Bursicon is an insect neuropeptide hormone that is secreted from the central nervous system into the hemolymph and initiates cuticle tanning. The receptor for bursicon is encoded by the rickets (rk) gene and belongs to the G protein-coupled receptor (GPCR) superfamily. The bursicon and its receptor regulate cuticle tanning as well as wing expansion after adult eclosion. However, the molecular action of bursicon signaling remains unclear. We utilized RNA interference (RNAi) and microarray to study the function of the bursicon receptor (Tcrk) in the model insect, Tribolium castaneum. The data included here showed that in addition to cuticle tanning and wing expansion reported previously, Tcrk is also required for development and expansion of integumentary structures and adult eclosion. Using custom microarrays, we identified 24 genes that are differentially expressed between Tcrk RNAi and control insects. Knockdown in the expression of one of these genes, TC004091, resulted in the arrest of adult eclosion. Identification of genes that are involved in bursicon receptor mediated biological processes will provide tools for future studies on mechanisms of bursicon action.  相似文献

Background  

The protective external cuticle of insects does not accommodate growth during development. To compensate for this, the insect life cycle is punctuated by a series of molts. During the molt, a new and larger cuticle is produced underneath the old cuticle. Replacement of the smaller, old cuticle culminates with ecdysis, a stereotyped sequence of shedding behaviors. Following each ecdysis, the new cuticle must expand and harden. Studies from a variety of insect species indicate that this cuticle hardening is regulated by the neuropeptide bursicon. However, genetic evidence from Drosophila melanogaster only supports such a role for bursicon after the final ecdysis, when the adult fly emerges. The research presented here investigates the role that bursicon has at stages of Drosophila development which precede adult ecdysis.  相似文献

The mosquito Anopheles gambiae is an important vector for malaria, which is one of the most serious human parasitic diseases in the world, causing up to 2.7 million deaths yearly. To contribute to our understanding of A. gambiae and to the transmission of malaria, we have now cloned four evolutionarily related G protein-coupled receptors (GPCRs) from this mosquito and expressed them in Chinese hamster ovary cells. After screening of a library of thirty-three insect or other invertebrate neuropeptides and eight biogenic amines, we could identify (de-orphanize) three of these GPCRs as: an adipokinetic hormone (AKH) receptor (EC50 for A. gambiae AKH, 3 × 10−9 M), a corazonin receptor (EC50 for A. gambiae corazonin, 4 × 10−9 M), and a crustacean cardioactive peptide (CCAP) receptor (EC50 for A. gambiae CCAP, 1 × 10−9 M). The fourth GPCR remained an orphan, although its close evolutionary relationship to the A. gambiae and other insect AKH receptors suggested that it is a receptor for an AKH-like peptide. This is the first published report on evolutionarily related AKH, corazonin, and CCAP receptors in mosquitoes.  相似文献

BACKGROUND: At the end of each molt, insects shed their old cuticle by performing the ecdysis sequence, an innate behavior consisting of three steps: pre-ecdysis, ecdysis, and postecdysis. Blood-borne ecdysis-triggering hormone (ETH) activates the behavioral sequence through direct actions on the central nervous system. RESULTS: To elucidate neural substrates underlying the ecdysis sequence, we identified neurons expressing ETH receptors (ETHRs) in Drosophila. Distinct ensembles of ETHR neurons express numerous neuropeptides including kinin, FMRFamides, eclosion hormone (EH), crustacean cardioactive peptide (CCAP), myoinhibitory peptides (MIP), and bursicon. Real-time imaging of intracellular calcium dynamics revealed sequential activation of these ensembles after ETH action. Specifically, FMRFamide neurons are activated during pre-ecdysis; EH, CCAP, and CCAP/MIP neurons are active prior to and during ecdysis; and activity of CCAP/MIP/bursicon neurons coincides with postecdysis. Targeted ablation of specific ETHR ensembles produces behavioral deficits consistent with their proposed roles in the behavioral sequence. CONCLUSIONS: Our findings offer novel insights into how a command chemical orchestrates an innate behavior by stepwise recruitment of central peptidergic ensembles.  相似文献

