Summary A chemically defined medium was developed that supported the growth ofAgrobacterium radiobacter K84 and the production of agrocin 84. Various supplements were investigated for their effect on growth rate and production of agrocin 84 using a well-diffusion assay method. Mannitol was found to be a better substrate for growth ofA. radiobacter K84 compared to the other sugar alcohols and sugars tested. By contrast,d-fructose was the better substrate for the production of agrocin 84. Biotin supplementation stimulated production of agrocin 84 but did not eliminate the diauxic lag seen with the basal medium. The opine, octopine, inhibited growth ofA. radiobacter K84 and production of agrocin 84 as did coenzyme B12. By contrast, the cytokinin, isopentenyl adenosine, was marginally stimulatory to production, as was vitamin B12. Acetosyringone supplementation had a negligible effect on growth rate and production of agrocin 84.  相似文献

Agrobacterium radiobacter K84 is an effective, commercially applied, biological control agent for the plant disease crown gall, yet little is known about the survival and dissemination of K84. To trace K84 in the environment, spontaneous antibiotic-resistant mutants were used. Growth rates and phenotypes of streptomycin- or rifampin-resistant K84 were similar to those of the parental K84, except the rifampin-resistant mutant produced less agrocin 84 as determined by bioassay. K84 and a strain of Agrobacterium tumefaciens established populations averaging 10(5) CFU/g in the rhizosphere of cherry and persisted on roots for 2 years. K84 established rhizosphere populations between 10(4) and 10(6) CFU/g on cherry, ryegrass, and 11 other herbaceous plants. Populations of K84 declined substantially in fallow soil or water over a 16-week period. K84 was detected in the rhizosphere of ryegrass located up to 40 cm from an inoculum source, indicating lateral dissemination of K84 in soil. In gall tissue on cherry, K84 established populations of 10(5) CFU/g, about 10- to 100-fold less than that of the pathogen. These data demonstrate that K84 persists for up to 2 years in a field environment as a rhizosphere inhabitant or in association with crown gall tissue.  相似文献

Levels of root colonization by K84 and K1026 of Montclar and Nemaguard peach seedlings were similar during the 21 days of the experiment. Four strains of A. tumefaciens bv. 1 were used for soil inoculations in biological control experiments on GF677 and Adafuel peach x almond rootstocks; two were sensitive and two were resistant to agrocin 84. Both strains K84 and K1026 were very efficient in controlling the sensitive strains, but some tumors appeared with both treatments. In the biocontrol of resistant strains, no galls were observed in K1026-treated plants, but some K84-treated plants had galls. Recovery of agrobacteria from galls in experiments with sensitive and resistant strains showed that all of the isolates from the controls or K1026-treated plants and most of the isolates from K84-treated plants had the same characteristics as the inoculated strains. Nine isolates from the K84-treated plants growing in soil inoculated with one resistant strain were virulent and produced agrocin 84. These isolates had a plasmid that hybridized with a probe prepared with the BamHI C fragment from pAgK84. These results show the efficiency of K1026 in biocontrol of agrocin 84-sensitive and -resistant strains of A. tumefaciens and suggest the use of K1026 as a safer organism than K84 for biological control of crown gall.  相似文献

Rhizobium rhizogenes strain K84 is a commercial biocontrol agent used worldwide to control crown gall disease. The organism binds tightly to polypropylene substrate and efficiently colonizes root surfaces as complex, multilayered biofilms. A genetic screen identified two mutants in which these surface interactions were affected. One of these mutants failed to attach and form biofilms on the abiotic surface although, interestingly, it exhibited normal biofilm formation on the biological root tip surface. This mutant is disrupted in a wcbD ortholog gene, which is part of a large locus predicted to encode functions for the biosynthesis and export of a group II capsular polysaccharide (CPS). Expression of a functional copy of wcbD in the mutant background restored the ability of the bacteria to attach and form normal biofilms on the abiotic surface. The second identified mutant attached and formed visibly denser biofilms on both abiotic and root tip surfaces. This mutant is disrupted in the rkpK gene, which is predicted to encode a UDP-glucose 6-dehydrogenase required for O-antigen lipopolysaccharide (LPS) and K-antigen capsular polysaccharide (KPS) biosynthesis in rhizobia. The rkpK mutant from strain K84 was deficient in O-antigen synthesis and exclusively produced rough LPS. We also show that strain K84 does not synthesize the KPS typical of some other rhizobia strains. In addition, we identified a putative type II CPS, distinct from KPS, that mediates cell-surface interactions, and we show that O antigen of strain K84 is necessary for normal cell-cell interactions in the biofilms.  相似文献

