Establishment of an agricultural water recycling system known as the wetland reservoir subirrigation system (WRSIS) results in the creation of two different types of wetlands adjacent to agricultural fields. Each WRSIS consists of one treatment wetland designed to process agricultural contaminants (WRSIS wetlands) and one storage wetland for holding subirrigation water (WRSIS reservoirs). Previous WRSIS related research has focused on the filtration ability and development of aquatic plants within WRSIS wetlands. The fauna of the WRSIS reservoirs and how its aquatic community structure compares with WRSIS wetlands is unknown. We compared fish, amphibian, and reptile community structure between WRSIS wetlands and reservoirs in northwestern Ohio. Fishes, amphibians, and reptiles were sampled by seining, hoop netting, and gee minnow trapping in three WRSIS wetlands and three WRSIS reservoirs in June of 2006, 2007, and 2008. No difference in species richness, abundance, percent fish, percent reptiles, fish abundance, or reptile abundance occurred between the smaller shallower WRSIS wetlands and the deeper larger WRSIS reservoirs. Percent amphibians and amphibian abundance was greater in WRSIS wetlands than reservoirs. Jaccard’s index scores ranged from 0 to 0.5 and indicated species composition differed between WRSIS wetlands and reservoirs. Our results assisted with the development of design and management criteria incorporating wetland size, hydrology, and upland habitat intended to enable WRSIS wetlands to function primarily as amphibian habitat and the reservoirs to function as fish habitat.  相似文献

通过研究杭州湾自然潮滩湿地和围垦湿地土壤有机碳含量及其分布格局,揭示湿地植被演替、外来物种入侵和围垦活动对土壤有机碳分布的影响.结果表明:潮滩湿地土壤表层有机碳含量在4.41~8.58 g·kg-1,平均值6.45 g·kg-1.不同植被类型下表层土壤有机碳表现为:芦苇(8.56±0.04 g·kg-1)>互花米草(7.31±0.08 g·kg-1)>海三棱蔗草(5.48±0.29 g·kg-1)>光滩(4.47±0.09 g·kg-1);围垦湿地表层土壤有机碳表现为:20世纪60年代(7.46±0.25 g·kg-1)>2003年(5.12±0.16 g·kg-1)>20世纪80年代(1.96±0.46 g·kg-1),即土壤有机碳含量随围垦时间延长表现为先降低后升高的趋势;土壤有机碳在垂直剖面上均表现为由表向下逐渐降低的趋势.潮滩湿地和围垦湿地的土壤有机碳与pH呈显著负相关,与总氮呈显著正相关,表明在土壤中氮主要以有机氮的形态存在.潮滩湿地有机碳与碳氮比相关性不明显,而围垦湿地具有显著正相关性,说明围垦利用对湿地土壤碳氮比产生了一定影响.研究表明,潮滩湿地土壤固碳能力随着植物群落演替逐步增强,而外来入侵种互花米草的大量入侵和扩散将有可能降低潮滩湿地生态系统土壤的储碳功能.围垦引起的土壤水分、颗粒组成的变化以及耕作、土地利用和利用历史是影响围垦湿地土壤有机碳分布的主要原因.  相似文献

As a commensal and opportunistic pathogen, Candida albicans possesses a range of determinants that contribute to survival, persistence and virulence. Among this repertoire of fitness and virulence attributes are iron acquisition factors and pathways, which allow fungal cells to gain this essential mineral in the iron-poor environment of the host. The aim of this review is to present the strategies used by C. albicans to exploit host iron reservoirs and their impact on C. albicans pathogenicity. Because iron in the human host is mostly linked to host proteins, pathogens such as C. albicans must possess mechanisms to gain iron from these proteins. Here, we introduce the most important groups of human proteins, including haemoglobin, transferrin, lactoferrin and ferritin, which contain iron and that are potential iron sources for invading microorganisms. We then summarize and discuss the known and proposed strategies by which C. albicans exploits or may exploit iron from host proteins and compare these with strategies from other pathogenic microorganisms.  相似文献