Using the adult Calliphora bioassay, we found that the tanning hormone, bursicon, is present in the blood of pupal and adult Tenebrio only at the time of ecdysis, when it is released massively from the thoracic and abdominal central nervous system. The hormone's half life in the blood is short (about 1–2 h). Contrary to the findings of other workers, we could find no evidence for the presence of the hormone in the haemolymph during pharate adult development, before ecdysis begins. When newly ecdysed pupae were ligated about the neck, adult development of the thorax and abdomen proceeded normally, but postecdysial tanning of the adult cuticle was almost completely prevented. This failure to tan was not due to lack of bursicon as the hormone was released normally in the ligated animals at the time of ecdysis. This suggests that a pre-ecdysial signal may be required for the development of epidermal competence to respond to bursicon.  相似文献

A simple and rapid extraction procedure was developed to determine simultaneously the molting hormone (MH) and juvenile hormone (JH) activity in a single insect tissue sample. From the onset of the last larval stage to adult eclosion of the greater wax moth, Galleria mellonella, three JH peaks were noted: at the time of the sixth larval ecdysis, 1 day before the seventh larval ecdysis, and at the time of adult eclosion. Three MH peaks were recorded for the male: at 1 day before the sixth larval ecdysis, 1 day before the seventh larval ecdysis, and 2 days after pupation. In the female, a fourth peak was shown at the time of adult eclosion. This fourth peak exhibits the highest molting hormone activity of all samples, 1600 Musca units/g of fresh tissue or an equivalent of 5.6 μg/g of ecdysterone. Eighty per cent of this MH accumulated in the ovary. The significance of MH and JH titers as related to the endocrine regulation of development is discussed in the light of this finding.  相似文献

Daily light and temperature cycles entrain adult eclosion rhythms in many insect species, but little is known about their interaction. We studied this problem in the onion fly, Delia antiqua. Pupae were subjected to various combinations of a photoperiod of 12L:12D and thermoperiods. The thermoperiods consisted of 12 h warm phase (W) and 12 h cool phase (C), giving a mean temperature of 25 °C with different temperature steps of 8, 4 and 1 °C. As the phase relation of the two Zeitgebers was varied, the phase of eclosion rhythm was shifted, depending on the phase angle with the light cycle and the amplitude of the temperature cycle. When the temperature step in the thermoperiod was 8 °C (WC 29:21 °C), the eclosion rhythm was entrained mainly to thermoperiod rather than photoperiod. In the regime with a 4 °C temperature step (WC 27:23 °C), both thermoperiod and photoperiod affected eclosion rhythm, and a phase jump of the eclosion rhythm occurred when the warm phase of thermoperiod was delayed 15-18 h from light-on. In regimes with a 1 °C temperature step (WC 25.5:24.5 °C), the eclosion rhythm was completely entrained to photoperiod. The observed interacting effect of light and temperature cycle on the eclosion rhythm in D. antiqua can be explained by the two-oscillator model proposed by Pittendrigh and Bruce (1959).  相似文献

Light and electron microscope observations have been made on the perisympathetic organs (P.O.) situated on the dorsal somatic abdominal nerves in pupae of Tenebrio molitor.Tests carried out with extracts of P.O. using a bioassay method on pupae of Tenebrio at various developmental stages indicate that these organs have an important role in the storage and release of the tanning hormone, bursicon. The P.O. extracts from the abdominal segments (2 to 5) of 5-day-old pupae were found effective but not those from pharate adult (2 days before ecdysis). The storage and release of bursicon occur later from the P.O. of the posterior abdominal segments (6 to 8).The activity of the blood begins to be effective in the pharate adult 2 days before ecdysis, reaches a maximum 1 hr after the adult ecdysis, and decreases thereafter.This study carried out on the various parts of the nervous system shows that all these parts are effective at a certain stage of the pupal and adult development following an antero-posterior gradient. In the cephalic region, this activity is early and long-lasting, but it appears at later stages and only for a short time in the thorax and abdomen. This phenomenon indicates that bursicon is either passed along the ventral nervous chain or it is synthesized in different parts of the nervous system at successive developmental stages. In the present paper, we have tried to give a comprehensive summary of the dynamics of bursicon in insects.  相似文献

