Summary Three polyclonal antisera raised in rabbits against the mammalian molecular form of gonadotropin-releasing hormone (GnRH) were tested in enzyme-linked immunosorbent assays for crossreactivity with naturally occurring GnRHs and with GnRH analogues. Antisera were then tested immunocytochemically in order (i) to identify amino acids essential for the binding of each antiserum, and (ii) to evaluate the specificity of the immunocytochemical reaction in brain sections from various species of cyclostomes, amphibians, reptiles, and birds. Antiserum GnRH 80/1, recognizing mainly a discontinuous determinant including the NH2- and COOH-termini, crossreacts with GnRHs the molecular bending of which enables the spatial approach of both terminal amino acid residues. Antiserum GnRH 80/2, by requiring the COOH-terminus for binding and not tolerating substitutions by aromatic amino acids in the middle region of the molecule, recognizes chicken I GnRH, however, not the salmon form. The use of this antiserum is appropriate in species synthesizing the mammalian and/or the chicken I form of GnRH. GnRH antiserum 81/1 is specific mostly for mammalian GnRH. The results obtained by ELISAs are confirmed by immunocytochemical studies. A comparison between the results obtained in ELISA and in immunocytochemistry involving mammalian-, chicken I-, chicken II-, salmon-, and lamprey-directed GnRH antisera resulted in the following conclusions: (1) An antiserum recognizing the discontinuous antigen determinant including both NH2- and COOH-termini may be reactive in most vertebrate brain sections thus being appropriate for phylogenetically directed immunocytochemical studies. (2) Moreover, this discontinuous determinant seems to be immunocytochemically reactive in all parts of the neurons in the GnRH system, whereas, in some species, determinants located in the middle region of the molecule(s) tend to become reactive only during the axonal transport. (3) A crossreaction between tissue-bound antigen and antibodies recognizing the above cited discontinuous determinant indicates an appropriate bending of the molecule even in case of severe molecular differences, e.g., in lamprey form of GnRH. (4) It follows that in phylogenetic studies, an immunologically well characterized antiserum can be substituted for a species-directed antiserum.  相似文献

Spiders are common generalist predators, and understanding their potential in biological control is important for the development of integrated pest management programs. In this study, predation by three groups of spiders on the mirid bug Stenotus rubrovittatus (Hemiptera: Miridae) in rice paddies was investigated using DNA-based gut-content analysis. A laboratory feeding study revealed that the detection half-lives of bug DNA in the spider gut at 25 °C was 3.4 days for Lycosidae and 1.5 days for Tetragnathidae. Individual spider predation on the mirid bug was investigated by detecting DNA of prey in field-collected spiders. In total, 1199 spiders were assayed from three spider groups: Pirata subpiraticus (Lycosidae), Tetragnatha spp. (Tetra-gnathidae), and Pachygnatha clercki (Tetra-gnathidae), which each differ in their preferred microhabitat as well as their predatory habits. Detection rates of prey DNA in spiders increased significantly with the density of prey across all spider groups. P. subpiraticus and Tetragnatha spp. predation showed a better fit to a saturated response curve to increasing prey density, while P. clercki showed a simple linear relationship with prey density. Densities of alternative prey species did not affect the detection rates of mirids. These results suggest that predation on pests by generalist predators in an agroecosystem is affected not only by prey abundance but also by predator preference for specific prey. Predator preference is therefore an important factor to consider when estimating the role of natural enemies as biological control agents.  相似文献

