Summary The growth and hyoscyamine production of transformed roots of Datura stramonium have been examined in a modified 14-1 stirred tank reactor in both batch and continuous fermentations on media containing half or full strength Gamborg's B5 salts and at three different temperatures. Under a range of conditions, roots grown on half strength B5 salts with 3% w/v sucrose had a higher dry matter content (up to 8.3% w/w) and a higher hyoscyamine content (up to 0.52 mg·g–1 wet weight) than roots grown on full strength B5 salts with the same level of sucrose (up to 4.6% w/w dry matter and up to 0.33 mg hyoscyamine g–1 wet weight). Growth at 30°C was initially faster than at either 25°C or 35°C and by day 12, the drained weight of roots in the fermentor at 30°C was about fourfold greater than at 25°C and twice that at 35°C. The ultimate hyoscyamine levels attained (approximately 0.5 mg·g–1 wet weight) were similar at both 25°C and 30°C but some 40% lower at 35°C. Final packing densities of 70% w/v were achieved for roots after 37 days growth at 25°C and the highest production rate of 8.2 mg hyoscyamine l–1 per day was obtained for roots grown at 30°C. In continuous fermentation at 25°C, the release of hyoscyamine into the culture medium was low (less than 0.5% w/w of the total) but was up to sevenfold higher in fermentors operated at 30°C or 35°C. Cellulose in wastewater was converted into H2 by a mixed culture in batch experiments at 55°C with various wastewaters pH (5.5–8.5) and cellulose concentrations (10–40 g l–1). At the optimal pH of 6.5, the maximum H2 yield was 102 ml g–1 cellulose and the maximum production rate was 287 ml d–1 for each gram of volatile suspended solids (VSS). Analysis of 16S rDNA sequences showed that the cellulose-degrading mixed culture was composed of microbes closely affiliated to genus Thermoanaerobacterium.  相似文献

Candida shehatae NCL-3501 utilized glucose and xylose efficiently in batch cultures. The specific rate of ethanol production was higher with mixtures of glucose and xylose (0.64–0.83 g g–1 cells d–1) compared to that with individual sugars (0.38–0.58 g g–1 cells d–1). Although the optimum temperature for growth was 30°C, this strain grew and produced appreciable levels of ethanol at 45°C. A stable ethanol yield (0.40–0.43 g g–1 substrate utilized) was obtained between 10 g L–1 and 80 g L–1 of initial xylose concentration. Conversion efficiency was further improved by immobilization of the cells in calcium alginate beads. Free or immobilized cells ofC. shehatae NCL-3501 efficiently utilized sugars present in rice straw hemicellulose hydrolysate, prepared by two different methods, within 48 h. Ethanol yields of 0.45 g g–1 and 0.5 g g–1 from autohydrolysate, and 0.37 g g–1 from acid hydrolysate were produced by free and immobilized cells, respectively.  相似文献

A novel design of a solid-state bioreactor, operated with periodic pressure oscillation coupled with forced aeration through the medium, gave efficient control of temperature. The evaluation of the bioreactor assembly with respect to temperature and cellulase production by Penicillium decumbens JUA10 showed that, at 4 atm and the bed depth of 6 cm, the maximal temperature variation in the reactor was +1.5 °C at a set value of 30 °C compared with +6.8 °C in a static tray system. The highest cellulase and -glucosidase activities were 15 IU g–1 and 51 IU g–1 substrate dry matter at 96 h, respectively, while only 10 IU g–1 and 24 IU g–1 were obtained in the static tray culture system.  相似文献

Young reptiles have higher relative energy demands than adults, but the proposed ontogenetic changes in diet to fulfil these demands were not found in the algae-eating Galápagos marine iguanas on Santa Fé. Feeding and digestion rates were investigated to analyse how young achieve higher energy intake. Daily food intake of free ranging marine iguana hatchlings (6–11 months old) was about one third that of adults, but relative intake (g dry mass · g–1 wet mass · day–1) was four times higher in the hatchlings. During feeding experiments, relative daily food intake of hatchling marine iguanas was approximately three times higher than that of adults (0.042 vs 0.013 g dry mass · g–0.8 wet mass · day–1), and mean gut passage time was two times shorter (5 vs 10 days). The hatchlings also maintained high body temperatures (36.7° C) even under relatively cool day-time air temperatures of 32° C. Apparent digestibility of algal food measured both during feeding trials and by Mn2+ AAS (atomic absorption spectrometry) for free-ranging iguanas was 70%, independent of body size and temperature. The red algae prevailing in the diet were high in protein (30% dry mass) and energy (12.1 kJ/g dry mass). Diving iguanas had higher rates of energy intake than intertidal foragers, but daily intake was less. Maintenance of high body temperature enabled hatchlings to achieve high digestion rates and, combined with high relative intake, thus achieve sufficient energy intake for rapid growth despite higher mass specific metabolic rates. Estimates of biomass of marine iguanas and their algal food are given for a section of coastline on Santa Fé.  相似文献

