A simple and novel procedure for the acceleration of fungal spore production was developed. A net of hydrophobic polymer such as polypropylene (PP) and polytetrafluoroethylene (PTFE) was embedded in a nutrient agar plate, and effect of the polymer net on spore production by 6 fungal strains, such as Aspergillus terreus, Penicillium multicolor, and Trichoderma virens were estimated. The effect of hydrophobic polymer net was insufficient in a liquid-surface immobilization (LSI) system with fungal cells immobilized on a ballooned microsphere layer formed on a liquid medium surface. On the other hand, the embedding of a PTFE net in an agar plate remarkably enhanced the spore production in all 6 strains tested to produce 2.0–8.5 × 107 spores/cm2-agar plate surface. Especially, the spore production by A. terreus ATCC 20542 in the presence of a PTFE net was 7.7 times as much than that in no net. Positive correlations between the hydrophobicity of net and the spore production were observed in all 6 strains (R2, 0.653–0.999).  相似文献

研究宿主植物栽培容器对丛枝菌根(Arbuscularmycorrhizae,AM)真菌Glomusmosseae生长发育的影响。结果表明:小容积容器的根系密度相对较大,在菌根共生体建立初期,菌根真菌繁殖体与根接触的机会增大,对于菌根真菌的迅速侵染及共生体的迅速建立非常有利,同时还增大了根外菌丝二次侵染的机会,从而使菌根真菌生长发育形成了一个良性循环,最终有利于根外孢子的形成。容器对共生体的影响决不是简单的盆的体积问题,而与其面积和体积之比有关,也和种植密度有密切关系。  相似文献

Fungi are a megadiverse group of organisms, they play major roles in ecosystem functioning and are important for human health, food production and nature conservation. Our knowledge on fungal diversity and fungal ecology is however still very limited, in part because surveying and identifying fungi is time demanding and requires expert knowledge. We present a method that allows anyone to generate a list of fungal species likely to occur in a region of interest, with minimal effort and without requiring taxonomical expertise. The method consists of using a cyclone sampler to acquire fungal spores directly from the air to an Eppendorf tube, and applying DNA barcoding with probabilistic species identification to generate a list of species from the sample. We tested the feasibility of the method by acquiring replicate air samples from different geographical regions within Finland. Our results show that air sampling is adequate for regional‐level surveys, with samples collected >100 km apart varying but samples collected <10 km apart not varying in their species composition. The data show marked phenology, and thus obtaining a representative species list requires aerial sampling that covers the entire fruiting season. In sum, aerial sampling combined with probabilistic molecular species identification offers a highly effective method for generating a species list of air‐dispersing fungi. The method presented here has the potential to revolutionize fungal surveys, as it provides a highly cost‐efficient way to include fungi as a part of large‐scale biodiversity assessments and monitoring programs.  相似文献

林火是森林生态系统的一种主要干扰因子,以青岛市三标山林火迹地为研究对象,采集荆条(Vitex negundo)、胡枝子(Lespedeza bicolor)、花木蓝(Indigofera kirilowii)、青花椒(Zanthoxylum schinifolium)和野青茅(Deyeuxia arundinacea)5种优势植物根围土壤,研究不同林火强度对丛枝菌根(AM)真菌多样性的影响。结果表明,AM真菌侵染率和孢子密度随火灾强度的加强而降低;非过火区植物根围土壤中,分离鉴定出AM真菌3属11种,轻度过火区分离鉴定出AM真菌3属10种,中度过火区分离鉴定出AM真菌3属9种,重度过火区分离鉴定出AM真菌3属8种。过火区AM真菌种丰度低于非过火区。过火区和非过火区AM真菌的重要值和优势种不同,非过火区植物根围的优势种是地球囊霉(Glomus geosporum)、台湾球囊霉(G.taiwanensis)、分支巨孢囊霉(Gigaspora ramisporophora)、极大巨孢囊霉(Gi.gigantean)、福摩萨球囊霉(G.formosanum)、悬钩子球囊霉(G.rubiforme)、柯氏无梗囊霉(Acaulospora koskei)和松蜜无梗囊霉(A.thomii);轻度过火区植物根围的优势种是地球囊霉和台湾球囊霉;中度过火区的是台湾球囊霉和地球囊霉(野青茅除外);重度过火区植物根围的优势种是地球囊霉。不同强度的过火区对AM真菌群落组成有不同程度的影响。认为林火降低植物根围土壤中AM真菌多样性。  相似文献