The genus Ganoderma has a worldwide distribution causing root and stem rot of many plantation crops. A limiting factor in controlling the BSR disease is the lack of reliable diagnostic method(s) for early diagnosis. In this study, we developed polyclonal antiserum for Ganoderma mycelial and extracellular protein, and evaluated its efficacy with different plant samples collected from artificially inoculated coconut seedlings and Ganoderma infected field palms. We also tested the cross-reactivity with the soil-borne and saprophytic fungus collected from different parts of coconut palm. The antisera developed against the crude mycelial protein (CMP) and extracellular protein (ECP) showed a 1:1000 titre value for the detection of Ganoderma. The CMP antisera developed showed more cross-reaction when compared to ECP antisera of Ganoderma. In the DIBA test, at a 1:10 dilution of antigen, 1:1000 dilution of CMP and ECP antisera, 1:5000 dilution of secondary antibody gave clear distinctions in colour development between healthy and diseased samples. In the DIBA test, ECP antisera detected positive control (ECP of Ganoderma MTP and CRS-1 isolate), artificially inoculated roots, infected field roots, infected basal trunk and additionally lesions gave positive reactions which were not found in the CMP antisera tested. Therefore, both ELISA and DIBA tests may be useful for screening a large number of samples and help in the detection of infection at the earliest stage of disease development and this will certainly help to adopt suitable management strategies against Ganoderma disease in palm crops in advance.  相似文献

  • 1 Landscape management for enhanced natural pest control requires knowledge of the ecological function of the habitats present in the landscape mosaic. However, little is known about which habitat types in agricultural landscapes function as reproduction habitats for arthropod pests and predators during different times of the year.
  • 2 We studied the arthropod assemblage on six crops and on the seven most abundant native plant species in two landscapes over 1 year in Australia. Densities of immature and adult stages of pests and their predators were assessed using beat sheet sampling.
  • 3 The native plants supported a significantly different arthropod assemblage than crops. Native plants had higher predator densities than crops over the course of the year, whereas crops supported higher pest densities than the native plants in two out of four seasonal sampling periods. Crops had higher densities of immature stages of pests than native plants in three of four seasonal sampling periods, implying that crops are more strongly associated with pest reproduction than native plants. Densities of immature predators, excluding spiders, were not different between native plants and crops. Spiders were, however, generally abundant and densities were higher on native plants than on crops but, because some species disperse when immature, there is less certainty in identifying their reproduction habitat.
  • 4 Because the predator to pest ratio on native plant species showed little variation, and spatial variation in arthropod assemblages was limited, the predator support function of native vegetation may be a general phenomenon. Incentives that maintain and restore native remnant vegetation can increase the predator to pest ratio at the landscape scale, which could enhance pest suppression in crops.

Antisera were produced to clover phyllody- and Spiroplasma citri-associated antigens partially purified from infected Vinca rosea plants. Separate antisera were made to ‘membrane fraction’ (MF) preparations comprising the resuspended pellet obtained by high speed centrifugation, and to ‘soluble fraction’ (SA) preparations, comprising the supernatant from high speed centrifugation concentrated by freeze-drying. All antisera showed considerable activity against normal plant antigens but after cross-absorption with extracts of healthy plants the MF antisera were used in F(ab')2based ELISA tests to detect S. citri- or clover phyllody-associated antigens in infected plants. The ‘clover phyllody’ antiserum also reacted specifically with extracts of clover plants with phyllody, and with naturally-infected strawberry plants showing symptoms of green petal disease. Both the ‘clover phyllody’ and S. citri antisera were specific for their respective homologous antigens. No cross-reactions were observed in heterologous tests or between either antiserum and extracts of V. rosea infected with various MLOs obtained from different host plants.  相似文献

  1. The possible impact of arthropod predation on inter-population variation in adult density of a thistle-feeding lady beetle, Henosepilachna niponica (Lewis ) was evaluated by means of predator exclusion experiments conducted in the field.
  2. The population density of newly-emerged adults at one habitat in the upstream area (site F) was significantly lower than at another in the downstream area (site A) although the egg density was nearly identical in the two habitats.
  3. In the habitat with lower adult density, egg mortality was higher due to higher levels of predation. A predator exclusion experiment demonstrated that arthropod predation was the main factor causing high mortality during the immature stages, and physical factors such as heavy rains were unlikely to influence larval survivals.
  4. Earwigs, ground beetles, predaceous stink bugs, and spiders were identified as the main predators in the study area. Of these, an earwig, Anechura harmandi (Burr ) was more predominant than other predators and was significantly more abundant in the habitat with low adult densities.

