Isolation and characterization of methanogenic bacteria from rice paddies   总被引:3,自引:0,他引:3  
Abstract Enrichment cultures for H2-CO2, methanol- or acetate-utilizing methanogens were prepared from two rice field soil samples. All the cultures except one acetate enrichment showed significant methane production. Pure cultures of Methanobacterium - and Methanosarcina -like organisms were isolated from H2-CO2 and methanol enrichment cultures, respectively, and were characterized for various nutritional and growth conditions. The organisms had an optimal pH range of 6.4–6.6 and a temperature optimum of 37°C. The Methanobacterium isolates were able to utilize H2-CO2 but no other substrates as sole energy source, while the Methanosarcina isolates were able to utilize methanol, methylamines or H2-CO2 as sole energy sources. Both Methanobacterium isolates and one isolate of Methanosarcina were able to use dinitrogen as the sole source of nitrogen for growth. The isolates used several sulfur compounds as sole sources of sulfur.  相似文献

Abstract The effect of temperature on CH4 production, turnover of dissolved H2, and enrichment of H2-utilizing anaerobic bacteria was studied in anoxic paddy soil and sediment of Lake Constance. When anoxic paddy soil was incubated under an atmosphere of H2/CO2, rates of CH4 production increased 25°C, but decreased at temperatures lower than 20°C. Chloroform completely inhibited methano-genesis in anoxic paddy soil and lake sediment, but did not or only partially inhibit the turnover of dissolved H2, especially at low incubation temperatures. Cultures with H2 as energy source resulted in the enrichment of chemolithotrophic homoacetogenic bacteria whenever incubation temperatures were lower than 20°C. Hydrogenotrophic methanogens could only be enriched at 30°C from anoxic paddy soil. A homoacetogen  相似文献

S. MARIN, V. SANCHIS, I. VINAS, R. CANELA AND N. MAGAN. 1995. The effect of different water activities ( a w, 0.968, 0.956, 0.944, 0.925) and temperature (25°C and 30°C) on colonization and production of fumonisin B1 (FB1) and B2 (FB2) on sterile layers of maize by Fusarium proliferatum and F. moniliforme isolates was determined over periods of 6 weeks. Generally, both F. moniliforme and F. proliferatum grew faster with increasing a w and best at 30°C. All three isolates produced more FB 1 than FB2 regardless of a w or temperature. Very little FB1 and FB2 were produced at 0.925 a w, with maximum produced at 0.956 and 0.968 a w at both temperatures tested. Most FB1 and FB2 were produced by F. moniliforme (25N), followed by F. proliferatum isolates (73N and 131N). At all a w levels and both temperatures there was an increase in FB1 and FB2 concentration with time. Statistical analyses of a w, temperature, time, two- and three-way interactions showed some significant differences between isolates and FB1 and FB2 production.  相似文献

Zoige wetland of Tibetan plateau is characterized by being located at a low latitude (33°56'N, 102°52'E) region and under the annual temperature around 1°C. Previous studies indicated that Zoige wetland was one of the CH4 emission centres in Qinghai-Tibetan plateau; in this study, the methanogen community in this low-latitude wetland was analysed based on the homology of 16S rRNA and mcrA genes retrieved from the soil. The results indicated that members of Methanosarcinales and Methanomicrobiales constituted the majority of methanogens, and a novel uncultured methanogen cluster, Zoige cluster I (ZC-I) affiliated to Methanosarcinales , could be dominant. Using quantitative polymerase chain reaction (qPCR) assay, ZC-I methanogens were estimated to be 107 cells per gram of soil, accounting for about 30% of the total Archeae . By combining culturable enrichment with qPCR assay, the quantity of ZC-I methanogens in the methanogenic enrichment with acetate, H2/CO2, methanol or trimethylamine was determined to increase to 108 cells ml−1, but not with formate, which indicated that ZC-I methanogens could use the four methanogenic substrates. The growth rates at 30°C and 15°C were not pronounced different, implying ZC-I to be the cold-adaptive methanogens. The broad substrate spectrum identified the ZC-I methanogens to be a member of Methanosarcinaceae , and could represent a novel sub-branch specifically inhabited in cold ecosystems. Fluorescence in situ hybridization (FISH) images also visualized ZC-I methanogens the sarcina-like aggregate of the spherical cells. The prevalence and flexibility in substrate utilization and growth temperature suggested ZC-I methanogens to be an important player in the methanogenesis of Zoige wetland.  相似文献

