Hemolymph plasma of the snail Helix aspersa which inhibits maturation and reproduction of its mantle cavity-inhabiting nematode, Rhabditis maupasi, was separated biochemically for the active proteinaceous component. Isolation of the active inhibitor was performed using ion-exchange chromatography in combination with subsequent gel filtration. The isolated peaks were bio-assayed in vitro on nematode larvae. The fractions harboring inhibitory protein suppressed larval growth and adult reproduction in vitro. The isolated fraction was purified by gel filtration and characterized on the basis of a single band on starch zone electrophoresis and positive reaction only with folin-phenol reagent.  相似文献

Required for a nematode's reproduction in a chemically defined medium are the nine mammalian essential amino acids (sensu strictu). Needed in addition to lysine, tryptophane, histidine, phenylalanine, leucine, isoleucine, threonine, methionine, and valine is arginine which is marginally essential for mammals. Tyrosine, nonessential for mammals, is not absolutely required by the nematode but is beneficial in terms of onset time and quantity of reproduction. Aspartic acid, nonessential for the nematode and mammals, is toxic for adult nematodes at and above 4.8 mg/ml medium. The nematode, Neoaplectana glaseri, is parasitic in insect grubs but the strain used has been cultivated in species isolation on kidney slices continuously since (1944).  相似文献

The bacterium Photorhabdus luminescens is a symbiont of the entomopathogenic nematode Heterorhabditis bacteriophora. The nematode requires the bacterium for infection of insect larvae and as a substrate for growth and reproduction. The nematodes do not grow and reproduce in insect hosts or on artificial media in the absence of viable P. luminescens cells. In an effort to identify bacterial factors that are required for nematode growth and reproduction, transposon-induced mutants of P. luminescens were screened for the loss of the ability to support growth and reproduction of H. bacteriophora nematodes. One mutant, NGR209, consistently failed to support nematode growth and reproduction. This mutant was also defective in the production of siderophore and antibiotic activities. The transposon was inserted into an open reading frame homologous to Escherichia coli EntD, a 4'-phosphopantetheinyl (Ppant) transferase, which is required for the biosynthesis of the catechol siderophore enterobactin. Ppant transferases catalyze the transfer of the Ppant moiety from coenzyme A to a holo-acyl, -aryl, or -peptidyl carrier protein(s) required for the biosynthesis of fatty acids, polyketides, or nonribosomal peptides. Possible roles of a Ppant transferase in the ability of P. luminescens to support nematode growth and reproduction are discussed.  相似文献

Amino acids emitted and extracted from surface-sterilized larvae and adults of Heterodera glycines were identified by paper chromatography and quantitatively analyzed by column chromatography. Five amino acids (alanine, aspartic acid, glutamic acid, glycine and serine) were emitted by H. glycines larvae and eight others (asparagine, glutamine, leucine/isoleucine, lysine, methionine sulfoxide, threonine, tyrosine, valine/methionine) were found in extracts from crushed larvae.In addition to the amino acids emitted or extracted from larvae, four others were emitted by adults (γ-aminobutyric acid, histidine, phenylalanine, and proline). Four different amino acids (arginine, cystathionine, hydroxyproline, and ornithine) were found only in the extract from crushed adults. Greater quantities of alanine, aspartic acid and glycine were emitted than could be detected in nematode extracts suggesting selective emission.Subsamples of nematode populations were taken from growing plants 19, 26, 33, and 40 days after inoculation and extracted to determine whether changes in specific amino acid content correlated with aging. Proline content shifted most, increasing from 4.1% to 21.5% of the total amino acid complement from the 19th to the 40th days.  相似文献

