Abstract

Talorchestia quoyana, a sand beach amphipod, shows a rhythm of locomotor activity controlled by a circadian clock and an inhibitory circatidal clock. This article reports on an investigation of the entrainment of the circadian dock to skeleton photoperiods. Four important mathematical models for circadian rhythms are examined with respect to the results of the entrainment experiments and to predictions from the phase response curve for Talorchestia. Significant differences between the models are described, and properties of circadian rhythms not accounted for by present models are outlined.  相似文献

Abstract

Neonatal treatment with monosodium glutamate (MSG) results in a substantial degeneration of the inner layer of the retina and a decreased diameter of the optic nerves. Nevertheless, MSG‐treated animals entrain and re‐entrain to a light dark cycle. The question arises whether MSG selectively destroys the optic pathways which are involved in vision but not the retinohypothalamic trart that mediates entrainment. In these experiments not only entrainment and re‐entrainment of the circadian food intake rhythm of MSG‐treated rats was investigated but also the freerunning period under continuous bright and dim light It appears that MSG‐treated rats have shorter freerunning periods under continuous illumination than controls. Therefore, these results suggest that also those pathways involved in entrainment of the circadian food intake rhythm are affected by neonatal treatment with MSG.  相似文献

Abstract

Sugar Gliders (Petaurus breviceps) re‐entrain faster after 8‐h delay shifts of an LD 12:12 and an LD 8:16 (31–56:0.3 lux each) than after 8‐h advance shifts of these Zeitgeber cycles. In order to test whether this asymmetric re‐entrainment behavior is related to, or even caused by the phase response characteristics of the circadian system, the phase response of the activity rhythm to short and long light pulses was studied. Short light pulses (15 min of 31–56 lux against a background intensity of 0.3 lux) caused only relatively small delay shifts when applied around the onset, and more pronounced advance shifts when given at the end of the activity time (α). Onset and end of activity shifted by different amounts. Long light pulses produced by 8‐h advances and delays of one single lighttime of an LD 12:12 elicited pronounced phase delays when applied at the beginning of the activity time, but only minor phase advances when given at the posterior part of α. These results indicate that in Petaurus breviceps the phase response characteristics to long light pulses exerting parametric effects of light are responsible for the pronounced asymmetry effect in re‐entrainment. Differing phase responses of onset and end of activity point to a two‐oscillator structure of the circadian pacemaker system in this marsupial.  相似文献

In 1936, Erwin Bünning suggested that photoperiodic time measurement was a function of the circadian system. Colin Pittendrigh became an ardent supporter of Bünning's hypothesis, drawing parallels between photoperiodism and his own group's investigations of adult eclosion rhythmicity in the fruit fly Drosophila pseudoobscura. They developed several more modern versions of Bünning's general hypothesis based on the entrainment of circadian oscillations to the light cycle, including ‘external coincidence’, which is a derivation of Bünning's original model, and ‘internal coincidence’, which relied upon seasonal changes in the mutual phase relationship of oscillators within a multi‐oscillator circadian system. This review considers the experimental evidence for the central role of the circadian system in photoperiodic timing and, in some species, for both external and internal coincidence. Pittendrigh, however, pursued the idea of internal coincidence further with his analysis of the pacemaker–slave organization of eclosion rhythmicity in D. pseudoobscura and proposed a similar theoretical model for photoperiodism comprising a group of slave oscillators driven by a light‐sensitive pacemaker. In this model, the phase relationships of the slaves to the pacemaker were affected by (i) the relative periods of the pacemaker and slave(s); (ii) the strength(s) of the coupling between the two; and (iii) the dampening coefficients of the various slaves. Manipulation of these variables showed that the slaves adopted different internal phase relationships (both to each other and to the pacemaker) under the influence of changes in daily photophase, the period of the Zeitgeber and phase shifts of the entraining light cycle.  相似文献

