In this paper, we report the estimates of outcrossing rates using open-pollinated progeny arrays of 40 BC1 individuals of Jatropha developed as a result of interspecific hybridization between J. curcas and J. integerrima. For analysis PCR-based dominant AFLP and codominant SSR markers were used. The multilocus outcrossing rate (tm) estimated from AFLP markers (0.892 ± 0.112) are almost in the same range with SSR (0.884 ± 0.293) markers which indicate a high level of heterozygosity. A low value of inbreeding coefficient (F) also points out to the fact that outcrossing was the prevalent mode of reproduction in Jatropha and suggests maintenance of adequate genetic variability within families.

利用RAPD显性标记位点对泽苔草(Caldesia parnassifolia)湖南茶陵湖里居群进行了交配系统研究。采用10个家系共240个子代样品测定了该居群花粉库基因频率、固定指数(F)、多位点异交率(tm)和单位点异交率(ts)。结果显示该居群异交率处于较高的水平(tm=1.008),多位点异交率与单位点异交率差异不显著(tm-ts=-0.021),固定指数(F)的估计值小于0(F=-0.200),表明该居群双亲近交不明显,而以异交为主。这可能是受居群密度、传粉者的活动能力等因素影响的结果。  相似文献

The objectives of this study were to estimate the outcrossing rate and to explain genetic consequences of the development of seed in the endocarp in a natural population of neem in Bangladesh. Cotyledons of germinated open-pollinated seeds of individual trees were analyzed by starch-gel electrophoresis to examine allozymes. Three loci with clear Mendelian segregation were used to estimate outcrossing rate. A multilocus mixed mating model was used to evaluate the mating system. The population exhibited high outcrossing rates both for multilocus (tm=0.90±0.024) and mean single-locus (ts=0.92±0.020) estimates. The difference between these two parameters (tm–ts=0±SE 0.038) was insignificant, indicating that there was no ’biparental inbreeding’ in the population. The degree of variance of the estimates of multilocus outcrossing rates decreased when two or more loci were included. In order to elucidate the significance of polycarpy a total of 471 seeds were counted out of 440 endocarps. This mechanism appears to be a possible way of avoiding inbreeding. The mating-system was investigated in a natural population of the tetraploid taxon Acacia nilotica ssp. leiocarpa using open-pollinated seeds from 15 families. Six-day-old germinated seeds were used for starch-gel electrophoresis. Three enzyme systems (ADH, EST-1, and 6PGD) were scored. Isozyme banding patterns and segregation of isozyme variants within families suggest that the species is an autotetraploid displaying tetrasomic inheritance. Estimates of single-locus (ts=0.358) and multilocus (tm=0.384) population outcrossing rates were homogeneous, and indicate substantial selfing in the population. Heterogeneity of outcrossing estimates among loci and families were marked, suggesting departure from the assumptions of the mixed-mating model. Implications of the result for the utilization of germplasm in tree-improvement programmes are noted.  相似文献

Summary A multilocus procedure was used to estimate outcrossing rates in ten roadside populations of Trifolium hirtum in California. Three groups of populations were studied: cultivars, hermaphroditic, and gynodioecious (sexually dimorphic) populations. The multilocus outcrossing rate (tm) varied from 0.05 to 0.43 among populations. Population level tm estimates were significantly correlated with the observed heterozygosity in gynodioecious populations but not in hermaphroditic populations. The outcrossing rate of hermaphrodites and females was estimated in three gynodioecious populations; the estimates of tm varied from 0.09 to 0.23 for hermaphrodites and from 0.73 to 0.80 for females. The distribution of outcrossing rates in gynodioecious populations is bimodal. Our results indicate that for the levels of selfing observed among hermaphrodites, inbreeding depression is likely to be a major factor in the maintenance of females in gynodioecious populations.  相似文献

