The inhibition of photosynthetic activity by bisulphite was studied in intact leaves of abscisic acid (ABA)-treated and non-treated (control) barley plants. ABA inhibited the photosynthetic process as evidenced by lower values of chlorophyll fluorescence kinetic parameters Fv/Fm (photosystem 2 activity) and Rfd (vitality index, related to the whole photosynthetic activity) compared with ABA-non-treated plants. After bisulphite treatment, the extent of inhibition was smaller in ABA-treated plants than in the control ones indicating a protective effect of ABA. The protective action sites of ABA were the QA reduction and the Calvin cycle. This revised version was published online in September 2006 with corrections to the Cover Date.  相似文献

Natural senescence of Cucurbita pepo (zucchini) cotyledons was accompanied by a gradual degradation of reserve proteins (globulins) and an intensive decrease in the content of both large subunit (LSU) and small subunit (SSU) of ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco). The net photosynthetic rate, the primary photochemical activity of PSII, estimated by the variable fluorescence (Fv)/maximal fluorescence (Fm) ratio (Fv/Fm) and the actual quantum yield of PSII electron transport in the light-adapted state (ΦPSII) also progressively decreased during natural senescence. In contrast, the fraction of the absorbed light energy, which is not used for photochemistry (LNU) increased with progression of senescence. The decline in the photosynthetic rate started earlier in ontogenesis compared with the down-regulation of the functional activity of PSII, thus suggesting the existence of protective mechanisms which maintain higher efficiency of the photochemical electron transport reactions of photosynthesis compared with the dark reactions of the Calvin cycle during earlier stages of natural senescence. Decapitation of the epicotyl above the senescing cotyledons resulted in full recovery of the polypeptide profile in the rejuvenated cotyledons. In addition, the photosynthetic rate increased reaching values that exceeded those measured in juvenile cotyledons. The photochemical efficiency of PSII also gradually recovered, although it did not reach the maximum values measured in the presenescent cotyledons.  相似文献

Sulfite treatment of pea leaf disks in light caused a significant decrease in the relative quantum yield of photosynthetic oxygen evolution and energy storage (ES) as measured by photoacoustic (PA) spectroscopy. The inhibition was concentration dependent and was less in darkness than in light, indicating light-dependent inhibitory site(s) on the photosynthetic electron transport chain. Further, in darksulfite-treated leaves, the energy storage was more affected than the relative quantum yield of oxygen evolution, suggesting that photophosphorylation and/or cyclic electron transport around PS I are sites of sulfite action in darkness. The Rfd values, the ratio of fluorescence decrease (fd) to the steady-state fluorescence (fs), decreased significantly in leaves treated with sulfite in light but were not affected in dark-treated ones, confirming the photoacoustic observations. Similarly, the ratio of variable fluorescence (Fv) to maximum fluorescence (Fm), a measure of PS II photochemical efficiency, was affected by sulfite treatment in light and not changed by treatment in darkness. In this study, effects of yellow (Y), purple (P), red (R), blue (B), green (G), and white (W) light on growth and development of tobacco plants were evaluated. We showed that monochromatic light reduced the growth, net photosynthetic rate (P N), stomatal conductance, intercellular CO2, and transpiration rate of tobacco. Such a reduction in P N occurred probably due to the stomatal limitation contrary to plants grown under W. Photochemical quenching coefficient (qP), maximal fluorescence of dark-adapted state, effective quantum yield of PSII photochemistry (ΦPSII), and maximal quantum yield of PSII photochemistry (Fv/Fm) of plants decreased under all monochromatic illuminations. The decline in ΦPSII occurred mostly due to the reduction in qP. The increase in minimal fluorescence of dark-adapted state and the decrease in Fv/Fm indicated the damage or inactivation of the reaction center of PSII under monochromatic light. Plants under Y and G showed the maximal nonphotochemical quenching with minimum P N compared with the W plants. Morphogenesis of plants was also affected by light quality. Under B light, plants exhibited smaller angles between stem and petiole, and the whole plants showed a compact type, while the angles increased under Y, P, R, and G and the plants were of an unconsolidated style. The total soluble sugar content increased significantly under B. The reducing sugar content increased under B but decreased significantly under R and G compared with W. In conclusion, different monochromatic light quality inhibited plants growth by reducing the activity of photosynthetic apparatus in plants. R and B light were more effective to drive photosynthesis and promote the plant growth, while Y and G light showed an suppression effect on plants growth. LEDs could be used as optimal light resources for plant cultivation in a greenhouse.  相似文献

