hyperprolactinemia activities of substances isolated from Fructus Viticis (the fruit of Vitex rotundifolia), which may be effective in the treatment of pre-menstrual symptoms, using acetic-acid-induced writhing and metoclopramide-dihydrochloride-induced hyperprolactinemia in mice. The fractions effective in terms of anti-nociceptive and anti-hyperprolactinemia activities were obtained from Fructus Viticis by elution through macro-porous resin, and polyamide and silica gel column chromatography. The standardization of the fractions obtained from the separation procedures was carried out by means of high-performance liquid chromatography (HPLC)-fingerprint. In this study, the flavone-enriched fraction (Fraction 6) showed a higher inhibitory rate than indomethacin (69.4% vs. 56.4%) at a dose of 50 mg/kg body wt., and significantly reduced the prolactin level as compared to HPRL-treated mice (8.2 ng/ml vs. 25.5 ng/ml). Furthermore, this fraction showed anti-nociceptive activity in a dose-dependent manner (10-50 mg/kg body wt., i.g.). On further purification with silica gel, Casticin was isolated from this fraction and it decreased abnormal serum levels of prolactin by approximately 50% (p < 0.01). Using bioassay-screening methods, our results indicate that the presence of flavonoids such as Casticin in this plant may be responsible for the activity effects. Casticin has potent analgesic and anti-hyperprolactinaemia properties, is likely to be one of the active components of Fructus Viticis, and may have a role in treating PMS (premenstrual syndrom).  相似文献

林生山黧豆幼苗用[3H]-天门冬氨酸标记后,高丝氨酸在6h内迅速增加。高丝氨酸合成速率降低后,2,4-二氨基丁酸的合成量上升,于18h达到高峰。赖氨酸和苏氨酸与二氨基丁酸的合成表现有协同反馈机制。结果支持了天门冬半醛转氨生成二氨基丁酸的假说。盐胁迫、渗透胁迫和热激增加了二氨基丁酸的合成,可能是因为不同胁迫条件都造成了细胞脱水,从而促进了二氨基丁酸的合成。  相似文献