Mass production of sporangiospores (spores) of Rhizopus oryzae NBRC 5384 (identical to NRRL 395 and ATCC 9363) on potato‐dextrose‐agar medium was studied aiming at starting its L (+)‐lactic acid fermentation directly from spore inoculation. Various parameters including harvest time, sowed spore density, size of agar plate, height of air space, and incubation mode of plate (agar‐on‐bottom or agar‐on‐top) were studied. Ordinarily used shallow Petri dishes were found out to be unsuitable for the full growth of R. oryzae sporangiophores. In a very wide range of the sowed spore density, the smaller it was, the greater the number of the harvested spores was. It was also interesting to find out that R. oryzae grown downward vertically with a deep air space in an agar‐on‐top mode gave larger amount of spores than in an agar‐on‐bottom mode at 30°C for 7‐day cultivation. Scale‐up of the agar plate culture from 26.4 to 292 cm2 was studied, resulting in the proportional relationship between the number of the harvested spores/plate and the plate area in the deep Petri dishes. The number of plates of 50 cm in diameter needed for 100 m3 industrial submerged fermentation started directly from 2 × 105 spores/mL inoculum size was estimated as about 6, from which it was inferred that such a fermentation would be feasible. Designing a 50 cm plate and a method of spreading and collecting the spores were suggested. Bioprocess technological significance of the “full‐scale industrial submerged fermentation started directly from spore inoculation omitting pre‐culture” has been discussed. © 2013 American Institute of Chemical Engineers Biotechnol. Revegetation following dam removal projects may depend on recovery of arbuscular mycorrhizal (AM) and ectomycorrhizal (EM) fungal communities, which perform valuable ecosystem functions. This study assessed the availability and function of AM and EM fungi for plants colonizing dewatered reservoirs following a dam removal project on the Elwha River, Olympic Peninsula, Washington, United States. Availability was assessed via AM fungal spore density in soils and EM root tip colonization of Salix sitchensis (Sitka willow) in an observational field study. The effect of mycorrhizal fungi from 4 sources (reservoir soils, commercial inoculum, and 2 mature plant community soils) on growth and nutrient status of S. sitchensis was quantified in a greenhouse study. AM fungal spores and EM root tips were present in all field samples. In the greenhouse, plants receiving reservoir soil inoculum had only incipient mantle formation, while plants receiving inoculum from mature plant communities had fully formed EM root tips. EM formation corresponded with alleviation of phosphorus stress in plants (lower shoot nitrogen:phosphorus). Thus, revegetating plants have access to AM and EM fungi following dam removal, and EM formation may be especially important for plant P uptake in reservoir soils. However, availability of mycorrhizal fungi declines with distance from established plant communities. Furthermore, EM fungal communities in recently dewatered reservoirs may not be as effective at forming beneficial mycorrhizae as those from mature plant communities. Whole soil inoculum from mature plant communities may be important for the success of revegetating plants and recovery of mycorrhizal fungal communities.  相似文献

Porcine pancreatic lipase immobilized on celite particles has been employed as a catalyst for the esterification of dodecanol and decanoic acid in a predominantly organic system. Solvent influence on the equilibrium position and on the catalyst activity has been studied using 20 solvents, including aliphatic and aromatic hydrocarbons, ethers, ketones, nitro- and halogenated hydrocarbons, and esters. The equilibrium constant for esterification correlates well with the solubility of water in the organic solvent, which in turn shows a good relationship with a function of Guttman's donor number and the electron pair acceptance index number of the solvent. This may be rationalized in terms of the requirements for solvation of water and of the reactants. The catalyst activity, measured as the initial rate of the esterification reaction, is best correlated as a function of both n-octanol-water partition coefficient (log P) and either the electron pair acceptance index or the polarizability.  相似文献

