Axinoscymnus cardilobus (Homoptera: Aleyrodidae) is an important predator of Bemisia tabaci (Coleoptera: Coccinellidae) that occurs in high population density of B. tabaci. Temperature among other factors is observed to play an important role in the development of arthropods. The effect of temperature on the development of A. cardilobus was studied at seven constant temperature regimes (14, 17, 20, 23, 26, 29, 32 °C). The results indicated that the duration of egg, larval and pupal stages were significantly influenced by increased temperature. The rate of development gradually increased with increase in temperature from 14 °C to 26 °C, but declined from 26 °C to 32 °C. The survival rates of different insect stages were stable at temperatures between 20 °C and 26 °C, but at extreme temperatures of 32 °C and 14 °C, a sharp decrease was evident. Ovipositional period of the female decreased when temperatures were increased from 17 °C to 32 °C. The highest fecundity of the female (225.7 eggs per female) was recorded at 23 °C. Life tables of A. cardilobus were constructed based on the experimental results at temperatures of 14–32 °C. The reproductive rate (R0), the innate capacity for increase (rm) and the finite rate of increase (λ) reached the maximum values at 23 °C, of 70.7, 0.059 and 1.062, respectively. The mean generation time (T) decreased with increased temperature from 17 °C to 32 °C, the highest and least values recorded at 17 °C and 32 °C were 112.7 and 38.7, respectively. These results offer valuable insight on the importation and establishment of A. cardilobus into new environments with diverse temperature regimes.  相似文献

Glucose self-exchange flux (Jex) and net efflux (Jnet) in human red cells and ghosts were studied at 25°C and pH 7.2 by means of the combined use of the Millipore-Swinnex filtering method and the continuous flow tube method to show the dependence of time of storage after aspiration, ATP and insulin. In fresh cells (RBC), ghosts (G), ghosts with 2 mM ATP (G+), and cells stored at 4°C > 60 days (OC) both Jex and Jnet follow simple Michaelis-Menten kinetics where

Jexmax and Jnetmax (nmol · cm−2 · s−1), respectively, was: (RBC) 0.27 and 0.19, (G) 0.24 and 0.27, (G +) 0.23 and 0.24, (OC) 0.23 and 0.20. and (mM), respectively, was: (RBC) 7.5 and 1.3, (G) 4.8 and 14.2, (G +) 11.6 and 6.8, (OC) 3.8 and 9.0. In ghosts, the ATP-dependent fraction of the permeability shows a hyperbolic dependence on glucose concentrations lower than 80 mM. Insulin up to 1 μM had effect on neither Jex nor Jnet in RBC. Based on reported values of cytochalasin B binding sites the turnover rate per site in RBC appears to be as high as in maximally insulin-stimulated fat cells. Our results suggest that the number of transport sites remains constant, independent of age, ATP and insulin.  相似文献

Changes were measured in the rates of respiration and in the levels of glycolytic intermediates during the first 5 min after addition of 1.6 mM glucose to a suspension (5%, v/v) of respiring Ehrlich ascites carcinoma cells incubated in an isotonic 50 mM tris(hydroxymethyl)methylglycine buffer (pH 7.4) at 38 °C. The rates of accumulation of lactate and glycolytic intermediates were used to calculate the in vitro velocities of glycolytic enzymes.The initial velocities of hexokinase (EC 2.7.1.1), fructose-6-phosphate kinase (EC 2.7.1.11) and lactate dehydrogenase (EC 1.1.1.27) in μmoles glucose equivalents/ ml cells per min were 14, 11 and 4, respectively. The velocities of the two kinases fell sharply to less than 5 between 5 and 10 s, while the velocity of the dehydrogenase declined gradually over the first minute. The initial burst of activity in the kinases, which lasted for about 8 s, was associated with a rapid accumulation of phosphate ester and a negative net ATP generation by glycolysis. The accumulation of phosphate ester is almost exactly matched by the generation of ATP by the “tail end” of glycolysis (triose-P to lactate) in this period. After this time (10–25 s) the rate of oxidative phosphorylation calculated as six times the rate of O2 consumption, is nearly identical to the combined rate of ATP utilization by hexokinase and fructose-6-phosphate kinase. As observed previously, oxamate (42 mM) blocked lactate dehydrogenase but did not depress the rate of phosphate ester accumulation.These various observations and correlations can be interpreted in terms of a dual glycolytic system. The accumulation of phosphate ester during the first 8 s is attributed to the operation of a partial glycolytic system, System B, which includes only the first three or four enzymes of glycolysis, and which draws upon an ATP pool (Pool I) previously employed in assorted cytoplasmic phosphorylations. The ADP generated by System B is rephosphorylated by and regulates the rate of a complete glycolytic system A, which converts glucose to lactate with little intermediate accumulation. The tail end of System A generates a new pool of ATP (Pool II) and controls the rate of glucose input through its head end, which is supplied by ATP being produced by oxidative phosphorylation. This scheme of interlocking controls is transient and alters after 8 s, when System B slows to a stop.  相似文献