The Ramsar Convention has gradually expanded the scope of the term ‘wetland’ to bring under its umbrella all kinds of inland freshwater (and saline) ecosystems as well as many marine ecosystems. It is not possible to develop a common framework for the study, management or policy of such a large and divergent assemblage of habitats with water being a single shared feature. In this paper, I argue that wetlands are distinct from deep open water systems such as rivers, lakes and reservoirs. The restriction of macrophytes (except the free floating plants like salvinia and water hyacinth) to shallow water habitats helps distinguish between wetlands and deep water systems. Following an ecosystem service approach, I discuss that wetlands are generally characterized by the occurrence of macrophytes, which critically contribute to their provisioning, regulating, supporting and cultural ecosystem services that differ significantly from those of the microphyte (phytoplankton)-dominated deep water habitats. I argue that wetlands do lie adjacent to deep and open water systems (including large rivers), which interact with them regularly and influence their biodiversity, hydrology, water quality and functioning, depending upon their relative areal extent and characteristics of the macrophyte community, but that only the littoral zones between the mean highest and lowest water levels (and stream banks and the floodplains beyond them in the case of rivers) should be treated as wetlands. Shallow lakes devoid of macrophytes because of eutrophication are degraded wetlands that need to be restored.  相似文献

Spatial patterns of planktonic production and respiration in the surface mixed layer were examined in eight Texas, USA reservoirs to test the hypothesis that P:R ratios are lowest in upreservoir inflow zones and highest in downreservoir open-water zones, as predicted by the heuristic reservoir zonation model. We measured summer planktonic metabolism with light–dark bottles and physical–chemical conditions in epilimnetic water at 85 sites distributed among sixteen longitudinal transects within the eight reservoirs (2 transects per reservoir). Volumetric production and plankton biomass were lowest in the open-water zones and increased upreservoir; however, that pattern was reversed for areal production due to greater photic depths at open-water sites. Volumetric respiration was similar in the three zones; however, corresponding planktonic P:R ratios in the surface mixed layer were significantly lower at open-water sites, which is opposite than hypothesized. Based on linear regressions of production and respiration rates on chlorophyll a, open-water sites were net heterotrophic during the summer regardless of trophic state; whereas inflow and mid-reservoir zone sites were heterotrophic when chlorophyll concentrations were respectively less than 9.5 and 35?mg?m?3. Although variation among reservoirs was high, five of the eight reservoirs had inflow zones that were net carbon sinks while seven had open-water zones that were carbon sources. Mean (±standard error) carbon flux rates of inflow, mid-reservoir, and open-water zones were ?0.22?±?0.12 (C sink), 0.39?±?0.44 (moderate C source), and 1.33?±?0.50 (strong C source) g C m?2 day?1 respectively. Inflow and mid-reservoir zones comprised approximately 45% of the total reservoir area studied. Therefore, omitting their contribution as often done when a single open-water site is sampled may substantially overestimate reservoir carbon flux.  相似文献

对湖库湿地进行生态健康评价,可为湖库湿地生态系统修复和管理提供决策依据。本文以天津市为例,选取20个典型的湖库湿地采样点,基于2018年8—9月期间调查获取的物理、化学和生物群落指标(浮游动物、浮游植物、底栖动物、鱼类、水生大型植物、河岸带植物)数据,构建包含物理完整性、化学完整性和生物完整性在内的生态完整性指数(IEI)对采样点生态健康状况进行评价。根据栖息地环境质量(QHEI)、水质状况和人类活动干扰3方面选取参照点,采用标准化方法筛选候选指标,应用层次分析法计算各指标权重,最终得出天津市典型湖库湿地生态完整性评价结果。结果表明: 1)所有样点中,“健康”点位占5.0%,“较好”点位占20.0%,“一般”点位占35.0%,“较差”点位占30.0%,“差”点位占10.0%。天津市典型湖库湿地生态健康状况整体处于一般水平,呈现出西部优于东部的趋势,空间差异显著;2)基于栖息地评分、水质状况与人类活动干扰相结合选取参照点是可行的,依据水质指标可降低选择参照点时的主观性;3)适用性验证结果表明,IEI可较好表征各点位的健康状况,区分效率明显,适用于评价研究区湖库湿地生态健康状况。  相似文献