目的探讨真菌荧光法在皮肤浅部真菌筛查的敏感性。方法收集皮肤科门诊疑似真菌感染的患者标本,同时用KOH湿片法和真菌荧光染色法进行检测,并对检验结果进行对比。结果 1 209例患者中真菌荧光染色法阳性的阳性率为42.76%,KOH湿片法的阳性率为24.81%,结果差异有统计学意义(χ^2=87.094,P<0.05)。荧光染色法和KOH法在患者不同部位的阳性率:头面部为55.59%、7.90%,体股部为46.93%、30.06%,手足部为27.37%、26.25%,外阴部为31.85%、14.07%,指甲(趾甲)部为51.64%、48.36%;在不同疾病中的阳性率:马拉色毛囊炎为77.00%、8.50%,手足癣为48.04%、46.08%,体癣为50.31%、33.44%,甲癣为51.64%、48.36%,花斑癣为66.67%、48.00%,湿疹为10.24%、5.42%,皮炎为48.11%、9.43%。结论荧光染色法是一种简单、快速、准确的真菌镜检方法,值得在临床上推广使用。其总体的阳性率比KOH湿片法高,毛囊虫、疥疮等真菌荧光染色法在镜下更容易辨认。头面部、外阴等油脂分泌旺盛的部位以及马拉色毛囊炎、湿疹和皮炎等疾病推荐使用荧光染色法来排除是否有真菌的感染。  相似文献

The effect of organic solvents on the equilibrium position of lipase-catalyzed esterification of glycerol and decanoic acid has been investigated. The reaction is carried out in an aqueous-organic two-phase system. In polar solvents, high mole fractions of monoacylglycerol and low mole fractions of triacylglycerol and measured, while in nonpolar solvents, the measured differences in the mole fractions of monodi-, and triacylglycerols are less. There is a good correlation between the ester mole fractions at equilibrium and the log P of the solvent (partition coefficient in n-octanolwater), however, only if the group of tertiary alcohols is excluded. In the plot of the easter mole fractions as a function of the logarithm of hte solubility of water in the organic solvent, the tertiary alcohols can be included; however, in this case other deviations appear.For the prediction of the effect of organic solvents on the ester mole fractions at reaction equilibrium in nondilute reaction systems with a water activity below 1, the program TREP (Two-phase Reaction Equilibrium Prediction) is developed, which is based on the UNIFAC group contribution method. With this model the equilibrium data are essentially predicted from basic thermodynamic data. The required equilibrium constants are estimated from experiments without an organic solvent in the reaction medium. The mole fractions calculated by TREP show the same trends as the experimentally measured mole fractions; however, some variation is observed in the absolute values. These deviations may be due to inaccuracies in the UNIFAC group contribution method. TREP is found to be a correct method to predict within some limits the ester mole fractions at equilibrium for all mixtures of solvents, substrates, and products. The production of monoester can be enhanced in reaction system with a sufficient high concentration of a polar solvent. Mobility represents a key innovation in the evolution of complex animal life. The ability to move allows for the exploration of new food sources, escapes from unfavorable environmental conditions, enhanced ability to exchange genetic material, and is one of the major reasons for the diversity and success of animal life today. The oldest widely accepted trace fossils of animal mobility are found in Ediacaran‐aged rocks (635–539 Ma). The earliest definitive evidence for movement associated with exploitation of resources for feeding occurs in the White Sea assemblage of the Ediacara Biota—macroscopic, soft‐bodied fossils of Ediacaran age. Here, we evaluate potential support for mobility in dickinsoniomorphs, presenting new data regarding abundant Dickinsonia and associated trace fossils from the Ediacara Member, South Australia. Results quantitatively demonstrate that Dickinsonia was capable of mobility on relatively short, ecological timescales. This organism was bilaterally symmetrical, likely moved via muscular peristalsis, and left trace fossils due to active removal of the organic mat related to feeding. Analogous structures associated with Yorgia indicate that it was also mobile and fed in a similar manner. Morphological evidence suggests that two other modular taxa, Andiva and Spriggina, were able to move but did not feed in a manner that impacted the organic mat. Together, these data suggest that mobility was present in multiple disparate bilaterally symmetrical Ediacaran taxa.  相似文献

