SYNOPSIS. A new peritrichous ciliate, Epistylis helicostylum n. sp. is described. It lives attached to the extremities of the ostracod Eucypris virens as its specific symphoriont. A spiral structure forming a protective theca for the colony of daughter animals is produced by the allometric growth of the outer sheath of the stalk. This review discusses the structure and properties of the isolated components of troponin, their interaction, and the mechanisms of regulation of contractile activity of skeletal and cardiac muscle. Data on the structure of troponin C in crystals and in solution are presented. The Ca2+-induced conformational changes of troponin C structure are described. The structure of troponin I is analyzed and its interaction with other components of actin filaments is discussed. Data on phosphorylation of troponin I by various protein kinases are presented. The role of troponin I phosphorylation in the regulation of contractile activity of the heart is analyzed. The structural properties of troponin T and its interaction with other components of thin filaments are described. Data on the phosphorylation of troponin T are presented and the effect of troponin T phosphorylation on contractile activity of different muscles is discussed. Comparative modeling methods are described that can be used to construct a three-dimensional model structure of a new protein from knowledge of its sequence and of the experimental structures and sequences of other members of its homology family. The methods are illustrated with the mammalian serine protease family, for which seven experimental structures have been reported in the literature, and the sequences for over 35 different protein members of the family are available. The strategy for modeling these proteins is presented, and criteria are developed for determining and assigning the reliability of the modeled structure. Criteria are described that are specially designed to help detect cases in which it is likely that the local structure diverges significantly from the usual conformation of the family.  相似文献

The most recently described member of the ErbB receptor tyrosine kinase family is ErbB-4. In general, the structure of this receptor and its mechanism of action is similar to that described for ErbB-1. However, significantly less is known about ErbB-4 and there are several novel aspects to its structure, mechanism of action, and biology. This includes the spectrum of ligands that activate ErbB-4, the presence of functionally distinct isoforms, a proteolytic processing pathway to the nucleus, and the capacity to induce a spectrum of cellular responses such as mitogenesis, differentiation, growth inhibition, and survival.  相似文献

A new octoflagellated species of Pyramimonas is described from three localities in Denmark. It is characterized by its ecology, being a marine psammophilic species associated with sand grains, and by details of the scaly covering on the cell surface. In the scale cover there are similarities to P. amylifera and to P. tetrarhynchus. P. octopus is distinguished by the possession of circular body scales of a type not previously found in the genus.
The general fine structure of the new species is described with emphasis on scale structure, the internal structure of the cell body and the flagella.  相似文献

The crystal structure of the monoclinic form of yeast phenylalanine tRNA has been redetermined at a resolution of 1.93 A. The structure of yeast tRNAphe described here is more accurate than its predecessors not only because it incorporates higher resolution data, but also because it has been refined using techniques that had not been developed when its predecessors were determined more than 20 years ago. The 1.93 A resolution version of this structure differs interestingly from its predecessors in its details. In loop regions particularly, the backbone torsion angles in the new structure are not the same as those reported earlier. Several new divalent cation binding sites have been identified, and the water structure that has emerged is also different.  相似文献

High resolution electron microscopy reveals that fully active alpha 2-macroglobulin (α2M) from fresh human plasma presents a very characteristic tetrameric structure. This native conformation of the α2M molecule is described here for the first time, along with its various orientations in negatively stained preparations. Although the native form is sensitive to inactivation, glutaraldehyde fixation is not necessary for its observation except when ammonium salts are used. The tetrameric structure of α2M undergoes a drastic conformational change when the protein is treated either with trypsin, thrombin or methylamine, as evidenced by the appearance of the typical)+(structure already described in the literature. The various aspects of this second conformation correspond to different orientations of the molecules in the stain film, and depend upon the nature of the support.  相似文献

The fine structure of the stinging cells (nematocytes) and stinging capsules (nematocysts) is described for Polypodium hydriforme. a freshwater coclenterate with a prominent endoparasitie stage in its life cycle. All the nematocysts belong to the type of lesser glutinants (atrichous isorhiza) and fall into three size classes. The internal structure of the capsules is similar in the three classes. A novel type of organization of the cnidocil apparatus of the nematocysts is described. The cnidocil lacks a root fibre and its kinctosome sits directly on the operculum of the nematocyst, so that the entire cnidocil apparatus has a radial rather than bilateral symmetry. It is compared with that of other types of nematocytes and its similarity with the mechanoreccptors of the coelentcratcs is noted. The possible place of the Polypodium nematocytes in the evolution of the collar receptors of the Metazoa is discussed.  相似文献

SYNOPSIS The fine structure of Pneumocystis carinii Delanoë is described in detail and is compared to the fine structure of protozoa and fungi. Pneumocystis does not have ultrastructural affinities to Protozoa but rather to fungi. Specifically, the process of intracystic body formation in the cyst is similar to the formation of ascospores inside a yeast ascus. The significance of the structural similarity between Pneumocystis and fungi is discussed. It is concluded from the ultrastructural evidence that Pneumocystis may indeed be a yeast or have a yeast-like stage in its life cycle. However, Pneumocystis is not completely similar to any fungus whose ultrastructure has been described so far. Perhaps its particular structure may be an adaptation to the parasitic way of life in mammalian lungs. It should be cultivated in order to be sure of its exact taxonomic position.  相似文献

In a previous paper (8) an organized structure was described in the macronuclei of certain old organisms of Tokophrya infusionum. It was found that the same honeycomb structure appears in great abundance in the macronuclei of overfed organisms. This permitted a better three-dimensional reconstruction of the described structure. Since the defined structure may be experimentally induced, it offers an opportunity for further more detailed studies as to its nature and meaning.  相似文献

A previously unrecognized bacterial flagellar structure present in Campylobacter jejuni and Campylobacter fetus is described. The structure is located just below the insertion of the flagellar apparatus through the bacterial wall. It is disc shaped, approximately 100 nm in diameter and has eleven spokes radiating from a central hole. The function of this new structure and its possible importance in the taxonomy of campylobacters and other spiral micro-organisms are discussed.  相似文献

What is considered to be Podochytrium cornutum Sparrow is recorded from the plankton of Crosemere, Cheshire, where it parasitizes the diatom Stephanodiscus rotula (Kütz) Hendey. In the main essentials of its sporangial development and its rhizoidal system the Crosemere material agrees well with that of the type described from U.S.A. Observations concerning dehiscence and the structure of living zoospores, previously unknown, have been made and resting spores are described for the first time.  相似文献

The detailed structure of the cockroach spermatheca is described and discussed firstly as an example of an insect integumentary gland, and secondly, from the standpoint of its role in reproduction. The gland comprises a cortical rank of separate secretory units, each associated with an epithelial duct cell responsible for receiving secreted materials and transporting them through the cuticular intima lining the reproductive tract. Secretory activity is cyclic, and the probable mode of elaboration and release of secretory material is described, together with the fine structure of the markedly differing intimas associated respectively with the glandular and conducting units.  相似文献

The fine structure of the mouthparts of the collembolan Folsomia Candida is described, largely on the basis of a study with the scanning electron microscope. The structure of the maxilla and of die mandibular molar plate is clarified and its interpretation corrected. The grinding function, usually attributed to the collembolan molar plate is disputed. This possible change in function of the mouthparts may also mean that these insects play a slightly different role than hitherto believed in the breakdown of the litter and humus in the soil.
An alternative method to replace freeze-drying in the preparation of Collembola, and possibly other small arthropods for scanning electron microscope studies is described.  相似文献