Abstract

We have studied the pattern for resetting the circadian rhythm in the spontaneous motor activity of the crayfish. Spontaneous motor activity was recorded continously at a constant temperature and under free running conditions in complete darkness. The effect of single light pulses applied at different circadian times, on the circadian rhythm of motor activity was measured in both transient stage and steady state. The results led us to construct a phase‐transition curve and phase‐response curve which were analyzed to obtain information about the oscillators which underlie the circadian rhythm of motor activity.  相似文献

Summary Japanese quail have a circadian rhythm of locomotor activity whose free-run period () in constant darkness (DD) was 22.5±0.1 h (45). A phase response curve of typical form was obtained by illuminating the free-running rhythm with single 1 h light pulses. Using entrainment theory a derived phase response curve was calculated from the phase relationships between the locomotor rhythm and 1 h light periods in light-dark cycles of various lengths (T). Although the limits of entrainment to theseT cycles differed slightly from those predicted, there was a close correlation between the two phase response curves. The phase relationships between the locomotor rhythm and 1–9 h photoperiods in 24 h cycles were in general accord with a prediction based on the short free-run period and the relative sizes of the delay and advance portions of the phase response curve for 1 h light pulses.  相似文献

We know that entrainment, a stable phase relationship with an environmental cycle, must be established for a biological clock to function properly. Phase response curves (PRCs), which are plots of phase shifts that result as a function of the phase of a stimulus, have been created to examine the mode of entrainment. In circadian rhythms, single-light pulse PRCs have been obtained by giving a light pulse to various phases of a free-running rhythm under continuous darkness. This successfully explains the entrainment to light-dark cycles. Some organisms show circannual rhythms. In some of these, changes in photoperiod entrain the circannual rhythms. However, no single-pulse PRCs have been created. Here we show the PRC to a long-day pulse superimposed for 4 weeks over constant short days in the circannual pupation rhythm in the varied carpet beetle Anthrenus verbasci. Because the shape of that PRC closely resembles that of the Type 0 PRC with large phase shifts in circadian rhythms, we suggest that an oscillator having a common feature in the phase response with the circadian clock, produces a circannual rhythm.  相似文献

The pineal gland plays a key role in the control of the daily and seasonal rhythms in most vertebrate species. In mammals, rhythmic melatonin (MT) release from the pineal gland is controlled by the suprachiasmatic nucleus via the sympathetic nervous system. In most non‐mammalian species, including birds, the pineal gland contains a self‐sustained circadian oscillator and several input channels to synchronize the clock, including direct light sensitivity. Avian pineal glands maintain rhythmic activity for days under in vitro conditions. Several physical (light, temperature, and magnetic field) and biochemical (Vasoactive intestinal polypeptide (VIP), norepinephrine, PACAP, etc.) input channels, influencing release of melatonin are also functional in vitro, rendering the explanted avian pineal an excellent model to study the circadian biological clock. Using a perifusion system, we here report that the phase of the circadian melatonin rhythm of the explanted chicken pineal gland can be entrained easily to photoperiods whose length approximates 24 h, even if the light period is extremely short, i.e., 3L:21D. When the length of the photoperiod significantly differs from 24 h, the endogenous MT rhythm becomes distorted and does not follow the light‐dark cycle. When explanted chicken pineal fragments were exposed to various drugs targeting specific components of intracellular signal transduction cascades, only those affecting the cAMP‐protein kinase‐A system modified the MT release temporarily without phase‐shifting the rhythm in MT release. Abstract

The aim of this study was to analyse the effect of light respectively illumination cycles on the activity rhythm of the insectivorous bat Myotis myotis (M.m.). A new electronic registration system devised for this purpose can be applied almost universally in recording the activity of terrestrial animals.

M.m. is a strictly dark‐active species with a bimodal activity pattern of bigeminus character. Under constant conditions, M.m. reveals a free running circadian activity rhythm. Its period length varies between 22.6 and 27.8 h and is positively correlated with the intensity of illumination. The range of entrainment of the circadian activity rhythm of M.m. is unusually wide. Accordingly, M.m. resynchronizes very quickly after phase shifts of the Zeitgeber LD. Activity maxima in the range of 10‐4 lx occur in LD 12:12 with constant illumination in L and varied illumination in D as well as in LL with varying intensity.