A new method allowing control of rhizosphere pH and mineral nutrition was applied to study depletion of various organic and inorganic phosphorus fractions extractable sequentially with 0.5M KHCO3 (pH 8.5), 0.1M NaOH and residual P extractable with 6M H2SO4 from the rhizosphere soil.Soil pH was affected about 2 mm from the root mat. Depletion zones of inorganic P (KHCO3-Pi) extractable with 0.5M KHCO3 extended up to about 4 mm but the depletion zones of all other P fractions were about 1 mm only. The root-induced decrease of soil pH from 6.7 to 5.5 increased the depletion of total P from all fractions by 20% and depletion of KHCO3-Pi and residual P by 34% and 43%, respectively. Depletion of organic P (KHCO3-Po) extractable with 0.5M KHCO3 was not affected by a change in rhizosphere pH. With constant or increased pH, depletion of inorganic P (NaOH-Pi) was 17% and organic P (NaOH-Po) was 22% higher than with decreased pH. Only 54–60% of total P withdrawn from all fractions was from KHCO3-Pi. Substantial amounts of KHCO3-Po and NaOH-Po were mineralized and withdrawn from the rhizosphere within 1 mm from the root mat, as 11–15% of total P withdrawn originated from the organic P fractions. A remaining 11–16% was derived from NaOH-Pi, and 15–18% from residual P fractions likely to be rather immobile. Thus, 40–46% of the P withdrawn near the root mat of rape originated from non-mobile P fractions normally not included in 0.5M NaHCO3 extraction used to obtain an index of plant-available soil P.  相似文献

The effect of solvent on the activity of mushroom tyrosinase toward three substrates was studied at a constant water activity of either 0.74 or 0.86. No simple correlation was observed between enzyme activity and log P, but partition coefficients of substrate (P(s)) and product (P(p)) gave systematic relations with enzyme activity. When initial reaction rates were considered, there was a bellshaped relationship between enzyme activity and P(s) with an optimal P(s) for each substrate. This can be explained by assuming that the solvent affected the enzyme activity primarily by affecting the substrate concentration in the aqueous layer around the catalyst where the enzymic reaction occurs. When long-term reaction rates were considered, a high P(p)/P(s) ratio was consistent with preservation of enzyme activity. (c) 1994 John Wiley & Sons, Inc.  相似文献

