We here describe further studies on the action of bonellin on sea-urchin eggs. Bonellin brings about Some of the changes that are known to occur in the egg upon fertilization. In particular, it appears to cause the increased rate of incorporation of amino acids into proteins, the increase of the voltage noise, and the exocytosis of some of the cortical granules. A comparison with the effect of ammonia is discussed. 相似文献

Bonellin, a chlorin extracted fromBonellia viridis (Echiura), and hemaatoporphyrin exhibit a strong antibiotic and bactericidal activity on marine and terrestrial bacteria. This action is enhanced by light. Oxygen consumption and motility of bacteria are also inhibited, while no chemotactic effects are observed. The drugs induce lysis onBacillus subtilis protoplasts, but they are ineffective onMicrococcus lysodeikticus protoplasts.The results are discussed and compared with those obtained with eukariotic cells. Several naturally-occurring lipids but not n-propanol, guanidine-HCl or a variety of synthetic detergents stimulate the 3′,5′-cyclic AMP-phosphodiesterase activities of a supernatant fraction of brain at 1.25 × 10?7 M cAMP. The time courses of the reaction are linear in the presence and absence of lipid. On the other hand, lipid has different effects on various phosphodiesterase activities in fractions obtained after gel filtration of the crude extract. It stimulates the phosphodiesterase activities measured at 1.25 × 10?7 M and 10?4 M 3′,5′-cyclic-AMP and 1.25 × 10?7 M 3′,5′-cyclic GMP in two of the fractions partially retained in the gel. However, lipid has little effect on the enzymatic hydrolysis of low concentrations of cAMP or cGMP and markedly inhibits the hydrolysis of high concentrations of cAMP by the fraction excluded from the gel. 相似文献

In this paper we report on the mechanism of action of bonellin on sea urchin eggs. Bonellin acts in the same way as porphyrins, ie, by causing the crosslinking of membrane proteins and the peroxocompounds formation. Removal of lipids from submitochondrial particles or detergent-solubilized mitochondrial preparations of rat liver resulted in a 90% loss of ferrochelatase (protochemeferro-lyase, EC 4.99.1.1) activity. The addition of either a fatty acid or phospholipid restored enzyme activity; the extent of reactivation being correlated with the degree of unsaturation of the fatty acid or acyl chain and independent of the polar head group of the phospholipid, Arrhenius plots of the ferrochelatase activities of submitochondrial particles and detergent-solubilized mitochondrial preparations showed transition temperatures of 37 and 28.5 degrees C, respectively. Ferrochelatase of submitochondrial particles or detergent-solubilized preparations had an absolute requirement for Ca2+. The ferrous salt of oxalic acid, a Ca2+ chelator, was a very poor substrate for these preparations. In contrast, ferrochelatase activities of fatty acid- or lipid-supplemented acetone extracts of these preparations were not dependent on the presence of Ca2+ and ferrous oxalate served as substrate for these extracts.20 相似文献

(particularly in nonpigmented yeasts). In carotene-synthesizing yeasts, these Stressors decreased the amount of carotenoids

Three-week-old plants of safflower (Carthamus tinctorius L.) were subjected to a water-deficit stress. The lipid composition of the shoot parts of both control (well-watered) and water stressed plants was analyzed. Experimental data revealed that moderate stress induced an increase in total lipid content within all lipidic classes. However, severe water-deficit induced a sharp decrease in the total lipid content and specially in polar lipids, particularly in phosphatidylethanolamine, phosphatidylcholine, monogalactosyl-diacylglycerol and digalactosyl-diacylglycerol. Also, the content of neutral lipids was increased. Concerning the fatty acid composition, water-deficit induced a decrease in their degree of unsaturation expressed by a reduction in the proportions of linolenic (18:3) and linoleic (18:2) acids and most of lipidic classes. 相似文献

Previous work has shown that the undissociated form of 2,4-dinitrophenol (DNP) increases the permeability of barley (Hordeum vulgare var. trebi) roots to ions. The present studies were undertaken to determine whether the effects of undissociated DNP were directly on membrane lipids. Relative amounts of the principal fatty acids from the lipids of barley root membranes were assayed as a function of DNP concentration, pH, and time of treatment under conditions similar to the previous studies of DNP effects on permeability. Undissociated DNP increases the proportions of palmitic and oleic acids and decreases linoleic and linolenic acids with no changes in the amounts of total fatty acids. The effects are immediate, as are the effects on permeability. Only the undissociated DNP is effective. Anionic DNP has no effect, although it is the major species taken up by the roots both at pH 5 and pH 7. DNP has no effect on respiration at either pH, indicating that undissociated DNP effects are on the membranes and not a general metabolic effect. The close parallelism between the effects of DNP on the composition of membrane lipids and on permeability suggests that the increase in permeability produced by undissociated DNP is due to a direct effect on the root membranes. 相似文献