 To evaluate how environmental and genetic factors influence mating-system evolution, accurate estimates of outcrossing rates of individual plants (families) are required. Using isozyme markers, we observed wide variation in family outcrossing rates in three natural populations of Asclepias incarnata using three statistical methods: (1) a multilocus maximum-likelihood procedure (t m); (2) a multilocus method-of-moments procedure (t a); and (3) a direct comparison of progeny phenotypes against maternal phenotypes (t d). Neighborhood floral-display size was positively correlated with t a in one population, but showed no relationship with any of the other estimates of outcrossing for any population. Monte-Carlo simulations revealed that statistical variation associated with these estimation procedures can be large enough to explain all of the observed variation in outcrossing. We also found that significant, spurious correlations with neighborhood floral display could arise, on average, 7% of the time by chance alone. Our observations suggest that it is difficult to obtain accurate estimates of outcrossing in naturally pollinated plants using the estimation procedures currently available. Moreover, we caution that attempts to interpret observed variation in family outcrossing estimates by observing variation in ecological parameters could be misleading. Mating system and inbreeding depression in quantitative traits of whitebark pine (Pinus albicaulis Engelm.) was determined using isozymes and a seedling common garden experiment. Simultaneous isozyme analysis of embryo and haploid megagametophyes from progeny arrays of families in three distinct geographic regions (Oregon, Montana, and southern British Columbia) was used to estimate parental and progeny inbreeding coefficients, as well as regional and family mean multilocus outcrossing rates (t m). Quantitative trait family means of seedlings from the same families growing in two temperature treatments in a common garden experiment were regressed on the estimated inbreeding coefficient to determine the presence and magnitude of inbreeding depression. Regional estimates of t m ranged from 0.73 to 0.93, with a mean over all regions of 0.86. Family mean t m values indicated predominant outcrossing; however, some individuals experienced substantial inbreeding. The Oregon region had a significant excess of heterozygotes in the parental generation relative to Hardy–Weinberg equilibrium, while both the Oregon and southern BC regions had a heterozygote deficiency in progeny, suggesting selection against inbred individuals. Biomass in the ambient temperature treatment for the southern BC region was the only trait significantly related to inbreeding coefficient. The mean inbreeding coefficient for this region was 0.25, and based on this relationship, mean predicted biomass would be reduced by 19.6% in this region if inbred individuals are not removed by selection. The estimated outcrossing rate of whitebark pine is slightly lower than most wind-pollinated conifers, and while most individuals are highly outcrossing, some experience substantial inbreeding.  相似文献

Five allozyme polymorphisms were used to analyze the mating system in a Sitka spruce seed orchard in Saanichton, British Columbia. Allelic frequencies differed between the pollen and maternal pools at three of the five loci, with alleles rare in the maternal pool being even rarer in the effective pollen pool. Minor differences in pollen allelic frequencies were observed in the upper vs. lower crown. The multilocus outcrossing rate of the upper crown (tm = 0.909) exceeded that of the lower crown (tm = 0.764). Single-locus estimates of the outcrossing rate were significantly heterogeneous, with the lowest estimate of outcrossing, t = 0.773, observed for PGM-2 locus. Analyses of the mating system for the three maternal PGM-2 genotypes revealed heterogeneous pollen allelic frequencies and heterogeneous outcrossing rates, possibly due to assortative mating at this locus.  相似文献

The mating system and seed variation of Acacia hybrid (A. mangium × A. auriculiformis) were studied using allozymes and random amplified polymorphic DNA (RAPD) markers, respectively. Multi-locus outcrossing rate estimations indicated that the hybrid was predominantly outcrossed (mean±s.e. t m = 0.86±0.01). Seed variation was investigated using 35 polymorphic RAPD fragments. An analysis of molecular variance (AMOVA) revealed the highest genetic variation among seeds within a pod (66%–70%), followed by among pods within inflorescence (29%–37%), and the least variation among inflorescences within tree (<1%). In addition, two to four RAPD profiles could be detected among seeds within pod. Therefore, the results suggest that a maximum of four seeds per pod could be sampled for the establishment of a mapping population for further studies.  相似文献

The mating system and allozyme variation at 20 loci in three Klamath Mountains and two Sierra Nevada populations of Jeffrey pine (Pinus jeffreyi Grev. & Balf.) were investigated. On average, multilocus estimates of the proportion of viable progeny due to outcrossing (tm) were high in all populations (mean tm = 0.935, range 0.881 to 0.971). Despite differences in stand structure, tm did not differ (P > 0.05) between the Klamath (mean tm = 0.933) and Sierra Nevada (mean tm = 0.937) populations. At all but one locus in one population and at two in another, genotype frequencies fit (P > 0.05) Hardy-Weinberg expectations. Mean estimates of observed heterozygosity in Klamath (0.182) and Sierra Nevada (0.327) populations were comparable to values reported for other conifers.  相似文献