Dissipation of light energy absorbed by photosystem II (PSII) in assimilating shoots of an evergreen shrub Ephedra monosperma was investigated during its transition from the vegetative to frost-tolerant state under natural conditions of Central Yakutia. The dynamics of modulated chlorophyll fluorescence and carotenoid content was analyzed during seasonal decrease in ambient temperature. The seasonal cooling was accompanied by a stepwise decrease in photochemical activity of PSII (F v/F m = (F m ? F 0)/F m). The decrease in F v/F m occurred from the beginning of September to the end of October, when the temperature was lowered from 10 to ?8°C. During winter period the residual activity of PSII was retained at about 30% of the summer values. The seasonal decrease in temperature was accompanied by a significant stimulation of pH-independent dissipative processes in reaction centers and antenna of PSII. The increase in energy losses was paralleled by a proportional increase in zeaxanthin content on the background of decreasing content of violaxanthin and β-carotene as possible zeaxanthin precursors. At the same time, inhibition of light-induced non-photochemical quenching in the PSII antenna was observed. The results suggest that principal photoprotective mechanisms during seasonal lowering of temperature are: (1) inactivation of PSII and dissipation of excitation energy in PSII reaction centers and (2) zeaxanthin-mediated energy dissipation in the antenna complexes. The first mechanism seems to prevail at early stages of seasonal cooling, whereas both mechanisms are recruited from the onset of sustained freezing temperatures.  相似文献

To assess the role of redox state of photosystem II (PSII) acceptor side electron carriers in PSII photochemical activity, we studied sub-millisecond fluorescence kinetics of the wild type Synechocystis PCC 6803 and its mutants with natural variability in the redox state of the plastoquinone (PQ) pool. In cyanobacteria, dark adaptation tends to reduce PQ pool and induce a shift of the cyanobacterial photosynthetic apparatus to State 2, whereas illumination oxidizes PQ pool, leading to State 1 (Mullineaux, C. W., and Holzwarth, A. R. (1990) FEBS Lett., 260, 245-248). We show here that dark-adapted Ox mutant with naturally reduced PQ is characterized by slower QA reoxidation and O2 evolution rates, as well as lower quantum yield of PSII primary photochemical reactions (Fv/Fm) as compared to the wild type and SDH–mutant, in which the PQ pool remains oxidized in the dark. These results indicate a large portion of photochemically inactive PSII reaction centers in the Ox mutant after dark adaptation. While light adaptation increases Fv/Fm in all tested strains, indicating PSII activation, by far the greatest increase in Fv/Fm and O2 evolution rates is observed in the Ox mutant. Continuous illumination of Ox mutant cells with low-intensity blue light, that accelerates QA reoxidation, also increases Fv/Fm and PSII functional absorption cross-section (590 nm); this effect is almost absent in the wild type and SDH–mutant. We believe that these changes are caused by the reorganization of the photosynthetic apparatus during transition from State 2 to State 1. We propose that two processes affect the PSII activity during changes of light conditions: 1) reversible inactivation of PSII, which is associated with the reduction of electron carriers on the PSII acceptor side in the dark, and 2) PSII activation under low light related to the increase in functional absorption cross-section at 590 nm.  相似文献

