The involvement of the glycosylphosphatidylinositol/inositol phosphoglycan (gly-PtdIns/IPG) system in the stimulation of macromolecular syntheses in human fibroblasts has been investigated. The study demonstrates that an insulin sensitive gly-PtdIns/IPG system is present in human fibroblasts, that IPG can significantly stimulate DNA, RNA, and protein synthesis, and that the action of insulin on DNA synthesis as well as that of IPG can be significantly reduced by a specific anti-IPG antibody. These results strongly support the hypothesis that the gly-PtdIns/IPG system is involved in the signal transduction pathway leading to the stimulation of cell proliferation.  相似文献

为了探讨中国H3N2亚型人流感病毒血凝素基因变异与流行关系,对1995~2005年中国分离的550株H3N2流感病毒的HA1序列进行分析.HA1基因进化树显示出以很长的主干和很短的侧枝为特征.HA1的氨基酸位点变异主要位于5个已知的抗原位点及其附近,同时其它位点也有改变.通过对HA1序列资料分析发现这期间导致H3N2流行的序列改变的三种可能,第一种是同时出现多位点变化,第二种是位点变化逐渐发生累积到多个位点变化,第三种是单个抗原位点和受体结合位点同时改变,均可以引起H3N2流行.  相似文献

利用1988年1月、7月和2004年3月、5月、9月、12月在大亚湾海域开展的7个航次63个站次的大型底栖动物调查数据,通过Brey经验公式对两个时段的大型底栖动物生产力和P/B值的变化特征进行研究.结果表明: 目前该海域大型底栖动物年生产力为10.22 g AFDM ·m-2 ,平均P/B值为0.85·a-1,在我国各海域中处于中上水平.两个指标的数值均高于1988年的7.25 g AFDM ·m-2·a-1和0.76·a-1,印证了大亚湾高输出系统是依赖快速周转来维持系统的高生产力的结论;大亚湾大型底栖动物4个主要类群的生产力和P/B值变化存在较大差异;湾内生产力的平面分布也发生了较大变化,生产力和P/B值的区域差异与人类活动影响密切相关, 生产力变化最显著的区域内人类活动影响显著.  相似文献

Chaetomellic acids are a class of alkyl dicarboxylic acids that were isolated from Chaetomella acutiseta. They are potent and highly specific farnesyl-pyrophosphate (FPP) mimic inhibitors of Ras farnesyl-protein transferase. We have previously described the first biogenetic type aldol condensation-based total synthesis of chaetomellic acid A. Modification of the later steps of that synthesis resulted in the efficient syntheses of chaetomellic acids A and B in three steps with 75-80% overall yield. In this report, details of the original total syntheses of chaetomellic acids A, B and C, the new syntheses of acids A and B and structure-activity relationship of these compounds against various prenyl transferases including human and yeast FPTase and bovine and yeast GGPTase I are described. Chaetomellic acids are differentially active against human and yeast FPTase. Chaetomellic acid A inhibited human and yeast FPTase activity with IC50 values of 55 nM and 225 microM, respectively. In contrast, chaetomellic acid C showed only a 10-fold differential in inhibitory activities against human versus yeast enzymes. In keeping with molecular modeling-based predictions, the compounds with shorter alkyl side chains (C-8) were completely inactive against FPTase.  相似文献

以往,在东亚大陆发现的更新世中期人类化石被分别归入直立人和古老型智人。这种分类的主要依据是化石形态特征以及年代。魏敦瑞对周口店第一地点人类化石研究描述的一些头骨、下颌骨和牙齿特征通常被作为判定直立人的标准。根据这些化石的年代分布,一般将30万年前的中更新世晚期作为划分直立人与古老型智人的大致年代界限。近20年来,在非洲、欧洲和东亚新发现了一些更新世中期人类化石,目前古人类学界对中国更新世中期人类化石特征及演化有了与以往不同的认识。最近对大荔、许家窑、盘县大洞、许昌、华龙洞等人类化石的研究显示,近30万年以来东亚大陆人类演化呈现复杂的多样性,将这一时期人类全部归入古老型智人难以准确反映更新世中期中国古人类演化模式及规律。本文结合近年中国更新世中期人类演化研究进展,选择部分具有演化及分类价值的形态特征,分析这些特征在更新世中期中国古人类化石的表现特点。在此基础上,对更新世中期中国古人类演化模式做了尝试性探讨。本研究发现,周口店、和县、沂源、南京等中更新世早期人类化石呈现有较多的区域性特征,形态特征表现相对稳定;而大荔、金牛山、许家窑、许昌、华龙洞、马坝、盘县大洞等中更新世晚期人类在化石形态特征表现复杂多样,变异范围大。此外,在这一时期人类化石上发现较多与生存活动、健康、环境适应有关的证据。根据这些发现,作者认为中国中更新世早期组人类演化以形态连续性为主;进入中更新世晚期,中国古人类演化区域性特征减弱,演化模式以多样性为主。一系列新的化石发现和研究证据提示中更新世晚期东亚大陆可能生存有不同的古人类成员。根据目前掌握的化石形态和年代证据,大约30万年前是中国更新世中期演化变化关键时间节点。  相似文献

