Barley mild mosaic virus inside its fungal vector, Polymyxa graminis   总被引:2,自引:0,他引:2  
In an electron microscope study to investigate the association of barley mild mosaic virus (BaMMV) with its fungal vector, Polymyxa graminis, thin sections were made of zoospores of the vector and of barley roots containing different stages in the life cycle of the fungus. Immunogold labelling was used to identify the virus in sections. Labelled bundles of presumed virus particles were seen in c. 1% of zoospores liberated from plant roots and in zoospores inside zoosporangia. A few zoosporangial plasmodia had localised labelling but no bundles were seen. No virus particles were seen in sections of resting spores.  相似文献

The generic names of the brown algaeMyagropsis, Cystoseira andCystophyllum have been used in Japan, Korea and China for a small group of seaweeds whose limits have not been clearly understood. Studies on the development of the eggs and subsequent germlings show that substantial differences occur betweenCystoseira andMyagropsis. Myagropsis Kützing is distinguished fromCystoseira C. Agardh by the following characteristics: (1) the tongue cell is undivided during development of the conceptacle; (2) paraphyses are projected from the conceptacle ostiole and become entangled; (3) during development, oospore germlings are mixed among paraphyses projecting from the ostiole; (4) oospores are large, with eight nuclei at maturity; (5) thirty-two primary rhizoids are produced on the germlings; and (6) the thallus is bilateral in organization. The shape and size of vesicles, their formation, and the presence of cryptostomata have been used as specific characters, but their use cannot be continued. It is concluded that the genusMyagropsis is monotypic, with a single species,M. myagroides (Turner) Fensholt. The status of this species is also discussed.  相似文献

The brown alga Cladosiphon zosterae (J. Ag.) Kylin (Chordariaceae) is found to exist in culture in a variety of forms, including filaments, discoid or crustose forms, and two types of erect cylindrical thalli. Asexual spores from plurilocular sporangia of each form can produce any and all forms. Three factors controlling morphological expression in C. zosterae have been identified. These factors include: (1) heteroblasty (formation of filamentous and discoid germlings as a consequence of alternate modes of spore germination), which was independent of all environmental factors tested including temperature, photoperiod, nitrogen source, and general fertility of the growth medium; (2) influence of certain nitrogen sources, notably ammonium, which induce a compact discoid or crustose form; and (3) a morphogenetic factor presumed to be bacterial which determines mode of development and subsequent morphology of the erect Cladosiphon thallus.  相似文献

Groundnut (Arachis hypogaea) plants from Nigeria with chlorotic rosette disease contained a manually transmissible virus, considered to be a strain of groundnut rosette virus (GRV(C)). GRV(C) infected nine out of 32 species in three out of nine families. It caused local lesions without systemic infection in Chenopodium amaranticolor, C. murale and C. quinoa, and systemic symptoms in Glycine max, Nicotiana benthamiana, N. clevelandii and Phaseolus vulgaris as well as in groundnut. Some ‘rosette-resistant’ groundnut lines were also infected. GRV(C) was transmitted by Aphis craccivora, but only from groundnut plants that were also infected with an aphid-transmissible second virus, which was not manually transmissible and was considered to be groundnut rosette assistor virus (GRAV). Plants infected with GRAV contained isometric particles c. 25 nm in diameter which were detectable by immunosorbent electron microscopy on grids coated with antisera to several luteoviruses, especially with antisera to bean leaf roll, potato leafroll and beet western yellows viruses. No virus-like particles were observed in extracts from plants infected with GRV(C) alone. A single groundnut plant obtained from Nigeria with symptoms of green rosette contained luteovirus particles, presumed to be of GRAV, and yielded a manually transmissible virus that induced symptoms similar to those of GRV(C) in C. amaranticolor but gave only mild or symptomless infection of N. benthamiana and N. clevelandii. It was considered to be a strain of GRV and designated GRV(G).  相似文献