Summary A strain ofFusarium moniliforme, previously used for microbial protein production, excreted lactase (-D-galactosidase, EC.3.2.1 23) when cultivated either in a whey liquid medium or on a wheat bran solid medium. The enzyme produced in both media had pH and temperature optima of 4–5 and 50–60°C respectively and was particularly suitable for processing acid whey.In the whey culture, maximum lactase yield was observed after 95 h of growth at 30°C and whey lactose concentration of 9%. The addition of ammonium, potassium and sodium ions to the growth medium considerably enhanced lactase production. A maximum enzyme yield corresponding to hydrolysis of 3 nmoles o-nitrophenyl--D-galactopyranoside sec–1 ml–1 of growth medium, at pH 5 and 60°C, was obtained.In the wheat bran culture, the maximum enzyme yield was obtained after 140 h of growth at 28–30°C. A marked increase in the enzyme production was observed when nitrate or phosphate was added to the growth medium. Also, the addition of certain agricultural by-products (molasses, whey) enhanced lactase production. The observed maximum yield corresponding to the hydrolysis of 182 nmoles of ONPG sec–1 g–1 of wheat bran, at pH 5 and 60°C, is comparable to that reported for certain microorganisms used commercially for lactase production.  相似文献

Wheat straw impregnated with a nutrient solution was used to culture Rhizobium leguminosarum. The fermentation was carried out in a periodic pressure, oscillating, solid-state fermenter. At 30 °C and 3 atm, Rhizobium leguminosarum grew to 5.3×1010 c.f.u. g–1 substrate dry matter in about 36 h, while only 1.8×1010 c.f.u. g–1 substrate dry matter was obtained in a conventional static tray fermenter.  相似文献

Summary In order to prepare fermentable sugars from cellulosic raw materials, we have tested the effect of different pretreatments (heat, chemicals, pectinase) on enzymatic hydrolysis of sugar beet pulp. Best results (90% hydrolysis) have been obtained after heat treatment (30 mn at 120°C) with 16 IU.Fp.g–1 of fresh pulp in 48 hours.  相似文献

Summary The nonsporulating extreme thermophile Thermus thermophilus was grown in continuous culture at dilution rates up to 2.65 h–1 at 75°C and pH 6.9 on complex medium. Concomitantly very low yield (Y=0.12 g cell dry weight g–1 utilized organic carbon) and incomplete substrate utilization (always less than 45%) were found. In batch cultures T. thermophilus could be grown with max =h–1, in shake flasks only with max =h–1 with the same low yield and incomplete substrate utilization. Stable steady states at 84C and 45°C were realized at a dilution rate of 0.3 h–1 whereas at 86°C and 40°C no growth could be detected. Artefacts arising from wall growth (in bioreactors) or improper materials must be ruled out. Inhibition of growth by organic substrates was demonstrated at low concentrations: a decrease in the yield obtained was found when more than 0.7 gl–1 of meat extract were supplied in the medium. The maintenance requirement for oxygen is potentially very high and was determined to be 10 to 15 mmol g–1 h–1.  相似文献

