The interaction of the heat shock factor (HSF) with the heat shock element (HSE) was determined by a non-radioactive electrophoretic mobility shift assay, in order to analyze HSF regulation in Neurospora crassa. HSF binds to HSE under normal, non-stress conditions and is thus constitutively trimerized. Upon heat shock, the HSF-HSE complex shows a retarded mobility. This was also observed in Saccharomyces cerevisiae, where this mobility shift was shown to be due to HSF phosphorylation [Sorger and Pelham (1988) Cell 54, 855-864]. In N. crassa, HSE-dependent electrophoretic mobility shift is temperature- and time-dependent. Under normal growth conditions, the HSF is located in the cytoplasm as well as in the nucleus. In germinating conidia the HSF shows a retarded mobility typical for heat shock even at normal growth temperatures. No HSF-dependent mobility shift was detectable in aerial hyphae.  相似文献

The electrophoretic mobilities of Ehrlich ascites, sarcoma 37 ascites, mouse liver cells and their isolated nuclei were measured under similar environmental conditions. No differences in mobility were detected between cells and homologous nuclei from the same cell population and it was concluded that their surface charge densities were probably the same. The effect of neuraminidase on Ehrlich ascites and liver cells and nuclei was also determined; neuraminidase reduced the mobility of Ehrlich ascites cell nuclei as well as cells. The reduction in mobility of cells and nuclei prepared by a sucrose method was the same; however, the reduction in mobility of citric acid prepared nuclei was less than that of citric acid treated cells. The reduction in mobility of both liver cells and nuclei was small or insignificant. It is suggested that although cells and nuclei have similar electrophoretic mobilities, possibly different groups contribute to their surface charge.  相似文献

DNA synthesis intensity and spectral and fluorescent properties of leucemic, PHA-induced and intact normal mouse spleen cells and of nuclei isolated from these cells were investigated. The cell electrophoretic mobility and DNA-protein interaction in the nuclei were studied. Similarity in cell and nuclei fluorescence, fluorescence of the probe ANS conjugated with the cells, the electrophoretic mobility and tightness of DNA--protein interaction for leucemic and PHA--induced cells and also the similarity of the tightness of DNA--protein interaction for leucemic and normal intact cells were found inspite of the differences in DNA synthesis intensity and cell functional peculiarities.  相似文献

A study was made of the heat shock puff activity in salivary glands of Drosophila melanogaster larvae after 5 and 20 min treatments with anoxia (dipping into physiological solution), heat shock (37 degrees C), and simultaneously with both the agents. The simultaneous treatment with heat shock and anoxia, as well as treatment with anoxia only blocked the induction of heat shock puffs. They appeared 10-15 min after the treatment during recovery under aerobic conditions. There was a super-additive effect of the simultaneous treatment on the heat shock puffing duration. A specific regulation of the 93D locus was observed. The 93D puff was induced by a 5 min simultaneous treatment with anoxia and heat shock and, as a rule, was not induced by the analogous 20 min treatment. The role of anoxia in blocking heat shock puff induction under simultaneous effects of heat shock and anoxia is discussed.  相似文献

When Drosophila salivary glands are disrupted in cytoplasm from heat-shocked Drosophila Kc cells in culture, puffs are induced in the polytene nuclei at the heat shock loci. The in vitro response increases with time of incubation, reaching in vivo levels after 2 hr, and is sensitive to α-amanitin. Cytoplasm from control Kc cells induces poorly, but prolonged exposure to air or the addition of hydrogen peroxide stimulates induction to the levels observed with cytoplasm from heat-shocked Kc cells. Autoradiography confirms that the heat shock loci induced to puff in vitro are the most actively transcribing sites in the genome.  相似文献

