Induced rifampicin-resistant mutants of Frankia were isolated by treatment of spores from strain ChI1 with 2 mg/ml of nitrosoguanidine. The mutagenic treatment was followed by growth for 72 h on non-selective medium to allow the expression of the mutation, before plating on selective medium. Spontaneous rifampicin-resistant mutants were isolated from strain ReI4 and chloramphenicol-resistant mutants were derived from strains ReI6 and TtI42. The mutants grew in medium containing up to 20 g/ml of antibiotics. They showed similar morphology and growth pattern compared with their parent strains. However, they exhibited differences in symbiotic properties, such as infectivity and nitrogenase activity, from their parent strains.  相似文献

Frankia strain BCU110501 was isolated from root nodules of the native Patagonian actinorhizal plant Discaria trinervis. The strain was grown on BAP medium with sodium propionate or glucose as carbon sources. Colonies grown in nitrogen-free medium showed branched hyphae bearing polymorphic sporangia and vesicles, which were capable of nitrogen fixation. Old cultures produced a red pigment. The infectivity and effectivity of a Frankia strain isolated from Discaria on its own host, D. trinervis and also in D. chacaye, is reported for the first time. Frankia BCU110501 has physiological properties that are intermediate between categories proposed by Lechevalier et al. (1983) to classify Frankia.  相似文献

 We studied the reproductive behavior of Colletia spinosissima Gmel. at six sites in two regions of Argentina during the 1998–2000 flowering seasons. Flowering takes place in winter. The flowers are homogamous, fragrant, and entomophilous. There was evidence of self-incompatibility and cross-incompatibility between nearby partners. Fruit set of open-pollinated flowers (13.3%) seemed pollen-limited, since low fecundity was consistently associated with small stigmatic pollen loads and with low numbers of successful pollen tubes, and fecundity increased with abundance of efficient pollinators. Reproductive success (percent ovules becoming viable seeds) was 3.6. It is hypothesized that homogamy was established in Colletia because in an impoverished pollination environment – such as the one likely prevailing when this lineage evolved – achieving pollen export and receipt in a single pollinator visit has selective advantages. A large stigma and an extragynoecial compitum would compensate for the homogamy-driven, increased pollen-stigma interference. Received March 8, 2001 Accepted July 19, 2001  相似文献

The time course of initiation and development of root nodules was investigated in the South American actinorhizal shrub Discaria trinervis (Rhamnaceae). A local strain of Frankia (BCU110501) which was isolated from D. trinervis nodules, was used as inoculum. Inoculated seedlings were periodically studied under the light microscope after clearing with aqueous NaClO. In parallel, semithin and ultrathin sections were analysed by light and electron microscopy. Infection by Frankia BCU110501 involved intercellular penetration among epidermal and cortical root cells. Nodule primordia were detected from 6 d after inoculation, while bacteria were progressing through intercellular spaces of the outer layers of cortical cells. Invasion of host cells by the symbiont occurred 7–9 d after inoculation, and hypertrophy of the primordium cells was associated with Frankia penetration. Root hairs were not deformed during the early events of nodule formation. From 13 to 16 d after inoculation, the proximal cellular zone of the primordia behaved differently from the other tissues after NaClO treatment and remained darkly pigmented. At the same time, differentiation of Frankia vesicles started to occur inside already infected cells. By 16 d after inoculation, spherical vesicles of BCU110501 were homogeneously distributed in the host cells. These vesicles were septate and surrounded by void space. Frankia spores or sporangia were not observed in the nodule tissue. This study has clarified the mode of Frankia penetration in D. trinervis , one of the Rhamnaceae which also includes Ceanothus . The events involved in infection, nodule induction, host-cell infection and vesicle differentiation have been characterized and identified as time-segregated developmental processes in the ontogeny of D. trinervis root nodules.  相似文献

应用原核生物16SrDNA特异性引物rD1和fD1,通过ARDRA(amplified ribosomal DNA restriction analysis)法直接扩增自中国云南、东北地区赤杨属3种植物和沙棘属1种植物根瘤内FrankiaDNA,得到一长约1500bp的扩增产物,选用两种内切酶HaeⅢ、AfaⅠ联合对扩增产物进行酶切,得到稳定的酶切图谱,将所测48个感染赤杨的Frankia样本区分为3个不同的组,所测43个感染沙棘的Frankia样本区分为3个不同的组,显示根瘤内Frankia存在丰富的遗传多样性。  相似文献