Total methylated fatty acid patterns of various developmental stages (third-stage larvae (L3), L3 and fourth-stage larvae (L4) cultured in vitro, L4 and female and male adults derived from intestinal contents) of the porcine nodular worms Oesophagostomum dentatum and Oesophagostomum quadrispinulatum and their cultivation medium were analysed by gas chromatography using Microbial Identification computer software. Fatty acids ranging from C-12 to C-20 could be separated. For each stage and species, characteristic patterns were found. The most prevalent fatty acids were C-18. The freshly exsheathed larvae contained the greatest variety of fatty acids (including short-chain fatty acids C-12 to C-15) with approximately equal amounts of fatty acids with odd and even chain lengths, whereas more advanced stages consisted of a lower number of fatty acids with mostly even chain lengths >/=C-16. Intestinal stages contained less odd-numbered fatty acids and less branched fatty acids than others. In contrast to intestinal L4, cultivated L4 had high amounts of C-15:0 and C-17:0. Sheathed L3 contained more C-18 than freshly exsheathed ones, and medium incubated for 7 days in the presence of parasites contained C-13 to C-15 and monounsaturated C-16, but less C-18 and C-20:4 than fresh medium or medium incubated without worms. Based on the evaluation of stage- and species-specific fatty acid patterns random samples could be assigned to the correct stage and species. In a dendrogram based on fatty acid patterns the same stages of the two species formed the closest relationships, and the intestinal stages formed a clade distinct from the cultivated larvae and L3. All stages contained considerable relative amounts of arachidonic acid, the main precursor of eicosanoids. The fixed differences between species and stages indicate genetic regulation of fatty acid patterns, while environmental influences are mirrored by differences between cultivated and intestinal stages. Regulation of fatty acid patterns probably plays a role in worm physiology and host-parasite interaction.  相似文献

Evolution of pollen feeding in Heliconius has allowed exploitation of rich amino acid sources and dramatically reorganized life‐history traits. In Heliconius, eggs are produced mainly from adult‐acquired resources, leaving somatic development and maintenance to larva effort. This innovation may also have spurred evolution of chemical defence via amino acid‐derived cyanogenic glycosides. In contrast, nonpollen‐feeding heliconiines must rely almost exclusively on larval‐acquired resources for both reproduction and defence. We tested whether adult amino acid intake has an immediate influence on cyanogenesis in Heliconius. Because Heliconius are more distasteful to bird predators than close relatives that do not utilize pollen, we also compared cyanogenesis due to larval input across Heliconius species and nonpollen‐feeding relatives. Except for one species, we found that varying the amino acid diet of an adult Heliconius has negligible effect on its cyanide concentration. Adults denied amino acids showed no decrease in cyanide and no adults showed cyanide increase when fed amino acids. Yet, pollen‐feeding butterflies were capable of producing more defence than nonpollen‐feeding relatives and differences were detectable in freshly emerged adults, before input of adult resources. Our data points to a larger role of larval input in adult chemical defence. This coupled with the compartmentalization of adult nutrition to reproduction and longevity suggests that one evolutionary consequence of pollen feeding, shifting the burden of reproduction to adults, is to allow the evolution of greater allocation of host plant amino acids to defensive compounds by larvae.  相似文献

本文收集了虫草蝠蛾2—6龄五个龄期组幼虫,四组处于不同发育阶段的虫草菌丝体、被虫草菌感染后的僵虫体、全虫草体及子实体共计12组样品的氨基酸,微量元素分析资料,应用聚类分析方法对各样品变量的分类单元及其贡献值进行初步探讨。结果显示:各样品间的氨基酸、微量元素组成水平各不相同,5、4龄幼虫的总体组成水平在比例关系上更接近全虫草体,虫草菌丝体愈培养至晚期就愈接近子实体的组成。分析认为,5、4龄虫易被感染与其氨基酸、微量元素的组成背景值有关;以培养至晚期的菌丝体或有性生殖孢子更易感染幼虫。  相似文献