The PSII photochemical activity in a terrestrial cyanobacterium Nostoc commune Vaucher ex Bornet et Flahault during rewetting was undetectable in the dark but was immediately recognized in the light. The maximum quantum yield of PSII (Fv/Fm) during rewetting in the light rose to 85% of the maximum within ~30 min and slowly reached the maximum within 6 h, while with rewetting in the darkness for 6 h and then exposure to light the recovery of Fv/Fm required only ~3 min. These results suggested that recovery of photochemical activity might depend on two processes, light dependence and light independence, and the activation of photosynthetic recovery in the initial phase was severely light dependent. The inhibitor experiments showed that the recovery of Fv/Fm was not affected by chloramphenicol (CMP), but severely inhibited by 3‐(3,4‐dichlorophenyl)‐1,1‐dimethylurea (DCMU) in the light, suggesting that the light‐dependent recovery of photochemical activity did not require de novo protein synthesis but required activation of PSII associated with electron flow to plastoquinone. Furthermore, the test indicated that the lower light intensity and the red light were of benefit to its activation of photochemical activity. In an outdoor experiment of diurnal changes of photochemical activity, our results showed that PSII photochemical activity was sensitive to light fluctuation, and the nonphotochemical quenching (NPQ) was rapidly enhanced at noon. Furthermore, the test suggested that the repair of PSII by de novo protein synthesis played an important role in the acclimation of photosynthetic apparatus to high light, and the heavily cloudy day was more beneficial for maintaining high photochemical activity.  相似文献

Development of chlorosis and loss of PSII were compared in young spinach plants suffering under a combined magnesium and sulphur deficiency. Loss of chlorophyll could be detected already after the first week of deficiency and preceded any permanent functional inhibition of PSII as detected by changes in the chlorophyll fluorescence parameter Fv/Fm. A substantial decrease in Fv/Fm was observed only after the second week of deficiency. After 4 weeks, the plants had lost about 70% of their original chlorophyll content, but fluorescence data indicated that 80% of the existing PSII centers were still capable of initiating photosynthetic electron transport. The degradation of the photosynthetic apparatus without loss of PSII activity was due to changes in protein turnover, especially of the PSII D1 reaction center protein. Already by day 7 of deficiency, a 1.4-fold increase in D1 protein synthesis was observed measured as incorporation of 14C-leucine. Immunological determination by western-blotting did not reveal a change in D1 protein content. Thus, D1 protein was also degraded more rapidly. The increased turnover was high enough to prevent any loss or inhibition of PSII. After 3 weeks, D1 protein synthesis on a chlorophyll basis was further increased by a factor of 2. However, this was not enough to prevent a net loss of D1 protein of about 70%. Immunological determination revealed that together with the D1 protein also other polypeptides of PSII became degraded. This process prevented a large accumulation of photo-inactivated PSII centers. However, it initiated the breakdown of the other thylakoid proteins, especially of LHCII, resulting in the observed chlorosis. Together with the change in protein turnover and stability, a characteristic change in thylakoid protein phosphorylation was observed. In the deficient plants steady state phosphorylation of both LHCII and PSII proteins was increased in the dark. In the light phosphorylation of PSII proteins was stimulated and after 3 weeks of deficiency was even higher in the deficient leaves than in the control plants. In contrast, the phosphorylation level of LHCII decreased in the light and could hardly be detected after 3 weeks of deficiency. Phosphorylation of the reaction center polypeptides presumably increased their stability against proteolytic attack, whereas phosphorylated LHCII seems to be the substrate for proteolysis.  相似文献

利用不同剂量的137Cs-γ射线对毛竹(Phyllostachys heterocycla ‘Pubescens’)种子进行辐射,测定实生苗叶片中的光合色素含量和叶绿素荧光参数等指标,探讨辐射对毛竹幼苗生长的影响,为筛选有利的突变单株奠定良好基础。结果表明:30或60Gy的137Cs-γ射线辐射后,毛竹幼苗的光合色素含量以及最大荧光强度(Fm)、可变荧光强度(Fv)、PSII最大光化学效率(Fv/Fm)、PSII的潜在活性(Fv/Fo)、PSII实际光化学效率(Yield)和表观光合电子传递速率(ETR)等荧光参数值均高于90Gy辐射处理,说明较低剂量辐射后PSII反应中心的能量捕获效率高,且具有较强的光合能力;而90Gy的137Cs-γ射线辐射对毛竹的影响则与之相反。不同处理剂量之间叶片光能耗散程度以及表观光合电子传递速率-光合有效辐射(ETR-PAR)响应曲线的分析结果也进一步证实了以上结论。  相似文献