It is to be discussed whether the wide variability of different parameters in the circadian system of M.m. compared with those of other Chiroptera can be interpreted as an adaptation to ecological factors.  相似文献

Circadian rhythms possess the ability to robustly entrain to the environmental cycles. This ability relies on the phase synchronization of circadian rhythm gene regulation to different environmental cues, of which light is the most obvious and important. The elucidation of the mechanism of circadian entrainment requires an understanding of circadian phase behavior. This article presents two phase analyses of oscillatory systems for infinitesimal and finite perturbations based on isochrons as a phase metric of a limit cycle. The phase response curve of circadian rhythm can be computed from the results of the analyses. The application to a mechanistic Drosophila circadian rhythm model gives experimentally testable hypotheses for the control mechanisms of circadian phase responses and evidence for the role of phase and period modulations in circadian photic entrainment.  相似文献

Abstract

The aim of this study was to explore the individual development of the circadian rhythm of human body temperature. Dailyperiodicchanges of body temperature were examined longitudinally in four infants from 3 to 18 months of age. At the 3rd month of life, the day‐night rhythm of the body temperature was obscure but at the 6th month it became moreevident. From the 12th month on, the circadian temperature rhythm with phase similar to that of the adult was discerned. However, the amplitude in circadian rhythm was significantly larger in children between 6 and 18 months of age than in the adult. These findings suggest that the adult type of circadian rhythm of human body temperature is established during the first year of life with regard to phase but not to amplitude.  相似文献

In an effort to understand the cellular basis of entrainment of circadian oscillators we have studied the role of membrane potential changes in the neurons which comprise the ocular circadian pacemaker of Bulla gouldiana in mediating phase shifts of the ocular circadian rhythm. We report that: 1. Intracellular recording was used to measure directly the effects of the phase shifting agents light, serotonin, and 8-bromo-cAMP on the membrane potential of the basal retinal neurons. We found that light pulses evoke a transient depolarization followed by a smaller sustained depolarization. Application of serotonin produced a biphasic response; a transient depolarization followed by a sustained hyperpolarization. Application of a membrane permeable analog of the intracellular second messenger cAMP, 8-bromo-cAMP, elicited sustained hyperpolarization, and occasionally a weak phasic depolarization. 2. Changing the membrane potential of the basal retinal neurons directly and selectively with intracellularly injected current phase shifts the ocular circadian rhythm. Both depolarizing and hyperpolarizing current can shift the phase of the circadian oscillator. Depolarizing current mimics the phase shifting action of light, while hyperpolarizing current produces phase shifts which are transposed approximately 180 degrees in circadian time to depolarization. 3. Altering BRN membrane potential with ionic treatments, depolarizing with elevated K+ seawater or hyperpolarizing with lowered Na+ seawater, produces phase shifts similar to current injection. 4. The light-induced depolarization of the basal retinal neurons is necessary for phase shifts by light. Suppressing the light-induced depolarization with injected current inhibits light-induced phase shifts. 5. The ability of membrane potential changes to shift oscillator phase is dependent on extracellular calcium. Reducing extracellular free Ca++ from 10 mM to 1.3 X 10(-7) M inhibits light-induced phase shifts without blocking the photic response of the BRNs. The results indicate that changes in the membrane potential of the pacemaker neurons play a critical role in phase shifting the circadian rhythm, and imply that a voltage-dependent and calcium-dependent process, possibly Ca++ influx, shifts oscillator phase in response to light.  相似文献