Biological and biochemical parameters of a flightless strain of Harmonia axyridis, fed on a pork liver-based artificial diet and on Ephestia kuehniella eggs as controls, were compared. The diet-grown larvae showed a significantly longer developmental time and a lower adult emergence rate compared to control larvae. The weights of the newly emerged adults were significantly higher for adults fed E. kuehniella eggs during their larval stages than fed the artificial diet. In contrast, larval food source had no effect on the duration of the pre-oviposition period or adult longevity. For adults fed on E. kuehniella eggs as larvae, a significantly longer pre-oviposition period, lower daily weight gain and fecundity were found for the diet-fed females compared to those fed on E. kuehniella eggs throughout the life span. The adult food source had no significant effect on longevity and fertility. Lower amino acid and fatty acid contents (in particular C16:1 and C18:3n-3) were found for the prepupae and newly emerged females obtained from diet-reared larvae compared to controls. Deficiencies in fatty acids C16:1 and C18:3n-3 were also observed in females obtained from E. kuehniella egg-reared larvae and fed on diet from adult emergence. The analyses of the foods showed deficiencies in artificial diet, especially for some amino and fatty acids. The results suggest a non-optimal composition of the artificial diet and some possibilities for its improvement. However, this polyphagous predator could be reared from first instar larvae to fully reproductive adults on a pork liver-based artificial diet.  相似文献

The nematode Caenorhabditis elegans arrests development at the first larval stage if food is not present upon hatching. Larvae in this stage provide an excellent model for studying stress responses during development. We found that supplementing starved larvae with ethanol markedly extends their lifespan within this L1 diapause. The effects of ethanol-induced lifespan extension can be observed when the ethanol is added to the medium at any time between 0 and 10 days after hatching. The lowest ethanol concentration that extended lifespan was 1 mM (0.005%); higher concentrations to 68 mM (0.4%) did not result in increased survival. In spite of their extended survival, larvae did not progress to the L2 stage. Supplementing starved cultures with n-propanol and n-butanol also extended lifespan, but methanol and isopropanol had no measurable effect. Mass spectrometry analysis of nematode fatty acids and amino acids revealed that L1 larvae can incorporate atoms from ethanol into both types of molecules. Based on these data, we suggest that ethanol supplementation may extend the lifespan of L1 larvae by either serving as a carbon and energy source and/or by inducing a stress response.  相似文献

The efficacy of soil treatments of three native entomopathogenic nematodes (Steinernema carpocapsae, S. feltiae and Heterorhabditis bacteriophora) against Tuta absoluta larvae, pupae and adults was determined under laboratory conditions. The effect of three insecticides commonly used against T. absoluta, in the survival, infectivity and reproduction of these nematode strains was also evaluated. When dropped into soil to pupate, soil application of nematodes resulted in a high mortality of larvae: 100, 52.3 and 96.7 % efficacy for S. carpocapsae, S. feltiae and H. bacteriophora respectively. No mortality of pupae was observed and mortality of adults emerging from soil was 79.1 % for S. carpocapsae and 0.5 % for S. feltiae. The insecticides tested had a negligible effect on nematode survival, infectivity and reproduction. No sublethal effects were observed. Infective juveniles that survived to insecticide exposition were able to infect Galleria larvae with no significant differences from the control. The Galleria larvae affected by the three insecticides tested served as suitable hosts for the infection and reproduction of the nematodes. These results suggest that larvae of T. absoluta, falling from leaves following insecticide application, could be suitable hosts for nematodes, thereby increasing their concentration and persistence in the soil.  相似文献

Heterodera betulae, a cyst-forming nematode, originally recovered from roots of river birch in Arkansas, appears to have a limited host range. Of 80 plant species in 20 families tested, only five species of Betula and Cleome spinosa supported reproduction. The minimum time for a complete life cycle was 52 days at 28 C. No reproduction occurred at 31 C or above, and development was very slow below 20 C. Successful population propagations from single larvae demonstrated that males were not necessary for reproduction.  相似文献