The survival data showed that it is possible to preserve A. radiobacter cells on dry solid supports for a long time provided that the storage temperature is 4 degrees C and that the inoculation volume for 4 x 10(9) CFU g-1 is not less than 0.15 ml g of carrier-1. On the other hand, a substantial carrier water content was necessary for room temperature storage. Many materials proved to be suitable as microbial carriers; in some cases, vermiculite allowed long storage times comparable to those reported for peat or carboxymethyl cellulose, which are already employed in some commercial A. radiobacter K84 products. Furthermore, vermiculite assured full and immediate biological activity in the prevention of crown gall, showing that it is suitable for a new formulation of strain K84. A hypothesis to explain the different survival abilities in wet and dry conditions is presented.  相似文献

Cholinergic stimulation of chloride secretion involves the activation of a basolateral membrane potassium conductance, which maintains the electrical gradient favoring apical Cl efflux and allows K to recycle at the basolateral membrane. We have used transepithelial short-circuit current (I SC), fluorescence imaging, and patch clamp studies to identify and characterize the K channel that mediates this response in T84 cells. Carbachol had little effect on I SC when added alone but produced large, transient currents if added to monolayers prestimulated with cAMP. cAMP also enhanced the subsequent I SC response to calcium ionophores. Carbachol (100 m) transiently elevated intracellular free calcium ([Ca2+] i ) by 3-fold in confluent cells cultured on glass coverslips with a time course resembling the I sc response of confluent monolayers that had been grown on porous supports. In parallel patch clamp experiments, carbachol activated an inwardly rectifying potassium channel on the basolateral aspect of polarized monolayers which had been dissected from porous culture supports. The same channel was transiently activated on the surface of subconfluent monolayers during stimulation by carbachol. Activation was more prolonged when cells were exposed to calcium ionophores. The conductance of the inward rectifier in cell-attached patches was 55 pS near the resting membrane potential (–54 mV) with pipette solution containing 150 mm KCl (37°C). This rectification persisted when patches were bathed in symmetrical 150 mm KCl solutions. The selectivity sequence was 1 K > 0.88 Rb > 0.18 Na Cs based on permeability ratios under bi-ionic conditions. The channel exhibited fast block by external sodium ions, was weakly inhibited by external TEA, was relatively insensitive to charybdotoxin, kaliotoxin, 4-aminopyridine and quinidine, and was unaffected by external 10 mm barium. It is referred to as the KBIC channel based on its most distinctive properties (Ba-insensitive, inwardly rectifying, Ca-activated). Like single KBIC channels, the carbachol-stimulated I SC was relatively insensitive to several blockers on the basolateral side and was unaffected by barium. These comparisons between the properties of the macroscopic current and single channels suggest that the KBIC channel mediates basolateral membrane K conductance in T84 cell monolayers during stimulation by cholinergic secretagogues.We thank Dr. Marcel Crest (Laboratoire de Neurobiologie, CNRS, Marseille) for providing a sample of kaliotoxin. Wild-type Agrobacterium radiobacter strain 84 and its Tra- derivative K1026, used for biological control of crown gall disease, each contain the plasmid pAtK84b. It confers incompatibility to tumor-inducing (Ti) plasmids of pathogenic A. tumefaciens, thus preventing transfer of Ti plasmids into K84 and K1026, and the consequent development of pathogens resistant to the specific antibiotic, agrocin 84 produced by K84 and K1026. pAtK84b also resembles one group of Ti plasmids in its capacity for directing nopaline catabolism. A study of the DNA homology among pAtK84b, pTiC58, and pTiAch5 was carried out. pAtK84b was transferred by conjugation to a plasmidless recipient and, after isolation, was hybridized with Ti plasmid DNA. Areas of DNA homology were located on published maps of pTiC58 and pTiAch5, a restriction enzyme map of pAtK84b was constructed, and areas of homology with DNA of known genetic function were located on the map. Strong and extensive (over 50%) homology was found between pAtK84b and pTiC58 (nopaline catabolic, Noc), but much less between pAtK84b and pTiAch5 (octopine catabolic). There was no detectable homology between pAtK84b and the oncogenic T-DNA and virulence (Vir) regions of either Ti plasmid. The size of pAtK84b was 173 kb and the orientation of regions of identified gene function (Noc, incompatability/origin of replication, and conjugal transfer) on pTiC58 was matched by the locations of homologous areas on pAtK84b. It is concluded that pAtK84b may be a deletion product of a pTiC58-type plasmid which has been disarmed in the oncogenic T-DNA and Vir regions.  相似文献