Shoots of two species of moss, Plagiomnium undulatum (Hedw.) Kop. and Plagiomnium affine (Funck) Kop., were subjected to freezing at various temperatures. After thawing, the activities of different photosynthetic reactions were determined in relation to the ages of the leaves. Analysis of the fast kinetics of chlorophyll-a fluorescence of individual leaves showed that young and old tissues were considerably less frost tolerant than mature ones. In principle, the pattern of freeze inactivation of photosynthetic reactions resembles that observed in higher plants. The decreases in the amplitude of Fv (variable fluorescence) and the ratio of Fv to Fm (maximum fluorescence) with increasing freezing stress reflect a progressive inactivation of photosystem II (PSII)-mediated electron transport, i.e. inhibition of photoreaction to photochemistry and-or electron donation to the photochemical reaction, and thus a decline in the potential photochemical efficiency of PSII. The insignificant change in the F0 (constant fluorescence) level during progressive decline of Fv indicates that the excitation-energy transfer between antenna pigments and from those to reaction centres of PSII was little impaired by lethal freezing stress. The photosynthetic performances of regenerated protoplasts of Bryopsis hypnoides, which were incubated in seawater for 1, 6, 12, and 24 h, were studied using chlorophyll (Chl) fluorescence and oxygen measurements. Results showed that for the regenerated protoplasts, the pigment content, the ratios of photosynthetic rate to respiration rate, the maximal photosystem II (PSII) quantum yield (Fv/Fm), and the effective PSII quantum yield (ΦPSII) decreased gradually along with the regeneration progress, indicated that during 24 h of regeneration there was a remarkable reduction in PSII activity of those newly formed protoplasts. We assumed that during the cultivation progress the regenerated protoplasts had different photosynthetic vigor, with only some of them able to germinate and develop into mature thalli. The above results only reflected the photosynthetic features of the regenerated protoplasts at each time point as a whole, rather than the actual photosynthetic activity of individual aggregations. Further investigation suggested a relationship between the size of regenerated protoplasts and their viability. The results showed that the middle-sized group (diameter 20–60 μm) retained the largest number of protoplasts for 24 h of growth. The changes in Fv/Fm and ΦPSII of the four groups of differently sized protoplasts (i.e. < 20, 20–60, 60–100, and > 100 μm) revealed that the protoplasts 20–60 μm in diameter had the highest potential activity of the photosynthetic light energy absorption and conversion for several hours.  相似文献

In a previous study, we characterized a high chlorophyll fluorescence Ipal mutant of Arabidopsis thallana, in which approximately 20% photosystem (PS) Ⅱ protein is accumulated. In the present study, analysis of fluorescence decay kinetics and thermoluminescence profiles demonstrated that the electron transfer reaction on either the donor or acceptor side of PSII remained largely unaffected in the Ipa1 mutant. In the mutant, maximal photochemical efficiency (Fv/Fm, where Fm is the maximum fluorescence yield and Fv is variable fluorescence) decreased with increasing light intensity and remained almost unchanged in wildtype plants under different light conditions. The Fv/Fm values also increased when mutant plants were transferred from standard growth light to low light conditions. Analysis of PSll protein accumulation further confirmed that the amount of PSll reaction center protein is correlated with changes in Fv/Fm in Ipal plants. Thus, the assembled PSll in the mutant was functional and also showed increased photosensitivity compared with wild-type plants.  相似文献

