To find a simple and reliable oxygen electrode-based method to estimate the values of alternative pathway activity (V alt) and its contribution to total respiration V alt/V t) in aged potato (Solanum tuberosum L.) tuber slices, we compared conventional hydroxamate-inhibiting method, improved hydroxamate-inhibiting method with 2,6-dichlorophenol indophenol (DCPIP), and the oxygen isotope discrimination (OID) method. The values of V alt and V alt/V t obtained with an improved hydroxamate-inhibiting method with DCPIP in 12-h- and 24-h-aged slices were about twice higher than those with the conventional hydroxamate-inhibiting method. Only a relatively small difference in the values of V alt and V alt/V t obtained by the OID method and the improved hydroxamate-inhibiting method with DCPIP in 12-h and 24-h-aged slices was observed. Twelve peptides, including eight conservative amino acid residues in the amino acid sequence of hydrophilic S helix of the alternative oxidase (AOX), were synthesized by solid-phase method. The polypeptide was coupled with α-chymotrypsinogen, and the antibodies were obtained through immunizing domestic rabbit by injecting this complex. By using these antibodies, which were raised to immunoreact with total proteins of purified mitochondria from different organs of mung bean seedlings, we find that there are two hybridizable AOX bands in mitochondria. Their molecular weights are about 35 and 38 ku, respectively. Moreover, the respiratory parameters of hypocotyl, true leaf and cotyledon of mung bean seedlings show that true leaf has the highest total respiration (Vt), alternative pathway (AP) capacity (Valt) and the activity of AP (ρValt) among the three organs. Vt and ρValt of cotyledon ranked the second. Hypocotyl has the lowest V, and ρValt, but its Valt is higher than that of cotyledon. These result  相似文献

Green soybean cells (Glycine max L.) were grown in suspension culture on B5-medium containing various glucose concentrations (2%, 4% or 8% glucose and 2% glucose + 2% or 6% mannitol). Respiratory parameters have been determined in these cells in relation to growth.The length of the growth phase and the amount of dry weight produced increased with higher glucose concentrations. In cells cultured in the presence of 2% glucose, 4% glucose or 2% glucose + 2% mannitol, the activity of the cytochrome pathway (V cyt )and the capacity of the alternative pathway (V alt )were not significantly different. Also the contribution of the alternative pathway to total respiration was essentially the same.The glucose content of cells grown in 2% or 4% glucose was roughly equal, which might explain the absence of any effect of the increased glucose concentration on the respiratory parameters.In cells grown in 8% glucose or on 2% glucose + 6% mannitol, both the activity of the cytochrome pathway and the capacity of the alternative pathway were lower. High values of (approaching 1) were found in these cells, meaning that the alternative pathway is nearly fully operative. This is probably caused by the impairment of the cytochrome pathway due to the high osmotic value of the medium.ATP-production per batch per day has been calculated from oxygen uptake data, integrated and compared with cellular dry weight production to give values of 13-15.3 g dry weight produced mole ATP-1.The described experiments produce no direct evidence for a function of the alternative pathway as energy overflow in soybean cells. The callus of Nicotiana rustica cv Gansu yellow flower and N. tabacum cv willow leaf were cultured on ordinary subculture medium (M-1) and on regeneration medium (M-2), respectively. No differentiation was observed in Gansu yellow flower tobacco callus cultures grown on both M-1 and M-2 medium. The respiration of both cultures was partially resistant to cyanide and markedly inhibited by m-chlorobenzhydroxamic acid. The relative contributions of alternative and cytochrome pathway were 31% and 47% of the total respiration, respectively, in M-1 callus cultures. The relative O2 uptake of the two pathways was not changed significantly in M-2 callus cultures. In subcultured M-1 callus cultures of Willow leaf tobacco, the respiration mediated via alternative pathway was about 29 to 38% of the total respiration, and the cytochrome pathway still was the major respiratory pathway. In M-2 callus cultures in which differentiation occurred, the relative contribution of alternative pathway increased to 41 to 47% of the total respiration, and the cytochrome pathway decreased considerably. These results suggested that the change of respiratory electron transport pathway was probably related to the differentiation of tobacco callus cultures.  相似文献