Creatine kinase (CK) was isolated from the white muscle of the Antarctic icefish Chaenocephalus aceratus, which is deficient in glycolytic capacity. C. aceratus white myotomal creatine kinase (MMCK) displayed an apparent Km at 0.5 °C of 0.06 mM for ADP and 17 mM for Phosphocreatine. These Km values are similar to those reported for other vertebrate MMCKs at their physiologically relevant body temperatures. C. aceratus MMCK exhibited optimal activity at pH of 7.6–7.7 at 0.5 °C, in contrast to rabbit MMCK which had optimum activity at pH 6.2 at 30 °C. The apparent Vmax of C. aceratus MMCK at 0.5 °C is 94±4 S.D. (n=9) μmol ATP/min/mg (i.e. U/mg), which is comparable to rabbit MMCK assayed at 20 °C and 8-fold greater than rabbit MMCK measured at 0.5 °C. DEAE chromatography of C. aceratus white muscle CK resolved two distinct activity peaks. Cloning and sequencing of C. aceratus CK cDNAs confirmed that two muscle-specific isoforms of CK were expressed that were distinct from the mitochondrial and brain isoforms. Icefish MMCK was sensitive to transient temperature elevation, and the DEAE-fractionated forms were highly unstable. These results indicate that C. aceratus MMCK displays significant activity at physiological temperature and intracellular pH of icefish muscle that could contribute to sustaining energy charge during burst-swimming.  相似文献

We studied how the introduction of an additional ATP-consuming reaction affects the metabolic fluxes in Lactococcus lactis. Genes encoding the hydrolytic part of the F1 domain of the membrane-bound (F1F0) H+-ATPase were expressed from a range of synthetic constitutive promoters. Expression of the genes encoding F1-ATPase was found to decrease the intracellular energy level and resulted in a decrease in the growth rate. The yield of biomass also decreased, which showed that the incorporated F1-ATPase activity caused glycolysis to be uncoupled from biomass production. The increase in ATPase activity did not shift metabolism from homolactic to mixed-acid fermentation, which indicated that a low energy state is not the signal for such a change. The effect of uncoupled ATPase activity on the glycolytic flux depended on the growth conditions. The uncoupling stimulated the glycolytic flux threefold in nongrowing cells resuspended in buffer, but in steadily growing cells no increase in flux was observed. The latter result shows that glycolysis occurs close to its maximal capacity and indicates that control of the glycolytic flux under these conditions resides in the glycolytic reactions or in sugar transport.  相似文献

We studied the effect of exogenous adenosine in isolated perfused normoxic rat hearts on glycolytic flux through pyruvate kinase (PK). We compared its effect with that of myxothiazol, an inhibitor of mitochondrial ATP production. Moreover, we tested whether an increase of membrane ionic flux with monensin is linked to a stimulation of glycolytic flux through PK. After a 20-min stabilization period adenosine, myxothiazol or monensin were administrated to the perfusate continuously at various concentrations during 10 min. The contraction was monitored and the lactate production in coronary effluents evaluated. The amount of adenine nucleotides and phosphoenolpyruvate was measured in the frozen hearts. Myxothiazol induced a decrease of the left ventricular developed pressure (LVDP : −40%) together with a stimulation of glycolytic flux secondary to PK activation. In contrast, adenosine primarily reduced heart rate (HR: −30%) with only marginal effects on LVDP. This was associated with an inhibition of glycolysis at the level of PK. The Na+ ionophore monensin affected HR (+14%) and LVDP (+25%). This effect was associated with a stimulation of glycolysis secondary to the stimulation of PK. These results provide new information of action of adenosine in the heart and support the concept of a direct coupling between glycolysis and process regulating sarcolemmal ionic fluxes.  相似文献