Visible light synchronizes the human biological clock in the suprachiasmatic nuclei of the hypothalamus to the solar 24‐hour cycle. Short wavelengths, perceived as blue color, are the strongest synchronizing agent for the circadian system that keeps most biological and psychological rhythms internally synchronized. Circadian rhythm is important for optimum function of organisms and circadian sleep–wake disruptions or chronic misalignment often may lead to psychiatric and neurodegenerative illness. The beneficial effect on circadian synchronization, sleep quality, mood, and cognitive performance depends not only on the light spectral composition but also on the timing of exposure and its intensity. Exposure to blue light during the day is important to suppress melatonin secretion, the hormone that is produced by the pineal gland and plays crucial role in circadian rhythm entrainment. While the exposure to blue is important for keeping organism's wellbeing, alertness, and cognitive performance during the day, chronic exposure to low‐intensity blue light directly before bedtime, may have serious implications on sleep quality, circadian phase and cycle durations. This rises inevitably the need for solutions to improve wellbeing, alertness, and cognitive performance in today's modern society where exposure to blue light emitting devices is ever increasing.   相似文献

Abstract

The effect of continuous infusion of NG‐methylarginine, an inhibitor of nitric oxide synthase, into the third cerebral ventricle above the suprachiasmatic nucleus (SCN) of the hypothalamus on the circadian rhythm of water intake was examined in rats maintained under either a 12‐h light and 12‐h dark cycle or in constant darkness. NG‐methylarginine disrupted the circadian rhythm in both conditions. In constant darkness, however, the effect of the inhibitor on the rhythm was found to be due to change in its phase. These findings suggest that nitric oxide is involved in the mechanism of the generation and/or synchronization of the circadian rhythm driven by the circadian oscillator in the SCN.  相似文献

An attempt was made to identify the neurophysiological processes involved in entrainment of the circadian rhythm of spontaneous optic nerve potentials from the Aplysia eye by determining whether pharmacological agents or ion substitutions could block phase shifts produced by single light pulses. Knowing which physiological processes are involved in entrainment should help define the morphological pathway traveled by entrainment information. A secretory step does not appear to be involved in the flow of entrainment information from the environment to the circadian oscillator. A treatment (HiMg LoCa) capable of inhibiting secretion did not interfere with phase shifting by light. Furthermore, treating eyes with putative transmitters or extracts of eyes did not phase shift the free running rhythm. Also, the phase shifting information is not translated into action potentials before reaching the oscillator since TTX–HiMg LoCa solutions did not block the light-induced phase shift. The photoreceptor potential does seem to be important for light-induced phase shifts. A correlation was found between the effects of treatments on the ERG and their effects on the light-induced phase shift. Solutions which decreased the ERG by 90% or more blocked phase shifting whereas solutions which decreased the ERG by less than 74% had no effect on phase shifting by light. The results from these studies are consistent with two pathways for the flow of phase shifting information to the circadian oscillator. The circadian oscillator may be associated with receptor cells and the entrainment pathway would include a step involving the photoreceptor potential. Alternatively, the circadian oscillator may be associated with secondary cells and receive entrainment information via the photoreceptor potential and passive spread of current through a gap junction. Higher order cells than second-order ones are probably not involved in the entrainment pathway.  相似文献

Abstract

The mammalian circadian pacemaker can be phase shifted by photic, pharmacological, and behaviorally‐derived stimuli. The phase‐response curves (PRCs) characterizing these diverse stimuli may comprise two distinct families; a photic PRC typified by the response to brief light pulses, and a non‐photic PRC, typified by the response to dark pulses and to behavioral activation. The present study examined the phase shifting effects of acute systemic treatment with the alpha2‐adrenoceptor agonist, clonidine, in Syrian hamsters. Clonidine injections (0.25 mg/kg, ip) delivered during subjective night mimicked the phase shifting effects of light pulses in animals housed in both constant darkness (DD) and constant red light (RR), but similar effects were not seen in saline‐treated controls. Both clonidine and saline injections resulted in phase advances during subjective day, but only in RR‐housed animals. Clonidine‐induced phase shifting was dose‐dependent, but rather high doses were required to induce phase shifts. Pretreatment with the selective noradrenergic neurotoxin, DSP‐4, blocked clonidine‐induced phase shifting. These results suggest that clonidine acts at presynaptic alpha2‐adrenergic autoreceptors to disinhibit spontaneous and/or evoked activity in the photic entrainment pathway.  相似文献