Total respiration (vT) increased after exposure to UV, but a decrease in the capacity of SHAM-sensitive-alternative respiration (Valt) was accompanied by an increase in residual respiration (vres). The capacity for CN sensitive-cytochrome c respiration (Vcyt) was not inhibited by UV-A. After 4 h of irradiation of high-CO2-grown cells of Chlamydomonas reinhardtii with UV-A (2 μW. CM?2) in the presence of white light (300μE.m?2.s?1), the capacity of Vast was reduced from 10 to 4 μmol O2. mg?1Chl.h?1, a 60 % reduction. After a similar exposure to UV-A, the capacity of Valt in pea leaves was reduced from 13 to 5 μmol O2.g?1 fr wt.h?1. Exposure to UV-C was not inhibitory, but UV-B caused up to 25% inhibition of the Valt. Twenty to 48 h after exposure to UV-A radiation, the capacity of alternative respiration had recovered. UV-A inhibition of the alternative respiration was consistent with UV-A absorption by quinones, except that UV-A did not inhibit the cyt c pathway of electron transport that also involves the ubiquinones.  相似文献

Treatment of intact potato (Solanum tuberosum L.) tubers with acetaldehyde, ethanol or acetic-acid vapors led to a respiratory upsurge which was further increased when the volatiles were applied in 100% O2. Mitochondria from tubers held in 100% O2 (O2 control) displayed a substrate state, state 3, and state 4 in respiration, whereas in mitochondria from the volatile-treated tubers the respiratory rate of the different states was virtually indistinguishable. This respiratory pattern was companied by the development of a cyanide-resistant respiration since these mitochondria exhibited resistance to CN and sensitivity to CN+salicylhydroxamic acid. Callus-forming discs from potato tubers lose 80% of their starch during one month of incubation on nutrient medium containing either 0, 3 or 6% (w/v) sucrose. The content of soluble sugar in the discs varies from 5 mg (incubated without sucrose) to 22 mg (on 3% sucrose) and 40 mg (on 6% sucrose) per g fresh weight. The activity of the cytochrome pathway (Vcyt) increases during the first week of incubation on all media. Thereafter Vcyt decreases again on 0% sucrose medium, while it remains constant on 3 and 6% sucrose media. Alternative pathway capacity (Valt), absent in freshly sliced tissue, shows a sharp increase during the first days of incubation, independent of the sucrose concentration in the medium. This capacity further increases during prolonged incubation on 3 and 6% sucrose but decreases on 0% sucrose. The in vivo activity of the alternative pathway (the participation in uninhibited respiration, ?Valt) varies with the sucrose concentration and with the culture time. In tissue incubated for 2-3 weeks on 6% sucrose as much as 45% of the electrons are transfered to oxygen via the alternative pathway. In this tissue the factor Q (the part of the alternative pathway capacity that is operative) is about 0.8, while in tissue incubated on 0 and 3% sucrose media p generally does not exceed 0.5. When chloramphenicol, an inhibitor of mitochondrial protein synthesis, is added to the medium together with 3% sucrose, the increase in Vyet does not occur, while the induction of Valt during the first week of incubation is the same as without chloramphenicol. A greater part of the alternative pathway capacity becomes operative in this tissue, leading to values of Q of almost 1 after prolonged incubation. Osmotic dehydration of wheat seedlings in-0.5 MPa polyethylene glycol (PEG) solutions for 24, 48 and 72 h resulted in mild, moderate and severe water stress respectively in leaves, but only caused mild water stress in roots as reflected by the changes in relative water content (RWC). In response to the above water stress conditions, leaf total respiratory rate (V_t) decreased progressively, and the alternative pathway (AP) capacity (V_(alt)) and its actual operation activity (ρV_(alt)) decreased more severely. Water stress also led to continuous reduction in cytochrome pathway (CP) activity ((ρ' V_(cyt)) and different changes in the contribution of ρV_(alt) and ρ' V_(cyt) to V_t in leaves, with ρV_(alt)/V_t decreasing and ρ' V_(cyt)/V_t increasing. The change pattern of root V_t was similar to that of its RWC, while root V_(alt) and ρV_(alt) were found to decrease during the first 24 h of stress and thereafter recover to a level close to that of the control (O h). 