Abscisic acid (ABA) inhibits the production of ethylene induced by water stress in excised wheat leaves and counteracts the stimulatory effect of 6-benzyladenine (BA) on this process. The stimulatory effect of BA and the inhibitory effect of ABA were equally pronounced whether external or endogenous ethylene levels were determined. When leaves were sprayed or floated on solutions of BA, indole-3-acetic acid (IAA), gibberellic acid (GA3), or ABA, the relative activities of these growth regulators on stress-induced ethylene at 10-4 mol l-1 were BA>IAA >GA3>controls>ABA. In non-stressed leaves, however, where the levels of ethylene produced were 2–20 times smaller, the relative activities were IAA >BA>GA3>controls>ABA. The effects of BA and ABA spray treatment on water stress induced ethylene were closely similar whether the solutions were applied 2 or 18 h prior to the initiation of water stress. Light was found to inhibit substantially (i.e. up to 88%) the production of ethylene induced by water stress in excised wheat leaves and from the shoots of intact plants. The relatively small amounts of ethylene emanating fron non-stressed leaves were also inhibited by light but to a smaller degree (i.e. up to 61%). In water-stressed leaves the degree of light inhibition of ethylene production was shown to be related to the age of the leaves; the amounts of ethylene diffusing from young leaves (i.e. 6-days old) was inhibited 52% by light whereas in older leaves (i.e. 9-days old) it was inhibited by 85%. Previous studies [Wright (1979) Planta 144, 179–188 and (1980) Planta 148, 381–388] had shown that application of 6-benzyladenine (BA) to leaves a day before wilting, greatly increases the amount of ethylene diffusing from the leaves following wilting (e.g. 8-fold), and to smaller degrees do applications of indole-3-acetic acid (IAA) and gibberellic acid (GA3). On the other hand abscisic acid (ABA) treatment reduces the amount of ethylene produced. In these earlier experiments the ethylene was collected from leaves held under dark or near-dark conditions, so in the present study the activities of these growth regulators (10-4 mol l-1 solutions) under dark and light conditions were compared. It was found that they maintained the same relative activities on ethylene emanation (i.e. BA>IAA>GA3>water controls>ABA) under both light and dark conditions. However, because of the inhibitory effect of light, the absolute amounts of ethylene produced from all treatments were always much higher in the dark than in the light (usually about a 6-fold difference). An interesting effect of light treatment on ethylene biosynthesis was found when water-stressed leaves were kept in dark chambers for 41/2 h and then transferred to light. Quite unexpectedly, instead of the rate of ethylene production falling immediately, it continued to be produced at the dark rate (i.e. no light inhibition!) for over 2 h before the rate began to decline, and for a much longer period (i.e. in excess of 41/2 h) if the leaves had previously been sprayed with BA. Predictably, leaves placed in the light (i.e. in leaf chambers) and then transferred to darkness, immediately or very soon produced ethylene at the dark rate. One explanation of these results, which is discussed, would be that the biosynthesis of an ethylene precursor requires an obligatory dark stage. The amount of diffusible ethylene from excised wheat leaves (Triticum aestivum L. cv. Eclipse) increased when they were subjected to water stress. The quantity of ethylene produced was related to the severity of the stress, reaching a maximum at a leaf water potential leaf of approximately-12 bars. Irrespective of the severity of the stress, the maximum rate of ethylene production usually occurred between 135–270 min after applying the stress and then the rate declined. Part of the decline may have been due to an oxygen deficiency in the leaf chambers. In excised water-stressed leaves there was a sigmoid relationship between increasing ethylene and abscisic acid (ABA) levels and decreasing leaf water potential values. The two curves were displaced from each other by approximately 1 bar, with ethylene evolution leading that of ABA accumulation. The maximum rate of increase in ethylene occurred between-8 and-9 bars and for ABA between-9 and-10 bars. A significant increase in the levels of these two plant growth regulators was found when the leaf decreased outside the normal diurnal leaf range by 1 bar for ethylene and 2 bars for ABA. Because of the sigmoid nature of the curves there was no distinct threshold leaf value triggering-off an increase in ethylene or ABA, but with ABA the curve became very steep at a leaf value of-9.3 bars and this could be looked upon as a kind of threshold value.It seems unlikely that the stress-induced ethylene evolution in excised wheat leaves stimulated the accumulation of ABA, because when the leaves were subjected to a substantial water stress (e.g. leaf bars) ABA increased immediately and at a faster rate than ethylene.Abbreviations ABA abscisic acid - GLC gas-liquid chromatography - RWC relative water content - TLC thin-layer chromatography - leaf leaf water potential  相似文献