Edhazardia aedis is a polymorphic microsporidium of mosquitoes that is both horizontally and vertically transmitted to its host. Because it is being developed for biological control of mosquitoes, detailed knowledge is needed regarding the longevity and germation of its fragile, mosquito‐infectious spore. Spores responsible for horizontal transmission were extracted from 7–10‐day‐old larvae (reared from infected Aedes aegypti eggs) and purified by Ludox density gradient centrifugation. Mature spores were variable in specific gravity, being found throughout the 20 and 60% zone in Ludox gradients. The optimal environment for spore germination was dilute KCl at pH 10.0–11.0; ammonia inhibited germination. Osmotic inhibition was almost complete in both sucrose and polyethylene glycol at concentrations equivalent to 40 atm. The spores retained their viability for a maximum of 21 days at 23±2°C, whereas when held at 5±2°C, their viability was completely lost within two days post‐harvest. Potential for germination decreased along with infectivity, providing a simple assay for spore viability.  相似文献

Nine nativeFrankia strains were isolated from root nodules of four chilean actinorhizal plants (Rhamnaceae). The strains were designated as ChI1, ChI2, ChI3 and ChI4 fromColletia hystrix; ReI4 and ReI6 fromRetanilla ephedra; TqI12 and TqI15 fromTalguenea quinquinervis and TtI42 fromTrevoa trinervis. By scanning electron microscopy, all the strains exhibited similar actinomycetal structures: hyphae, sporangia and vesicles. The growth patterns of the isolates in BAP medium were similar. All showed a lag phase of approximately 6–7 days, then exhibited a logarithmic phase, except the ReI4 strain which seems to follow a linear growth pattern. A common feature of all the strains was a rapid loss of biomass at the end of the growth phase. All native strains grew on BAP medium supplemented with glucose. In six out of nine strains, the glucose was the best of the carbon sources tested. However, the strains differed in their ability to use other carbon sources such as arabinose, mannitol, maltose, succinate, sucrose, pyruvate, propionate and galactose. The isolates were sensitive to six antibiotics assayed (ampicillin, penicillin G, rifampicin, chloramphenicol, erythromycin and kanamycin). Using the acetylene reduction assay, the nitrogenase activity of the strains was determined. All strains grown in BAP medium lacking a combined nitrogen source were able to reduce acetylene ‘in vitro’.  相似文献

Abstract Seven Frankia strains from Casuarina and Allocasuarina were analyzed by polyacrylamide gel electrophoresis to determine the patterns of several enzymes. No relatedness could be established between the strains as far as polyphenol oxidase, esterase and diaphorase were concerned. A first attempt at grouping the Frankia isolates could be achieved with catalase. It was confirmed by the study of superoxide dismutase: only one activity band, with the same mobility, was found in all cases. We propose to use this enzyme as a marker for identification of Frankia strains issued from Casuarinaceae.  相似文献

Summary The genetic diversity of microsymbiont Frankia from Colletia hystrix (Clos.) plants growing in a restricted area, were investigated by PCR-restriction fragment length polymorphism (RFLP) technique. DNA from field-collected nodules was amplified by PCR with primers targeting two genomic regions; one included a large portion of the 3′ end of the 16S rRNA gene, the intergenic spacer, and the 5′ end of the 23S rRNA gene and the other in the nifD–nifK intergenic region in the nif operon as a means to estimate molecular diversity. A HaeIII digestion of the PCR product allowed us to identify PCR-RFLP groups or haplotypes among the Colletia-infective Frankia strains tested. An exhaustive small-scale sampling permitted us to detect haplotypes with a low frequency in the microsymbiont population and showed that Frankia microsymbionts have a higher genetic diversity than previously reported. Fifteen haplotypes were recognized on the basis of combining the restriction patterns in each region analyzed. The haplotype designated as A3 was found with a high frequency in the five microsymbiont Frankia groups studied indicating a dominant haplotype. This haplotype was also exhibited by strain ChI4, which was isolated in 1991 in the same locality suggesting that it is the most common haplotype in this area and very stable over time.  相似文献

以经过3年低温储藏的粗茎鳞毛蕨孢子为实验材料,从孢子离心、孢子消毒、培养基种类、光质等4方面对孢子萌发进行研究,结果表明:在离心转数≤14 000 r.min-1、离心时间≤30 min条件下,离心处理对孢子萌发基本无影响;对孢子进行1%NaClO水溶液浸泡处理20~30 min为最佳消毒条件;改良Knop’s培养基为最佳孢子萌发培养基;黑暗条件下孢子不能萌发,但是黑暗处理能够明显提高孢子萌发整齐性;红光比白光能促进孢子提早萌发1 d左右,但对提高萌发率效果不显著。  相似文献