Resistance to pyrethroid insecticides in the malaria vector Anopheles gambiae is a major threat to malaria control programmes. Cytochome P450-mediated detoxification is an important resistance mechanism. CYP6M2 is over-expressed in wild populations of permethrin resistant A. gambiae but its role in detoxification is not clear. CYP6M2 was expressed in Escherichia coli and a structural model was produced to examine its role in pyrethroid metabolism. Both permethrin and deltamethrin were metabolized. Rates were enhanced by A. gambiae cytochrome b5 with kinetic parameters of KM = 11 ± 1 ??M and kcat = 6.1 ± 0.4 per min for permethrin (1:1 cis-trans) and KM = 2.0 ± 0.3 ??M and kcat = 1.2 ± 0.1 per min for deltamethrin. Mass spectrometry and NMR analysis identified 4′-hydroxy deltamethrin and hydroxymethyl deltamethrin as major and minor deltamethrin metabolites respectively. Secondary breakdown products included cyano(3-hydroxyphenyl)methyl deltamethrate and deltamethric acid. CYP6M2 was most highly transcribed in the midgut and Malpighian tubules of adult A. gambiae, consistent with a role in detoxification. Our data indicates that CYP6M2 plays an important role in metabolic resistance to pyrethroids and thus an important target for the design of new tools to combat malaria.  相似文献

The eclosion of the adult Manduca sexta moth is followed by a wave of cell death that eliminates up to 50% of the neurons of the central nervous system within the first few days of imaginal life. While the identity of some of the dying motoneurons has been established, that of most doomed neurons is unknown. Here, we show that the dying cells include peptidergic neurons involved in the control of ecdysis behavior. These cells belong to a small population of 50 neurons that express crustacean cardioactive peptide (CCAP), a potent regulator of the ecdysis motor program, and show increases in cyclic 3′,5′-guanosine monophosphate at each ecdysis. First, we describe new markers for these neurons and show that they are expressed in these CCAP-immunoreactive neurons in a complex temporal pattern during development. We then show that these neurons die within 36 h after adult eclosion, the last performance of ecdysis behavior in the life of the animal, via the active, genetically determined process of programmed cell death. SYNOPSIS. The end of the molting process in the tobacco hornwormincludes the rapid digestion of the old cuticle, molting fluidresorption, ecdysis of the old cuticle, and expansion and hardeningof the new cuticle. The coordination of these processes is accomplishedby three hormones. Each ecdysis during the life of Manduca appearsto be triggered by eclosion hormone. Depending on developmentalstage, the hormone comes either from the brain-corpora cardiacacomplex or from the chain of ventral ganglia. The neural programstriggered by eclosion hormone include a neuroendocrine event,the release of the tanning hormone, bursicon, thereby ensuringthat tanning of the new cuticle must follow ecdysis. Ecdysis,itself, appears to be controlled by the ecdysteroid levels sinceecdysteroid injections delay ecdysis at physiological concentrationsand in a dose dependent fashion. This delay is due to inhibitionof eclosion hormone secretion and to the retardation of theterminal phases of the molt including the digestion of the oldcuticle and the onset of sensitivity to eclosion hormone. Thus,eclosion hormone secretion and the ecdysis it triggers are coordinatedwith the end of development because both are influenced by thesame endocrine signal—the decline in the ecdysteroid titer.  相似文献