Microsclerotium formation by six isolates of Verticillium dahliae was studied at different temperatures both in vitro and in Arabidopsis thaliana . In vitro mycelial growth was optimal at 25°C, but microsclerotium formation was greatest at 20°C (two isolates) or 15–20°C (one isolate). Seedlings of A. thaliana were root-dipped in a conidial suspension, planted, and either placed at 5, 10, 15, or 25°C, or left at 20°C until the onset of senescence, after which some of the plants were placed at 5, 10, 15, or 25°C. The amount of microsclerotia per unit of shoot weight was assessed in relation to isolate and temperature. The optimal temperature for production of microsclerotia was 15–25°C. Two isolates each produced about 10 times more microsclerotia than each of the other four isolates. For these isolates, high R 2adj.-values of 0.77 and 0.66 were obtained, with temperature and its square as highly significant (P   < 0.001) independent variables. R 2adj.-values for the other isolates varied between 0.28 and 0.39. Moving plants to different temperatures at the onset of senescence led to microsclerotial densities that were intermediate between densities on plants that had grown at constantly 20°C and plants grown at other temperatures. This suggests that vascular colonization rate and rate of microsclerotium formation are similarly affected by temperature. The senescence rate of plants appeared unimportant except for plants grown at 25°C, which showed the highest amounts of microsclerotia per unit of plant weight in the most rapidly senescing plants.  相似文献

ABSTRACT. One of the two methanogenic endosymbionts of the giant sapropelic amoeba Pelomyxa palustris was isolated in pure culture. The cells were slender non-motile rods (3 × 0.4 μm), sometimes occurring in chains of 3–4 cells. Ultrathin sections revealed a Gram-positive cell wall and conically pointed ends with mesosome-like structures in the cytoplasm. The isolate had a generation time of 10 and 12 h during growth on H2/CO2 and formate, respectively. The optimum growth temperature was 40°C and the optimum pH was 7.8. The G+C content of its DNA was found to be 37.7% mole percent. The isolate was identified as Methanobacterium formicicum .  相似文献

The production and stability of pediocin N5p from Pediococcus pentosaceus , isolated from wine, were examined in grape juice medium. Maximum growth and higher titre (4000 U ml-1) were observed at a initial pH of 7·5 and 30°C. The activity of the inhibitory substance was stable between pH values from 2·0 to 5·0 at 4° and 30°C. At pH 10·0 it was completely inactivated. When submitted to 30 min at 80°, 100° and 115°C, maximal stability was observed at pH 2·0. Ethanol up to 10% did not affect pediocin activity at acid pH, nor did 40–80 mg 1-1 SO2, independently or combined with different ethanol concentrations, affect inhibitory activity.  相似文献

Ananas comosus L. (Merr.) (pineapple) was grown at three day/night temperatures and 350 (ambient) and 700 (elevated) μ mol mol–1 CO2 to examine the interactive effects of these factors on leaf gas exchange and stable carbon isotope discrimination ( Δ ,‰). All data were collected on the youngest mature leaf for 24 h every 6 weeks. CO2 uptake (mmol m–2 d–1) at ambient and elevated CO2, respectively, were 306 and 352 at 30/20 °C, 175 and 346 at 30/25 °C and 187 and 343 at 35/25 °C. CO2 enrichment enhanced CO2 uptake substantially in the day in all environments. Uptake at night at elevated CO2, relative to that at ambient CO2, was unchanged at 30/20 °C, but was 80% higher at 30/25 °C and 44% higher at 35/25 °C suggesting that phosphoenolpyruvate carboxylase was not CO2-saturated at ambient CO2 levels and a 25 °C night temperature. Photosynthetic water use efficiency (WUE) was higher at elevated than at ambient CO2. Leaf Δ -values were higher at elevated than at ambient CO2 due to relatively higher assimilation in the light. Leaf Δ was significantly and linearly related to the fraction of total CO2 assimilated at night. The data suggest that a simultaneous increase in CO2 level and temperature associated with global warming would enhance carbon assimilation, increase WUE, and reduce the temperature dependence of CO2 uptake by A. comosus .  相似文献

