Overexpression of genes from thermophiles in Escherichia coli is an attractive approach towards the large-scale production of thermostable biocatalysts. However, various factors can challenge efficient heterologous protein expression--one example is the formation of stable 5' mRNA secondary structures that can impede an efficient translation initiation. In this work, we describe the expression optimization of purine nucleoside phosphorylase from the thermophilic microbe Deinococcus geothermalis in E. coli. Poor expression levels caused by stable secondary 5' mRNA structure formation were addressed by two different approaches: (i) increasing the cultivation temperature above the range used typically for recombinant protein expression and (ii) optimizing the 5' mRNA sequence for reduced secondary structures in the translation initiation region. The increase of the cultivation temperature from 30°C to 42°C allowed a more than 10-fold increase of activity per cell and optimizing the 5' mRNA gene sequence further increased the activity per cell 1.7-fold at 42°C. Thus, the combination of high-temperature cultivation and 5' sequence optimization is described as an effective approach to overcome poor expression levels resulting from stable secondary 5' mRNA structure formation. We suggest that this method is especially suitable for improving the expression of proteins derived from thermophiles in E. coli.  相似文献

The gene (HSD-1) coding a human sperm membrane protein (hSMP-1) was isolated from a human testis cDNA expression library using antibodies found in the serum of an infertile woman. HSD-1 was localized to a single locus on chromosome 9 and assigned to band 9p12-p13 by fluorescent in situ hybridization (FISH) mapping and DAPI (4,6-diamidino-2-phenylindole) banding, using rat/human somatic cell hybrids and metaphase chromosomes of human lymphocytes. In rescreening a testis lambdagt10 cDNA expression library, the full-length cDNA (HSD-1) and several truncated cDNAs with heterologous regions were isolated from positive clones. The heterology consisted of deletion, insertion and alteration of the 5'-end. These heterologous truncated fragments may be produced by alternative splicing of mRNAs. Two recombinant prokaryotic expression vectors were constructed with one of the heterologous fragment (clone #26) with and without the alternative 5'-end. Escherichia coli transfected with the construct containing the alternative 5'-end failed to produce the recombinant product, whereas those transfected with the vector lacking the 5'-end produced hSMP-1. DNASIS analysis of the structure of #26 mRNA suggests that the 5'-end has a stable secondary configuration that may maintain the mRNA in an inactivated state, whereby hindering its translation and preventing the expression of the gene.  相似文献

This study addresses the problem of poor expression of somatotropin (ST) gene in E. coli and describes expression enhancement through silent and non-silent gene modifications. A series of constructs with codon optimization, substitution, deletion or addition in the 5'-region of the sequence encoding bubaline ST (BbST) were prepared. In the native form, the BbST expression was barely discernible on SDS-gel of the total E. coli cellular proteins (TCP). Introduction of silent and non-silent mutations in +2 to +8 codons, however, raised the expression levels to varying extents. In some constructs, a single base variation, i.e., G-->A or G-->C led to a remarkable increase in BbST expression (up to 28% of the TCP) whereas in the case of G-->T substitution the expression dropped to undetectable levels. Deletion of native GCC codon and addition of CAUCAC repeat thrice at +2 position enhanced the expression up to 48%, while insertion of NGG codons at the same position caused just a modest increase in expression. Differences in expression appeared as if related to the nature of early downstream codons (especially +2) and the stability of mRNA secondary structure although the levels of intracellular mRNA pools, as analyzed by real-time RT-PCR were quite similar. Overall, the study highlights the importance of 5'-end codon adaptations in solving the problems encountered in expressing the eukaryotic genes in E. coli.  相似文献

Codon usage and thermodynamic optimization of the 5'-end of mRNA have been applied to improve the efficiency of human protein production in Escherichia coli. However, high level expression of human protein in E. coli is still a challenge that virtually depends upon each individual target genes. Using human interleukin 10 (huIL-10) and interferon alpha (huIFN-alpha) coding sequences, we systematically analyzed the influence of several major factors on expression of human protein in E. coli. The results from huIL-10 and reinforced by huIFN-alpha showed that exposing AUG initiator codon from base-paired structure within mRNA itself significantly improved the translation of target protein, which resulted in a 10-fold higher protein expression than the wild-type genes. It was also noted that translation process was not affected by the retained short-range stem-loop structure at Shine-Dalgarno (SD) sequences. On the other hand, codon-optimized constructs of huIL-10 showed unimproved levels of protein expression, on the contrary, led to a remarkable RNA degradation. Our study demonstrates that exposure of AUG initiator codon from long-range intra-strand secondary structure at 5'-end of mRNA may be used as a general strategy for human protein production in E. coli.  相似文献