When any of the ten “rat essential” amino acids was omitted singly from a fully-defined synthetic dietary medium, newly-hatched Culex pipiens larvae were unable to develop to the second instar. With proline omitted, development was greatly retarded and survival to the adult stage reduced. Without aspargine (but not aspartic acid) growth and development ceased in most individuals before larval-pupal ecdysis, and no adults were obtained. These twelve amino acids are considered nutritionally essential for this mosquito. With glycine omitted singly, development was markedly retarded, but survival to the adult stage was not affected; thus this amino acid is required for good growth, but these experiments do not demonstrate it as essential. Single omission of alanine, aspartic acid, cysteine, glutamic acid or amide, serine or tyrosine had virtually no effect on development and they are therefore considered nutritionally non-essential. With diets containing the twelve culex-essential amino acids only, very little development occurred, but augmentation with either glycine or serine allowed growth and development almost as good as with the complete amino acid mixture. Augmentation of the essential twelve with alanine, cysteine, glutamic acid/amide, or tyrosine singly failed to improve development. The requirement for dietary asparagine shown by these studies appears to be unique among insects so far studied. In particular, another mosquito, Aedes aegypti, has no such requirement.  相似文献

【目的】了解金银花尺蠖Heterolocha jinyinhuaphaga Chu幼虫、蛹和成虫的主要营养成分,评价其营养价值,为金银花尺蠖的食用开发提供理论依据。【方法】采用国家标准检测方法,测定金银花尺蠖幼虫、蛹和成虫的水分、蛋白质、脂肪、总糖、氨基酸、脂肪酸、微量元素和维生素的含量,分析金银花尺蠖幼虫、蛹和成虫的氨基酸分、必需氨基酸指数、必需氨基酸与非必需氨基酸的比值、不饱和脂肪酸与饱和脂肪酸的比值,并与常见食物进行比较分析。【结果】结果表明,金银花尺蠖幼虫、蛹和成虫的水分含量分别为72.93%、64.92%和56.16%;蛋白质含量分别为50.64%、60.12%和48.38%;脂肪含量分别为21.98%、23.84%和20.60%;总糖含量分别为3.32%、5.35%和2.79%;总氨基酸含量分别为394.31、413.54和356.09 mg/g;必需氨基酸总含量分别为158.80、166.83、132.67 mg/g;必需氨基酸占总氨基酸的比例分别为40.27%、40.34%和37.26%;必需氨基酸与非必需氨基酸的比值分别为67.43%、67.62%和59.38%;必需氨基酸指数分别为1.02、1.07和0.87;不饱和脂肪酸的含量分别为68.45%、69.09%和66.29%,其P/S值分别为2.17、2.24和1.95;必需脂肪酸含量分别为34.35%、34.77%和32.71%;微量元素Zen的含量分别为11.29、98.42、66.98 mg/100 g,Fe的含量分别为32.31、19.84、13.04 mg/100 g,幼虫中Cu的含量为18.54 mg/100 g,蛹和成虫中均不含Cu;维生素以B6的含量最高,分别为7.93、25.82、26.21 mg/100 g。【结论】金银花尺蠖具有较高的营养价值和食用价值,具有极大的开发潜力。  相似文献

Three distinct types of storage hexamerins are expressed in the "last-instar" larvae of the rice moth, Corcyra cephalonica. A cDNA expression library was constructed from fat body-RNA and screened with a polyclonal antibody raised against purified hexamerin (SP2) of Corcyra cephalonica. Two slightly different "full-length" hexamerin cDNA clones (Hex2a and Hex2b) were isolated and sequenced. Both include open reading frames of 2109 bp which are translated into polypeptides of 703 amino acids with 92.5% identity. Signal peptides of 19 amino acids are present at the N-termini. The 684 amino acids native proteins have a high content of aryl groups (17.6%). According to both the criteria for amino acid composition and the phylogenetic analysis, Hex2a and Hex2b belong to the lepidopteran arylphorins. Northern blot studies revealed that the Hex2 genes are species- and tissue-specifically expressed in fat body cells of "last-instar" (= 5th) larvae.  相似文献

The root-knot nematode Meloidogyne spp. includes important plant pathogens worldwide. This study has considered nematode Meloidogyne javanica second stage larvae activity in the extracts of Pseudomonas fluorescens strains UTPF5 and cytotoxic effect of the strain on the nematode. The movement of second stage larvae of nematodes in water agar medium at four concentrations of bacterial extracts and second stage larvae mortality rate of hatching nematode and bacterial strains in vitro were affected. Different concentrates of the strain UTPF5 effect nematode larvae movement and disposal of the same. Bacterial extraction kills almost 100% of the larvae hatching after 24?h and a complete ban on egg hatch of biocontrol nematodes and nematode indicated that root-knot nematode larvae movement on the right attract the bacteria P. fluorescens to extract in the first place.  相似文献