The goal of this project was the development of a serological test for the detection of Sclerotinia sclerotiorum on oilseed rape petals. Since the fungus exists in two forms on petals, as ascospores and mycelium, the responses of anti-mycelium and anti-ascospore antisera against these two kinds of antigens were compared. Two anti-mycelium sera, Smy and Smy', were produced against mycelial soluble extracts at different concentrations (0.3 mg and 0.1 mg of protein ml-1). Smy gave the greatest response level with eight S. sclerotiorum mycelial extracts tested. It had a very superior level of recognition to that of anti-ascospore serum, Ssp, when mycelium was tested as antigen. In contrast, Ssp and Smy were equally reactive when exposed to ascospores but the sensitivity of the assay was low. For each antiserum, the solution from which ascospores had been removed reacted similarly to the original suspension containing ascospores. A collection of fungi likely to be found on oilseed rape petals was examined. Cross-reactions were produced with both antisera, especially with Botrytis cinerea for which greater cross-reactivities were produced with Smy. The cross-absorption of antiserum Smy with a mycelial extract of B. cinerea considerably reduced this cross-reaction. The choice of antiserum for the development of a reliable detection system is discussed.  相似文献

Abstract.
  • 1 The interactions between the univoltine mirid bug Cupsodes infuscatus and its food plant, the geophyte Asphodelus ramosus, were studied in the Negev desert for a 5 year period. The bug feeds mainly on Asphodelus inflorescence meristems, flowers and fruits, and in some years may destroy more than 95% of the plant population expected fruit production.
  • 2 Asphodelus expected fruit production fluctuated widely during the study period, but was not related to precipitation. Cupsodes density was related to the plant expected fruit production, but with a 1 year time lag. In years of high inflorescence production, a high per-capita reproduction of the bug resulted in a dense bug population in the following year. This dense population then decimated the plant fruit production, became food limited and had a low per-capita reproduction.
  • 3 This kind of time lag is expected to be common among desert insect herbivores that specialize in using ephemeral resources. The rare years of high plant production are in general preceded and followed by years of low plant production. Hence, in years which contribute most to plant reserves (seed, underground storage organs), insect herbivores are relatively rare as a result of food limitation in preceding low production years. But the insect populations which build up during years of high plant production decimate their food resources and become food limited in subsequent years with low plant production.
  • 4 Thus, herbivorous insects seem to have a limited ability to affect plant population dynamics in desert ecosystems. In contrast, the potential appears to be much greater for herbivorous insects to be regulated by their food plants.

Among the various fungal diseases affecting plantation crops viz., coconut, aracanut, oil palm, etc. in India, basal stem rot (BSR) caused by species of Ganoderma is the most destructive. A limiting factor in controlling the BSR disease is the lack of reliable diagnostic method(s) for early diagnosis. In this study we generated two different types of antiserum for diagnosis of Ganoderma using the purified monospecific protein (62 kDa) (MS) and crude sporophore extract (SE). We also tested the cross-reactivity with the soil-borne and saprophytic fungus collected from different parts of coconut palm. The antiserum developed against the MS and SE showed 1:700 and 1:3000 titre values for the detection of Ganoderma. The MS antisera developed showed very low or almost no cross-reaction when compared to SE antisera of Ganoderma. In the DIBA test, at a 1:10 dilution of antigen, 1:1000 dilution of CMP and ECP antisera, 1:5000 dilution of secondary antibody gave clear distinctions in colour development between healthy and diseased samples. In DIBA test, both types of antisera were used separately for pathogenicity tests. MS antisera showed a positive reaction for purified protein, artificially infected roots and infected field palm. A mild reaction was observed against infected field trunk but a negative reaction was observed for lesions and leaf samples. In the case of SE antisera, a negative reaction was observed for all leaf samples, healthy roots and healthy trunk samples but positive reactions were observed for positive control, artificially inoculated roots, infected field roots, infected trunk and lesions samples. Therefore, both ELISA and DIBA tests may be useful in the detection of infection at the earliest stage of disease development and this will certainly help in the development of management strategies against Ganoderma disease in palm crops in advance.  相似文献