The crown gall biocontrol agent strain K84 and three mutants derived from it, K1026 (Tra deletion mutant of pAgK84), K84 Agr (lacking pAgK84), and K1143 (lacking pAgK84 and pNoc), significantly reduced gall formation caused by two pathogenic strains resistant to agrocin 84 in peach × almond seedlings planted in infested soil. Cocolonization of roots by pathogenic and nonpathogenic strains was observed in these biocontrol experiments under field conditions. In spite of the efficient biocontrol observed, average populations consisting of 102 and 106 pathogenic agrobacteria per g of root were found 8 months after planting. The total numbers of pathogenic bacteria on roots were similar for plants treated with the biocontrol strains and for the untreated plants. Strain K84 and the genetically engineered organism K1026 survived at a level of 106 agrocin 84-producing bacteria per g of root. The population size of genetically engineered strain K1026 was not significantly different than the population size of wild-type strain K84 8 months after root inoculation. Strains K84 and K1026 controlled two pathogens resistant to agrocin 84 without reducing the total number of pathogenic bacteria in the root system. In addition, this study shows that some biological control activity of strain K84 against agrocin 84-resistant pathogens is independent of plasmids pAgK84 and pNoc.  相似文献

Both drought and freezing-thawing of stems induce a loss of hydraulic conductivity (percentage loss of conductivity [PLC]) in woody plants. Drought-induced PLC is often accompanied by physical damage to pit membranes, causing a shift in vulnerability curves (cavitation fatigue). Hence, if cavitated stems are flushed to remove embolisms, the next vulnerability curve is different (shifted to lower tensions). The 84K poplar (Populus alba × Populus glandulosa) clone has small vessels that should be immune from frost-induced PLC, but results demonstrated that freezing-thawing in combination with tension synergistically increased PLC. Frost fatigue has already been defined, which is similar to cavitation fatigue but induced by freezing. Frost fatigue caused a transition from a single to a dual Weibull curve, but drought-fatigued stems had single Weibull curves shifted to lower tensions. Studying the combined impact of tension plus freezing on fatigue provided evidence that the mechanism of frost fatigue may be the extra water tension induced by freezing or thawing while spinning stems in a centrifuge rather than direct ice damage. A hypothesis is advanced that tension is enhanced as ice crystals grow or melt during the freeze or thaw event, respectively, causing a nearly identical fatigue event to that induced by drought.Water transport in xylem conduits of trees occurs while water is under tension (negative pressure; Tyree and Zimmermann, 2002). The xylem water-transport system is vulnerable to cavitation and embolism because tensile water is metastable, so if a gas bubble appears in a conduit it will rapidly expand to fill the conduit whenever the fluid tension is 0.1 MPa or greater, where a tension of 0.1 MPa is equivalent to vacuum pressure. A cavitation event occurs whenever a tensile water column breaks, which results in a water vapor-filled void. Because of Henry’s law of gas solubility in water, this vapor void will eventually equilibrate with air at atmospheric pressure, at which point the conduit is fully embolized (Tyree and Zimmermann, 2002). Embolism has been identified as a limiting factor of primary production (Hubbard et al., 2001). As a result, tree growth and fitness are probably negatively impacted temporarily or seriously limited permanently if embolism is extensive (Christensen-Dalsgaard and Tyree, 2013).The two main factors causing cavitation and embolism are drought and frost (Mayr et al., 2003; Christensen-Dalsgaard and Tyree, 2013). Drought-induced cavitation is caused by the high xylem tension attributed to water stress. The high tension in the sap forces air bubbles into functional conduits from neighboring