目的:研究人β神经生长因子(β-NGF)基因中稀有密码子及其mRNA二级结构对其在大肠杆菌中表达量的影响。方法:根据对人β-ngf中稀有密码子及其mRNA二级结构的研究,同义突变人β-ngf基因,通过PCR得到人β-ngf的5’端同义突变基因rh-β-ngfp32和全同义突变基因rh-β-ngfmu,将这2个序列克隆入载体pET3a中,得到重组质粒pET3a-ngfp32和pET3a-ngfmu,分别转化大肠杆菌BL21(DE3)感受态,IPTG诱导表达,收集菌体,SDS-PAGE检测其表达量的改变。结果:构建的pET3a-ngfp32和pET3a-ngfmu表达载体酶切和测序结果正确,SDS-PAGE结果显示,与在重组菌pET3a-NGF总蛋白中的表达量相比,目的蛋白rh-β-NGF在重组菌pET3a-NGFP32和pET3a-NGFmu中的表达量均明显增高,并且在重组菌pET3a-NGFmu中的表达量高于重组菌pET3a-NGFP32。结论:目的蛋白rh-β-NGF在重组菌pET3a-NGFP32和pET3a-NGFmu中表达量的增高,说明人β-ngf基因中稀有密码子和mRNA的二级结构对其在大肠杆菌中的表达有较为明显的影响,结果为构建rh-β-NGF的大肠杆菌工程菌株奠定了基础。  相似文献

The codon that follows the AUG initiation triplet (+2 codon) affects gene expression in Escherichia coli. We have extended this analysis using two model genes lacking any apparent Shine-Dalgarno sequence. Depending on the identity of the +2 codon a difference in gene expression up to 20-fold could be obtained. The effects did not correlate with the levels of intracellular pools of cognate tRNA for the +2 codon, with putative secondary mRNA structures, or with mRNA stability. However, most +2 iso-codons that were decoded by the same species of tRNA gave pairwise similar effects, suggesting that the effect on gene expression was associated with the decoding tRNA. High adenine content of the +2 codon was associated with high gene expression. Of the fourteen +2 codons that mediated the highest efficiency, all except two had an adenine as the first base of the codon. Analysis of the 3540 E. coli genes from the TransTerm database revealed that codons associated with high gene expression in the two expression systems are over-represented at the +2 position in natural genes. Codons that are associated with low gene expression are under-represented. The data suggest that evolution has favored codons at the +2 position that give high translation initiation.  相似文献

The use of the polymerase chain reaction was proposed for intron excision from genomic genes with known nucleotide sequences. Three exons (5, 6 and 7) of genomic interleukin 1 beta gene were amplified by means of thermostable DNA polymerase TthI from Thermus thermophilus on the base of cloned in M13 phage human genomic interleukin 1 beta gene. Synthetic oligonucleotides complementary to sequences flanking exons were used as primers. The fragments obtained by exon DNA amplification were joined in the correct order due to reciprocal complementation of end sequences, that was foreseen during synthesis of oligonucleotide primers followed by amplification of the enlarged fragments. As a result the structural interleukin-1 beta gene consisting of three exons was assembled. DNA sequences carrying the ATG initiation codon and XbaI recognition site at the 5'-end, and PstI recognition site at the 3'-end (essential for insertion into the expression vector) were formed by the additional end sequences of primers. The nucleotide sequence analysis of the obtained structural gene revealed its complete identity with natural interleukin 1 beta human gene. We created the expression vector pPR114 with phage lambda promoter PR thermo-inducible in case of the cIts857 repressor presence in cells. It was used for expression of the present gene. The interleukin 1 beta synthesized in E. coli had biological activity.  相似文献