Habitat management and planning strategies for nonbreeding ducks are focused on providing enough energy to support a desired number of individuals. Therefore, regional estimates of energy availability for nonbreeding ducks are required to determine if sufficient habitat exists for them. I used core sampling to estimate food and energy density in 6 types of water bodies (i.e., actively and passively managed emergent wetlands, playas, small and large reservoirs, and sloughs) in northeastern Colorado, USA, during 3 sampling occasions throughout 2 nonbreeding seasons, 2015–2016 and 2016–2017. Also, I used precise depth measurements to estimate the percentage of each site that was shallow enough to facilitate feeding by dabbling ducks as a way to correct overall energy density to reflect availability to ducks. Emergent wetlands contained the greatest food and energy density, followed by playas and sloughs, and reservoirs contained little food or energy. Fall depletion of food was greatest in actively managed emergent wetlands and spring depletion was greatest in sloughs and passively managed emergent wetlands. Mean percentage of passively managed emergent wetlands, actively managed emergent wetlands, small reservoirs, large reservoirs, and sloughs shallower than 50 cm was 37%, 77%, 10%, 4%, and 83%, respectively. Incorporating these estimates into the energetic carrying capacity model developed by the Playa Lakes Joint Venture for eastern Colorado resulted in a 54% decrease in overall duck energy day estimates, which is below what is needed to support population goals. This research identifies the need for additional wetland restoration in eastern Colorado to meet energy requirements of nonbreeding ducks and provides information to conservation planners to make more informed decisions about the extent and location of wetland restoration activities. © 2020 The Authors. Wetland restoration aims to recreate or enhance valuable ecosystem services lost during wetland destruction. Regaining wetland ecosystem services depends on restarting basic wetland functions, like carbon (C) storage, which are unmeasured in many Wetlands Reserve Program (WRP) restoration sites. We collected soil and plant data from 17 WRP sites in western New York that were used for tillage or non-tillage agriculture and then actively restored as isolated depressional wetlands by excavating basins and disabling drainage systems. Sites had been restored for 0–15 years when sampled in August-October 2010. We analyzed data as chronosequences and tested whether soil and vegetation parameters in restored wetlands, over time, (1) departed from pre-restoration baselines, estimated using active agricultural fields paired to each WRP site, and (2) converged towards “natural” benchmarks, estimated from four naturally-occurring wetlands. Restored WRP soils remained similar to agricultural soils in organic matter, density, moisture, and belowground plant biomass across chronosequences, indicating negligible C storage and belowground development for 15 years following restoration. Soil changes were limited in sites restored after both tillage and non-tillage agriculture and throughout the upland meadow, emergent shoreline, and open-water habitat zones that characterize these sites. Many plant metrics like aboveground biomass matched natural wetlands within 15 years, but recovered inconsistently among tilled and untilled sites and across all habitat zones, suggesting land-use history impacts and/or zonation effects. Disparities in recovery times exists between vegetation, which can respond quickly to wetland restoration, and underlying soils, which show limited signs of recovery 15 years after being restored.  相似文献

Lipolytic activity of 40 strains of Candida spp. was tested on API ZYM system and on RPMI agar plates supplemented with 1% Tween 40. Lipolytic activity was indicated by opaque zones around the inoculum cylindrical holes were punched in the medium. Clearing of the medium around the bacterial colonies indicated that an isolate produce lipase. Only 4 (21.1%) strains of C. albicans, and 3 (14.1%) strains of non-C. albicans which hydrolyzed 2-naftylomirystylan by use of the API ZYM system was observed. In contrast, 16 (78.9%) strains of C. albicans and 17 (80.7%) strains of non-C. albicans produced lipases on the agar plate using RPMI agar plates supplemented with 1.0% Tween 40. Determination oflipase activities with the API ZYM system were in no agreement with lipase tests in RPMI supplemented with Tween 40. Our study verify greater usefulness of RPMI supplemented with Tween 40 for detection of lipolytic enzymes of Candida species in comparison to the API ZYM.  相似文献

 2003年6~9月采用静态箱_气相色谱法,对三江平原生长季不同淹水条件下沼泽湿地CH4、N2O的排放进行了同步对比研究,并探讨了影响气体排放的主要影响因素。结果表明, 生长季沼泽湿地CH4和N2O排放具有明显的时空变化特征。长期淹水的毛果苔草(Carex lasiocarpa)和漂筏苔草(Carex pseudocuraica)植物带CH4的平均排放强度分别为259.2和273.6 mg•m-2•d-1,高于季节性淹水的小叶章(Deyeuxia angustifolia)植物带的排放强度(38.16 mg•m-2•d-1)(p<0.00 0 1);而生长季N2O的平均排放强度分别为0.969、0.932 和0.983 mg•m-2•d-1, 植物带间无显著差异(p=0.967)。相关分析表明,气温和5 cm深地温对沼泽湿地CH4生长季排放通量的影响较大,而水位则是影响长期淹水沼泽N2O排放通量的主要因素;不同类型湿地间CH4平均排放强度的差异主要受水位的控制,而强烈的还原环境可能是导致不同类型湿地具有近似的N2O排放强度的原因。  相似文献