The chromatin structure of the ribosomal DNA in Xenopus laevis was studied by micrococcal nuclease digestions of blood, liver and embryonic cell nuclei. We have found that BglI-restricted DNA from micrococcal nuclease-digested blood cell nuclei has an increased electrophoretic mobility compared to the undigested control. Micrococcal nuclease digestion of liver cell nuclei causes a very slight shift in mobility, only in the region of the spacer containing the "Bam Islands". In contrast, the mobility of ribosomal DNA in chromatin of embryonic cells, under identical digestion conditions, remains unaffected by the nuclease activity. Denaturing gels or ligase action on the nuclease-treated DNA abolishes the differences in the electrophoretic mobility. Ionic strength and ethidium bromide influence the relative electrophoretic migration of the two DNA fragment populations, suggesting that secondary structure may play an important role in the observed phenomena. In addition, restriction analysis under native electrophoretic conditions of DNA prepared from blood, liver and embryonic cells shows that blood cell DNA restriction fragments always have a faster mobility than the corresponding fragments of liver and embryo cell DNA. We therefore propose that nicking activity by micrococcal nuclease modifies the electrophoretic mobility of an unusual DNA conformation, present in blood cell, and to a lesser extent, in liver cell ribosomal chromatin. A possible function for these structures is discussed. The differences of the ribosomal chromatin structures in adult and embryonic tissues may reflect the potential of the genes to be expressed.  相似文献

At least four proteins of 70,000 to 75,000 molecular weight (70-75K) were synthesized from mRNA which hybridized with a cloned heat shock gene previously shown to be localized to the 87A and 87C heat shock puff sites. These in vitro-synthesized proteins were indistinguishable from in vivo-synthesized heat shock-induced proteins when analyzed on sodium dodecyl sulfate-polyacrylamide gels. A comparison of the pattern of this group of proteins synthesized in vivo during a 5-min pulse or during continuous labeling indicates that the 72-75K proteins are probably not kinetic precursors to the major 70K heat shock protein. Partial digestion products generated with V8 protease indicated that the 70-75K heat shock proteins are closely related, but that there are clear differences between them. The partial digestion patterns obtained from heat shock proteins from the Kc cell line and from the Oregon R strain of Drosophila melanogaster are very similar. Genetic analysis of the patterns of 70-75K heat shock protein synthesis indicated that the genes encoding at least two of the three 72-75K heat shock proteins are located outside of the major 87A and 87C puff sites.  相似文献

Using indirect immunofluorescence visualization techniques we investigated the distribution of RNA polymerase B (or II) and histone H1 at heat shock puff loci in Drosophila melanogaster polytene chromosomes at different times during and after heat shock. After heat treatments of from 5 to 45 min, the heat shock puff displayed intense fluorescence when stained for RNA polymerase B, but relatively little fluorescence when stained for histone H1. Returning heat shocked larvae to room temperature resulted in the appearance of a distinctive pattern of RNA polymerase-associated fluorescence in the heat shock puff at 87C, presumably reflecting events associated with the inactivation and regression of this puff. Large differences observed in the apparent RNA polymerase B content of puffs of similar size suggest that the interaction of RNA polymerase B with chromosomal loci does not depend on simply the state of condensation or decondensation of the chromatin.  相似文献

Effect of mutations at the ebony or black locus on induction of heat shock puffs in polytene nuclei of salivary glands ofDrosophila melanogaster larvae were examined by [3H]uridine autoradiography. The levels of-alanine in the body are known to be increased by mutation at the ebony locus but decreased by mutation at the black locus. The presence of mutant allele/s at either locus in the homo- or heterozygous condition prevented induction of the 93D puff by heat shock. Elimination of the mutant allele at the ebony or black locus by recombination or by reversion of a P element insertion mutant allele of ebony restored the heat shock inducibility of the 93D puff. In vivo or in vitro administration of excess-alanine to salivary glands of wild-type larvae also resulted in the 93D site being refractory to heat shock induction. In agreement with earlier results, noninduction of the 93D puff during heat shock due to the-alanine effect was accompanied by unequal puffing of the 87A and 87C loci. The selective inducibility of the 93D puff by benzamide was not affected by ebony or black mutations or by excess-alanine in wild-type larvae  相似文献

It is reported that chromatin can be prepared from highly purified polytene nuclei from the salivary glands of third instar larvae of Drosophila hydei; such chromatin differs from that of diploid nuclei mainly by deficiencies in certain nonhistone chromosomal proteins. It is suggested that these proteins are important components of constitutive heterochromatin, which is severely underrepresented in polytene chromosomes. Chromosome morphology, including the pattern of induced puffs, is maintained throughout the mass isolation of glands and sucrose gradient purification of nuclei, as indicated by studies on temperature-shocked and control larvae. No significant alteration in the chromosomal proteins following puff induction by heat shock could be detected on analysis of the isolated protein fractions by disc gel electrophoresis. More sensitive techniques must be developed to study the apparent rearrangement or accumulation of protein at puff sites, and to elucidate the role of this protein in gene activation.  相似文献