  1. Crop production sequences influence arthropod populations in temporally unstable row crop systems. Winter wheat (Triticum aestivum L.) represents one of the earliest abundant crops in south-eastern United States. This study aims to understand primary source habitats driving brown stink bug, Euschistus servus (Say), and tarnished plant bug, Lygus lineolaris (Palisot de Beauvois), population abundance in wheat.
  2. To better understand these relationships, adult and nymphal densities were in wheat fields weekly from flowering through harvest in 2019 and 2020. Geospatial data were used to measure landscape composition surrounding sampled fields. We investigated the influence of landscape predictors on E. servus and L. lineolaris abundance using generalized linear mixed modelling.
  3. Field size, proportion of agriculture, proportion of wheat area, and proportion of soybean Glycine max L.) area from the previous year in the surrounding landscape were associated with E. servus abundance in wheat. Similarly, L. lineolaris abundance was associated with proportion of wheat area and soybean area from the previous year.
  4. These results reveal the influence of soybean area planted the previous year on insect pest densities the following spring in wheat. Further, results suggest agricultural landscapes dominated by wheat are associated with decreased pest abundance across the sampled region.

An enzyme-linked immunosorbent assay (ELISA) was developed to detect remains of Pieris rapae L. (Lepidoptera: Pieridae) immature stages in the guts of field collected arthropod predators. The assay can be used to help ascertain the relative importance of arthropod predator species in suppressing P. rapae in cabbage, Brassica oleracea var. capitata L. The ELISA is sensitive to all immature stages of P. rapae, although first and fifth instars can be detected more readily than eggs or pupae and third instars showed intermediate detectability. Assays on whole body homogenates of predators readily detected predation on P. rapae first instars by all seven of the predator species tested, although response generally declined with increasing predator size. Together the results show that the P. rapae ELISA possesses a sufficiently high level of sensitivity and specificity to be a useful tool in helping to elucidate the roles of arthropod predator species in reducing populations of P. rapae in cabbage.  相似文献

The winter cherry bug, Acanthocoris sordidusThunberg , lives in aggregation especially in their early larval instars. Using the 1st-instar larvae of this species, the author tried to clarify both the processes and the mechanisms of the breakup and later re-formation of colony in relation to the defence against their enemies. The results obtained were summarized as follows.
  1. In the field population, there is a high possibility of dispersal of the 1st-instar larvae from a colony possibly through the disturbance by some predators but they can re-form a colony with each other or join, with colonies of different instar larvae.
  2. The individuals in a colony immediately disperse through the attack of predatory coccinellid beetle, Harmonica axyridis but tend to re-form a colony in a short time.
  3. The breakup of colony is caused by the secretion from the attacked individual.
  4. The formation of colony is attributed to the habit closely related with the senses of smell and/or contact.
From these results, it was concluded that the dispersal of 1st-instar larvae from a colony, followed by the re-formation of a colony, is an an adaptive behaviour to escape from the attack by their predators.  相似文献

Polyclonal antibodies were raised in rabbits against MLO associated with faba bean (Vicia. faba L.) phyllody which exists in the Sudan. Two indirect ELISA methods were able to detect the MLO antigens. In the former, the whole antigen was directly coated onto plates, while in the second, only the F(ab')2, fragments of the IgG were used to coat the ELISA plates. Higher detectable efficiency was obtained when the F(ab')2 method was used. Moreover the obtainable antiserum was found to exhibit a high degree of specificity through which the MLO associated with faba bean phyllody in the Sudan, are serologically differentiated from other isolates of MLO existing in the Sudan as well as European MLO isolates maintained at Versailles, and Spiroplasma citri, causal agent of Citrus Stubborn Disease. The positive reactions obtained with this antiserum against MLO phyllody naturally existing in the Sudan on Crotalaria saltiana and some Catharanthus roseus demonstrate that these plants are potential reservoirs of the disease in the Sudan. The same antiserum was used in order to distinguish healthy and diseased plant preparations using the membrane ELISA method (dot-blot).  相似文献