The effect of water deficit on nodulation, N2 fixation, photosynthesis, and total soluble sugars and leghemoglobin in nodules was investigated in cowpea and groundnut. Nitrogenase activity completely ceased in cowpea with a decrease in leaf water potential ( leaf) from –0.4 MPa to –0.9 MPa, while in groundnut it continued down to –1.7 MPa. With increasing water stress, the acetylene reduction activity (ARA) declined very sharply in cowpea, but ARA gradually decreased in groundnut. Even with mild water stress ( leaf of 0.2 MPa), nodule fresh weight declined 50% in cowpea partly due to a severe nodule shedding whereas nodule fresh weight declined in groundnut only when leaf decreased by 1.0 MPa. No nodule shedding was noticed even at a higher stress level in groundnut. Photosynthesis and stomatal conductance were also more stable in groundnut than in cowpea under water stress. There was a sharp increase in total soluble sugars and leghemoglobin in the nodules of groundut with water stress, but no definite trend could be found in cowpea.  相似文献

Water and salt stress promote betaine accumulation in leaves of barley (Hordeum vulgare L.) by accelerating the de-novo synthesis of betaine, via choline. Previous radiotracer kinetic studies have implicated stress-enhanced turnover of the choline moiety of phosphatidylcholine (PC) as a major source of choline for betaine synthesis. Two approaches have therefore been followed to show whether stress-induced PC turnover is a cellor organelle-specific phenomenon, or a generalized one. In the first approach, [3H]ethanolamine of high specific activity was supplied to second leaves of unstressed and water-stressed barley plants; after 1 h, paired sections of tissue were excised from each leaf, one for extraction and analysis of [3H]metabolites and the other for autoradiography. The3H-activity remaining in the leaf tissue after washing out the water-soluble3H-metabolites during preparation for autoradiography was taken to be mainly in phospholipids. In unstressed leaves, [3H]phosphatidylethanolamine (PE) was the major labeled phospholipid, whereas there were approximately equal amounts of [3H]PE and [3H]PC in stressed leaves. At the light-microscope level, silver grains were associated with all living cells in both unstressed and stressed leaves; grains were concentrated in the cytoplasmic regions of highly vacuolate mesophyll cells, and were distributed throughout densely cytoplasmic vascular parenchyma. At the electron-microscope level, silver grains were not confined to any particular types of membranes in unstressed or stressed leaves. In the second approach, second leaves of stressed plants received a 1-h pulse of [14C]ethanolamine, and were then homogenized. The brei was subjected to sucrose density gradient centrifugation. The specific radioactivity of [14C]PC was quite similar in the gradient fractions, whether they contained microsomes or mitochondrial plus chloroplast membranes. Excised wheat (Triticum aestivum L.) leaves, when subjected to drought stress, increased ethylene production as a result of an increased synthesis of 1-aminocyclopropane-1-carboxylic acid (ACC) and an increased activity of the ethyleneforming enzyme (EFE), which catalyzes the conversion of ACC to ethylene. The rise in EFE activity was maximal within 2 h after the stress period, while rehydration to relieve water stress reduced EFE activity within 3 h to levels similar to those in nonstressed tissue. Pretreatment of the leaves with benzyladenine or indole-3-acetic acid prior to water stress caused further increase in ethylene production and in endogenous ACC level. Conversely, pretreatment of wheat leaves with abscisic acid reduced ethylene production to levels produced by nonstressed leaves; this reduction in ethylene production was accompanied by a decrease in ACC content. However, none of these hormone pretreatments significantly affected the EFE level in stressed or nonstressed leaves. Suboptimal nitrogen nutrition, leaf aging, and prior exposure to water stress all increased stomatal closure in excised cotton (Gossypium hirsutum L.) leaves supplied abscisic acid (ABA) through the transpiration stream. The effects of water stress and N stress were partially reversed by simultaneous application of kinetin (N6-furfurylaminopurine) with the ABA, but the effect of leaf aging was not. These enhanced responses to ABA could have resulted either from altered rates of