Olfaction plays an important role in the host-seeking behavior of the malaria mosquito Anopheles gambiae. After a complete blood meal, female mosquitoes will not engage in host-seeking behavior until oviposition has occurred. We investigated if peripheral olfactory sensitivity changed after a blood meal by recording electroantennograms (EAGs) of female mosquitoes at three time points (2 h, 48 h and 72 h) to 15 volatile kairomones of either human origin or documented to emanate from oviposition sites. The EAG-sensitivity was compared with that of females of similar age post eclosion. As is common practice in electrophysiological studies, the EAG recordings were obtained by repeated stimulation of the same antennal preparations. We introduce mixed linear modeling as an improved statistical analysis for electrophysiological data. Two hours after blood ingestion, olfactory sensitivity as quantified through EAG-recording increased significantly and selectively, i.e. for seven compounds, compared to unfed females of the same age. Such short-term electrophysiological sensitization in the olfactory system as a result of feeding has not been documented before for insects. Sensitization to six compounds persisted until 48 h or 72 h post-blood meal at one or more concentrations. Desensitization was observed at 48 and 72 h pbm in response to two and three kairomones, respectively. For several compounds, sensitization at the EAG-level corresponded with sensitization found previously in single sensillum studies on olfactory neurons in antennal sensilla trichodea of An. gambiae females. These effects are likely to reflect sensitization to oviposition cues, as eggs have matured 48–72 h pbm. Knowledge of changes in olfactory sensitivity to kairomones can be applied to increase trap catches of malaria mosquitoes that have taken a blood meal and need to locate oviposition sites.  相似文献

1. Transmitter plasticity, the ability to alter transmitter expression, has been demonstrated in a wide range of neurons in the vertebrates and invertebrates.2. One preparation where this issue has been investigated in detail is a set of four neurosecretory neurons in the tobacco hawkmoth Manduca sexta.3. These neurons undergo a transmitter switch during the transition from caterpillar to moth.4. In larvae these neurons, known as the lateral neurosecretory cells (LNCs), express CAP2, a myomodulatory neuropeptide involved in the neuroregulation of the activity of visceral muscles.5. By the adult moth stage, however, the LNCs are no longer producing CAP2 and instead are expressing a different peptide, bursicon, which is involved in tanning of the cuticle at adult emergence.6. In fourth instar larvae of Pieris brassicae the haemolymph tyrosine level begins to rise about 1 day before apolysis to reach a level about treble that in the middle of the instar. Between apolysis and ecdysis the haemolymph tyrosine level appears to decline, until just before ecdysis another steep rise occurs. About 30 min after ecdysis a steep decline starts, levelling off gradually until the level in the middle of the instar is restored.Bursicon assays show that this hormone operates in the haemolymph both during apolysis and after ecdysis; but during the actual ecdysis no bursicon activity can be demonstrated in the haemolymph.Indications have been found that the bursicon activity can restore itself spontaneously in the haemolymph of newly ecdysed larvae. Insect growth and metamorphosis is punctuated by molts, during which a new cuticle is produced. Every molt culminates in ecdysis, the shedding of the remains of the old cuticle. Both the timing of ecdysis relative to the molt and the actual execution of this vital insect behavior are under peptidergic neuronal control. Based on studies in the moth, Manduca sexta, it has been postulated that the neuropeptide Crustacean cardioactive peptide (CCAP) plays a key role in the initiation of the ecdysis motor program. We have used Drosophila bearing targeted ablations of CCAP neurons (CCAP KO animals) to investigate the role of CCAP in the execution and circadian regulation of ecdysis. CCAP KO animals showed specific defects at ecdysis, yet the severity and nature of the defects varied at different developmental stages. The majority of CCAP KO animals died at the pupal stage from the failure of pupal ecdysis, whereas larval ecdysis and adult eclosion behaviors showed only subtle defects. Interestingly, the most severe failure seen at eclosion appeared to be in a function required for abdominal inflation, which could be cardioactive in nature. Although CCAP KO populations exhibited circadian eclosion rhythms, the daily distribution of eclosion events (i.e., gating) was abnormal. Effects on the execution of ecdysis and its circadian regulation indicate that CCAP is a key regulator of the behavior. Nevertheless, an unexpected finding of this work is that the primary functions of CCAP as well as its importance in the control of ecdysis behaviors may change during the postembryonic development of Drosophila.  相似文献