Kalanchoë daigremontiana, a CAM plant grown in a greenhouse, was subjected to severe water stress. The changes in photosystem II (PSII) photochemistry were investigated in water‐stressed leaves. To separate water stress effects from photoinhibition, water stress was imposed at low irradiance (daily peak PFD 150 μmol m?2 s?1). There were no significant changes in the maximal efficiency of PSII photochemistry (Fv/Fm), the traditional fluorescence induction kinetics (OIP) and the polyphasic fluorescence induction kinetics (OJIP), suggesting that water stress had no direct effects on the primary PSII photochemistry in dark‐adapted leaves. However, PSII photochemistry in light‐adapted leaves was modified in water‐stressed plants. This was shown by the decrease in the actual PSII efficiency (ΦPSII), the efficiency of excitation energy capture by open PSII centres (Fv′/Fm′), and photochemical quenching (qP), as well as a significant increase in non‐photochemical quenching (NPQ) in particular at high PFDs. In addition, photoinhibition and the xanthophyll cycle were investigated in water‐stressed leaves when exposed to 50% full sunlight and full sunlight. At midday, water stress induced a substantial decrease in Fv/Fm which was reversible. Such a decrease was greater at higher irradiance. Similar results were observed in ΦPSII, qP, and Fv′/Fm′. On the other hand, water stress induced a significant increase in NPQ and the level of zeaxanthin via the de‐epoxidation of violaxanthin and their increases were greater at higher irradiance. The results suggest that water stress led to increased susceptibility to photoinhibition which was attributed to a photoprotective process but not to a photodamage process. Such a photoprotection was associated with the enhanced formation of zeaxanthin via de‐epoxidation of violaxanthin. The results also suggest that thermal dissipation of excess energy associated with the xanthophyll cycle may be an important adaptive mechanism to help protect the photosynthetic apparatus from photoinhibitory damage for CAM plants normally growing in arid and semi‐arid areas where they are subjected to a combination of water stress and high light.  相似文献

The photosynthetic activity of two Syrian barley landraces, Arabi (A.) Aswad and A. Abiad, grown under 120 mM NaCl, was studied, using gas exchange and chlorophyll (Chl) a fluorescence transient (OJIP) measurements. Salt treatment of barley seedlings decreased both the rates of photosynthesis and photosystem II (PSII) activity, as evaluated from chlorophyll fluorescence data. However, the noted decrease was dependent on the duration of the salt treatment and the barley cultivar. Several parameters (e.g., light absorption flux per cross section of leaf; time to reach maximum chlorophyll a fluorescence intensity; plastoquinone pool size; yield of heat loss; rate of reaction center closure; and the so-called Performance Index), calculated and inferred from Chl fluorescence measurements, and related to PSII activity, were affected after 24 h of salt application, but these changes were much more pronounced after 7 days of salt treatment. Similar changes were found for measured gas exchange parameters: CO2 uptake (photosynthetic) rate and stomatal conductance. The photosynthetic apparatus of the cultivar variety (c.v.) Arabi Aswad was found to be much more tolerant to salt treatment, compared with c.v. Arabi Abiad. After 7 days of salt treatment, the latter showed a very high value of the initial (minimal) fluorescence (Fo) and then essentially almost flat fluorescence transient curve; this result may be due to several causes that include structural changes as well as changes in the rate constants of different dissipative processes. The parameters that were most affected, by salt treatment, were: the time needed to reach the maximal chlorophyll fluorescence (Fm), and the inferred oxygen evolving complex activity (Fv/Fo, where Fv, is Fm  Fo), and the calculated Performance Index (PIABS) that depends on the efficiency and the yield of energy transfer and primary photochemistry. We suggest that the early reactions of the photosynthetic apparatus of barley plants could play a key role in their tolerance to salt stress. Further, we found that the first stage of salinity effect on photosynthesis of barley plants is related to stomatal conductance limitation rather than to PSII activity reduction. Thus, on the basis of our results on the two barley landraces, we recommend the use of a combination of gas exchange measurements along with the analysis of the OJIP fluorescence transient for the detection of salt stress-induced changes in plants.  相似文献