ABA release from symplast to apoplast, or from some post-release effect involving ABA transport to, or detection by, the guard cells. Excised leaves were preloaded with [14C]ABA and subjected to overpressures in a pressure chamber to isolate apoplastic solutes in the exudate. Small quantities of 14C were released into the exudate, with the amount increasing greatly with increasing pressure. Over the range of pressures from 1 to 2.5 MPa, ABA in the exudate contained about 70% of the total 14C, and a compound co-chromatographing with phaseic acid contained over half of the remainder. At a low balancing pressure (1 MPa), release of 14C into the exudate was increased by N stress, prior water stress, and leaf aging. Kinetin did not affect 14C release in leaves of any age, N status, or water status. Net photosynthetic rate (PN), transpiration rate (E), and stomatal conductance (gs) during water stress and after rehydration were measured in Phaseolus vulgaris, Beta vulgaris, and Zea mays. Immediately before imposition of water stress by cessation of watering, plants were irrigated with water (control), 100 M abscisic acid (ABA), and/or 10 M N6-benzyladenine (BA). In all three species, application of ABA decreased gs, E, and PN already 1 h after application. However, during water stress gs, E, and PN in plants pre-treated with ABA remained higher than in plants pre-treated with water. Positive effects of ABA application were observed also after rehydration. In contrast, the effects of pre-treatment with BA were species-specific. While in bean plants BA application ameliorated negative effect of water stress, only very slight effects were observed in maize, and in sugar beet BA even aggravated the effects of water stress.  相似文献

The activity of nitrate reductase and the pool sizes of some amino acids and some sugars were measured in relation to the leaf water potential () of maize leaves. The activity of nitrate reductase was severely inhibited in water-stressed maize leaves. This was not due to substrate shortage or the presence of an inhibitor at reduced leaf water potential. While the typical proteinogenic amino acids valine, tyrosine, leucine and isoleucine were almost undetectable in the leaves of the control plants, their concentrations markedly increased with declining , thus indicating protein degradation. The concentrations of serine, glycine and glutamate increased upon water stress, their total amount in severely stressed leaves ranging 5- to 6-fold higher than the total amount of valine, tyrosine, leucine and isoleucine at this stage of water deficit. The pool sizes of glucose, fructose and sucrose decreased in relation to decreasing . Leaf disks from peppermint, spearmint, orange mint, lavender mint and Scotch spearmint were cultured on various Murashige-Skoog-based media in order to regenerate shoots. A significantly larger average number of orange mint leaf disks regenerated shoots on basal medium containing 44.4 M benzyladenine (BA) and 250 ml l-1 coconut water (CW). Shoots regenerated from peppermint leaf disks cultured on basal medium containing 44.4 M BA and 250 ml or 450 ml l-1 CW. The most shoots regenerated from orange mint leaf disks cultured on medium containing 10 g l-1 washed Difco Bacto-agar. Disks excised from the bases of the first expanding pair of orange mint leaves cultured under dark conditions regenerated a significantly larger average number of shoots. Yields of 106–108 peach mesophyll cells and protoplasts · gfw-1 were obtained depending on factors such as digesting enzymes, and leaf size. Onozuka R-10 (2%) in combination with Macerase (0.5%) was found best for protoplast isolation and mediocre for cell isolation among several enzyme combinations tested. Viability was 90% for protoplasts and 60% for cells. Pectolyase Y23 was found to be ineffective in our investigation. Small leaves, 4–10 mm in length, were a superior source for protoplast isolation than medium or big expanded leaves, 22–30 mm in length. The high yields of protoplasts could be obtained only when keeping the ratio of leaf biomass to volume of digesting enzyme solution under 20 mg ml-1. Purification of protoplasts on a sucrose gradient yielded about 107 protoplasts · gfw-1, however, the preparation was still contaminated by intact cells. Protoplasts were cultured under different growth regulators and physical conditions. Apical flower buds of Cymbidium goeringli Reichenbach fil. (ca 2 mm long) exeised from infloreseences (ca 5 cm long) were explanted on modified Murashige & Skoog medium (=MS medium) supplemented with N6-benzyladenine (BA) and -naphthaleneacetic acid (NAA). Within 107 days of culture, swelling growth, chlorophyll synthesis, and subsequent rhizome differentiation were observed. MS medium containing 0.1 mg l-1 BA and 10 mg l-1 NAA was found