Summary Wood frogs,Rana sylvatica, were sampled after freezing at –4°C (a short time course from 2 to 70 min after the appearance of the freezing exotherm) and thawing (20 h at 3°C after 70 min of freezing) and the regulation of liver glycolysis with respect to cryoprotectant glucose synthesis was examined. Within 5 min of the initiation of freezing, cryoprotectant concentrations in blood and liver had begun to increase. This was correlated with a rapid rise in the levels of hexose monophosphates in liver, including a 2.5 fold increase in glucose-6-P and 10 fold rise in fructose-6-P contents within the first 5 min post-exotherm. Contents of fructose-1,6-P2, fructose-2,6-P2, triose phosphates, P-enolpyruvate, and pyruvate did not significantly change over the course of freezing. Thawing sharply reduced the levels of hexose monophosphates in liver but raised P-enolpyruvate content by 2.3 fold. Changes in the contents of glycolytic intermediates over the freeze/thaw course are consistent with an inhibitory block of glycolysis at phosphofructokinase during freezing in order to facilitate a rapid glycogenolysis and production of cryoprotectant; during thawing, however, glycolysis appears to be inhibited at the level of pyruvate kinase.Possible regulatory control of cryoprotectant synthesis by covalent modification of liver glycolytic enzymes was examined. Glycogenolysis during freezing was facilitated by an increase in the percentage of glycogen phosphorylase in the activea (phosphorylated) form and also by an increase in the total amount (a+b) of enzyme expressed. For phosphofructokinase, kinetic changes as a result of freezing included a 40% reduction inK m for fructose-6-P, a 60% decrease inK a for fructose-2,6-P2, and a 2 fold increase in I50 for ATP. These changes imply a freezing-induced covalent modification of the enzyme but are not, apparently, the factors responsible for inhibition of glycolytic flux at the phosphofructokinase locus during glucose synthesis. Kinetic parameters of pyruvate kinase were not altered over the freeze/thaw course.  相似文献

The kinetic parameters of net exit of d-glucose from human red blood cells have been measured after the cells were loaded to 18 mM, 75 mM and 120 mM at 2°C and 75 mM and 120 mM at 20°C. Reducing the temperature, or raising the loading concentration raises the apparent Km for net exit. Deoxygenation also reduces the Km for d-glucose exit from red blood cells loaded initially to 120 mM at 20°C from 32.9 ± 2.3 mM (13) with oxygenated blood to 20.5 ± 1.3 mM (17) (P<0.01). Deoxygenation increases the ratio Vmax/Km from 5.29 ± 0.26 min−1 (13) for oxygenated blood to 7.13 ± 0.29 min−1 (17) for deoxygenated blood (P < 0.001). The counterflow of d-glucose from solutions containing 1 mM 14C-labelled d-glucose was measured at 2°C and 20°C. Reduction in temperature, reduced the maximal level to which labelled d-glucose was accumulated and altered the course of equilibration of the specific activity of intracellular d-glucose from a single exponential to a more complex form. Raising the internal concentration from 18 mM to 90 mM at 2°C also alters the course of equilibration of labelled d-glucose within the cell to a complex form. The apparent asymmetry of the transport system may be estimated from the intracellular concentrations of labelled and unlabelled sugar at the turning point of the counterflow transient. The estimates of asymmetry obtained from this approach indicate that there is no significant asymmetry at 20°C and at 2°C asymmetry is between 3 and 6. This is at least 20-fold less than predicted from the kinetic parameter asymmetries for net exit and entry. None of the above results fit a kinetic scheme in which the asymmetry of the transport system is controlled by intrinsic differences in the kinetic parameters at the inner and outer membrane surface. These results are consistent with a model for sugar transport in which movement between sugar within bound and free intracellular compartments can become the rate-limiting step in controlling net movement into, or out of the cell.  相似文献