Progress in the breeding of plantain and banana has been restricted by the complex genetic structure and behaviour of cultivated polyploid Musa. Genetic improvement has been hindered due to the large amount of space required for growth and maintenance of plant populations, in addition to the long growth cycle and the low levels of fertility and seed viability characteristic of cultivated genotypes. Molecular marker assisted breeding has the potential to dramatically enhance the pace and efficiency of genetic improvement in Musa. This study was conducted to compare different PCR-based marker systems (RAPD, VNTR and AFLP) for the analysis of breeding populations generated from two diverse Musa breeding schemes. All three assays detected a high level of polymorphism between parental genotypes and within progeny populations. As expected, AFLP assays had by far the highest multiplex ratio while VNTR analysis detected the highest levels of polymorphism. AFLP analysis of a full-sib tetraploid hybrid population confirmed previous reports based on VNTR analysis, of a high frequency of recombination during 2n (3x) gamete formation by a triploid plantain landrace. In addition, both VNTR and RAPD analyses of a full-sib triploid hybrid population suggested a high frequency of homoeologous recombination during n (2x) gamete formation by tetraploid hybrids. In general, there was a poor correlation between estimates of genetic similarity based on different types of marker. Microsatellites were used to evaluate the mating system of the remaining trees in a logged population of Swietenia macrophylla, a highly valuable and threatened hardwood species, in the Brazilian Amazon. A total of 25 open pollinated progeny arrays of 16 individuals, with their mother trees, were genotyped using eight highly polymorphic microsatellite loci. Genotypic data analysis from the progeny arrays showed that 373 out of the 400 seedlings (93.25%) were unambiguously the result of outcrossed matings and that the remaining 6.75% had genotypes consistent with self-fertilisation. Apomixis could be ruled out, since none of the 400 seedlings analysed had a multi-locus genotype identical to its mother tree. The high estimate of the multi-locus outcrossing rate (t m = 0.938 ± 0.009) using the mixed mating model also indicated that the population in this remnant stand of S. macrophylla was predominantly allogamous. The relatively large difference between the multi-locus and single-locus outcrossing estimates (t m −t s = 0.117 ± 0.011) provides evidence that, in spite of the high outcrossing rate, a considerable degree of biparental inbreeding has contributed to the genetic structure of this population. Levels of outcrossing were not evenly distributed among maternal trees (t m ranging from 0.39 to 1.00), suggesting the occurrence of a variable degree of self-incompatibility and/or dichogamy among individual trees of this monoecious species. Due to its generalist pollination system and some level of tolerance for selfing, S. macrophylla seems to be resilient to environmental disturbances such as those caused by logging, since it sets fruits with predominantly outcrossed seeds even at low stand densities. Therefore, the remaining individuals in logged areas or in relict fragments may be very important for long-term population recovery and genetic conservation programmes.  相似文献

The aim of this study was to determine and compare the mating systems among Psychotria tenuinervis populations at anthropogenic edges, natural edges, and the forest interior using allozyme electrophoresis of naturally pollinated progeny arrays. P. tenuinervis showed low outcrossing rates, varying from 37% to 50% of the mating attributable to outcrossing and 50% to 63% attributable to self-fertilization, in the three habitats. The forest interior had the highest outcrossing rate (t m = 0.50 and t s = 0.43) among the three habitats. However, there were no differences in either multilocus or single-locus rates among the three habitats, indicating that the contribution of biparental inbreeding to the apparent selfing rate in these populations was very low. The multilocus (t m) and single-locus (t s) outcrossing rates for the P. tenuinervis in the sample plots within each habitat showed great heterogeneity. In conclusion, edge creation seems not to influence its mating systems. Additionally, although P. tenuinervis is a distylous species, the population’s inbreeding can be attributed almost entirely to self-fertilization.  相似文献