To study the effects of and approaches to technological progress on China’s regional environmental performance, this study first estimates China’s regional environmental performance and its variation indexes by applying a slack-based model (SBM) and an entropy-based model (EBM). The results indicate that the environmental performance in different regions of China has improved, but the rate of improvement differs greatly. This may be attributed to heterogeneous characteristics and changes in the green technology innovation level in different regions. Considering the overflow effect of environmental pollution among different regions, we study the impact of various technological progress patterns on China’s regional environmental performance using spatial econometrics, and we find that there are significant spatial effects of technology innovation, technology transfer, and imitative innovation on China’s regional environmental performance. Also, different technological progress patterns have different effects. Specifically, independent innovation has failed to effectively improve regional environmental performance, whereas the introduction of technology and imitative innovation have significantly improved this performance. Moreover, after the cross-items of independent innovation and human capital are introduced, the effects of technology introduction and imitative innovation on China’s regional environmental performance through the absorptive capacity of human capital remain significant, whereas the effect of independent innovation on regional environmental performance via the absorptive capacity of human capital becomes more obvious. Based on this and from the perspective of environmental enhancement, we believe that China should strengthen human capital accumulation and give consideration to imitative innovation and technology introduction while emphasizing independent innovation.  相似文献

Large-scale preparation of 5-bromo-1-mesyluracil (BMsU) 4 has been optimized. BMsU was synthesized by condensation of silylated 5-bromouracil and MsCl in acetonitrile or by the reaction of 5-bromouracil with MsCl in pyridine. The same product was obtained by bromination of 1-mesyluracil. The purpose of this study was to elucidate the effects of BMsU on the biosynthetic activity of tumor cell enzymes involved in DNA, RNA and protein syntheses, and in de novo and salvage pyrimidine and purine syntheses. Investigations were performed in vitro on human cervix carcinoma cells (HeLa). BMsU displayed inhibitory effects on DNA and RNA syntheses in HeLa cells after 24 h of treatment. De nova biosynthesis of pyrimidine and purine was also affected. Antitumor activity of BMsU is closely associated with its inhibitory activity on the enzymes that play an important role in the metabolism of tumor cells. In vivo antitumor activity of BMsU was also investigated. The model used in investigations was a mouse anaplastic mammary carcinoma transplanted into the thigh of the right leg of CBA mice. Significant reduction in tumor growth time was achieved with BmsU administered at a dose of 50 mg/kg.  相似文献

应用空间自相关分析方法对中国栗疫病菌17个居群RAPD遗传变异的空间结构进行研究,以探讨栗疫病菌居群遗传变异的分布特征及其形成机制。结果表明:中国栗疫病菌居群缺乏空间结构,绝大多数RAPD位点变异为随机分布的空间模式,但部分位点表现出渐变、斑块和双向渐变的非随机分布模式,又显示了一定的空间结构。推测其形成原因可能是长距离的基因流、人类活动、地理隔离以及栗疫病菌本身的繁殖特性综合作用的结果,并依据部分位点呈单向渐变的模式推测西南地区为中国栗疫病菌的起源中心。  相似文献

This study examined variation in leaf traits and water relations in 12 evergreen and semideciduous woody species that occur in both seasonal wet and dry forests in Costa Rica and compared intra-specific leaf–trait correlations to those found in inter-specific global studies. The following traits were measured in both forests across seasons for 2 years: leaf nitrogen (N), leaf carbon (C), specific leaf area (SLA), toughness, cuticle thickness, leaf thickness, and leaf lifespan (LLS). Leaf water potential (LWP) and water content (LWC) were measured as indices of plant available water. Canopy openness, soil moisture, and herbivory were also measured to compare environmental variation across sites. Although species contributed the greatest amount to variation in traits, season, forest, and their interaction had a large influence on patterns of intra-specific leaf–trait variation. Leaf traits that contributed most to variation across sites were C, LWP, leaf thickness, and SLA. Traits that contributed most to variation across seasons were leaf toughness, LWP, and LWC. Furthermore, leaf traits were more correlated (i.e., number and strength of correlations) in the dry than in the wet forest. In contrast to results from global literature syntheses, there was no correlation between LLS and N, or LLS and SLA. Both light and water availability vary seasonally and may be causing variation in a number of leaf traits, specifically those that relate to water relations and leaf economics. Strong seasonality may cause leaf–trait relationships at the local scale to differ from those documented in continental and global-scale studies.  相似文献