Fructose-1,6-bisphosphate (FBP) aldolase, is a glycolytic enzyme that catalyzes the reversible condensation reaction of FBP to dihydroxyacetone phosphate (DHAP) and glyceraldehyde-3-phosphate (G3P). The aldolase gene from Aquifex aeolicus was subcloned, overexpressed in E. coli and purified to 95% homogeneity. The purified enzyme was activated by high concentrations of NH4+ and low concentrations of Co2+. The native molecular weight of the purified FBP aldolase was identified as 67 kDa (dimer) by gel filtration chromatography. The enzyme exhibits optimum pH at 6.5 and temperature at 90 °C. Based on the kinetic characterizations, the apparent Km was calculated to be 4.4 ± 0.07 mM, while Vmax was found to be 100 ± 0.02 μM min−1 mg protein−1. The recombinant protein showed extreme heat stability; no activity loss was observed even at 100 °C for 2 h. In addition, the thermophilic enzyme also showed higher stability against several organic solvents viz. acetonitrile, 1,4-dioxane, and methanol. With higher stability against both heat and organic solvents than any other class II aldolase, the A. aeolicus FBP aldolase is an attractive enzyme for use as a biocatalyst for industrial applications.  相似文献

The ratios of incorporation and retention of tritium compared to protium into metabolites in Chlorella pyrenoidosa and in Anacystis nidulans growing in water labeled with tritium have been determined. The algae were continuously supplied during growth with CO2 labeled with 14CO2, and the 14C content of metabolites were used to determined their concentrations. The tritium/protium ratios (R) of metabolites in Chlorella were determined following growth at 10 °C, 20 °C and 25 °C.As previously reported, variations in R in Chlorella, range from 0.5–0.7 for most metabolites, to values of R around 1 for metabolites of the tricarboxylic acid pathway. The R value for fumarate has now been measured. The increased R values for tricarboxylic acid cycle intermediates and related amino acids can be accounted for in terms of specific isotope effects of several enzyme-mediated steps. Very different R values for certain metabolites were found in A. nidulans. For example, R for citrate was 1.81 (the highest value observed in these studies) while aspartate was only 0.59, comparable to other metabolites in both organisms not related to the tricarboxyclic acid cycle. This lower value for aspartate is explainable in terms of the in complete tricarboxylic acid cycle in A. nidulans.No significant differences in R values for C. pyrenoidosa grown at 20 °C and 25 °C were observed, but in cells grown at 10 °C, there was a small but significant increase in R for tricarboxylic acid cycle metabolites.If the increase in R from sugar phosphates to tricarboxylic acid cycle intermediates seen in these two types of algae may be taken as an indication of likely discriminatory retention of tritium in organisms higher in the food chain, it would appear that no serious concentration of tritium due to isotopic discrimination should occur in the biosphere. However, research workers using compounds labeled with hydrogen isotopes for studies of in vivo metabolism should take into account the likelihood of such discriminatory uptake and retention during specific metabolic steps.  相似文献

To investigate the short-term (30–240 min) interactions among nitrogenase activity, NH4+ assimilation, and plant glycolysis, we measured the concentrations of selected C and N metabolites in alfalfa (Medicago sativa L.) root nodules after detopping and during continuous exposure of the nodulated roots to Ar:O2 (80:20, v/v). Both treatments caused an increase in the ratios of glucose-6-phosphate to fructose-1,6-bisphosphate, fructose-6-phosphate to fructose-1,6-bisphosphate, phosphoenolpyruvate (PEP) to pyruvate, and PEP to malate. This suggested that glycolytic flux was inhibited at the steps catalyzed by phosphofructokinase, pyruvate kinase, and PEP carboxylase. In the Ar:O2-treated plants the apparent inhibition of glycolytic flux was reversible, whereas in the detopped plants it was not. In both groups of plants the apparent inhibition of glycolytic flux was delayed relative to the decline in nitrogenase activity. The decline in nitrogenase activity was followed by a dramatic increase in the nodular glutamate to glutamine ratio. In the detopped plants this was coincident with the apparent inhibition of glycolytic flux, whereas in the Ar:O2-treated plants it preceded the apparent inhibition of glycolytic flux. We propose that the increase in the nodular glutamate to glutamine ratio, which occurs as a result of the decline in nitrogenase activity, may act as a signal to decrease plant glycolytic flux in legume root nodules.  相似文献

In vitro studies were undertaken to determine the effect of pH, temperature, water availability and carbon dioxide (CO2) concentration on germination and growth of Colletotrichum musae, the causal pathogen of anthracnose of bananas. The optimum pH for germination and growth varied between 4·0 and 5·0 depending on temperature. At low pH (< 3·0) and 15°C, both germination and growth were significantly reduced, with a marked increase in the lag time, in days, prior to growth. C. musae germinated and grew over a wide range of water activities (aw; 0·995−0·94 and 0·995−0·92, respectively) at 20, 25 and 30°C. In all cases where germination occurred appresoria were subsequently produced. Optimum growth occurred at 30°C and 0·995 aw, although this changed to 0·98 aw at 35°C. Increasing CO2 concentration to 15% or reducing oxygen concentration to 1% resulted in a significant (P < 0·05) reduction in growth, but did not inhibit growth completely.  相似文献