Summary Gametophytic germlings ofAllomyces macrogynus are examined with regard to the sequence of developmental events which have been suggested to be effected by long-lived messenger RNA. During encystment of the meiospore, the cytoplasmic triplet microtubules are depolymerized, the gamma bodies are mobilized, the nuclear cap envelope is fragmented and the cap ribosomes are dispersed, the side body complex breaks into its components, and a cyst wall is deposited. All these processes take place prior to the appearance of significant amounts of protein synthesis. The first fission of the mitochondria occurs shortly after the onset of protein synthesis but before detectable RNA synthesis. The developmental sequence of gametophytic and sporophytic germlings was found to be very similar.  相似文献

Summary The leucine-lysine synchronization technique of Dill and Fuller (1970) has been further refined and used to study various biosynthetic events of pre-mitotic germlings of Allomyces neo-moniliformis (the time of DNA replication, RNA synthesis, and protein synthesis), and various morphogenetic changes (germling development, nuclear cap breakdown, and the first mitotic nuclear division). The degree of synchrony induced in a population of germlings appears to be determined by the time when the zoospores are induced to encyst and germinate rather than by the duration of the swimming period of the zoospore. DNA replication, nuclear cap breakdown, early protein synthesis, and morphogenetic development appear to occur prior to messenger RNA synthesis in developing thalli and thus would be under the control of pre-existing messenger RNA. The degree of synchrony of particular morphogenetic or biosynthetic developmental changes induced in a population of A. neo-moniliformis germlings must be determined for each aspect of development which is to be studied.  相似文献

Summary Dikaryotic cells of S. commune synthesized polyols throughout the life cycle when grown on glucose, cellobiose, or cellulose. Basidiospores contained arabitol and mannitol which were depleted during germination. The mannitol content of the young germlings rose to normal levels within a day; arabitol accumulation remained depressed for 5 to 7 days and then returned to normal levels characteristic of vegetative cells. Individual homokaryons differed in their production of intracellular polyols, which, unlike germlings, remained constant with cultural age. Homokaryon (str. 699) produced low levels of arabitol but high levels of glycerol while another homokaryon (str. 845) was the reverse. Mixtures of these homokaryons as well as the dikaryon (699×845) produced arabitol and glycerol levels intermediate between the parent homokaryons. High concentrations of glucose did not change the nature of the polyols produced. Arabitol formation could be induced prematurely in germlings or elevated in the dikaryon by growth on acetate or ethanol. Both homokaryons responded to growth on acetate with elevated arabitol production; acetate induction of arabitol formation was repressed in all types of cells if glucose were added simultaneously with acetate. Maltose, cellobiose, and trehalose also stimulated arabitol formation in young germlings, suggesting that glucose repression was the cause of decreased arabitol formation in basidiospore germlings. There was no correlation between the formation of arabitol and the derepression of isocitrate lyase or change in specific activities of alkaline and acid phosphatase in germlings grown on various carbon sources.  相似文献

We have been working to characterize viruses that infect the HAB‐forming pelagophyte Aureococcus anophagefferens Hargraves et Sieburth. Field samples were collected during brown‐tide events in 2002 and tested for the presence of lytic agents. Here, we describe a recently isolated, lytic virus‐like particle (VLP) that is morphologically similar to particles observed in thin sections of infected A. anophagefferens cells from natural samples. TEM and SEM have revealed VLPs consistent with the morphological characteristics of previously described Phycodnaviridae. Large icosahedral particles (~140 nm) of similar shape and morphology dominate cell lysates and are accompanied by smaller phage‐like particles and heterotrophic prokaryotes that appear to be incurable from our cultures. To determine which of these particles interacts with the Aureococcus cells, we preserved cultures during the early stage of infection so that SEM could be used to visualize those particles that attach to the surface of naïve cultures. SEM revealed that 63% of the large icosahedral‐shaped particles attached to A. anophagefferens cells after only 30 min of exposure, while no significant frequency of attachment to the alga was observed for the phage‐like particles. The results of these observations are in contrast to previous studies, where phage‐like particles were reported to infect cells. When considered in conjunction with field observations, the results suggest that this newly isolated virus represents the dominant virus‐morphotype associated with bloom collapse and termination.  相似文献

The life history of Callithamnion byssoides Arnott ex Harv. in Hook. has been shown to comprise a regular sequence of gametophytic, carposporophytic, and tetrasporophytic phases in unialgal culture using supplemented seawater media. Tetraspore germlings gave rise to gametophytes bearing either antheridia or carpogonia in 13 days. Fertilization, carposporophyte development, and carpospore release took place within 5 days. Carpospore germlings produced mature tetrasporophytes in 13 days. The life history thus required approximately 1 month for completion. No deviations from this pattern were observed.  相似文献

