Unhatched, but fully developed larvae of two solitary ascidians Ascidia mentula O.F. Müller and Ascidiella scabra (O.F. Müller) were incubated in a variety of adult tissue extracts, which an earlier study had shown to accelerate metamorphosis. Our study confirmed this observation and showed, furthermore, that tissue extracts induce tail resorption in unhatched larvae of Ascidiella. For both species, the most active induction of metamorphosis was obtained with tunic tissue extracts. The results indicate that chemical stimulation without the presence of tactile cues may initiate metamorphosis. We suggest that the ability of tunic extracts to induce metamorphosis may explain juvenile establishment on adult ascidians and their aggregated distribution found in nature. 相似文献

Glycine (Gly) is considered an obligatory co-agonist at NMDA receptors. Müller glia from the retina harbor functional NMDA receptors, as well as low and high affinity Gly transporters, the later identified as GLYT1. We here studied the regulation of Gly transport in primary cultures of Müller glia, as this process could contribute to the modulation of NMDA receptor activity at glutamatergic synapses in the retina. We demonstrate that neither glutamate stimulation nor the activation or inhibition of protein kinases A or C modify transport. In order to assess a function for Ca2+ and calmodulin (CaM)-dependent processes in the regulation of Gly transport, we explored the participation of Ca2+ concentration, CaM and Ca2+/CaM-dependent enzymes on Gly transporter activity. ATP and carbachol, known to induce Ca2+ waves in Müller cells, as well as caffeine-induced Ca2+ release from intracellular stores stimulated transport, whereas Ca2+ chelation by BAPTA-AM markedly reduced transport. CaM inhibitors W-7, ophiobolin A, R-24571 and trifluoperazine, induced a specific dose-dependent inhibition of transport. The inhibition of CaMKII by the autocamtide-2-related inhibitory peptide or by KN62 caused a decrease in transport which, in the case of KN62, was due to the abolition of the high affinity component, ascribed to GLYT1. Our results further suggest that Gly transport is under cytoskeletal control, as activation of calpain by major increases in [Ca2+]i induced by ionophores, as well as actin destabilization clearly inhibit uptake. We here demonstrate for the first time the participation of CaM, CaMKII and the actin cytoskeleton in the regulation of Gly transport in glia. Ca2+ waves are induced in Müller cells by distinct neuroactive compounds released by neurons and glia, hence the regulation of [Gly] by this system may be of physiological relevance in the control of retinal excitability. 相似文献

Summary Numerous immunochemical and immunohistochemical studies have shown a wide distribution of several families of neuropeptides in invertebrates as well as vertebrates. There are relatively few data available for Annelida: Polychaeta. Therefore, we undertook an immunohistochemical investigation in the marine worm Nereis.Among the vertebrate type antibodies tested, those against met-enkephalin, LH-RH, vasopressin, oxytocin and ACTH had negative or only very slight effects. Slight to moderate reactions were obtained for VIP, SRIF, CRF, GRF, and leu-enkephalin. Moderate to very strong responses were found with anti-CCK/gastrin, -substance P, and --MSH sera. Immunopositive reactions were usually observed in the entire CNS (except, until now, in neurosecretory cells, type II, in nuclei 20, and in nerve fibres located in the infracerebral neurohemal area). The immunoreactivity was, however, more or less abundant according to different CNS regions. For example, it appeared that the immunostaining for CRF is more important in the VNC while the leu-enkephalin family is more abundant in the brain (particularly in fuchsinophilic neurosecretory cells, type I, in nuclei 20). Moreover, several vertebrate type peptides (such as CRF/GRF and CCK/gastrin) may coexist in a single neurone. Several antisera may elicit a positive reaction in some specific area (for example, substance P in the nuchal organ; SRIF in oocytes; CCK/gastrin in the gastrointestinal tract).Nothing is known about the role of the different substances immunologically detected in Nereis. It is suggested that CCK/gastrin-, -MSHand substance P-like materials transmit external stimuli to neurosecretory centres located in the caudal part of the brain.Principal Abbreviations used ACTH

Cells of Müller glia and microglia react to neuronal injury in glaucoma. The change to a reactive phenotype initiates signaling cascades that may serve a neuroprotective role, but may also proceed to promote damaging effects on retinal neurons. Both effects appear to occur most likely in parallel in glaucoma, but the underlying mechanisms and signaling pathways that specifically promote protective versus destructive roles of reactive glial cells are mostly unclear. More research is needed to understand the homeostatic signaling network in which retinal glia cells are embedded to maintain or restore neuronal function after injury. 相似文献