Oxygen consumption (o2) and respiratory variables were measured in the Prochilodontid fish, Prochilodus scrofa exposed to graded hypoxia after changes in temperature. The measurements were performed on fish acclimated to 25°C and in four further groups also acclimated to 25°C and then changed to 15, 20, 30 and 35°C. An increase in o2 occurred with rising temperature, but at each temperature o2 was kept constant over a wide range of O2 tensions of inspired water ( Pi o2). The critical oxygen tensions ( Pc o2) were Pi o2= 22 mmHg for 25°C acclimated specimens and after transfer from 25°C to 15, 20, 30 and 35°C the Pc o2 changed to Pi o2= 28, 22, 24 and 45 mmHg, respectively. Gill ventilation ( G ) increased or decreased following the changes in o2 as the temperature changed and was the result of an accentuated increase in breath frequency. During hypoxia the increases in G were characterized by larger increases in breath volume. Oxygen extraction was kept almost constant at about 63% regardless of temperature and ambient oxygen tensions in normoxia and moderate hypoxia ( P o2∼70 mmHg). P. scrofa showed high tolerance to hypoxia after abrupt changes in temperature although its survival upon transfer to 35°C could become limited by the capacity of ventilatory mechanisms to alleviate hypoxic stress.  相似文献

We investigated the combined effects of pressure, temperature, pH, initial spore concentration and the presence of nisin on the survival of spores of Bacillus coagulans. Spores were more sensitive to pressure both at lower pH and at higher treatment temperatures. An additional 1.5-log10 reduction in cfu ml-1 was observed when pH was lowered from 7.0 to 4.0 during pressurization at 400 Mpa and 45°C. A 4-log10 cfu ml-1 reduction was observed when the temperature was increased from 25°C to 70°C during pressurization at 400 Mpa. The spores were sensitive to nisin at concentrations as low as 0.2 IU ml-1. At least a 6-log10 reduction was generally achieved with pressurization at 400 Mpa in pH 4.0 buffer at 70°C for 30 min when plated in nutrient agar containing 0.8 IU ml-1 nisin.  相似文献

Abstract Acremonium persicinum grown in batch culture with ammonium tartrate as the nitrogen source possessed an NADP+-dependent glutamate dehydrogenase and a glutamine synthetase. Glutamate synthase was not detected under the culture conditions used. Kinetic studies of the NADP+-dependent glutamate dehydrogenase at 25°C and pH 7.6 revealed an apparent K m of 3.2 × 10−4 M for 2-oxoglutarate and an apparent K m of 1.0 × 10−5 M for ammonium ions, with corresponding apparent V max values of 0.089 and 0.13 μmol substrate converted/min/mg of protein, respectively. Glutamine synthetase was measured by the γ-glutamyl transferase reaction at 30°C and pH 7.55. This transferase reaction of glutamine synthetase had a higher rate at 30°C than at 25°C or 37°C.  相似文献

The soil temperature in flooded Italian rice fields is generally lower than 30°C. However, two temperature optima at ≈ 41°C and 50°C were found when soil slurries were anoxically incubated at a temperature range of 10–80°C. The second temperature optimum indicates the presence of thermophilic methanogens in the rice field soil. Experiments with 14C-labelled bicarbonate showed that the thermophilic CH4 was exclusively produced from H2/CO2. Terminal restriction fragment length polymorphism (T-RFLP) of archaeal SSU rRNA gene fragments revealed a dramatic change in the archaeal community structure at temperatures > 37°C, with the euryarchaeotal rice cluster I becoming the dominant group (about 80%). A clone library of archaeal SSU rRNA gene fragments generated at 49°C was also dominated (10 out of 11 clones) by rice cluster I. Our results demonstrate that Italian rice field soil contains thermophilic methanogenic activity that was most probably a result of members of the as yet uncultivated euryarchaeotal rice cluster I.  相似文献

Portions of skinless chicken breast meat (pH 5·8) were inoculated with a strain of Listeria monocytogenes and stored at 1, 6 or 15°C in (1) aerobic conditions; (2) 30% CO2+ air; (3) 30% CO2+ N2; and (4) 100% CO2. When samples were held at 1°C the organism failed to grow under any of the test conditions, despite marked differences between treatments in spoilage rate and ultimate microflora. At 6°C counts of L. monocytogenes increased ca 10-fold in aerobic conditions before spoilage of the meat, but only when the inoculum culture was incubated at 1°C rather than 37°C. In CO2 atmospheres growth of L. monocytogenes was inhibited on meat held at 6°C, especially under 100% CO2. By contrast, storage at 15°C led to spoilage of the meat within 2 d, in all gaseous environments, and listeria levels increased up to 100-fold. Differences in the behaviour of L. monocytogenes on poultry and red meats are discussed.  相似文献

S ummary : The heat resistance of spores of Bacillus coagulans strain Sp. 33, isolated from canned chopped spinach, has been determined. It appeared to be higher in 1/40 M phosphate than in 1/15 M phosphate and higher at pH 6°8 than at pH 6°0. In 1/40 M phosphate buffer of pH 6°8 the calculated D 110 was 2°4 min. In chopped spinach of pH 6°2 the calculated D 110 was 0°84 min. In addition a simple and reliable method of assessing the thermal resistance of micro-organisms in thermal death time tubes is reported.  相似文献