A network model for the determination of tumor metabolic fluxes from 13C NMR kinetic isotopomer data has been developed and validated with perfused human DB-1 melanoma cells carrying the BRAF V600E mutation, which promotes oxidative metabolism. The model generated in the bonded cumomer formalism describes key pathways of tumor intermediary metabolism and yields dynamic curves for positional isotopic enrichment and spin-spin multiplets. Cells attached to microcarrier beads were perfused with 26 mm [1,6-13C2]glucose under normoxic conditions at 37 °C and monitored by 13C NMR spectroscopy. Excellent agreement between model-predicted and experimentally measured values of the rates of oxygen and glucose consumption, lactate production, and glutamate pool size validated the model. ATP production by glycolytic and oxidative metabolism were compared under hyperglycemic normoxic conditions; 51% of the energy came from oxidative phosphorylation and 49% came from glycolysis. Even though the rate of glutamine uptake was ∼50% of the tricarboxylic acid cycle flux, the rate of ATP production from glutamine was essentially zero (no glutaminolysis). De novo fatty acid production was ∼6% of the tricarboxylic acid cycle flux. The oxidative pentose phosphate pathway flux was 3.6% of glycolysis, and three non-oxidative pentose phosphate pathway exchange fluxes were calculated. Mass spectrometry was then used to compare fluxes through various pathways under hyperglycemic (26 mm) and euglycemic (5 mm) conditions. Under euglycemic conditions glutamine uptake doubled, but ATP production from glutamine did not significantly change. A new parameter measuring the Warburg effect (the ratio of lactate production flux to pyruvate influx through the mitochondrial pyruvate carrier) was calculated to be 21, close to upper limit of oxidative metabolism.  相似文献

The thermal coefficient of expansion of egg lecithin bilayer thickness, αd1, was measured as a function of its cholesterol content up to mole ratio lecithin/cholesterol of 1:1, and over the temperature range 0–40 °C. At all cholesterol contents αd1 changes abruptly at approximately 12 °C indicating a structural transition at this temperature. Above 12 °C, αd1 decreases monotonically from −2·10−3 for pure egg lecithin to −1·10–3 at mole ratio 1:1. Below 12 °C αd1 is walways higher than above 12 °C and shows a sharp, anomalously high value of −6·10−3 at the mole ratio 2:1. The results have been interpreted as the movement of cholesterol into the bilayer or the formation of lecithin-cholesterol “complexes” at temperatures below 12 °C. Similar studies with phosphatidylinositol containing cholesterol showed no structural transition and lysolecithin containing cholesterol behaved differently giving two lamellar phases in equilibrium.  相似文献

The reproductive and developmental biology of Gonatocerus ashmeadi Girault, a parasitoid of the glassy-winged sharpshooter Homalodisca coagulata (Say), was determined at five constant temperatures in the laboratory: 15; 20; 25; 30; 33 °C. At 30 °C, G. ashmeadi maintained the highest successful parasitism rates with 46.1% of parasitoid larvae surviving to adulthood. Lifetime fecundity was greatest at 25 °C and fell sharply as temperature either increased or decreased around 25 °C. Temperature had no effect on sex ratio of parasitoid offspring. Mean adult longevity was inversely related to temperature with a maximum of 20 days at 15 °C to a minimum of eight days at 33 °C. Developmental rates increased nonlinearly with increasing temperatures. Developmental rate data were fitted with the modified Logan model for oviposition to adult development times across each of the five experimental temperatures to determine optimal and upper lethal temperature thresholds. The lower developmental threshold estimated by the Logan model and linear regression were 1.10 and 7.16 °C, respectively. Linear regression of developmental rate for temperatures 15–30 °C indicated that 222 degree-days were required above a minimum threshold of 7.16 °C to complete development. A temperature of 37.6 °C was determined to be the upper development threshold with optimal development occurring at 30.5 °C. Demographic parameters were calculated and pseudo-replicates for intrinsic rate of increase (rm), net reproductive rates (Ro), generation time (Tc), population doubling time (Td), and finite rate of increase (λ) were generated using the bootstrap method. Mean bootstrap estimates of demographic parameters were compared across temperatures using ANOVA and nonlinear regression.  相似文献