The glutathione content of retinal Müller (glial) cells: effect of pathological conditions 总被引:2,自引:0,他引:2
Maintenance of isolated retinal Müller (glial) cells in glutamate-free solutions over 7 h causes a significant loss of their initial glutathione content; this loss is largely prevented by the blockade of glutamine synthesis using methionine sulfoximine (5 mM). Anoxia does not reduce the glutathione content of Müller cells when glucose (11 mM), glutamate and cystine (0.1 mM each) are present. In contrast, simulation of total ischemia (i.e., anoxia plus removal of glucose) decreases the glutathione levels dramatically, even in the presence of glutamate and cystine. Less severe effects are caused by high extracellular K+ (40 mM). Reactive oxygen species are generated in the retina under various conditions, such as anoxia, ischemia, and reperfusion. One of the crucial substances protecting the retina against reactive oxygen species is glutathione, a tripeptide constituted of glutamate, cysteine and glycine. It was recently shown that glutathione can be synthesized in retinal Müller glial cells and that glutamate is the rate-limiting substance. In this study, glutathione levels were determined in acutely isolated guinea-pig Müller cells using the glutathione-sensitive fluorescent dye monochlorobimane. The purpose was to find out how the glial glutathione content is affected by anoxia/ischemia and accompanying pathophysiological events such as depolarization of the cell membrane. Our results further strengthen the view that glutamate is rate-limiting for the glutathione synthesis in glial cells. During glutamate deficiency, as caused by e.g., impaired glutamate uptake, this amino acid is preferentially delivered to the glutamate-glutamine pathway, at the expense of glutathione. This mechanism may contribute to the finding that total ischemia (but not anoxia) causes a depletion of glial glutathione. In situ depletion may be accelerated by the ischemia-induced increase of extracellular K+, decreasing the driving force for glutamate uptake. The ischemia-induced lack of glutathione is particularly fatal considering the increased production of reactive oxygen species under this condition. Therefore the therapeutic application of exogenous free radical scavengers is greatly recommended. 相似文献

The seasonal changes in body weight, DNA, total carbohydrate, total lipid, and two nitrogen fractions are described for Verruca stroemia (O. F. Müller). Events are dominated by the fact that the spring brood is virtually synchronous throughout the population. Values for all the above rise during the autumn and early winter and fall at copulation during February as the animals are fertilized. Subsequently there may be some storage of nutrients which is later utilized. In general, there are only small changes in all these constituents during the summer. Although this species has escaped from the limitation of a single annual brood the seasonal changes in weight and composition closely resemble those in Balanus balanoides and B. balanus although their phasing is different. 相似文献

Glial cells are thought to protect neurons from various neurological insults. When there is injury to retina, Müller cells, which are the predominant glial element in the retina, undergo significant morphological, cellular and molecular changes. Some of these changes reflect Müller cell involvement in protecting the retina from further damage. Müller cells express growth factors, neurotransmitter transporters and antioxidant agents that could have an important role in preventing excitotoxic damage to retinal neurons. Moreover, Müller cells contact to endothelial cells to facilitate the neovascularization process during hypoxic conditions. Finally, recent studies have pointed to a role of Müller cells in retina regeneration after damage, dedifferentiating to progenitor cells and then giving rise to different neuronal cell types. In this article we will review the role of Müller glia in neuroprotection and regeneration after damage in the retina. 相似文献

The ionic regulation of the haemolymph of larvae of Aeshna cyanea (Müller) was studied by means of two types of experiments. In the first the change in internal ionic composition was followed as a function of the time spent in a given experimental medium. These experiments led to the conclusion that: 1. the haemolymph composition does not change when larvae are starved in tap water for 10 days; 2. the haemolymph ionic concentrations (Na and Cl) have initial marked increase when the animals are kept in hypertonic media of diluted sea water; after 80 hours however, both concentrations stay constant. In a second series of experiments the internal ionic concentration was compared to a series of different concentrations of external media. From this, the relation between the internal and external ionic concentration was elaborated : in hypotonic media the internal Na and Cl concentrations stay constant, in hypertonic media there is a parallelism between the increase of the external concentration and haemolymph concentration, the internal Na concentration being always slightly hypertonic, the Cl concentration markedly hypotonic. Finally, when larvae are placed in glass distilled water, the internal Na and Cl concentrations begin to decrease after 60 hours. 相似文献