alpha-OH-PdG, an acrolein-derived deoxyguanosine adduct, inhibits DNA synthesis and miscodes significantly in human cells. To probe the cellular mechanism underlying the error-free and error-prone translesion DNA syntheses, in vitro primer extension experiments using purified DNA polymerases and site-specific alpha-OH-PdG were conducted. The results suggest the involvement of pol eta in the cellular error-prone translesion synthesis. Experiments with xeroderma pigmentosum variant cells, which lack pol eta, confirmed this hypothesis. The in vitro results also suggested the involvement of pol iota and/or REV1 in inserting correct dCMP opposite alpha-OH-PdG during error-free synthesis. However, none of translesion-specialized DNA polymerases catalyzed significant extension from a dC terminus when paired opposite alpha-OH-PdG. Thus, our results indicate the following. (i) Multiple DNA polymerases are involved in the bypass of alpha-OH-PdG in human cells. (ii) The accurate and inaccurate syntheses are catalyzed by different polymerases. (iii) A modification of the current eukaryotic bypass model is necessary to account for the accurate bypass synthesis in human cells.  相似文献

Due to its haploid nature and its predominantly uniparental mode of inheritance, the mitochondrial genome has been analyzed extensively in population and evolutionary genetic studies of eukaryotes. Among the mitochondrial DNA (mtDNA) fragments, the region surrounding the origin of replication is the most commonly studied. However, most of the studies have focused on animals and little is known about the extent and patterns of sequence variation around the mtDNA origin of replication (mtOri) in fungi. In this study, we found abundant variation in a 597bp fragment surrounding the mtOri for 53 isolates of the pathogenic yeast Candida albicans obtained from a diverse group of hosts in Hainan, a tropical island of China. Within this DNA fragment, a total of 17 haplotypes were found for the 53 isolates. The extent of sequence variation for this group of strains was similar to that for 24 strains that represented the global nuclear genotype diversity. In contrast to those in animals where there were significant biases in favor of transitional mutations (e.g. the transition to transversion ratio is about 20 in human mtDNA), our data showed a transition to transversion ratio of approximately 0.5 around mtOri in C. albicans. Our analysis revealed no apparent geographic pattern of sequence variation based on the birthplaces of the analyzed hosts. However, the sample from patients had a lower genotypic diversity than those from healthy hosts borne either in Hainan or elsewhere in China. Our results suggest that C. albicans mtDNA has a base substitution pattern different from its nuclear genome and that sequences from the mtOri region could enhance our understanding of C. albicans genome evolution and population structuring.  相似文献

鉴定老龄树的古树等级并了解其生长历史是国家古树名木普查建档工作的重要内容.本研究利用树木年轮学方法,调查估算了西藏拉萨市尼木县20棵胸径较大的核桃和10棵大果圆柏的树龄,分析其生长历史.结果表明: 20棵核桃树中,一级古树、二级古树及三级古树分别为4、7及9棵;10棵柏树中,有2棵一级古树、4棵二级古树、4棵三级古树.历史上的持续低生长现象在某些时间段内2个树种间表现出明显的同步性,主要集中在20世纪40至80年代,但古树生长在个体间表现出较大的生长差异.古树生长与帕默尔干旱指数(PDSI)表现出显著的正相关关系.研究区古树主要分布于村庄和道路两旁,人类活动频繁.古树的径向生长受到该区域气候调控的同时也受到人类活动的干扰,这导致了古树的径向生长变化在不同树种间表现出空间同步性,而在个体间表现出较大的生长差异.因此,相关部门在制定古树保护措施时应同时考虑气候变化和人类活动对古树生长的影响.  相似文献