Hydraulic residence time (HRT) is one of the key design parameters controlling the removal efficiency of contaminants and nutrients in stormwater and wastewater wetlands. The paper presents a new approach to the estimation of HRT using the variable residence time (VART) model. The VART model is employed to simulate the major processes (including advection, dispersion, and transient storage of contaminants/nutrients in vegetated zones) affecting HRT and thereby to produce a hydraulic residence time distribution (HRTD) for a design wetland. The HRTD in combination with a moment-based method is then utilized to find a mean design HRT for the design wetland. Methods for estimation of parameters governing the HRTD are proposed. The new approach to HRT computation is demonstrated through a case study for the Tres Rios Demonstration (TRD) Wetlands in Arizona, USA. Modeling results show that the design HRTs for the Hayfield wetland (H1) and the Cobble wetlands (C1 and C2) are 4.04, 4.66, and 2.65 days, respectively. The computed HRTs agree well with those reported by previous studies, confirming the efficacy of the new approach to hydraulic design of constructed wetlands.  相似文献

The mode of action of aqueous garlic extract (AGE) was studied in Candida albicans. The minimum inhibitory concentration (MIC) of AGE against six clinical yeast isolates ranged between 0.8 and 1.6 mg ml-1. Scanning electron microscopy and cell leakage studies showed that garlic treatment affected the structure and integrity of the outer surface of the yeast cells. Growth of C. albicans in the presence of AGE affected the yeast lipid in a number of ways: the total lipid content was decreased; garlic-grown yeasts had a higher level of phosphatidylserines and a lower level of phosphatidylcholines; in addition to free sterols and sterol esters, C. albicans accumulated esterified steryl glycosides; the concentration of palmitic acid (16:0) and oleic acid (18:1) increased and that of linoleic acid (18:2) and linolenic acid (18:3) decreased. Oxygen consumption of AGE-treated C. albicans was also reduced. The anticandidal activity of AGE was antagonized by thiols such as L-cysteine, glutathione and 2-mercaptoethanol. Interaction studies between AGE and thiols included growth antagonism, enzymic inhibition and interference of two linear zones of inhibition. All three approaches suggest that AGE exerts its effect by the oxidation of thiol groups present in the essential proteins, causing inactivation of enzymes and subsequent microbial growth inhibition.  相似文献

Microcystins (MCs) are cyclic heptapeptides produced by cyanobacteria present in water contaminated reservoirs. Reported toxic effects for microcystins are liver injury and tumour promotion. In this study, we evaluated the effects of two MCs, MC-LR and [Asp(3)]-MC-LR, on human neutrophil (PMN). We observed that even at concentrations lower than that recommended by World Health Organization for chronic exposure (0.1 nM), MCs affect human PMN. Both MCs have chemotactic activity, induce the production of reactive oxygen species, and increase phagocytosis of Candida albicans. MC-LR also increased C. albicans killing. The effect of MCs on PMN provides support for a damage process mediated by PMN and oxidative stress, and may explain liver injury and tumour promotion associated to long-term MCs exposures.  相似文献

Aims:  This study investigated the anti-fungal activity of coptisine on Candida albicans growth.
Methods and Results:  The metabolic power-time curves of Candida albicans growth at 37°C affected by coptisine were measured by microcalorimetry using an LKB-2277 Bioactivity Monitor with stop-flow mode. Then, the diameter of inhibitory zones in the agar layer was observed using agar cup method, and the minimal inhibitory concentration (MIC) of coptisine on Candida albicans growth was determined by serial dilution method. From the principal component analysis on nine quantitative parameters obtained from the power-time curves, we could easily evaluate the anti-fungal activity of coptisine by analysing the change of values of the main two parameters, growth rate constant k and maximum power output in the log phase P m, log. The results showed that coptisine had strong anti-fungal activity: at a low concentration (45  μ g ml−1) began to inhibit the growth of Candida albicans and at a high concentration (500  μ g ml−1) completely inhibited Candida albicans growth. Coptisine gave big inhibitory zones with diameters between 11 and 43 mm within test range, and the MIC of it was 1000  μ g ml−1.
Conclusions:  Coptisine had strong anti-fungal activity on Candida albicans growth. The method of microcalorimetry applied for the assay of anti-fungal activity of coptisine was quantitative, sensitive and simple.
Significance and Impact of the Study:  This work will provide useful information for the development of chemical biology policy in the use of anti-microbials in food and drug production.  相似文献

Lippia (Phyla canescens: Verbenaceae) is a serious weed of wetlands, riparian zones and floodplains, particularly in eastern Australia where many Ramsar wetlands are threatened by hydrological changes precipitated by soil-accreting lippia mats. Enriched genomic DNA libraries were used to develop nine informative microsatellite markers. These markers will be valuable tools to understand the genetic structure of the lippia populations in different regions throughout the world.  相似文献