To determine the contribution of food ingestion (trophic pathway) to PCB contamination of zooplankton in the river Meuse (Belgium), we used 14C-labelled algae (Dictyosphaerium ehrenbergianum) to measure ingestion and assimilation rates in the rotifer species Brachionus calyciflorus. When the concentration of algae in the culture medium varied from 20 103 to 200 103 algal cells ml–1 (0.12 to 1.18 mg Cl–1), the Brachionus calyciflorus ingestion rate varied from 0.25 ± 0.12 to 1.52 ± 0.43 ng C ind–1 h–1 at 15 °C and from 0.74 ± 0.17 to 5.93 ± 0.61 ng C ind–1 h–1 at 20 °C. The assimilation efficiency (ratio of the assimilation rate to the ingestion rate) measured in a culture medium containing 200 103 algal cells ml–1 was 55.7 ± 5.8%. Since the PCB concentration measured in the phytoplankton of the river Meuse is about 3 µg PCBs g–1 D.W., the estimated PCB contamination of zooplankton ascribable to the trophic pathway ranges from 0.22 ± 0.17 to 1.31 ± 0.77 µg PCBs g–1 D.W. at 15 °C and from 0.64 ± 0.34 to 5.10 ± 2.10 µg PCBs g–1 D.W. at 20°C. The lower figure based on measurements effected at 20 °C is comparable to the actual level measured in zooplankton samples collected in the river Meuse (0.69 ± 0.20 µg PCBs g–1 D.W.). The applicability of the formula used in our estimate was checked in a 48-hour in vitro experiment in which the rotifers were fed contaminated algae. The PCB accumulation measured in the rotifers was found to coincide with the calculated PCB contamination. Additional experiments were carried out to determine the contribution of the direct pathway to PCB contamination of zooplankton living in the river Meuse (0.02 µg PCBs l–1 of water; average dissolved organic matter: 3 mg C 1–1). The PCB concentration in zooplankton resulting from direct uptake of PCBs from the water was estimated at 0.19 ± 0.05 µg PCBs g–1 D.W. These results show that in zooplankton living in polluted ecosystems, PCBs are likely to accumulate via the trophic pathway to concentrations up to 30 times higher than by direct contamination. Furthermore, our estimates of PCB contamination via the trophic pathway coincide quite well with actual concentrations measured in situ.  相似文献

During the hydrolysis of an exopolysaccharide (EPS) produced by Citrobacter sp., the maximum liberation of hexosamine was obtained with 6 m HCl at 115 °C in an autoclave for 1 h. The glycosidic bond energy and degree of acetylation of the hexosamine in EPS were approximately 77 kJ mol–1 and 61%, respectively. Thermal destruction of the hexosamines and the effect of salt on the hexosamine determination could be minimized under the optimized hydrolytic conditions. Sweating rate, rectal and skin temperatures and respiration rate were measured at weekly intervals from 7 days of age (for 4 weeks in Experiment 1; 6 weeks in Experiment 2) in winter- and summer-born Friesian calves exposed to a temperature of 39°C dry bulb and 32°C wet bulb in a climate chamber. Four calves were studied in each season in both experiments. In Experiment 1, ambient temperatures were from 3° to 9°C higher in early summer than in late winter. During each 39°C exposure, sweating rate increased from basal levels of 40–90 to plateau levels of 120–300 g/m2 per h after 90–120 min. The increase in sweating rate with age was most pronounced in winter-born calves, but summer-born calves had higher values at 1 week of age (167±52.4 vs 94.4±30.1 g/m2 per h). Seasonal differences in ambient temperature were greater in Experiment 2 (11° to 17°C). In this case summer-born calves had higher sweating rates at each age (plateau values of 220–320 g/m2 per h), and showed a more rapid increase in sweating rate during each 39°C exposure than winter-born calves (plateau values of 100–250 g/m2 per h). The results demonstrate major changes in sweating competence during the first 4–6 weeks of life in Friesian calves, a quite pronounced effect of season (ambient temperature) on the levels of sweating achieved, and indicate that low sweating rates in newborn calves are a contributing factor in deaths due to hyperthermia in semi-arid grazing areas.  相似文献

A strain of Kluyveromyces marxianus was grown in batch culture in lactose-based media at varying initial lactose concentrations (10–60 g L–1) at 30°C, pH 5.0, dissolved oxygen concentrations greater than 20%. Increasing the concentration of mineral salts three-fold at 40 g L–1 and 60 g L–1 initial lactose concentration showed only a small increase in the yield of biomass, from 0.38 g g–1 to 0.41 g g–1, indicating that the initial batch cultures were not significantly nutrient- (mineral salts)-limited. A relatively high biomass concentration (105 g L–1) was obtained in fed-batch culture following extended lactose feeding. An average specific growth rate (0.27 h–1), biomass yield (0.38 g g–1) and overall productivity (2.9 g L–1 h–1) were obtained for these fed-batch conditions. This fed-batch protocol provides a strategy for achieving relatively high concentrations and productivities of K. marxianus on other lactose-based substrate streams (e.g., whey) from the dairy industry.  相似文献

The modified sulfite oxidation method was adapted for estimation of the overall oxygen transfer rate in a pressure oscillating, solid-state fermenter. At 4.5 atm and 30 °C, the oxygen transfer rate reached 717 mmol kg–1 initial dry matter h–1 in this system against 37 mmol kg–1 initial dry matter h–1 in a static tray fermenter. At 30 °C and 3 atm, Azotobacter vinelandii grew on wheat straw and reached 4.7×1010 c.f.u. g–1 substrate dry matter after 36 h, while only 8.2×109 c.f.u. g–1 substrate dry matter was obtained in a static tray system.  相似文献