Chloroplasts or chromoplasts were purified from sweet-pepper (Capsicum annuum L. cv. Yolo Wonder) fruits and analysed with respect to their enzymic equipment, the transport properties across the envelope membrane, and for the presence of a functional oxidative pentose-phosphate pathway (OPPP). It was demonstrated that both types of plastid contain enzyme activities that allow glycolysis and OPPP. During the developmental conversion from chloroplasts to chromoplasts the activities of enzymes catalysing potentially rate-limiting reactions in glycolysis increased considerably. Most enzyme activities involved in the plastidic OPPP stayed constant or decreased during ripening, but transaldolase activity increased by more than 500%. To analyse whether pepper fruit chromoplasts are able to use exogenously supplied carbohydrates for the OPPP we measured the rate of 14CO2 release after application of radioactively labelled precursors. Isolated pepper fruit chromoplasts used exogenously supplied [U14C]glucose- 6-phosphate (Glc6P) as a precursor for the OPPP. The metabolic flux through this pathway was stimulated by the presence of additional compounds which require reducing equivalents for further conversion, e.g. nitrite, or 2-oxoglutarate plus glutamine. The [14C]Glc6P-driven OPPP in isolated chromoplasts exhibited saturation with rising concentrations of Glc6P, reaching highest rates at an external concentration of about 2 mM. Exogenously given [U14C]glucose 1-phosphate (Glc1P)′ did not lead to a release of 14CO2, indicating that this hexose phosphate is not taken up into the intact plastid. Using a proteoliposome system in which the envelope membrane proteins from sweet-pepper chromoplasts were functionally reconstituted we demonstrated that Glc6P is transported in counter-exchange with inorganic phosphate (Pi) or other phosphorylated intermediates. The Glc6P was taken up into proteoliposomes with an apparent K m of 0.34 mM. Surprisingly, in contrast to tomato fruit plastids, isolated chromoplasts from sweet-pepper fruits do not possess a phosphate translocator allowing the uptake of Glc1P. Rising exogenous concentrations of dihydroxyacetone phosphate strongly inhibited the metabolic flux through the OPPP. This observation is discussed with respect to the presence of two phosphate translocator proteins in the envelope of sweet-pepper chromoplasts and with respect to possible metabolic changes occurring in heterotrophic tissues during development. Ehrlich ascites carcinoma cells depleted of K+ and provided with 5.5 mM K+ in isosmotic 50 mM tris(hydroxymethyl)methylglycine buffer at pH 7.4 and 38 °C take up K+ from the medium at a rate of 6 μmoles/ml intracellular fluid per min. Depleted cells exposed to K+ for 2 min prior to glucose addition exhibit a higher initial rate of glycolysis, a lower glycose-6-P accumulation, and a higher fructose-1,6-P2 accumulation than depleted cells incubated in a K+-free medium. Both the K+ transport and the effect of K+ on glycolysis are blocked by 2 mM oubain.Calculation of thein vitro velocities of glycolytic enzymes from the rates of accumulation of lactate and glycolytic intermediates shows that the presence of K+ accelerates the velocities of fructose-6-phosphate kinase and lactate dehydrogenase about 2-fold and the velocity of hexokinase about 1.5-fold during the first 15 s. In either the presence or absence of K+, the hexokinase velocity is highest immediately after glucose addition and declines sharply with time; this decline is greater than would be predicted by product inhibition by the accumulated glucose-6-P. The maximal stimulation of fructose-6-phosphate kinase attibutable to the increasing intarcellular K+ concentration is only 1.25-fold. These observations indicate that the initial acceleration in glycolysis is not simply mediated through a direct K+ activation of fructose-6-phosphate kinase.The calculated theoretical rate of ATP generation by glycolysis shows that glycolysis is an ATP-utilizing system for the first 5–10 s both in the presence and in the absence of K+. Hence, the initial stimulation of glycolysis by K+ is not a consequence of an increased rate of ATP hydrolysis associated with K+ transport, although this mechanism may be responsible for the stimulation of steady-state glycolysis.The initial rate of phosphate ester (hexose and triose phosphates) accumulation corresponds to be rate of ATP generation by the “tail-end” of glycolysis, or twice the rate of lactate accumulation, in either the absence or presence of K+, but both the rate and the maximal level of ester accumulated are higher in the presence of K+. This implies that the oxidatively generated pool of ATP which is diverted from endogenous reactions to hexokinase and fructose-6-phosphate kinase on the introduction of glucose is larger in the presence of K+.Valinomycin (0.27 μM) under certain conditions can produce effects on the glycolysis of non-depleted cells which superficially resemble the effects of K+ on depleted cells. However, unlike K+, valinomycin stimulates the initial rate of glycolytic ATP generation, and abolishes the initial correspondence between the ATP generation by the “tail-end” of glycolysis and phosphate ester accumulation. These observations are interpreted to mean that valinomycin introduces an ATPase activity effective on glycolytically generated ATP.Comparison of the theoretical ATP generation in the presence and absence of K+ indicates that approximately one ATP is hydrolyzed for each K+ transported.  相似文献