为了进一步了解光照下植物呼吸作用的内在机理以及呼吸作用和光合作用的关系,该文研究了在光照下菜豆(Phaseolus vulgaris)叶片抗氰呼吸与光合作用的关系。研究发现,将黑暗下生长的菜豆幼苗叶片转到光照下10 h,总呼吸、抗氰呼吸以及抗氰呼吸在总呼吸中的比例均逐步上升;光照也导致了叶片叶绿体光合放氧和CO2固定的出现及其速率的增加,但光合放氧和CO2固定速率的增加均滞后于抗氰呼吸的增加。将黑暗下生长的叶片转到光照下之前用抗氰呼吸的抑制剂水杨基氧肟酸(SHAM)处理叶片,发现用SHAM处理并没有导致叶片在光照下光合放氧和CO2固定速率的明显变化,这也提示了黑暗下生长的叶片转至光照的过程中,抗氰呼吸和光合作用没有产生偶联。进一步研究发现,在黑暗中对叶片施加短时间的光照能够增加抗氰呼吸在总呼吸中的比例,但短时间的光照对叶片光合CO2固定速率没有影响。这些结果表明了光照对抗氰呼吸的诱导可以不依赖于光合作用,光照可能是作为一种直接的信号去诱导抗氰呼吸。  相似文献

Dark respiration in the red macroalga Chondrus crispus was studied under a variety of conditions. The components of respiration were examined using selective inhibitors in order to characterise pathways of respiration and examine regulation of respiration in marine macroalgae.In comparison to respiration rates generally reported for higher-plant leaves and roots, the steady-state rate of O2 consumption by this alga, after 30 min dark pretreatment, was found to be quite low (three- to sixfold lower than in higher plants). The addition of uncoupler had only a slight effect on the basal respiration rate, indicating that in these conditions, substrate supply could be limiting respiration. The addition of KCN inhibited respiration by approx. 60%, indicating the presence of alternative oxidase activity. The coefficient of engagement of the alternative pathway (calculated from the data herein) showed that under normal conditions there was little participation of the alternative pathway in O2 consumption. The response of respiration to O2 tension was examined with and without inhibitors and the apparent K m was 17 to 21 M. The addition of KCN plus salicylhydroxamic acid almost completely blocked respiration in C. crispus. The hypothesis that respiratory substrate limits respiration in this alga was investigated by measuring respiration rates immediately after periods of photosynthetic activity. It was found that the respiration rate was dependent on the duration of the light period and could increase up to twofold. This stimulated rate of respiration declined in a first-order fashion during the next 20 to 30 min, finally reaching the basal, zero-order rate measured before illumination. These results strongly indicate a change in the nature of the respiratory substrates during this period. An 15N off-resonance R 1 spin relaxation study of an L99A point mutant of T4 lysozyme is presented. Previous CPMG-based relaxation dispersion studies of exchange in this protein have established that the molecule interconverts between a populated ground state and an excited state (3.4%) with an exchange rate constant of 1450 s–1 at 25°C. It is shown that for the majority of residues in this protein the offset dependence of the R 1 relaxation rates cannot be well fit using models which are only valid in the fast exchange regime. In contrast, a recently derived expression by Trott and Palmer (J. Magn. Reson., 154, 157–160, 2002) which is valid over a wider window of exchange than other relations, is shown to fit the data well. Values of (signed) chemical shift differences between exchanging sites have been extracted and are in reasonable agreement with shift differences measured using CPMG methods. A set of simulations is presented which help establish the exchange regimes that are best suited to analysis by off-resonance R 1 techniques.  相似文献