A Brassica juncea mapping population was generated and scored for seed coat colour. A combination of bulked segregant analysis and AFLP methodology was employed to identify markers linked to seed coat colour in B. juncea. AFLP analysis using 16 primer combinations revealed seven AFLP markers polymorphic between the parents and the bulks. Individual plants from the segregating population were analysed, and three AFLP markers were identified as being tightly linked to the seed coat colour trait and specific for brown-seeded individuals. Since AFLP markers are not adapted for large-scale application in plant breeding, our objective was to develop a fast, cheap and reliable PCR-based assay. Towards this goal, we employed PCR-walking technology to isolate sequences adjacent to the linked AFLP marker. Based on the sequence information of the cloned flanking sequence of marker AFLP8, primers were designed. Amplification using the locus-specific primers generated bands at 0.5 kb and 1.2 kb with the yellow-seeded parent and a 1.1-kb band with the brown-seeded parent. Thus, the dominant AFLP marker (AFLP8) was converted into a simple codominant SCAR (Sequence Characterized Amplified Region) marker and designated as SCM08. Scoring of this marker in a segregating population easily distinguished yellow- and brown-seeded B. juncea and also differentiated between homozygous (BB) and heterozygous (Bb) brown-seeded individuals. Thus, this marker will be useful for the development of yellow seed B. juncea cultivars and facilitate the map-based cloning of genes responsible for seed coat colour trait. Inbreeding depression is one of the major selective forces driving the evolution of mating systems. Previous theories predict that long-lived plants will show a negative correlation between inbreeding depression and the level of inbreeding (as determined by an inbreeding coefficient) at maturity, but the extent of this correlation may vary among life stages because of variation in the genetic basis for inbreeding depression at different stages. To test this prediction, I used electrophoretic allozyme analysis and pollination experiments to examine the fixation index (F is) at maturity and inbreeding depression in the early and late life stages of two populations with different outcrossing rates of a highly self-fertilizing tree, Magnolia obovata. The magnitude of inbreeding depression for early survival (δ e) in an outcrossing population (t m = 0.51; F is = −0.015) was higher (δ e = 0.97) than that in an inbreeding population (t m = 0.18; F is = 0.15; δ e = 0.38). From these results, I estimated that both populations exhibited high inbreeding depression for late survival (δ l) (0.94 in the outcrossing population and 0.93 in the inbreeding one) and lifetime survival (δ t) (0.99 and 0.96, respectively). My results and previously published data demonstrate the predicted relationship between inbreeding depression and the level of inbreeding for early survival, but not for late survival. This suggests that there is a differential genetic basis for inbreeding depression at different life stages. The inbreeding depression for late survival appears to play a central role in the maintenance of reproductive traits that promote outcrossing in M. obovata.  相似文献

The aim of this study was to assess the genetic basis of rust mite (Aculus schlechtendali) resistance in apple (Malus × domestica). A. schlechtendali infestation of apple trees has increased as a consequence of reduced side effects of modern fungicides on rust mites. An analysis of quantitative trait loci (QTLs) was carried out using linkage map data available for F1 progeny plants of the cultivars ‘Fiesta’ × ‘Discovery’. Apple trees representing 160 different genotypes were surveyed for rust mite infestation, each at three different sites in two consecutive years. The distribution of rust mites on the individual apple genotypes was aggregated and significantly affected by apple genotype and site. We identified two QTLs for A. schlechtendali resistance on linkage group 7 of ‘Fiesta’. The AFLP marker E35M42-0146 (20.2 cM) and the RAPD marker AE10-400 (45.8 cM) were closest positioned to the QTLs and explained between 11.0% and 16.6% of the phenotypic variability. Additionally, putative QTLs on the ‘Discovery’ chromosomes 4, 5 and 8 were detected. The SSR marker Hi03a10 identified to be associated to one of the QTLs (AFLP marker E35M42-0146) was traced back in the ‘Fiesta’ pedigree to the apple cultivar ‘Wagener’. This marker may facilitate the breeding of resistant apple cultivars by marker assisted selection. Furthermore, the genetic background of rust mite resistance in existing cultivars can be evaluated by testing them for the identified SSR marker. Understanding the genetic structure of the population of Alternaria solani (AS) is an important component of epidemiological studies of early blight, a severe disease that affects potato (Po) and tomato (To) worldwide. Up to 150 isolates obtained from both hosts were analysed with RAPD and AFLP markers to estimate the amount and distribution of genetic variability of AS in Brazil. Using RAPD, gene diversity (h = 0.20) and scaled indices of diversity of Shannon (H′ = 0.66) and Stoddart and Taylor’s (G = 0.31) for the Po population were higher than those of the To (h = 0.07, H′ = 0.34, G = 0.17). For AFLP, the statistics for the Po (h = 0.17, H′ = 0.86, G = 0.49) and To (h = 0.17, H′ = 0.85, G = 0.36) populations were similar. For each RAPD and AFLP locus, the allele frequency for the overall population ranged from 0.006 to 0.988, and 0.007 to 0.993, respectively. Genetic differentiation was high (GST = 0.41 and θ = 0.59) and moderately high (GST = 0.23 and θ = 0.37) when estimated with RAPD and AFLP, respectively. Based on cluster analyses, there was strong evidence of association of pathogen haplotypes with host species. The null hypothesis of random association of alleles was rejected in the analysis of both RAPD (IA = 13.1, P < 0.001) and AFLP (IA = 2.2, P < 0.001) markers. The average number of migrants was estimated to be around one and two individuals per generation, using RAPD and AFLP, respectively. There was no correlation between genetic distance and geographical origin of AS haplotypes for RAPD (r = ?0.07, P = 0.84) and AFLP (r = ?0.03, P = 0.70). The AS population is clonal with high genetic variability, and there is genetic differentiation between the populations that affect To and Po.  相似文献