The mature female conceptacle of Sargassum horneri (Turner) C. Agardh has an ostiole filled with a gelatinous plug. The oogonium in the conceptacle has cell walls that can be differentiated into a dense outer and a less dense inner microfibrillar layer. Just prior to egg release, stalk material is produced inside the outer layer and the inner layer disappears. At this stage the gelatinous plug is extruded and mucilage is released through the ostiole. The released eggs are retained on the receptacle by the stalk and are surrounded by a large amount of the mucilage. Three-celled germlings form a primary wall with a polylamellated structure of microfibril layers. In multicellular germlings that have differentiated into thallus and rhizoids, the peripheral thallus cells have an outer cell wall consisting of a microfibril layer under the primary wall, while the cell wall of the rhizoid tip has an amorphous structure. The germlings are released from the stalk and become attached to the substratum by an adhesive substance secreted from rhizoidal cells.  相似文献

DNA probes, made by cloning double-stranded forms of each of the genome parts (DNA-1 and DNA-2) of the Kenyan type isolate of African cassava mosaic virus (ACMV-T), reacted strongly with extracts from Nicotiana benthamiana plants infected with ACMV-T, or with Angolan or Nigerian isolates that are closely serologically related to the type isolate. However, only the DNA-1 probes reacted with extracts of TV. benthamiana infected with a Kenyan coast isolate (ACMV-C), which is serologically less closely related to ACMV-T. DNA-1 and DNA-2 probes also reacted with extracts of mosaic-affected Angolan cassava plants, including some which have not yielded ACMV particles detectable by immunosorbent electron microscopy and from which virus isolates have not been transmitted to TV. benthamiana. These anomalous plants, unlike other naturally infected cassava plants, showed mosaic symptoms on all their leaves which, however, contained only traces of virus particle antigen detectable by enzyme-linked immunosorbent assay. They contain isolates of ACMV that are probably defective for particle production. ACMV-T particles accumulated optimally in N. benthamiana at 20–25°C. At 30°C fewer particles, which apparently had a slightly greater specific infectivity, were produced. At 15°C, considerable quantities of virus particle antigen, virus DNA and virus particles were produced but the particles were poorly infective, and the few that could be purified contained an abnormally large proportion of polydisperse linear DNA molecules, and fewer circular molecules than usual. Angolan isolates, whether particle-producing or not, likewise replicated better in cassava plants at 23 °C than at 30 °C. In contrast, ACMV-C attained only very low concentrations in N. benthamiana, but these were greater at 30 °C than at 23°C.  相似文献

The relationship between median grain size of sediments and the abundance in the wild of green algal mats (Ulva prolifera) on the intertidal flats of Muan, Korea, were investigated. The impact of substratum particle size on the growth and survival of germlings was examined in the laboratory. In the wild, the average annual density of algal mats was 7,950 ind m−2. The algal mats mainly occurred in sands and exhibited patchy distribution. Statistical analysis indicates significant spatial analysis differences and a significant relationship between density and the ratio of sands to silts, suggesting that the distribution and density of this species were related to particle size. In laboratory experiments, the survival rate of U. prolifera germlings was the lowest value (22%) on sediments with a median grain size of 63–125 μm. Laboratory experiments have generally shown a positive relationship between attachment or survival of the alga and substratum particles size. Our laboratory results indicate a clear link between germling settlement/survival and substratum particle size. These results explain the spatial differences in abundance observed in the field in relation to the distribution and ratio of sands to silt on the Muan flats.  相似文献