to be optimal for initiating rhizome development and subsequent plantlet regeneration.Explants cultured on MS medium supplemented with 1 mg l-1 NAA alone formed a mass of rhizome branches. In preparation for gene transfer experiments we investigated factors that might affect the production of shoots and somatic embryos from the wound callus of cultured sugarbeet leaf discs. A complex interaction was found between the leaf disc plating density, the disc culture medium, the source-shoot culture medium and the frequency of disc transfer to fresh medium. The most productive protocol utilized: source shoots maintained on MS medium containing 0.25 mg 1-1 BA; multiple leaf discs (ten 4-mm discs/plate) plated onto an enriched modification of MS medium (RV) containing 1.0 mg 1-1 BA and solidified with 0.3% Gelrite (not permitted to dry during hardening); and transfer of the discs to fresh medium every two weeks during the first month. This standard protocol produced more callus per plate and higher rates of morphogenesis per unit dry weight of callus than did the one-step method of Saunders and Doley. Water availability considerations were found to be critical to obtaining high morphogenic rates. Metabolism of abscisic acid (ABA) was studied after wilting and upon recovery from water stress in individual, detached leaves of Phaseolus vulgaris L. (red kidney bean). Loss of turgor was correlated with accumulation of ABA and its metabolites, resulting in a 10-fold increase in the level of phaseic acid (PA) and a doubling of the level of conjugated ABA. The level of conjugated ABA in turgid leaves was no higher than that of the free acid. These results indicate that accumulation of ABA in wilted leaves resulted from a stimulation of ABA synthesis, rather than from a release from a conjugated form or from inhibition of the metabolism of ABA. The rate of synthesis of ABA was at its maximum between 2.5 and 5 h after turgor was lost, and slackened there-after. In wilted leaves, the rate of conversion of ABA to PA climbed steadly until it matched the rate of synthesis, after about 7.5 h. Upon rehydration of sections from wilted leaves, the rate of synthesis of ABA dropped close to zero within about 3 h, while the rate of conversion to PA accelerated. Formation of PA was two to four times faster than in sections maintained in the wilted condition; it reached a rate sufficient to convert almost one-half of the ABA present in the tissue to PA within 1 h. In contrast, the alternate route of metabolism of ABA, synthesis of conjugated ABA, was not stimulated by rehydration. The role of turgor in the stimulation of the conversion of ABA to PA was investigated. When leaves that had been wilted for 5 h were rehydrated to different degrees, the amount of ABA which disappeared, or that of PA which accumulated during the next 3 h, did not depend linearly on the water potential of the rehydrated leaf. Pretreatment of detached carnation petals (Dianthus caryophyllus cv White Sim) for 24 hours with 0.1 millimolar of the cytokinins n6-benzyl-adenine (BA), kinetin, and zeatin blocked the conversion of externally supplied 1-aminocyclopropane-1-carboxylic acid (ACC) to ethylene and delayed petal senescence by 8 days. The normal enhanced wilting and increase in endogenous levels of ACC and ethylene production following exposure of petals to ethylene (16 μl/l for 10 hours), were not observed in BA-pretreated petals. In carnation foliage leaves pretreated with 0.1 mm BA, a reduction rather than inhibition of the conversion of exogenous ACC to ethylene was observed. This indicates that foliage leaves respond to cytokinins in a different way than petals. A constant 24-hour treatment with BA (0.1 mm) was not able to reduce ethylene production of senescing carnation petals, while 2 mm aminoxyacetic acid, a known inhibitor of ACC synthesis, or 10 mm propyl gallate, a free radical scavenger, decreased ethylene production significantly.  相似文献

Cytokinins inhibit hypocotyl elongation in darkness but have no obvious effect on hypocotyl length in the light. However, we found that cytokinins do promote hypocotyl elongation in the light when ethylene action is blocked. A 50% increase in Arabidopsis thaliana (L.) Heynh. hypocotyl length was observed in response to N6-benzyladenine (BA) treatment in the presence of Ag+. The level of the ethylene precursor 1-aminocyclopropane-1-carboxylic acid was strongly increased, indicating that ethylene biosynthesis was up-regulated by treatment with cytokinin. Furthermore, the effects of cytokinins on hypocotyl elongation were also tested using a series of mutants in the cascade of the ethylene-signal pathway. In the ethylene-insensitive mutants etr1-3 and ein2-1, cytokinin treatment resulted in hypocotyl lengths comparable to those of wild-type seedlings treated with both Ag+ and BA. A similar phenotypical response to cytokinin was observed when auxin transport was blocked by -naphthylphthalamic acid (NPA). Applied cytokinin largely restored cell elongation in the basal and middle parts of the hypocotyls of NPA-treated seedlings and at the same time abolished the NPA-induced decrease in indole-3-acetic acid levels. Our data support the hypothesis that, in the light, cytokinins interact with the ethylene-signalling pathway and conditionally up-regulate ethylene and auxin synthesis.  相似文献