ABSTRACT

Melatonin, an essential pineal hormone, acts as a marker of the circadian clock that regulates biological rhythms in animals. The effects of exogenous melatonin on the circadian system of nocturnal rodents have been extensively studied; however, there is a paucity of studies on the phase-resetting characteristics of melatonin in diurnal rodents. We studied the phase shifting effects of exogenous melatonin as a single melatonin injection (1 mg/kg) at various phases of the circadian cycle on the circadian locomotor activity rhythm in the palm squirrel, Funambulus pennantii. A phase response curve (PRC) was constructed. Adult male squirrels (N = 10) were entrained to a 12:12 h light-dark cycle (LD) in a climate-controlled chronocubicle with food and water provided ad libitum. After stable entrainment, squirrels were transferred to constant dark condition (DD) for free-running. Following stable free run, animals were administered a single dose of melatonin (1 mg/kg in 2% ethanol-phosphate buffered saline (PBS) solution) or vehicle (2% ethanol-PBS solution) at circadian times (CTs) 3 h apart to evoke phase shifts. The phase shifts elicited at various CTs were plotted to generate the PRC. A dose response curve was generated using four doses (0.5, 1, 2 and 4 mg/kg) administered at the CT of maximum phase advance. Melatonin evoked maximum phase advances at CT0 (1.23 ± 0.28 h) and maximum phase delays at CT15 (0.31 ± 0.09 h). In the dose response experiment, maximal phase shifts were evoked with 1 mg/kg. In contrast, no significant shifts were observed in control groups. Our study demonstrates that the precise timing and appropriate dose of melatonin administration is essential to maximize the amelioration of circadian rhythm–related disorders in a diurnal model.  相似文献

Obesity is increasing in industrialized countries at an alarming rate. Recent studies have linked this condition with changes in the circadian regulation, and circadian clock dysfunctions have also been linked to metabolic disorders. When in captivity and fed a regular rodent chow diet ad libitum some volcano mice, Neotomodon alstoni (endemic species of Mexico) become overweight and display symptoms equivalent to metabolic syndrome. The aim of this work was to observe whether there are significant changes in the functional properties of the circadian system, namely in the entraining circadian locomotor activity rhythm, between mice that became obese and normal adult mice. Freely moving circadian rhythms of locomotor activity were tested under constant conditions as well as under conditions of discrete and continuous entrainment. Our results show that volcano lean mice present a phase response curve with larger delays than advances, indicating that re-entrainment is more easily achieved by delays. Volcano obese mice are less efficient at re-entraining because they show smaller phase shifts in delays than control mice. These results indicate that obesity in N. alstoni has a negative effect on the circadian mechanisms that integrate the photic entrainment.  相似文献

Summary Photoperiod plays an important role in controlling the annual reproductive cycle of the male lizard Anolis carolinensis. The nature of photoperiodic time measurement in Anolis was investigated by exposing anoles to 3 different kinds of lighting paradigms (resonance, T cycles, and night breaks) to determine if photoperiodic time measurement involves the circadian system. Both the reproductive response and the patterns of entrainment of the activity rhythm were assessed. The results show that the circadian system is involved in photoperiodic time measurement in this species and that a discrete photoinducible phase resides in the latter half of the animals' subjective night. Significantly, the ability of the circadian system to execute photoperiodic time measurement is crucially dependent on the length of the photoperiod. Resonance, T cycle and night break cycles utilizing a photoperiod 10–11 h in duration reveal circadian involvement whereas these same cycles utilizing 6 or 8 h photoperiods do not.Abbreviation CRPP circadian rhythm of photoperiodic sensitivity  相似文献