The viability of mycelial fragments of Trametes versicolor and Irpex lacteus and their growth on selected hydrogels are described. The size of mycelial fragments of the fungi did not significantly influence their viability. Alginate hydrogel films supported fungal growth better than agarose, carrageenan, chitosan and gelatin films, and had the highest mechanical strength but were less hydrophilic than the other hydrogels. All commercial alginates that were tested supported aseptic growth of fungal fragments without prior sterilization of the hydrogel solution. The viability of mycelial fragments in the hydrogel solutions was higher for some commercial alginates than that in laboratory grade alginate. The mechanical strength and hydrophilicity of hydrogels from alginate type Sobalg FD 155 and Meer HV were comparable to that of laboratory grade alginate. Sterilization and pH of the alginate hydrogel did not significantly influence the growth of T. versicolor mycelial fragments but affected the growth of I. lacteus. Concentrations of alginate in the range of 1–2% in the hydrogel did not affect the growth of entrapped mycelial fragments of these fungi. Aims:  To maximize spore production by Bacillus amyloliquefaciens B128, and its antagonism to the fungal pathogen Botrytis elliptica B061. Methods and Results:  In the 5-l stirred-tank bioreactor (STR), with the 0·5 vvm aeration rate, an agitation rate of 200 rev min−1 significantly enhanced the spore yield compared to the same in 300 rev min−1 cultivations. In a 20-l airlift bioreactor (ALR) the maximal spore production was further increased with a controlled aeration rate of 2·5 vvm operated in a 24-mesh net-draft tube mode, and no pH control cultivation. This spore yield in the 20-l ALR was five- and eightfold higher; in addition the cultivation period was 19 h shorter, compared to that obtained from shaker flask and in the 5-l STR cultivations respectively. Conclusions:  Although culture conditions are still to be optimized, by using an ALR with net-draft tube, a scaling up from shaker flasks and STR to ALR of spore production by the strain B128 is technically feasible. Significance and Impact of the Study:  The spore yields obtained using bioreactors were much higher than those previously reported. The freshly produced spore preparations from the B128 strain significantly antagonized the grey mould pathogen B. elliptica.  相似文献

Cross-talk of subchondral bone and articular cartilage could be an important aspect in the etiology of osteoarthritis. Previous research has provided some evidence of transport of small molecules (~370 Da) through the calcified cartilage and subchondral bone plate in murine osteoarthritis models. The current study, for the first time, uses a neutral diffusing computed tomography (CT) contrast agent (iodixanol, ~1550 Da) to study the permeability of the osteochondral interface in equine and human samples. Sequential CT monitoring of diffusion after injecting a finite amount of contrast agent solution onto the cartilage surface using a micro-CT showed penetration of the contrast molecules across the cartilage-bone interface. Moreover, diffusion through the cartilage-bone interface was affected by thickness and porosity of the subchondral bone as well as the cartilage thickness in both human and equine samples. Our results revealed that porosity of the subchondral plate contributed more strongly to the diffusion across osteochondral interface compared to other morphological parameters in healthy equine samples. However, thickness of the subchondral plate contributed more strongly to the diffusion in slightly osteoarthritic human samples.  相似文献

评估极端干旱对温带荒漠土壤真菌群落的影响有助于进一步认识干旱导致的区域荒漠化特征。本研究利用在古尔班通古特沙漠建立的干旱三年和干旱十年样地, 分析了长期极端干旱对温带荒漠土壤真菌群落和生态网络的影响。结果显示, 干旱三年与干旱十年处理对总真菌和丰富真菌的Chao1和Shannon多样性指数均无显著性影响, 而对稀有真菌的Shannon多样性指数有显著促进作用; 干旱三年和干旱十年处理显著影响总真菌、丰富和稀有真菌的群落组成, 且极端干旱对稀有真菌群落变异的影响(ANOSIM, R = 0.378-0.595, P < 0.01)大于对丰富真菌的影响(ANOSIM, R = 0.282-0.555, P < 0.01), 表明丰富真菌具有更强的干旱抵抗力; 另外, 极端干旱显著降低了总真菌、丰富和稀有真菌的β多样性, 表明极端干旱具有生态过滤作用。分子生态网络结果显示, 干旱三年与干旱十年处理降低了荒漠土壤真菌群落网络复杂性, 表明极端干旱减弱了真菌物种间的相互作用; 相比稀有真菌, 丰富真菌具有更高的节点拓扑参数(P < 0.05), 表明丰富真菌对维持极端干旱下的真菌物种间相互作用的重要性。综上所述, 极端干旱显著改变了荒漠表层土壤真菌群落组成, 减弱了真菌物种间的相互作用; 稀有真菌敏感响应极端干旱, 有利于减缓荒漠土壤真菌群落响应的滞后性; 丰富真菌作为网络的核心菌群, 对维持极端干旱下的真菌群落稳定性以及物种间的相互作用很关键。  相似文献