AIMS: To use bovicin HC5 to inhibit predominant bacteria isolated from spoiled mango pulp. METHODS AND RESULTS: Bovicin HC5 and nisin were added to brain heart infusion (BHI) medium (40-160 AU ml(-1)) or mango pulp (100 AU ml(-1)) and the growth of Bacillus cereus and Bacillus thuringiensis was monitored. Cultures treated with bovicin HC5 or nisin showed longer lag phases and grew slower in BHI medium. Bovicin HC5 and nisin were bactericidal and showed higher activity in mango pulp at acidic pH values. To determine the effect on spore germination and D values, mango pulp containing bovicin HC5 was inoculated with 10(6) and 10(9) spores per ml(-1), respectively, from each strain tested. Bovicin HC5 reduced the outgrowth of spores from B. cereus and B. thuringiensis, but thermal sensitivity was not affected. CONCLUSIONS: Bovicin HC5 was bactericidal against B. cereus and B. thuringiensis isolated from spoiled mango pulp. SIGNIFICANCE AND IMPACT OF THE STUDY: Bacillus cereus and B. thuringiensis had not been previously isolated from spoiled mango pulp and bovicin HC5 has the potential to inhibit such bacteria in fruit pulps.  相似文献

BACKGROUND AND AIMS: Although allopolyploidy is a prevalent speciation mechanism in plants, its adaptive consequences are poorly understood. In addition, the effects of allopolyploidy per se (i.e. hybridization and chromosome doubling) can be confounded with those of subsequent evolutionary divergence between allopolyploids and related diploids. This report assesses whether fern species with the same ploidy level or the same altitudinal distribution have similar germination responses to temperature. The effects of polyploidy on spore abortion and spore size are also investigated, since both traits may have adaptive consequences. METHODS: Three allotetraploid (Dryopteris corleyi, D. filix-mas and D. guanchica) and three related diploid taxa (D. aemula, D. affinis ssp. affinis and D. oreades) were studied. Spores were collected from 24 populations in northern Spain. Four spore traits were determined: abortion percentage, size, germination time and germination percentage. Six incubation temperatures were tested: 8, 15, 20, 25 and 32 degrees C, and alternating 8/15 degrees C. KEY RESULTS: Allotetraploids had bigger spores than diploid progenitors, whereas spore abortion percentages were generally similar. Germination times decreased with increasing temperatures in a wide range of temperatures (8-25 degrees C), although final germination percentages were similar among species irrespective of their ploidy level. Only at low temperature (8 degrees C) did two allotetraploid species reach higher germination percentages than diploid parents. Allotetraploids showed faster germination rates, which would probably give them a competitive advantage over diploid parents. Germination behaviour was not correlated with altitudinal distribution of species. CONCLUSIONS: The results of this study suggest that (i) relative fitness of allopolyploids at sporogenesis does not differ from that of diploid parents and (ii) neither does allopolyploidization involve a change in the success of spore germination.  相似文献

AIMS: The objective of the study was to evaluate the variability of germination response of 10 strains of proteolytic Clostridium botulinum. METHODS AND RESULTS: An automated turbidometric method was used to follow the fall in optical density. Spores of proteolytic Cl. botulinum germinated in response to l-alanine alone, with rate and extent of germination increased by addition of l-lactate or bicarbonate ions. Other hydrophobic amino acids also triggered germination of spores of proteolytic Cl. botulinum but not AGFK and inosine, germinants for Bacillus subtilis or B. cereus. CONCLUSIONS: Unlike spores of nonproteolytic Cl. botulinum, all proteolytic Cl. botulinum germinate in hydrophobic l-amino acids without l-lactate. However, a great variability of response to germinant is evidenced between the species. SIGNIFICANCE AND IMPACT OF THE STUDY: The selection of a model strain to study germination of Cl. botulinum spores should consider the variability in sensitivity to germinants shown in this work. In particular, the sequenced strain ATCC 3502 may not be the most appropriate model for germination studies.  相似文献

通过对采自河北雾灵山海拔1500m的仙鹤藓(Atrichum undulatum)的孢子萌发以及原丝体发育的观察,发现仙鹤藓孢子无休眠现象,孢子接种3天左右萌发:其原丝体发育分为绿丝体和轴丝体两个阶段。扩大培养实验结果表明。仙鹤藓茎叶体在添加2%葡萄糖的MS培养基上,置于25℃/20℃、14小时光照/10小时黑暗、36μmol·m^-2·s^-1条件下培养.产生新生茎叶体最多,且茎叶体长势最好,可以获得大量无菌材料。仙鹤藓愈伤组织诱导实验显示,形成愈伤组织的最佳培养基为添加2%葡萄糖和1.0mg·L^-16-BA的MS培养基。  相似文献