To test wether chlorosis is induced by photoinhibitory damage to photosystem II (PSII), onset of chlorosis and loss of PSII function were compared in young spinach (Spinaciae oleracea L.) plants suffering under a combined magnesium and sulphur deficiency. Loss of chlorophyll already occurred after the first week of deficiency and preceded any permanent functional inhibition of the photosynthetic apparatus. Permanent disturbancies of photosynthetic electron transport measured in isolated thylakoids and of PSII function, determined via the ratio of variable fluorescence to maximal fluorescence, Fv/Fm, could be detected only after the second week of deficiency. After the third week, the plants had lost about 60% of their chlorophyll; even so, fluorescence data indicated that 85% of the existing PSII was still capable of initiating photosynthetic electron transport. However, quenching analysis of steady-state fluorescence showed an early increase in non-photochemical quenching and in down-regulated PSII centres with low steady-state quantum efficiency. Together with the down-regulation of PSII centres, a 1.4-fold increase in D1-protein synthesis, measured as incorporation of [14C]leucine, could be observed at the end of the first week before any loss of D1 protein, chlorophyll or photosynthetic activity could be detected. Immunological determiation by Western-blotting did not show a change in D1-protein content; thus, at this time, D1 protein was not only faster synthesised but was also faster degraded than before the imposition of mineral deficiency. The increased turnover was high enough to prevent any loss or functional inhibition of PSII. After 3 weeks, D1-protein synthesis on a chlorophyll basis was further stimulated by a factor of 2. However, this was not enough to prevent a net loss of D1 protein of about 70%, showing that the D1-protein was now degraded faster than it was synthesised. Immunological determination and electron-transport measurements showed that together with the loss of D1 protein the other polypetides of PSII were also degraded, resulting in a specific loss of PSII centres. The degradation of PSII centres prevented a large accumulation of damaged PSII centres. Effects of root treatment with 5-aminolevulinic acid (ALA) on leaf photosynthesis in strawberry (Fragaria ananassa Duch.) plants were investigated by rapid chlorophyll fluorescence and modulated 820 nm reflection using 3-(3,4-dichlorophenyl)-1,1-dimethyl urea (DCMU) and methyl viologen (MV). Our results showed that ALA treatments increased the net photosynthetic rate and decreased the intercelluar CO2 concentration in strawberry leaves. Under DCMU treatment, trapping energy for QA reduction per PSII reaction center increased greatly, indicating DCMU inhibited electron transfer from QA ?. The maximum photochemical efficiency of PSII (Fv/Fm) decreased under the DCMU treatment, while a higher Fv/Fm remained in the ALA-pretreated plants. Not only the parameters related to a photochemical phase, but also that one related to a heat phase remained lower after the ALA pretreatment, compared to the sole DCMU treatment. The MV treatment decreased PSI photochemical capacity. The results of modulated 820 nm reflection analysis showed that DCMU and MV treatments had low re-reduction of P700 and plastocyanin (PSI). However, the strawberry leaf discs pretreated with ALA exhibited high re-reduction of PSI under DCMU and MV treatments. The results of this study suggest that the improvement of photosynthesis by ALA in strawberry was not only related to PSII, but also to PSI and electron transfer chain.  相似文献

The effect of four different NaCl concentrations (from 0 to 102 mM NaCl) on seedlings leaves of two corn (Zea mays L.) varieties (Aristo and Arper) was investigated through chlorophyll (Chl) a fluorescence parameters, photosynthesis, stomatal conductance, photosynthetic pigments concentration, tissue hydration and ionic accumulation. Salinity treatments showed a decrease in maximal efficiency of PSII photochemistry (Fv/Fm) in dark-adapted leaves. Moreover, the actual PSII efficiency (ϕPSII), photochemical quenching coefficient (qp), proportion of PSII centers effectively reoxidized, and the fraction of light used in PSII photochemistry (%P) were also dropped with increasing salinity in light-adapted leaves. Reductions in these parameters were greater in Aristo than in Arper. The tissue hydration decreased in salt-treated leaves as did the photosynthesis, stomatal conductance (g s) and photosynthetic pigments concentration essentially at 68 and 102 mM NaCl. In both varieties the reduction of photosynthesis was mainly due to stomatal closure and partially to PSII photoinhibition. The differences between the two varieties indicate that Aristo was more susceptible to salt-stress damage than Arper which revealed a moderate regulation of the leaf ionic accumulation.  相似文献