Melon necrotic leaf spot virus (MNSV) caused a major outbreak of a leaf necrosis disease of hydroponically-grown cucumber plants at Humberside in 1983. The virus had c. 33 nm diam. particles which reacted serologically with MNSV antiserum of Dutch or American origin. Virus particles, which contained a single polypeptide (mol. wt 45 × 103) and a presumed RNA species (mol. wt 1.5 × 106), had a sedimentation coefficient (s20.w) of 134 S and a buoyant density in caesium chloride of 1.35 g/cm3. The virus was mechanically transmissible, confined to species of Cucurbitaceae, transmitted by zoospores of Olpidium radicale and retained in the resting spores of the fungus. MNSV is thus both water-borne and soil-borne. O. radicale zoospores were killed in <5 min in suspensions containing 20 μg/ml of the surfactant Agral (alkyl phenol ethylene oxide). The disease did not reappear in 1984 when the cucumber crops were fed with nutrients containing 20μg/ml Agral.  相似文献

Virus-like particles (VLPs) have been found in cells of the colonial, chrysophycean alga Hydrurus foetidus (Vill.) Trev. The infected cells, probably zoospores, were discovered in material fixed directly in the field. The VLPs appear either hexagonal or pentagonal when sectioned and are therefore presumed to be icosahedra, They are 50–60 nm diam. and demonstrate distinct trilaminar coals. The VLPs were observed in the cytoplasm, but not in the chloroplast or nucleus.  相似文献

The rhabdovirus causing maize mosaic in Shiraz, Iran, is transmitted by Ribautodelphax notabilis Logvinenko (Homoptera, Delphacidae). Average size of bullet-shaped virus particles in negatively stained leaf-dip preparations of naturally or experimentally infected plants was 81 × 179 nm. The virus is transmitted to wheat and barley causing mosaic and severe stunting. Similar virus particles have been observed in leaf-dip preparations of naturally infected wheat, barley and Sudangrass. This is believed to be the first record of the involvement of R. notabilis in virus transmission. The relationship of the described isolate with similar viruses infecting gramineous plants is discussed.  相似文献

Morphological and biochemical changes associated with synchronous germination of mature, aged and desiccated akinetes of two alkaliphilic cyanobacteria, Cyanospira rippkae and Cyanospira capsulata, are described. Akinetes of both strains proved to be highly resistant to desiccation, being able to germinate, in the presence of either N2 or nitrate as nitrogen source, with a germination frequency of more than 90% after seven years of storage in a dried state. The first cell division occurred after 8–10 h of incubation, thereafter the germlings of the two strains followed a different pattern of cell differentiation. Heterocysts were first noted, in a terminal position, at 16–18 h in three-celled germlings of C. capsulata and at 21–24 h in C. rippkae, when germlings were at least seven cells in length. Akinetes of both species possessed, on a per cell basis, almost identical amounts of all photosynthetic pigments but, under nitrogen fixing conditions, photosynthetic activity (oxygen evolution) was detected only after new proteins had been synthesized, before a functional heterocyst was developed and while total nitrogen remained constant. With energy provided by aerobic respiration, a wide range of intracellular amino acids characteristic of proteins was utilised to sustain the new protein synthesis. The end of this biosynthetic activity coincided with the timing of the first cell division. From this stage on, no changes in protein concentration occurred until mature heterocysts were developed. In the presence of nitrate, no significant changes in the major germination events were observed.  相似文献