为在大肠杆菌中非融合表达肝癌相关抗原HAb18G胞外区片段(HAb18GEF),将HAb18GEF基因的cDNA插入原核表达载体pET21a+。通过计算机辅助设计,对重组的HAb18GEF/pET21a+的mRNA翻译起始区(TIR)的二级结构和密码子偏性同时进行预测。结果发现其存在稳定的茎环结构和许多稀有密码子。通过优化二级结构和优化密码子偏性二种策略分别来降低HAb18GEF/pET21a+的mRNA翻译起始区(TIR)的稳定性。在不改变氨基酸序列的前提下,利用密码子的简并性,通过非连续定点突变实现这两种优化。将突变前后的重组子经酶切鉴定和测序验证后,转化感受态JM109DE3宿主菌后,随机挑菌37℃下用IPTG诱导表达。SDSPAGE、间接ELISA、Western blot 和细胞分级分离法分析这些重组子的诱导表达情况。RNA dot blot对比分析优化前后目的基因mRNA的量。结果证明,成功地构建了HAb18GEF/pET21a+及其二种优化突变体。仅优化TIR区二级结构或仅优化TIR区密码子偏性均能实现HAb18GEF蛋白的非融合表达,而未优化的重组子不表达任何HAb18GEF。非融合表达产物在大肠杆菌中主要以包涵体形式存在,高达293%。由于过表达和细胞渗漏,培养基和周质腔中也可检测到少许的HAb18GEF。优化二级结构和优化密码子偏性二种策略的HAb18GEF的非融合表达量基本相同。优化前后HAb18GEF转录的mRNA量没有差别。这些结果表明,降低mRNA翻译起始区的稳定性可实现肝癌相关抗原HAb18G胞外区片段在大肠杆菌中的非融合表达。  相似文献

The synthetic C5a gene was initially found to be expressed poorly in Escherichia coli. We undertook studies to determine the reasons for poor expression and to increase expression. The work was focused on the role of the mRNA structure in C5a expression and stability of its product in E. coli. We present data on the effects of varying the sequence at the 5' end of mRNA as well as different ribosome-binding sites on expression. Evaluation of the stability of C5a showed rapid degradation of C5a in wild-type E. coli (half-life 3-5 min). Screening of several protease-deficient strains of E. coli showed that C5a was much more stable in an htpR strain carrying a mutation in the sigma subunit of RNA polymerase that is specific for heat shock promoters. The mutation is associated with a proteolytic deficiency. The half-life of C5a was increased to 20 min. By manipulating the expression vector, the regulatory region for the C5a gene, the host strain, growth conditions and methods for recovering the protein, C5a levels were increased 300-fold over previously reported amounts to about 3% of total cellular protein.  相似文献

大肠杆菌(Escherichia coli)共表达系统常要求质粒具有不同抗生素抗性以及不同的复制子。利用粘性末端PCR技术,以含有大肠杆菌分子伴侣基因GroEL、GroES和唧E的pR—GESP质粒为模板,设计两对引物,通过两次独立的PCR反应扩增3个基因的多顺反子,将形成粘性末端的PCR产物插入NcoI和Xho1酶切的pACY.CDuet-1质粒,构建的pA—GESP质粒具有p15A复制子及氯霉素抗性,和具有ColE1复制子及卡那霉素抗性表达载体pET28b相容。SDS—PAGE显示含有pA—GESP质粒的大肠杆菌细胞中3个分子伴侣蛋白的表达水平和含有pR—GESP质粒的大肠杆菌细胞没有明显差异,它们对玉米丝氨酸消旋酶的可溶性表达有部分促进作用,但对N端含有组氨酸标签的玉米铁氧还蛋白还原酶的表达没有作用,在三个含有不同抗生素基因的质粒中共表达分子伴侣、5-氨基乙酰丙酸合酶和尿卟啉原III甲基化酶,两个酶连续催化的荧光产物在细胞内积累量为562.13±3.17/OD600,而没有分子伴侣的积累量为457.66±4.98/OD600,表明分子伴侣改善部分蛋白在大肠杆菌的可溶性表达和催化功能。  相似文献