Several Candida albicans morphological mutants were obtained by a procedure based on a combined treatment with nitrous acid plus UV irradiation and a double-enrichment step to increase the proportion of mutants growing as long filamentous structures. Altered cell morphogenesis in these mutants correlated with an altered colonial phenotype. Two of these mutants, C. albicans NEL102 and NEL103, were selected and characterized. Mutant blastoconidia initiated budding but eventually gave rise to filamentous hypha-type formations. These filaments were long and septate, and they branched very regularly at positions near septa. Calcofluor white (which is known to bind chitin-rich areas) stained septa, branching zones, and filament tips very intensely, as observed under the fluorescence microscope. Wild-type hybrids were obtained by fusing protoplasts of strain NEL102 with B14, another morphological mutant previously described as being permanently pseudomycelial, indicating that genetic determinants responsible for the two altered phenotypes are different. The mutants characterized in this work seemed to sequentially express the morphogenic characteristics of C. albicans, from blastoconidia to hyphae, in the absence of any inducer. Further characterization of these strains could be relevant to gain understanding of the genetic control of dimorphism in this species.  相似文献

We studied patterns in avian cholera mortality, the presence of Pasteurella multocida in the water or sediment, and water chemistry characteristics in 10 wetlands at the Sacramento National Wildlife Refuge Complex (California, USA), an area of recurrent avian cholera epizootics, during the winters of 1997 and 1998. Avian cholera outbreaks (>or=50 dead birds) occurred on two wetlands during the winter of 1997, but no P. multocida were recovered from 390 water and 390 sediment samples from any of the 10 wetlands. No mortality events were observed on study wetlands during the winter of 1998; however, P. multocida was recovered from water and sediment samples in six of the 10 study wetlands. The pH levels were higher for wetlands experiencing outbreaks during the winter of 1997 than for nonoutbreak wetlands, and aluminum concentrations were higher in wetlands from which P. multocida were recovered during the winter of 1998. Water chemistry parameters (calcium, magnesium, sodium, and dissolved protein) previously linked with P. multocida and avian cholera mortality were not associated with the occurrence of avian cholera outbreaks or the presence of P. multocida in our study wetlands. Overall, we found no evidence to support the hypothesis that wetland characteristics facilitate the presence of P. multocida and, thereby, allow some wetlands to serve as long-term sources (reservoirs) for P. multocida.  相似文献

Water quality in many stream catchments and river basins is severely impacted by nutrient enrichment as a result of agriculture. Water-resource managers worldwide are considering the potential role of riparian zones and floodplain wetlands in improving stream-water quality, as there is evidence at the site scale that such wetlands are efficient at removing nutrients from through-flowing water. However, recent studies have highlighted disadvantages of such use of wetlands, including emissions of greenhouse gases and losses of biodiversity that result from prolonged nutrient loading. Here, we discuss the water purification function of wetlands at the site and catchment scale and suggest ways in which these disadvantages could be overcome.  相似文献

Few mating-regulated genes have been characterized in Candida albicans. C. albicans FIG1 (CaFIG1) is a fungus-specific and mating-induced gene encoding a putative 4-transmembrane domain protein that shares sequence similarities with members of the claudin superfamily. In Saccharomyces cerevisiae, Fig1 is required for shmoo fusion and is upregulated in response to mating pheromones. Expression of CaFIG1 was also strongly activated in the presence of cells of the opposite mating type. CaFig1-green fluorescent protein (GFP) was visible only during the mating response, when it localized predominantly to the plasma membrane and perinuclear zone in mating projections and daughter cells. At the plasma membrane, CaFig1-GFP was visualized as discontinuous zones, but the distribution of perinuclear CaFig1-GFP was homogeneous. Exposure to pheromone induced a 5-fold increase in Ca(2+) uptake in mating-competent opaque cells. Uptake was reduced substantially in the fig1Δ null mutant. CaFig1 is therefore involved in Ca(2+) influx and localizes to membranes that are destined to undergo fusion during mating.  相似文献

Human luteinizing hormone (hLH) and the GTP analogue guanosine 5'-O-(3-thio)triphosphate stimulated morphogenesis in the dimorphic fungal pathogen Candida albicans. hLH bound specifically to subcellular fractions from C. albicans and stimulated adenylate cyclase activity in C. albicans microsomes. We provide the first demonstration of guanine-nucleotide-binding proteins (G-proteins) in C. albicans, and propose that the stimulation of C. albicans morphogenesis by hLH is mediated by a receptor-coupled adenylate cyclase system involving G-proteins.  相似文献