Summary The effect of 2-day cycles of osmotically induced leaf moisture stress followed by partial recovery on the nodulation and nitrogenase activity of 2 soya cultivars was studied. Fourteen days after plant inoculation (mid-growth stage) the total leaf electrochemical water potential (wleaf) of control plants ranged from –0.8 to –1.9 bars, whereas the concentrations of osmoticum (polyethylene glycol 4000) induced wleaf values ranging from –1.4 (recovery value) to –3.1 bars (low stress), –1.8 to –4.4 bars (mild stress), and –2.2 to –6.2 bars (medium stress). The low stress treatment reduced nodule numbers and their specific activity in both cultivars, without affecting nodule size or the time required for nodule initiation. Nodule initiation was delayed in both cultivars by the mild and medium stress treatments, the former treatment reducing the number and size of the nodules and such nodules exhibited very low specific activity. The medium stress treatment prevented the further development of nodule initials, which remained inactive throughout the experiment. Such results imply an effect of water stress on the infection process and on nodule morphogenesis. The reduction in nodule numbers observed in water stressed plants was not associated with a reduced number of rhizobia in the rhizoplane nor due to an effect on root growth or root hair formation.At a stage prior to the formation of macroscopic nodule initials, the roots of plants under medium stress (wleaf=–5.5 bar)s) had a higher content of abscisic acid (ABA) (4-fold increase) and a lower content of gibberellin (GA)-like substances (21.4% reduction) as compared to control plants (wleaf=–1.0 bar). Although the medium stress treatment slightly increased the stomatal resistance of leaves, photosynthetic and transpiration rates were unaffected. Similar alterations of the hormononal balance occurred in the nodulated roots of plants subjected to naturally induced leaf moisture stress.Since the foliar application of ABA (1.92×10–5 M) to unstressed plants inhibited nodulation (45% reduction in nodule numbers), the increased endogenous content of thishormone in the roots of plants under leaf moisture stress may provide some physiological insight into the inhibitory effect of water stress on the nodulation process.  相似文献

Leaf water potentials of Phaseolus vulgaris L. plants exposed to a -3.0 bar root medium were reduced to between -7 and -9 bars within 25 min and remained constant for the next several hours. This treatment led to considerable variation between leaves in both abscisic-acid (ABA) content and Rs, although the two were well correlated after a 5-h treatment. There was an apparent 7-fold increase in leaf ABA levels necessary to initiate stomatal closure when plants were exposed to a -3.0 bar treatment, but when plants were exposed to a -5.0 bar stress Rs values increased prior to any detectable rise in ABA levels. An efficient method was developed using floating membrane rafts (Liferaft) for the micropropagation of Aconitum napellus (Ranunculaceae), a cut flower crop with a low natural propagation rate. This was achieved by introducing shoot tips into culture on Murashige and Skoog's (1962) solid medium, or liquid medium-supported rafts, supplemented by different levels of benzyl adenine (BA). Optimum shoot proliferation on solid medium required 4mg/l BA, whereas for expiants supported on rafts optimal proliferation was achieved at 0.25mg/l BA. Maximum shoot proliferation was found using the floating rafts (propagation ratio of 4.2 per month), 45% higher than the maximum value on solid medium. A similar value could be obtained on solid medium after a period of 2 months. The optimal response to BA was similar for fresh weight gain and shoot length. Summary The simultaneous presence of 6-benzyladenine (BA) and sucrose in a Murashige and Skoog medium (SIM) during the initial stages of shoot initiation have been found to be obligatory for high-frequency shoot formation in the Capsicum annuum L. var. Sweet Banana upper hypocotyl explants. The explants are determined for shoot formation following a minimum of 8 days of culture on SIM. Deprivation of exogenous sucrose from day 6 to day 20 of culture had no effect on the shoot forming response of the explants.