The present study in solid-liquid contactors, an attempt has been made to calculate the minimum/critical impeller speed required for complete suspension of solids. A new correlation, involving Reynolds number, modified Froude number, along with the agitation characteristics is proposed. Cyanide-resistant, alternative respiration in Nicotiana tabacum L. cv Xanthi-nc was analyzed in liquid suspension cultures using O2 uptake and calorimetric measurements. In young cultures (4-8 d after transfer), cyanide inhibited O2 uptake by up to 40% as compared to controls. Application of 20 μm salicylic acid (SA) to young cells increased cyanide-resistant O2 uptake within 2 h. Development of KCN resistance did not affect total O2 uptake, but was accompanied by a 60% increase in the rate of heat evolution from cells as measured by calorimetry. This stimulation of heat evolution by SA was not significantly affected by 1 mm cyanide, but was reduced by 10 mm salicylhydroxamic acid (SHAM), an inhibitor of cyanide-resistant respiration. Treatment of SA-induced or uninduced cells with a combination of cyanide and SHAM blocked most of the O2 consumption and heat evolution. Fifty percent of the applied SA was taken up within 10 min, with most of the intracellular SA metabolized in 2 h. 2,6-Dihydroxybenzoic and 4-hydroxybenzoic acids also induced cyanide-resistant respiration. These data indicate that in tobacco cell-suspension culture, SA induces the activity and the capacity of cyanide-resistant respiration without affecting the capacity of the cytochrome c respiration pathway.  相似文献

Total dark respiration (vt), cytochrome pathway (veyt), alternativepathway (valt) and residual (vres) respiration were measuredin Lemna gibba plants, by the use of pathway inhibitors. NaCNwas used to inhibit veyt and SHAM (salicylhydroxamic acid) toinhibit valt. Residual respiration (vres) was about 5% of vt.The effect of high (100 Pa) and low (0 Pa) carbon dioxide partialpressure ([CO2]) on vt, veyt and valt was determined from bothCO2 efflux and O2 uptake measurements. The higher [CO2] suppressedvt by about 30%. When respiration operated through the cytochromepathway only, in the absence of valt, it was suppressed by about12% as measured by the O2 uptake of submerged Lemna fronds orby about 40% as measured by CO2 efflux from floating fronds.The higher [CO2] treatment had only a small effect on respiration,when valt alone operated. There was no evidence of a specificsuppression of the valt pathway by high [CO2]. Succinic dehydrogenaseactivity of the mitochondria of roots of Medicago sativum wasreduced by 18%, when the mitochondria were pre-treated with120 as compared to 34 Pa [CO2]. There was no such effect oncytochrome c oxidase activity of mitochondria under the sameconditions. Growth and oxygen uptake of potato callus is faster in an oxygen-enriched atmosphere (70% oxygen, v/v; oxygen-callus) than in air (20% oxygen, v/v; air-callus). Especially the non-mitochondrial, so-called residual respiration is increased in oxygen-callus. The capacities of the mitochondrial respiratory pathways (cytochrome pathway, Vcyt and alternative pathway, Valt) are also higher in this callus. In both callus types only a small part of the alternative pathway capacity is used in uninhibited respiration. The lower oxygen uptake of air-callus at normal air oxygen pressures is partially due to diffusional impedance. Measurement of the respiratory parameters of air-callus in oxygen-saturated medium leads to higher values than measurement in air-saturated medium, although these values are still lower than those of oxygen-callus.ATP-production was calculated from the oxygen-uptake data and compared with the dry weight production of the callus to give values of 10.0 and 10.8 g dry weight produced.-mol ATP-1, for air-callus and oxygen-callus respectively. Several screening methods at the so-called ready biodegradability level are suitable to test poorly soluble substances. Typical for these tests is that mineralization is evaluated from monitoring oxygen uptake or carbon dioxide production. Unfortunately, they suffer from a rather low precision in the calculated percentage of mineralization caused by subtracting a too high inoculum control measurement from the response in the test system. Criteria for blank oxygen consumption, due to the metabolic activity of the inoculum, are proposed from which maximum amounts of activated sludge or secondary effluent per litre test medium can be derived to be used as an appropriate inoculum. Both for current and future standardized tests the precision of the method can be kept within acceptable margins. Inoculum material was sampled from 40 communal biological waste water treatment plants.