Spontaneous locomotor activity of cod Gadus morhua maintained at 6° C tripled from February to May. In contrast, locomotor activity of cod held at 2° C was significantly lower than at 6° C (between 25 and 65% lower) and the seasonal increase was smaller. Plasma levels of both thyroxine (T4) and triiodothyronine (T3) did not differ between 2 and 6° C. T4 injection increased locomotor activity by 10% for both temperature regimes. These data indicate that low water temperature reduces locomotor activity associated with migration in cod and that thyroid hormones are not involved in this decrease. This study provides a possible mechanism through which cold waters may affects migration and distribution of cod via its Effects on locomotor activity and swimming speed.  相似文献

Abstract During the first stage of the preparation of mushroom compost oxygen is believed to be readily available. However we measured methane in the evoking air above the compost piles and were able to isolate thermophilic methanogenic bacteria from this compost. The isolates grow only on H2 and CO2 as energy and carbon source and do not require complex factors for growth. On the basis of nutritional and morphological characteristics these methanogens were identified as strains of Methanobacterium thermoautotrophicum .  相似文献

Abstract Alkali-tolerant Aspergillus fischeri Fxn1 produced two extracellular xylanases. The major xylanase ( M r 31000) was purified to electrophoretic homogeneity by ammonium sulfate precipitation, anion exchange chromatography and preparatory PAGE. Xylose was the major hydrolysis product from oat spelt and birch wood xylans. It was completely free of cellulolytic activities. The optimum pH and temperature were 6.0 and 60 °C, respectively. pH stability ranged from 5 to 9.5 and the t1 / 2 at 50 °C was 490 min. It had a K m of 4.88 mg ml−1and a V max of 588 μmol min−1 mg−1. The activity was inhibited (95%) by AlCl3 (10 mM). This enzyme appears to be novel and will be useful for studies on the mechanism of hydrolysis of xylan by xylanolytic enzymes.  相似文献

Bifidobacterium adolescentis Int-57 isolated from human faeces produced extracellular amylase. The enzyme was purified from the culture supernatant fluids by ammonium sulphate precipitation, gel-filtration chromatography (Sephadex-G-75), ion-exchange chromatography (CM-cellulose) and FPLC. SDS-PAGE of the purified enzyme revealed a major band with an apparent molecular weight of 66 kDa. The pI was 5·2. Enzyme activity was optimal at 50°C, and at pH 5·5. The enzyme was stable at 20–40°C, and at pH 5–6 with a K m value of 2·4 g l−1 soluble starch. The activation energy was 42·3 kJ mol−1. The enzyme was significantly inhibited by maltose (10%), glucose (10%), Cu2+ (5 mmol l−1), Zn2+ (5 mmol l−1), N- bromosuccinimide (5 mmol l−1), EDTA (5 mmol l−1), I2 (1 mmol l−1) and activated by β-mercaptoethanol (10 mmol l−1).  相似文献

At four temperatures ( T= 15, 20, 25 and 28° C) swimming performance of Dicentrarchus labrax was significantly correlated with total length (23–43 mm L T); r2=0.623–0.829). The relative critical swimming speed ( RU crit= U crit L T−1), where U crit is the critical swimming speed, was constant throughout the L T range studied. The significant effect of temperature on the relative critical swimming speed was described binomially: RU crit=−0.0323T2+ 1.578 T −10.588 (r2=1). The estimated maximum RU crit (8.69 L T s−1) was achieved at 24.4° C, and the 90% performance level was estimated between 19.3 and 29.6° C.  相似文献

Abstract. Cyperus longus L. , which has a widespread but disjunct distribution throughout Europe and extends northwards into Britain, was found to be a C4 species based upon its Kranz leaf anatomy, low CO2 compensation point and the labelling of malate as an early product of 14CO2 fixation. The photosynthetic characteristics of C. longus are similar to many other C4 species with a high maximum rate of photosynthesis (> 1.5 mg CO2 m −2 s −1) and a relatively high temperature optimum (30–35°C), but unlike many C4 species the rate of photosynthesis does not decline rapidly below the optimum temperature and a substantial rate (0.6 mgCO2 m−2s−1)occursat 15°C. Leaf extension is very slow at 15°C and shows a curvilinear response to temperatures between 15 and 25°C. Leaves extend at a rate of almost 4 cm d−1 at 25°C.  相似文献