The reproductive morphology of Dumontia incrassata (O. F. Müller) Lamouroux is described from New Hampshire, U.S.A. populations. Mature carpogonial and auxiliary branches are usually 5 and 6 celled, respectively, but both may range from 4 to 9 cells. Auxiliary branches are more numerous than carpogonial branches; however, the ratio of carpogonial to auxiliary branches increases towards the apex of the frond. A linear relationship exists between the position of the generative auxiliary cell and the number of cells in the auxiliary branches. In most cases the third cell from the top of the auxiliary branch becomes the generative auxiliary cell. The probabilities for different positions of the generative auxiliary cells are summarized. During spermatangial development, four mother cells are initially produced per cortical cell initial; subsequently, successive spermatangia are cut off obliquely and on opposite sides of each mother cell. The frequency of occurrence of Haementeria costata in the waters of north-eastern Poland is 66.5%. This leech is usually found in shallow water zones frequented by water fowl and mammals.The different numbers of individuals taking food and different amounts of food consumed at one meal during successive feedings resulted in individuals of a cohort reaching sexual maturity at the age of 1, 2, and 3 years and surviving for 1.5, 2.5 and 3.5 years, respectively. Seasonal variations of pollution biomarkers have been studied in two populations of Corbicula fluminea (Müller) for 2 years. Clams were collected from the Sanguinet lake and the Dronne river (Southwest France), sites characterised by water of excellent and passable quality, respectively. Biological parameters studied were enzymatic activity (catalase, propionylcholinesterase, glutathione S-transferase, NADPH-cytochrome c reductase and NADH-cytochrome c reductase activities) and indicators of physiological (protein, carbohydrate, lipid and ash contents, fresh and dry tissue weights, condition factor) and reproductive (amount of oocytes present in gonads and clams incubating larvae in gills) status. Environmental parameters (e.g. water temperature, chlorophyll a concentration) were also monitored. Effects of site, sampling date and season (from spring to winter) were studied, as well as relationships between abiotic and biotic factors. Finally, an index of variability was calculated for each biomarker. Catalase, propionylcholinesterase, NADH-cytochrome c reductase and lipids experienced the most pronounced seasonal fluctuations, whereas glutathione S-transferase, NADPH-cytochrome c reductase and proteins suffered less pronounced ones. For some parameters, results differed depending on the site. Most seasonal changes were related to the reproductive cycle and/or water temperature. Implications of such variations in the use of pollution biomarkers in environmental surveys are discussed. 相似文献

Summary The sequence of the appearance of mixis in the rotifer Brachionus plicatilis was followed among the descendents of amictic rotifers transferred from a high salinity media (40 S) to a low one (9 S). All the neonates that hatched from the amictic eggs, after being transferred to a low salinity, were amictic. Each one of these neonates was cultured individually and its offspring removed periodically every 8–10 h. It was observed that throughout their reproductive phase, these parental females retained their potential to produce either mictic or amictic offspring. All the first produced neonates developed into amictic females, but among those produced later, three patterns were prevalent. The prevalent pattern (type A) was one in which the probability of a neonate being mictic increased towards the middle of the parents' reproductive phase and was followed by a slow decline. In the second pattern (type B), the probability of a daughter being mictic was constant throughout the parents' reproductive phase. It is suspected that the quality of food supplied to the rotifers determines the appearance of patterns, A, B or C. It is postulated that the innate capacity of rotifers to undergo mixis is genetically controlled, while its expression is modulated by environmental conditions. 相似文献

The radular morphology of the patellid species Testudinalia testudinalis (O. F. Müller, 1776) from the White Sea was studied using light, electron, and confocal microscopy. The radula is of the docoglossan type with four teeth per row and consisting of six zones. We characterize teeth formation in T. testidinalis as follows: one tooth is formed by numerous and extremely narrow odontoblasts through apocrine secretion; this initially formed tooth consists of numerous vesicles; the synthetic apparatus of the odontoblasts is localized in the apical and central parts of the cells throughout the cytoplasm and is penetrated by microtubules which are involved in the transport of the synthesized products to the apical part of the odontoblast; the newly formed teeth consist of unpolymerized chitin. Mitotic activity is located in the lateral parts of the formation zone. The first four rows contain an irregular arrangement of teeth, but the radular teeth are regularly arranged after the fifth row. The irregularly arranged teeth early on could be a consequence of the asynchronous formation of teeth and the distance between the odontoblasts and the membranoblasts. The morphological data obtained significantly expands our knowledge of the morphological diversity of the radula formation in Gastropoda. 相似文献