The strength by which species interact can vary throughout their ontogeny, as environments vary in space and time, and with the density of their populations. Characterizing strengths of interaction in situ for even a small number of species is logistically difficult and may apply only to those conditions under which the estimates were derived. We sought to combine data from field experiments estimating interaction strength of life stages of the barnacle, Semibalanus balanoides, on germlings of Ascophyllum nodosum, with a model that explored the consequences of variability at per capita and per population levels to the abundance of year-old algal recruits. We further simulated how this interaction affected fucoid germling abundance as the timing of their respective settlements varied relative to one another, as occurs regionally across the Gulf of Maine, USA. Juvenile S. balanoides have a weak estimated per capita effect on germlings. Germling populations are sensitive to variation in per capita effects of juvenile barnacles because of the typically large population sizes of the latter. However, high mortality of juvenile barnacles weakens the population interaction strength over time. Adult barnacles probably weakly facilitate fucoid germlings, but greater survival of adults sustains the strength of that interaction at the population level. Germling abundance is positively associated with densities of adult barnacles and negatively associated with that of juvenile barnacles. Metamorphosing cyprid larvae have the strongest per capita effect on germling abundance, but the interaction between the two stages is so short-lived that germling abundance is altered little. Variation in the timing of barnacle and A. nodosum settlement relative to one another had very little influence on the abundance of yearling germlings. Interactions between barnacles and germlings may influence the demographic structure of A. nodosum populations and the persistence of fucoid-dominated communities on sheltered rocky shores in New England. Summary Exposure of germlings of Allomyces neo-moniliformis to colchicine for 0 to 5 min after zoospore encystment was found to block 30% of germlings derived from flagellated zoospores and 55% of germlings derived from deflagellated zoospores in C-metaphase configurations at the first mitotic division. The zoospore lacks a pool of colchicine binding protein, and protein synthesis is absent during the time when colchicine first becomes effective in inducing C-metaphase. From these observations it is concluded that the microtubule subunit protein of the spindle apparatus of the first mitotic division to a large extent is derived from the depolymerization of the cytoplasmic microtubules of the zoospore. GTP, Mg2+, and ATP were observed to be antagonistic to the action of colchicine in vivo. It is suggested that these compounds may compete with colchicine for binding to the subunit protein in vivo. Germlings derived from flagellated zoospores are appreciably less subject to the action of colchicine in the presence of the antagonistic compounds than are germlings derived from deflagellated zoospores. This differential sensitivity to colchicine is interpreted as reflecting a difference in the quantity of microtubule subunit protein present at the time of exposure to colchicine.  相似文献

Porphyra haitanensis is one of the most economical nutritive marine algae; however, its production and quality are significantly jeopardized by high temperatures. Selection of heat-resistant strains will greatly reduce the economic risks and benefit to the nori industry. Three previously isolated and improved strains with a high yield were screened at 28°C and identified, of which one strain, ZS-1, showed significantly improved heat tolerance. Upon further characterizing of the cultures of the ZS-1 strain and the wild-type (WT) strain at 28°C and 30°C, the ZS-1 conchospore germlings survived at rates of 69.9% and 59.6%, while the WT conchospore germlings survived at significantly lower rates of 15.9% and 6.7%, respectively, over a period of 15 days. Furthermore, ZS-1 conchospore germlings divided at significantly higher rates of 100% and 88.6% compared to the WT conchospore germlings with 90.4% and 63.8%, respectively. When the 35-day-old conchospore germlings were transferred from the optimal temperature of 24°C to higher temperatures of 28°C and 30°C, the ZS-1 blades sustained growth over a 25-day period without decay and increase of blade lengths with a factor of 18.5 and 10.3 times, respectively. The blade lengths of the WT germlings only increased by a factor of 1.7 and 0.9 times and began to decay after being cultured for 15 days at 28°C and 30°C. At 24, 28, and 30°C, the ZS-1 blades grew 3.4, 8.6, and 8.0 times faster than those of the WT. Evidently, ZS-1 is a fast-growing and heat-resistant strain compared to the WT strain and may offer an alternative for the nori industry.  相似文献

Previous results have indicated that incorporation of surface glycoprotein into retroviral particles is not a specific process and that many heterologous viral and cellular glycoproteins can be incorporated as long as they do not have long cytoplasmic C-terminal regions which were presumed to be sterically inhibitory. In this study, this concept has been directly examined by analyzing the incorporation of the wild-type human epidermal growth factor receptor (Wt-EGFR) and of a C-terminally truncated mutant of Wt-EGFR (Tr-EGFR) into human immunodeficiency virus (HIV)-like particles. Incorporation was directly analyzed at the protein level and by immunogold labelling of enriched HIV-like particles. In agreement with the above concept, Tr-EGFR, with only 7 C-terminal amino acids (aa), was efficiently incorporated into HIV-like particles. Incorporation of the Wt-EGFR species, with 542 C-terminal cytoplasmic aa, was reduced by a factor of about 5 in comparison to that of the Tr-EGFR species. However, the Wt-EGFR species was still very significantly present in the HIV-like particles. A series of control experiments verified that this represents genuine incorporation of Wt-EGFR into the membrane of HIV-like particles. These observations allow further speculation as to the processes governing glycoprotein incorporation into retroviral particles and indicate that the internal virus structure of HIV (in particular the matrix layer [MA]) can accommodate much larger heterologous cytoplasmic domains in incorporated glycoproteins than previously assumed.  相似文献