 A PCR-based marker (E20570) linked to the gene Gm4t, which confers resistance to a dipteran pest gall midge (Orseolia oryzae), has been mapped using the restriction fragment length polymorphism (RFLP) technique in rice. Gm4t is a dominant resistance gene. We initially failed to detect useful polymorphism for this marker in a F3 mapping population derived from a cross between two indica parents, ‘Abhaya’בShyamala’, with as many as 35 restriction enzymes. ‘Abhaya’ carries the resistance gene Gm4t and ‘Shyamala’ is susceptible to gall midge. Subsequently, E20570 was mapped using another mapping population represented by a F2 progeny from a cross between ‘Nipponbare’, a japonica variety, and ‘Kasalath’, an indica variety, in which the gene Gm4t was not known to be present. Gm4t mapped onto chromosome 8 between markers R1813 and S1633B. Our method, thus, presents an alternative way of mapping genes which otherwise would be difficult to map because of a lack of polymorphism between closely related parents differing in desired agronomic traits.  We have constructed a genetic linkage map within the cultivated gene pool of cowpea (2n=2x=22) from an F8 recombinant inbred population (94 individuals) derived from a cross between the inbreds IT84S-2049 and 524B. These breeding lines, developed in Nigeria and California, show contrasting reactions against several pests and diseases and differ in several morphological traits. Parental lines were screened with 332 random RAPD decamers, 74 RFLP probes (bean, cowpea and mung bean genomic DNA clones), and 17 AFLP primer combinations. RAPD primers were twice as efficient as AFLP primers and RFLP probes in detecting polymorphisms in this cross. The map consists of 181 loci, comprising 133 RAPDs, 19 RFLPs, 25 AFLPs, three morphological/classical markers, and a biochemical marker (dehydrin). These markers identified 12 linkage groups spanning 972 cM with an average distance of 6.4 cM between markers. Linkage groups ranged from 3 to 257 cM in length and included from 2 to 41 markers, respectively. A gene for earliness was mapped on linkage group 2. Seed weight showed a significant association with a RAPD marker on linkage group 5. This map should facilitate the identification of markers that “tag” genes for pest and disease resistance and other traits in the cultivated gene pool of cowpea. The importance of pollinator visitation for determining both maternal reproductive success and outcrossing rates was investigated in the facultatively autogamous annual, Lupinus nanus. Sixty plants in each of two adjacent sites were assigned to either a pollinator-exclusion, pollen-augmentation, or open-pollinated (control) treatment. Flower, fruit, and seed production were recorded for each plant, and outcrossing rates were determined for plants in all three treatments at each site. Pollinator-exclusion reduced the reproductive success of plants at Site 1, but had no effect on reproduction for plants at Site 2. Pollen-augmentation increased fruit production by 22% and seed production by 45% at Site 2, but had no effect on reproduction for plants at Site 1. Plants in the open-pollinated, control treatment outcrossed at intermediate rates at both sites, with plants at Site 2 (outcrossing rate, t = 0.66) outcrossing at a significantly higher rate than plants at Site 1 (t = 0.40). The pollen-augmentation treatment increased the outcrossing rate for plants at both sites (Site 1, t = 0.72; Site 2, t = 0.78). These results indicate that pollinator availability can limit maternal reproductive success for this facultatively autogamous plant species. Additionally, the intermediate outcrossing rates observed in nature for this species are not a consequence of plant characters alone, but instead result from plant characters combined with the local pollination ecology.  相似文献