embolized ones through shared pit membranes (Jarbeau et al., 1995; Sperry et al., 1996; Hacke et al., 2001; Stiller and Sperry, 2002; Christman et al., 2012) according to an air-seeding mechanism (Sperry and Tyree, 1988; Cochard et al., 1992). Hence, the continuity of water flow is disrupted due to cavitation. Frost-induced cavitation, on the other hand, occurs when dissolved gases in the sap freeze out and create bubbles during ice formation because air is not soluble in ice (Mayr et al., 2003; Christensen-Dalsgaard and Tyree, 2013, 2014) but remains entrapped between ice crystals. Once the sap melts and tension is regenerated, the entrapped bubbles may expand to embolize the conduits instead of dissolving (Pittermann and Sperry, 2006). Current thinking is that freezing-induced embolism occurs when the tension exceeds a critical value determined by the surface tension of the bubbles, which mainly depends on the xylem water potential and the bubble radius (Yang and Tyree, 1992; Tyree et al., 1994; Hacke and Sperry, 2001). Larger bubbles may form in conduits with a larger diameter, so species with larger conduits are more vulnerable to frost-induced embolism (Langan et al., 1997; Davis et al., 1999; Pittermann and Sperry, 2006). Furthermore, enhanced loss of hydraulic conductivity (Kh) of trees may occur when stems are subjected to a combination of frost and drought causing low xylem water potential (Mayr et al., 2003; Willson and Jackson, 2006) and repeated freeze-thaw cycles (Sperry and Sullivan, 1992; Cox and Zhu, 2003; Mayr et al., 2003). However, even trees with small conduits are found to suffer severe embolism in winter (Sperry et al., 1988; Améglio et al., 2002) due mostly to freezing-drying of stems.Resilient species are those that suffer no significantly different cavitation resistance before and after a cavitation-refilling cycle (Hacke et al., 2001; Christensen-Dalsgaard and Tyree, 2013). In contrast, species that are weakened by cavitation or frost are said to suffer cavitation fatigue or frost fatigue. Cavitation or frost fatigue is quantified by how much the vulnerability curve is shifted before versus after a fatigue-inducing event, and it is typically reported as a shift in P50, which is either the pressure (negative value) or tension (positive value) that produces 50% loss of Kh. In the rest of this article, we will use T50 to indicate the tension at 50% loss of conductivity or Tx to indicate the tension that induces x% loss of conductivity. Vulnerability curves (VCs) are usually measured by a centrifuge technique (Alder et al., 1997; Cochard et al., 2005), but most researchers measure just five or six points to determine a VC. High-resolution VC curves with nine to 27 points per curve can be collected quickly using the Cochard rotor. Recent studies have successfully used high-resolution VC to characterize the detailed shape of VCs, revealing dual Weibull curves (e.g. r- and s-shaped or dual s-shaped curves; Cai et al., 2014; Wang et al., 2014a) because a complex shape to a VC cannot be identified with just a few points. Furthermore, we used a centrifuge to induce tension while simultaneously freezing in order to study the combined impact of tension and freezing-thawing on frost fatigue and freeze-thaw-induced embolism.In this article, we intend to investigate whether drought and freeze-thaw cycles could have an effect on the cavitation resistance in terminal shoots from adult trees of 84K poplar (Populus alba × Populus glandulosa), with high-resolution analysis of VCs and an artificial freeze-thaw simulation technique. Populus spp. are known to be water-demanding, drought-sensitive species with T50 ranging from 1.07 to 2.5 MPa (Fichot et al., 2015) and vulnerable to winter damage (Feng et al., 2010). Among poplars, 84K poplar is known by foresters to be relatively resistant to water stress, low temperature, diseases, and insects (Zhou et al., 2007). As the main afforestation species in Shaanxi, Gansu, and Qinghai Province, 84K poplar is of great ecological importance.  相似文献