Preimaginal development and adult longevity and reproduction of Dichochrysa prasina Burmeister were studied at six constant temperatures (15, 20, 25, 27, 30 and 33 °C) and a photoperiod of 16:8 (L:D). Eggs of the flour moth Ephestia kuehniella (Zeller) were used as food throughout preimaginal development, whereas the adults of D. prasina fed on a liquid diet of water, yeast hydrolysate, sugar and honey. At the highest tested temperature of 33 °C no larvae completed their development. At the rest of the tested temperatures the egg to adult developmental period ranged from approximately 92 days at 15 °C to 25 days at 30 °C. Percentages of adult emergence ranged from 36% at 15 °C to 84% at 30 °C. Both adult longevity and fecundity were significantly affected by temperature and the intrinsic rate of increase (rm) reached its maximum value at 27 °C. These results could be useful for the establishment of a small scale rearing and mass production of D. prasina.  相似文献

Fourier transform infrared spectroscopy was used to characterize the lamellar phases of 1,3-dipalmitoyl-sn-glycero-2-phosphocholine (1,3-DPPC), a positional isomer of 1,2-dipalmitoyl-sn-glycero-3-phosphocholine (1,2-DPPC). The molecule exists in three distinct phases over the temperature interval 0–70°C. In the low-temperature (Lc) phase, the spectra are indicative of acyl chains packed in an orthorhombic subcell, while the carbonyl groups and phosphate ester at the head group show evidence of only partial hydration. The transition from the low-temperature (Lc) phase to the intermediate-temperature (Lβ) phase at 25°C corresponds to a temperature-induced head-group hydration in which the hydration of the phosphate and carbonyl ester groups results in the reorganization of the hydrocarbon chain-packing subcell from orthorhombic to hexagonal. The transition from the intermediate (Lβ) to the high-temperature (Lα) phase at 37°C is a gel-to-liquid-crystalline phase transition analogous to the 41.5°C transition of 1,2-DPPC. The spectra of the acyl-chain carbonyl groups show evidence of significant differences in molecular conformation at the carbonyl esters in the Lc phase. In the Lβ and Lα phases, the carbonyl band contour becomes much more symmetric. However, two components are clearly present in the spectra indicating that the sn-1 and sn-3 carbonyls experience slightly different environments. The observed differences are likely due to a preferred conformation of the phosphocholine group relative to the glycerol backbone. Indications from the infrared spectra of differences in the structure of the C=O groups provide a possible explanation for the selection of the sn-1 chain of 1,3-DPPC by phospholipase A2 on the basis of a preferred head group conformation.  相似文献

The toxic effects of high pressure oxygen on the isolated toad urinary bladder have been studied. Sodium transport in this system is reversibly inhibited by high pressure O2. This inhibition is potentiated by adrenal steroid hormones, and occurs despite both increased glycolytic and Kreb's cycle flux and tissue ATP content. High pressure O2 leads to increased pyruvate/lactate and pyruvate/malate redox couples, as well as to a decrease in the weight percentage of phospholipid long-chain unsaturated fatty acids and [2-14C]pyruvate incorporation into tissue lipid. During recovery from high pressure O2 treatment, [2-14C]Pyruvate incorporation into lipid is increased and the weight percentage of long-chain unsaturated fatty acids increases. These data indicate that high pressure O2 poisoning in this tissue does not result from an inhibition of carbohydrate metabolism, but may result from the formation of toxic lipid peroxides.  相似文献