The Drosophila larval nervous system is radically restructured during metamorphosis to produce adult specific neural circuits and behaviors. Genesis of new neurons, death of larval neurons and remodeling of those neurons that persistent collectively act to shape the adult nervous system. Here, we examine the fate of a subset of larval motor neurons during this restructuring process. We used a dHb9 reporter, in combination with the FLP/FRT system to individually identify abdominal motor neurons in the larval to adult transition using a combination of relative cell body location, axonal position, and muscle targets. We found that segment specific cell death of some dHb9 expressing motor neurons occurs throughout the metamorphosis period and continues into the post‐eclosion period. Many dHb9 > GFP expressing neurons however persist in the two anterior hemisegments, A1 and A2, which have segment specific muscles required for eclosion while a smaller proportion also persist in A2–A5. Consistent with a functional requirement for these neurons, ablating them during the pupal period produces defects in adult eclosion. In adults, subsequent to the execution of eclosion behaviors, the NMJs of some of these neurons were found to be dismantled and their muscle targets degenerate. Our studies demonstrate a critical continuity of some larval motor neurons into adults and reveal that multiple aspects of motor neuron remodeling and plasticity that are essential for adult motor behaviors. © 2016 Wiley Periodicals, Inc. The BGL1 gene, encoding β-glucosidase in Saccharomycopsis fibuligera, was intracellular, secreted or cell-wall associated expressed in an industrial strain of Saccharomyces cerevisiae. The obtained recombinant strains were studied under aerobic and anaerobic conditions. The results indicated that both the wild type and recombinant strain expressing intracellular β-glucosidase cannot grow in medium using cellobiose as sole carbon source. As for the recombinant EB1 expressing secreted enzyme and WB1 expressing cell-wall associated enzyme, the maximum specific growth rates (μmax) could reach 0.03 and 0.05 h−1 under anaerobic conditions, respectively. Meanwhile, the surface-engineered S. cerevisiae utilized 5.2 g cellobiose L−1 and produced 2.3 g ethanol L−1 in 48 h, while S. cerevisiae secreting β-glucosidase into culture broth used 3.6 g cellobiose L−1 and produced 1.5 g ethanol L−1 over the same period, but no-full depletion of cellobiose were observed for both the used recombinant strains. The results suggest that S. cerevisiae used in industrial ethanol production is deficient in cellobiose transporter. However, when β-glucoside permease and β-glucosidase were co-expressed in this strain, it could uptake cellobiose and showed higher growth rate (0.11 h−1) on cellobiose.  相似文献

We describe the molecular analysis and cellular expression of the insect peptide neurohormone, bursicon. Bursicon triggers the sclerotization of the soft insect cuticle after ecdysis. Using protein elution analyses from SDS gels, we determined the molecular weight of bursicon from different insects to be approximately 30 kDa. Four partial peptide sequences of Periplaneta americana bursicon were obtained from purified nerve cord homogenates separated on two-dimensional gels. Antibodies produced against one of the sequences identified the cellular location of bursicon in different insects and showed that bursicon is co-produced with crustacean cardioactive peptide (CCAP) in the same neurons in all insects tested so far. Additionally, using the partial peptide sequences, we successfully searched the Drosophila genome project for the gene encoding bursicon. With Drosophila as a tool, we can now verify the function of the sequence using transgenic flies. Sequence comparisons also allowed us to verify that bursicon is conserved, corroborating the older data from bioassays and immunohistochemical analyses. The sequence of bursicon will enable further analysis of its function, release, and evolution.  相似文献