A Ca2+-dependent actin-severing 84K Mr protein prepared from bovine aorta caused four-fold activation of smooth muscle actin-activated myosin ATPase at a 1/10(2) molar ratio to actin in the presence of tropomyosin and light chain kinase-calmodulin in a Ca2+-dependent manner, while it inhibited it markedly at a 1/5 molar ratio. Purified actin-tropomyosin filaments under the experimental ATPase conditions were distributed in a range of more than 10 micron in length and the addition of the 84K Mr protein changed the filament length to around 1 micron at a 1/10(2) molar ratio to actin or less than 50 nm at a 1/5 molar ratio in the presence of Ca2+. However, the apparent length of actin filaments alone does not appear to be responsible for the activation of ATPase activity, since in the absence of tropomyosin, the ATPase activation was much less in spite of actin filament length changes. These results indicate the possibility that the 84K Mr protein plays an important role with tropomyosin in at least in vitro smooth muscle actin-myosin interaction.  相似文献

We usedsingle-channel recording techniques to identify and characterize alarge-conductance,Ca2+-independentK+ channel in the colonicsecretory cell line T84. In symmetric potassium gluconate, this channelhad a linear current-voltage relationship with a single-channelconductance of 161 pS. Channel open probability(Po) wasincreased at depolarizing potentials. Partial substitution of bathK+ withNa+ indicated a permeability ratioof K+ toNa+ of 25:1. ChannelPo was reduced byextracellular Ba2+. Event-durationanalysis suggested a linear kinetic model for channel gating having asingle open state and three closed states: C3C2C1O.Arachidonic acid (AA) increased thePo of thechannel, with an apparent stimulatory constant(Ks)of 1.39 µM. Neither channel open time (O) nor the fast closed time(C1) was affected by AA. Incontrast, AA dramatically reduced mean closed time by decreasing bothC3 andC2. Thecis-unsaturated fatty acid linoleate increased Poalso, whereas the saturated fatty acid myristate and thetrans-unsaturated fatty acid elaidatedid not affectPo. Thus we determined the effect of thestretch-activated channel blockers amiloride and Gd3+ on theK+ channel after activation by AA.Amiloride (2 mM) on the extracellular side reduced single-channelamplitude in a voltage-dependent manner, whereasGd3+ (100 µM) had no effect onchannel activity. Activation of this K+ channel may be important duringstimulation of Cl secretionby agonists that use AA as a second messenger (e.g., vasoactiveintestinal polypeptide, adenosine) or during the volume regulatoryresponse to cell swelling.

TheCl secretory response ofcolonic cells to Ca2+-mediatedagonists is transient despite a sustained elevation of intracellular Ca2+. We evaluated the effects ofsecond messengers proposed to limit Ca2+-mediatedCl secretion on thebasolateral membrane,Ca2+-dependentK+ channel(KCa) in colonic secretorycells, T84. Neither protein kinase C (PKC) nor inositoltetrakisphosphate (1,3,4,5 or 3,4,5,6 form) affectedKCa in excised inside-out patches.In contrast, arachidonic acid (AA; 3 µM) potently inhibitedKCa, reducingNPo, the productof number of channels and channel open probability, by 95%. Theapparent inhibition constant for this AA effect was 425 nM. AAinhibited KCa in the presence ofboth indomethacin and nordihydroguaiaretic acid, blockers of thecyclooxygenase and lipoxygenase pathways. In the presence of albumin,the effect of AA on KCa wasreversed. A similar effect of AA was observed onKCa during outside-out recording.We determined also the effect of thecis-unsaturated fatty acid linoleate,the trans-unsaturated fatty acidelaidate, and the saturated fatty acid myristate. At 3 µM, all ofthese fatty acids inhibited KCa,reducing NPo by 72-86%. Finally, the effect of the cytosolic phospholipaseA2 inhibitorarachidonyltrifluoromethyl ketone(AACOCF3) on thecarbachol-induced short-circuit current(Isc) responsewas determined. In the presence ofAACOCF3, the peakcarbachol-inducedIsc response wasincreased ~2.5-fold. Our results suggest that AA generation inducedby Ca2+-mediated agonists maycontribute to the dissociation observed between the rise inintracellular Ca2+ evoked by theseagonists and the associatedCl secretory response.