Abstract— Concentrations in whole rat brains of lipids (total lipids, phospholipids, galactolipids, cholesterol, plasmalogens) and of proteolipid protein were not altered after feeding for 2 and 6 weeks of diets containing 5 per cent excess l -phenylalanine. After 2 weeks of diet with 7 per cent excess l -phenylalanine there was a slight reduction (5–10 per cent) in the concentrations in whole brain of cholesterol and galactolipids. No significant effects were noted in cerebral hemispheres after 3 weeks of diets with 7 per cent excess l -phenylalanine. In the 5 and 7 per cent supplemented groups of animals, the total amounts of the various lipids were initially reduced to levels which were within 10 per cent of those in diet-matched controls. The results for rats indicate that after 3 weeks of age only very moderate effects on accumulation of cerebral lipids can be produced by excess dietary l -phenylalanine fed at the most toxic levels of supplementation, while lower levels of dietary supplementation are without effect. The results suggest further that the more mature brain is resistant to alteration of deposition of myelin lipids by high levels of phenylalanine. 相似文献

Cold hardened and unhardened 8- or 16-month-old citrus plants were examined for differences in fatty acid (FA) content. Unhardened leaves from 8-month-old Valencia scion budded on sour orange rootstock had 29% less FAs than leaves from seedling sour orange. After cold hardening triacylglycerol (TAG) FAs increased 4-fold in Valencia on sour orange and 6-fold in sour orange seedling. The percentage of FAs associated with TAGs for unhardened-hardened 16-month-old Valencia on sour orange tissues were: upper leaves 7–20, lower leaves 6–17, bark 6–9, and roots 57–73%. Cold hardening increased the amount of TAG FAs of 16-month-old Valencia on sour orange in upper leaves by 226% and in lower leaves by 173%. Concentrations of linoleic acid increased by 479% in upper leaves and by 303% in lower leaves. Quantities of lionolenic acid in monogalactosyl diacylglycerols declined by 27% in upper leaves and by 20% in lower leaves. 相似文献

Biologic membranes are not simply inert physical barriers, but complex and dynamic environments that affect membrane protein structure and function. Residing within these environments, ion channels control the flux of ions across the membrane through conformational changes that allow transient ion flux through a central pore. These conformational changes may be modulated by changes in transmembrane electrochemical potential, the binding of small ligands or other proteins, or changes in the local lipid environment. Ion channels play fundamental roles in cellular function and, in higher eukaryotes, are the primary means of intercellular signaling, especially between excitable cells such as neurons. The focus of this review is to examine how the composition of the bilayer affects ion channel structure and function. This is an important consideration because the bilayer composition varies greatly in different cell types and in different organellar membranes. Even within a membrane, the lipid composition differs between the inner and outer leaflets, and the composition within a given leaflet is both heterogeneous and highly dynamic. Differential packing of lipids (and proteins) leads to the formation of microdomains, and lateral diffusion of these microdomains or "lipid rafts" serve as mobile platforms for the clustering and organization of bilayer constituents including ion channels. The structure and function of these channels are sensitive to specific chemical interactions with neighboring components of the membrane and also to the biophysical properties of their membrane microenvironment (e.g., fluidity, lateral pressure profile, and bilayer thickness). As specific examples, we have focused on the K+ ion channels and the ligand-gated nicotinicoid receptors, two classes of ion channels that have been well-characterized structurally and functionally. The responsiveness of these ion channels to changes in the lipid environment illustrate how ion channels, and more generally, any membrane protein, may be regulated via cellular control of membrane composition. 相似文献

Neonatal rats, thyroidectomized at one day of age, received either no replacement therapy or daily subcutaneous injections of 100 μg/kg body weight of thyroxine (T4) or equal molar amounts of triiodothyroacetic acid (T3AC) from day 6 to day 26. Myelin lipid was isolated from the rat cerebrums and the cholesterol, cerebroside and total phospholipid content were determined. Cerebral myelin lipid weights were decreased in the neonatally thyroidectomized and T4 treated rats. The myelin lipid weights of the T3AC treated rats were not different from those treated with T4. The lipid composition remained unchanged in the T3AC, T4, and thyroidectomized groups when compared with the normal rats. These results indicate that T3AC and T4 administration have similar effects on CNS myelination in brains of neonatally thyroidectomized rats. 相似文献

Unusual chemicals produced by the-'blue oyster' diatom, Haslea ostrearia, include the water-soluble blue pigment marennine and numerous polyunsaturated sesterterpene oils or haslenes. Aqueous extracts of the alga exhibit in vitro and in vivo activities against human lung cancer cells and anti-HIV effects. Here we report that three haslenes also demonstrate in vitro cytostatic action against a human lung cancer cell line. The most active haslene is the most unsaturated and unsaturation in the haslenes increases with increasing algal growth temperature. 相似文献

The lipid composition of rat spinal cord undergoing postmortem autolysis for 3 min and for 4 h at 38 degrees C was investigated as a model for lipid changes in total spinal cord ischemia. The only change in cords incubated for 3 min was an 11.7% decrease in cholesterol/g fresh weight. The cords incubated for 4 h showed a similar 11.6% decrease in cholesterol/g fresh weight as well as a 5.6% increase in water content and a 22% decrease in phosphatidyl serine. Changes of marginal statistical significance included a 15% increase in lipid phosphorus/g dry wt. and a 15% decrease in G4 (GM1)1 in the 4 h incubated cords. Therefore, autolytic processes are of little consequence in total spinal cord ischemia and attention should now be focused on exogenous pathogenetic factors to explain such ischemic changes in spinal cord.