In biological oceanography, it has been widely accepted that the maximum quantum yield of photosynthesis is influenced by nutrient stress. A closely related parameter, the maximum quantum yield for stable charge separation of PSII, (φ PSII )m, can be estimated by measuring the increase in fluorescence yield from dark-adapted minimal fluorescence (Fo) to maximal fluorescence (Fm) associated with the closing of photosynthetic reaction centers with saturating light or with a photosynthetic inhibitor such as 3′-(3,4-dichlorophenyl)-1′,1′-dimethyl urea (DCMU). The ratio Fv/Fm (= (Fm− Fo)/Fm) is thus used as a diagnostic of nutrient stress. Published results indicate that Fv/Fm is depressed for nutrient-stressed phytoplankton, both during nutrient starvation (unbalanced growth) and acclimated nutrient limitation (steady-state or balanced growth). In contrast to published results, fluorescence measurements from our laboratory indicate that Fv/Fm is high and insensitive to nutrient limitation for cultures in steady state under a wide range of relative growth rates and irradiance levels. This discrepancy between results could be attributed to differences in measurement systems or to differences in growth conditions. To resolve the uncertainty about Fv/Fm as a diagnostic of nutrient stress, we grew the neritic diatom Thalassiosira pseudonana (Hustedt) Hasle et Heimdal under nutrient-replete and nutrient-stressed conditions, using replicate semicontinuous, batch, and continuous cultures. Fv/Fm was determined using a conventional fluorometer and DCMU and with a pulse amplitude modulated (PAM) fluorometer. Reduction of excitation irradiance in the conventional fluorometer eliminated overestimation of Fo in the DCMU methodology for cultures grown at lower light levels, and for a large range of growth conditions there was a strong correlation between the measurements of Fv/Fm with DCMU and PAM (r2 = 0.77, n = 460). Consistent with the literature, nutrient-replete cultures showed consistently high Fv/Fm (∼0.65), independent of growth irradiance. Under nutrient-starved (batch culture and perturbed steady state) conditions, Fv/Fm was significantly correlated to time without the limiting nutrient and to nutrient-limited growth rate before starvation. In contrast to published results, our continuous culture experiments showed that Fv/Fm was not a good measure of nutrient limitation under balanced growth conditions and remained constant (∼0.65) and independent of nutrient-limited growth rate under different irradiance levels. Because variable fluorescence can only be used as a diagnostic for nutrient-starved unbalanced growth conditions, a robust measure of nutrient stressed oceanic waters is still required.  相似文献

The chlorophyll (Chl) fluorescence imaging technique was applied to cashew seedlings inoculated with the fungus Lasiodiplodia theobromae to assess any disturbances in the photosynthetic apparatus of the plants before the onset of visual symptoms. Two-month-old cashew plants were inoculated with mycelium of L. theobromae isolate Lt19 or Lt32. Dark-adapted and light-acclimated whole plants or previously labelled, single, mature leaf from each plant were evaluated weekly for Chl fluorescence parameters. From 21 to 28 days, inoculation with both isolates resulted in the significantly lower maximal photochemical quantum yield of PSII (Fv/Fm) than those for control samples, decreasing from values of 0.78 to 0.62. In contrast, the time response of the measured fluorescence transient curve from dark-acclimated plants increased in both whole plants and single mature leaves in inoculated plants compared with controls. The Fv/Fm images clearly exhibited photosynthetic perturbations 14 days after inoculation before any visual symptoms appeared. Additionally, decays in the effective quantum yield of PSII photochemistry and photochemical quenching coefficient were also observed over time. However, nonphotochemical quenching increased during the evaluation period. We conclude that Fv/Fm images are the effective way of detecting early metabolic perturbations in the photosynthetic apparatus of cashew seedlings caused by gummosis in both whole plants and single leaves and could be potentially employed in larger-scale screening systems.  相似文献