Gap junction-enriched fractions were prepared from larvae of the tobacco budworm Heliothis virescens using the NaOH procedure in the presence or absence of protease inhibitors and were analyzed by SDS-PAGE, immunoblotting and EM immunocytochemistry. Protease inhibitor fractions contained a 48-kDa protein in addition to the 10 proteins in fractions with and without inhibitors. Three polyclonal antibodies were used as probes for gap junction plaques and proteins: R16, against an 40-kDa candidate gap junction protein from Drosophila melanogaster; R17, against the 40-kDa candidate gap junction protein from H. virescens; and R18AP, an affinity purified antibody against a consensus sequence of N-terminal amino acids 2–21 of the H. virescens 40-kDa protein. R16, R17, and R18AP stain the 40- and 48-kDa proteins, R16 and R18AP stain a 64-kDa protein, and R16 stains an 30-kDa protein in the absence of inhibitors. Inclusion of protease inhibitors had no effect on gap junction ultrastructure. R16 and R17 label gap junction plaques in crude membrane and NaOH fractions, whereas R18AP exhibits only a low level of reactivity with gap junctions in crude membrane fractions and none with gap junctions in NaOH fractions. The results show that the 30-, 40-, 48- and 64-kDa proteins are immunologically related and are associated with gap junctions in H. virescens, the N-terminus of the 40-kDa protein is relatively inaccessible or easily lost, and the 48-kDa protein is protease-sensitive.  相似文献

In social insects, colony nutrition depends upon the volume and quality of food distributed, ingested, and assimilated by its members. The ability of Solenopsis invicta workers and larvae to regulate the volume of food ingested individually has been well documented. In this paper, the ability of fire ant workers and larvae to regulate the quality and type of food ingested is demonstrated. Larvae displayed independent appetites for solid protein, amino acid solution, and sucrose solution. When larvae that had fed on one food type were switched to another, they fed on the second food type at rates characteristic of that food type, not of the volume of food previously ingested. Larvae preferred concentrated sucrose and amino acids solutions over dilute solutions. Larval fullness was thus a relative property, depending upon the nature of food as well as the volume ingested. The number of workers recruiting to food sites also depended upon food concentration and food type. Workers preferred sucrose to amino acids, concentrated to dilute solutions, and novel to accustomed food. The absence of protein in the worker diet rather than the presence of larvae caused workers to switch their preference from sugar to amino acids solutions. When the colony was offered sucrose and amino acids solutions simultaneously, individual workers ingested from one or the other site, but not both. Little mixing of crop contents occurred when workers solicited from one another inside the nest. Workers tended to regurgitate to larvae after ingesting amino acids and to other workers after ingesting sucrose. The mechanism regulating the distribution of protein pellets, which workers do not ingest, among larvae is unknown. In summary, colony nutrition was regulated by a chain of demand. Forager hunger determined the rate at which food flowed from the environment into the nest. Larval hunger and nest-worker hunger determined the rate and direction in which food moved within the nest.  相似文献

Free, peptide-bound and total amino acids were determined in symbiotic and aposymbiotic Sitophilus oryzae. The concentrations of free and peptide amino acids were higher, that of protein amino acids lower in aposymbiotic larvae (+30, +66 and −35%, respectively). In the free fraction, tyrosine concentration was the only one to be lower in aposymbiotic larvae (−41%) and could be a limiting factor for protein synthesis.In nymphs and adults, the compositions in symbiotic and aposymbiotic insects were very similar. A deficiency of tyrosine occurred however in aposymbiotic nymphs and of aspartate, glutamate, arginine, lysine and tyrosine in aposymbiotic adults. On the other hand, the composition in ovaries was unchanged.It is suggested that symbiotes could favour the synthesis of proteins in the larvae by increasing the efficiency of the conversion of phenylalanine in tyrosine.  相似文献