Water absorption by and seedling emergence of barley (Hordeum vulgare) seeds was studied in a two layer drying out system. Seeds were placed 3 cm below surface in sandy loam (Typic Ustochrept) soil having 4 or 7g.100g–1 water underlain by wet (10g.100g–1) layer 2, 4 or 6cm below seed. The study was carried out at 18°, 23°, 28° and 33°C with and without a thin liquid-flow barrier placed on top of the wet layer.Water absorption by seed and coefficient of rate of emergence showed parabalic relation with temperature and strong soil-water × temperature interactions. Liquid-flow barrier considerably reduced the seed water absorption, percent emergence and coefficient of rate of emergence showing thereby that liquid flow was the principal mode of upward water transport from the wet soil to the seed. Influence of both the wet soil and the liquid-flow barrier was detectable up to about 8 cm; shorter the distance greater the effect. It is concluded that in a drying out seed-zone, in addition to wetness of the soil surrounding the seed the wetness of the soil several cm below the seed is also crucial for seedling emergence. Also indicated that the optimum temperatures in drying out seed-zones are different from those in the absence of evaporation.  相似文献

The combined effects of water activity (aw) and temperature on mycotoxin production by Penicilium commune (cyclopiazonic acid — CPA) and Aspergillus flavus (CPA and aflatoxins — AF) were studied on maize over a 14-day period using a statistical experimental design. Analysis of variance showed a highly significant interaction (P 0.001) between these factors and mycotoxin production. The minimum aw/temperature for CPA production (2264 ng g–1 P. commune, 709 ng g–1 A. flavus) was 0.90 aw/30 °C while greatest production (7678 ng g–1 P. commune, 1876 ng g–1 A. flavus) was produced at 0.98 aw/20 °C. Least AF (411 ng g–1) was produced at 0.90 aw/20 °C and most (3096 ng g–1) at 0.98 aw/30 °C.  相似文献

Studies were carried out for the production of aroma compounds by Kluyveromyces marxianus grown on cassava bagasse in solid state fermentation using packed bed reactors, testing two different aeration rates. Respirometric analysis was used to follow the growth of the culture. Headspace analysis of the culture by gas chromatography showed the production of 11 compounds, out of which nine were identified. Ethyl acetate, ethanol and acetaldehyde were the major compounds produced. Lower aeration rate (0.06l h–1 g–1 of initial dry matter) increased total volatile (TV) production and the rate of production was also increased at this aeration rate. Using an aeration rate of 0.06l h–1 g–1 maximum TV concentrations were reached at 24 h and at 40 h with 0.12l h–1 g–1.  相似文献

Yarrowia lipolytica, grown in a nitrogen-limited continuous culture (D = 0.032 h–1), produced 9.3 g dry biomass l–1, which contained 0.25 g oil g–1. When an aqueous extract from Teucrium polium L. was added, the biomass concentration remained constant while the oil content increased to 0.33 g oil g–1 dry weight. The specific rate of oil formation increased from 7.9 to 10.6 mg oil g–1 biomass h.  相似文献

Summary The formation and localization of the -lactamase of Acinetobacter calcoaceticus CCM 5593 is strongly affected by cultivation and induction conditions. Optimal parameters for enzyme yield are cultivation on minimal salts medium with acetate (10 g·1–1) as carbon source and addition of yeast extract (5–10 g·l–1), induction by cefotaxime (50g·ml–1) immediately after inoculation and growth for 24 h at 25° C. The strain forms a basal level of -lactamase constitutively [70 units (U)·g–1]. Nearly all of this was found to be cell-bound. However, -lactamase activity additionally produced after induction (up to 500 U·g–1 wet bacteria) was located in the culture medium (up to 96%). Extraction of squalene from yeast by supercritical carbon dioxide   总被引:2,自引:0,他引:2  
Squalene produced under anaerobic conditions, by a strain of Torulaspora delbrueckii was extracted from the biomass using supercritical carbon dioxide. Minimum use of solvent, lower time of isolation and a higher selectivity of extraction merit use of supercritical fluid extraction (SFE) technique over solvent extraction of squalene, as optimized and reported previously. A maximum squalene yield of 11.12 g g–1 (dry weight) of yeast cells was obtained at a temperature of 60 °C and pressure of 250–255 bar at a constant flow rate of 0.2l min–1 of carbon dioxide. Lyophilization prior to SFE increased the squalene yield to 430.52 g g–1 dry weight of yeast cells, an amount that is far greater than that obtained by (2:1) chloroform–methanol solvent extraction.  相似文献