河西走廊植被净初级生产力时空变化及其影响因子研究   总被引:3,自引:0,他引:3  
干旱半干旱区植被NPP变化对全球碳循环有重要影响,该区域NPP对气候变化的响应表现出较大的时空异质性,其驱动机制并不十分清楚。选择中国河西走廊,利用随机森林算法估算了2002-2018年的NPP,基于偏导数法计算了气候与人类活动对NPP的影响。结果表明:(1)随机森林算法能较好的适用于干旱半干旱区NPP估算。(2)2002-2018年间河西走廊年NPP的平均值为153.32 gC m-2 a-1,总量为37.468 Tg C/a,呈东南向西北递减的分布特征,研究期间NPP呈2.37 gC m-2 a-1P=0.09)增长趋势。(3)河西走廊NPP变化52.51%由气候因子贡献,47.49%由人类活动贡献。(4)在气候变化对NPP的影响中,降水主导了该区72.21%的区域,温度对NPP变化量的贡献占73.71%,前者影响着NPP变化格局,后者主导NPP变化数量。升温和增湿均有利于该区NPP增加,随着西北地区气候暖湿化,河西走廊植被会持续改善,该研究有助于理解干旱半干旱区NPP对气候变化的响应机制,为适应气候变化政策制定提供理论依据。  相似文献

Aim To document the patterns of fish species richness and their possible causes in China's lakes at regional and national scales. Location Lakes across China. Methods We compiled data of fish species richness, limnological characteristics and climatic variables for 109 lakes across five regions of China: East region, Northeast region, Southwest region, North‐Northwest region, and the Tibetan Plateau. Correlation analyses, regression models and a general linear model were used to explore the patterns of fish species richness. Results At the national scale, lake altitude, energy availability (potential evapotranspiration, PET) and lake area explained 79.6% of the total variation of the lake fish species richness. The determinants of the fish richness pattern varied among physiographic regions. Lake area was the strongest predictor of fish species richness in the East and Southwest lakes, accounting for 22.2% and 82.9% of the variation, respectively. Annual PET explained 68.7% of the variation of fish richness in the Northeast lakes. Maximum depth, mineralization degree, and lake area explained 45.5% of the fish variation in the lakes of the North‐Northwest region. On the Tibetan Plateau, lake altitude was the first predictor variable, interpreting 32.2% of the variation. Main conclusions Lake altitude was the most important factor explaining the variation of fish species richness across China's lakes, and accounted for 74.5% of the variation. This may stem in part from the fact that the lakes investigated in our study span the largest altitudinal range anywhere in the world. The effects of the lake altitude on fish species richness can be separated into direct and indirect aspects due to its collinearity with PET. We also found that the fish diversity and its determinants were scale‐dependent. Fish species richness was probably energy‐determined in the cold region, while it was best predicted by the lake area in the relatively geologically old region. The independent variables we used only explained a small fraction of the variations in the lake fish species richness in East China and the Tibetan Plateau, which may be due to the effects of human activity and historical events, respectively.  相似文献

Activated hepatic stellate cells produce vascular endothelial growth factor (VEGF). VEGF has been shown to act on mesenchymal cells as well. If hepatic stellate cells can express FLT tyrosine receptor family, flt-1 and KDR/flk-1, their function might be regulated by VEGF in an autocrine manner. This hypothesis was tested using hepatic stellate cells isolated from normal rats. Northern blot analysis and immunocytochemical study revealed that hepatic stellate cells cultured for 3 days on plastic dishes expressed both flt-1 and KDR/flk-1. When the culture was prolonged to 10 days, the flt-1 mRNA expression was increased, whereas both KDR/flk-1 mRNA and protein expressions diminished. DNA and collagen syntheses were minimal in the cells cultured for 3 days, but marked in those cultured for 10 days. Addition of recombinant human VEGF to the culture medium did not change both syntheses but attenuated an increase of smooth muscle alpha-actin expression in the cells during culture on plastic dishes and also contraction of collagen gels on which the cells were cultured. We conclude that VEGF may inhibit contraction of hepatic stellate cells appearing during activation by culture, probably through attenuation of smooth muscle alpha-actin expression via upregulated VEGF receptor, flt-1.  相似文献

Abstract: Human glioma cell extracts were found to elicit a marked growth-promoting activity on human neuroblastoma cells. This activity was also detected in the extracts of neurofibroma type 1 (NF1; von Recklinghausen neurofibromatosis) comprising aberrant Schwann cell growth. The purified substance from the NF1 extracts by HPLC on ODS columns was identical to a pyrimidine nucleoside, uridine, the chemical structure of which was identified by gas chromatography-mass spectrometry. The authentic uridine showed a strong growth-promoting activity on human neuroblastoma cells. Other purine or pyrimidine nucleotides, their derivatives, and ribose sources for their syntheses were employed to test the activity; a purine nucleoside, adenosine, showed a stronger activity than uridine. The current study raises the possibility that human neuroblastoma cells may be affected by dysfunctions of the de novo pathway of both purine and pyrimidine nucleotide biosyntheses.  相似文献
